The mutagenic effect of low-dose gamma-irradiation was studied in Drosophila melanogaster systems of hybrid dysgenesis by estimating polytene chromosome rearrangements, recombination frequency, and viability at the embryonic and postembryonic developmental stages. A dose of gamma-irradiation which had no effect detectable by routine line crossing proved to significantly reduce the number of recombinants in the H-E and P-M systems and mortality at postembryonic stages. However, this combined effect was obtained if irradiation followed transposition, i.e., it depended on the application sequence of the mutagenic factors. The reverse order of the mutagenic treatment led to summation of the effects: as compared to either control, the frequencies of the dominant allele mutations as well as the larval and pupal mortality in F2 increased significantly (at the level of 99.9%). This allowed us to estimate the contribution of extremely low-dose gamma-irradiation into the mutagenic effect, which was impossible under routine conditions.  相似文献

Summary The influence of repair and replication on the frequency of spontaneous chromosome aberrations and of those induced by gamma-irradiation is reported.Using the technique of labelling DNA with radioactive 3H-thymidine and measuring the radioactivity of DNA isolated from embryos, the time of initiation and the duration of DNA synthesis in barley seeds was studied after the soaking of the seeds had begun. The average duration of each phase of the first DNA synthesis cycle in soaking barley seeds was found to be as follows: pre-DNA synthesis stage, 10–11 hrs; DNA synthesis stage, 8 hrs. After gamma-irradiation, the intensity of DNA synthesis decreased and the beginning of DNA synthesis was delayed.It was found that the inhibition of repair by caffeine led to an increase in the frequency of both spontaneous and induced chromosome aberrations. Caffeine enhanced several times the frequency of chromosome and chromatid aberrations at the time of the maximal activity of repair enzymes. During DNA replication, caffeine had a lower effect on the realization of premutational lesions.An inhibitor of DNA replication — hydroxyurea — had no influence on the frequency of spontaneous chromosome aberrations during the replication period, whereas after gamma-irradiation, hydroxyurea enhanced the frequency of aberrations mainly at the stage of DNA replication.The relatively small mutagenic action of both agents (caffeine and hydroxyurea) was observed during all stages of the cell cycle of germinating barley seeds.  相似文献

Aprostocetus vaquitarum (Wolcott) causes 78–91 percent mortality to eggs of Diaprepes abbreviatus (L.), under field conditions in southern Florida. In the laboratory, A. vaquitarum was reared on D. abbreviatus eggs at 25 °C, a photoperiod of 12:12 (L:D) and with abundant hosts, A. vaquitarum adult females lived around 15 days. Oviposition was significantly affected by the age of the host egg mass. Egg masses aged 0- to 3-day-old were accepted significantly better than those aged 4–6 days. The mean number of eggs deposited per female was around 53, with extreme values of 124 and 19 eggs per female. Using these data in combination with the sex ratio observed in the field (0.16) and the duration of the preimaginal stages, rm (0.168–0142 day−1), T (22.39–22.89 days), and R0 (43.03–25.81 females per female) were calculated.  相似文献

The effect of theyellow (y) locus on germ cell sensitivity to the alkylating agent ethyl methanesulfonate (EMS) has been studied in Drosophila. Since DNA repair is one of the most important factors that control cell sensitivity to mutagens, the approaches used in our experiments aimed at evaluating the relationship between germ-cell mutability and activity of DNA repair. Germ-cell mutability and repair activity were assessed using several parameters, the most important of which was the frequency of the sex-linked recessive lethals (RSLLM). In one series of experiments, the adult males of various genotypes (Berlin K; y; y ct v; and y mei-9 a) were treated by mutagenic agents and then crossed to Bascfemales. Comparative analysis of germ-cell mutability as dependent on genotype and the stage of spermatogenesis showed that theyellow mutation significantly enhanced the premeiotic cell sensitivity to EMS, presumably, due to the effect on DNA repair. In the second series of experiments, the effect of the maternal DNA repair was studied and, accordingly, mutagen-treated Bascmales were crossed to females of various genotypes including y and y mei-9 a ones. The crosses involving y females yielded F1 progeny with high spontaneous lethality, whereas in F2, the frequency of spontaneous mutations was twice higher. The germ cell response to EMS depended also on female genotype: the effect of yellow resulted in increased embryonic and postembryonic lethality, whereas the RSLLM frequency decreased insignificantly. The latter result may be explained by elimination of some mutations due to 50% mortality of the progeny F1. The results obtained using the above two approaches suggest that theyellow locus has a pleiotropic effect on the DNA repair systems in both males and females of Drosophila.  相似文献

A collection of bacterial strains obtained from a wide-range origin was screened for ability to promote growth in two types of Prunus rootstocks in a commercial nursery. Only few strains promoted growth significantly and consistently, and a strong specificity for the rootstock cultivar was observed. Irrigation of plants with Pseudomonas fluorescens EPS282 and Pantoea agglomerans EPS427 significantly increased plant height and root weight of the plum Marianna 2624 and the peach–almond hybrid GF-677, respectively. Plant height showed a higher rate of growth in early stages of development (2.6–3.5 times the non-treated controls), but the effect decreased with plant age. However, in aged plants growth promotion was more significant on root weight (1.9 times the non-treated controls) than on plant height. The efficacy of growth promotion and the persistence of strains in the root environment were dependent on the bacterial inoculum concentration applied. Increases in root development were maximum at inoculum concentrations of up to 8 log10 CFU ml–1 (ca 10 log10 CFU L–1 of potting mix). Population levels at the optimum inoculum concentration were around 7 log10 CFU g f.w.–1 root material at early stages of development and decreased to 4 log10 CFU g f.w.–1 after several months of development. The best plant growth-promoting strains were very diverse in secondary metabolite production and antagonistic ability against several plant pathogens.  相似文献

We examined the effects of UVB radiation on hatching success of eggs, survival of various naupliar and copepodite stages, and feeding and egg production of adult females of the brackish-water copepod, Sinocalanus tenellus, by exposure to varying doses of UVB irradiance in the laboratory. Artificial UVB radiation resulted in an increased mortality of eggs, nauplii and copepodites with increasing UVB doses. UVB induced damage was stage-specific with eggs being most susceptible (LD50= 4.1 kJ m–2 ) and adult females being least susceptible (LD50= 16.7 kJ m–2). Effects on feeding and egg production of adult females were significantly evident at UVB doses higher than 11.0 kJ m–2 and 7.0 kJ m–2, respectively. We also examined the photorepair response of eggs and various developmental stages in simultaneous irradiation of UVB and enhanced PAR. With enhanced PAR there was a considerable recovery against UVB damage, being higher for younger animals than older ones. In nature, however, solar UVB radiation may rarely cause appreciable damage to S. tenellus population due to optically high attenuation properties of their habitat waters.  相似文献

The partition behaviour of cutinase on poly(ethylene glycol) (PEG)–hydroxypropyl starch aqueous two-phase systems was characterized. The effect of molecular mass of PEG, the pH of the system and tie-line length on cutinase partition coefficient and cutinase yield to the top phase was investigated for systems prepared with a purified hydroxypropyl starch (Reppal PES 100) and a crude one (HPS). The effect of the presence of different salts, such as sodium chloride, sodium sulphate and ammonium sulphate, on cutinase partition was also studied. The results lead to the conclusion that aqueous two-phase systems composed of PEG and hydroxypropyl starch are not efficient in the purification of cutinase. In the majority of cases, the partition coefficients were very close to 1, with pH being the factor which affects most cutinase partition. Partition coefficients were significantly improved when salts were added to the systems. For PEG 4000–Reppal PES 100 [at pH 4.0; 0.5 M (NH4)2SO4], the partition coefficient for cutinase was 3.7, while a value of 12 was obtained for PEG 4000–HPS (at pH 4.0; 1 M NaCl). An isoelectric point (pI) of 7.8 was confirmed for cutinase by constructing a cross partition graphic from the results obtained in the experiments with different salts.  相似文献

Energy metabolism in early life stages of the shrimp Farfantepenaeus paulensis subjected to temperature reduction (26 and 20 °C) was determined using the activities of citrate synthase (CS) and pyruvate kinase (PK). At both temperatures, weight-specific activity of CS decreased throughout the ontogenetic development from protozoea II (PZ II) to postlarva XII–XIV (PL XII–XIV). PK activity reached a pronounced peak in PL V–VI, followed by a further decrease in PL XII–XIV. Temperature reduction produced variation in oxygen consumption rates (QO2), ammonia–N excretion and in enzyme activities. Ammonia–N excretion was higher at 20 °C in mysis III (M III), PL V–VI and PL XII–XIV, resulting in substantially lower O:N ratios in these stages. QO2 was increased in protozoea II (PZ II) and mysis I (M I) at 26 °C, while no difference in QO2 was detected in the subsequent stages at either temperature. This fact coincided with higher CS and PK activities in M III, PL V–VI and PL XII–XIV at 20 °C compared with 26 °C. Regressions between individual enzyme activities and dry weight exhibited slope values of 0.85–0.92 for CS and 1.1–1.2 for PK and temperature reduction was reflected by higher slope values at 20 than at 26 °C for both enzymes. Weight-specific CS activity was positively correlated with QO2 at 20 and 26 °C, and may thus be used as an indicator of aerobic metabolic rate throughout the early stages of F. paulensis. The variation in enzyme activities is discussed in relation to possible metabolic adaptations during specific ontogenetic events of the F. paulensis life cycle. Here, the catalytic efficiency of energy-metabolism enzymes was reflected in ontogenetic shifts in behaviour such as larval settlement and the adoption of a benthic existence in early postlarvae. In most cases, enhanced enzyme activities appeared to counteract negative effects of reduced temperature.  相似文献

The mutagenic effect of an industrial enterprise (tungsten and molybdenum factory) was studied in three stages. At the first stage, the putative impact of the industrial sewage of the factory was studied using three plant test systems: Crepis capillaris L., Tradescantia sp. clone 02, and Glycine max (L.) Merill. It was found that the sewage increased the mutation level by a factor of 11–45. At the second stage, the rate of mutation was studied in the native vegetation growing on solid waste piles of the enterprise. It exceeded the corresponding index of uncontaminated areas by a factor of 2.0–4.5. At the third stage, the rates of children with birth defects and miscarriages were studied in the vicinity of the enterprise. The rate of miscarriages proved to be higher than the value averaged over the autonomous republic by a factor of 2.4. The impact of different doses of artificial ultraviolet (UV) radiation on the growth stages of a marine zooplankton was investigated using laboratory microcosms. Mortality percentages of naupliar and adult samples of Artemia franciscana were recorded in relation to different UV doses (single exposure: 75, 150, 300, 600, 1,200, 2,400, 3,900, 7,800 J m–2) at specific observation times after exposure (24, 48, 72, 96 and 120 h). The relationship between mortality percentage and UV dose showed significant differences in relation to the zooplankton growth stage. The elevated susceptibility of the naupliar samples to UV radiation is described through a mortality model based on a logistic equation. The data analysis shows that the slope of mortality versus dose remains the same for the two growth stages while the lethal dose in the naupliar stage was 3.3 smaller than that determined for the adult stage. The slope of the UV mortality rate versus post-incubation time was found to be significantly different (P<0.05) at low UV doses for the two life stages examined, i.e. naupliar and adult. The lower value of LD50 in naupliar stages compared to that for adults confirms that in the early growth stage this marine zooplankton is more susceptible to UV radiation.  相似文献

Summary A selection of mono- and polyfunctional alkylating agents as well as a folic acid antagonist and an acridine derivate were tested with the host-mediated assay, and as far as not known from the literature, with the dominant lethal test for mutagenic activity in mice. In the host-mediated assay system the indicator organisms Salmonella typhimurium G46 His , Serratia marcescens a 13 His and a 21 Leu were used as back mutation systems and E. coli 343 as a forward mutation system. We found indications that polyfunctional alkylating agents induce dominant lethal mutations to a larger extent, whereas monofunctional alkylating agents revealed more mutagenic activity on the molecular level. No definite mutagenicity could be observed for amethopterine, which is mutagenic in cytogenetic investigations. Trypaflavin which is known to be mutagenic in the dominant lethal test, did not induce point mutations in our indicator strains. Food concentration (0.5 × 103 – 5 × 105 Scenedesmus cells m1–1) significant influenced the somatic growth, maturation and survivorship ofS. proboscideus larvae. A density of 5 × 104 cells m1–1 was optimal for early larval stages. Of four temperature tested (20–35 °C), 30 °C resulted in the best growth and survival. Maturation time was inversely related to temperature, and was size- rather that age-dependent. Larvae were tolerant of a wide conductivity range, but optimal growth and survival were observed at 260 µS cm–1. Nitrate-Nitrogen (NO2-N) caused a larval mortality of 50% after 24 h at 0.58 mg1–1.  相似文献

The large bowel intestinal flora of mammals is made up mostly of O2-intolerant anaerobic microorganisms which are irreversibly damaged by brief exposure to air. The aim of our work was to investigate the effect of atmospheric O2 on human intestinal anaerobic microorganisms. Thirty O2-intolerant bacterial strains that reached 100% mortality after 120 min of air exposure were isolated. Ten of these strains were tested for their atmospheric O2 sensitivity as a function of air exposure time; all tested microorganisms showed a similar mortality trend on exposure to air. In fact, 50% of cells survive, on the average, after 4–5 min of atmospheric O2; this percentage decreases to 3–5% after only 20 min, and after 40 min only one cell in a thousand survives; all strains reached 100% mortality in a time range of 100–120 min. The strains examined were identified as belonging to the generaEubacterium, Peptostreptococcus, andCoprococcus.  相似文献

The susceptibility of immature stages of the legume flower thrips, Megalurothrips sjostedti, to the entomopathogenic fungus Metarhizium anisopliae, was investigated under laboratory conditions. The adult stage was found to be more susceptible to infection than the larval and pupal stages. Mortality at all stages was dose-dependent, with the highest concentration of 1×108 conidia ml–1 producing the highest mortality (26, 46 and 100% for larvae, pupae and adults, respectively) at 8 days post-inoculation. At the same concentration, daily pollen consumption was significantly reduced at 2 days after treatment in infected adults but more slowly in infected larvae. Fecundity, egg fertility and longevity in adults surviving infection as larvae were significantly reduced compared to the control.  相似文献

Acrylamide (AA) can be formed in certain foods by heating, predominantly from the precursor asparagine. It is a carcinogen in animal experiments, but the relevance of dietary exposure for humans is still under debate. There is substantial evidence that glycidamide (GA), metabolically formed from AA by Cyp 2E1-mediated epoxidation, acts as ultimate mutagenic agent. We compared the mutagenic potential of AA and GA in V79-cells, using the hprt mutagenicity-test with N-methyl-N′-nitro-N-nitroso-guanidine (MNNG) as positive control. Whereas MNNG showed marked mutagenic effectivity already at 0.5 μM, AA was inactive up to a concentration of 10 mM. In contrast, GA showed a concentration dependent induction of mutations at concentrations of 800 μM and higher. Human blood was used as model system to investigate genotoxic potential in lymphocytes by single cell gel electrophoresis (comet assay) and by measuring the induction of micronuclei (MN) with bleomycin (BL) as positive control. AA did not induce significant genotoxicity or mutagenicity up to 6000 μM. With GA, concentration dependent DNA damage was observed in the dose range of 300–3000 μM after 4 h incubation. Significant MN-induction was not observed with AA (up to 5000 μM) and GA (up to 1000 μM), whereas BL (4 μM) induced significantly enhanced MN frequencies. Thus, in our systems GA appears to exert a rather moderate genotoxic activity.  相似文献

This study was conducted to examine the effects of varying N rates and cropping systems (mixedversus pure stand) on the suitability of oats (Avena sativa L.) for estimating N2 fixed in sequentially harvested vetch (Vicia sativa L.) over two growing seasons (1984–85 and 1985–86). The N rates were, 20 and 100 kg N ha–1 in 1984–85 and 15 and 60 kg N ha–1 in 1985–86. In the 1984–85 season, vetch at maturity derived 76 and 63% N from fixation at the high and low N rates respectively. The corresponding values for the second season were 66 and 42%. Except in the 1985–86 season when some significantly higher values of % N2 fixed were estimated by using the reference crop grown at the higher (A-value approach) than at the lower N rate (isotope-dilution approach), both approaches resulted in similar measurements of N2 fixed. In the 1984–85 season, similar values of N2 fixed were obtained using either the pure or mixed stand oats reference crops. Although in the 1985–86 season, the mixed reference crop occasionally estimated lower % N2 fixed than pure oats, total N2 fixed estimates were always similar (P<0.05). Thus, in general, N fertilization and cropping system of the reference crop did not significantly influence estimates of N2 fixation.  相似文献

A method for chromatographic separation and quantitative determination of individual components of the antibiotic virginiamycin, produced by microbiological synthesis (Streptomyces virginiaestrain 147), is described. The components, M1–2and S1–5, were isolated from fermentation broth and identified by HPTLC and HPLC (the results obtained using the two methods correlate well with each other). The conditions of culturing of the producer and the composition of nutritive media were optimized. Using UV irradiation as a mutagenic factor, the producer was selected for increased level of synthesis of the antibiotic; this was achieved by inducing mutations that impart resistance to virginiamycin and meta-fluorophenylalanine, an analog of phenylalanine.  相似文献

We measured the toxicity and mutagenicity induced in human diploid lymphoblasts by various radiation doses of X-rays and two internal emitters. [125I]iododeoxyuridine ([125I]dUrd) and [3H]thymidine ([3H]TdR), incorporated into cellular DNA. [125I]dUrd was more effective than [3H]TdR at killing cells and producing mutations to 6-thioguanine resistance (6TGR). No ouabain-resistant mutants were induced by any of these agents. Expressing dose as total disintegrations per cell (dpc), the D0 for cell killing for [125I]dUrd was 28 dpc and for [3H]TdR was 385 dpc. The D0 for X-rays was 48 rad at 37°C. The slopes of the mutation curves were approximately 75 × 10−8 6TGR mutants per cell per disintegration for [125I]dUrd and 2 × 10−8 for [3H]TdR. X-Rays induced 8 × 10−8 6TGR mutants per cell per rad. Normalizing for survival, [125I]dUrd remained much more mutagenic at low doses (high survival levels) than the other two agents. Treatment of the cells at either 37°C or while frozen at −70°C yielded no difference in cytotoxicity or mutation for [125I]dUrd or [3H]TdR, whereas X-rays were 6 times less effective in killing cells at −70°C.Assuming that incorporation was random throughout the genome, the mutagenic efficiencies of the radionuclides could be calculated by dividing the mutation rate by the level of incorporation. If the effective target size of the 6TGR locus is 1000–3000 base pairs, then the mutagenic efficiency of [125I]dUrd is 1.0–3.0 and of [3H]TdR is 0.02–0.06 total genomic mutations per cell per disintegration. 125I disintegrations are known to produce localized DNA double-strand breaks. If these breaks are potentially lethal lesions, they must be repaired, since the mean lethal dose (D0) was 28 dpc. The observations that a single dpc has a high probability of producing a mutation (mutagenic efficiency 1.0–3.0) would suggest, however, that this repair is extremely error-prone. If the breaks need not be repaired to permit survival, then lethal lesions are a subset of or are completely different from mutagenic lesions.  相似文献

Stratiolaelaps ( = Hypoaspis) miles (Berlese) (Acari: Mesostigmata: Laelapidae) is a polyphagous soil-dwelling predatory mite that is widely marketed for use in greenhouse production systems to manage populations of dark-winged fungus gnats, Bradysia spp. (Diptera: Sciaridae) and for supplemental control of thrips. The suggestion by Walter and Campbell (2003, Biol. Control 26: 253–269) that North American commercial cultures of S. miles may actually be S. scimitus was confirmed. The development and reproduction at 21–23 °C of S. scimitus provided ad libidum with one of three different prey – the fungus gnat Bradysia aff. coprophila (Lintner), potworms (Enchytraeidae), or Sancassania aff. sphaerogaster (Zachvatkin, 1937) (Acari: Astigmata: Acaridae) – were compared. Developmental duration of the egg and non-feeding larval stages were 2.47 and 1.11 days, respectively; mortalities were 8.3 and 5.5%. Stratiolaelaps scimitus failed to develop beyond the protonymphal stage when provided with S. aff. sphaerogaster alone, although some feeding was observed. Development and reproduction of S. scimitus was successful on both fungus gnat larvae and enchytraeids, with no influence of prey on protonymphal duration (4.70 days) and mortality (8.3%), or on deutonymphal duration (4.61 days) and mortality (6.1%). Adult female S. scimitus feeding on potworms, compared to those feeding on fungus gnat larvae, had a significantly shorter pre-oviposition period (2.69 vs. 4.59 days). However, diet did not influence other adult female developmental or reproductive characteristics (oviposition period, 18.6 days; post-oviposition period, 6.2 days; total adult longevity, 27.3 days; total number of eggs, 26.5). S. scimitus reared on potworms tended (p = 0.06) to have a higher intrinsic rate of increase, a higher finite rate of increase and a shorter doubling time (r m = 0.142 day−1, λ = 1.153, Dt = 4.85 days) than those reared on fungus gnat larvae (r m = 0.105 day−1, λ = 1.110, Dt = 6.58 days), but differences in net reproductive rate (R 0) and generation time (G) were not significant.  相似文献

Mini UV lamps were installed over antarctic plants at Léonie Island, Antarctic peninsula, and shoot length measurements of Deschampsia antarctica were performed during the austral summer January–February 1999.We studied the response of the antarctic hairgrass, Deschampsia antarctica to enhanced UV-B. In a climate room experiment we exposed tillers of Deschampsia antarctica, collected at Léonie Island, Antarctic peninsula, to ambient and enhanced levels of UV-B radiation. In this climate room experiment with 0, 2.5 and 5 kJ m–2 day–1 UV-BBE treatments we observed that length growth of shoots at 2.5 and 5 kJ m–2 day–1 UV-BBE was markedly reduced compared to 0 kJ m–2 day–1 UV-BBE. In addition, there was an increased number of shoots and increased leaf thickness with enhanced UV-B. The Relative Growth Rate (RGR) was not affected by UV-B, possibly because reduced shoot length growth by enhanced UV-B was compensated by increased tillering. Light response curves of net leaf photosynthesis of plants exposed to 5 kJ m–2 day–1 UV-BBE did not differ from those exposed to 0 kJ m–2 day–1 UV-BBE. The content of UV-B absorbing compounds of plants exposed to increasing UV-B did not significantly change.Mini UV-B lamp systems were installed in the field, to expose the terrestrial antarctic vegetation at Léonie Island to enhanced solar UV-B. In that study, the increment of shoot length of tagged plants of Deschampsia antarctica during the January-February 1999 at Léonie Island, was recorded and compared to shoot length growth under controlled conditions.The consequences of enhanced UV-B radiation as a result of ozone depletion for the terrestrial antarctic ecosytems are discussed.  相似文献
