A series of eight histrionicotoxins and two synthetic analogs inhibit binding of [3H]batrachotoxinin B to sites on voltage dependent sodium channels in brain membranes. Perhydrohistrionicotoxin (IC50 0.33 M) and octahydrohistrionicotoxin (IC50 1.2 M) are comparable in activities to potent local anesthetics. Histrionicotoxin (IC50 17 M) and the other histrionicotoxins are much less potent. The histrionicotoxins also inhibit binding of [3H]phencyclidine to putative potassium channels in brain membranes. Histrionicotoxin (IC50 15 M) and the other histrionicotoxins are much more potent than perhydrohistrionicotoxin (IC50 200 M), but are at least 200-fold less potent than phencyclidine. The histrionicotoxins enhance binding of [3H]nitrendipine to sites on calcium channels in brain membranes, with the exception of perhydrohistrionicotoxin, which inhibits binding. Structure activity relationships at these channel sites and at the sites for noncompetitive blockers on the nicotinic acetylcholine receptor channel (AChR) complex differ. The histrionicotoxins are more potent at the sites on the AChR complex than at sites on other channels with the exception of perhydrohistrionicotoxin, which has comparable potency at the AChR complex and sodium channels.  相似文献

The binding of the calcium antagonist [3H] nitrendipine ([3H] NDP) to brain and heart is described and the brain site is characterized. The binding is saturable, specific and of very high affinity with KD values of 0.16 nM in brain and 0.21 nM in heart. Our kinetic results are similar to those recently reported by two other groups (1,2), indicating a saturable, high affinity binding site in brain. In brain the binding sites are enriched in crude nuclear and synaptosomal fractions. The highest levels of binding are seen in the hippocampus, caudate and cerebral cortex with much lower levels in the cerebellum and pons. Calcium has a marked stimulatory effect on [3H] NDP binding at 10?4 M. Addition of 0.5 mM CaCl2 to EDTA treated membranes nearly doubles the number of binding sites. Of the many drugs and neurotransmitters tested only other calcium antagonists, i.e., verapamil, inhibit binding (IC50 = 250 nM). The inhibition of [3H] NDP binding by verapamil is apparently non-competitive and not complete, suggesting that [3H] NDP binds to several sites, only some of which are inhibited by verapamil. The [3H] NDP binding site is probably a protein since it is very sensitive to trypsin, heat and sulfhydryl reagents.  相似文献

Abstract

Cooperative modulation of [3H]MK-801 binding to extensively washed pig cortical brain membranes in the presence of various concentrations of L-glutamate, glycine, spermine, CPP and DCKA was evaluated in association experiments. In saturation experiments [3H]MK-801 labelled a homogeneous population of binding sites with a Kd-value of 1.26 ± 0.18 nmol 1?1 and a Bmax-value of 2130 ± 200 fmol/mg protein. The pharmacological profile of this site was further evaluated in competition experiments with known NMDA receptor channel blockers. In nonequilibrium binding experiments EC50-values of reference compounds acting at the L-glutamate, at the glycine, and at the polyamine site, were determined by increasing or decreasing [3H]MK-801 binding. Ifenprodil reduced [3H]MK-801 binding in a biphasic manner. All the data obtained are in agreement with results from [3H]MK-801 binding to rodent as well as human brain membranes. This study therefore strongly suggests, that pig cortical membranes are a suitable alternative to rodent brain membranes, and an acceptable substitute for human brain membranes in [3H]MK-801 binding experiments.  相似文献

Abstract

The effects of incubation temperature and allosteric modulators were studied on [3H]nitrendipine binding to guinea-pig cardiac membranes. Incubation temperature only slightly affected the ability of nifedipine and verapamil derivatives to inhibit binding. By contrast, the Ca2+ channel blockers d-cis-diltiazem and fostedil (KB-944) stimulated [3H]nitrendipine binding in a temperature-dependent manner (37° > 25° > 4° C). The stimulatory effect of fostedil could be related to a decrease (2.3-fold at 37° C) in the rate of radioligand binding site dissociation, without significant effects on association kinetics. Both fostedil and d-cis-diltiazem caused a shift to the right of the concentration-inhibition curve of tiapamil, a negative allosteric modulator of [3H]nitrendipine binding. Neither compound affected the ability of nifedipine, a competitive antagonist, to inhibit radioligand binding. This selective effect of fostedil or d-cis-diltiazem may be useful for testing whether potential Ca2+ channel blockers interact in a competitive as opposed to allosteric manner with the dihydropyridine site. Varying the incubation temperature may also be useful in detecting compounds which act as positive allosteric modulators (stimulators) of dihydropyridine binding.  相似文献

Abstract

A method for measuring [3H]-AMPA binding in rat cortex membranes is described. Specific binding was saturable and accounted for 95% of total binding at 5 nM of [3H]-AMPA. Non linear curve fitting of [3H]-AMPA saturation isotherms suggested the presence of two binding sites: the high affinity site showed a pKd of 8.26 ± 0.07 (Kd = 5.49 nM) and a Bmax of 0.19 ± 0.03 pmol/mg protein, whereas the low affinity site indicated a pKd of 7.28 ± 0.05 (Kd = 52 nM) and a Bmax of 1.30 ± 0.23 pmol/mg protein. The pharmacological profile of [3H]-AMPA binding has been determined by studying a series of compounds in binding displacement experiments: Quisqualate was the most potent inhibitor of [3H]-AMPA binding (IC50 = 9.7 nM), followed by AMPA (19 nM), CNQX, DNQX and L-Glutamate (272–373 nM). Kainate was a moderate displacer (6.2 μM); Ibotenic acid and glycine were very weak inhibitors (74 and 92 μM, respectively). CPP, GAMS and L-Aspartic acid showed IC50-values of over 400 μM and MK-801, DL-AP5 and NMDA were almost inactive at the maximal concentration used in our experiments.  相似文献

The binding of the dihydropyridine calcium channel antagonist [3H]nitrendipine to whole rat brain synaptosomes was studied. Binding was specific, saturable, and of high affinity (Kd = 170 pM). The calcium channel antagonist diltiazem enhanced [3H]nitrendipine binding in synaptosomes in concentrations of 1 and 10 μM. Equilibrium saturation analysis demonstrated that this effect was mediated by a decrease in the dissociation constant, due to a 3-fold reduction in the rate of ligand-receptor complex dissociation. It is concluded that diltiazem allosterically modulates the calcium channel drug receptor labeled by [3H]nitrendipine in this preparation.  相似文献

The binding of [3H]nitrendipine to membrane preparation from turtle and rabbit brain was studied. A single population of [3H]nitrendipine binding sites was detected in both species. [3H]nitrendipine bound with high affinity to brain membrane from both rabbit and turtle, revealing a significant population of binding sites (K d values of 0.55±0.05 nM and 0.56±0.04 nM and Bmax values of 122±11 and 275±18 fmol/mg of protein, respectively). Displacement studies showed a similar order of potency of various unlabeled ligands against [3H]nitrendipine both in rabbit or in turtle: nitrendipine > nifedipine ≥ nicardipine ≫ verapamil ≥ diltiazem. Our results show that a two fold increment of [3H]nitrendipine binding sites exists in the turtle brain respect to the rabbit.  相似文献

The sarcoplasmic reticulum (S.R.) of rabbit skeletal muscle has been found to contain a single, high affinity binding site for the Ca antagonist drug [3H] -nitrendipine. Two subfractions of the reticulum were studied, the heavy (HSR) and light (LSR) preparations, which exhibited similar nitrendipine equilibrium dissociation constants (KD) of 1nM. Crude cardiac and brain membranes assayed under the same conditions exhibited KD values of 0.2–0.3nM. The concentration of binding sites per mg. protein (Bmax) in HSR was found to be very high, namely 6.7 picomoles/mg, some four times greater than that of LSR. [3H] -nitrendipine binding to HSR was reversible and inhibited by the Ca antagonists flunarizine and verapamil, and by the intracellular Ca release antagonist TMB-8 (8-diethylamino-octyl 3,4,5- trimethylbenzoate hydrochloride). However, unlabelled nitrendipine at 2 × 10?5M had no effect on contraction of isolated electrically stimulated rabbit lumbrical or rat diaphragm muscles, nor did it affect the neuromuscular junction as studied in rat phrenic nerve-diaphragm preparations. Also, little effect of 2 × 10?5M nitrendipine was seen on net 45Ca uptake by HSR. These results suggest that [3H] -nitrendipine binding to skeletal muscle S.R. resembles that of brain membranes, which also contain a high affinity binding site for [3H] -nitrendipine and which similarly are pharmacologically insensitive to this dihydropyridine type of Ca channel blocking agent. Since HSR is also enriched in calsequestrin and terminal cysternae from which Ca is released in vivo, it seems likely that the [3H]- nitrendipine binding sites in S.R. are associated with Ca channels in the S.R.  相似文献

The binding properties of the calcium channel antagonist, [3H]nitrendipine, were investigated in homogenates of the rat cerebral cortex, heart and ileum. The specific component of [3H]nitrendipine binding was consistent with mass-action behavior and was characterized by a high affinity dissociation constant in the range of 0.1 ? 0.3 nM. A variety of other calcium channel antagonists inhibited the binding of [3H]nitrendipine with Ki's that agree generally with the ability of these drugs to block contractions of cardiac and smooth muscle. The inhibition of [3H]nitredipine binding by other dihydropyridines was consistent with competitive antagonism whereas the inhibition caused by verapamil and D600 resembled negative heterotropic cooperativity. Consistent with this latter postulate was the observation that the kinetics of [3H]nitrendipine binding are altered by verapamil, with both the association rate and the dissociation rate being increased. La+3 and several divalent cations caused an inhibition of [3H]nitrendipine with the rank order of potency being Cd+2 > La+3 > Ni+2 > Co+2 ? Mn+2 > Mg+2 ? Ba+2 > Ca+2.  相似文献

High affinity binding sites for the calcium channel inhibitor [3H]nitrendipine have been identified in microsomes from pig coronary arteries (KD=1.6 nM; Bmax=35 fmol/mg) and in purified sarcolemma from dog heart (KD=0.11 nM; Bmax=230 fmol/mg). [3H]nitrendipine binding to coronary artery microsomes was completely inhibited by nifedipine, partially by verapamil and D600 and, surprisingly, was stimulated by d-cis-diltiazem but not by 1-cis-diltiazem, a less active isomer. Half-maximal relaxation of KCl-depolarized coronary rings occurred in a slow process at 1 nM nitrendipine or 100 nM d-cis-diltiazem. In dog trabecular strips, nitrendipine caused a negative inotropic response (ED50=1μM). These results suggest that there may be multiple binding sites for different “subclasses” of calcium channel inhibitors, and that drug binding sites may be different molecular entities from the putative calcium channels.  相似文献

Summary. NPS Pharmaceuticals, Inc. (NPS) has synthesized a series of open-channel blockers with varying potencies at the NMDA receptor. NPS 1506 (Fig. 1) is a moderate affinity antagonist that inhibits NMDA/glycine-induced increases in cytosolic calcium in cultured rat cerebellar granule cells (IC50 = 476 nM) and displaces the binding of [3H]MK-801 to rat cortical membranes (IC50 = 664 nM). Abstract

The ionic and drug specificities of the [3H]nifedipine binding site in rabbit cardiac homogenates were investigated. Divalent cations inhibited specific [3H]nifedipine binding in the potency order: Ni+2 > Ca+2 ≥ Mg+2. Monovalent cations did not affect binding. The inorganic calcium entry blocker La+3 (IC50 = 1.1 mM) was the most potent cation in inhibiting radioligand binding. Calcium entry blocking drugs of different chemical classes inhibited [3H]-nifedipine binding, with a rank potency order of: nifedipine >> D600 = verapamil > tiapamil > cinnarizine = prenylamine. The same potency order was observed for these drugs in inducing negative inotropic activity of isolated, electrically stimulated rabbit papillary muscle. The stereoselectivity of verapamil and D600 ((?) >> (+) isomers) in depressing papillary muscle contractions was not seen in [3H]nifedipine competition experiments. This presents an obstacle to accepting the equivalence of the [3H]nifedipine binding site with the myocardial Ca+2 channel. It is, however, possible that the myocardial Ca+2 channel may be associated with multiple sites of action for calcium entry blockers.  相似文献

Abstract: The effects of some GABA analogues and some drugs on the binding of [3H]muscimol (3.08 nM) to thoroughly washed subcellular particles prepared from a neuron-enriched culture of embryonic rat brain were examined using Na+-free Tris-citrate medium and a centrifugation method. Competition for [3H]muscimol binding sites by excess(10?5 M) unlabelled GABA provided estimates of “specific” binding. In accord with in vivo neuropharmacological studies on GABA receptors and with in vitro studies on cerebral membrane preparations, [3H]muscimol binding was potently inhibited by muscimol itself (IC50, 2.5 nM), GABA (1C50, 43 nM), isoguvacine (IC50, 61 nM), and 3-aminopropanesulphonic acid (IC50, 160 nM), and less potently inhibited by the GABA antagonist bicuculline methobromide (IC50, 800 nM). δ- Aminovaleric acid (IC50, 2.6 μM), the glycinelp-alanine antagonist strychnine (IC50, 6.6 μM), and the predominantly glial GABA uptake inhibitors β-alanine (IC50, 23 μM) and p-proline (IC50, 66 μM) also inhibited [3H]muscimol binding. Other inhibitors of Na+-dependent GABA uptake, (±)-nipecotic acid, L- 2,4-diaminobutyric acid, and guvacine, as well as picrotoxinin, were relatively inactive as inhibitors of [3H]muscimol binding (IC50≥ 1 mM). In addition to revealing that GABA receptors are present on neuronal membranes before the formation of most synapses, this binding of [3H]muscimol that occurs to neuronal, but not to glial, membranes might be useful as a “neuronal marker” and for the further characterization and isolation of GABA receptors.  相似文献

The aryl imidazoline compound UK-14, 304 (5-bromo-6-[2-imidazolin-2-yl-amino]-quinoxaline) is a potent and selective α2-adrenoceptor agonist with full intrinsic activity, unlike other imidazolines. We examined the characteristics of high specific activity (84 Ci/mmol) [3H] UK-14, 304 binding to rat cerebral cortex membranes. [3H] UK-14, 304 specific binding was enhanced by Mn2+ ion, and associated and dissociated moderately rapidly at 25°C. Norepinephrine-displaceable binding was saturable and monophasic, with a KD of 1.4 nM, in agreement with rate and competition experiments, and a Bmax of 200 fmol/mg protein. Competition studies revealed that binding was α2-adrenoceptor-specific, with yohimbine being 12 times more potent than prazosin. [3H] UK-14, 304 appeared to label predominantly the R(H) state of the brain α2-adrenoceptor, as judged by the high affinity of catecholamine and imidazoline agonists (IC50, 1–13 nM), and the relatively low affinity of yohimbine and rauwolscine (IC50, 100–300 nM), at the binding site. [3H] UK-14,304 compares favorably with other α2-adrenoceptor ligands because of its high affinity and specific activity.  相似文献

The intent of the present study was to investigate adenosine receptor sites in brain membranes of the saltwater teleost fish, Mullus surmuletus, using the A1 receptor selective agonist, [3H]CHA, and A2a receptor selective agonist [3H]CGS 21680. The A1 selective agonist, [3H]CHA, bound saturably, reversibly and with high affinity to a single-class of binding sites (Kd 1.47 nM; Bmax 100–190 fmol/mg protein, dependent on fish length). The A2a selective agonist, [3H]CGS 21680, also bound saturably, reversibly and with relative high affinity to a single-class of binding sites (Kd 44.2 nM; Bmax 150–300 fmol/mg protein dependent on fish length). In equilibrium competition experiments, adenosine analogous, NECA, CGS 21680, CHA, CPA, S-PIA, R-PIA, CPCA, DPMA, and xanthine antagonists, DPCPX, XAC, and THEO all displaced [3H]CHA and [3H]CGS 21680 specifically bound to brain membranes from Mullus surmuletus. Specific binding of both [3H]CHA and [3H]CGS 21680 was inhibited by GDPβS. For [3H]CHA the IC50 value was 2.5 ± 0.1 μM, while for [3H]CGS 21680 the IC50 value was 7.7 ± 0.3 μM. Our results indicate that the high affinity binding sites for [3H]CHA have some pharmacological characteristics of mammalian A1 adenosine receptors, while the binding sites for [3H]CGS 21680 appear to be virtually identical to the binding sites for [3H]CHA.  相似文献

Specific binding of the calcium antagonist [3H]verapamil to a microsomal fraction, a presumptive plasma membrane fraction and an intracellular membrane fraction of the phototactic unicellular green alga Chlamydomonas reinhardtii has been demonstrated. The specific activity of the plasma membrane marker enzyme K+-stimulated, Mg2+-dependent ATPase was severalfold higher in the upper (polyethylene glycol-rich) than in the lower (dextran-rich) phase, and the reverse was established for the marker enzymes of intracellular membranes such as cytochrome c oxidase for mitochondria and antimycin Aresistant NADPH-cytochrome c reductase for endoplasmic reticulum. Chlorophyll as a marker for thylakoid fragments was exclusively found in the lower phase. In the microsomal fraction two specific binding sites of [3H]verapamil were found at 22°C, one with higher and a second with lower affinity to [3H]verapamil. Separation of plasma membranes from intracellular membranes revealed that the highaffinity binding site is attributed to the plasma membrane fraction whereas the low-affinity binding site can be attributed to the intracellular membrane fraction. Specific binding to both separated membrane fractions is saturable and reversible. [3H]Verapamil binding to plasma membranes was not inhibited by the calcium channel blockers diltiazem and nifedipine. However, in the intracellular membrane fraction [3H]verapamil could be displaced by diltiazem but not by nifedipine. A radioreceptor assay using [3H]nitrendipine and rat cerebral cortical membranes, in conjunction with equilibrium dialysis, measures the unbound (free) level of nitrendipine in human sera. The sensitivity of the assay is 0.1–0.2 picomoles/ml and is linear from 4 × 10?11 to 4 × 10?9 M nitrendipine. Other dihydropyridine calcium channel antagonists may be measured using this assay if these compounds are used to generate the standard curve. Blank serum interferes with specific [3H]nitrendipine binding (24 percent inhibition per 20 μ1 serum) whereas serum dialysates do not. Total serum nitrendipine levels may be measured, but the sensitivity of the assay is decreased due to interference by serum. Nitrendipine is highly protein bound in serum (93 – 99 percent). This protein binding is essentially unchanged over a serum concentration from 1 to 100 ng/ml. This assay is suitable for pharmacokinetic and pharmacodynamic studies.  相似文献

The binding of [3H]phencyclidine ([3H]PCP) to a preparation of housefly thoracic muscle membranes was studied. Specific [3H]PCP binding saturated with both time and at concentrations of the radiolabeled ligand greater than 20 nM. One binding site with a KD of 10.7 nM and a Bmax of 3.9 pmol/mg protein was observed. Specific [3H]PCP binding was also readily reversible with a half-time of dissociation (t1/2) of 13.8 min and varied proportionately with tissue concentration. Of the drugs tested, specific [3H]PCP binding was inhibited by PCP analogs, antipsychotics, antidepressants, Ca2+ channel antagonists, and K+ channel blockers. [3H]PCP binding was unaffected by addition of carbamylcholine or L-glutamate in absence or presence of ATP, GTP, cAMP, or cGMP. Though the identity of the [3H]PCP binding protein in housefly muscle membranes is still unclear, it is more likely to be an ionic channel such as K+ or Ca2+ channels than a neurotransmitter receptor.  相似文献

The binding properties of the 1,4-dihydropyridine calcium channel antagonist, [3H](+)PN 200-110, were studied in rat cerebral cortical and cardiac homogenates (37°C, Krebs phosphate buffer). Specific binding of [3H](+)PN 200-110 was saturable, reversible, and of high affinity (Kd values are 35 and 64 pM for the cerebral cortex and heart, respectively). In parallel studies with [3H](+)PN 200-110, the dissociation constant of [3H]nitrendipine was 10–12 times higher. Substituted dihydropyridine calcium channel antagonists and agonists competitively inhibited specific [3H](+)PN 200-110 binding, but d-cis diltiazem enhanced and verapamil incompletely inhibited [3H](+)PN 200-110 binding in both the cerebral cortex and the heart. The effects of diltiazem and verapamil on [3H](+)PN 200-110 binding were due mainly to alterations in the dissociation constant (Kd), without alterations in the binding density (Bmax). The new [3H](+)PN 200-110 receptor binding assay is remarkable for its low degree of nonspecific binding as compared to [3H]nitrendipine at physiological temperatures. [3H(+)PN 200-110 is a useful ligand for the further analysis of the dihydropyridine binding sites associated with calcium channels.  相似文献

[3H]Nitrendipine was used to label sites in homogenates of bovine anterior and neurointermediate lobes of the pituitary gland. The amount of specific binding in the anterior lobe was 1.82 +/- 0.30 pmol/g wet weight of tissue and the KD was 1.44 +/- 0.02 X 10(-10) M. Preliminary experiments indicated a similar amount of binding in bovine neurointermediate lobe. In competition studies nimodipine and nisoldipine (two potent voltage-sensitive calcium channel blockers) displayed IC50 values of 1.6 and 6.8 X 10(-10) M, respectively. Verapamil and the verapamil-like calcium channel blockers D-600 and tiapamil competed in a complex manner for the [3H]nitrendipine specific binding to bovine anterior pituitary homogenates. Autoradiographical studies demonstrated specific [3H]nitrendipine binding sites distributed approximately equally in the anterior and posterior lobes, but not in the intermediate lobe of the rat pituitary. In general the properties of [3H]nitrendipine binding in the pituitary tissue resemble strongly the properties of [3H]nitrendipine binding in the brain which is believed to be to voltage-sensitive calcium channels. These results provide support for the hypothesis that calcium channels are involved in pituitary hormone secretion and that drugs that interact with calcium channels may modulate the secretory process directly at the level of the pituitary.  相似文献
