Although dielectric relaxation can significantly affect the intrinsic fluorescence properties of a protein, usually it is fast compared to fluorescence timescales and needs to be slowed down by adding viscogens or lowering temperature before its impact on fluorescence can be studied. We report here a remarkable blue shift in fluorescence upon bimolecular quenching in the single-tryptophan thermostable protein Bj2S, the 2S seed albumin from Brassica juncea, at ambient temperature and viscosity. The magnitude of the blue shift ( approximately 5 nm at 50% quenching by acrylamide) is striking in a single-tryptophan protein and is attributed to a slowly relaxing dielectric environment in Bj2S from red edge excitation, steady-state polarization and time-resolved fluorescence experiments. Our results have important implications on interpretation of fluorescence of proteins with highly constrained backbones and in designing model systems for studying slow protein solvation dynamics using Trp fluorescence as the reporter probe.  相似文献

The effects of ultradry storage on the starch mobilization in maize (Zea mays L.) seed after aging were investigated. The results indicated that there were no significant differences in the content of ATP,starch, and soluble sugar, as well as the activity of amylase, between ultradried seeds and seeds stored at -20 ℃ during germination. These results were consistent with the higher level of vigor of the ultradried seed. Sieve tube introduction of a fluorescence dye (carboxyl fluoresceindiacetate) and laser confocal microscopy were used to study the development of plasmodesmata in the ultradried seeds. The results indicated that plasmodesmata developed well in ultradried seeds. Fluorescence analysis also showed that the fluorescence intensity in the radicle of ultradried seeds was stronger than that in seeds with a higher moisture content. This suggests that ultradry treatment has no adverse effects on the seeds. After seed imbibition, cell orgaelles could be resumed. It is concluded that ultradry seed storage is beneficial for maintaining seed vigor and that starchy mobilization proceeds regularly during germination.  相似文献

Seed security is of prime importance for agriculture. To protect true seeds from being faked, more secure dual anti-counterfeiting technologies for tobacco (Nicotiana tabacum L.) pelleted seed were developed in this paper. Fluorescein (FR), rhodamine B (RB), and magnetic powder (MP) were used as anti-counterfeiting labels. According to their different properties and the special seed pelleting process, four dual-labeling treatments were conducted for two tobacco varieties, MS Yunyan85 (MSYY85) and Honghua Dajinyuan (HHDJY). Then the seed germination and seedling growth status were investigated, and the fluorescence in cracked pellets and developing seedlings was observed under different excitation lights. The results showed that FR, RB, and MP had no negative effects on the germination, seedling growth, and MDA content of the pelleted seeds, and even some treatments significantly enhanced seedling dry weight, vigor index, and shoot height in MS YY85, and increased SOD activity and chlorophyll content in HHDJY as compared to the control. In addition, the cotyledon tip of seedlings treated with FR and MP together represented bright green fluorescence under illumination of blue light (478 nm). And the seedling cotyledon vein treated with RB and MP together showed red fluorescence under green light (546 nm). All seeds pelleted with magnetic powder of proper concentration could be attracted by a magnet. Thus, it indicated that those new dual-labeling methods that fluorescent compound and magnetic powder simultaneously applied in the same seed pellets definitely improved anti-counterfeiting technology and enhanced the seed security. Controlled hand pollinations and field observations were usedin conjunction with fluorescence and scanning electron microscopyand fruit and seed set to investigate the timing of stigma receptivity,pollen tube growth and self-incompatibility in relation to fertilityin B. coccinea. The species showed both protandry and partialself-incompatibility. Peak stigma receptivity as measured bypollen germination was recorded at 3 d after anthesis and maximumproduction of stigmatic exudate at 6 d. Pollen tubes reachedthe base of the style by 6 d after pollination. A 5 x 5 diallelexperiment was conducted and the results measured by pollentube growth. Self-pollinations generally resulted in poorertube growth than crosses and there was significant specificand general combining ability as well as reciprocal effects.Cross-pollination resulted in improved fruit set and seed toflower ratio over both selfing and open pollination. Spatiallimitations to fertility due to infructescence size were alsorecorded, but the combination of outcrossing mechanisms andspatial limitation did not entirely account for the low fertility.It is suggested that environmental conditions and the availabilityof resources may also exert an influence. 苇状羊茅内生真菌Neotyphodium coenophialum和多年生黑麦草内生真菌N.lolii对美国、新西兰等国家的畜牧业曾经造成过巨大损失。以N.coenophialum和N.lolii及其近似种N.huerfanum、N.chisosum、N.aotearoae、N.sp.共6种18个菌株,以及苇状羊茅和多年生黑麦草8个品种种子为供试材料,根据Tub-2基因设计了通用Taqman探针及引物,根据NC25基因设计了N.coenophialum和N.lolii的特异Taqman探针及共用引物,通过通用探针的单色荧光PCR和特异探针的双色荧光PCR,建立了N.coenophialum和N.lolii的菌丝及单粒种子稳定可靠、特异性强的荧光PCR检测方法,检测灵敏度达到单粒种子,使检测时间由至少一个月缩短至7~8个小时。  相似文献

The proteolytic activities of α-chymotrypsin, trypsin, pepsin, bromelain, and an extract from germinating pumpkin seeds were determined by their ability to effect the release of 1-anilino-8-naphthalenesulfonate bound to internal hydrophobic sites in intact protein substrates resulting in a decline in fluorescence. Casein, glyceraldehyde-3-P dehydrogenase, urease, catalase, pumpkin seed globulin, and bovine serum albumin enhanced the fluorescence of 1-anilino-8-naphthalenesulfonate sufficiently to be used as proteolytic substrates in this assay procedure. The activity of 1 μg chymotrypsin or trypsin and 100 ng pepsin could be easily detected by this method within 4 to 8 min depending upon the protein substrate. The digestive enzymes and bromelain exhibited activity against most if not all six of the protein substrates used. In contrast, the extract from germinating pumpkin seeds exhibited significant activity only against pumpkin seed globulin, with maximal activity at pH 7.4. Compared with the assay method for proteolytic activity utilizing ninhydrin analysis of the reaction products, this method was at least 10 times more rapid and gave significant detectable activity with much lower quantities of proteolytic enzyme.  相似文献

The seed coat structure and histochemistry of Abelmoschus esculentuswere studied by bright-field, fluorescence and scanning electronmicroscopy. The seed coat was typical of species of the Malvaceae.The endotesta cells had inner tangential walls which were verythick and autofluorescent. The occurrence of phenolic substancesat this level has been related to seed coat imposed dormancy.The palisade cells were composed of three differently shapedparts: an upper ‘prismatic part’, a medium ‘transitionpart’ and a lower ‘twisted part’. The formerwas rich in hydrophilic substances, the latter was lignified.The swelling of the ‘prismatic parts’ was relatedto seed coat cracks. The region controlling onset of water entrywas thought to be the chalazal area. Thanks to the presenceof a large amount of highly acidic polysaccharide, water wasable to penetrate from the permeable maternal tissue, throughthe chalazal cap and plug as far as the boundary between thepalisade and underlying mesophyll. During imbibition of watera kidney-shaped ‘blister’ was seen to rise, formedby separation of the palisade cells from an underlying singlelayer of subpalisade cells. The palisade layer forming the blisterroof showed the same histochemical characteristic of other seedregions. The single layer of the blister floor showed an affinitywith Toluidine Blue O and Alcian blue. Both blister roof andfloor were strongly autofluorescent. Abelmoschus esculentus (L.) Inhibition of protein glycation by skins and seeds of the muscadine grape   总被引:1,自引:0,他引:1  
The formation of advanced glycation end products (AGEs) leading to protein glycation and cross-linking is associated with the pathogenesis of diabetic complications. The inhibition of protein glycation by phenolic and flavonoid antioxidants demonstrates that the process is mediated, in part, by oxidative processes. In this study, the effects of seed and skin extracts of the muscadine grape on AGEs formation were examined. Seeds and skins were extracted (10% w/v) with 50% ethanol and incubated at 37 degrees C with a solution containing 250 mM fructose and 10 mg/ml albumin. After 72 h, fluorescence was measured at the wavelength pair of 370 and 440 nm as an index of the formation of AGEs. Both seed and skin extracts were found to be efficacious inhibitors of AGE formation. A 1:300 dilution of the seed extract decreased fluorescence by approximately 65%, whereas muscadine grape skin extract produced a 40% lowering. This difference correlates with the greater antioxidant activity found in muscadine seeds in comparison to skins, however, on a mass basis, the inhibitory activities of the seeds and skins were found to be nearly equivalent. Gallic acid, catechin and epicatechin, the three major polyphenols in the seeds, all significantly decreased the AGE product related fluorescence at a concentration of 50 microM. Neither muscadine seed extract nor skin extract inhibited the methylglyoxal-mediated glycation of albumin. These results suggest that consumption of the muscadine grape may have some benefit in altering the progression of diabetic complications.  相似文献

 利用叶片体内荧光测定技术,检测了南北5个地区华山松种源的荧光诱导动力学参数。结果表明,南方种源具有较强的最大荧光(Fm)和可变荧光(Fv),其光系统Ⅱ(PSⅡ)的潜在活性(Fv/Fo)与原初光能转换效率(Fv/Fm)也明显高于北方种源,并与其树高生长成正相关。另一方面,北方种源的非光化学荧光猝灭系数(QN)比南方种源要大,与它们的树高生长成反相关,而与纬度成正相关。这些实验结果说明,利用荧光诱导动力学技术,可以检测华山松的生长量潜力,在华山松种源选择中有较大的应用前景。  相似文献

The Maillard reaction and oxidative stress during aging of soybean seeds   总被引:8,自引:0,他引:8  
The chemical reactions that may lead to the loss of seed viability were investigated both during the accelerated aging and natural aging of soybeans ( Glycine max Merrill cv. Chippewa 64). Under conditions of accelerated aging (36°C and 75% RH), fluorescence of soluble proteins accumulated, which was closely correlated with the loss of seed germinability and vigor. We were able to show this correlation by using partially purified proteins for the assay. Fluorescence also increased in seeds under good storage conditions (5°C for up to 21 years), although there was a less significant correlation between seed viability and the accumulation of fluorescent products during the time of natural aging. The rise in protein fluorescence is interpreted as an increase of Maillard products. The carbonyl content of soluble proteins (a measure of the oxidative damage) did not change significantly during either accelerated aging or natural aging: however the elimination of carbonyls during germination seemed to be hindered in seeds that had poor germination. The Maillard reaction may be a consequence of the formation of reducing sugars through a gradual hydrolysis of oligosaccharides during aging. Preliminary evidence from the natural aging study showed that, when seeds were in the glassy state, the sugar hydrolysis was inhibited. These results suggest that the Maillard reaction and oxidative reaction may play an important role in seed deterioration.  相似文献

BACKGROUND: Xanthomonas campestris pv. campestris (Xcc) is a seed-transmitted plant pathogenic bacterium that causes black rot of crucifers. Seed lots and plants are screened for contamination with this pathogen using plating or serological assays. These methods, however, are time consuming and not very sensitive, respectively. Therefore, flow cytometry (FCM) was evaluated as a tool for the rapid detection and quantification of Xcc cells labeled with a mixture of specific fluorescein isothicyanate (FITC)-monoclonal antibodies (mAb) in pure culture, in mixed cultures of Xcc with either the common saprophyte Pseudomonas fluorescens (Psf) or a nonpathogenic X. campestris isolate (Xc), and in crude seed extracts. METHODS: The mAb 18G12, conjugated with FITC, was tested at dilutions of 1:50, 1:100, 1:200, and 1:400. For mixed suspensions of Xcc and Psf, mAb 18G12 was used at a dilution of 1:100. The combination of mAbs 18G12, 2F4, and 20H6, all conjugated with FITC, was used at a dilution of 1:100 for the detection and quantification of Xcc cells in mixed suspensions containing Xcc and Xc and in crude seed extracts. The analyses were performed with a Coulter EPICS XL-MCL flow cytometer, at low flow rate during 2 min. RESULTS: Using FCM, Xcc cells labeled with FITC-conjugated mAbs (18G12, 2F4, and 20H6) were detected and quantified rapidly at low numbers, i.e., 10(3) colony-forming units per milliliter in pure and in mixed cultures with Psf. The presence of the nonpathogenic Xc in the seed extracts did not interfere with the FCM results. Xcc cells were distinguished from the cells of other organisms and from small particles present in the seed extract based on the high-intensity fluorescence of the labeled cells. CONCLUSION: The application of FCM in combination with FITC-conjugated mAbs appears to be a promising technique for the detection and quantification of Xcc cells in seed extracts of crucifers.  相似文献

The seed protein IAP1 from bean (PvIAP1; Phaseolus vulgaris L.) that is modified by the phytohormone indole-3-acetic acid (IAA) was heterologously expressed in the two reference plant species Arabidopsis thaliana and Medicago truncatula. For the transformation of Medicago we devised a novel protocol using seedling infiltration. When PvIAP1 was overexpressed under the control of the constitutive 35SCaMV promoter in Arabidopsis, the plants showed signs of earlier bolting and enhanced branching. Expression of a fusion protein of PvIAP1 with both a green fluorescence protein (GFP) as reporter and 6× histidine (His) tag under the control of the native bean IAP1 promoter resulted in the accumulation of the protein in both plant species exclusively in seeds as shown by immunoblotting and by fluorescence microscopy. During seed development, PvIAP1 was first expressed in the vascular bundle of Arabidopsis, whereas in later stages GFP fluorescence was visible essentially in all tissues of the seed. Fluorescence decreased rapidly after imbibition in the seeds for both Arabidopsis and Medicago, although the fluorescence persisted longer in Arabidopsis. GFP fluorescence was distributed evenly between an organelle fraction, the microsomal membrane fraction, and the cytosol. This was also confirmed by immunoblot analysis. Clusters of higher GFP fluorescence were observed by confocal microscopy. Although PvIAP1 protein accumulated in seeds of both Arabidopsis and Medicago, neither species post-translationally modified the protein with an indoleacyl moiety as shown by quantitative GC–MS analysis after alkaline hydrolysis. The interaction of different saccharides with the snake gourd (Trichosanthes anguina) seed lectin (SGSL) was investigated by fluorescence spectroscopy. Binding of 4-methylumbelliferyl-beta-D-galactopyranoside (MeUmb beta Gal) to SGSL resulted in a significant increase in the fluorescence emission intensity of the sugar at 376 nm, and this change was used to estimate the association constants for the binding interaction. Interestingly, the increase in emission intensity changed with a change in temperature, increasing from 19.2% at 20 degrees C to 80.2% at 40 degrees C. At 20 degrees C the association constant, K(a), for the MeUmb beta Gal-SGSL interaction was found by fluorescence titration to be 5.8 x 10(4) M(-1). From the temperature dependence of the association constants, the changes in enthalpy (Delta H) and entropy (Delta S) associated with binding of MeUmb beta Gal to SGSL were estimated to be -80.85 kJ.mol(-1) and -184.0 J.mol(-1).K(-1), respectively. Binding of unlabeled sugars was investigated by monitoring the decrease in fluorescence intensity when they were added to a mixture of SGSL and MeUmb beta Gal. The Ka values for different sugars were determined at several temperatures, and Delta H and Delta S were determined from the van't Hoff plots. Enthalpy-entropy compensation was noticed in all cases. The results indicate that saccharide binding to SGSL is enthalpy-driven and the negative contribution from entropy is, in general, quite high.  相似文献

Inhibitors from (Pinus pinea L.) seed coats were separated using paper chromatography, thin layer chromatography, and a Sephadex G-10 column. The inhibitory activity was resolved into several fractions. One of these behaved similarly to abscisic acid. It has exhibed the same properties as ABA in thin-layer chromatography, paper chromatography, and Sephadex G-10 chromatography and in UV absorption and fluorescence spectra. The embryo envelope tissues in both mature dry seed and duringearly germination of Phacelia tanacetifolia were investigatedby bright-field and fluorescence light microscopy and scanningelectron microscopy. The ruminate seed had an irregularly reticulatesurface owing to the presence of polygonal areas, correspondingto the cells of the seed coat. The raised margins of these cellsjoined at the lobe tips, where radially arranged thickeningsoccurred. The unitegmic seed coat was made up of three distinctlayers: the frayed outer layer, the middle layer with portionsrising outwards to form the radial thickenings, and the innerlayer, the thickness of which was greatest in the micropylarzone. The endosperm tissue had two regions, the micropylar andthe lateral endosperm, which differed in polysaccharide composition,thickness and metachromasy intensity, and presence (in the lateralendosperm) or absence (in the micropylar endosperm) of birefringenceof the cell walls. Moreover, in the micropylar region, wherethe embryo suspensor remnant was found, Ca-oxalate crystalswere scarce or absent. The presence of a partially permeablecuticle covering the seed endosperm was observed. Incubationof seeds in Lucifer Yellow CH indicated that water was ableto penetrate quickly into the seed coat along the pathway formedby the radial thickenings, the raised margins of the polygonalcells and the middle layer. Afterwards, LY-CH readily infiltratedthe apical portions of the seed lobes and then the whole endosperm.Following imbibition, morphological changes were found in themicropylar endosperm, such as the initial digestion of proteinbodies. In addition, both in the seed coat and in the endosperm,a weaker fluorescence, probably due to leaching of polyphenolicsubstances, was observed. Once the seed coat was broken at themicropylar end of the seed, the endosperm cap surrounding theradicle tip had to be punctured by it so that complete germinationcould occur. 目的探究间歇式轴向压应力对组织工程骨种子细胞黏附、增殖与成骨分化能力的影响。 方法构建表达绿色荧光蛋白的兔骨髓间充质干细胞(rBMSCs)作为示踪种子细胞,运用旋转细胞培养仪将松质骨支架和种子细胞共培养7 d获得组织工程骨(TEB),实验组在第7 ~ 14天施加大小10 N、频率1 Hz、4 h/d的间歇式轴向压应力刺激,对照组常规培养,14 d后胰酶消化法获取两组种子细胞并比较其黏附、增殖和成骨分化能力。采用两组独立样本t检验进行统计学分析。 结果(1)流式细胞术显示rBMSCs被成功提取分离。(2)倒置荧光显微镜及扫描电镜显示TEB中种子细胞与支架相容性良好。(3)活体荧光成像系统及扫描电镜显示应力刺激组种子细胞的生长状况要优于非应力刺激组,前者平均荧光密度及细胞数/500倍视野均大于后者,差异均具有统计学意义(平均荧光密度:(3.75±0.34)×108 vs (2.91±0.22)×108,t = 2.90,P = 0.04;细胞数/500倍视野:30.50±4.43 vs 21.00±5.13,t = 3.14,P = 0.01)。(4)细胞黏附实验显示,应力刺激组种子细胞的75%细胞贴壁时间短于非应力刺激组,两组时间分别为(3.00±0.41)h、(13.33±1.70)h,差异具有统计学意义(t = 8.20,P < 0.01),前者的最终细胞贴壁率高于后者(99.97%±0.34% vs 85.83%±1.18%),差异具有统计学意义(t = 11.31,P < 0.01)。(5)CCK-8检测显示,在培养第48 ~ 96 h,应力刺激组种子细胞的增殖能力优于非应力刺激组,将两者的450 nm吸光度值在第48小时(0.49±0.02、0.40±0.02)、72 h(0.76±0.07、0.64±0.04)和96 h(1.58±0.07、1.34±0.13)分别进行比较,差异均具有统计学意义(t = 5.15、2.57、2.86,P均< 0.01)。(6)在成骨诱导14 d后,应力刺激组种子细胞的ALP和Ca结节染色阳性率要强于非应力刺激组:两组ALP染色阳性率分别为26.73%±4.56%、16.68% ± 3.89%,差异具有统计学意义(t =3.33,P = 0.03);两组Ca结节染色阳性率分别为41.81%±3.56%、27.40% ± 2.35%,差异具有统计学意义(t = 3.68,P = 0.02)。 结论间歇性轴向压应力可促进组织工程骨种子细胞的黏附、增殖与成骨分化。  相似文献

Counterfeit seeds cause a large loss in seed industry and crop production. Present study was undertaken to find a new anti-counterfeiting technology by labeling seeds with a fluorescent compound, safranine T (ST). Response surface analysis (RSA) experiment with two factors (ST concentration and seed soaking time) of 14 combinations was performed to investigate the ST effects on seed vigor and fluorescence showing in seedlings of two tobacco varieties, MS Yunyan 85 and Honghua Dajinyuan. After soaking in ST solutions, tobacco seeds were germinated for 16 days, then the germination index (GI), vigor index (VI), length of roots and shoots were determined. The optimum combinations of ST concentration and soaking time on GI, VI and length of shoots and roots were 0.57–0.69 mg/ml and 6.77–9.34 h for MS Yunyan 85 and 0.51–0.80 mg/ml and 5.81–7.39 h for Honghua Dajinyuan, respectively. Meanwhile, tobacco seedlings, treated with ST solutions from 0.3 to 1.5 mg/ml showed obviously fluorescence under illumination of green light. It suggests that soaking tobacco seeds in safranine T solution of suitable concentration can be used as a labeling technique for anti-counterfeiting in tobacco seeds.  相似文献

Biochemical and fluorescence microscopic imaging approach has been adopted to investigate the accumulation of oil bodies at specific stages of seed development in Helianthus annuus L. cv. Morden. Seed filling in sunflower is marked with a rapid accumulation of proteins and lipids upto 30 DAA, after which protein accumulation declines whereas lipids continue to accumulate. Earliest signs of lipid accumulation are evident as early as during globular stage of embryo development. Spatially, a developing seed exhibits enhanced lipid deposition in peripheral cells. Oil body biogenesis is observed as early as 10 DAA, as is evident from the fluorescence microscopic detection of Nile red-positive entities in the protoplasts. To begin with, expression of one of the oleosin (the principal oil body membrane proteins) isoforms (16 kDa), is slower than the other two (17.5 and 20 kDa). Fatty acid composition of oil body lipids is quite similar to that of total seed lipids. An enhanced accumulation of linoleic acid is evident during later stages of seed filling. The proportion of major saturated fatty acids, palmitic (16:0) and stearic (18:0), however, do not alter much during the later phases of seed development. Present findings provide new information on oil body development, lipid accumulation and fatty acid composition, for a better understanding of the phasing of physiological and biochemical events associated with oilseed development.  相似文献
