Three new compounds, 17β-cevanin-6-oxo-5α,20β-diol yibeinine (1), 2-(tetrahydro-5-(2-hydroxyphenyl)-2H-pyran-3-yl) phenol (2), 1,3-O-diferuloyl-2-methoxypropane diol (3), as well as four known compounds (47), have been isolated from the ethanol extract of dried bulbs of Fritillaria pallidiflora Schrenk. All structures were determined based on their spectroscopic data (1D and 2D NMR (including 1H NMR, 13C NMR, 1H-1H COSY, HMBC, HSQC, HSQC-TOCSY, and NOESY experiments), and MS). Biological evaluation showed that compounds 14 inhibited the production of nitric oxide (NO) in LPS-stimulated RAW 264.7 cells with IC50 values of 18.0, 38.7, 29.5, and 47.1 μM, respectively. These results indicated that compound 1 has potential anti-inflammatory activity.  相似文献

Two new isoflavanones (1 and 13), along with 25 known compounds (212, 1427), were isolated from the EtOAc-soluble fraction of the heartwood of Dalbergia odorifera by following their potential to inhibit the LPS-induced nitric oxide production in RAW 264.7 cells. The structures of the isolated compounds were established by spectroscopic data such as 1D, 2D NMR and MS spectrometry. Among the isolated compounds, (2S)-pinocembrin (26), showed the most potent inhibitory effect with IC50 value of 18.1 μM.  相似文献

Two new compounds named cleroserroside C (1), schisphenlignan O (2), as well as twenty-one known compounds (3–23) were isolated from the roots of Tripterygium regelii. The structures of the new compounds were determined by 1D and 2D NMR spectra and HR-ESI-MS data. The known compounds were determined by comparing the 1D NMR data in the literature. All compounds were evaluated for anti-inflammatory activity using the LPS-induced RAW264.7 inflammatory cell model, and the results showed that compounds 1, 8-11, 15-16, and 20-21 had good anti-inflammatory activity (IC50 < 20 μM). The cytotoxicity of all compounds was tested by CCK-8 assay, using RAW264.7 cells. The results showed that all compounds had no cytotoxicity to RAW264.7 in the range of 0 ~ 200 μM.  相似文献

Eighteen secondary metabolites were isolated from the fermentation broth of the endophytic fungus Xylaria sp. SYPF 8246, including four new compounds, xylarianins A-D (14), three new natural products, 6-methoxycarbonyl-2-methyl-3,5,4′,6-tetramethoxy-diphenyl ether (5), 2-chlor-6-methoxycarbonyl-2-rnethyl-3,5,4′,6-tetramethoxy-diphenyl ether (6), and 2-chlor-4-hydroxy-6-methoxy carbonyl-2-methyl-3,5,6-trimethoxy-diphenyl ether (7), and eleven known compounds (818). Their structural elucidations were conducted by using 1D and 2D NMR, HRESIMS, and Rh2(OCOCF3)4-induced electronic circular dichroism (ECD) spectra analyses. The integrated 1H and 13C NMR data of three new natural products 57 were reported for the first time. All the isolated compounds were assayed for their inhibitory activities against human carboxylesterase 2 (hCE 2). Compounds 1, 59, and 18 displayed significant inhibitory activities against hCE 2 with IC50 values of 10.43 ± 0.51, 6.69 ± 0.85, 12.36 ± 1.27, 18.25 ± 1.78, 29.78 ± 0.48, 18.86 ± 1.87, and 20.72 ± 1.51 µM, respectively. The interactions between compounds 1 and 5 with hCE 2 were anaylzed by molecular docking.  相似文献

Two new compounds lepidodromos A-B and (1-2) have been isolated from the methanol-soluble extract of the aerial parts of Lepidogrammitis drymoglossoides (Bak.) Ching (Family: Polypodiaceae) and their structures were determined by 1D, 2D NMR, and HR-ESI–MS. The comparisons of experimental and calculated CD curves were used to determine their absolute configurations. The antiproliferative test indicated that both compounds 1-2 displayed moderate biological activity against Hela cells with IC50 values of 17.8 and 20.4 μM, respectively.  相似文献

In our continuing effort to discover more new cytotoxic sesquiterpenoid from Solanum lyratum collected from different districts of China, three new sesquiterpenoids, attributable to eudesmane-type (1, named solajiangxin D) and vetispirane-type (23, named solajiangxins E and 2-hydroxysolajiangxin E), respectively, were isolated. Their structures were elucidated on the basis of integrated spectroscopic techniques, mainly HR-FABMS, 1D and 2D NMR (1H–1H COSY, HMQC, HMBC and ROESY). In vitro, compounds 13 were found to show significant cytotoxicities against three human cancer lines (P-388, HONE-1, and HT-29), and gave ED50 values in the range 2.1–3.7 μg/ml.  相似文献

Two undescribed furanoxanthones, cratocochinones A (1) and B (2), along with eight known xanthones (3–10), were isolated from the bark of Cratoxylum cochinchinense. Their structures were elucidated using spectroscopic methods, mainly 1D and 2D NMR. The isolated compounds were investigated for α-glucosidase inhibitory activity. Seven compounds, 3-6 and 8-10, displayed stronger inhibitory effects than the positive control, acarbose. Especially, cochinxanthone A (6) showed remarkable inhibition with an IC50 value of 59.6 μM, which was approximately 16-fold more potent than acarbose.  相似文献

Three undescribed iridoid glycosides, cyc(7β-O-6′)-morroniside (1), 6′-methyl succinate-7β-O-methylmorroniside (2), and 7β-O-methyl phenyllactate morroniside (3) were isolated from 50% ethanol extract of Cornus officinalis fruits. The structures of the isolated compounds were determined by HRESIMS, 1D NMR, 2D NMR, UV and IR spectroscopic methods. Compounds 1-3 exhibited moderate anti-inflammatory activities in vivo in a CuSO4-induced zebrafish inflammation model (when evaluated at 50 μM).  相似文献

The new alkene lactone, (3E)-5,6-dihydro-5-(hydroxymethyl)-3-docdecylidenefuran-3(4H)-one (1), named majoranolide B, and three alkene lactones known as majorenolide (2), majoranolide (3) and majorynolide (4) were obtained from the aerial parts of Persea fulva (Lauraceae). The structures were elucidated in light of extensive spectroscopic analysis, including 1D, 2D NMR (1H, 13C, 1H-1H-COSY, HMBC and HSQC) and HR-ESI-MS. These compounds were screened for their in vitro antiproliferative activity in rat C6 glioma and astrocyte cells using MTT assay and in silico by molecular docking against targets that play a central role in controlling glioma cell cycle progression. Majoranolide (3) is the most active compound with IC50 6.69 µM against C6 glioma cells, followed by the compounds 1 (IC50 9.06 µM), 2 (IC50 12.04 µM) and 4 (IC50 41.90 µM). The alkene lactones 13 exhibited lower toxicity in non-tumor cells when compared to glioma cells. Molecular docking results showed that majoranolide establishes hydrogen bonds with all targets through its α,β-unsaturated-γ-lactone moiety, whereas the long-chain alkyl group binds by means of several hydrophobic bonds. In the present study, it can be concluded from the anti-proliferative activity of isolates against C6 glioma cells that lactone constituents from P. fulva could have a great potential for the control of C6 glioma cells.  相似文献

One new α-pyrone derivative (S)-3-(4-methoxy-3-methyl-2-oxo-2H-pyran-6-yl)butyl acetate (1) and one new fatty acid derivative (4S,5S)-5-acetoxy-4-hydroxyhexanoic acid (2), along with six known compounds (3–8) were obtained from the mangrove-derived fungus Phomopsis sp. HYP11. Their structures and absolute configurations were determined by detailed 1D and 2D NMR, HR-ESI-MS spectral, and quantum chemical electronic circular dichroism (ECD) calculations. Compounds 2, 3, 5, 6 and 8 showed antioxidant activity with the IC50 values range of 0.09–0.67 mM, especially compounds 5 and 6 showed stronger antioxidant activity than the positive control trolox (IC50 = 0.29 mM). Compound 8 showed weak antagonistic effect on Ralstonia solanacearum with the MIC value of 50 μg/mL.  相似文献

Two new compounds, lasdiplactone (1) and lasdiploic acid (2) and one known compound 3 were isolated from the chloroform extract of cell free filtrate of the endophytic fungus Lasiosdiplodia pseudotheobromae. The structures of new compounds were determined by interplay of spectral techniques (IR, mass, 1H NMR, 13C NMR, DEPT, and 2D NMR). The absolute configuration at C-4 position of 1 was established as S using a process similar to modified Mosher’s method. The absolute configuration of 2 was established by comparing its ECD spectrum with the calculated ECD spectra of all possible isomers. In the in vitro XO inhibition assay, the highest inhibition was exhibited by 3 with an IC50 of 0.38 ± 0.13 μg/ml, followed by 2 with an IC50 of 0.41 ± 0.1 μg/ml and the least in 1. The oxidized form of 1 also showed high XO inhibition with IC50 of 0.35 ± 0.13 μg/ml.  相似文献

Five new lactones, litsenolide F1 (1), lisealactone H1 (10), lisealactone H2 (11), akolactone D (13), and akolactone E (14), along with thirteen known compounds were isolated from the pericarps of Litsea japonica (Thunb.) Jussieu. Their chemical structures were elucidated by extensive spectroscopic analyses, including 1D and 2D NMR, HRMS, and chemical methods. The isolated compounds were evaluated for their inhibitory effects on NO production in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Among them, 2-alkylidene-3-hydroxy-4-methylbutanolide derivatives (compounds 19) exhibited the most potent activity, with IC50 values in the range of 2.9–12.8?μM. In additon, compounds 1, 3, 4, and 6 showed inhibition of iNOS and COX-2 expression in concentration-dependent manner. Compound 3 suppresses mRNA expression of iNOS, COX-2, IL-6, and TNF-α in LPS-stimulated RAW264.7 cells. Based on these evidence, the isolated lactones from L. japonica could be promissing candidates for the development of new anti-inflammatory agents.  相似文献

Phytochemical investigation of the oleo-gum resins from Boswellia papyrifera afforded one new triterpene, named 3α-hydroxyurs-5:19-diene (1) together with twelve known compounds including eight triterpenoids (29), two diterpenoids (10 and 11) and two straight chain alkanes (12 and 13). Similarly ten more known compounds were isolated from the resin of Boswellia sacra including one triterpene (20) and nine boswellic acids (1419 and 2123). Herein the compound 2 was first time reporting from natural source along with complete NMR assignment, while compounds 311 are known, but reported for the first time from the resin of B. papyrifera. The structure elucidation was done by advance spectroscopic 1D and 2D NMR techniques viz., 1H, 13C, DEPT, HSQC, HMBC, and COSY, and NEOSY, ESI-MS and compared with the reported literature. All compounds were evaluated for their α-glucosidase inhibitory activity and as result eight of them 1, 3, 10, 11, 15, and 17–19 were found significantly active against α-glucosidase with an IC50 value ranging from 15.0 ± 0.84 to 80.3 ± 2.33 µM, while 21 exhibited moderate activity with IC50 of 799.9 ± 4.98 µM. Furthermore, two compounds 24 and 25 were synthesised from 16 and 17 to see the effect of carboxyl group in structural-activity relationship (SAR) study. Compounds 24 and 25 retained good α-glucosidase inhibition as compared to 16 and 17, indicating that carboxylic group play a key role in SAR. In addition, the aforementioned activity of all the active compounds was first time reported for their α-glucosidase inhibition potential. The molecular docking studies showed that all the active compounds well accommodate in the active site of the enzyme. Moreover pharmacokinetic properties of the compounds were predicted in silico, suggesting that the compounds possess drug like properties and excellent ADMET profile.  相似文献

In the course of our ongoing search for biologically active compounds from marine-derived fungi, the organic extract of the marine-derived fungus Penicillium vinaceum species was investigated. Seven compounds including a new alkaloid with an unprecedented carbon skeleton, penicillivinacine (1) together with six previously reported ones were isolated. The known compounds were identified as indol-3-carbaldehyde (2), α-cyclopiazonic acid (3), terretrione A (4), brevianamide F (5), cyclo-d-Trp-l-pro (6) and citreoisocoumarin (7). Their structures were established by different spectroscopic data including 1D (1H NMR and 13C NMR) and 2D NMR (COSY, HSQC, HMBC and NOESY) studies as well as high-resolution mass spectral data. The isolated compounds were evaluated for their antimigratory activity against the human breast cancer cell line MDA-MB-231 as well as antimicrobial activities against different pathogens. Compounds 1 and 4 displayed potent antimigratory activities against the highly metastatic triple negative human breast cancer cells MDA-MB-231 with IC50 of 18.4 and 17.7 μM, respectively. Four novel 2-styrylchromones, 4′,5,7-trihydroxy-6-isopentene-2-styrylchromone (1), 4′,5,7-trihydroxy-8-isopentene-2-styrylchromone (2), 4′,5,7-trihydroxy-6-(2-hydroxy-3-methylbut-3-enyl)-2-styrylchromone (3) and 4′,5,7-trihydroxy-8-(2-hydroxy-3-methylbut-3-enyl)-2-styrylchromone (4), were isolated from shed bark of Platanus × acerifolia (Aiton) Willd., as well as four known compounds, 4′,5,7-trihydroxy-2-styrylchromone (5), scutellarein (6), 4′,5,7-trihydroxy-6-prenylflavone (7), and 4′,5,7-trihydroxy-8-prenylflavone (8). The structures of compounds 14 were established by direct interpretation of their spectral data, mainly high resolution electrospray ionization mass spectrometry (HR-ESI-MS), 1D and 2D NMR (1H–1H COSY, HSQC and HMBC). The cytotoxicity of the compounds 18 was evaluated in four human carcinoma cell lines, including HepG2, SMMC-7721, MDA-MB-231, and KB. Compounds 14 exhibited significantly cytotoxic activity toward HepG2 and KB cells, with IC50 values ranging from 3.0 to 9.7 μM.  相似文献

Lam., along with seven known compounds. H2O2-induced neurotoxicity on PC12 cells model were conducted to evaluate the neuro-protective capability of these compounds. New series of benzimidazole ring core conjugated with either dithiocarbamate or thiopropyl linkers, hybridized with different secondary amines were synthesized; 515 and 2231; respectively. The new compounds were characterized by different spectroscopic techniques (1H, 13C 1D & 2D NMR, ESI-MS and IR). They were screened for in vitro anticancer activity against breast cancer using MCF7 cell line. The results obtained revealed that compounds 5, 12, 15 and 25 were the most active among the synthesized series exhibiting IC50 < 10 µg/ml against DOX. To characterize targeting breast cancer on molecular level, binding to 15N-labeled Pin1 enzyme was conducted using state-of-the-art 2D NMR binding experiments. Results showed promising binding between compounds 5, 12, and 25 by chemical shift perturbation (peak shifting or peak disappearance). Molecular docking study were quite valuable to explain the binding mode of active derivatives via hydrogen bonding. Additional contact preferences and surface mapping studies stated the similarity pattern between active candidates which may pave the way for more precise anti breast cancer target optimization.  相似文献

Fifteen compounds, including five new phorbol esters (1-5) and ten known metabolites were isolated from the leaves of Croton tiglium. The structures of new compounds 1-5 were determined by comprehensive analysis of the HRESIMS, IR, 1D and 2D NMR spectral data. The isolates were assayed for their larvicidal and α-glucosidase inhibitory activity; results suggested the new compounds 1-4 possessed significant insecticidal activity, inhibiting the Plutella xylostella with LC50 values ranging from 0.081 to 0.114 μg/mL, while the sesquiterpenoids 11 and 15 showed noticeable α-glucosidase inhibitory activity with IC50 values of 11.27 and 8.13 μM, respectively.  相似文献

Bio-assay guided fractionation of the methanolic extract of Aloe vera resin and Lycium shawii stem successively afforded twenty three compounds; fourteen (114) from A. vera and nine (1523) from L. shawii. All these compounds were characterized by 1D and 2D NMR spectroscopic techniques viz., 1H, 13C, DEPT, HSQC, HMBC, and COSY, and NEOSY, ESI-MS and compared with the reported literature. These compounds were assessed for their potential as urease inhibitors targeted in peptic ulcer. Among crude extracts and fractions of A. vera resin, n-butanol fraction (23.5 ± 1.7 μg·mL−1) showed the most potent urease inhibition followed by methanol (30.9 ± 0.3 μg/mL) and ethyl acetate (31.7 ± 0.5 μg·mL−1). In case of L. shawii, ethyl acetate fraction exhibited the highest urease activity (41.0 ± 1.4 μg/mL) trailed by dichloromethane (55.2 ± 1.5 μg/mL) fraction. Among the isolates, compounds 7, 11 and 23 were found to be excellent urease inhibitors with IC50 values of 14.5 ± 0.90 µM, (16.7 ± 0.16 µM) and 14.0 ± 0.8 µM, respectively. To the best of our knowledge, this is the first report on the urease enzyme inhibitory activity of the said compounds excluding compound 18. In addition, the urease activity of different fractions of L. shawii stem was also reported for the first time. The molecular docking studies showed that all the active compounds well accommodate in the active site of the urease enzyme by interacting with key amino acids.  相似文献
