A high proportion of nonreproductive (NR) Varroa destructor Anderson & Trueman (Mesostigmata: Varroidae), is commonly observed in honey bee colonies displaying the varroa sensitive hygienic trait (VSH). This study was conducted to determine the influence of brood removal and subsequent host reinvasion of varroa mites on mite reproduction. We collected foundress mites from stages of brood (newly sealed larvae, prepupae, white-eyed pupae, and pink-eyed pupae) and phoretic mites from adult bees. We then inoculated these mites into cells containing newly sealed larvae. Successful reproduction (foundress laid both a mature male and female) was low (13%) but most common in mites coming from sealed larvae. Unsuccessful reproductive attempts (foundress failed to produce both a mature male and female) were most common in mites from sealed larvae (22%) and prepupae (61%). Lack of any progeny was most common for mites from white-eyed (83%) and pink-eyed pupae (92%). We also collected foundress mites from sealed larvae and transferred them to cells containing newly sealed larvae, prepupae, white-eyed pupae, or pink-eyed pupae. Successful reproduction only occurred in the transfers to sealed larvae (26%). Unsuccessful reproductive attempts were most common in transfers to newly sealed larvae (40%) and to prepupae (25%). Unsuccessful attempts involved the production of immature progeny (60%), the production of only mature daughters (26%) or the production of only a mature male (14%). Generally, lack of progeny was not associated with mites having a lack of stored sperm. Our results suggest that mites exposed to the removal of prepupae or older brood due to hygiene are unlikely to produce viable mites if they invade new hosts soon after brood removal. Asynchrony between the reproductive status of reinvading mites and the developmental stage of their reinvasion hosts may be a primary cause of NR mites in hygienic colonies. Even if reinvading mites use hosts having the proper age for infestation, only a minority of them will reproduce.  相似文献

The enzyme spectrum of an ectoparasitic mite of the honeybee,Varroa destructor (Anderson and Trueman) was studied usinga semi-quantitative method, especially designed for complex samples which havenot been purified. Exopeptidases and phosphatases are shown present. Achitinaseand enzymes able to transform carbohydrates are also present with a largerange in the intensity of the reaction. The role of the chitinase can berelatedto the supply of nutritional needs or/and the piercing and sucking behaviour ofthe adult parasite. The ectoparasitic honey bee mite Varroa destructor was originally confined to the Eastern honey bee Apis cerana. After a shift to the new host Apis mellifera during the first half of the last century, the parasite dispersed world wide and is currently considered the major threat for apiculture. The damage caused by Varroosis is thought to be a crucial driver for the periodical colony losses in Europe and the USA and regular Varroa treatments are essential in these countries. Therefore, Varroa research not only deals with a fascinating host–parasite relationship but also has a responsibility to find sustainable solutions for the beekeeping.This review provides a survey of the current knowledge in the main fields of Varroa research including the biology of the mite, damage to the host, host tolerance, tolerance breeding and Varroa treatment. We first present a general view on the functional morphology and on the biology of the Varroa mite with special emphasis on host–parasite interactions during reproduction of the female mite. The pathology section describes host damage at the individual and colony level including the problem of transmission of secondary infections by the mite. Knowledge of both the biology and the pathology of Varroa mites is essential for understanding possible tolerance mechanisms in the honey bee host. We comment on the few examples of natural tolerance in A. mellifera and evaluate recent approaches to the selection of Varroa tolerant honey bees. Finally, an extensive listing and critical evaluation of chemical and biological methods of Varroa treatments is given.This compilation of present-day knowledge on Varroa honey bee interactions emphasizes that we are still far from a solution for Varroa infestation and that, therefore, further research on mite biology, tolerance breeding, and Varroa treatment is urgently needed.  相似文献

The mite Varroa destructor is an obligatory ectoparasite of the honey bee (Apis mellifera) and is one of the major threats to apiculture worldwide. We previously reported that honey bees fed on double-stranded RNA (dsRNA) with a sequence homologous to that of the Israeli acute paralysis virus are protected from the viral disease. Here we show that dsRNA ingested by bees is transferred to the Varroa mite and from mite on to a parasitized bee. This cross-species, reciprocal exchange of dsRNA between bee and Varroa engendered targeted gene silencing in the latter, and resulted in an over 60% decrease in the mite population. Thus, transfer of gene-silencing-triggering molecules between this invertebrate host and its ectoparasite could lead to a conceptually novel approach to Varroa control.  相似文献

Reproducing Varroa females begin oviposition on a host larva by laying an unfertilized (male) egg, followed by fertilized (female) offspring. Using transfer experiments, we examined whether the sequence of sexes in the brood cell is triggered by a host stimulus. When reproducing Varroa females were transferred from white-eyed pupae (worker brood) into freshly capped worker brood cells, 77% (n = 22 fertile mites after the transfer) began a new reproductive cycle by laying a male egg. The proportion of brood cells with male offspring was similar to naturally infested brood cells. Varroa females transferred into brood cells with young pupae reproduced, but only 6% (n = 16 fertile mites after the transfer) produced male offspring. This was significantly different from male production in naturally reproducing Varroa females and those transferred into freshly capped brood cells. We conclude that a host stimulus present in freshly capped brood cells triggers both the start of reproduction and the sequence of sexes.  相似文献

There has been growing concern over declines in populations of honey bees and other pollinators which are a vital part to our food security. It is imperative to identify factors responsible for accelerated declines in bee populations and develop solutions for reversing bee losses. While exact causes of colony losses remain elusive, risk factors thought to play key roles are ectoparasitic mites Varroa destructor and neonicotinoid pesticides. The present study aims to investigate effects of a neonicotinoid pesticide Imidacloprid and Varroa mites individually on survivorship, growth, physiology, virus dynamics and immunity of honey bee workers. Our study provides clear evidence that the exposure to sublethal doses of Imidacloprid could exert a significantly negative effect on health and survival of honey bees. We observed a significant reduction in the titer of vitellogenin (Vg), an egg yolk precursor that regulates the honey bees development and behavior and often are linked to energy homeostasis, in bees exposed to Imidacloprid. This result indicates that sublethal exposure to neonicotinoid could lead to increased energy usage in honey bees as detoxification is a energy‐consuming metabolic process and suggests that Vg could be a useful biomarker for measuring levels of energy stress and sublethal effects of pesticides on honey bees. Measurement of the quantitative effects of different levels of Varroa mite infestation on the replication dynamic of Deformed wing virus (DWV), an RNA virus associated with Varroa infestation, and expression level of immune genes yields unique insights into how honey bees respond to stressors under laboratory conditions.  相似文献

Background

Recent elevated winter loss of honey bee colonies is a major concern. The presence of the mite Varroa destructor in colonies places an important pressure on bee health. V. destructor shortens the lifespan of individual bees, while long lifespan during winter is a primary requirement to survive until the next spring. We investigated in two subsequent years the effects of different levels of V. destructor infestation during the transition from short-lived summer bees to long-lived winter bees on the lifespan of individual bees and the survival of bee colonies during winter. Colonies treated earlier in the season to reduce V. destructor infestation during the development of winter bees were expected to have longer bee lifespan and higher colony survival after winter.

Methodology/Principal Findings

Mite infestation was reduced using acaricide treatments during different months (July, August, September, or not treated). We found that the number of capped brood cells decreased drastically between August and November, while at the same time, the lifespan of the bees (marked cohorts) increased indicating the transition to winter bees. Low V. destructor infestation levels before and during the transition to winter bees resulted in an increase in lifespan of bees and higher colony survival compared to colonies that were not treated and that had higher infestation levels. A variety of stress-related factors could have contributed to the variation in longevity and winter survival that we found between years.

Conclusions/Significance

This study contributes to theory about the multiple causes for the recent elevated colony losses in honey bees. Our study shows the correlation between long lifespan of winter bees and colony loss in spring. Moreover, we show that colonies treated earlier in the season had reduced V. destructor infestation during the development of winter bees resulting in longer bee lifespan and higher colony survival after winter.  相似文献

The present study analyzes the fatty acid (FA) profile of lipids isolated from Varroa destructor Anderson & Trueman, a parasitic mite of the honey bee (Apis mellifera L.), uninfected and infected worker prepupae of the Carnolian subspecies Apis mellifera carnica Pollmann, and bee bread fed to the worker brood. Significant differences are observed in the FA profiles of lipids isolated from parasites, hosts and bee bread. Parasitism by V. destructor (henceforth, varroosis) induces visible changes in the lipid profile of worker prepupae. In infected prepupae, the percentage of total saturated FAs is lower and the percentage of unsaturated FAs is higher than in uninfected insects. These differences result from significant changes in the percentages of FAs that are most abundant in the evaluated groups (i.e. C16:0, C18:1 9c, C18:2n‐6 and C18:3n‐3 FAs). In mites and in uninfected and infected prepupae, the predominant FAs are oleic acid (41.07 ± 2.26%, 42.79 ± 1.21% and 45 ± 0.20%, respectively) and palmitic acid (22.62 ± 0.87%, 39.48 ± 0.43% and 36.84 ± 0.22%, respectively). Highly significant differences in FA composition are noted between bee bread and worker brood. The results suggest specific mechanisms of FA uptake, accumulation and metabolism in the food chain of this parasitic association, beginning from the food processed by nurse bees for larval feeding, through host organisms (worker brood) to V. destructor mites.  相似文献

Current high losses of honeybees seriously threaten crop pollination. Whereas parasite exposure is acknowledged as an important cause of these losses, the role of insecticides is controversial. Parasites and neonicotinoid insecticides reduce homing success of foragers (e.g. by reduced orientation), but it is unknown whether they negatively affect flight capacity. We investigated how exposing colonies to the parasitic mite Varroa destructor and the neonicotinoid insecticide imidacloprid affect flight capacity of foragers. Flight distance, time and speed of foragers were measured in flight mills to assess the relative and interactive effects of high V. destructor load and a field-realistic, chronic sub-lethal dose of imidacloprid. Foragers from colonies exposed to high levels of V. destructor flew shorter distances, with a larger effect when also exposed to imidacloprid. Bee body mass partly explained our results as bees were heavier when exposed to these stressors, possibly due to an earlier onset of foraging. Our findings contribute to understanding of interacting stressors that can explain colony losses. Reduced flight capacity decreases the food-collecting ability of honeybees and may hamper the use of precocious foraging as a coping mechanism during colony (nutritional) stress. Ineffective coping mechanisms may lead to destructive cascading effects and subsequent colony collapse.  相似文献

1. Varroa destructor Linnaeus (Acari: Varroidae) is one of the greatest threats to apiculture. This study examines the role of host density, natural resource availability for bees, the management and climate in driving spatial and annual variability in the abundance of Varroa, and the occurrence of colony losses, recorded in apiaries across a Mediterranean island over a 2‐year period, using a hierarchical generalised linear model framework. 2. The seasonal abundance of Varroa showed a bimodal pattern with two peaks, the first one being in spring and larger than the second one located in summer/autumn. In contrast, bee colony losses were mainly concentrated in autumn/winter. 3. The abundance patterns of Varroa were best explained by models combining host, climate, and resource availability. A key novel finding was that low availability of natural flowering resources leads to high levels of infestation of Varroa, highlighting the importance of preserving natural resources around apiaries for the maintenance of pollination services in the landscape. Varroa abundance was also found to increase as bee density increased, probably as a result of the greater brood availability. Moreover, Varroa abundance increased as temperatures decrease and decreases as relative humidity increases, which is consistent with previous studies. Anti‐varroa treatments were only found to impact Varroa levels in the second 6 months of the year. Organic treatments outperformed synthetic treatments. 4. Empirical research on optimal seasonal timing and combinations of treatments, as well as impacts of climate and resource availability on natural dynamics of bees and Varroa in different climate zones, is urgently required.  相似文献

The ectoparasitic mite, Varroa destructor, shifted host from the eastern honeybee, Apis cerana, to the western honeybee, Apis mellifera. Whereas the original host survives infestations by this parasite, they are lethal to colonies of its new host. Here, we investigated a population of A. cerana naturally infested by the V. destructor Korea haplotype that gave rise to the globally invasive mite lineage. Our aim was to better characterize traits that allow for the survival of the original host to infestations by this particular mite haplotype. A known major trait of resistance is the lack of mite reproduction on worker brood in A. cerana. We show that this trait is neither due to a lack of host attractiveness nor of reproduction initiation by the parasite. However, successful mite reproduction was prevented by abnormal host development. Adult A. cerana workers recognized this state and removed hosts and parasites, which greatly affected the fitness of the parasite. These results confirm and complete previous observations of brood susceptibility to infestation in other honeybee host populations, provide new insights into the coevolution between hosts and parasites in this system, and may contribute to mitigating the large‐scale colony losses of A. mellifera due to V. destructor.  相似文献

Some DNA viruses infect host animals usually by integrating their DNAs into the host genome. However, the mechanisms for integration remain largely unknown. Here, we find that Cotesia vestalis bracovirus (CvBV), a polydnavirus of the parasitic wasp C. vestalis (Haliday), integrates its DNA circles into host Plutella xylostella (L.) genome by two distinct strategies, conservatively and randomly, through high-throughput sequencing analysis. We confirmed that the conservatively integrating circles contain an essential “8+5” nucleotides motif which is required for integration. Then we find CvBV circles are integrated into the caterpillar’s genome in three temporal patterns, the early, mid and late stage-integration. We further identify that three CvBV-encoded integrases are responsible for some, but not all of the virus circle integrations, indeed they mainly participate in the processes of early stage-integration. Strikingly, we find two P. xylostella retroviral integrases (PxIN1 and PxIN2) are highly induced upon wasp parasitism, and PxIN1 is crucial for integration of some other early-integrated CvBV circles, such as CvBV_04, CvBV_12 and CvBV_24, while PxIN2 is important for integration of a late-integrated CvBV circle, CvBV_21. Our data uncover a novel mechanism in which CvBV integrates into the infected host genome, not only by utilizing its own integrases, but also by recruiting host enzymes. These findings will strongly deepen our understanding of how bracoviruses regulate and integrate into their hosts.  相似文献

世界各地大范围的西方蜜蜂Apis mellifera蜂群损失现象已引起科学界和公众的持续关注。狄斯瓦螨Varroa destructor和蜜蜂残翅病毒(Deformed wing virus,DWV)是西方蜜蜂群中最主要的两大生物威胁。尽管二者侵害蜜蜂均已有较长历史,但直至近十年来的研究才发现两者间的协同效应对蜂群健康的影响远超过其单独作用时所造成的危害:(1)蜜蜂残翅病毒可在狄斯瓦螨体内大量复制,继而进一步传播;(2)狄斯瓦螨的刺吸行为使病毒粒子跨越寄主的生理屏障而直接进入蜜蜂血淋巴;(3)狄斯瓦螨的寄生促使蜜蜂残翅病毒的高毒力毒株在蜂群中优势扩增和盛行;(4)狄斯瓦螨影响蜜蜂个体发育与免疫系统等生理机能,以致降低了蜂群对病毒的抵抗力;(5)蜜蜂残翅病毒对宿主造成的免疫抑制有利于狄斯瓦螨的寄生与繁殖。狄斯瓦螨、蜜蜂残翅病毒和西方蜜蜂间的关系已经成为昆虫外寄生物、病原体与寄主相互作用研究的一个典型模型。本文对近十年该领域的相关研究进行综述,以期为蜂群损失的原因调查以及昆虫寄生虫、病原微生物与寄主间关系的研究提供参考和借鉴。  相似文献

BLM and WRN, the products of the Bloom’s and Werner’s syndrome genes, are members of the RecQ family of DNA helicases. Although both have been shown previously to unwind simple, partial duplex DNA substrates with 3′→5′ polarity, little is known about the structural features of DNA that determine the substrate specificities of these enzymes. We have compared the substrate specificities of the BLM and WRN proteins using a variety of partial duplex DNA molecules, which are based upon a common core nucleotide sequence. We show that neither BLM nor WRN is capable of unwinding duplex DNA from a blunt-ended terminus or from an internal nick. However, both enzymes efficiently unwind the same blunt-ended duplex containing a centrally located 12 nt single-stranded ‘bubble’, as well as a synthetic X-structure (a model for the Holliday junction recombination intermediate) in which each ‘arm’ of the 4-way junction is blunt-ended. Surprisingly, a 3′-tailed duplex, a standard substrate for 3′→5′ helicases, is unwound much less efficiently by BLM and WRN than are the bubble and X-structure substrates. These data show conclusively that a single-stranded 3′-tail is not a structural requirement for unwinding of standard B-form DNA by these helicases. BLM and WRN also both unwind a variety of different forms of G-quadruplex DNA, a structure that can form at guanine-rich sequences present at several genomic loci. Our data indicate that BLM and WRN are atypical helicases that are highly DNA structure specific and have similar substrate specificities. We interpret these data in the light of the genomic instability and hyper-recombination characteristics of cells from individuals with Bloom’s or Werner’s syndrome.  相似文献
