Dark fixation of 14CO2 was followed in potato disks under varyingsalt treatments at 0° C and 25° C. It is shown thatthe specific activity of the 14CO2 supplied is heavily dilutedby endogenously produced CO2 and that the apparently greaterfixation of 14CO2, at 0° C as compared with that at 25 °C is due to the lower respiration rate at 0° C, with consequentlyless dilution of the 14CO2. supplied. At 25° C organic acidformation in response to different salt treatments fulfils thecommon expectation, 14CO2 fixation increasing in the presenceof K2SO4 and decreasing in CaCl2 relative to that in KCl. Therole of organic acids in maintaining ionic balance within thecell at 25° C is thereby indicated but at 0° C organicacid adjustments did not follow the normal pattern. At 25°C but not at o° C increasing external concentration of KCIresulted in an increased level of 14CO2 fixation.  相似文献

To find out whether the decrease in muscle performance of isolated mammalian skeletal muscle associated with the increase in temperature toward physiological levels is related to the increase in muscle superoxide (O2) production, O2 released extracellularly by intact isolated rat and mouse extensor digitorum longus (EDL) muscles was measured at 22, 32, and 37°C in Krebs-Ringer solution, and tetanic force was measured in both preparations at 22 and 37°C under the same conditions. The rate of O2 production increased marginally when the temperature was increased from 22 to 32°C, but increased fivefold when the temperature was increased from 22 to 37°C in both rat and mouse preparations. This increase was accompanied by a marked decrease in tetanic force after 30 min incubation at 37°C in both rat and mouse EDL muscles. Tetanic force remained largely depressed after return to 22°C for up to 120 min. The specific maximum Ca2+-activated force measured in mechanically skinned fibers after the temperature treatment was markedly depressed in mouse fibers but was not significantly depressed in rat muscle fibers. The resting membrane and intracellular action potentials were, however, significantly affected by the temperature treatment in the rat fibers. The effects of the temperature treatment on tetanic force, maximum Ca2+-activated force, and membrane potential were largely prevented by 1 mM Tempol (4-hydroxy-2,2,6,6-tetramethylpiperidine-1-oxyl), a membrane-permeable superoxide dismutase mimetic, indicating that the increased O2 production at physiological temperatures is largely responsible for the observed depression in tetanic force at 37°C by affecting the contractile apparatus and plasma membrane. intact mammalian muscle; physiological temperature; superoxide; excitation-contraction coupling; maximum Ca2+-activated force; muscle excitability; cytochrome c assay  相似文献

Fidgeon, C. and Wilson, G. 1988. Uptake and accumulation ofa-naphthalene acetic acid by cell suspensions of Galium mollugoL.—J. exp. Bot. 39: 241-249. Galium mollugo cell suspensions require -NAA for continued growthand cell division. The kinetics of -NAA uptake from the medium(B5) by Galium cells was assessed using 1-14C -NAA in a standardratio of cells to medium (0.25 g: 2.5 cm3). It was found thatthe uptake of -NAA was rapid, over 90% being taken up within4 h. Cells which had accumulated -NAA for 4 h or more did notrelease it back into the medium. It was found that Galium cellsaccumulated -NAA against a significant concentration gradient;suggesting the participation of an active component in the uptakemechanism. The effect of free-space and surface adsorption onthe uptake of -NAA was determined by means of a repeated washtechnique. These two factors were found to be of importanceonly during the first hour of uptake. Neither dead cells norplasmolysed cells absorbed -NAA. It is clear that, in the normal growth cycle, Galium cells cantake up the available -NAA within 3 or 4 h of inoculation andthat this can stimulate a cell division response of 3-4 generationsover the subsequent 14 d. The use of the oxygen electrode in measuring the respirationrate of potato slices between o° C. and 27° C. is described.When slices are transferred from 27° C. to o° C. therespiration rate adjusts to the level characteristic of thelow temperature within 2 or 3 minutes. The slices have a highoxygen affinity and the respiration rate at o° C. is approximately10 per cent. of the rate at 24° C. in both freshly cut slicesand slices aged 24 hours, during which time respiration increasesfourfold.  相似文献

The regulation of ethylene synthesis in pear fruits was investigated.During storage for 60 d at — 1 °C the rate of ethylenesynthesis increased 100-fold but the concentration of 1-aminocyclopropane-l-carboxylicacid (ACC) increased only 2-fold and ACC synthase activity waslow. On transfer to 15 °C after storage at — 1 °Cethylene synthesis increased 10-fold within 10 h but ACC synthaseactivity only increased rapidly after 24 h; the decline in ACClevels during the first 16 h at 15 °C was insufficient tosustain ethylene synthesis. Ethylene synthesis was further investigatedusing discs cut from the mid cortex of pear fruits. Synthesiswas inhibited by aminoethoxyvinylglycine (AVG) and amino-oxyaceticacid at all stages of ripening. The rate of synthesis and ACCsynthase activity increased rapidly after slicing of pears heldat — 1 °C but more slowly in discs cut from pearsimmediately after harvest. Cycloheximide (CHI) inhibited theseincreases and reversed increases resulting from pre-incubationof discs. A combination of CHI and AVG abolished the capacityof discs to synthesize ACC and ethylene production was curtailed.Cordycepin and actinomycin-D were less effective as inhibitorsof the development of ethylene synthesis and ACC synthase activitythan as inhibitors of incorporation of 5-[3H] uridine into totalRNA or poly A rich RNA. The ability of discs to develop ethylenesynthesis and ACC synthase activity in the presence and absenceof cordycepin increased concurrently during storage of wholefruits at — 1 °C. We modeled changes in contractile element kinetics derived from the cyclic relationship between myoplasmic [Ca(2+)], measured by indo 1 fluorescence, and left ventricular pressure (LVP). We estimated model rate constants of the Ca(2+) affinity for troponin C (TnC) on actin (A) filament (TnCA) and actin and myosin (M) cross-bridge (A x M) cycling in intact guinea pig hearts during baseline 37 degrees C perfusion and evaluated changes at 1) 20 min 17 degrees C pressure, 2) 30-min reperfusion (RP) after 30-min 37 degrees C global ischemia during 37 degrees C RP, and 3) 30-min RP after 240-min 17 degrees C global ischemia during 37 degrees C RP. At 17 degrees C perfusion versus 37 degrees C perfusion, the model predicted: A x M binding was less sensitive; A x M dissociation was slower; Ca(2+) was less likely to bind to TnCA with A x M present; and Ca(2+) and TnCA binding was less sensitive in the absence of A x M. Model results were consistent with a cold-induced fall in heart rate from 260 beats/min (37 degrees C) to 33 beats/min (17 degrees C), increased diastolic LVP, and increased phasic Ca(2+). On RP after 37 degrees C ischemia vs. 37 degrees C perfusion, the model predicted the following: A x M binding was less sensitive; A x M dissociation was slower; and Ca(2+) was less likely to bind to TnCA in the absence of A. M. Model results were consistent with reduced myofilament responsiveness to [Ca(2+)] and diastolic contracture on 37 degrees C RP. In contrast, after cold ischemia versus 37 degrees C perfusion, A x M association and dissociation rates, and Ca(2+) and TnCA association rates, returned to preischemic values, whereas the dissociation rate of Ca(2+) from A x M was ninefold faster. This cardiac muscle kinetic model predicted a better-restored relationship between Ca(2+) and cross-bridge function on RP after an eightfold longer period of 17 degrees C than 37 degrees C ischemia.  相似文献

Vitamin C is one of the most abundant exogenous antioxidants in the cell, and it is of the utmost importance to elucidate its mechanism of action against radicals. In this study, the reactivity of vitamin C toward OH and \( {HO}_2/{O}_2^{-} \) radicals in aqueous medium was analyzed by ab initio molecular dynamics using CPMD code. The simulations led to results similar to those of static studies or experiments for the pair of \( {HO}_2/{O}_2^{-} \) radicals but bring new insights for the reactivity with hydroxyl radical: the reaction takes place before the formation of an adduct and consists of two steps: first an electron is transferred to hydroxyl radical and then the ascorbyl radical loses a proton.

The influence of environmental variables which are known tochange with altitude on the 13C of leaves of Nardus stricta,have been investigated in controlled environment experiments.Low temperature and an increased incidence of freezing temperaturescaused 13C to increase. In contrast, lowered atmospheric pressurereduced 13C. Increased leaf surface wetness and soil water contentalso caused a decrease. Plants from different altitudes didnot differ consistently or significantly in their responses. Truncated human coronavirus HCoV-229E spike glycoproteins containing amino acids 407 to 547 bound to purified, soluble virus receptor, human aminopeptidase N (hAPN). Soluble hAPN neutralized the infectivity of HCoV-229E virions at 37 degrees C, but not 4 degrees C. Binding of hAPN may therefore trigger conformational changes in the viral spike protein at 37 degrees C that facilitate virus entry.  相似文献

The kinetic parameters for transport of the nonmetabolizable glucose analogue 3-O-methyl-D-glucose and the relationship between transport and metabolism of D-glucose and D-fructose were determined in isolated rat hepatocytes at 37 degrees C and pH 7.4. 3-O-Methylglucose at a very low concentration (0.1 mM) equilibrated with the intracellular water with a rate constant of 0.41 s-1. Km for equilibrium exchange entry was 5.5 mM and Vmax was 2.2 mM X s-1 and similar results were obtained when using the zero-trans entry protocol. The rate constant for entry of tracer D-glucose was 0.15 s-1 and Km for glucose was about 20 mM. The phosphorylation rate for D-glucose was much slower than the transport rate. The rate constant for D-fructose entry was about 0.04 s-1, the apparent Km was about 100 mM and Vmax about 5 mM X s-1. The concentration dependence of 3-O-methylglucose inhibition of labelled fructose transport revealed biphasic kinetics indicating that fructose was transferred by both the glucose transporter and a fructose transporter. At concentrations lower than 1 mM, fructose metabolism appeared to be limited by the transport step.  相似文献

All cells of four Candida tropicalis strains IFO 0199 (Ct-0199), IFO 0587 (Ct-0587), IFO 1400 (Ct-1400), and IFO 1647 (Ct-1647), obtained by cultivation at 27 and 37 degrees C for 48 h in yeast extract-added Sabouraud liquid medium, showed the shapes of typical budding yeast and the same agglutination patterns against factor sera 1, 4, 5 and 6 in the commercially available kit 'Candida Check'. The cells of the C. tropicalis IFO 0589 strain display the same properties at 27 degrees C but formed hyphae at 37 degrees C. The cell wall mannan (Ct-0589-37-M) obtained from the strain cells cultured at 37 degrees C had lost most of its reactivity against factor sera 4, 5 and 6 in an enzyme-linked immunosorbent assay, in contrast to the mannan (Ct-0589-27-M) at 27 degrees C. The 1H-nuclear magnetic resonance patterns of the mannans obtained from the cells of the four C. tropicalis strains IFO 0199, IFO 0587, IFO 1400, and IFO 1647, obtained by cultivation at 37 degrees C, did not change compared to those at 27 degrees C. By contrast, the Ct-0589-37-M had significantly lost the beta-1,2-linked mannopyranose units, corresponding to the serum factors 5 and 6. These results show that the IFO 0589 strain is an unusual strain among the general C. tropicalis strains studied.  相似文献

Reactions between the activated cluster [Os3(CO)10(NCMe)2] and malonic acid, succinic acid and dicarboxylic acetylene, respectively, lead to the formation of the linked cluster complexes [{Os3H(CO)10}2(CO2CH2CO2)] (1), [{Os3H(CO)10}2(CO2C2H4CO2)] (2), and [{Os3H(CO)10}2(C4O4)] (3) in good yield. Cluster 3 was subsequently treated with [Co2(CO)8] and this results in the addition of a “Co2(CO)6” group giving [{Os3H(CO)10}2(C2O4){Co2(CO)6}] (4). The X-ray crystal structures are reported for 24. In each structure the two triangular triosmium units are linked by the carboxylate groups and within each complex the carboxylate groups are chelating and bridge two osmium atoms.  相似文献
