This study describes a novel strategy to improve the growth performance of Lactococcus lactis by heterologous production of food-grade transglutaminase. The mtg gene from Streptoverticillium mobaraense that encodes the transglutaminase mature protein was cloned into a nisin-inducible expression vector and transformed into L. lactis subsp. cremoris NZ9000. The leaky expression of the mtg gene from the nisA promoter resulted in ammonia formation and carbon flux redistribution at the pyruvate branch. As a consequence, medium acidification was lessened and energy utilization was improved. This led to significantly higher biomass production under aerobic conditions and particularly under non-pH-controlled conditions (up to a 12-fold increase). The results presented here provide a novel way to enhance the growth yield of L. lactis, which is an important step for the purposes of producing proteins of commercial interest using L. lactis as a host.  相似文献

Elongation factor Tu (EF-Tu) ofStreptoverticillium mobaraense, which produces pulvomycin, has been prepared to 90% purity. The purified protein differs significantly from the analogous protein found inEscherichia coli in molecular weight and antibiotic sensitivity. EF-Tu migrates in sodium dodecyl sulfate gel electrophoresis as a 46,000-dalton species. The protein is sensitive to pulvomycin, but highly resistant to kirromycin. EF-Tu fromStv. mobaraense exists in multiple forms as monomer and polymers. By contrast to the monomer, the polymers of EF-Tu are completely resistant to pulvomycin.  相似文献

Immobilization of aminoacylase from Streptoverticillium olivoreticuli by incorporation into acrylamide gel has been investigated. The data showed that the process should be carried out at constant pH. Thermal inactivation of the immobilized enzyme under the reaction conditions was studied. The kinetics of enzymatic stereospecific deacylation of N-acetyl-DL-phenylalanine was analyzed. The results of this study may be used in the synthesis of amino acid enantiomers.  相似文献

The lethal and mutagenic effects of N-nitrozo-N-methyl biuret (NMB), N-nitrozo-N-methyl urea (NMU) and UV light on Streptoverticillium mycoheptinicum, strains O883 and 852, were studied. The concentrations of NMB were 0.005, 0.1 and 0.25 per cent, the exposure time was 2, 4 and 6 hours. The concentration of NMU was 1 per cent and the exposure time was 1, 2, 3, 4 and 5 hours. The dose of UV light was 2000 erg/mm2. When the spores of Streptoverticillium mycoheptinicum, strain O883, were treated with NMB, the frequency of auxotrophic mutants increased from 0.63 to 3.4 per cent with an increase of the mutagen concentration from 0.05 to 0.25 per cent and the exposure time from 2 to 6 hours. More than 80 auxotrophic mutants were selected. When Streptoverticillium mycoheptinicum, strain 852, was treated with NMU, the frequency of auxotrophic mutants ranged from 0.5 to 2.4 per cent. Fifty-seven auxotrophic mutants were selected. The majority of the auxotrophic mutants selected with the use of NMB and NMU was unstable. Exposure of Streptoverticillium mycoheptinicum, strains 852, 10/69 Met and 54/100 Lys to UV light resulted in formation of groups of polyauxotrophic mutants.  相似文献

Bovine beta-lactoglobulin (BLG) is a major component in whey and its physical properties are important for the texture of many dairy-based foods. Modification of proteins with transglutaminase from Streptoverticillium mobaraense (MTGase) can be used to alter their physical properties. MTGase-mediated modification of native BLG was until now, however, not effective. Here we report a method that allows for the enzymatic modification of native BLG with MTGase. Lysines 8, 77, and 141 were modified with alpha-N-carbobenzyloxy-glutamine-glycine and glutamines 35, 59, 68,and 155 were modified with 6-aminohexanoic acid under nonreducing and nondenaturing conditions. MTGase-mediated BLG crosslinking is hampered by the low reactivity of the lysines and enzymatic deamidation of the glutamines prevails. Modification of BLG with poly-lysine yields a BLG derivative with increased affinity for the water-air interface and stronger surface tension lowering capacities than normal BLG. Hence, this modification method offers the opportunity to change the functional properties of BLG and to prepare novel protein foods.  相似文献

Abstract Members of the genera Streptomyces, Micromonospora, Nocardia and Streptosporangium were effectively inhibited by oxytetracycline at concentrations tolerated by a majority of the Streptoverticillium strains tested. Rubrivivax gelatinosus having both the spheroidene and spirilloxanthin biosynthetic pathways produces carotenoids (Cars) with a variety of conjugated chains, which consist of different numbers of conjugated double bonds (n), including the C=C (m) and C=O (o) bonds. When grown under anaerobic conditions, the wild type produces Cars for which n = m = 9-13, whereas under semiaerobic conditions, it additionally produces Cars for which n = m + o = 10 + 1, 13 + 1, and 13 + 2. On the other hand, a mutant, in which the latter pathway is genetically blocked, produces only Cars for which n = 9 and 10 under anaerobic conditions and n = 9, 10, and 10 + 1 under semianaerobic conditions. Those Cars that were extracted from the LH2 complex (LH2) and the reaction center (RC), isolated from the wild-type and the mutant Rvi. gelatinosus, were analyzed by HPLC, and their structures were determined by mass spectrometry and 1H NMR spectroscopy. The selective binding of Cars to those pigment-protein complexes has been characterized as follows. (1) Cars with a shorter conjugated chain are selectively bound to LH2 whereas Cars with a longer conjugated chain to the RC. (2) Shorter chain Cars with a hydroxyl group are bound to LH2 almost exclusively. This rule holds either in the absence or in the presence of the keto group. The natural selection of shorter chain Cars by LH2 and longer chain Cars by the RC is discussed, on the basis of the results now available, in relation to the light-harvesting and photoprotective functions of Cars.  相似文献

为改善乳酸乳球菌的生长性能,以轮枝链霉菌染色体DNA为模板,扩增得到编码谷氨酰胺转胺酶成熟酶的基因mtg,将其克隆到质粒pNZ8148中,电转化乳酸乳球菌NZ9000,获得乳酸乳球菌NZ9000(pFL001)(重组菌)。在不控制pH条件下,重组菌的胞外pH显著高于对照菌NZ9000(pNZ8148);前者的最高生物量可达4.13gL,而后者只有0.34gL。在控制pH为6.5±0.1的条件下,重组菌最高生物量为4.73gL,对葡萄糖的菌体最高平均得率为71.1gmol,而相同条件下对照菌最高生物量为2.6gL,对葡萄糖的菌体最高平均得率为27.3gmol。由此表明,重组菌与对照菌相比,好氧生长性能得到显著改善。可能的原因是mtg的活性表达升高了重组菌的胞内pH,原先用于泵出胞内H 所需的部分能量可能因此得到节省,这样相应增加了用于细胞生长的能量。  相似文献

Intergeneric crossing of the actinomycetes Streptoverticillium mycoheptinicum and Streptomyces coelicolor yielded recombinants which were mostly nonviable and unstable despite a relatively high frequency (10(-2)-10(-4)) at which their colonies appeared. The rare viable and stable recombinants were prototrophs. The structure of antibiotics in the hybrid cell may be modified as follows from the differences in the antibiotic activity of the LIA-973 hybrid and the parent strains.  相似文献

Complete 5S ribosomal RNA sequences were obtained for four acidophilic actinomycetes, seven neutrophilic streptomycetes and a strain of Streptoverticillium baldaccii. All of the organisms contained RNAs belonging to the 120 nucleotide type. An evolutionary tree was generated after combining the test data with results from similar studies on representative Gram-positive bacteria. The acidophilic, neutrotolerant and neutrophilic actinomycetes were recovered in a distinct cluster that was equated with the genus Streptomyces. The sequence data support the view that the genera Chainia, Elytrosporangium, Kitasatoa and Microellobosporia should be considered as synonyms of the genus Streptomyces. The recovery of the Streptoverticillium baldaccii strain on the fringe of the Streptomyces cluster is also consistent with current trends in the taxonomy of these organisms. Further work is needed to determine the taxonomic status of the two streptomycete subgroups that comprised the streptomycete cluster.  相似文献

The structural gene for phospholipase D (PLD) of an actinomycete, Streptoverticillium cinnamoneum, together with its promoter region was introduced into Streptomyces lividans using a shuttle vector—pUC702—for Escherichia coli and S. lividans. The transformant was found to secrete a large amount of PLD (about 2.0×104 U/l, 42 mg/l) when cultured in a jar fermentor. Both an initial glucose concentration of 17.5 g/l and the feeding of carbon and nitrogen sources are effective for efficient secretion of PLD; under these culture conditions, the amount of PLD secreted reached a maximum level (about 5.5×104 U/l, 118 mg/l) after about 60 h. In contrast to the original producer, Stv. cinnamoneum, which secretes only a small amount of PLD (about 1.1×103 U/l, 2 mg/l) along with other extracellular proteins, this heterologous expression system is markedly more efficient in production of secretory PLD.  相似文献

The fluorescence of (Na,K)-ATPase labeled with 5-iodoacetamidofluorescein was studied under turnover conditions. At 4 degrees C the hydrolysis of ATP is slowed sufficiently to permit study of the effects of Na+, K+, and ATP on the steady-state intermediates. With Na+ and Mg2+ (Na-ATPase conditions), addition of ATP produces a 7% drop in signal that reverts back to the initial, high fluorescence after a steady state of several minutes. K-sensitive phosphoenzyme is formed under these conditions, indicating that the fluorescence signal during the steady state is associated with E2P. Under (Na,K)-ATPase conditions (Na+, K+, Mg2+), micromolar ATP produces a steady-state signal that is 25% lower than the initial fluorescence, with no detectable phosphoenzyme formed. This low-fluorescence intermediate, which is also formed by adding K+ to enzyme in the Na-ATPase steady state described above, resembles the state produced by adding K+ directly to enzyme under equilibrium conditions, i.e. E2K. The K0.5(K+) for the fluorescence decrease and for keeping the enzyme dephosphorylated are nearly identical, indicating that the fluorescence change accompanies K+-dependent dephosphorylation. High ATP increases the steady-state fluorescence during the (Na,K)-ATPase reaction; while oligomycin produces still another steady-state fluorescent intermediate. These last two intermediates may be associated with the formation of E2P and E1P, respectively.  相似文献

Production of dihydrothymidine stereoisomers in DNA by gamma-irradiation   总被引:3,自引:0,他引:3  
5,6-Dihydrothymidine (dDHT) is a derivative thymidine formed during gamma-irradiation. This paper demonstrates the conditions under which dDHT is formed in solutions of DNA and that dDHT is produced in the DNA of HeLa cells during gamma-irradiation. The product of dDHT by gamma-irradiation of either thymidine or DNA has been quantitated by a sensitive and specific high-pressure liquid chromatography method. dDHT is a major product of the anoxic irradiation of thymidine (G value 0.5) but is produced in substantially smaller amounts in DNA irradiated under the same conditions (G value 0.026). The presence of oxygen reduces the yield of dDHT by at least 25-fold for both irradiation substrates. In HeLa cells, 60Co irradiation under anoxia produces (6.2 +/- 0.2) X 10(-8) mol of the R isomer of dDHT per mole of cell deoxynucleotide per gray (G value 0.11). gamma-Irradiation of thymidine produces equal quantities of the R and S stereoisomers of dDHT. Irradiation of DNA produces significantly more (69%) (R)- than (S)-dDHT. DNA isolated from cultured human cells following gamma-irradiation also contains more of the R than the S form of dDHT. The conformation of double-stranded DNA favors a stereospecific production of the R isomer. Among products of gamma-irradiation of DNA, dDHT is unique in its strict requirement for anoxia during irradiation and the preferential production of a particular stereoisomer.  相似文献

Females from female-biased populations of the notostracan Triops newberryi produce viable eggs when reared in isolation. Clutches produced under such conditions exhibit sex-ratio polymorphism. One category of females (monogenics) produces only female offspring; the second category (amphigenics) produces clutches with a sex ratio of 3 females: 1 male. The relative proportions of the two categories of females varies significantly between populations and is correlated with population sex ratio. This correlation, and the pattern of offspring distribution in amphigenics, suggests that sex is determined by an autosomal Mendelian gene locus for which the male-determining allele is recessive. Limited pedigree analysis of lineages under selfing support the genetic model.  相似文献

In order to delineate the roles of lignin and manganese peroxidases in the degradation of polycyclic aromatic hydrocarbons by Phanerochaete chrysosporium, the biodegradation of phenanthrene (chosen as a model for polycyclic aromatic hydrocarbons) was investigated. The disappearance of phenanthrene from the extracellular medium and mycelia was determined by using gas chromatography. The disappearance of phenanthrene from cultures of wild-type strains BKM-F1767 (ATCC 24725) and ME446 (ATCC 34541) under ligninolytic (low-nitrogen) as well as nonligninolytic (high-nitrogen) conditions was observed. The study was extended to two homokaryotic (basidiospore-derived) isolates of strain ME446. Both homokaryotic isolates, ME446-B19 (which produces lignin and manganese peroxidases only in low-nitrogen medium) and ME446-B5 (which totally lacks lignin and manganese peroxidase activities), caused the disappearance of phenanthrene when grown in low- as well as high-nitrogen media. Moreover, lignin and manganese peroxidase activities were not detected in any of the cultures incubated in the presence of phenanthrene. Additionally, the mineralization of phenanthrene was observed even under nonligninolytic conditions. The results collectively indicate that lignin and manganese peroxidases are not essential for the degradation of phenanthrene by P. chrysosporium. The observation that phenanthrene degradation occurs under nonligninolytic conditions suggests that the potential of P. chrysosporium for degradation of certain environmental pollutants is not limited to nutrient starvation conditions.  相似文献
