<正> The influence of social upbringing on the activity pattern of lion Panthera leo cubs was investigated at three sites. Inthis study, stimulus objects such as sticks, grass, fresh dung (elephant Loxondota africana, zebra Equus quagga, impala Aepycerosmelampus, duiker Sylvicapra grimmia, kudu Tragelaphus strepsiceros, giraffe Girajfa camelopardalis and wildebeestConnochaetes taurinus) and cardboard boxes, were utilized in an enrichment program aimed at encouraging active behaviors ofcaptive lion cubs at Antelope Park and Masuwe. Lion cubs at Chipangali were not behaviorally enriched. Activity patterns wererecorded for 10 days at each site. We recorded moving, resting, playing, grooming, visual exploration and display of hunting instincts.We found that behavioral enrichment enhanced the active behaviors of captive lion cubs. Orphan-raised cubs spent moretime moving, playing and displaying hunting instincts than mother-raised cubs, but the time spent grooming was similar acrossareas and suggests that grooming is not influenced by enrichment. Mother-raised cubs spent more time engaged in visual explorationthan orphan-raised cubs and this could be a behavior acquired from mothers or a result of confidence to explore because oftheir presence. Activity patterns were different among time treatments across our three study sites. Based on these findings, wesuggest that lion cubs raised in captivity could benefit from behavioral enrichment to encourage active behaviors essential foreventual reintroduction into the wild .  相似文献

The 5‘-region of the chitinase gene cabch29,derived from Brassica oleracea var.capitata,has been sequenced and analyzed for cis-acting elements important in controlling gene expression in transgenic tobacco plants.Different 5‘-deletion fragments were linked to reporter gene β-glucuronidase (GUS) as translational fusions,and the expression of these chimeric genes was analyzed in vegetative organs and tissues.Sequences up to-651 showed some basal GUS activity with nearly equal levels in wounded and intact tissues.The addition of further upstream sequences(-651 to-1284) enhanced expression level,and the expression driven by this fragment was inducible by a factor of two to three-fold by wounding.Histochemical analysis of different tissue from transgenic plants that contain cabch29 promoter-gus fusion gene demonstrated woundinducible and tissue-specific cabch29 promoter activity in plants containing the 1308 base pair fragment.The location of GUS activity appears to be cell-specific,being highest in vascular cells and epidermal cells of stem,leaf and roots.Meanwhile,the temporal and spatial expression of cabch29-GUS fusion gene has been investigated.Among the different vegetative organs,a high level of GUS activity was observed in stem and a moderate one in roots;whereas,wounding stress led to a high level of GUS in stem and moderate one in leaf.  相似文献

Presence of the red leg syndrome (RLS) was documented through bacterial and histological examinations in the endangered Kaiser's mountain newt Neurergus kaiseri obtained from a pet shop. The individuals which were severely infected showed lethargy, appetite loss, weight loss, abdominal skin redness and skin ulcers on hind legs. This study reveals the presence of two bacteria causing RLS on the skin of captive AT. kaiseri including Proteus vulgaris and Bacillus cereus. Sections of skin in affected areas and internal organs were examined through standard histological procedures. Histologically, epidermal necrosis and ulcers, epidermal gland depletion, myositis and subcutaneous edema, gastric submucosal edema and hepatomegaly were seen. There were also correlations between the microbial infection and structural changes in tissues of Kaiser's mountain newt. The severity of the structural changes are related to the level of microbial infection in the target organs and could be sustained by the isolation of P. vulgaris and other pathogens. The presence of the infective bacterial population and their interaction on the skin of the newt may have changed the normal skin flora and facilitate the prevalence of other disease.  相似文献

Chinese giant salamander ranavirus(CGSRV) is an emerging pathogen in captive populations of the Chinese giant salamander(Andrias davidianus). We processed 140 morbid Chinese giant salamanders from seven captive breeding populations over five years, and describe the disease associated with CGSRV infection. The most common gross signs were significant swelling of the legs and coelomic cavity, erythema of the legs and ventrum in juveniles; cutaneous erosions and ulcerations in adults, particularly the limbs and the head; and marked petechial or ecchymotic hemorrhages of the internal organs, particularly the liver, spleen and kidney. Histological examination showed degeneration, necrosis, and inflammation in many organs, particularly in the organs where hemorrhage was observed. There was evidence of eosinophilic inclusion bodies in degenerated and necrotic cells. We identified virus particles and empty capsids without viral nucleoid in the inclusion bodies using electron microscopy. Virus particles were hexagonal or round shape, and appeared in paracrystalline arrays, aggregates, or singly. All enveloped viral particles were 140–160 nm. Polymerase chain reaction followed by sequencing verified that the virus particles were CGSRV. These results collectively support that CGSRV was the etiologic agent responsible for these mass die-offs of the Chinese giant salamander. The pathology described herein will be useful in diagnosing cases of ranaviral disease caused by CGSRV, and provide evidence that this pathogen is a significant threat to the Chinese giant salamander.  相似文献

Woonyoungia septentrionalis (Dandy) Law is aceae. The floral morphology and structure of the species a dioecious species with unisexual flowers in Magnoliare conspicuously different from other species and are important to the study of floral phylogeny in this family. The floral anatomy and ontogeny were investigated to evaluate the systematic position of W. septentrionalis, using scanning electron microscopy and light microscopy. All of the floral organs are initiated acropetally and spirally. The carpels are of conduplicated type without the differentiation of stigma and style. The degenerated stamens in the female flowers have the same structures as the normal stamens at the earlier developmental stages, but they do not undergo successive development and eventually degenerate. The male floral apex was observed to have the remnants of carpels in a few investigated samples. As the bisexual flower features could be traced both in the male and female flowers in W. septentrionalis, it suggests that the flower sex in Magnoliaceae tends toward unisexual. As well as the unisexual flowers, the reduced tepals and carpels and concrescence of carpels conform to the specialized tendency in Magnoliaceae, which confirms the derived position of W. septentrionalis in this family. As the initiation pattern of floral parts of W. septentrionalis is very similar to other species in this family, it needs further investigation and especially comparison with species in Kmeria to evaluate the separation of Woonyoungia.  相似文献

In order to investigate the distribution of rare earth elements (REEs) in the natural hyperaccumulator fern Dicranopteris dichotoma Bernh. and to characterize this plant photosynthetically, concentrations of REEs in D. dichotoma from mines mining heavy and light REEs (HREEs and LREEs, respectively), as well as in D. dichotoma from an area in which no mining occurred, in southern Jiangxi Province were determined using inductively coupled plasma-mass spectrometry. The REE concentrations in the lamina of D. dichotoma were in the order LREEs mine 〉 HREEs mine 〉 non-mining area. The maximum REE content in the lamina of D. dichotoma from the LREE mine was approximately 2 648 mg/kg dry weight. The photosynthetic activity of D. dichotoma from areas of HREE and LREE mines was improved by the presence of high concentrations of REEs in the lamina compared with D. dichotoma from the non-mining area. However, this enhancement varied according to the concentrations of the REEs, as well as their type. In addition, 77K fluorescence, electron transport rate, and chlorophyll-protein complex studies showed that the enhancement of the photosynthetic activity of D. dichotoma from HREE mines was mainly due to an increase in the chlorophyll-protein complex of the reaction center of photosystem (PS) Ⅰ, whereas the enhancement observed in D. dichotoma from LREE mines was due to an increase in the internal antennae chlorophyll-protein complex of PS Ⅱ and greater light energy distribution to the light-harvesting chlorophyll-protein complex of PS Ⅱ.  相似文献

Innate immunity is expected to be very important in fish. Mannose-bingding lectin (MBL) participates in the innate immune system as an activator of the complement system and as an opsonin after binding to certain carbohydrate structures on microorganisms. In this experiment, total mRNA was isolated from spleen, liver, gills, thymus, head kidney and kidney of adult and immature grass carp Ctenopharygodon idllus. The cDNA of MBL was obtained by RT-PCR using total mRNA from the spleen of carp as template. Such cDNA was labled with ^32p and used as probe for Northern analysis, and autoradiographic signals were quantified by densitometry analysis. The results showed that MBL was high expressed in the spleen and liver and low in gills, thymus, head kidney and kidney of adult grass carp, and MBL was much lower expressed in spleen and liver of immature grass carp than those of adult grass carp. The results might partially explain why immature grass carp are vulnerable to grass carp hemorrhage virus (GCHV) whereas adult grass carp are not.This suggested that MBL mav be an imoortant anti-GCHV factor [Acta Zoologica Sinica 50 (1): 137 - 140. 2004].  相似文献

Two hundred and eighteen serum samples from 175 lung cancer patients and 43 healthy individuals were analyzed by using Surface Enhaced Laser Desorption/Ionization Time of Flight Mass Spectrome- try (SELDI-TOF-MS). The data analyzed by both Biomarker Wizard? and Biomarker Patterns? software showed that a protein peak with the molecular weight of 11.6 kDa significantly increased in lung cancer. Meanwhile,the level of this biomarker was progressively increased with the clinical stages of lung cancer. The candidate biomarker was then obtained from tricine one-dimensional sodium dodecyl sul- fate-polyacrylamide gel electrophoresis by matching the molecular weight with peaks on WCX2 chips and was identified as Serum Amyloid A protein (SAA) by MALDI/MS-MS and database searching. It was further validated in the same serum samples by immunoprecipitation with commercial SAA antibody. To confirm the SAA differential expression in lung cancer patients, the same set of serum samples was measured by ELISA assay. The result showed that at the cutoff point 0.446(OD value)on the Receiver Operating Characteristic (ROC) curve, SAA could better discriminate lung cancer from healthy indi- viduals with sensitivity of 84.1% and specificity of 80%. These findings demonstrated that SAA could be characterized as a biomarker related to pathological stages of lung cancer.  相似文献

Juvenile mortality in cheetahs was found to be extremely high compared to other large mammals, with approximately 72.2% of litters dying before they emerged from the lair at eight weeks of age. An average of 83.3% of cubs alive at emergence died by adolescence at 14 months of age, thus cheetah cubs were estimated to have only a 4.8% chance of reaching independence at birth. The instantaneous rate of mortality was highest immediately after cubs emerged from the lair. Before emergence, lion predation was the major source of this mortality, although some cubs died from starvation after they were abandoned by their mothers, or as a result of grass fires and inclement weather. After emergence, predation again accounted for virtually all cub mortality, with lions and spotted hyaenas taking approximately the same proportion of cubs. Overall predation accounted for 73.2% of cheetah cub deaths in this study, with 78.2% of these being killed by lions. The extent of maternal care, in the form of vigilance and antipredator behaviour, mirrored cub susceptibility to mortality and, in the case of vigilance, possibly also starvation. The probability of a cheetah mother responding aggressively to a predator was found also to depend on the species of predator. This study highlights the importance of the influence of juvenile mortality on patterns of parental care.  相似文献

We determined the body composition from the carcasses of three 3- to 4-months-old polar bear cubs found dead at Svalbard, Norway. Two of the cubs were killed by a male bear at the den and one cub died from starvation. These three carcasses weighed 70-77% of total body mass of cubs. The ash and protein content of the three carcasses were similar but the starving cub had higher water content and lower fat content than the two killed cubs. The fat content varied from 1.3% (starving cub) to 12.9% while the content of water and fat was similar (about 76%) in the three cubs. More data on the body composition of polar bear cubs at time of emergence from the den are needed to understand the breeding ecology and reproductive energetics of female polar bears.  相似文献

11 只5 ~ 11 月龄的断奶大熊猫幼兽先后患一种急性传染性、顽固性腹泻病,1 只大熊猫幼兽在发病4 d 后死亡,相同饲养条件下由母兽哺育的同龄大熊猫幼兽未见发病。取发病大熊猫幼兽粪便、血液以及呕吐物进行细菌学和寄生虫学检查,未检出病原;用轮状病毒粪便抗原ELISA 快速诊断,受检的8 只大熊猫幼兽腹泻便都多次检出阳性结果。采集发病大熊猫的病料进行病毒病原分离,从腹泻大熊猫幼兽的粪便中分离出病毒颗粒,通过电镜形态、理化特性和中和试验鉴定,确认分离病毒为轮状病毒。   相似文献

Based on in-vitro produced (IVP) bovine embryos, embryo proper and embryonic/fetal membranes were studied in 12 pregnancies from day 26 to 47. The embryos/fetuses displayed external as well as internal development of organs and structures according to the expectations from comparable in-vivo studies. However, the embryonic/fetal membranes were shorter than those reported for in-vivo-derived embryos/fetuses on days 26–35 of calculated age, whereas on days 41–47 they were of comparable lengths.  相似文献

Ureaplasma diversum was isolated from the lungs and/or stomach fluid and placentas of five aborted bovine fetuses and four newborn calves. All isolates were serotype D48. Placentitis was observed in all instances in which the placenta was examined. Gross lesions consisted of focal or diffuse reddening of the chorioallantois and amnion and thickening of the amnion. Microscopically there were fibrosis, edema and inflammation of the amnion. Microscopic lesions in the lung consisted of diffuse pneumonitis with thickening of the alveolar walls and in some cases peribronchiolar lymphoid accumulations. Macrophages and granulocytes were present in the alveoli. Inoculation of the vulva of a virgin heifer with one of the isolates from a fetal lung produced hyperemia and profuse purulent discharge with slight granularity.  相似文献

Although assisted reproductive techniques are commonly applied in humans and animals, they are frequently associated with major developmental deficits and reduced viability. To explore abnormalities associated with cloning or nuclear transfer (NT) as the most invasive of these methods, we used a bovine model to characterize abnormalities. Detailed necropsy examinations were done on 13 calves that died soon after birth; in addition, we included data from embryos and fetuses (produced by NT) that terminated prematurely. Bovine clones that survived until the neonatal period differed quantitatively and qualitatively from in-vivo-derived cattle. Although alterations affected a variety of organs (e.g. heart, lung and liver), there was a clear association with abberant vascular developmental during the early intrauterine phase. Therefore, we concluded that vascular problems were key alterations induced by cloning (presumably via epigenetic modifications).  相似文献

Pathology of brucellosis in bison from Yellowstone National Park   总被引:2,自引:0,他引:2  
Between February 1995 and June 1999, specimens from seven aborted bison (Bison bison) fetuses or stillborn calves and their placentas, two additional placentas, three dead neonates, one 2-wk-old calf, and 35 juvenile and adult female bison from Yellowstone National Park (USA) were submitted for bacteriologic and histopathologic examination. One adult animal with a retained placenta had recently aborted. Serum samples from the 35 juvenile and adult bison were tested for Brucella spp. antibodies. Twenty-six bison, including the cow with the retained placenta, were seropositive, one was suspect, and eight were seronegative. Brucella abortus biovar 1 was isolated from three aborted fetuses and associated placentas, an additional placenta, the 2-wk-old calf, and 11 of the seropositive female bison including the animal that had recently aborted. Brucella abortus biovar 2 was isolated from one additional seropositive adult female bison. Brucella abortus was recovered from numerous tissue sites from the aborted fetuses, placentas and 2-wk-old calf. In the juvenile and adult bison, the organism was more frequently isolated from supramammary (83%), retropharyngeal (67%), and iliac (58%) lymph nodes than from other tissues cultured. Cultures from the seronegative and suspect bison were negative for B. abortus. Lesions in the B. abortus-infected, aborted placentas and fetuses consisted of necropurulent placentitis and mild bronchointerstitial pneumonia. The infected 2-wk-old calf had bronchointerstitial pneumonia, focal splenic infarction, and purulent nephritis. The recently-aborting bison cow had purulent endometritis and necropurulent placentitis. Immunohistochemical staining of tissues from the culture-positive aborted fetuses, placentas, 2-wk-old calf, and recently-aborting cow disclosed large numbers of B. abortus in placental trophoblasts and exudate, and fetal and calf lung. A similar study with the same tissue collection and culture protocol was done using six seropositive cattle from a B. abortus-infected herd in July and August, 1997. Results of the bison and cattle studies were similar.  相似文献

Using cub growth as an index, I examine the influence of maternalnutrition, litter size, and cub sex on maternal care in cheetahs(Acinonyx jubatus) and compare cub and litter growth rates withthose of other large feilds. Seventy-nine free-living cheetahcubs in 21 litters from 15 mothers were weighed at least oncebetween 6 and 48 days of age. Eleven litters were weighed atthe beginning and end of a 5-day observation of their mothers.The mean cub growth rate varied significantly between litters,due primarily to differences in maternal food intake. Growthdeclined sharply when maternal food intake was less than 1.5kg/ day, but did not increase with greater levels of food intake.Lower limits of growth rates may therefore have been set bythe mother's food intake, whereas upper limits may be set bythe intrinsic physiological ability of cubs to grow. Althoughmale cubs were heavier than female cubs in the same litter whenfirst weighed, major differences in growth rate between thesexes were not apparent at this stage. Both cheetah cubs andlitters grow fast relative to other large felids, and I arguethat this may be an adaptation to the high rate of cheetah juvenilemortality from predation.  相似文献
