与拟除虫菊酯抗性相关的烟粉虱钠通道基因突变及其检测   总被引:12,自引:2,他引:12

通过RT-PCR克隆了烟粉虱Bemisia tabaci (Gennadius) 南京种群(B-生物型)的钠离子通道结构域ⅡS4-6 cDNA片段,证实了与拟除虫菊酯抗性相关的是位于第925位亮氨酸到异亮氨酸的突变(L925I),并建立了L925I突变的PASA检测技术。与SUD-S敏感品系相比,2002年采自南京棉花上的烟粉虱种群对氯氰菊酯具有77倍的抗性,用氯氰菊酯对该种群进行多次筛选后,该种群对氯氰菊酯的抗药性提高到227倍。PASA检测结果表明筛选后的南京种群中100%个体都具有L925I突变(61.1%的个体为L925I突变纯合子,38.9%的个体为杂合子),而未筛选的南京种群只有75%个体具有L925I突变(35%个体为L925I突变纯合子,40%的个体为杂合子,25%的个体为野生型)。该结果表明了烟粉虱钠离子通道L925I突变与对拟除虫菊酯抗性密切相关。还讨论了烟粉虱对拟除虫菊酯抗性的代谢机理。  相似文献

温带臭虫Cimex lectularius L.是一种重要的卫生害虫,严重影响人们的生活和健康。目前有关该虫的防治措施主要以喷洒化学杀虫剂为主,而杀虫剂的使用不当极易导致该虫抗药性的产生或进一步加重,还将对人身安全构成极大的威胁。因此,探索高效、安全的防治方法成为当前温带臭虫防治过程中亟待解决的问题。本研究在实验室条件下采用行为观察法评估了高分子聚合物-聚四氟乙烯对温带臭虫垂直和水平爬行能力的影响。结果表明,随着聚四氟乙烯涂层宽度的增加,温带臭虫的垂直攀爬能力逐渐降低,当涂层宽度为6 cm时可100%阻隔温带臭虫垂直攀爬,且涂层的持效期可达112 d以上。而聚四氟乙烯涂层无法阻止温带臭虫的水平扩散。研究结果可为温带臭虫的高效、安全物理预防提供理论依据和技术支持。  相似文献

为明确使用新烟碱类杀虫剂烯啶虫胺、噻虫胺和噻虫嗪防治入侵害虫西花蓟马的抗性风险及抗性稳定性,本研究采用芸豆浸药法对西花蓟马敏感种群初羽化雌成虫进行连续筛选获得抗性种群,根据抗性现实遗传力计算公式分析西花蓟马对上述3种杀虫剂的抗性风险,预测其抗性发展速度,并测定抗性稳定性。结果表明: 经过30代抗性筛选,西花蓟马对烯啶虫胺、噻虫胺和噻虫嗪均达到高水平抗性(44.7、45.5和32.7倍)。西花蓟马对噻虫胺、烯啶虫胺和噻虫嗪的抗性发展速度依次降低,抗性现实遗传力分别为0.1503、0.1336和0.1258。对抗性种群在无选择压力下继续饲养10代,西花蓟马对烯啶虫胺、噻虫胺和噻虫嗪的抗性水平均出现一定程度的下降,但均未能恢复到敏感性水平。抗性选育后,西花蓟马若虫与成虫对杀虫剂的敏感性差异显著缩小,西花蓟马敏感种群及抗性种群若虫对上述3种杀虫剂的敏感性显著高于成虫。西花蓟马对烯啶虫胺、噻虫胺和噻虫嗪均存在高抗风险,噻虫嗪的抗性上升速度较慢且抗性稳定性最低。因此,在西花蓟马若虫期使用噻虫嗪有利于西花蓟马防治。  相似文献

采用稻茎浸渍法,测定了广州市本地褐飞虱种群对噻虫嗪和烯啶虫胺的室内毒力,评估褐飞虱对其的抗性风险。结果表明:广州本地褐飞虱种群对噻虫嗪和烯啶虫胺的LC50分别为02857mg/L和05022 mg/L,分别是敏感品系LC50的267倍和106倍,仍属敏感水平。室内抗性筛选结果表明:经过30代的连续筛选后,噻虫嗪的抗性上升82980倍,达到极高抗性水平,烯啶虫胺的抗性上升3170倍,达到中等抗性水平,表明褐飞虱对噻虫嗪和烯啶虫胺存在抗性风险的可能。根据试验结果,对褐飞虱噻虫嗪和烯啶虫胺抗性的预防治理提出了应用策略。  相似文献

噻虫胺是具有内吸和触杀等多种作用方式的新烟碱类杀虫剂,常用于防治入侵害虫西花蓟马。为明确抗性风险,本文研究了西花蓟马抗噻虫胺种群对多种杀虫剂的交互抗性及其机制。经过45代筛选,西花蓟马对噻虫胺产生了高水平抗性(56.8倍)。生物测定结果表明: 西花蓟马高抗噻虫胺种群与噻虫嗪、吡虫啉、毒死蜱、三氟氯氰菊酯、甲维盐存在中等水平交互抗性(18.6>RR50>11.3),对辛硫磷及灭多威具有低水平交互抗性,与溴虫腈和多杀菌素不存在交互抗性。胡椒基丁醚(PBO)与磷酸三苯酯(TPP)对杀灭西花蓟马抗噻虫胺种群(CL)、云南田间种群(YN)和敏感种群(S)均有显著增效作用。西花蓟马抗噻虫胺种群细胞色素P450含量(3.6倍)、细胞色素b5含量(2.9倍)及O-脱甲基酶活性(4.9倍)和羧酸酯酶活性(2.5倍)均显著高于敏感种群,表明多功能氧化酶及羧酸酯酶活性增强是西花蓟马对噻虫胺产生抗性的重要机制。  相似文献

应用成虫浸叶生测法研究了江苏省扬州、无锡和东台3个地区Q型烟粉虱Bemisia tabaci(Gennadius)田间种群对5种杀虫剂的抗药性。结果表明,与相对敏感种群相比,Q型烟粉虱田间种群已对氯氰菊酯和吡虫啉产生了低到中等水平抗性,对阿维菌素仍然表现敏感。进一步通过RCR扩增获得了长度分别为287bp和184bp的烟粉虱乙酰胆碱酯酶ace1和para-同源钠离子通道基因片断。序列分析表明,江苏Q型烟粉虱存在与有机磷抗性相关的乙酰胆碱酯酶F331W突变和与拟除虫菊酯抗性相关的钠离子通道L925I和T929V突变。  相似文献

为明确河北省推广种植植转Bt基因抗虫棉(简称Bt棉)后, 棉铃虫Helicoverpa armigera (Hübner)对常用杀虫剂的抗药性水平及其生化机理, 2011-2012年采用点滴法对保定南郊、 沧州南皮、 邢台巨鹿3个地区的田间种群以及敏感种群进行了室内毒力测定, 并采用生化分析法对4个种群相关的羧酸酯酶(carboxylesterase, CarE)、 谷胱甘肽S 转移酶(glutathione S-transferases, GSTs)和乙酰胆碱酯酶(acetylcholinesterase, AChE)的活性进行了研究。结果表明: 3个田间种群对高效氯氰菊酯和氰戊菊酯处于中至高抗水平, 抗性倍数为20.02~73.70倍; 对灭多威处于低至中抗水平, 抗性倍数为6.27~11.84倍; 对高效氯氟氰菊酯(抗性倍数: 1.07~4.20倍), 辛硫磷、 毒死蜱和马拉硫磷(抗性倍数: 1.00~2.69倍), 以及氯虫苯甲酰胺(抗性倍数: 2.00~3.67倍)均处于敏感水平。3个田间种群的CarE, GSTs和AChE活性分别是敏感种群的1.06~1.23, 1.20~1.63和1.15~1.23倍, 这可能与其对高效氯氰菊酯、 氰戊菊酯和灭多威产生的抗性有关。  相似文献

室内采用浸叶法测定了柑橘木虱Diaphorina citri敏感种群与田间种群对广东常用药剂的敏感性,明确了柑橘木虱田间种群对阿维菌素、高效氯氰菊酯、吡虫啉、噻虫嗪、啶虫脒和甲维盐6种药剂的抗药性.柑橘木虱田间种群对70%吡虫啉水分散粒剂、20%啶虫脒可溶液剂、3%甲维盐微乳剂处于低水平抗药性,LC50抗性倍数分别为5.89倍、8.53倍和9.86倍;对25%噻虫嗪水分散粒剂、10%高效氯氰菊酯乳油、1.8%阿维菌素乳油仍具有较高的敏感性.田间防治柑橘木虱推荐不同作用机理的药剂交替使用,综合运用多种防控措施,以延缓柑橘木虱抗药性的产生.  相似文献

本研究明确了kdr突变和解毒代谢在B型烟粉虱Bemisia tabaci对高效氯氰菊酯抗性中的作用。B型烟粉虱NJ品系相对于烟粉虱敏感品系(SUD-S,非B型)对高效氯氰菊酯有266倍的抗性。对NJ品系用高效氯氰菊酯进行群体筛选获得抗性为811倍的NJ-R1品系,对NJ品系进行单对交配筛选获得抗性达2 634倍的NJ-R2品系。在NJ,NJ-R1和NJ-R2品系间,酯酶、多功能氧化酶和谷胱甘肽S-转移酶活性无显著差异,说明在筛选过程中解毒代谢没有发生变化。PASA检测结果表明,NJ-R2品系钠离子通道基因L925I突变(kdr突变)频率为100%,NJ-R1品系为80.6%,NJ品系为55%。由此可见,kdr突变频率的增加是B型烟粉虱种群对高效氯氰菊酯抗性上升的主要原因。在NJ,NJ-R1和NJ-R2品系中,增效醚(PBO)对高效氯氰菊酯的增效作用均为20倍左右,而PBO对SUD-S品系没有任何增效作用。PBO能同时抑制烟粉虱的多功能氧化酶和酯酶,通过与TPP增效作用进行对比表明,在B型烟粉虱中PBO所产生的增效作用主要来源于对酯酶的抑制。因此,B型烟粉虱品系(NJ-R2,NJ-R1和NJ)与非B型SUD-S品系相比存在20倍左右的先天抗性,该先天抗性主要与B型烟粉虱的特有酯酶有关。在B型烟粉虱品系对高效氯氰菊酯的抗性中,抗性水平完全由kdr突变频率高低所决定。  相似文献

几类杀虫剂对灰飞虱的相对毒力及田间种群的抗药性现状   总被引:8,自引:0,他引:8  
采用浸苗法测定了6类11种杀虫剂对灰飞虱Laodelphax striatellus (Fallén)3龄若虫的毒力,并分析比较了江苏句容、通州、楚州、大丰、南京和苏州以及安徽庐江等7地灰飞虱种群对10种杀虫剂的抗药性水平。对云南种群而言,在所测杀虫剂中,以乙酰甲胺磷为标准药剂,氟虫腈的相对毒力最高;噻嗪酮、阿维菌素和噻虫嗪次之;高效氯氰菊酯、IPP(硝基亚甲基类化合物)、毒死蜱、敌敌畏和三唑磷间毒力处于同一数量级,低于阿维菌素等杀虫剂;吡虫啉的相对毒力最低。与云南种群相比,2007年采自苏、皖7个不同地区的灰飞虱种群对噻嗪酮产生了极高水平的抗性,其抗性倍数均超过200倍;对高效氯氰菊酯产生了中高水平的抗性,其抗性倍数为7.8~108.8;安徽庐江灰飞虱种群对三唑磷产生了7.7倍的抗性,对毒死蜱产生了12.0倍的抗性,江苏楚州、南京、大丰和句容灰飞虱种群对毒死蜱产生了5.7~12.6倍的抗性; 所有灰飞虱种群对敌敌畏仍然敏感,对氟虫腈、阿维菌素和新烟碱类杀虫剂吡虫啉、噻虫嗪和IPP等也比较敏感。  相似文献

Of 479 eggs laid by female Cimex lectularius Linnaeus and C.hemipterus (Fabricius) mated by males of the other species, one was fertile and gave rise to a first stage nymph. The egg showed sculpturing typical of C.hemipterus, the female parent, and the nymph conformed to the narrow pronotum and abdomen of this species, being significantly different from C.lectularius in the width of the abdomen. Because the orientation and lengths of the bristles on the sides of the abdomen were distinctly different from C.hemipterus and closely resembled C.lectularius, the single nymph obtained from the cross C.hemipterus x lectularius was interpreted as being a hybrid rather than a product of parthenogenesis.  相似文献

Interspecific mating between male Cimex hemipterus and female C. lectularius bedbugs (Hemiptera: Cimicidae) occurs freely in the laboratory, even when many female C. hemipterus are available. Longevity and fertile egg production of female C. lectularius are reduced by mating with male C. hemipterus. In KwaZulu, South Africa, a correlation exists between the percentage of C. lectularius females which are interspecifically mated and the predominance of C. hemipterus in mixed domestic infestations of these bedbugs. When C. hemipterus constitutes over 75% of the infestation, the majority of C. lectularius females lay only sterile eggs.  相似文献

The interspecific cross-mating of female Cimex lectularius Linnaeus with male Cimex hemipterus (Fabricius) results in a visible mark developing in the female ectospermalege. This mark has been used to record the frequency of mating between female C.lectularius and male C.hemipterus in sympatric wild populations in a DDT-sprayed area of northern KwaZulu, South Africa. A high percentage (11/16 = 69%) of C.lectularius females were marked, showing that these two species mate in nature.  相似文献

When Tanzanian villages were provided with pyrethroid-treated bednets, bedbugs (Cimicidae) disappeared; however, after about 6 years they have re-appeared in these villages. Using a newly devised test-kit, susceptibility tests of bedbugs Cimex hemipterus (Fabricius) from five of these villages showed that there is resistance to permethrin and alphacypermethrin in bedbugs from each of the villages, in contrast to those from five villages without treated nets. Circumstantial evidence indicates that bedbug resistance to pyrethroid insecticides may evolve more readily in villages with incomplete coverage rates of treated bednets, allowing bedbug infestations to become re-established. Bedbugs have not returned to a village where nearly all the beds have been provided with pyrethroid-treated bednets for 14 years.  相似文献

Accurate and timely surveillance of bed bug infestations is critical for the development of effective control strategies. Although the bed bug‐produced volatiles (E)‐2‐hexenal and (E)‐2‐octenal are considered as defensive secretions, the present study demonstrates, using ethovision® video‐tracking software (Noldus Information Technology Inc., Leesburg, Virginia), that low amounts of these commercially‐obtained aldehydes function as attractants, and high amounts function as local repellents, against the common bed bug Cimex lectularius L. In behavioural assays, both males and female C. lectularius are attracted to 0.04 µg of an aldehyde blend (1 : 1) for up to 1 h after initial treatment of filter paper disks. Males differ from females in their response to higher amounts of aldehydes, with females and males exhibiting maximum local repellency at 40 µg and 400 µg, respectively. The results suggest that these bed bug secretions may be candidates for lures and monitors.  相似文献

Abstract. Adult bedbugs, Cimex lectularius, were exposed for 24 h (25oC) to filter paper treated with various dilutions of the technical grade of nine insecticides dissolved in acetone to determine the concentration-response relationships. The order of toxicity, from most to least based on the LC50's was: dichlorvos, pirimiphos methyl, lambda-cyhalothrin, bendiocarb, permethrin, malathion, carbaryl, tetrachlorvinphos, and fenvalerate. The residual toxicities of commercial formulations of six of the chemicals diluted with water and applied to wood, cardboard, cloth and galvanized metal, were determined by exposing adult bedbugs at 3,7 and 12 weeks after treatment. The formulation of bendiocarb (FICAM® 76% W) had little residual activity on all surfaces at 12 weeks after treatment. The formulation of carbaryl (SEVIN® 21.5% L) was toxic to bedbugs on all surfaces at 12 weeks after treatment, but required high concentrations on wood, cardboard, and cloth. The formulation of pirimiphos methyl (ACTELLIC® 57% EC) had no residual activity on any of the surfaces at 12 weeks after treatment. The formulation of tetrachlorovinphos (RABON® 50% W) had residual activity for 12 weeks on all surfaces except metal. The formulation of permethrin (ATROBAN® 11% EC) had residual activity on only metal and wood while the formulation of lambda-cyhalothrin (KARATE® 13.1% EC) had residual activity 12 weeks on all surfaces.  相似文献

The common bedbug, Cimex lectularius L. (Hemiptera: Cimicidae), is a globally re-emerging pest that is playing an increasing role in legal disputes and compensation claims as a result of its unpleasant feeding activity. However, there is little information about the feeding frequency of bedbugs outside controlled laboratory cultures. Here, we present a simple method of estimating the average time since the last bloodmeal of individual female bedbugs in a single sampling event, applicable to a single bedbug harbourage or an entire room. Using the temperature-dependent rate of decrease in the abdomen size of the bedbug after a bloodmeal, we found that, in a highly infested room kept at a constant temperature of 26°C, females fed every 2.5 days on average. Our method corrects for variations in body size across different populations and determines the shrinkage that occurs when individuals are preserved in ethanol. This method should, therefore, be widely applicable. It is cheap, rapid and, if coupled with information on the total number of bedbugs present in a room, allows for the estimation of the minimum number of times persons lodging in a room have been bitten by bedbugs. This method can also be used to calculate the feeding rate of other blood-sucking insects on their hosts. The sex ratio in the infestation was female-biased. Finally, our case study suggests that individual female bedbugs within a harbourage do not seem to feed at a regular rate, but tend to synchronize feeding patterns.  相似文献
