Natural product's properties are related to certain classes of compounds such as alkaloids, flavonoids, essential oils and others. Traditionally, separation techniques including thin layer chromatography (TLC), liquid chromatography (LC), gas chromatography (GC) and capillary electrophoresis (CE) even hyphenated to mass spectrometry (MS) were used for the elucidation, qualitative and quantitative analysis of individual compounds.In food industry, spectroscopic investigations using infrared radiation have been used to monitor and evaluate the composition and quality already since the early sixties. During the last four decades near-infrared spectroscopy (NIR; 800–2500 nm; 12,500–4000 cm−1) has become one of the most attractive and used methods for analysis for the following reasons: it represents a non-invasive analytical tool allowing a fast and simultaneous qualitative and quantitative characterization of natural products and their constituents. Additionally, the development of custom-made hand-held instruments enables in-field measurement for determining the optimum harvest time.Attenuated total reflection (ATR) and Fourier transform infrared (FTIR) spectroscopic imaging are suitable not only for the differentiation of different plant species, but also to distinct various ingredients within a plant. FTIR spectroscopic microscopy enables molecular imaging of complex botanical samples and therefore the detection and characterization of the molecular components of biological tissue.In the present contribution, the principle, technique and methodology of the different infrared spectroscopic methods are described followed by a discussion of quantitative and qualitative application possibilities in the field of natural product analysis.  相似文献

Functional indicators are being increasingly used to assess waterway health but their responses to pressure in non-wadeable rivers have not been widely documented or applied in modern survey designs that provide unbiased estimates of extent. This study tests the response of river metabolism and loss in cotton strip tensile strength across a land use pressure gradient in non-wadeable rivers of northern New Zealand, and reports extent estimates for river metabolism and decomposition rates. Following adjustment for probability of selection, ecosystem respiration (ER) and gross primary production (GPP) for the target population of order 5–7 non-wadeable rivers averaged −7.3 and 4.8 g O2 m−2 d−1, respectively, with average P/R < 1 indicating dominance by heterotrophic processes. Ecosystem respiration was <−3.3 g O2 m−2 d−1 for 75% of non-wadeable river length with around 20% of length between −10 and −20 g O2 m−2 d−1. Cumulative distribution functions of cotton strength loss estimates indicated a more-or-less linear relationship with river km reflecting an even spread of decay rates (range in k 0.0007–0.2875 d−1) across non-wadeable rivers regionally. A non-linear relationship with land cover was detected for GPP which was typically <5 g O2 m−2 d−1 where natural vegetation cover was below 20% and greater than 80% of upstream catchment area. For cotton strength loss, the relationship with land cover was wedge-shaped such that sites with >60% natural cover had low decay rates (<0.02 d−1) with variability below this increasing as natural cover declined. Using published criteria for assessing waterway health based on ER and GPP, 232–298 km (20–29%) of non-wadeable river length was considered to have severely impaired ecosystem functioning, and 436–530 km (42–50%) had no evidence of impact on river metabolism.  相似文献

The phosphorus (P) fractions and bioavailable P in the sediments from the Quanzhou Bay Estuarine Wetland Nature Reserve were investigated using chemical extraction methods for the first time to study the distribution and bioavailability of P in the reserve sediments. A hypothesis was presented suggesting that the bioavailable P in the sediments could be evaluated using the P fractions. The total phosphorus (TP), inorganic phosphorus (IP), organic phosphorus (OP), non-apatite phosphorus (NAIP), and apatite phosphorus (AP) contents in the sediments were in the ranges of 303.87–761.59 mg kg−1, 201.22–577.66 mg kg−1, 75.83–179.16 mg kg−1, 28.86–277.90 mg kg−1, and 127.36–289.94 mg kg−1, respectively. The water soluble phosphorus (WSP), readily desorbable phosphorus (RDP), algal available phosphorus (AAP), and NaHCO3 extractable phosphorus (Olsen-P) contents in the sediments were in the ranges of 0.58–357.17 mg kg−1, 80.77–586.75 mg kg−1, 1.09–24.12 mg kg−1, and 54.96–676.82 mg kg−1, respectively. The correlation analysis results showed that the NAIP was the major component of the bioavailable P and that the impact of the AP on the bioavailable phosphorus may be minimal. Due to the low TP content in the sediments of the Quanzhou Bay Estuarine Wetland Nature Reserve, the potential pollution risks of P in the sediments may not be very high. The results also show that the bioavailable P concentrations in the sediments of the Quanzhou Bay Estuarine Wetland Nature Reserve could not be evaluated by measuring the P fractions and that the hypothesis was untenable.  相似文献

Pure glycerol and glycerol-rich product (GRP) obtained from the biodiesel industries were used as carbon source for the production of a new extracellular polysaccharide (EPS) by Pseudomonas oleovorans NRRL B-14682. The influence of temperature (20–40 °C) and pH (6.0–8.0) was studied. A temperature of 30 °C and pH control at 6.8 gave the maximum cell growth and EPS production. The culture attained a maximum cell dry weight (CDW) of 9.55 g l−1 and an EPS concentration of 11.82 g l−1 when cultivated with pure glycerol. GRP was a suitable carbon source, as shown by the slightly higher EPS concentration (12.18 g l−1). The EPS productivity obtained with GRP (3.85 g l−1 d−1) was almost twice that obtained with pure glycerol (2.00 g l−1 d−1). Also, the yield on glycerol was higher for the cultivation with GRP (0.36 g g−1) than for pure glycerol (0.28 g g−1). The EPS was a high molecular weight heteropolysaccharide, composed by neutral sugars (37–80 wt% galactose, 2–30 wt% glucose, 0.5–25 wt% mannose and 0.5–20 wt% rhamnose) and containing acyl group substituents (pyruvil, acetyl and succinyl were identified). The EPS forms highly viscous aqueous dispersions with many potential commercial applications.  相似文献

Two experiments were undertaken to adapt the in vitro gas production technique in syringes, used for ruminants, to fibre fermentation studies in the large intestine of pigs.In a first experiment, two inocula (faeces and large intestine content) were compared at four dilution levels in a buffer solution (0.025, 0.05, 0.1 and 0.2 g ml−1) with two substrates: wheat bran and sugar–beet pulp. The accumulated gas produced over 72 h was modelled and the kinetics parameters compared. The time to half asymptote was lower for the intestinal inoculum (5.5 versus 8.0 h, P<0.02), but the 2 inocula yielded similar fractional rates of degradation (0.16 h−1) and gave equal final gas production (252 ml g−1 substrate). No interaction (P>0.05) was observed between inocula and substrates. The dilution of the samples in the buffer solution increased (P<0.001) the lag time (from 0.9 to 2.1 h for dilution rates ranging from 0.2 to 0.025 g ml−1, respectively) and decreased (P<0.001) the rates of substrate degradation (from 0.18 to 0.13 h−1).A second experiment aimed to study the effect of an in vitro pepsin–pancreatin hydrolysis of the sample prior to the gas test. Six substrates were tested: maize, wheat bran, sugar–beet pulp, lupins, peas and soybean meal. The enzymatic hydrolysis affected (P<0.001) the kinetics parameters and the ranking order of the fermented substrates. The lag times also increased for all ingredients. The rate of degradation decreased when peas, lupins, maize and wheat bran were hydrolysed (P<0.001) but it increased with soybean meal (P=0.014) and sugar–beet pulp (P<0.001). Final gas production increased with peas and soybean meal (P<0.001), remained unchanged for lupins and decreased for the other substrates (P<0.001).In conclusion, the method using faeces as a source of microbial inoculum is reliable to characterise the fermentation kinetics of ingredients in the large intestine of pigs. However, it is important to hydrolyse the substrates with pepsin and pancreatin before the gas tests.  相似文献

The effect of nutrient addition on the growth of E. najas was evaluated in a dose response experiment using sand amended with phosphorus (P) and nitrogen (N), and in enrichment trials with N and P amendments to natural sediments. Plants, water and sediment came from lagoons of the Upper Paraná River Floodplain and from Itaipu Reservoir (Brazil). Relative growth rates (RGRs) of E. najas shoots, based on dry mass (DM), varied from 0.03 to 0.060 d−1 for both nutrients. Root:shoot biomass ratios were related to sediment exchangeable P (r = −0.419; P = 0.03) and N (r = −0.54; P = 0.006), however root RGR was not related to sediment nutrient concentrations. When natural sediments were amended with N and P, neither shoot nor root RGRs differed among treatments for substrata from either the reservoir or the floodplain lagoons (P > 0.05). Comparison of nutrient concentrations measured in natural sediments collected from several sites in both the Upper Paraná River Floodplain (range 49–213 μg P g−1 DM; 36–373 μg N g−1 DM) and Itaipu Reservoir (range 43–402 μg P g−1 DM; 7.9–238 μg N g−1 DM) showed that sediment N and P from these systems usually exceeded minimum requirements necessary for E. najas growth, as measured in the dose response experiment. Together, these results indicate that E. najas, at least in early stages of development, responds to sediment nutrient amendments and relies upon bottom sediments to meet its N and P requirements and that for at least two Brazilian ecosystems, growth of this species is not limited by insufficient sediment N or P. Thus, reducing N and P in water is not enough to control E. najas growth in short time periods in these ecosystems.  相似文献

Experiments were conducted to determine the optimum requirements of non-phytin phosphorus (NPP) in commercial broilers and White Leghorn layers. Five levels of NPP (2.5, 3.0, 3.5, 4.0 and 4.5 g kg−1 diet) were tested to assess the NPP requirement of commercial broilers (3–30 days of age) fed maize–soya diets containing 10 g Ca kg−1. Each level of NPP was fed to quadruplicate groups of ten chicks each. Inclusion of graded levels of NPP significantly (P < 0.01) influenced body weight gain, feed intake, tibia ash content, phosphorus content in serum, tibia ash and phosphorus retention. The predicted NPP requirements for body weight gain, P content in serum and tibia ash were 4.4, 4.48 and 4.1 g kg−1 diet, respectively. The NPP requirement for tibia ash was the highest (7.4 g kg−1 diet). Similarly, four levels of NPP (2.0, 2.5, 3.0 and 3.5 g kg−1 diet) were tested with maize–soya diets containing 35 g Ca kg−1 for White Leghorn layers (266–350 days of age). Each diet was tested on four groups of 12 hens in each. Egg production was not influenced by the variation in dietary NPP levels. The predicted NPP requirements for better egg weight and shell thickness were 2.6 and 2.4 g kg−1 diet, respectively, while for the serum inorganic P level the value was 3.42 g kg−1 diet. Therefore, it can be concluded that commercial broilers need about 4.4 g NPP kg−1 diet for better performance, whereas, White Leghorn layers need not more than 2.0 g NPP kg−1 diet for better egg production. However, layers require 2.6 g NPP kg−1 diet to produce eggs with better egg size and shell quality.  相似文献

Pseudomonas luteola was immobilized by entrapment in alginate–silicate sol–gel beads for decolorization of the azo dye, Reactive Red 22. The influences of biomass loading and operating conditions on specific decolorization rate and dye removal efficiency were studied in details. The immobilized cells were found to be less sensitive to changes in agitation rates (dissolved oxygen levels) and pH values. Michaelis–Menten kinetics could be used to describe the decolorization kinetics with the kinetic parameters being 36.5 mg g−1 h−1, 300.1 mg l−1 and 18.2 mg g−1 h−1, 449.8 mg l−1 for free and immobilized cells, respectively. After five repeated batch cycles, the decolorization rate of the free cells decreased by nearly 54%, while immobilized cells still retained 82% of their original activity. The immobilized cells exhibited better thermal stability during storage and reaction when compared with free cells. From SEM observation, a dense silicate gel layer was found to surround the macroporous alginate–silicate core, which resulted in much improved mechanical stability over that of alginate beads when tested under shaking conditions. Alginate–silicate matrices appeared to be the best matrix for immobilization of P. luteola in decolorization of Reactive Red 22 when compared with previous results using synthetic or natural polymer matrices.  相似文献

In the present study, interactions of Au(III) and Ga(III) ions on human serum albumin (HSA) were studied comparatively via spectroscopic and thermal analysis methods: UV–vis absorbance spectroscopy, fluorescence spectroscopy, Fourier transform infrared (FT-IR) spectroscopy and isothermal titration calorimetry (ITC). The potential antitumor effects of these ions were studied on MCF-7 cells via Alamar blue assay. It was found that both Au(III) and Ga(III) ions can interact with HSA, however; Au(III) ions interact with HSA more favorably and with a higher affinity. FT-IR second derivative analysis results demonstrated that, high concentrations of both metal ions led to a considerable decrease in the α-helix content of HSA; while Au(III) led to around 5% of decrease in the α-helix content at 200 μM, it was around 1% for Ga(III) at the same concentration. Calorimetric analysis gave the binding kinetics of metal–HSA interactions; while the binding affinity (Ka) of Au(III)–HSA binding was around 3.87 × 105 M−1, it was around 9.68 × 103 M−1 for Ga(III)–HSA binding. Spectroscopy studies overall suggest that both metal ions have significant effects on the chemical structure of HSA, including the secondary structure alterations. Antitumor activity studies on MCF7 tumor cell line with both metal ions revealed that, Au(III) ions have a higher antiproliferative activity compared to Ga(III) ions.  相似文献

A study was conducted on H2S removal in a biotrickling filter packed with open-pore polyurethane foam. Thiobacillus denitrificans was used as inoculum and a mixed culture population was developed during the process. The inhibitory effect of sulphate concentration (1.8–16.8 g L−1), pH (6.9–8.6), trickling liquid velocity (TLV, 9.1–22.8 m h−1), H2S inlet concentration (20–157 ppmv) and the empty bed residence time (EBRT, 9–57 s) on the H2S removal efficiency (RE) were thoroughly investigated. An increase in pH from 6.9 to 8.5 led to a corresponding increase in H2S removal. In addition, an inhibitory effect of sulphate concentration was observed from 16.8 g L−1 and the maximum elimination capacity was found to be 22 gS m−3 h−1 (RE 98%). The RE was constant (98.8 ± 0.30%) for EBRT  16 s, but a decrease in the EBRT from 16 to 9 s led to a corresponding decrease in RE from 98.2 to 89.6% for a TLV of 9.1 m h−1 and from 97.9 to 94.9% for a TLV of 22.8 m h−1 (inlet load of 11.0 ± 0.2 gS m−3 h−1). The sulphur oxidation capacity in the biotrickling filter was not diminished by the presence of other bacteria.  相似文献

Effects of some precursors on phenylethanoid glycosides (PeGs) accumulation in Cistanche salsa cell suspension cultures were investigated. Precursors such as tyrosine, phenylalanine, caffeic acid and cucumber juice at proper concentrations could increase the total accumulation of PeGs (echinacoside, acteoside, 2′-acetylacteoside) by 50%, 12%, 12% and 23%, respectively. Under the combined feeding of precursors at proper concentrations, the total production of PeGs in bio-staged culture reached the highest amount of 1358.1 mg l−1 (640.8 mg echinacoside l−1, 689.4 mg acteoside l−1 and 54.9 mg 2′-acetylacteoside l−1), which was about two-fold of that in the control. This study showed promise for obtaining large-scale production of active ingredients in plant cells by the solid–liquid two step culture (SLTSC) technique and also provided for the first time an example for producing PeGs by C. salsa cell culture. The improved production of PeGs was higher than that in previous reports on PeG production by Cistanche deserticola cell culture fed with precursors.  相似文献

Anthropogenic impact on CO2 levels was studied in the Bear Chamber of the Výpustek Cave, a show cave in the Moravian Karst (Czech Republic), during a period of active ventilation and enhanced attendance. The study showed that the natural CO2 levels were controlled by (i) the natural CO2 influxes from soils/epikarst (up to ∼5.64 × 10−2 mol s−1); and, (ii) the advective CO2 fluxes out of cave atmosphere (up to 4.66 × 10−2 mol s−1). During visitor presence, the anthropogenic CO2 flux into the chamber reached up to ∼0.13 mol s−1 and exceeded all other CO2 fluxes. The reachable anthropogenic steady states at sufficient duration of stay (up to 2.65 × 10−1 mol m−3) could exceed the natural CO2 levels by factor of more than nine based on the number of visitors. Recession analysis of anthropogenic pulses showed that intervals between individual visitor groups would have to be up to ∼6 h long if the cave environment has to return to natural conditions. As such pauses between individual tours are hardly realizable, a risk analysis was conducted to find the consequences of breaking natural conditions. It showed that the condition under which dripwater becomes aggressive to calcite (i.e., the point when PCO2 in cave atmosphere exceeds the hypothetical CO2 concentrations in epikarst that has participated on the water formation, PCO2(H) = 10−1.56) is potentially reachable under extreme conditions only (enormous visitor stay period and visitor number). In case of condensed water, however, any increase in CO2 concentration will cause an increase of water aggressiveness to calcite. Therefore, in the periods and sites of enhanced condensation, it is important to strive for preservation of natural conditions.  相似文献

The aim of this study was to develop a bioprocess for l- and d-lactic acid production from raw sweet potato through simultaneous saccharification and fermentation by Lactobacillus paracasei and Lactobacillus coryniformis, respectively. The effects of enzyme and nitrogen source concentrations as well as of the ratio of raw material to medium were investigated. At dried material concentrations of 136.36–219.51 g L−1, yields of 90.13–91.17% (w/w) and productivities of 3.41–3.83 g L−1 h−1 were obtained with lactic acid concentrations as high as 198.32 g L−1 for l-lactic acid production. In addition, d-lactic acid was produced with yields of 90.11–84.92% (w/w) and productivities of 2.55–3.11 g L−1 h−1 with a maximum concentration of 186.40 g L−1 at the same concentrations of dried material. The simple and efficient process described in this study will benefit the tuber and root-based lactic acid industries without requiring alterations in plant equipment.  相似文献

Anaerobic bioenergy production processes including fermentative biohydrogen (BioH2), anaerobic digestion (AD) and bioelectrochemical system have been investigated for converting municipal waste or various biomass feedstock to useful energy carriers. However, the performance of a microbial fuel cell (MFC) fed on the effluent from a two-stage biogas production process has not yet been investigated extensively in continuous reactor operation on complex substrates. In this study we have investigated the extent to which a microbial fuel cell (MFC) can reduce COD and recover further energy from the effluent of a two-stage biohydrogen and biomethane system. The performance of a four-module tubular MFC was determined at six different organic loadings (0.036–6.149 g sCOD L−1 d−1) in terms of power generation, COD removal efficiency, coulombic efficiency (CE) and energy conversion efficiency (ECE). A power density of 3.1 W m−3 was observed at the OLR = 0.572 g sCOD L−1 d−1, which resulted in the highest CE (60%) and ECE (0.8%), but the COD removal efficiency decreased at higher organic loading rates (35.1–4.4%). The energy recovery was 92.95 J L−1 and the energy conversion efficiency, based on total influent COD was found to be 0.48–0.81% at 0.572 g sCOD L−1 d−1. However, the energy recovery by the MFC is only reported for a four-module reactor and improved performance can be expected with an extended module count, as chemical energy remained available for further electrogenesis.  相似文献

Studies were conducted with crossbred weanling pigs to determine the level of phosphorus needed to be fed when a maize–soyabean meal–whey diet was supplemented with exogenous phytase (Natuphos™). In Trial 1, phytase was added at 1200 phytase units (PTU) kg−1 as phosphorus decreased. The control diet in Phase I (0–14 days) contained 7.3 g kg−1 phosphorus and in Phase II (14–28 days) contained 6.5 g kg−1 phosphorus. Dietary phosphorus was calculated to decrease by 0.8, 1.6 or 2.4 g kg−1 when phytase was supplemented. Chromic oxide was added for estimation of apparent absorption of phosphorus. Performance was optimum when 5.7 and 4.8 g kg−1 phosphorus (analysed levels) were fed with 1200 PTU kg−1 phytase in Phases I and II, respectively. The lowest dietary phosphorus levels did not reduce performance for the overall 28-day period. Apparent phosphorus digestibility was increased by phytase in Phase I when 5.7 g kg−1 phosphorus was fed compared to the control diet and in Phase II when 6.0 g kg−1 phosphorus was fed with phytase. Faecal phosphorus excretion decreased in both phases as dietary phosphorus decreased. Faecal phosphorus excretion was minimized at the lowest phosphorus level with no decrease in performance. The estimated requirement for dietary phosphorus, as determined by the NLIN procedure, is 5.0 g kg−1 in Phase I and 4.3 g kg−1 in Phase II when 1200 PTU kg−1 is used. In Trial 2, phytase was supplemented at 500 PTU kg−1 when phosphorus was decreased in the diet. The control diet contained 6.6 and 6.0 g kg−1 phosphorus in Phases I and II, respectively, and phosphorus was calculated to decrease by 0.5, 1.0, 1.5, or 2.0 g kg−1 when phytase was added. Daily gain decreased when 5.0 g kg−1 phosphorus was fed in Phase I and when 4.6 or 4.2 g kg−1 (analysed levels) phosphorus was fed in Phase II with 500 PTU kg−1. Faecal phosphorus excretion decreased as dietary phosphorus decreased, but there were no treatment effects on apparent phosphorus digestibility. The dietary phosphorus requirement was estimated to be 5.7 and 5.0 g kg−1 in Phases I and II, respectively, when phytase is fed at 500 PTU kg−1. At the present recommendation of 500 PTU kg−1 in starter feed, phosphorus can be decreased by 0.10 g kg−1. However, higher levels of phytase are needed to actually increase apparent phosphorus digestibility.  相似文献

Patch dynamics of the Mediterranean slow-growing seagrass Posidonia oceanica was studied in two shallow sites (3–10 m) of the Balearic Archipelago (Spain) through repeated censuses (1–2 year−1). In the sheltered site of Es Port Bay (Cabrera Island), initial patch density (October 2001) was low: 0.05 patches m−2, and the patch size (number of shoots) distribution was bimodal: most of the patches had less than 6 shoots or between 20 and 50 shoots. Mean patch recruitment in Es Port Bay (0.006 ± 0.002 patches m−2 year−1) exceeded mean patch loss (0.001 ± 0.001 patches m−2 year−1), yielding positive net patch recruitment (0.004 ± 0.003 patches m−2 year−1) and a slightly increased patch density 3 years later (July 2004, 0.06 patches m−2). In the exposed site of S’Estanyol, the initial patch density was higher (1.38 patches m−2, August 2003), and patch size frequency decreased exponentially with size. Patch recruitment (0.26 patches m−2 year−1) and loss (0.24 patches m−2 year−1) were high, yielding a slightly increased patch density in the area 1 year later (October 2004, 1.40 patches m−2). Most recruited patches consisted of rooting vegetative fragments of 1–2 shoots. Seedling recruitment was observed in Summer 2004 at both sites. Episodic, seedling recruitment comprised 30% and 25% of total patch recruitment in Es Port Bay and S’Estanyol, respectively. Patch survival increased with patch size and no direct removal was observed among patches of 5 shoots or more. Most patches grew along the study, shifting patch distribution towards larger sizes. Within the size range studied (1–150 shoots), absolute shoot recruitment (shoots year−1) increased linearly with patch size (R2 = 0.64, p < 4 × 10−5, N = 125), while specific shoot recruitment was constant (about 0.25 ± 0.05 year−1), although its variance was large for small patches. Given the slow growth rate and the high survival of patches with 5 or more shoots, even the low patch recruitment rates reported here could play a significant role in the colonisation process of P. oceanica.  相似文献

Marine toxic dinoflagellates of the genus Gambierdiscus are the causative agents of ciguatera fish poisoning (CFP), a form of seafood poisoning that is widespread in tropical, subtropical and temperate regions worldwide. The distributions of Gambierdiscus australes, Gambierdiscus scabrosus and two phylotypes of Gambierdiscus spp. type 2 and type 3 have been reported for the waters surrounding the main island of Japan. To explore the bloom dynamics and the vertical distribution of these Japanese species and phylotypes of Gambierdiscus, the effects of light intensity on their growth were tested, using a photoirradiation-culture system. The relationship between the observed growth rates and light intensity conditions for the four species/phylotypes were formulated at R > 0.92 (p < 0.01) using regression analysis and photosynthesis-light intensity (P-L) model. Based on this equation, the optimum light intensity (Lmax) and the semi-optimum light intensity range (Ls-opt) that resulted in the maximum growth rate (μmax) and ≥80% μ max values of the four species/phylotypes, respectively, were as follows: (1) the Lmax and Ls-opt of G. australes were 208 μmol photons m−2 s−1 and 91–422 μmol photons m−2 s−1, respectively; (2) those of G. scabrosus were 252 and 120–421 μmol photons m−2 s−1, respectively; (3) those of Gambierdiscus sp. type 2 were 192 and 75–430 μmol photons m−2 s−1, respectively; and (4) those of Gambierdiscus sp. type 3 were ≥427 and 73–427 μmol photons m−2 s−1, respectively. All four Gambierdiscus species/phylotypes required approximately 10 μmol photons m−2 s−1 to maintain growth. The light intensities in coastal waters at a site in Tosa Bay were measured vertically at 1 m intervals once per season. The relationships between the observed light intensity and depth were formulated using Beer’s Law. Based on these equations, the range of the attenuation coefficients at Tosa Bay site was determined to be 0.058–0.119 m−1. The values 1700 μmol photons m−2 s−1, 500 μmol photons m−2 s−1, and 200 μmol photons m−2 s−1 were substituted into the equations to estimate the vertical profiles of light intensity at sunny midday, cloudy midday and rainy midday, respectively. Based on the regression equations coupled with the empirically determined attenuation coefficients for each of the four seasons, the ranges of the projected depths of Lmax and Ls-opt for the four Gambierdiscus species/phylotypes under sunny midday conditions, cloudy midday conditions, and rainy midday conditions were 12–38 m and 12–54 m, 1–16 m and 1–33 m, and 0 m and 0–16 m, respectively. These results suggest that light intensity plays an important role in the bloom dynamics and vertical distribution of Gambierdiscus species/phylotypes in Japanese coastal waters.  相似文献

The research on the function and mechanism of selenium (Se) is of great significance for the development of Se-enriched agricultural products. In this paper, uptake, speciation distribution, the effects on the flue-cured tobacco growth and antioxidant system of Se at different levels (0–22.2 mg Se kg−1) were studied through a pot experiment, aiming to clarify flue-cured tobacco's response to Se stress and the relationship between Se speciation and antioxidant system. The results showed that the leaf area and number, the biomass and the chlorophyll content reached the maximum at 4.4 mg kg−1 of Se treatment. Selenium at low levels (≤4.4 mg kg−1) stimulated the growth of flue-cured tobacco by elevating the capability of antioxidant stress and reducing the malondialdehyde (MDA) content to 0.6–0.8 times of that of the control. However, high Se levels (≥11.1 mg kg−1) depressed the capability of antioxidant stress and raised the MDA content to 1.5-fold of that of the control, and meanwhile the biomass of the aboveground parts and underground parts declined notably. The Se content in different parts of flue-cured tobacco significantly increased with the growth of Se levels. The range of Se content in roots, leaves and stems at 2.2–22.2 mg kg−1 of Se treatment were 16.7–58.6 mg kg−1, 2.6–37.3 mg kg−1 and 2.2–10.3 mg kg−1, respectively. According to the detection of different Se speciation, only selenocysteine (SeCys) was detectable in leaves at 2.2 mg kg−1 Se treatment; SeCys, selenite [Se(IV)]and selenate [Se(VI)] were detected in flue-cured tobacco leaves at Se treatment (≥4.4 mg kg−1), which accounted for 4.6–10%, 9–18.7% and 71–86% respectively; SeCys, selenomethionine (SeMet) and Se(IV) were detected in roots, and organic selenium(66–84%) was the main Se species at Se  11.1 mg kg−1 treatment; four Se species [SeCys, SeMet, Se(IV) and Se(VI)] were detected in flue-cured tobacco roots, and the main Se species was inorganic Se (60%) at 22.2 mg kg−1 Se treatment. That was to say, the percentage of organic Se species (SeCys and SeMet in flue-cured tobacco leaves and root) declined, whereas the ratio of inorganic Se species [Se(IV) and Se(VI)] increased with the growth of Se levels. The correlation analysis showed that the superoxide dismutase (SOD) activity as well as the glutathione (GSH) and MDA contents were positively correlated with the Se(IV) and Se(VI) contents at P < 0.01 and excessive inorganic Se might destruct the reactive oxygen species (ROS) balance and enhance the MDA content, thus causing damage to the plant growth. In a word, the present study suggested that the ratio of inorganic Se [Se(IV) and Se(VI)] was closely related with the growth and the antioxidant capacity of flue-cured tobacco and the excessive application of Se led to the higher proportion of inorganic Se and poorer antioxidant capacity, which ultimately inhibited the growth of flue-cured tobacco.  相似文献

The dairy industry is generally considered to be the largest source of food processing wastewater in many countries. The highly variable nature of dairy wastewaters in terms of volumes and flowrates and in terms of high organic materials contents such as COD 921–9004 mg L−1, BOD 483–6080 mg L−1, TN of 8–230 mg L−1 and SS of 134–804 mg L−1 makes the choice of an effective wastewater treatment regime difficult. A high performance bioreactor, an aerobic jet loop reactor, combined with a ceramic membrane filtration unit, was used to investigate its suitability for the treatment of the dairy processing wastewater. The oxygen transfer rates of the bioreactor were found to be very high (100–285 h−1) on the operating conditions. A loading rate of 53 kg COD m−3 d−1 resulted in 97–98% COD removal efficiencies under 3 h hydraulic retention time. The high MLSS concentrations could be retained in the system (up to 38,000 mg L−1) with the contribution of UF (ultrafiltration) unit. During the filtration of activated sludge, the fluxes decreased with increasing MLSS. Cake formation fouling was determined as dominant fouling mechanisms. The results demonstrate that jet loop membrane bioreactor system was a suitable and effective treatment choice for treating dairy industry wastewater.  相似文献

The photosynthetic and repiratory metabolism of Zostera marina and maerl communities was compared, in the same area of the Bay of Brest in March–April, using benthic chambers. PE curves for both oxygen and carbon were established for bottom irradiances between 0 and 525 μmol m−2 s−1. An exponential function was fitted to calculate daily production. Community metabolic quotients did not differ for maerl and seagrass beds. Community photosynthetic quotients were significantly higher (1.19) whereas community respiratory quotients were lower (0.70) than 1. Maerl and seagrass bed PE curves mainly differed by the minimum saturating irradiance (Ek). Net community production was estimated to 26.8 mmol C m−2 d−1 for Z. marina meadows and 8.6 mmol C m−2 d−1 for maerl beds. The two communities can, therefore, be considered as autotrophic during the March–April period. Community respiration did not differ between Z. marina meadows and maerl beds, with an average value of 53.8 mmol C m−2 d−1 during a day. In similar environmental conditions, the production of maerl beds corresponds to approximately one third that of seagrass meadows. The maerl communities, therefore, form productive ecosystems, relevant to temperate coastal ecosystems functioning.  相似文献
