为进一步探讨高温致神经管畸形的机制, 本文利用我们已经建立的高温致神经管畸形金黄地鼠模型, 在神经管发育的不同阶段, 用免疫组织化学法检测了高温对神经管及其周围间充质中神经生长因子(Nerve Grow th Factor, NGF) 和NGF受体 (trkA) 表达的影响。结果显示: NGF及其受体广泛分布于神经管及其周围间充质内, 并随胎龄增加而呈规律性变化, 高温处理后的胚胎神经管上皮及其周围间充质中NGF及其受体的免疫组织化学反应不同程度的减弱。结果提示, 神经管NGF及其受体的减少, 可能是高温致神经管畸形的一个重要因素  相似文献

A study was made of the role of the cotyledons, embryo orientation, surgical treatment, darkness, light, and autoclaved coconut milk on the growth of Pinus lambertiana Dougl. embryos in vitro. The embryos did not require an haustorial function of the cotyledons in vitro. Removal of the shoot meristem drastically altered the developmental physiology of the embryo and the function of the root meristem was severly inhibited. Positional effects on embryo growth were reversed by darkness. In the light vertical-inverted tube cultures displayed better growth than horizontal-inverted tube cultures, whereas in the dark the horizontal-inverted tube cultures displayed better growth than the ver ical-inverted tube cultures. Autoclaved coconut milk had no statistically demonstrable effect on embryo growth as measun d by the analysis of variance and Student-Newman-Keul's range test; however, the graphical analysis suggests that, in conjunction with the presence of the shoot meristem, there may be a slight beneficial response to autoclaved coconut milk.  相似文献

The stigma papillae in Gladiolus are of the “dry” type and are highly vacuolated cells with an organelle-rich peripheral cytoplasm. The cell wall of each papilla is overlain by a distinctive cuticle possessing an irregularly scalloped inner margin. Between the cell wall and cuticle is a layer of amorphous sub-cuticular material. Lipids are detected on the papilla surface. A pollen grain will hydrate and germinate only on a papilla and not on any other (non-papillate) portion of the stigma. The pollen tube penetrates the papilla cuticle, which is forced away from the papilla cell wall by sub-cuticular pollen tube growth. As the cuticle lifts away, the sub-cuticular material disperses. At the base of the papilla, the pollen tube grows onto the adaxial non-papillate surface of the stigma lobe. At this site, the cuticle has been lifted away from the underlying cells by release of a mucilaginous substance from the latter, and the pollen tube grows within this substance beneath the detached cuticle. The cytological features of Gladiolus papillae are compared with other stigma papillae described in the literature. Also, a review of the literature, as well as some of the findings of the present study, suggest that certain prevalent interpretations of dry stigma structure and function may be open to question.  相似文献

In some circumstances peritoneal infection causes distention and crowding of the bowel to such an extent that the effect is that of intestinal obstruction and it is impossible to introduce a tube far enough by physiologic routes to bring about deflation. The pressure upon the diaphragm then may cause pain in breathing, with the result that the patient does not fully expand the lungs and atelectasis develops in the lower lobes. In such circumstances surgical intervention to relieve the pressure in the bowel may be indicated.An ordinary urethral catheter introduced through an incision in the intestine cannot be extended far enough to relieve pressure in more than a few loops.A method of using a Foley catheter and of advancing it for greater distances inside the bowel was used in five cases with good results and without complications.  相似文献

The ultrastructure and composition of the synergids of Capsella bursa-pastoris were studied before and after fertilization. The synergids in the mature embryo sac contain numerous plastids, mitochondria, dictyosomes and masses of ER and associated ribosomes. Each synergid contains a large chalazal vacuole, a nucleus with a single nucleolus and is surrounded by a wall. This wall is thickest at the micropyle end of the cell where it proliferates into the filiform apparatus. At the chalazal end of the cell the wall thins and may be absent for small distances. The pollen tube grows into one of the two synergids through the filiform apparatus and extends one-third the length of the cell before it discharges. Following discharge of the pollen tube, mitochondria and plastids of the tube can be identified in the synergid as can hundreds of 0.5 μ polysaccharide spheres liberated by the tube. The method by which the sperm or sperm nuclei enter the egg or central cell is not known although an apparent rupture was found in the wall of the egg near the tip of the pollen tube. The second synergid changes at the time the pollen tube enters the first synergid. These changes result in the disorganization of the nucleus and loss of the chalazal wall and plasma membrane. Eventually this synergid loses its identity as its cytoplasm merges with that of the central cell.  相似文献

Tidal stresses are thought to be involved in maintaining airway patency in vivo. The present study examined the effects of normal stresses exerted by the lung parenchyma during tidal ventilation on recovery from agonist-induced airway constriction. In seven anesthetized dogs, one lung was selectively ventilated with a Univent endotracheal tube (Vitaid, Lewiston, NY). Airway tone was increased either transiently (intravenous bolus) or continuously (intravenous infusion) with methacholine (MCh). During one-lung ventilation, changes in the airway size of both lungs were measured for up to 40 min during recovery from constriction by using high-resolution computed tomography. After recovery to baseline, the alternate lung was ventilated, and the protocol was repeated. The absence of tidal stresses led to an attenuated recovery from either transient or steady-state airway constriction. The effectiveness or lack thereof of normal tidal stress in stabilizing airway size may be one factor that contributes to the lack of reversal with tidal breathing and deep inspiration seen in asthmatic subjects.  相似文献

Patients with prion diseases can live for long periods of time in a state of akinetic mutism given appropriate management of their symptoms. To study symptom support in these cases, we performed gastrostomies on 3 patients with V180I genetic Creutzfeldt-Jakob disease (CJD) who had become akinetic and mute, and compared them to 14 other similar patients being fed by tube. In the 3 gastrostomy cases, there were no direct complications due to the gastrostomy or tube feeding, nor were there episodes of discontinuation of tube feeding or initiation of continuous drip infusion due to severe complications. Antibiotics were administered for mild infections, a complication of CJD, with 0.2% and 8.8% of the total time after gastrostomy being used for intravenous or transluminal administration, respectively. We compared the present patient series with that of our previous report statistically, and found that patients undergoing gastrostomy required significantly fewer discontinuations of tube feeding than those who did not. No significant difference in antibiotic administration was found between groups, however. It is our conclusion that gastrostomy should be allowed for symptom support in akinetic patients with prion disease, but adequate informed consent must be provided to the patient's family.  相似文献

Small-animal intubation is often necessary during inhalation anesthesia to allow steady-state conditions for large operations and in vivo experiments in all fields of experimental surgery. In rats, placing an orotracheal tube is technically difficult primarily because of the small size of the subject and the lack of equipment specifically designed for this task. We describe a simple rat intubation technique in which the animal is suspended in dorsal recumbency on an inclined metal plate. The animal, anesthetized with ether, is fixed to a 70 degrees-inclined metal plate in a dorsal position by means of a Mersilene ribbon hooked around the upper incisors. This method of positioning the animal is the most important step in the intubation process and further facilitates the technique already described by other authors. A human otoscope was used as a laryngoscope, intubation was performed using the Seldinger technique, and a 14-gauge intravenous catheter served as an endotracheal tube. This inexpensive technique is quickly learned and can be used in any laboratory. Safe and reliable airway management can thus be achieved, permitting in vivo examinations and operations.  相似文献

陕南晚震旦世高家山生物群中骨骼化石的电镜扫描显示,Sinotubulites levis壳壁中具有丰富的磷酸盐化球状和丝状体,可分为3种不同类型的微生物组构:1)由相互缠绕的丝状体所组成的藻席状组构;2)由外表面成莓球状,内部中空的球状体紧密堆叠所形成的组构类型;3)由表面极为光滑的球状体堆叠所形成的组构。它们可能分别代表不同类型的矿化蓝藻,依此推断微生物在S.levis壳壁磷酸盐化过程中起着重要作用。在共生的Sinotubulites baimatuoensis和Cloudina壳壁上,极少或完全不存在这些磷酸盐化组构表明微生物的磷酸盐化作用具有专属性,可能由壳壁微结构所决定,具多层状壁的S.levis可能与具许多有机质纹层的多毛类壳壁关系密切。  相似文献

Laryngeal spasm is a problem constantly confronting the anesthetist. It can be serious and may produce fatal cerebral or cardiac complications. Etiologic agents include primary vagal hypertonicity, anoxemia, and painful stimulation of whatever source.Laryngeal spasm must be differentiated from simple obstruction by the tongue or foreign bodies, epiglottic impaction, laryngeal edema, tracheal spasm and collapse, and bronchial spasm.Proper checking of the patient before anesthesia and adequate premedication with atropine or scopolamine are preventive measures of great value. Once spasm has developed the etiologic agent should be removed if possible. Other measures include intravenous administration of atropine or curare, tracheal intubation, and tracheotomy.  相似文献

A duodenal tube was introduced into the duodenum of the fasted baboon via the nasal passage. After the application of vacuum for several minutes, the baboon received an intravenous injection of pancreozymin and cholecystokinin which caused contraction of the gall bladder. Even mild head injuries may cause cerebral swelling and vascular alterations, including vasoparalysis and increased vascular permeability. The severer the injury, the more pronounced these changes. They may cause death.Maintenance of adequate oxygen supply to the brain is the most effectual means of preventing or reducing the severity of this secondary effect of cerebral trauma, and the preservation of a good respiratory exchange is therefore essential in a patient who is comatose as a result of a head injury.The commonly employed measures such as an oral airway, suction and oxygen therapy may be tried first, but if the patient continues to have respiratory distress or cyanosis, an intratracheal tube should be employed for 24 hours; and then if there is no improvement, tracheotomy should be carried out. If the injury is severe, tracheotomy from the beginning may be a life-saving measure.  相似文献

These experiments serve to show that neutral salts in amounts considerably below those commonly employed in culture solutions may be very injurious to pollen. It has been found, for example, that NaCl, one of the least toxic salts tried, excepting CaCl2, added to a sucrose solution in a concentration of 0.0002 M, or about 11 parts per million, reduces the growth of sweet pea pollen tubes 15 per cent. When it is considered that MgCl2 and BaCl2 are about fifteen times as toxic as NaCl it becomes evident that the susceptibility of pollen tubes to injury by these substances amounts virtually to hypersensitiveness. On the other hand calcium salts in concentrations ranging from 0.02 to 0.002 M markedly enhance the growth of sweet pea pollen tubes. MgCl2 has a similar action in the case of Nicotiana. Calcium, moreover, exerts a strong protective action in the presence of the injurious monovalent cations Na and K. So far as can be determined by microchemical means these salts do not alter the wall of the pollen tube; presumably, their effect is on the protoplast itself. In the light of recent experimentation (Osterhout) with other forms better adapted to precise investigation of these phenomena it seems probable that the explanation of the facts presented here lies in changes brought about in the permeability of the cells. Since several gaps exist in our evidence, however, conclusions drawn at this time must necessarily be provisional. The highly injurious action manifested by the cations of several of the salts used indicates that they penetrate the protoplast very rapidly. Possibly in pure sucrose cultures, exosmosis is a limiting factor in pollen tube growth. The addition of salts of calcium or magnesium may favor development by retarding or preventing this outward diffusion. The protective effect of calcium in the presence of the toxic cations K and Na is best interpreted on the assumption that the entry of these latter into the protoplast is retarded by the calcium. The mode by which hydrogen ion concentration affects pollen tube growth is largely a matter of speculation. It has previously been been shown by Brink that the time relations of the growth process simulate those of an autocatalytic reaction. It has been demonstrated also that elongation of the tubes in artificial media is related to the digestion of the reserve food materials contributed by the pollen grain. In the case of the sweet pea these stored substances are largely fats and their hydrolysis may constitute the most important chemical reaction in growth. If, as seems not improbable, the other reactions involved wait upon this one, it is the "master reaction" according to Robertson''s hypothesis. If this conception really applies to the case in hand as outlined, the effect of the concentration of hydrogen ions on growth may be a direct one. It is known that the action of the fat-splitting enzyme lipase is favored by a certain amount of free acid. The maximum rate of germination of the pollen and the greatest amount of growth of the pollen tubes occur at pH 6.0. This may be due in large part to the immediate effect of this concentration of hydrogen ions upon the digestion of the reserve food.  相似文献

Techniques are described for detection of pollen grain and pollen tube nuclei using the fluorescent DNA probes ethidium bromide or Hoechst 33258, in conjunction with the aniline blue fluorochrome sirofluor, which stains the callose component of pollen tube walls and plugs. The DNA probes, which may be used either as vital stains or following fixation, permit discrimination between vegetative and generative or sperm nuclei. Double staining with sirofluor allows location of nuclei within pollen tubes grown in vitro, and when used after pollination enables the viewer to discriminate between nuclei within the pollen tube vs. nuclei of the pistil tissue.  相似文献

An equation is derived for the calculation of a constant which, experimental results indicate, may be a more reliable index of the antiproteolytic activity of serum than those equations hitherto used. 2. (a) Intramuscular administration of trypsin resulted in a slow rise in the antiproteolytic activity of the serum, followed by a lesser decline. (b) Intravenous administration resulted in no appreciable variation. (c) Oral administration resulted in a rapid rise, which was sustained during the period of administration. (d) Intramuscular, intravenous, or oral administration of denatured trypsin resulted in no appreciable variation. (e) The extent of the local necrosis following subcutaneous injection of trypsin varied inversely with the antiproteolytic activity of the serum. 3. The experimental results indicate that the products of protein hydrolysis in the intestine and parenterally are an important factor in the antiproteolytic activity of the serum. They also indicate that antibodies to trypsin are not an important factor in the antiproteolytic activity of the serum.  相似文献

While the number of experiments performed is perhaps somewhat limited the results obtained were quite definite and warrant the following conclusions. The toxicity of normal blood for living plant protoplasm as studied on the growth of Lupinus albus seedlings is definitely influenced by various radiations. Ultra-violet rays produce no effect on normal blood or may even render it slightly less toxic. Roentgen rays render normal blood more toxic. The toxicity is greater in the case of the blood plasma as compared with the blood cells and a more toxic effect is produced with the Coolidge tube as compared with the gas tube. Radium emanations in the few experiments performed produced changes very much the same as those given by the x-rays.  相似文献

Developmental phases surrounding the processes of gametic delivery and fusion were examined ultrastructurally in the reduced megagametophyte of Plumbago zeylanica, which lacks synergids. Gametic delivery occurs at the end of pollen tube growth and results in deposition of two male gametes, a vegetative nucleus, and a limited amount of pollen cytoplasm between the egg and central cell. Discharge of these materials from the tube is accompanied by loss of inner and outer pollen tube plasma membranes, loss of sperm-associated cell wall components, and disruption of the formerly continuous cell wall between the egg and central cell. The dispersion of egg cell wall components directly exposes female reproductive cell membranes to the unfused male gametes and pollen tube without disrupting gametic cell plasma membranes. Presence of unfused sperms within the female gametophyte appears to be a transitory phenomenon, lasting less than 5 min at the end of over 8½ hr of pollen tube growth. At the time of gametic deposition, plasma membranes of unfused sperm cells become directly appressed to plasma membranes of both the egg and central cell. Gametic fusion is initiated by a single fusion event between membranes of participating male and female cells, which is rapidly followed by subsequent, secondary fusion events between the same two cells at different locations along their surface. Gametic fusion results in the transmission of male gamete nuclei with co-transmission of nearly the entire sperm cytoplasmic volume and organellar complement, and it is possible to identify heritable male cytoplasmic organelles within both the incipient zygote and endosperm. Paternally originating plastids may be distinguished from maternal plastids by differences in morphology and staining characteristics, whereas paternal mitochondria may be distinguished from maternal mitochondria by populational differences in mitochondrial size which are statistically significant. Such observations further indicate that transmitted paternal mitochondria seem to remain viable, as judged by their ultrastructural appearance, and are transmitted exclusively by sperm cytoplasm rather than discharged pollen cytoplasm. The presence of anucleate, membrane-bounded cytoplasmic bodies between the egg and central cell are identifiable on the basis of their enclosed organelles and indicate that fragmentation of a small amount of the sperm cytoplasm associated with the vegetative nucleus commonly occurs. The presence and identification of sperm cytoplasmic organelles and associated membranes within female reproductive cells following gametic transmission represents strong evidence in support of the cellular basis of nuclear and cytoplasmic transmission during sexual reproduction in Plumbago.  相似文献
