Ustilago maydis, the causative agent of corn smut disease, is one of the most versatile model systems for the study of plant pathogenic fungi. With the availability of the complete genomic sequence there is an increasing need to improve techniques for the generation of deletion mutants in order to elucidate the functions of unknown genes. Here a method is presented which allows one to generate constructs for gene replacement without the need for cloning. The 5 and 3-regions of the target gene are first amplified by PCR, and subsequently ligated directionally to a marker cassette via two distinct Sfi I sites, providing the flanking homologies needed for homologous recombination in U. maydis. Then the ligation product is used as a template for the amplification of the deletion construct, which can be used directly for transformation of U. maydis. The use of the fragments generated by PCR drastically increases the frequency of homologous recombination when compared to the linearized plasmids routinely used for gene replacement in U. maydis.Communicated by G. NADP-glutamate dehydrogenase (NADP-GDH) and NAD-glutamate dehydrogenase (NAD-GDH) activities from Bipolaris maydis race T (ATCC 36180) were determined by measuring the change in absorbance at 340 nm of either reduced NADP or NAD in a reaction mixture of NH4C1, -ketoglutarate and a cell free extract of the fungus. NADP-GDH activity was high at 48 h, but low at 72 and 96 h when the fungus was incubated on a reciprocal shaker at 28 °C in a mineral salts medium containing 2 g/l glucose and 4 g/l Lasparagine. In contrast, in these cultures NAD-GDH activity was low at 48 h, but high at 72 and 96 h. At 72 and 96 h glucose was not detected in the culture medium. In addition, levels of ammonium and pH increased from 0.0 moles/ml and pH 5.8 at 48 h to 10.6 moles/ml and pH 7.2 at 72 h, and to 23.0 moles/ml and pH 8.4 at 96 h. Fungal mycelia were transferred after 48 h of incubation on media containing 2 g/l glucose and 4 g/l L-asparagine to fresh media containing 0, 2 or 5 g/l glucose with and without 4 g/l L-asparagine. Twenty-four h after transfer to fresh media containing 5 g/l glucose with L-asparagine or 2 or 5 g/l glucose without L-asparagine, NADP-GDH activity was high and NAD-GDH activity was low. Glucose was detected in the culture medium, ammonium was not detected and the pH remained unchanged or decreased. In contrast, 24 h after transfer to fresh media with 0 or 2 g/l glucose with L-asparagine and on media lacking glucose or L-asparagine, NADP-GDH activity was low and NAD-GDH activity was high. Glucose was not detected in the culture medium, ammonium levels were high and the pH increased. Thus, accumulation of ammonium and pH increases accompanying depletion of glucose in a L-asparagine medium could be related to a change in the capacity of B. maydis race T to assimilate and produce ammonium via pathways involving glutamate dehydrogenases.  相似文献

14-3-3 proteins are a family of highly conserved polypeptides that function as small adaptors that facilitate a diverse array of cellular processes by binding phosphorylated target proteins. One of these processes is the regulation of the cell cycle. Here we characterized the role of Bmh1, a 14-3-3 protein, in the cell cycle regulation of the fungus Ustilago maydis. We found that this protein is essential in U. maydis and that it has roles during the G2/M transition in this organism. The function of 14-3-3 in U. maydis seems to mirror the proposed role for this protein during Schizosaccharomyces pombe cell cycle regulation. We provided evidence that in U. maydis 14-3-3 protein binds to the mitotic regulator Cdc25. Comparison of the roles of 14-3-3 during cell cycle regulation in other fungal system let us to discuss the connections between morphogenesis, cell cycle regulation and the evolutionary role of 14-3-3 proteins in fungi.  相似文献

Thirty wild isolates belonging to five different locations in Mexico plus two laboratory strains of Ustilago maydis were characterized by restriction fragment length polymorphism (RFLP) analysis using 23 different clones as probes derived from a PstI library and two restriction enzymes. All loci analysed presented a high level of polymorphism, including one locus with thirty one different alleles. Geographical grouping of the populations was based on Nei's genetic distance and there was no correlation between genetic and geographic distances among these isolates. Our results suggest that DNA fingerprinting is a useful method for detecting genetic variation in populations of U. maydis. This work demonstrated that considerable genetic variation may be present within field populations of U. maydis.  相似文献

Summary. Ventricaria ventricosa and its relatives have intrigued cell biologists and electrophysiologists for over a hundred years. Historically, electrophysiologists have regarded V. ventricosa as a large single plant cell with unusual characteristics including a small and positive vacuole-to-outside membrane potential difference. However, V. ventricosa has a coenocytic construction, with an alveolate cytoplasm interpenetrated by a complex vacuole containing sulphated polysaccharides. We present a theory relating the coenocytic structure to the unusual electrophysiology of V. ventricosa. The alveolate cytoplasm of V. ventricosa consists of a collective of uninucleate cytoplasmic domains interconnected by fine cytoplasmic strands containing microtubules. The cytoplasm is capable of disassociating into single cytoplasmic domains or aggregations of domains that can regenerate new coenocytes. The cytoplasmic domains are enclosed by outer (apical) and inner (basolateral) faces of a communal membrane with polarised K+-transporting functions, stabilised by microtubules and resembling a tissue such as a polarised epithelium. There is evidence for membrane trafficking through endocytosis and exocytosis and so plasmalemma and tonoplast do not have fixed identities. Intra- and extracellular polysaccharide mucilage has effects on electrophysiology through reducing the activity of water and through ion exchange. The vacuole-to-outside potential difference, at which the cell membrane conductance is maximal, reverses its sign from positive under hypertonic conditions to negative under hypotonic conditions. The marked mirror symmetry of the characteristics of current as a function of voltage and conductance as a function of voltage is interpreted as a feature of the communal membrane with polarised K+ transport. The taxonomic position of the causal agent of fruit rot of olives was determined from fresh collections of the fungus from central Greece. In culture it formed two types of conidia, namely fusiform, hyaline, aseptate conidia typical of the genus Fusicoccum, and dark-walled, ovoid, ellipsoid or fusiform, 1–2 septate conidia that are not typically observed in Fusicoccum. A phylogenetic analysis based on ITS and EF1- sequences placed the fungus within the same clade as Fusicoccum aesculi, which is the anamorph of Botryosphaeria dothidea, and the type of the genus Fusicoccum.  相似文献

This paper provides an approach for optimizing the cell density (Xc) and dilution rate (D) in a chemostat for a Pichia pastoris continuous fermentation for the extracellular production of a recombinant protein, interferon (INF-). The objective was to maximize the volumetric productivity (Q, mg INF- l–1 h–1), which was accomplished using response surface methodology (RSM) to model the response of Q as a function of Xc and D within the ranges 150 Xc 450 g cells (wet weight) l–1 and 0.1 mD0.9 m (m=0.0678 h–1, the maximum specific growth rate obtained from a fed-batch phase controlled with a methanol sensor). The methanol and medium feed rates that resulted in the desired Xc and D were determined based on the mass balance. From the RSM model, the optimal Xc and D were 328.9 g l–1 and 0.0333 h–1 for a maximum Q of 2.73 mg l–1 h–1. The model of specific production rate (, mg INF- g–1 cells h–1) was also established and showed the optimal Xc=287.7 g l–1 and D=0.0361 h–1 for the maximum (predicted to be 8.92×10–3 mg–1 g–1 h–1). The methanol specific consumption rate (, g methanol g–1 cells h–1) was calculated and shown to be independent of the cell density. The relationship between and (specific growth rate) was the same as that discovered from fed-batch fermentations of the same strain. The approach developed in this study is expected to be applicable to the optimization of continuous fermentations by other microorganisms.  相似文献

Stable diatom populations in earthen ponds for fishpond effluent treatment supported fast rates of bivalve growth in integrated mariculture systems. However, when these ponds were lined with PVC plastic sheets to prevent seepage, the populations of benthic diatoms dwindled, and did not support, as before, a commercially acceptable rate of growth by oysters and clams. Experiments were undertaken to understand the problem and restore the diatom productivity of such ponds. Clones of Amphora luciae, A,cftenerrima, Cylindrotheca closterium, Navicula cf lineola, N.cf lenzii, N. salinicola, N. cf viminoides, and Nitzschia laevis were isolated in axenic culture from an earthen sedimentation pond. Their N, P, Si, and trace element requirements for growth in fully defined media, and in media formulated with mariculture effluent, were studied in axenic batch culture. In fully defined batch culture tests, most of the isolates achieved their highest density in media with 32 M P, 0.7 mM N, 20 M Fe, 10 nM–20 M Mn, 10–20 M Zn and Co, and 17.5 M Si. Enrichment by trace elements and Si stimulated the populations of these diatoms even in media based on nutrient-enriched mariculture effluents. However, in large flow-through agnotobiotic mesocosms (700 L), only Si enrichment was needed. Si concentration >100 M was required to promote the sustained blooms of diatoms in full-sized and commercial PVC-lined fishpond effluent treatment ponds (300 m2, 1 m depth). Except for Si, the requirements of the diatoms for micronutrients were apparently fully satisfied by the fishpond effluents (uneaten food and fish-waste). A molar ratio of 1:1 between Si and N is necessary to sustained dense diatom populations in the pond water. It is therefore recommended to enrich plastic lined mariculture effluent treatment/sedimentation ponds with Si, if the goal is to raise bivalves as a secondary crop.  相似文献

Background

Maternal or uniparental inheritance (UPI) of mitochondria is generally observed in sexual eukaryotes, however, the underlying mechanisms are diverse and largely unknown. Recently, based on the use of mutants blocked in autophagy, it has been demonstrated that autophagy is required for strict maternal inheritance in the nematode Caenorhabditis elegans. Uniparental mitochondrial DNA (mtDNA) inheritance has been well documented for numerous fungal species, and in particular, has been shown to be genetically governed by the mating-type loci in the isogamous species Cryptococcus neoformans, Phycomyces blakesleeanus and Ustilago maydis. Previously, we have shown that the a2 mating-type locus gene lga2 is decisive for UPI during sexual development of U. maydis. In axenic culture, conditional overexpression of lga2 triggers efficient loss of mtDNA as well as mitophagy. To assess a functional relationship, we have investigated UPI in U. maydis Δatg11 mutants, which are blocked in mitophagy.

Results

This study has revealed that Δatg11 mutants are not affected in pathogenic development and this has allowed us to analyse UPI under comparable developmental conditions between mating-compatible wild-type and mutant strain combinations. Explicitly, we have examined two independent strain combinations that gave rise to different efficiencies of UPI. We demonstrate that in both cases UPI is atg11-independent, providing evidence that mitophagy is not critical for UPI in U. maydis, even under conditions of strict UPI.

Conclusions

Until now, analysis of a role of mitophagy in UPI has not been reported for microbial species. Our study suggests that selective autophagy does not contribute to UPI in U. maydis, but is rather a consequence of selective mtDNA elimination in response to mitochondrial damage.

Summary. Filamentous fungi are an important group of tip-growing organisms, which include numerous plant pathogens such as Magnaporthe grisea and Ustilago maydis. Despite their ecological and economical relevance, we are just beginning to unravel the importance of endocytosis in filamentous fungi. Most evidence for endocytosis in filamentous fungi is based on the use of endocytic tracer dyes that are taken up into the cell and delivered to the vacuole. Moreover, genomewide screening for candidate genes in Neurospora crassa and U. maydis confirmed the presence of most components of the endocytic machinery, indicating that endocytosis participates in filamentous growth. Indeed, it was shown that in U. maydis early endosomes cluster at sites of growth, where they support morphogenesis and polar growth, most likely via endosome-based membrane recycling. In humans, such recycling processes to the plasma membrane involve small GTPases such as Rab4. A homologue of this protein is encoded in the genome of U. maydis but is absent from the yeast Saccharomyces cerevisiae, suggesting that Rab4-mediated recycling is important for filamentous growth. Furthermore, human Rab4 regulates traffic of early endosomes along microtubules, and a similar microtubule-based transport is described for U. maydis. These observations suggest that Rab4-like GTPases might regulate endosome- and microtubule-based recycling during tip growth of filamentous fungi. The responses to water stress of the bulk modulus of elasticity () and the apoplastic water fraction were examined using six sunflower cultivars of differing capacity for osmotic adjustment (OA). Water stress did not affect the partitioning of water between apoplastic (ca. 20%) and symplastic fractions in leaves which expanded during the exposure to stress in any genotype. Hence, no genotype-linked effects on either the buffering of cell water status during stress or on the estimates of bulk leaf osmotic potential could be expected. Genotypes differed in the degree of change in (estimated from pressure/volume [P/V] curves) and OA (estimated using both ln RWC/ ln o plots and P/V curves) induced by exposure to stress. In three genotypes increased significantly (p=0.05) as a consequence of stress, in another three change were small. OA was the only attribute of the three examined that could have contributed to turgor maintenance under stress. There was a strong negative association between leaf expansion and degree of OA across genotypes (r=–0.91) and a strong positive one between OA and (r=0.94). However all genotypes evidenced some degree of OA. These results are consistent with part of the genotype differences in OA being attributable to variations in leaf expansion during exposure to stress.  相似文献

Large immature and mature female bolas spiders of the genus Mastophora attract certain male moths by aggressive chemical mimicry of those moth species' sex pheromones. These older spiders capture moths by swinging a bolas (i.e., a sticky globule suspended on a thread) at the approaching male moths. Juvenile bolas spiders do not use a bolas, but instead use their first two pairs of legs to grab prey, which our field observations suggested were primarily nematocerous Diptera. Our field experiments over a 2-year period demonstrated that juvenile bolas spiders attract moth flies (Psychodidae), with each species apparently specializing on a particular prey species. In three experiments, sticky traps containing young Mastophora phrynosoma spiderlings consistently captured significantly more male Psychoda phalaenoides than were captured on traps containing spiderlings of other Mastophora species or no spiderlings (control traps). Results from two of the three experiments suggested that Mastophora hutchinsoni spiderlings attract male Psychoda trinodulosa. Only two of our experiments included Mastophora bisaccata and those produced contrasting results. In the first experiment, M. bisaccata appeared to attract P. phalaenoides, albeit in lower numbers than were captured on traps containing M. phrynosoma. However, in a second experiment the following year, M. bisaccata spiderlings attracted Psychoda satchelli, a species that had not been caught on any traps the previous year. As suggested by a systematist four decades ago, the taxon currently called M. bisaccata may consist of two or more sibling species, which could account for the contrasting results obtained from our two experiments involving M. bisaccata. This is the first reported evidence that, during early developmental stadia before these spiders attract moths, juvenile bolas spiders attract their prey.  相似文献

Infection of Hordeum vulgare L. by Pyrenophora teresand of Zea mays by Dreschslera maydis were characterized by green island formation, higher cytokinin levels and accumulation of metabolites in the infected areas. Higher cytokinin concentrations of the order 6-Y,Y-dimethylallylaminopurine > zeatinriboside > zeatin >dihydrozeatinriboside were detected at infection sites of susceptible hosts. By virtue of these cytokinins, infection sites may be acting as metabolic sinks helping proliferation of the pathogen. Existence of translocatory sinks at infection zones was confirmed from autoradiographic studies,where, accumulation of labeled metabolites was prominent at infection sites of susceptible hosts. Upon infection the lower cytokinin levels of resistant hosts decreased further with progress of infection. In the infected resistant hosts the concentrations of zeatin/zeatinriboside were the maximum among the four identified cytokinins. The pathogen is also capable of secreting cytokinins as evident from quantification of cytokinins in culture filtrate extracts using HPLC. Since detached leaves were used in the experiments the increase/decrease of various cytokinin levels may be attributed to pathogen influence. The increase in cytokinin levels in the susceptible host may be aiding the growth of the pathogen on one hand, while the decrease in the infected resistant host may signal the host to activate defenses against a potential pathogen at the early stage of infection.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

Mutual effects between the symbiotic bacteria of entomopathogenic nematodes, Photorhabdus luminescens and Xenorhabdus poinarii, and entomopathogenic fungi were investigated in vitro. A dual culture assay on nutrient agar supplemented with bromothymol blue and triphenyltetrazolium chloride (NBTA) medium revealed that P. luminescens is antagonistic to Metarhizium anisopliae, Beauveria bassiana, B. brongniartii and Paecilomyces fumosoroseus by inhibiting their growth and conidial production; the fungal growth was not inhibited by X. poinarii. In a second laboratory experiment, crude extract produced by M. anisopliae was tested for its activity against P. luminescens and X. poinarii. Crude extract from M. anisopliae was antibacterial to P. luminescens and X. poinarii at 1000 g/ml and inhibited their growth on NBTA, but had no effect at 100 or 10 g/ml. The influence of the crude extract of M. anisopliae on the dispersal of infective juveniles (IJs) of Heterorhabditis megidis and Steinernema glaseri was assayed on Sabouraud Dextrose Agar (SDA) plates. Results showed that the crude extract of M. anisopliae had no toxic effects even at highest concentration (1000 g/ml).  相似文献

The extracellular matrix (ECM) ofCochliobolus heterostrophus (anamorph:Bipolaris maydis) was made visible by gold/silver and FITC-lectin staining at different stages of germ tube development. A proteinaceous material was released from conidia as germ tubes began to emerge and continued to be released from the germ tube tip throughout elongation. A material that did not stain for protein was observed to surround germ tubes upon their elongation. At later stages of maturation, germ tubes were surrounded by a sheath of proteinaceous material. After 15 h of incubation, staining with the FITC-labeled Concanavalin A revealed that a carbohydrate material surrounded and extended between hyphae. The ECM extract was separated into two fractions which were shown by SDS-PAGE and HPLC analyses to consist of proteins and carbohydrates. The results demonstrate that the composition and physical structure of the ECM change over time. Thus, the ECM is not a static material. Rather, the components of the ECM appear to be laid down at different stages of fungal morphogenesis, possibly related to germ tube emergence, elongation, and maturation.  相似文献

Double-stranded RNA viruses of Ustilago maydis encode secreted killer toxins to which other cells of the same species and closely related species are sensitive. KP6 toxin consists of two polypeptides, and , produced from a single precursor preprotoxin. In this work, we cloned complementary DNA for the toxin-encoding segment of two of the KP6 nonkiller mutants NK3 and NK13 that secrete the and polypeptides, respectively. Both sequence analysis of the cDNA clones and in vitro translation of the toxin-encoding double-stranded RNAs showed that both mutants can produce full-length preprotoxins. Cys51 in is converted to Arg in NK3 and Thr25 and Lys42 in are changed to Pro and Arg, respectively, in NK13. Although and are encoded in a single prepropolypeptide, only the polypeptide is secreted by NK3 and only the polypeptide is secreted by NK 13. This differential expression of peptides from one precursor is a unique phenomenon. Neither of the nonsecreted polypeptides accumulated in the cytosol. The possible effects of these mutations on pre-protoxin folding and their consequences for toxin secretion are discussed.  相似文献

Microsomes from Kluyveromyces marxianus GK1005 examined by carbon monoxide difference spectroscopy showed no evidence of cytochrome P450, in contrast to microsomes isolated from a control strain of Saccharomyces cerevisiae. Benzo[a]pyrene produced a typical Type I-binding spectrum with microsomes of both yeasts, with K s values of 82 M (S. cerevisiae) and 70 M (K. marxianus). While aflatoxin B1 generated a typical Type I-binding spectrum with microsomes from S. cerevisiae (K s of 178 M), the toxin did not produce a recognisable binding spectrum with microsomes from K. marxianus.  相似文献

Interspecific ecophysiological differences in response to different light environments are important to consider in regeneration behavior and forest dynamics. The diurnal changes in leaf gas exchange and chlorophyll fluorescence of two dipterocarps, Shorea leprosula (a high light-requiring) and Neobalanocarpus heimii (a low light-requiring), and a pioneer tree species (Macaranga gigantea) growing in open and gap sites were examined. In the open site, the maximum net photosynthetic rate (Pn), photosystem II (PSII) quantum yield (; F/Fm), and relative electron transport rate (r-ETR) through PSII at a given photosynthetic photon flux density (PPFD) was higher in S. leprosula and M. gigantea than in N. heimii, while non-photochemical quenching (NPQ) at a given PPFD was higher in N. heimii. The maximum values of net photosynthetic rate (Pn) in M. gigantea and S. leprosula was higher in the open site (8–11 mol m–2 s–1) than in the gap site (5 mol m–2 s–1), whereas that in N. heimii was lower in the open site (2 mol m–2 s–1) than in the gap site (4 mol m–2 s–1), indicating that N. heimii was less favorable to the open site. These data provide evidence to support the hypothesis that ecophysiological characteristics link with plants regeneration behavior and successional status. Although Pn and stomatal conductance decreased at midday in M. gigantea and S. leprosula in the open site, both r-ETR and leaf temperature remained unchanged. This indicates that stomatal closure rather than reduced photochemical capacity limited Pn in the daytime. Conversely, there was reduced r-ETR under high PPFD conditions in N. heimii in the open site, indicating reduced photochemical capacity. In the gap site, Pn increased in all leaves in the morning before exposure to direct sunlight, suggesting a relatively high use of diffuse light in the morning.  相似文献
