The saline soda lakes of the Kulunda steppe (Altai krai) are small and shallow; they are characterized by a wide range of salinity and alkalinity, as well as by the extreme instability of their water and chemical regimes. Accumulations of anoxygenic phototrophic bacteria (APB) visible to the unaided eye were noted only in several lakes with high rates of sulfate reduction in their bottom sediments. However, enumeration of APB cells by inoculation revealed their presence in all 17 lakes. APB cell numbers varied from 103 to 109 CFU cm?3. In the APB communities of all lakes, purple sulfur bacteria of the family Ectothiorhodospi- raceae were predominant. In 14 out of the 17 lakes, purple nonsulfur bacteria of the family Rhodobacteraceae were also detected (103–107 CFU cm?3). Purple sulfur bacteria of the family Chromatiaceae were less abundant: Halochromatium sp. (104–107 CFU cm?3) were found in six lakes, while Thiocapsa sp. (104 CFU cm3) were detected in one lake. On the whole, the APB communities of the soda lakes of the Kulunda steppe were characterized by the low diversity and evenness of their species compositions, as well as by the pronounced dominance of the members of the family Ectothiorhodospiraceae. There was no correlation between the structures of the APB communities and alkalinity. However, the dependence of the species composition of APB (mainly ectothiorhodospiras) on water mineralization was revealed. High mineralization (above 200 g l?1) was a limiting factor that affected the APB communities on the whole, restricting the APB species diversity to extremely halophilic bacteria of the genus Halorhodospira.  相似文献

Here we describe the diversity and activity of sulfate reducing bacteria along a salinity gradient in four different soda lakes from the Kulunda Steppe (South East Siberia, Russia). For this purpose, a combination of culture-dependent and independent techniques was applied. The general bacterial and SRB diversity were analyzed by denaturing gradient gel electrophoresis (DGGE) targeting the 16S rDNA gene. DNA was used to detect the microbial populations that were present in the soda lake sediments, whereas ribosomal RNA was used as a template to obtain information on those that were active. Individual DGGE bands were sequenced and a phylogenetic analysis was performed. In addition, the overall activity of SRB was obtained by measuring the sulfate reduction rates (SRR) and their abundance was estimated by serial dilution. Our results showed the presence of minor, but highly active microbial populations, mostly represented by members of the Proteobacteria. Remarkably high SRR were measured at hypersaline conditions (200 g L−1). A relatively high viable count indicated that sulfate reducing bacteria could be highly active in hypersaline soda lakes. Furthermore, the increase of sodium carbonate/bicarbonate seemed to affect the composition of the microbial community in soda lakes, but not the rate of sulfate reduction.  相似文献

The activity and culturable diversity of sulfidogens were investigated in anoxic sediments of four hypersaline lakes with pH 7.6-8.2 in the Kulunda Steppe (Altai, Russia). Sulfate reduction rates were low, varying from 0.1 to 6.0 nmol HS(-) /(cm(3) h) with a maximum in the top 10 cm layer. Potential sulfidogenic rates with thiosulfate and sulfur as the e-acceptors were higher than with sulfate and were stimulated by formate, lactate, and acetate. Sulfidogenesis was optimal at salt concentrations below 2 M NaCl. Cultivation at 2 M NaCl resulted in the isolation of several strains of moderately halophilic SRB, but no growth of SRB was observed at 4 M NaCl. At lithotrophic conditions (i.e., with formate or H(2) as e-donors), several closely related alkalitolerant strains belonging to the genus Desulfonatronovibrio were isolated. Enrichments at heterotrophic conditions with lactate, propionate, acetate, or butyrate using sulfate or thiosulfate as e-acceptors yielded isolates related to Desulfosalsimonas propionicica, Desulfohalobium utahense, and Desulfocella halophila. Sulfur-reducing enrichments at 2 M NaCl with ethanol produced a member of the genus Halanaerobium, while enrichments at 4 M NaCl with acetate were dominated by archaea, demonstrating for the first time such type of catabolism in haloarchaea.  相似文献

The structure of benthic phototrophic communities of 24 soda lakes of the southeastern Transbaikal Region was studied. The physicochemical properties of the lakes were determined. The results of the cell count of anoxygenic phototrophic bacteria (APB) belonging to various groups are presented. The influence of salinity on the structure of APB communities was investigated. The APB reaction to environmental conditions was determined. Massive development of phototrophic microorganisms in the form of mats and films was observed in the majority of the investigated lakes. The APB communities were characterized by a wide diversity of species and evenness of species composition. Purple sulfur bacteria of the families Ectothiorhodospiraceae and Chromatiaceae were predominant. Purple nonsulfur bacteria of the family Rhodobacteraceae, green filamentous bacteria Oscillochloris sp., and heliobacteria were also detected. According to preliminary data, no less than 15 species of APB occur in the studied lakes. Among them, three novel genera and four species have already been described. Identification of other isolates is still in progress. The lakes make an almost continuous series of fresh, brackish, and saline water bodies, varying in their degree of mineralization. It was demonstrated that the structure of APB communities was unaffected by salinity ranging from 5 to 40 g/l. At salt concentrations of lower than 5 g/l, the level of water mineralization became a limiting factor. Experiments with the isolated cultures showed that the APB were obligately dependent on the presence of carbonate ions in the medium. They were haloalkalitolerant or haloalkaliphilic. Thus, they are well adapted to the conditions of soda lakes with a high mineralization. It was demonstrated that soda lakes of the southeastern Transbaikal Region represent a special type of habitat which harbors a peculiar autochthonous microflora and differs from both highly mineralized soda lakes and shallow saline water bodies of the sea origin.  相似文献

Bitter-1 is a shallow hypersaline soda lake in Kulunda Steppe (Altai region, Russia). During a study period between 2005 and 2016, the salinity in the littoral area of the lake fluctuated within the range from 85 to 400 g/L (in July of each year). Light-dependent nitrogen fixation occurred in this lake up to the salt-saturating conditions. The rates increased with a decrease in salinity, both under environmental conditions and in laboratory simulations. The salinities below 100 g/L were favorable for light-dependent nitrogen fixation, while the process was dramatically inhibited above 200 g/L salts. The analysis of nifH genes in environmental samples and in enrichment cultures of diazotrophic phototrophs suggested that anaerobic fermenting and sulfate-reducing bacteria could participate in the dark nitrogen fixation process up to soda-saturating conditions. However, we cannot exclude the possibility that haloalkaliphilic nonheterocystous cyanobacteria (Euhalothece sp. and Geitlerinema sp.) and anoxygenic purple sulfur bacteria (Ectothiorhodospira sp.) might also play a role in the process at light conditions. The heterocystous cyanobacterium Nodularia sp. develops at low salinity (below 80 g/L) that is not characteristic for Bitter-1 Lake and thus does not make a significant contribution to the nitrogen fixation in this lake.

The lakes under study can be classified as eutrophic-hypertrophic waterbodies according to the level of primary production. High concentrations of suspended matter (up to 7326 g/m3) and total phosphorus (up to 5625 mg/m3) were registered in the lakes. The values of the primary production of plankton and the destruction of organic matter do not depend on the absolute concentration of salts in water. Unlike the destruction of organic matter, the primary production of plankton is higher in winter than in summer. Thus, the accumulation of organic matter in saline lakes occurs mainly in winter. The negative relationship between the primary production of plankton and the biomass of the filter feeder Artemia sp. has been determined.  相似文献

We have studied the activity and composition of several geochemically significant physiological groups of bacteria in more than twenty alkaline salt lakes of the north-east Mongolia steppe with water salinity from 3 to 390 g l?1 and pH values ranging from 9.0 to 10.6. Active and diverse microbial communities have been found in most of the lakes. The methanotrophic bacteria were represented by the Type I members. Among the culturable forms of sulfur-oxidizing bacteria obligately chemolithoautotrophic and haloalkaliphilic representatives of the genera Thioalkalimicrobium and Thioalkalivibrio were detected in the sediments at high numbers (up to 106 cells ml?1). The largest population of anaerobic phototrophic bacteria was represented by purple sulfur bacteria of the Ectothiorhodospiraceae family. Salinity was the key factor in determining the activity and the composition of the microbial communities. The most diverse and active prokaryotic populations, including aerobic and anaerobic phototrophic, methanogenic, methanotrophic, sulfur-oxidizing, sulfate-reducing and nitrifying bacteria, were found in lakes with salinity less than 60 g l?1. In hypersaline lakes with a salinity >100 g l?1, the sulfur cycle remained active due to the activity of extremely halotolerant and alkaliphilic sulfur bacteria, while other important functional groups responsible for nitrification and methane oxidation processes were not detected. Overall, the prokaryotic communities of the Mongolian alkaline salt lakes represent an interesting new example of a diverse community of haloalkaliphilic bacteria well adopted to a broad salinity range.  相似文献

Samples of natural APB communities were inoculated into media that differed in pH values, which were 7, 9.5, or 10.5. The structure of the APB communities in the obtained enrichment cultures depended on the pH values and on the mineralization levels of the media, which were the same as in the lakes from which samples were taken. The same dependence of the community structure on salinity was observed as in the case of the natural communities that had been described previously. APB were most diverse in the enrichment cultures grown at pH 9.5. The shift of the pH to either neutral or extremely alkaline values affected the species diversity within the APB community, resulting in marked predominance of the most adapted forms. It was shown that the status of Ectothiorhodospira within the community could serve as an indicator of both salinity and pH in soda lakes with a water mineralization of higher than 5 g/l. The statuses of various APB groups in the community as dependent on pH and salinity are discussed, as well as possible changes in these statuses due to changes in the water level and other environmental parameters in the studied lakes.  相似文献

From reed biofilm samples of Kelemen-szék (Kiskunság National Park, KNP) and Nagy-Vadas (Hortobágy National Park, HNP) altogether 260 bacterial isolates were gained after serial dilutions and plating onto different media. Following a primary selection 164 strains were investigated by "traditional" phenotypic tests and clustered by numerical analysis. Fifty-six representative strains were selected to ARDRA and 16S rDNA sequence analysis for identification. Strains were identified as members of genera Agrobacterium, Paracoccus, Halomonas, Pseudomonas, Bacillus, Planococcus and Nesterenkonia. The species diversity was also investigated by a cultivation independent method. A clone library was constructed using the community DNA isolated from the biofilm sample of Kelemen-szék. Screening of the 140 bacterial clones resulted in 45 different ARDRA groups. Sequence analysis of the representatives revealed a great phylogenetic diversity. A considerable majority of the clones was affiliated with uncultured bacterial clones (with sequence similarity between 93 and 99%) originating from diverse environmental samples (for example salt marshes, compost or wastewater treatment plants). The DNA sequences of other clones showed the presence of genera Flavobacterium, Sphingobacterium, Pseudomonas and Agrobacterium.  相似文献

The presence of aerobic anoxygenic phototrophs (AAPs) has been repeatedly reported from various marine environments, but their distribution in freshwater lakes was neglected until recently. We investigated the phylogenetic composition of AAP communities in 10 lakes in Northeastern Germany with different trophic status including oligotrophic Lake Stechlin and humic matter rich Lake Grosse Fuchskuhle. The AAP community was composed by members of Alpha- and Betaproteobacteria, but their contribution varied largely among the studied lakes. Our results show that AAP community composition in the studied lakes was affected mostly by pH and humic matter content. While alkaline lakes were mostly composed of Betaproteobacteria, the acidic and humic matter rich south-west (SW) basin of Lake Grosse Fuchskule was dominated (87%) by Alphaproteobacteria. The most frequent group within Betaproteobacteria was a cluster of pufM genes which was phylogenetically related to Rhodoferax representing 38.5% of all retrieved sequences. Alphaproteobacteria-related sequences had a broader phylogenetic diversity including six different taxa dominated by Sphingomonas- and Rhodobacter-like bacteria in lakes with alkaline to neutral pH. In the acidic and humic matter-rich SW basin of Lake Grosse Fuchskuhle, however, Methylobacterium-related sequences dominated the AAP community. We suggest that the variable AAP community structure might reflect the potential of these bacteria to cope with the contrasting conditions in freshwater environments.  相似文献

SUMMARY. 1. A survey of dissolved organic carbon concentration [DOC] and optical absorbance at 340 nm (A340) was carried out for thirteen lakes and twenty-three streams in Cumbria over the period May 1986 to April 1987.
2. Values of [DOC] were in the range 0-6.2 mg 1-1-1, no water having a mean [DOC] greater than 3.6mgI-1. Streamwaters showed no gross seasonal dependence in [DOC], but there was a peak in [DOC] associated with the first major rain event of autumn 1986. Summer values of [DOC] in lakes were greater than winter values, probably because of the production of DOC by phytoplankton.
3. In streams, [DOC] was weakly positively correlated (r2 <0.3) with discharge (as estimated from rainfall); A340 was more strongly correlated (0.4 < r2 < 0.6). Linear regression of mean A340 against mean [DOC] for streams gave a slope consistent with the absorptivities of aquatic humic substances, and a positive intercept on the abscissa that suggests the presence of, on average 0.6 mg 1-1 of DOC that does not absorb at 340 nm. Lake samples taken in winter gave a similar A.340/[DOC] relationship to that for streams, but in summer A340/[DOC] ratios were lower, suggesting that phytoplankton-derived [DOC] absorbs weakly or not at all at 340nm.  相似文献

The population structure of bloodsucking mosquito larvae in temporary and constant water bodies of the northern Kulunda steppe was investigated. The seasonal dynamics of the population density, the number of species in different types of reservoirs, and average density of each species are given. The productivity of water reservoirs in relation to mosquitoes of the family Culicidae is analyzed. The basic factors affecting the distribution of larvae of dominant species between different water bodies were revealed. The degree of correlation between the structure of bloodsucking mosquito larva assemblages and the type of the reservoir was established.  相似文献

从土壤有机无机复合状况、腐殖质结合形态和腐殖质组成等方面,研究了供试土壤(草原淡黑钙土、草原风沙土和已开垦的风沙土)有机质的性质.结果表明,不同土壤类型及利用方式对土壤有机质性质的影响有很大的差异.与风沙土相比,淡黑钙土的全土碳、生物量碳、重组碳、可提取腐殖物质(HE)、松结态腐殖质的含量和全土腐殖化度较高.风沙土种向日葵后其全土碳、生物量碳、重组碳和HE含量及各种结合形态腐殖质含量均下降,但腐殖化程度相差不大.  相似文献
