Three new iridoid glycosides, 6″-O-trans-caffeoylgenipin gentiobioside (1), genipin 1-O-β-d-apiofuranosyl (1→6)-β-d-glucopyranoside (2), genipin 1-O-α-d-xylopyranosyl (1→6)-β-d-glucopyranoside (3), three new monocyclic monoterpenoids, jasminoside R (4), jasminoside S (5), jasminoside T (6), together with nine known iridoid glycosides (715) and three crocetin glycosides (1618), were isolated from the fruit of Gardenia jasminoides. Their chemical structures were established mainly by 1D and 2D NMR techniques and mass spectrometry. Inhibitory effects of the isolated compounds on nitric oxide production in lipopolysaccaride-activated macrophages were evaluated. Compounds 8 and 18 showed strong inhibitory activity on NO production with IC50 values of 11.14 ± 0.67 and 5.99 ± 0.54 μM, respectively.  相似文献

Two new butyrolactones: aspernolides F (6) and G (7), together with three stigmasterol derivatives: (22E,24R)-stigmasta-5,7,22-trien-3-β-ol (1), stigmast-4-ene-3-one (2), and stigmasta-4,6,8(14), 22-tetraen-3-one (3), two meroterpenoids: terretonin A (4) and terretonin (5), and a butyrolactone derivative: butyrolactone VI (8) have been isolated from the endophytic fungus Aspergillus terreus isolated from the roots of Carthamus lanatus (Asteraceae). Their structures were determined by spectroscopic means (1D, 2D NMR, and HRESIMS), as well as optical rotation measurement and comparison with literature data. The isolated compounds were evaluated for their anti-microbial, anti-malarial, anti-leishmanial, and cytotoxic activities. Compound 1 displayed a potent activity against MRSA and C. neoformans with IC50 values of 0.96 μg/mL and 4.38 μg/mL, respectively compared to ciprofloxacin (IC50 0.07 μg/mL) and amphotericin B (IC50 0.34 μg/mL), respectively. While, 6 showed good activity against C. neoformans (IC50 5.19 μg/mL) and mild activity against MRSA (IC50 6.39 μg/mL). Moreover, 1 and 2 exhibited very good anti-leishmanial activity towards L. donovani with IC50 values of 4.61 and 6.31 μg/mL, respectively and IC90 values of 6.02 and 16.71 μg/mL, respectively.  相似文献

A bioassay guided isolation of potential antimalarial molecules from the stem bark of Caesalpinia volkensii Harms (Fabaceae) achieved three new 11-oxocassane-type diterpenoids named voulkensin C (1), D (2) and E (3) together with one steroid glycoside named 3-O-[β-glucopyranosyl(1→2)-O-β-xylopyranosyl]-stigmasterol (4) and seven other known compounds including stigmasterol (5), β-sitosterol (6), oleanolic acid (7), 3-β-acetoxyolean-12-en-28-methyl ester (8), voucap-5-ol (9), caesadekarin C (10), deoxycaesaldekarin C (11). The structures of the new compounds were determined on the basis of extensive spectroscopic data (IR, MS, 1H and 13C NMR and 2D NMR) analyses. The polar extracts revealed moderate to good antiplasmodial activities against chloquine-sensitive (D6) and -resistant strains (W2) of Plasmodium falciparum. Whereas the pure isolates exhibited limited to moderate antiplasmodial activities with compound 4 showing the highest antiplasmodial activities (IC50 values of 4.44 ± 0.88 and 2.74 ± 1.10 μM against D6 and W2 strains, respectively). These results suggest a possible contribution of phytochemicals from C. volkensii stem bark towards inhibition of plasmodial parasites’ growth hence potential antimalarial.  相似文献

Suspension cultures of Abronia nana were established to produce C-methylisoflavones. A new C-methylrotenoid, named abronione A (2), was isolated along with three known rotenoids, boeravinone D (1), boeravinone A methyl ether (3), and mirabijalone D (4). The IC50 values of compounds 1, 2, and 4 on β-secretase (BACE1) were 4.77, 62.21, and 4.24 μM, respectively, whereas 3 was inactive. At concentrations up to 1.0 mM, the compounds did not inhibit other proteases such as trypsin, chymotrypsin, and elastase, indicating that they were specific inhibitors of β-secretase. Compounds 1 and 4 were non-competitive inhibitors based on the Dixon plot and with Ki values of 5.01 and 4.28 μM, respectively. At 50 μM, compound 4 inhibited Aβ1–42 production by 43.7% in APPSW-N2a cells.  相似文献

Subvellerolactones B (1), D (2), and E (3), structurally unusual lactarane sesquiterpenoids, were isolated from the fruiting bodies of Lactarius subvellereus together with four known lactarane sesquiterpenes (47). The chemical structures and stereochemistries of compounds 13 were determined on the basis of spectroscopic analyses, including 1D and 2D NMR experiments and a convenient Mosher ester procedure. Subvellerolactone B (1) exhibited cytotoxicity against the A549, SK-MEL-2, and HCT-15 cell lines with IC50 values of 26.5, 18.3, and 14.2 μM, respectively, and subvellerolactones D (2) and E (3) showed cytotoxicity against the A549 and HCT-15 cell lines (IC50 (2): 25.1 and 17.8 μM, and IC50 (3): 19.6 and 28.7 μM, respectively).  相似文献

Six new compounds including two oleanane-type triterpenoid saponins (1, 2) and four C-glycosyl flavones (36), along with a known saponin (7), three di-C-glycosyl flavones (810) and a glycosyl auronol (11), were isolated from the stem bark of Erythrina abyssinica Lam. The structures of the new compounds, identified as 3-O-[α-l-rhamnopyranosyl-(1  2)-β-d-galactopyranosyl-(1  2)-β-d-glucuronopyranosyl]-22-O-β-d-glucopyranosyl sophoradiol (1), 3-O-[α-l-rhamnopyranosyl-(1  2)-β-d-glucopyranosyl-(1  2)-β-d-glucuronopyranosyl]-22-O-β-d-glucopyranosyl sophoradiol (2), 6-C-β-glucopyranosyl-8-C-β-quinovopyranosyl apigenin (3), 6-C-β-quinovopyranosyl-8-C-β-glucopyranosyl apigenin (4), 8-C-[6″-O-α-l-rhamnopyranosyl-(1‴  6″)]-β-glucopyranosyl 7,4′-dihydroxyflavone (5) and 8-C-[6″-O-β-d-xylopyranosyl-(1‴  6″)]-β-glucopyranosyl 7,4′-dihydroxyflavone (6), were determined by comprehensive spectroscopic analysis, including 1D and 2D NMR techniques, mass spectrometry and acid hydrolysis. These new compounds together with the known saponins 7 were evaluated for their cytotoxic activity against MCF-7 (estrogen dependent) and MDA-MB-231 (estrogen independent) cell lines. The new saponin 2 exhibited the highest cytotoxic activity among tested compounds, exerting a selective inhibitory effect against the proliferation of MCF-7 cells, with lower IC50 value (12.90 μM) than that of the positive control, resveratrol (13.91 μM). Structure–activity relationship of these compounds is also discussed.  相似文献

Two new tirucallane triterpenoids, 21-methoxy-21,23-epoxy-tirucalla-7,24-dien-3α-ol (1) and 21-methoxy-21,23-epoxy-tirucalla-7,24-diene-1α,3α-diol (2), together with thirteen known compounds were isolated from the CH2Cl2 extract of the stem bark of Araliopsis synopsis. The structures of the compounds were determined by comprehensive analyses of their 1D and 2D NMR, mass spectral (EI and ESI) data and comparison with previously known analogs. Compounds 110 were tested against bacteria, fungi and plant pathogen oomycetes by the paper disk agar diffusion assay resulting in missing to low activities corresponding with MICs > 1 mg/mL. However, compounds 510 exhibited high cytotoxic activity against the human Caucasian prostate adenocarcinoma cell PC-3 line, with IC50 8.5–12.5 μM compared to the standard Doxorubicin with IC50 = 0.9 μM, while compounds 1, 3 and 4 showed low activity.  相似文献

A new lanostane aldehyde, charantal (1), was isolated from the ethanolic leaf extract of Momordica charantia together with the known compound, 2,4-bis(2-phenylpropan-2-yl)phenol (2). The structure of compound 1 was elucidated by extensive 1D and 2D NMR and MS experiments. Compound 2 displayed a moderately strong antitubercular activity against Mycobacterium tuberculosis H37Rv (MIC = 14 μg/mL) according to the MABA susceptibility assay.  相似文献

From the leaves of Nelumbo nucifera (an aquatic plant), one new compound, 24(R)-ethylcholest-6-ene-5α-ol-3-O-β-d-glucopyranoside (1), along with 11 known metabolites (212), were isolated and identified by spectroscopic methods including 1D- and 2D NMR. Antifungal activity for (R)-roemerine (3) (IC50/MIC = 4.5/10 μg/mL against Candida albicans) and antimalarial activity for (R)-roemerine (3) and N-methylasimilobine (5) (IC50 = 0.2 and 4.8 μg/mL for the D6 clone, respectively, and 0.4 and 4.8 μg/mL for the W2 clone, respectively) was observed. None of the compounds were cytotoxic to Vero cells up to a concentration of 23.8 μg/mL. NMR data for 10-eicosanol (7) and 7,11,15-trimethyl-2-hexadecanone (10) are presented for the first time. An analysis of the structure–activity relationship shows that the substituents in position C-1 and C-2 of aporphine alkaloids are crucial for the antimalarial activity.  相似文献

Bioassay guided fractionation of the roots of Lantana montevidensis (Verbenaceae) has resulted in the isolation and identification of three new triterpenoids; 13β-hydroxy-3-oxo-olean-11-en-28-oic acid (1), 12β,13β-dihydroxyolean-3-oxo-28-oic acid (2) and 12β,13β,22β-trihydroxyolean-3-oxo-28-oic acid (3) in addition to nine known compounds: oleanonic acid (4), oleanolic acid (5), 3β,25β-dihydroxy-olean-12-en-28-oic acid (6), lantadene A (7), 19α-hydroxy-3-oxo-olean-12-en-28-oic acid (8) pomolic acid (9), camaric acid (10) together with β-sitosterol (11) and β-sitosterol-3-O-β-d-glucoside (12). The structures of the isolated metabolites were elucidated based on comprehensive 1D and 2D NMR spectroscopic data as well as HR-ESI–MS. The extracts and the isolated metabolites were evaluated for their antiprotozoal and antimicrobial activities. Compound 2 showed antibacterial activity against Staphylococcus aureus and methicillin resistant S. aureus with IC50 values against both organisms of 2.1 μM and compound 10 showed activity against same organisms with IC50 values 8.74 and 8.09 μM, respectively, compared to the positive control ciprofloxacin (IC50 = 0.3 μM against S. aureus and MRSA). Compounds 1, 4, 5, 6, and 10 showed moderate antileishmanial activity with IC50 values ranging between (2.54–14.95 μM) and IC90 values ranging between (11.90–19.47 μM), using pentamidine as a control (IC50 values 2.09  16.8 μM) and IC90 values ranging between (4.72  16.8 μM). These compounds also showed highly potent antitrypanosomal activity with IC50 values ranging between (0.39–7.12 μM) and IC90 values ranging between (1.91–10.51 μM), which are more efficient than the DFMO, the antitrypanosomal drug employed as positive control (IC50 and IC90values 11.82 and 30.82 μM).  相似文献

Two new lignans, gymnothelignans V (1) and W (2), were isolated from a methanol extraction of Gymnotheca chinensis Decne. Their structures were established on the basis of extensive 1D and 2D NMR spectroscopy. Compound 1 exhibited moderate cytotoxicity against the HCT116, HCT15, A549, MCF-7 and HepG2 cancer cell lines with IC50 values of 45.1 μM, 26.9 μM, 49.6 μM, 30.0 μM and 49.7 μM, respectively. Compound 2 exhibited weak cytotoxicity against the A549 cancer cell line with an IC50 value of 41.3 μM.  相似文献

Six 1,3-diphenylpropanes exhibiting inhibitory activities against both the monophenolase and diphenolase actions of tyrosinase were isolated from the methanol (95%) extract of Broussonetia kazinoki. These compounds, 16, were identified as kazinol C (1), D (2), F (3), broussonin C (4), kazinol S (5) and kazinol T (6). The latter two species (5 and 6) emerged to be new 1,3-diphenylpropanes which we fully spectroscopically characterized. The IC50 values of compounds (1, 35) for monophenolase inhibition were determined to range between 0.43 and 17.9 μM. Compounds 1 and 35 also inhibited diphenolase significantly with IC50 values of 22.8, 1.7, 0.57, and 26.9 μM, respectively. All four active tyrosinase inhibitors (1, 35) were competitive inhibitors. Interestigly they all mainfested simple reversible slow-binding inhibition against diphenolase. The most potent inhibitor, compound 4 diplayed the following kinetic parameters k3 = 0.0993 μM?1 min?1, k4 = 0.0048 min-1, and Kiapp = 0.0485 μM.  相似文献

Three new cassane furanoditerpenes, phanginins N–P (13), together with four known ones were isolated from the seeds of Caesalpinia sappan. Their structures were elucidated on the basis of spectroscopic analysis including HRESIMS, 1D and 2D NMR techniques. Two new steroidal saponins, 25(R)-3β [(O-β-d-glucopyranosyl-(1  3)-β-d-glucopyranosyl-(1  2)-O-[β-d-xylopyranosyl-(1  3)-O-β-d-glucopyranosyl-(1  4)-β-d-galactopyranosyl)oxy]-5α, 15β, 22R, 25R-spirostan-3,15-diol (1, named parquispiroside) and 25R-26-[(β-d-glucopyranosyl)Oxy]-(3β [(O-β-d-glucopyranosyl-(1  3)-β-d-glucopyranosyl-(1  2)-O-[β-d-xylopyranosyl-(1  3)-O-β-d-glucopyranosyl-(1  4)-β-d-galactopyranosyl)oxy], 5α, 15β, 22R, 25R)-furostane-3,15,22-triol (2, named parquifuroside), along with the known saponins, capsicoside D (3) and 22-OMe-capsicoside D (4) and the known glycoside, benzyl primeveroside (5), were isolated from the leaves of Cestrum parqui. The structures of these compounds were elucidated by careful analysis of 1D and 2D NMR spectra and ESIMS data. Parquispiroside (1) exhibited moderate inhibition of Hela, HepG2, U87, and MCF7 cell lines with IC50 values in the range of 3.3–14.1 μM.  相似文献

Two new hydroxychavicol analogs nudibaccatumin A (1) and B (2), together with twenty known compounds were isolated from the methanol extract of Piper nudibaccatum. Their structures were elucidated by extensive spectroscopic analyses (1D and 2D NMR, HRESIMS, UV, IR and polarimetry). Hydroxychavicol is a known inhibitor of xanthine oxidase (XO). In the present study, hydroxychavicol and 5 natural analogs (15) were evaluated for their XO inhibitory activity. Neotaiwanensol B (3) (IC50 = 0.28 μM) showed a greater inhibitory effect than hydroxychavicol and allopurinol (the positive control). Two new compounds 1 and 2 showed a moderate inhibition activity with an IC50 value of 62.94 μM and 70.67 μM, respectively.  相似文献

Three new phenylethanoid glycosides, 2-(3-hydroxy-4-methoxyphenyl)ethyl 1-O-β-d-allopyranoside (hodgsonialloside A, 1), 2-(3-hydroxy-4-methoxyphenyl)ethyl 1-O-β-d-glucopyranosyl-(1  4)-β-d-allopyranoside (hodgsonialloside B, 2) and 2-(3-methoxy-4-hydroxyphenyl)ethyl 1-O-β-d-allopyranoside (hodgsonialloside C, 3) were isolated from the leaves of Magnolia hodgsonii in addition to six known compounds, tyrosol 4-O-β-d-xylopyranosyl-(1  6)-β-d-glucopyranoside (4), kaempferol 3-O-neohesperidoside (5), kaempferol 3-O-rutinoside (6), kaempferol 3-O-α-l-rhamnopyranosyl-(1  2)-[α-l-rhamnopyranosyl-(1  6)]-β-d-glucopyranoside (7), (+)-syringaresinol O-β-d-glucopyranoside (8), and oblongionoside C (9). The structure elucidation of these compounds was based on analyses of physical and spectroscopic data including 1D and 2D NMR experiments.  相似文献

Reactions of ligand 2-(1H-1,2,4-triazol-1-yl)acetic acid (HL) with varied metal salts of Cu(II), Co(II), Ni(II), Zn(II), Cd(II) and Ag(I) result in formation of six new coordination complexes, {[Cu(L)2] · 3H2O}n (1), [Co(L)2(H2O)2]n (2), [Ni(L)2(H2O)2]n (3), [Zn(L)2(H2O)2]n, (4), [Cd(L)2]n (5) and [Ag(L)]n (6), and their structures were determined by X-ray crystallography. Complexes 1, 2, 3 and 4 with square-planar or octahedral metal centers have similar two-dimensional (2D) network structure with (4, 4) topology, while complex 5 displays a 2D structure with (6, 3)-connected topology. Complex 6 has a three-dimensional (3D) structure, in which the Ag(I) has tetrahedral coordination geometry. Ligand L? acts as a 2-connected rod (bridging ligand) in 1, 2, 3 and 4, and acts as 3-connected nodes in 5 and 6. The results indicate that the coordination modes of the ligand and metal centers have great influence on the structures of the complexes. In addition, the photoluminescent properties of ligand HL and complexes 4 and 5 were studied in the solid state at room temperature.  相似文献

Phytochemical investigation of the EtOAc and MeOH extracts from flowers of Getonia floribunda Roxb., a Thai herbal medicine, resulted in the isolation of two new flavonols, 4′-hydroxy-6,7,8,3′-tetramethoxyflavonol (1) and 4′-hydroxy-6,7,8-trimethoxyflavonol (2), along with a known pachypodol (3). Their structures were elucidated by spectroscopic methods including UV, IR, 1D and 2D NMR techniques and MS analysis. Compound 1 showed cytotoxicity against the oral cavity cancer (KB) cell line with an IC50 value of 8.99 ± 2.00 μg/ml.  相似文献

A series of alkynylgold(I) bis(diphenylphosphino)alkyl- and aryl-amine complexes, [{Ph2PN(R)PPh2}Au2(CCR′)2] [R = nPr, R′ = Ph (1), C6H4OMe-p (2), C6H4Me-p (3), C6H4Cl-p (4); R = C6H4OMe-p, R′ = Ph (5)], has been synthesized. The X-ray crystal structures of 1 and 2 revealed the presence of short intramolecular Au⋯Au contacts with the distances of 2.8404(8) and 3.0708(7) Å.
