S ummary . During growth of Clostridium sporogenes in tryptone-salt-peptone-glucose medium the pH value of the medium varies due to formation of acid and CO2 and to subsequent production of NH3. Glucose concentrations of 0·2, 0·5 or 1·0% result in increasing sporulation times and in spores of low, extremely high ( D 110 c . 80 min) and negligible heat resistance, respectively. When the pH value is maintained at 7, a reproducible sporulation time of a few hours is observed and the resulting spores have a heat resistance ( D 110) of 13 min, regardless of the glucose concentration.  相似文献

In strictly anaerobic conditions in a culture medium adjusted to pH 5·2 with HCl and incubated at 30°C, inocula containing < 10 vegetative bacteria of Clostridium botulinum ZK3 (type A) multiplied to give > 108 bacteria per ml in 3 d. Growth from an inoculum of between 10 and 100 spores occurred after a delay of 10–20 weeks. Citric acid concentrations of 10–50 mmol/l at pH 5·2 inhibited growth from both vegetative bacteria and spore inocula, a concentration of 50 mmol/l increasing the number of vegetative bacteria or of spores required to produce growth by a factor of approximately 106. The citric acid also reduced the concentration of free Ca2+ in the medium. The inhibitory effect of citric acid on vegetative bacteria at pH 5·2 could be prevented by the addition of Ca2+ or Mg2+ and greatly reduced by Fe2+ and Mn2+. The addition of Ca2+, but not of the remaining divalent metal ions, restored the concentration of free Ca2+ in the medium to that in the citrate-free medium. The inhibitory effect of citric acid on growth from a spore inoculum was only partially prevented by Ca2+. Citric acid (50 mmol/l) did not inhibit growth of strain ZK3 at pH 6 despite the greater chelating activity of citrate at pH 6 than at pH 5·2. The effect of citric acid and Ca2+ at pH 5·2 on vegetative bacteria of strains VL1 (type A) and 2346 and B6 (proteolytic type B) was similar to that on strain ZK3.  相似文献

Spores of Bacillus cereus T germinated and formed vegetative cells in Tryptone Soya broth (TSB), pH 9-0 and 7-4 at 30oC. Spores germinated but did not form vegetative cells when suspended in hen egg white (pH 9-0) supplemented with L-alanine and inosine. Using a split image eyepiece, the volumes of germinating spores in egg white were seen to increase as a result of increases in both length and breadth. In TSB at the same pH, the major volume increase resulted from a progressive increase in cell length. Egg white supplemented with L-alanine and inosine (pH 7-6 30oC) allowed limited outgrowth to occur but the vegetative cells differed in morphology to those in TSB. Fe(NH4)2(SO4)2.6H2O overcame the inhibition of outgrowth in egg white at pH 7–8 but not in egg white at pH 9-1. Solutions containing trace elements, growth factors and casamino acids could not replace iron in this respect. Sporulation occurred in egg white only when iron was present.  相似文献

Escherichia coli shifted from broth at external pH (pH0) 7·0 to pH0 7·0 broth plus glucose rapidly induced marked acid tolerance which also appeared, albeit to a lesser extent, plus maltose, sucrose or lactose. Tolerance appeared without the medium pH becoming acidic. Tolerance was most substantial when glucose was added at pH0 7·0 but was also appreciable at pH0 7·5, 8·0 and 8·5. Induction of tolerance by glucose was markedly reduced by cyclic AMP and essentially abolished plus NaCl or sucrose ; the induction process was also reduced but not fully inhibited by chloramphenicol, tetracycline and nalidixic acid. Glucose-induced organisms showed less acid damage to DNA and β-galactosidase and it is likely that this is because glucose induces a new pH homeostatic mechanism which keeps internal pH close to neutrality at acidic pH0. In conclusion, it is clear that glucose induces a novel acid tolerance response in log-phase E. coli at pH0 7·0 ; it is now known that induction of this response involves the functioning of extracellular induction components including an extracellular induction protein.  相似文献

We investigated the combined effects of pressure, temperature, pH, initial spore concentration and the presence of nisin on the survival of spores of Bacillus coagulans. Spores were more sensitive to pressure both at lower pH and at higher treatment temperatures. An additional 1.5-log10 reduction in cfu ml-1 was observed when pH was lowered from 7.0 to 4.0 during pressurization at 400 Mpa and 45°C. A 4-log10 cfu ml-1 reduction was observed when the temperature was increased from 25°C to 70°C during pressurization at 400 Mpa. The spores were sensitive to nisin at concentrations as low as 0.2 IU ml-1. At least a 6-log10 reduction was generally achieved with pressurization at 400 Mpa in pH 4.0 buffer at 70°C for 30 min when plated in nutrient agar containing 0.8 IU ml-1 nisin.  相似文献

Growth and proteinase production by Micrococcus sp. INIA 528 in a batch-operated laboratory fermentor were investigated, with trypticase soy broth as the basal medium for studies on optimum temperature, pH and medium composition. Maximum growth was recorded at 34°C and pH 715, whereas optimum temperature and pH for proteinase production were 31°C and pH 6.25. Maximum rate of enzyme production occurred during the late log and early stationary phases of growth. Addition of 5.0 g 1-1 yeast extract, 1.0 g 1-1 glucose, 1.0 g 1-1 MgSO4 or 1.0 g 1-1 K2HPO4 to basal medium resulted in a lower enzyme yield, but supplementation of basal medium with 2.5 g 1-1 (NH4)2SO4 increased enzyme production by 45%. A high initial biomass added to fresh broth supplemented with 2.5 g 1-1 (NH4)2SO4 only increased enzyme activity by 19%, compared to the maximum enzyme activity achieved with the standard inoculum.  相似文献

Lactobacillus plantarum was found to produce extracellular polygalacturonase (EC 3.2.1.15.). Maximum enzyme production was obtained in a medium containing 0.5% glucose and 1.5% low methyl-pectin as inducer at 27°C at an initial pH of 6.8. Enzyme production was strongly inhibited by 5 μmol/l NiCl2, 5 μmol/l CoCl2, 5 μmol/l CuSO4, and 10 μmol/l ZnCl2. MnSO4 and MgSO4 at 200 μmol/l and 50 μmol/l respectively seemed to enhance enzyme biosynthesis. The optimal pH and temperature for enzyme activity were 4.5 and 30°C respectively. Enzyme production in batch culture accompanied growth.  相似文献

The most critical period at onset of feeding in pikeperch Stizostedion lucioperca is short (<5 days at 20° C). The larvae are sensitive to prey density during the first week of exogenous feeding. First-feeding larvae of 6.5 mm total length ( LT ) needed prey densities of >585 prey l−1 to maintain mass (Cmaint), whereas 5 days older larvae of 7 mm L T Cmaint= 55 prey l−1 and for 11 mm L T larvae Cmaint<10 prey 1−1. Changes in specific growth rates for larvae of 7 and 11 mm were similar to a type-II functional response curve reaching a specific growth rate of 26 and 30% day−1, respectively, at 1000 prey l−1, whereas the 6·5 mm larvae showed a linear growth response reaching a specific growth rate of only 9% day −1 at 1000 prey l−1. The results suggest that prey density is a limiting factor, which might contribute to the high variation between year-class Strengths.  相似文献

The production and stability of pediocin N5p from Pediococcus pentosaceus , isolated from wine, were examined in grape juice medium. Maximum growth and higher titre (4000 U ml-1) were observed at a initial pH of 7·5 and 30°C. The activity of the inhibitory substance was stable between pH values from 2·0 to 5·0 at 4° and 30°C. At pH 10·0 it was completely inactivated. When submitted to 30 min at 80°, 100° and 115°C, maximal stability was observed at pH 2·0. Ethanol up to 10% did not affect pediocin activity at acid pH, nor did 40–80 mg 1-1 SO2, independently or combined with different ethanol concentrations, affect inhibitory activity.  相似文献

Antibiotically active producer mutants derived from the spontaneous degenerate parent Aspergillus versicolor N5 possessed not only mutual but also self-sensitive activity. The producer mutants, like the inactive parent, were only 3·5-fold less sensitive than the most sensitive unrelated organism, Trichophyton rubrum. The germination of spores is generally more sensitive than growth of vegetative cells. The antifungal spectrum of these mutual and self-sensitive mutants was fairly wide, unlike the host range specificity of bacteriocinogenic strains acting on organisms closely related to the producers. The self and mutual growth inhibitory principle was finally identified as the antibiotics mycoversilin and versilin in the case of producer mutants (N5)17 and N5T10(7), respectively, or Vx, an antibiotic of unknown molecular species, in the case of another producer mutant N5T10(8). Thus self-sensitivity, instead of self-resistance, of these antibiotically active mutant derivatives is a unique property among filamentous fungi in having simultaneously expressed two loci of contradictory functions, one for synthesis of, and the other for sensitivity towards, the same or related antibiotics.  相似文献

Nitrogen dioxide at 4 to 8 ml 1−1 breaks dormancy of dehulled, unimbibed seeds of red rice ( Oryza sativa L.) with minimum exposure times to the gas of 20 min or less. The response is independent of incubation medium pH following treatment. Germination is complete within 2 days but proceeds 4 to 6 h more slowly than non-dormant controls. The magnitude of response at a constant NO2 dose is dependent upon initial seed moisture. There was less than 40% germination at 9.6% initial moisture while germination was 95% at 12% initial moisture. NO2 treatment also breaks dormancy of imbibing, dehulled seeds, but the response is dependent upon the pH of the medium. Dormancy is broken at pHs close to the pKa of nitrous acid. Intact, unimbibed or imbibing seeds respond to NO2 treatment only if partially dry-afterripened prior to exposure.  相似文献

The Behaviour of a Food Poisoning Strain of Clostridium welchii in Beef   总被引:5,自引:5,他引:0  
S ummary : An inoculum of 105 spores of Clostridium welchii F2985/50 in meat survived steaming at 100° for 5 h, the number being reduced sevenfold for every hour of steaming. They also survived for at least 6 months in frozen meat stored at -5° and -20°, whereas vegetative cells died more rapidly at -5° than at -20°. In beef stored for 13 days at 1°, 5°, 10° and 15° there was no multiplication but a slow destruction of vegetative cells, but there was little change in the spore count. Slow multiplication occurred at 20° but at 25° and 37° growth was rapid. Only about 3% of the spores germinated without prior heat shock, so the majority failed to germinate in raw meat stored at any temperature, but did so once the meat had been heated. In meat which had been heated and allowed to cool almost all of the spores had lost their heat resistance.
It was found that the minimal growth temperature was related to pH and medium, so that meat with a pH higher than that used in these experiments (pH 5°7–5°8) would probably have a lower minimal growth temperature for these organisms and would thus be more susceptible to spoilage.  相似文献

Abstract Eight strains of obligately anaerobic, mesophilic, chitinolytic bacteria were isolated from the sediment of an estuarine environment. The isolates were rod-shaped, Gram-negative, and formed terminal spherical spores that swelled the sporangium. The major products from the fermentation of chitin were: acetate, ethanol, formate, CO2, H2 and ammonia. Growth of the isolates was possible at pH values ranging from 5.0–9.0. During the fermentation of chitin, N -acetylglucosamine accumulated in the culture fluids and was not metabolized. No organic compounds other than chitin and its oligomers could be demonstrated to support growth of the isolates. Hydrolysis of chitin proceeded at a relatively low rate and was incomplete. Approximately 65% of the initial amount of chitin was hydrolyzed during a period of 5–10 days. Supplementation of the medium with yeast extract, casamino acids or peptone did not enhance the rate of chitin hydrolysis, but reducing agents such as Na2S2O4 and Ti (III)-NTA markedly stimulated the rate of chitin fermentation.
The ecological implications of the high degree of substrate specialization are discussed.  相似文献

Six- to eight-week greenhouse experiments with independent control of pH and dissolved CO2 evaluated the potential for CO2 enrichment to stimulate the accumulation of Al, Fe, P and N in shoots of Vallisneria americana , particularly at pH 5. These minerals were provided only as they occurred in natural lake sediments.
2. The effect of CO2 enrichment at pH 5 v pH 7.3 on growth and tissue N concentration was also determined.
3. CO2 enrichment at pH 5 effected 5.5- and 7-fold increases in total shoot accumulation of Al and Fe, respectively. In a two-way factorial experiment, CO2 enrichment yielded 6- to 11-fold greater total shoot P accumulation in plants grown on less and more fertile sediments, respectively.
4. In a three-way factorial experiment, CO2 enrichment stimulated Vallisneria growth, especially at pH 5, and resulted in a 31–58% reduction in tissue [N] for different pH × sediment combinations. These are greater reductions than previously reported. It also increased total shoot N accumulation up to 6-fold, and there were significant interactions with pH and sediment source: the CO2 enrichment effect on shoot N accumulation was greater at pH 5 than at pH 7.3, and it was greater with the more fertile sediment at pH 5.
5. Water chemistry (pH and/or [CO2]) and sediment fertility thus both indirectly influenced the accumulation of sediment-derived minerals in macrophyte shoots within the water column.  相似文献

This study describes a broad host transformation protocol that enables the uptake of plasmid DNA into 10 different species of Bifidobacterium , some of which have never been transformed before. The vector pNC7 (4·9 kb) was used to optimize the electroporation protocol. Transformation efficiencies ranged from 3·6×10−1 to 1·2×105 transformations per μg DNA. The impact of growth medium composition and electric field strength on transformation efficiency were independently optimized. Electrocompetent cells were grown in Iwata medium broth enriched with ActilightRP 16%, harvested during the early exponential growth phase, and pulsed at 12·5 kV cm−1, 100 Ω and 25 μF.  相似文献

Aims:  To evaluate the impact of modified atmosphere packaging on in vitro growth of Aspergillus carbonarius and Aspergillus niger , and possible effects on ochratoxin A (OTA) biosynthesis.
Methods and Results:  Ochratoxigenic isolates belonging to the species A. carbonarius and A. niger were grown on a synthetic grapejuice medium (SNM) and packaged in combinations of controlled O2 (1% and 5%) and CO2 levels (0% and 15%), and in air as a control. Colony diameters were recorded every 3 days up to 21 days, and OTA was analysed after 7, 14 and 21 days. The greatest reductions in mycelial growth rate were observed at 1% O2 followed by 1% O2/15% CO2, whereas 5% O2 stimulated the growth of all isolates. OTA production by A. carbonarius and A. niger isolates was minimized at 1% O2/15% CO2 and 1% O2, respectively, after 7 days of incubation. Maximal OTA accumulation after 7 days was observed for all isolates in the control pack and at 5% O2.
Conclusions:  Of the atmospheres tested, only 1% O2 combined with 15% CO2 consistently reduced fungal growth and OTA synthesis by A. carbonarius and A. niger .
Significance and Impact of the Study:  Storage under modified atmospheres is unlikely to be suitable as the sole method for OTA minimization and grape preservation; other inhibitory factors are necessary.  相似文献

Mucor circinelloides LU M40 produced 12·2 mU ml−1 of linamarase activity when grown in a 3 l fermenter in the following optimized medium (g l−1 deionized water): pectin, 10·0; (NH4)2SO4,
1·0; KH2PO4, 2·0; Na2HPO4, 0·7; MgSO4.7H2O, 0·5; yeast extract, 1·0; Tween-80,
1·0, added after 48 h of fermentation. The purified linamarase was a dimeric protein with a molecular mass of 210 kDa; the enzyme showed optimum catalytic activity at pH 5·5 and 40 °C and had a wide range (3·0–7·0) of pH stability. The enzyme substrate specificity on plant cyanogenic glycosides was wide; the Km value for linamarin was 2·93 mmol l−1. The addition, before processing, of the fungal crude enzyme to cassava roots facilitated and shortened detoxification; after 24 h of fermentation, all cyanogenic glycosides were hydrolysed.  相似文献

Abstract Factors that may initiate the biosynthesis of acetoacetate decarboxylase were investigated in resting cells of Clostridium acetobutylicum . Linear acids from C1 to C4 were inducers, whereas branched acids and linear acids from C5 to C7 were not inducers of acetoacetate decarboxylase biosynthesis. Induction of acetoacetate decarboxylase was maximal at pH 4.8 in the presence of acid concentrations comparable with those found during fermentation. In growth conditions repression of acetoacetate decarboxylase biosynthesis was found. This fact explains that acetone production by Clostridium acetobutylicum occurs when growth slows down.  相似文献

Rainbow trout body mucus dialysed with acidified distilled water at pH 7,5 and 3 experienced ion depletion which was greatest at pH 3 and minimal between pH 7 and 5. Mucus Na+ loss is exacerbated in the presence of 1 mg I−1 aluminium as A12(SO4), at pH 5 and 7. Al2(SO4), causes greater depletion of Na+ from mucus than A1C13. A lethal level of zinc (2 mg 1−1) does not deplete mucus Na or K+, unlike a lethal level of aluminium (1 mg 1−1) at pH 7. The results are discussed in terms of the ionoregulatory role of mucus in heavy metal and acid toxicity.  相似文献
