We had previously found that a perchloric acid-soluble protein (PSP1) occurs in rat liver, and that this novel protein inhibits protein synthesis in a rabbit reticulocyte lysate system (T. Oka, H. Tsuji, C. Noda, K. Sakai, Y.-H. Hong, I. Suzuki, S. Mu?oz, Y. Natori, J. Biol. Chem. 270 (1995) 30060-30067). In the present study, we analyzed lipid components bound to PSP1. Native PSP1 was purified from rat liver using Sephadex G-75, DE-52 cellulose and IgGPSP-affinity chromatography, and the lipid components were extracted. The components obtained from the purified PSP1 were shown to be free fatty acids by thin-layer chromatography. By GC-MS, six major fatty acids were identified as 14:0, 16:0, 18:0, 18:1, 18:2 and 20:4. 1 mol of PSP1 contained 1.26 mol of total fatty acid components. The fatty acid-binding assay of PSP1 showed that the Bmax was 1.25 mol fatty acid/mol PSP1 and the Kd value for palmitic acid was 6.03 microM. The concentration of PSP1 mRNA in rat liver increased 2.3-fold by the administration of peroxisome proliferator, bezafibrate. These findings show that PSP1 is a fatty acid-binding protein-like protein, which is involved in the intracellular metabolism of fatty acid and is quite different from the known fatty acid-binding proteins.  相似文献

Perchloric acid-soluble protein (PSP) is highly conserved during evolution from bacteria to mammals. Although PSP has been recognized as an inhibitor of translation and proliferation in vitro, its precise biological role has not yet been elucidated. Since we previously found similar distributions for PSP and the endoplasmic reticulum (ER) and Golgi complex, the intracellular distribution of PSP was analyzed in more detail. Immunofluorescence studies indicated that PSP co-localized with the ER and Golgi complex, since the distribution pattern of PSP was well matched to both of these organelles. An immunoelectron microscopic study revealed PSP was located not only in the cytosol but also on the surface of the outer ER membrane. Since PSP was present on the ER, we speculated that it may be associated with ER function. Therefore, we analyzed whether or not the ER stress response, which is one of the ER functions, affected PSP expression. The results showed that various ER stressors (thapsigargin, A23187, tunicamycin, brefeldin A, and cisplatin) provoked a dramatic change in the localization of PSP from outside of the nucleus to inside the nucleus within 3 h. Moreover, the ER stressors induced PSP expression. These results suggest that PSP is involved in the cellular response to ER stressors, and that the change in localization of PSP from the ER to the nucleus may be associated with ER stress responses.  相似文献

Divalent cation ionophore A23187 has a potent inhibitory effect on protein synthesis in the C6 rat glioma cell line. Treatment with 4 μM A23187 resulted in 93% inhibition of [1-14C]leucine label incorporation into proteins and a 61% increase in free pool labeling. Total RNA synthesis was not affected. Extracellular ionic calcium or magnesium are not required for these changes to occur. Therefore, these effects of A23187 may be a direct effect on protein synthesis or may result from release of internal stores of divalent cations. By comparison, ionophore X537A (4 μM) has only a slight inhibitory effect on protein synthesis.  相似文献

We examined the expression of perchloric acid-soluble protein (PSP) during liver regeneration after partial hepatectomy (PH) in rats. Liver regeneration was almost complete at 7-d after PH. Expression of PSP protein and mRNA decreased and then gradually increased during liver regeneration. An immunohistochemical study showed that PSP is distributed in cytosol and nuclei in normal liver, but localization in the nuclei was not be recognized in the regenerated liver.  相似文献

The effect of disulfiram on the 5-lipoxygenase activity from rat polymorphonuclear leukocyte cell-free lysates was determined and compared with that of other thiocarbamoyl and aryl disulfides. Disulfiram was a potent inhibitor of the soluble 5-lipoxygenase causing 50% inhibition at submicromolar concentrations (0.4-0.7 microM). The inhibition by disulfiram was similar to that of bis(diisopropylthiocarbamoyl) disulfide with both compounds being about 100-fold more potent as inhibitors than the structurally related bis(4-methyl-1-homopiperazinylthiocarbonyl) disulfide analog. The potency of 5-lipoxygenase inhibition by disulfiram was comparable to that of diphenyldisulfide (IC50 = 0.2-0.4 microM), in the same range or better than most typically used inhibitors. However, the degree of inhibition by disulfiram was more sensitive to thiols than that of diphenyldisulfide, as shown by the selective protection against disulfiram inhibition by low concentrations of thiols. Diethyldithiocarbamate, the reduction product of disulfiram, was a less potent inhibitor of the 5-lipoxygenase activity, causing only a partial inhibition (40-60%) over a wide range of concentrations (2-30 microM). The results demonstrate that disulfiram is a potent inhibitor of 5-lipoxygenase in vitro and provide the basis for further investigations on the effect of the drug on leukotriene biosynthesis inhibition and its contribution to the ethanol-disulfiram reaction. They also indicate that disulfiram represents a sensitive reagent to characterize the thiol requirement of the 5-lipoxygenase reaction.  相似文献

Sinefungin and a related metabolite, A9145C, which are antifungal antibiotics isolated from Streptomyces griseolus, were found to be potent inhibitors of both calf thymus and bovine adrenal S-adenosylmethionine-dependent protein O-methyl-transferase (EC 2.1.1.24). Sinefungin and A9145C exhibit inhibitory activity which are equal to or greater than S-adenosylhomocysteine, the endogenous inhibitor of this methyltransferase.  相似文献

Argatroban was synthesized in seven steps from 4-methylpiperidine. The condensation of (+/-)-trans-benzyl 4-methylpipecolic acid ester with N(alpha)-Boc-N(omega)-nitro-L-arginine led to two diastereomers that were separated. One of them is the precursor of argatroban.  相似文献

Flurbiprofen, 2-(2-fluoro-4-biphenylyl)propionic acid, inhibited the formation of prostaglandin E2 from arachidonic acid by bovine seminal vesicular microsomes. It was found that flurbiprofen was an approx. 12.5-fold better inhibitor than indomethacin by comparison of their I50 values. It was suggested that the inhibition of prostaglandin synthesis by flurbiprofen might be due to the inhibition of the endoperoxygenase which catalyzed conversion of arachidonic acid to cyclic endoperoxide. Other carboxylic acid compounds such as aspirin, ibuprofen and indomethacin showed the same type of inhibition as flurbiprofen. In contrast, phenylbutazone which was a pyrozolone derivative inhibited the formation of prostaglandin E2, but not affected the endoperoxygenase reaction. The kinetic studies for inhibition of prostaglandin E2 synthetase indicated that flurbiprofen competitively inhibited prostaglandin E2 synthesis, just like indomethacin. The Ki values were estimated to be 0.128 micron for flurbiprofen and 3.18 micron for indomethacin.  相似文献

Effects of five kinds of dopamine agonists on the activity of type 2A protein phosphatase in rat brain were studied. Apomorphine and SKF-38393 reduced the enzyme activity considerably and their effects were further enhanced in the presence of 10 microM Mn2+. Also, 6,7-ADTN slightly inhibited the activity. The present results suggest that type 2A protein phosphatase in the brain is possibly involved in dopamine mediated protein phosphorylation functions.  相似文献

Protein kinase C functions prominently in cell regulation via its pleiotropic role in signal transduction processes. Certain oncogene products resemble elements involved in transmembrane signaling, elevate cellular sn-1,2-diacylglycerol second messenger levels, and activate protein kinase C. Sangivamycin was unique among the nucleoside compounds tested in its ability to potently inhibit protein kinase C activity. Inhibition was competitive with respect to ATP for both protein kinase C and the catalytic fragment of protein kinase C prepared by trypsin digestion. Sangivamycin was a noncompetitive inhibitor with respect to histone and lipid cofactors (phosphatidylserine and diacylglycerol). Sangivamycin inhibited native protein kinase C and the catalytic fragment identically, with apparent Ki values of 11 and 15 microM, respectively. Sangivamycin was an effective an inhibitor of protein kinase C as H-7, an isoquinolinsulfonamide. Sangivamycin did not inhibit [3H]phorbol-12,13-dibutyrate binding to protein kinase C. Sangivamycin did not exert its action through the lipid binding/regulatory domain; inhibition was not affected by the presence of lipid or detergent. Unlike H-7, sangivamycin selectively inhibited protein kinase C compared to cAMP-dependent protein kinase. The discovery that protein kinase C is inhibited by sangivamycin and other antitumor agents suggests that protein kinase C may be a target for rational design of antitumor compounds.  相似文献

A product of mitochondrial protein synthesis in rat liver mitochondria, characterized by a low molecular weight (Mr is less than 10000) and an unusually high hydrophobicity, has been identified as the dicyclohexylcarbodiimide-binding protein and as a peptide of the hydrophobic sector of the mitochondrial ATPase complex.
