【目的】从印染污水处理厂污泥中筛选孔雀石绿脱色菌,并分析比较染料脱色前后毒性。【方法】采用平板稀释涂布法筛选分离纯化菌株,通过扫描电镜、16S r RNA基因序列分析对菌株进行初步鉴定,采用蚕豆根尖微核毒性试验和小球藻毒性试验对染料脱色前后毒性进行分析。【结果】从印染污水处理厂污泥中筛选出一株高效孔雀石绿脱色菌。经过16S r RNA基因序列分析,鉴定该菌株为Klebsiella sp.JD。菌株的菌落边缘规则,圆形,灰白色粘稠,以接种环挑之,易拉成丝,表面光滑。通过对比小球藻在脱色前后的生长率和抑制率以及蚕豆微核率和污染指数分析出脱色后的染料毒性大大降低。【结论】筛选得到一株新的孔雀石绿脱色菌,该菌降解效率高,可降低染料毒性,对修复染料污染的水体有一定的现实意义。  相似文献

The DnaB helicase from Bacillus stearothermophilus (DnaBBst) was a model protein for studying the bacterial DNA replication. In this work, a non-radioactive method for measuring ATPase activity of DnaBBst helicase was described. The working parameters and conditions were optimized. Furthermore, this method was applied to investigate effects of DnaG primase, ssDNA and helicase loader protein (DnaI) on ATPase activity of DnaBBst. Our results showed this method was sensitive and efficient. Moreover, it is suitable for the investigation of functional interaction between DnaB and related factors.  相似文献

The classical malachite green (MLG) assay of phosphate, which added MLG after molybdate to the acidified reaction solutions of phosphate, tolerated interference from papaverine, sildenafil, and some similar hydrophobic amines. Resonance Rayleigh scattering signals, the alleviation of interference by poly(vinyl alcohol), and the precipitation of some yellow complexes supported that the irreversible aggregation of the complexes of a hydrophobic amine of interference and phosphomolybdate reduced the amounts of phosphomolybdate accessible to MLG and caused the interference. By adding MLG before molybdate to the acidified reaction solutions of phosphate, the complexes of phosphomolybdate and MLG were preferentially formed before the complexes of phosphomolybdate and such a hydrophobic amine effectively aggregated; thereby, an improved MLG assay of phosphate with the resistance to common hydrophobic amines was developed. Using the improved MLG assay of phosphate and a phosphatase to release phosphate from AMP, a spectrometric method successfully estimated the half-inhibition concentrations of papaverine on the recombinant human cyclic nucleotide phosphodiesterase (PDE) isozyme 4 and the mixture of PDE isozymes from rabbit brain. Therefore, the improved MLG assay of phosphate was a favorable and universal technique for developing spectrometric methods for characterizing and screening inhibitors of enzymes that release phosphate during their actions.  相似文献

The inhibitory effects of the cationic triarylmethane (TAM+) dyes, pararosaniline (PR+), malachite green (MG+), and methyl green (MeG+) on human plasma cholinesterase (BChE) were studied at 25 degrees C in 100 mM Mops, pH 8.0, with butyrylthiocholine as substrate. PR+ and MG+ caused linear mixed inhibition of enzyme activity. The respective inhibitory parameters were K(i) = 1.9 +/- 0.23 microM, alpha = 13 +/- 48, beta = 0 and K(i) = 0.28 +/- 0.037 microM, alpha = 23 +/- 7.4, beta = 0. MeG+ acted as a competitive inhibitor with K(i) = 0.12 +/- 0.017 microM (alpha, infinity, beta, not applicable). The K(i) values were within the same range reported for a number of ChE inhibitors including propidium ion, donepezil, and the phenothiazines, suggesting that TAM+s are active site ligands. On the other hand, the alpha values failed to correlate with values previously reported for a number of ChE inhibitors. It appears that mixed inhibition is the combined result of more than one type of binding and S-I interference. The impact of ligands at the choline-specific and peripheral anionic sites (or, possibly, accessory structural domains) on BChE activity needs to be studied in further detail.  相似文献

A sensitive analytical procedure for the determination of residues of leucomalachite green (LMG)-malachite green (MG) and leucogentian violet (LGV)-gentian violet (GV) in catfish or trout tissue is presented. Frozen (−20°C) fish fillets were cut into small pieces and blended in a Waring blender. A 20-g amount of homogenized fish tissue was extracted with acetonitrile-buffer, partitioned against methylene chloride, and cleaned up on tandem neutral alumina and propylsulfonic acid cation-exchange solid-phase extraction cartridges. Samples of 100 μl (0.8 g equiv.) were chromatographed isocratically in 10 min using an acetonitrile-buffer mobile phase on a short-chain deactivated (SCD) reversed-phase column (250×4.6 mm I.D.) in-line with a post-column PbO2 oxidation reactor. The PbO2 post-column reactor efficiently oxidized LMG to the chromatic MG, and LGV to the chromatic GV permitting visible detection at 588 nm for all four compounds. Linearity was demonstrated with standards over the range of 0.5–50 ng per injection. Recoveries of LMG, MG, LGV and GV from catfish tissues fortified at 10 ng/g were 75.4±3.0, 61.3±4.1, 72.6±3.7 and 87.9±2.5, respectively, while trout tissues fortified at 10 ng/g yielded recoveries of 82.6±2.3, 48.6±1.8, 72.1±2.1 and 83.8±4.6 (mean±S.D., N=4), respectively.  相似文献

【目的】评价裂褶菌cfcc7252菌株降解孔雀石绿(Malachite Green,MG)的能力及其潜在的应用价值。【方法】采用单因子液体培养实验,研究了通气、p H、温度、碳源和氮源种类及浓度、金属离子、盐度、染料浓度对该菌降解效果的影响;采用平皿培养实验,利用植物种子萌发和微生物抑菌实验对降解产物进行毒性测试。【结果】研究结果表明,裂褶菌cfcc7252菌株在好氧和厌氧条件下均能高效降解MG。该菌在10.0 g/L葡萄糖,5.0 g/L酵母浸粉,0.01 mmol/L Zn2+,p H为4.0的液体培养基中培养36 h,对350 mg/L的MG降解率达67.8%;连续降解7次后,其降解率还能保持在95.4%以上。此外,该菌在盐度低于10.20%时,其对MG的降解率均达到98%以上。对植物、微生物的毒性测试结果表明,MG降解产物对红豆、豌豆等植物、金黄色葡萄球菌、枯草芽孢杆菌和铜绿假单胞杆菌等微生物基本没有毒性。【结论】裂褶菌cfcc7252菌株在处理以MG为主的染料废水时具有很强的应用潜力。  相似文献

Electrochemically active composite film that contains multiwalled carbon nanotubes (MWCNTs), Nafion (NF), and poly(malachite green) (PMG) has been synthesized on glassy carbon electrode (GCE), gold, and indium tin oxide (ITO) electrodes by potentiodynamic method. The presence of MWCNTs in the composite film (MWCNT–NF–PMG) enhances the surface coverage concentration (Γ) of PMG by fivefold. Similarly, an electrochemical quartz crystal microbalance study revealed enhancement in the deposition of PMG at MWCNT–NF film when compared with bare and only NF modified electrodes. The surface morphology of the composite film was studied using atomic force microscopy, which revealed that the PMG incorporated on MWCNT–NF film. The composite film exhibited enhanced electrocatalytic activity toward the mixture of biochemical compounds catechol and quinol. The electrocatalytic responses of analytes at MWCNT–NF–PMG composite film were measured using both cyclic voltammetry (CV) and differential pulse voltammetry (DPV). From electrocatalysis studies, well-separated voltammetric peaks were obtained at the composite film for catechol and quinol with a peak separation of 147 mV. The sensitivity values of the composite film toward catechol and quinol by the DPV technique were 0.4 and 3.2 mA mM−1 cm−2, respectively, which are higher than the values obtained by the CV technique. Similarly, the above-mentioned values are better than the previously reported electroanalytical values for the same analytes.  相似文献

We studied the decolorization of malachite green (MG) by the fungus Cunninghamella elegans. The mitochondrial activity for MG reduction was increased with a simultaneous increase of a 9-kDa protein, called CeCyt. The presence of cytochrome c in CeCyt protein was determined by optical absorbance spectroscopy with an extinction coefficient (E550-535) of 19.7 ± 6.3 mM−1 cm−1 and reduction potential of + 261 mV. When purified CeCyt was added into the mitochondria, the specific activity of CeCyt reached 440 ± 122 μmol min−1 mg−1 protein. The inhibition of MG reduction by stigmatellin, but not by antimycin A, indicated a possible linkage of CeCyt activity to the Qo site of the bc1 complex. The RT-PCR results showed tight regulation of the cecyt gene expression by reactive oxygen species. We suggest that CeCyt acts as a protein reductant for MG under oxidative stress in a stationary or secondary growth stage of this fungus.  相似文献

2-Cys peroxiredoxin (Prx) is the major subgroup of a family of Prx enzymes that reduce peroxide molecules such as hydrogen peroxide (H2O2). 2-Cys Prxs are inactivated when their active site cysteine residue is hyperoxidized to sulfinic acid. Sulfiredoxin (Srx) is an enzyme that catalyzes reduction of hyperoxidized 2-Cys Prxs in the presence of ATP, Mg2+, and thiol equivalent. Therefore, Srx activity is crucial for cellular function of 2-Cys Prxs. The method currently available for the determination of Srx activity relies on immunoblot detection using antibodies to hyperoxidized enzymes. Here we introduce a simple quantitative assay for Srx activity based on the colorimetric determination of inorganic phosphate released in Srx-dependent reduction of hyperoxidized Prx using the malachite green. The colorimetric assay was used for high-throughput screening of 25,000 chemicals to find Srx inhibitors.  相似文献

In this paper, waste sawdust was functionalized by monosodium glutamate for improving its cationic sorption capacity. The functionalized sawdust (FS) and crude sawdust (CS) were compared for their malachite green (MG) sorption behaviors with a batch system. The effects of various experimental parameters (e.g. initial pH, sorbent dose, dye concentration, contact time, and temperature etc.) were investigated and the sorption kinetic and thermodynamic characteristics were understood. The MG removal ratios on FS and on CS increased with increasing initial pH and came up to the maximum value beyond pH 6 for FS and pH 8 for CS, respectively. The ratio of sorbed MG kept above 95% for 250 mg/l of MG solution when 2.0 g/l or more of FS was used. The MG removal percentage decreased more on CS than on FS with increasing initial MG concentration. The isothermal data of MG sorbed on FS and on CS followed the Langmuir model. By functionalizing, the sorption capacity (Q(m)) of sawdust for MG was increased from 85.47 to 196.08 mg/g and the sorption equilibrium time of MG was shortened from 23 to 4.5 h. The MG sorption processes on FS and on CS followed the pseudo-second-order rate kinetics. The sorptions of MG on FS and on CS were spontaneous and exothermic processes and lower temperatures were favorable for the sorption processes.  相似文献

In this study, we investigated the efficacy of phenolic extract of wheat bran and lignin-related phenolic compounds as natural redox mediators on laccase-mediated transformation of malachite green (MG) using purified laccase from the white-rot fungus Ganoderma lucidum. G. lucidum laccase was able to decolorize 40.7% MG dye (at 25 mg l−1) after 24 h of incubation. Whereas, the addition of phenolic extract of wheat bran enhanced the decolorization significantly (p < 0.001) by two- to threefold than that of purified laccase alone. Among various natural phenolic compounds, acetovanillone, p-coumaric acid, ferulic acid, syringaldehyde, and vanillin were the most efficient mediators, as effective as the synthetic mediator 1-hydroxybenzotriazole. Characterization of MG transformation products by HPLC, UV–Vis, and liquid chromatography-mass spectrometry-electrospray ionization analysis revealed that N-demethylation was the key mechanism of decolorization of MG by laccase. Growth inhibition test based on mycelial growth inhibition of white rot fungus Phanerochaete chrysosporium revealed that treatment with laccase plus natural mediators effectively reduced the growth inhibitory levels of MG than that of untreated one. Among all the tested compounds, syringaldehyde showed the highest enhanced decolorization, as a consequence reduced growth inhibition was observed in syringaldehyde-treated samples. The results of the present study revealed that the natural phenolic compounds could alternatively be used as potential redox mediators for effective laccase-mediated decolorization of MG.  相似文献

Triphenylmethane dyes are considered to be one of the most recalcitrant pollutants in the environment. Malachite Green (MG) was successfully removed from aqueous solution by Pseudomonas sp. DY1 immobilization with Aspergillus oryzae. Inhibition test in the presence of sodium azide and nystatin indicated that A. oryzae was a natural immobilization reagent, and removal of MG by the immobilized cell pellets was attributed to the biodegradation by Pseudomonas sp. DY1. Optimum conditions of immobilization for maximum biodegradation were obtained using Taguchi design at 37 °C, inoculation size of Pseudomonas sp. DY1 (dry cell mass) 0.01 g, of A. oryzae (spore number) 1.0 × 109, initial pH 6.5. Decolorization and biodegradation of MG by immobilized pellets under optimum conditions were 99.5% and 93.3%, respectively. Immobilized pellets exhibited more than 96% decolorization after 16 days in batch condition, indicating it had stable and high biodegradation capabilities when immobilized for long-term operation.  相似文献

The sorption of heavy metals ions by immobilized Trichoderma viride biomass in a packed-bed column was studied. Fungal biomass T. viride was immobilized to Ca-alginate used for removal of Cr(VI), Ni(II) and Zn(II) ions from synthetic solutions and electroplating effluent. The experiments were conducted to study the effect of important design parameters such as bed height, flow rate and initial concentration of metal ions. The maximum sorption capacity was observed at flow rate 5 ml/min, bed height 20 cm and metal ions concentration 50 mg/L with immobilized biomass. Whereas, breakthrough time and saturation time decreased with increase flow rate and metal ions concentration and an inverse condition was found in bed height. The bed depth service time (BDST) Adams-Bohart model was used to analyze the experimental data. The regeneration efficiency was observed 40.1%, 75% and 53% for Cr(VI), Ni(II) and Zn(II) without any significant alteration in sorption capacity after 5th sorption-desorption cycles.  相似文献

研究了培养时间、初始pH、温度对禾本红酵母Y-5吸附结晶紫、孔雀石绿的影响,并对吸附剂的解吸和循环利用等进行考察.结果表明: 在染料浓度为50 mg·L-1、pH 7.0、摇床转速150 r·min-1、30 ℃、吸附10 h时,红酵母对结晶紫、孔雀石绿的吸附率分别达峰值,为93.8%和87.7%;解吸后的菌体对结晶紫、孔雀石绿的吸附率分别为85.5%和78.5%,说明菌体对染料的吸附是可逆的,循环利用效果良好,菌体可再生和循环利用;禾本红酵母Y-5对结晶紫、孔雀石绿的脱色机理为吸附作用,染料多被吸附在红酵母表面的羟基(-OH)上,其吸附染料快速、高效、可逆,在染料废水处理上具有潜在的应用价值.  相似文献

Dye removal from industrial waste water has become an important issue. The highvisibility, undesirability and recalcitrance are the significant environmental problemfor the dyes. In the present work,β‐cyclodextrin functionalized KCC‐1 (KCC‐1‐NH‐β‐CD)was synthesized and utilized to the removal of hazardous malachite green. In order to study the morphology of the synthesized nano adsorbent, Scanning Electron Microscopy (SEM) and Transmission Electron Microscopy (TEM) were obtained from the surface of the sample. Additionally, the functionalization of KCC‐1 with β‐cyclodextrin was confirmed with Furrier Transform Infrared spectroscopy (FTIR). The textural property of KCC‐1 was verified using nitrogen adsorption/ desorption analysis (BET equation). UV‐Vis spectroscopy utilized for the investigation of malachite green by KCC‐1‐NH‐β‐CD. Specific surface area of the adsorbent was calculated to be 140 m2/g and it can be stated that the synthesized nano adsorbent has high removal efficiency. It should be noted that the adsorption capacity of the employed nano adsorbent was more than 95%, which could be attributed to high porosity of β‐cyclodextrin functionalized KCC‐1.  相似文献

Endocrine disruptors refer to chemical compounds in the environment which interfere with the endocrine systems of organisms. Among them, environmental estrogens pose serious problems to aquatic organisms, in particular fish. It is therefore important and necessary to have a fast and low-cost system to screen the large number of different chemical compounds in the aquatic environment for their potential endocrine disrupting actions. In this study, a screening platform was developed to detect xenoestrogens in the aquatic environment using the fission yeast Schizosaccharomyces pombe, and applied for compound screening. The aim was to demonstrate any significant potential differences between the fish screening system and the human screening system. To this end, a yeast expression vector harboring a fish estrogen receptor alpha and a reporter vector containing the estrogen responsive element fused with the Escherichia coli LacZ gene were constructed. After transformation with these two vectors, the transformed yeast clones were confirmed by Western blotting and selected on the basis of the beta-galactosidase activity. In this transgenic yeast system, the natural estrogen (estradiol) and other known xenoestrogens such as diethylstilbestrol, bisphenol A, genistein and dichloro-diphenyl-trichloroethane exhibited dose-dependent activities. Using this system, more than 40 putative endocrine disruptors including phytoestrogens, pesticides, herbicides, industrial dyes and other industrial chemicals were screened. Ten of them were demonstrated to exhibit estrogenic actions. Industrial dyes such as malachite green (MG) that disrupt thyroid hormone synthesis are extensively used and are widely distributed in the aquatic environment. Using this system, MG did not show any estrogenic action, but was demonstrated to exhibit anti-estrogenic activity.  相似文献

Biosorption potential of novel lignocellulosic biosorbents Musa sp. peel (MSP) and Aegle marmelos shell (AMS) was investigated for the removal of toxic triphenylmethane dye malachite green (MG), from aqueous solution. Batch experiments were performed to study the biosorption characteristics of malachite green onto lignocellulosic biosorbents as a function of initial solution pH, initial malachite green concentration, biosorbents dosage, and temperature. Biosorption equilibrium data were fitted to two and three parameters isotherm models. Three-parameter isotherm models better described the equilibrium data. The maximum monolayer biosorption capacities obtained using the Langmuir model for MG removal using MSP and AMS was 47.61 and 18.86 mg/g, respectively. The biosorption kinetic data were analyzed using pseudo-first-order, pseudo-second-order, Elovich and intraparticle diffusion models. The pseudo-second-order kinetic model best fitted the experimental data, indicated the MG biosorption using MSP and AMS as chemisorption process. The removal of MG using AMS was found as highly dependent on the process temperature. The removal efficiency of MG showed declined effect at the higher concentrations of NaCl and CaCl2. The regeneration test of the biosorbents toward MG removal was successful up to three cycles.  相似文献

A method was validated for the determination of ochratoxin A (OTA) in soluble and green coffee. Performance parameters evaluated included selectivity, accuracy, intermediate precision, linearity, limit of detection, limit of quantitation, and ruggedness. The method was found to be selective for OTA in both matrices tested. Recovery rates from soluble coffee samples ranged from 73.5 to 91.2%, and from green coffee samples from 68.7 to 84.5%. The intermediate precision (RSDr) was between 9.1 and 9.4% for soluble coffee and between 14.3 and 15.5% for green coffee analysis. The linearity of the standard calibration curve (r2) was <0.999 for OTA levels of 1.0–20.0 μg/kg in coffee samples. The limit of detection was determined to be 0.01 ng of OTA on column, while the limit of quantitation was found to be 0.03 ng on column. The limit of quantitation is equivalent to 0.6 μg/kg in soluble coffee samples and 0.3 μg/kg in green coffee samples. The results of the ruggedness trial showed two factors are critical for soluble coffee analysis: the extraction method, and the flow rate of the mobile phase. For green coffee analysis two critical factors detected were the extraction method and the storage temperature of the immunoaffinity column. Five samples of soluble coffee and 42 of green coffee were analysed using the validated method. All soluble coffee samples contained OTA at levels that ranged from 8.4 to 13.9 μg/kg. Six of the 42 green coffee samples analysed (14.3%) contained OTA at levels ranging from 0.9 to 19.4 μg/kg. The validated method can be used to monitor OTA levels in Colombian coffee for export or for local consumption.  相似文献
