The present study shows the chemical profile and the in vitro properties (antioxidant and inhibition of nitric oxide production) of the Origanum heracleoticum L. (Lamiaceae). The ethanolic extract of the aerial parts is characterized by terpenes and fatty acids. The extract, with high total phenol and flavonoid content, showed a significant radical-scavenging activity (IC(50) value of 12.8 μg/mL) using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) test and an interesting antioxidant activity with the β-carotene bleaching test (IC(50) values of 12.9 and 14.1 μg/mL at 30 and 60?min of incubation, respectively). The test for the inhibition of NO production, performed using the murine monocytic macrophage RAW 264.7 cell line, showed that the extract had significant activity with an IC(50) value of 108.5 μg/mL. The cytotoxic effect of O. heracleoticum extract in presence of lipopolysaccharide (LPS) (1 μg/mL) was evaluated but found to be negligible.  相似文献

The present study shows for the first time the phenolic composition and the in vitro properties (antioxidant and inhibition of nitric oxide production) of Hypericum calabricum Sprengel collected in Italy. The content of hypericins (hypericin and pseudohypericin), hyperforin, flavonoids (rutin, hyperoside, isoquercetrin, quercitrin, quercetin and biapigenin) and chlorogenic acid of H. calabricum, have been determined. The ethyl acetate fraction from the aerial parts of H. calabricum exhibited activity against the radical 1,1-diphenyl-2-picrylhydrazyl (DPPH) with IC50 value of 1.6 jig/ml. The test for inhibition of nitric oxide (NO) production was performed using the murine monocytic macrophage cell line RAW 264.7. The ethyl acetate fraction had significant activity with an IC50 value of 102 jig/ml and this might indicate that it would have an anti-inflammatory effect in vivo.  相似文献

A combination of mineral nutrients and plant growth regulators should be assessed to improve crop performance under various abiotic stresses. There is a need to include plant growth regulators in fertilization regime of various crops along with essential mineral nutrients, especially when they are irrigated with polluted water with higher levels of heavy metals. The performance of pea was evaluated under cadmium (Cd) stress coupled with potassium (K) and jasmonic acid (JA) supplementation. The Cd stress (50 μM) was applied to soil (sandy loam) grown pea plants as basal dose after a month of sowing. The control and stressed plants were then supplemented with K (5 M), JA (0.5 mM) and their collective application along with control as distilled water. Cd stress showed a marked reduction in growth pattern, however, the collective supplementation sufficiently improved the growth pattern of stressed peas plants as evidenced by improvement in shoot length (cm), root length (cm), number of leaves per plant, leaf area (cm2), plant fresh and dry weight (gm). Potassium application under Cd stress significantly enhanced internodal distance (cm) while the number of seeds per pod and relative water contents remained nonsignificant. The applied treatment (JA + K) under Cd stress prominently improved enzymatic activities, which were measured as nitrate reductase activity (NRA), nitrite reductase activity (NiRA), superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT). Cd stress impacted the biochemical profile by enhancing antioxidant capacity (AC), antioxidant activity (AA), total phenols (TP), while reducing total soluble protein (TSP), chlorophyll ‘a’, chlorophyll ‘b’ and carotenoids. The combined application of JA and K under Cd stress enhanced AC, AA, TP, Chl a and b, TSP and carotenoids. The results indicate that foliar application of JA and K efficiently negated the harmful effects of Cd stress on peas.  相似文献

The mode of action of paraquat (1,1-dimethyl-4,4-bipyridinium) and the mechanism of resistance to it were studied in leaves of atrazine/paraquat co-resistant (R) and susceptible (S) biotypes of horseweed (Conyza canadensis) collected from Hungarian vineyards. The application of 0·5 mol m?3 paraquat by spraying onto the surface of the leaves of intact plants in the light rapidly led to typical symptoms of paraquat action in the initial period in both biotypes, i.e. inhibition of CO2 fixation, suppression of variable chlorophyll fluorescence (Fv), decrease of oxygen evolution and stimulation of ethane production. The inhibitory effect of paraquat in the S plants was irreversible, whereas it was transient in the R plants and those plants recovered gradually afterwards. The R plants recovered from the inhibitory effect of paraquat only in the light, and an increase in light intensity was found to have a pronounced effect on the recovery of Fv. The mechanism of resistance to paraquat in C. canadensis is discussed.  相似文献

The present study aims to investigate some physical attributes, total phenolics content, total flavonoids content, mineral composition, bioluminescence toxicity assay and antioxidant activity in terms of DPPH, HPS, TAC and FRAP assays in the kernel and pomace samples of six apricot cultivars grown in Balochistan, Pakistan. TFC and TPC determined by the AlCl3 and Folin-Ciocalteu assays in apricot kernel extracts of six cultivars varied from 1797.5 (Chagali) to 4778.9 (Badoghur) mg QUE/100 g DW and from 1750.0 (Chagali) to 5005.8 (Badoghur) mg GAE/100 g DW. Apricot kernels exhibited higher antioxidant activity than pomace; antioxidant activity in terms of IC50 in kernels ranged from 24.88 to 98.61 μg/ml for DPPH, 334.84 to 516.63 μg/ml for HPS, from 22.02 to 110.80 μg/ml for TAC and from 96.27 to 163.35 μg/ml for FRAP. The apricot kernels showed higher TPC, TFC, bioluminescence toxicity to V. logei and antioxidant activity than the pomace. The correlation analysis demonstrated substantial contributions of polyphenols and flavonoids to antioxidant assays. The sample type was the leading factor affecting the amounts of K, Na, Ca, Fe, and Mn in the tested samples; mineral contents were higher in pomace than kernels. The highest inhibition to V. logei was found in the kernels of Badoghur (IC50 = 1.61 mg/ml). The PCA analysis showed significant contributions of phenolic and flavonoid contents towards antioxidant bioluminescence toxicity assays. Our results suggest Badoghur, Shakarpara and Sardai kernels are rich sources of secondary metabolites and possess remarkable antioxidant and antiluminescence activity and can make a significant contribution to the treatment and prevention of chronic health problems.  相似文献

From the roots of Lindera strychnifolia, seven sesquiterpenes, named linderolides A-F and linderoline, were isolated together with six known compounds. The structures of linderolides A-F were elucidated to be eudesmane-type sesquiterpenes and linderoline was found to be a derivative of a germacrane-type sesquiterpene. Their structures were elucidated by means of spectroscopic evidence and that of linderolide A was confirmed by X-ray analysis. The inhibitory activity toward NO production was assayed using RAW264.7 cells with evaluating the cell growth by MTT method, linderolides C, D and F being found to be moderately active.  相似文献

Abstract

Immunoglobulin E (IgE) is involved in the onset of allergic reaction, and the suppression of IgE production leads to alleviation of allergic symptoms. We found that mango peel ethanol extract (MPE) significantly suppresses IgE production by human myeloma cell line U266 cells, suggesting that MPE has an anti-allergic effect by inhibiting the production of IgE. Although mangiferin is contained in mango, which suppresses IgE production by U266 cells, it was not contained in MPE. We investigated the suppressive effect of MPE in 2,4-dinitrofluorobenzene (DNFB)-induced allergic contact dermatitis model mice. The elevation of serum IgE level was significantly suppressed by oral administration of MPE. Intake of MPE also suppressed the expression level of IL-4 in the DNFB-challenged ears, suggesting that MPE suppresses the IL-4-mediated maturation into IgE-producing cells. Our findings indicate that MPE has a potential to alleviate the increase in serum IgE level that is feature of type I allergy.  相似文献

Abstract

Imbalance of free radicals over antioxidants in human body may result in oxidative damage to biomolecules (lipids, proteins, DNA) that causes severe chronic diseases. The aim of this proposed research was to examine the hypoglycemic and oxidative stress potential of fava bean (Vicia faba L.) seed extract. Acetone extract showed a significant effect on glucose uptake rate (77.28?±?2.42%) in yeast cells at 25?mM glucose concentration. Minimum glucose uptake rate was found to be 52.36?±?2.06% % by chloroform seed extract. Atomic force microscopy revealed that 3% hydrogen peroxide concentration results in roughness was found to be maximum (441?±?6.7?nm) and along with extract treatment showed a significant reduction in roughness (251?±?6.2?nm). Propidium iodide and DAPI staining showed apoptotic ratio as 0.40 (40?±?1.18%,) and 0.42 (42?±?1.16%) in hydrogen peroxide treated cell only as compared to other treatments. MTT assay showed that acetone extract had maximum survival rate (82.067%) and least survival rate was found in chloroform extract (70.48%). Hypoglycaemic potential and oxidative stress might be polyphenols (phenolics, flavonoids) present in seed extract or synergistic effect.  相似文献

Aim and objectiveHippophae rhamnoides is an edible, nutrient rich plant found in the northern regions of India. It belongs to the family Elaeagnaceae and is well known for its traditional pharmacological activities. The present study was aimed to investigate the antioxidant and neuroprotective activities of H. rhamnoides.MethodologyThe hydroalcoholic extract of H. rhamnoides was evaluated for free radical scavenging activity using DPPH, hydroxyl radical scavenging and ferric thiocyanate assays. In vitro neuroprotective activity was assessed on human neuroblastoma cell line-IMR32 against hydrogen peroxide (H2O2) induced cytotoxicity. The neuroprotective effect was determined by measuring the cell viability through tetrazolium dye MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) reducing assay and propidium iodide (PI) staining. Also the intracellular reactive oxygen species (ROS) activity was assessed using dichloro-dihydro-fluorescein diacetate (DCFDA) assay by flowcytometer.ResultsThe results of the study demonstrated that H. rhamnoides extract possesses potential free radical scavenging activity. The IC50 value for DPPH and OH radical scavenging assay was 70.92 μg/ml and 0.463 mg/ml, also the extract was also found to have considerable level of lipid peroxidation activity. The neuroprotective effect of H. rhamnoides was confirmed by its cell viability enhancing capacity against hydrogen peroxide induced cell cytotoxicity. The extract acted on IMR32 cells in a dose dependent manner as observed through PI and MTT assays. The percentage intracellular ROS activity was reduced by 60–70% in treated cells compared to H2O2 control.ConclusionThus the outcome of the study suggests that H. rhamnoides acts as a neuroprotectant against oxidative stress induced neurodegeneration.  相似文献

Dietary sunflower oil (SFO) was used to gradually replace fish oil (FO) in six diets (which also contained fish meal) for Atlantic salmon parr (initial mass: 21.7 g). The effect on growth performance, tissue fatty acid profiles and disease resistance was monitored after 63 days. At the conclusion of the trial, no significant differences were detected in growth between any of the feeds. Fatty acid composition of whole carcass, dorsal muscle and liver generally reflected that of the diets. Forty percent of the FO could be replaced by SFO before tissue 22:6n-3 was significantly reduced, although other essential and non-essential fatty acids were more susceptible to change. Significant differences were detected in cumulative mortality of Atlantic salmon challenged with Vibrio anguillarum at the trials conclusion, although this was not correlated to the inclusion level of SFO. Despite the changes observed to the tissue fatty acid profile, there was no significant effect on growth suggesting that SFO is a suitable alternative to FO in diets for Atlantic salmon parr when fish meal is also included.  相似文献
