Laboulbenia ophioneae is described as a new species with illustrations. It is closely related to Laboulbenia celestialis and Laboulbenia asiatica because of some similarities in the morphology of appendages and perithecia. The present species from the latter two species can be distinguished by the shorter, inflated perithecia, the shorter receptacles, and the appendages consisting of more or less darkly colored, broader branches. This new species was found on elytra of Ophionea indica from Taiwan, which has been also known as a host of Laboulbenia polymorpha. A mature thallus noticed by Terada (2004) on the slide 673b (M. Ishikawa collection) and a young thallus illustrated by Sugiyama (1978, fig. 1-G as L. polymorpha) on 673d (M. Ishikawa collection) are both identified as L. ophioneae.  相似文献

Summary Far-red light was demonstrated to accelerate senescence in mature, green tissue of intact thalli of Marchantia polymorpha. The red/far-red reversibility of this phenomenon proves the involvement of phytochrome. As a daily exposure to 5 min red light is sufficient to prevent aging of the tissue, photosynthesis does not play a specific role in this response.  相似文献

In this study, we observed the germination behaviour of airborne conidia from powdery mildews that settle on thalloid surfaces. We inoculated thalli (flat, sheet‐like leaf tissues) and gemmae (small, flat, sheet‐like leaf tissues that propagate asexually via bud‐like structures) of the common liverwort (Marchantia polymorpha) with conidia from tomato powdery mildew (Oidium neolycopersici; KTP‐02) and red clover powdery mildew (Erysiphe trifoliorum; KRCP‐4N) and examined their germination and subsequent appressorium formation under a high‐fidelity digital microscope. Conidial bodies and germ tubes of the inoculated KRCP‐4N conidia were destroyed on both the thalli and gemmae. The destruction of these fungal structures was observed only for KRCP‐4N conidia inoculated onto M. polymorpha on both leaf surfaces. No differences in destruction of the KRCP‐4N fungal structures between thalli and gemmae were observed. At 4 h post‐inoculation, destruction of the germ tube tip was observed when it reached the gemmae leaf surface. At 6 h post‐inoculation, the conidial bodies and germ tubes were destroyed. In contrast, KTP‐02 conidia were not destroyed and formed normal, well‐lobed appressoria on the surface of M. polymorpha gemmae.  相似文献

Glycogen synthase I (UDP glucose: glycogen alpha-4-glycosyltransferase, EC2.4.1.11) of the tapeworm Hymenolepis diminuta is the form of the enzyme which is active in vivo, while the D-form represents an inactive "storage form." Utilizing the differential effect of inorganic phosphate (Pi) on the I and D-forms, the ratio of the 2 forms in vivo has been determined under conditions of starvation of the host and refeeding of the parasite with glucose. This procedure reveals that conversion of the inactive D-form to the active I-form takes place when glycogen-depleted worms are incubated in glucose. The activity of glycogen synthase I also is affected by the molecular weight of the primer glycogen. With certain molecular weight fractions, enzymatic activity is higher than with others. This specificity of the glycogen primer could explain the relatively low concentrations of those molecular weight fractions which confer the highest synthase activity.  相似文献

A new species, Anthonomus dilatofemurus Poinar et Legalov, sp. nov., from Mexican amber is described and illustrated. The new species is close to A. cruraluma Poinar et Legalov and differs from it in the wider elytra with narrow striae and strongly swollen pro- and metafemora. It differs from A. browni Poinar et Legalov in the wide and short elytra, shorter and thicker weakly curved rostrum, and strongly swollen metafemur. It is distinguished from A. camoiranensis Clark by the metatitibia with uncus and from A. sulcatus Kirsch by the protibia with uncus, metatibia with ventral fringe of medium scales, narrower elongate pronotum, weakly convex elytra and weakly curved rostrum.  相似文献

Abstract 1 External gland openings and associated structures on the elytra of teneral and mature Diaprepes root weevils, Diaprepes abbreviatus (L.), were elucidated by scanning electron microscopy (SEM). 2 There were clear differences between teneral, callow adults and fully mature adults. In the field, teneral adults remain in the pupal chamber in the soil until sclerotization of the cuticle is complete or nearly so. 3 Phenotypic variation of the elytra in this species consists of varying patterns and coloration of scaled intervals between a variable number of raised ridges devoid of scales. In addition to being thinner and lighter in colour than fully mature adults, the elytra of teneral adults were devoid of waxy hydrocarbon secretions. 4 External gland openings at the base of each scale were observed on teneral elytra and mature elytra washed with methylene chloride. 5 SEM evidence to document the production of waxy filaments by these glands and partial characterization of these by gas chromatography and mass spectrometry are presented.  相似文献

Extracts of uninduced thalli, induced thalli, stalks, antheridiophore and archegoniophore disks of Marchantia polymorpha were subjected to starch-gel zone electrophoresis. Developed gels were treated with appropriate reaction mixtures to detect sites of activity for 12 enzyme systems; only phosphatases, esterases, and peroxidases were observed. Although common sites of phosphatase, peroxidase, and esterase activity were detected in all tested extracts, additional sites of peroxidase and esterase activity were found in extracts from antheridiophore disks. The antheridia provided the additional esterases as determined by the electrophoresis of antheridial extracts and by a histochemical test for esterases in sections of antheridiophores.  相似文献

In lower land plants, genes controlling the transition from vegetative growth to sexual reproduction have not yet been identified. In the dioecious liverwort Marchantia polymorpha, the transition to sexual reproduction accompanied by the formation of sexual organs on the gametophytic thallus is initiated under long-day conditions. By particle bombardment-mediated mutagenesis, we generated a mutant of M. polymorpha that constitutively forms sexual organs. This mutant is fully fertile, showing that the mutation does not affect formation of male or female sexual organs per se. Genetic analysis reveals that this phenotype is caused by mutation of a single autosomal locus, suggesting that this mutation defines or controls a gene regulating the transition to sexual reproduction in M. polymorpha.  相似文献

Thalli of the haploid liverwort Marchantia polymorpha were successfully used for direct particle bombardment with plasmid pMT, which carries a hygromycin phosphotransferase gene (hpt) controlled by the CaMV 35S promoter and the NOS polyadenylation region. Hygromycin-resistant cell masses arose from the thallus surface and developed directly into hygromycin-resistant thalli. Southern blot analyses indicated that these thalli carried at least 1–4 copies of the hpt gene, which were stably transmitted to their asexual thallus progenies via gemma propagation for three generations. This transformation and direct plant regeneration protocol is expected to be a valuable tool for the molecular analysis of this lower land plant.  相似文献

New lobe development and lobe division was studied in the foliose lichen Xanthoparmelia conspersa (Ehrh. ex. Ach.) Hale. In thalli with either the centre or margin removed, the inside edge of the perimeter, the outer edge of the reproductive centre, and fragments derived from the thallus perimeter all regenerated growing points (‘lobe primordia’) within a year. Thalli possessing isidia had the greatest ability to regenerate growing points. In reproductive thalli, there was a positive correlation between the density of new growing points and thallus size. When fragments were cut from the perimeters of mature X. conspersa thalli and glued to pieces of slate, the ratio of growing points to mature lobes increased over 54 months. Lobes within a thallus exhibited different degrees of bifurcation. In some bifurcating lobes, the point of origin of the bifurcation advanced at the same rate as the lobe tips over 4 months but in most lobes, the bifurcation point either advanced less rapidly than the lobe tips or retreated from its original location. Removing adjacent lobes had no significant effect on the radial growth of a lobe over 4 months or on the location of the bifurcation point but it increased the number of growing points. These results suggest that for X. conspersa: 1) all portions of of thalli can regenerate growing points, 2) few growing points actually develop into mature lobes, 3) individual lobes within a thallus grow and divide differently, and 4) adjacent lobes inhibit the development of growing points on their neighbours.  相似文献

Summary The effect of high temperatures on the photosynthetic apparatus of Preissia quadrata (Scop.) Nees, Conocephalum conicum (L.) Dum. and Marchantia polymorpha L. were investigated. changes in the activities of various photosynthetic reactions were followed by measuring light-dependent oxygen evolution, chlorophyll a fluorescence and light-induced absorbance changes at 518 nm.Mild heat treatment of the thalli led to reversible depression of photosynthesis; the period necessary for complete recovery depended on the extent of heat damage. Irreversible inactivation of photosynthesis after more severe heat stress was caused by damage of photosystem II. On principle, the pattern of reversible and irreversible heat inactivation of photosynthetic reactions in liverwort thalli resembles that observed in leaves of higher plants. However, in contrast to a number of Spermatophyta, exposure of liverwort thalli to high sublethal temperatures did not result in a significant increase in the heat stability of the photosynthetic apparatus indicating that the heat hardening capacity of hygrophytic hepatics is extremely low.  相似文献

Genomic, proteomic, phylogenetic and evolutionary aspects of a novel gene encoding a putative chloroplast-targeted sulfate permease of prokaryotic origin in the green alga Chlamydomonas reinhardtii are described. This nuclear-encoded sulfate permease gene (SulP) contains four introns, whereas all other known chloroplast sulfate permease genes lack introns and are encoded by the chloroplast genome. The deduced amino acid sequence of the protein showed an extended N-terminus, which includes a putative chloroplast transit peptide. The mature protein contains seven transmembrane domains and two large hydrophilic loops. This novel prokaryotic-origin gene probably migrated from the chloroplast to the nuclear genome during evolution of C. reinhardtii. The SulP gene, or any of its homologues, has not been retained in vascular plants, e.g. Arabidopsis thaliana, although it is encountered in the chloroplast genome of a liverwort (Marchantia polymorpha). A comparative structural analysis and phylogenetic origin of chloroplast sulfate permeases in a variety of species is presented.  相似文献

To test the premise that fertilization distances in bryophytes are limited by spermatozoid motility, we measured the distances between over 80 000 male and female thalli of Marchantia polymorpha on heathland over 2 successive years after a major fire and recorded the incidence of sporophytes. The first comparative data on spermatozoid numbers per antheridium across bryophytes and pteridophytes were calculated from antheridial sizes and spermatid diameters and cross‐checked with the likely numbers produced by successive antheridial mitoses. Individual antheridia of Marchantia produce over 200 000 spermatozoids. Extrapolating from individual antheridia to the numbers of mature antheridia per antheridiophore and then to individual male plants, we calculated that a single flooding event on a male thallus with 10–12 antheridiophores will result in the release of over 50 million spermatozoids. Assuming radial dispersal in surface water films by lipids released from the dehiscing antheridia, spermatozoids can reach distances exceeding 20 m from the parent plants, in line with our finding of 100% fertilization in female plants of Marchantia up to 19 m from the nearest males, far beyond published fertilization distances in dioicous bryophytes. We attribute this to (i) much greater spermatozoid production and numbers in Marchantia than in other bryophytes and (ii) highly effective sperm transport both within the antheridiophores and archegoniophores via their overlapping scales and grooves in the stalks and between male and female thalli via surface water films. These features, coupled with the massive production of small spores, explain the success of Marchantia as the primary colonist of open habitats.  相似文献

Aerobic dissimilation of l-rhamnose by various genera and species of yeasts was examined. Of 63 strains tested, 12 utilized l-rhamnose fairly well, and 7 strains, Debary-omyces klöckeri, Pichia pseudopolymorpha, P. rhodanensis, P. robertsii, P. wickerhamii, Candida polymorpha and Torulopsis famata, produced an appreciable amount of l-rhamnonic acid and/or 1,2-propanediol. These two main products were isolated in pure forms from the fermented broth of C. polymorpha and identified. Candida polymorpha produced l-rhamnonic acid and 1,2-propanediol from l-rhamnose in the presence of CaCO3 at good yield of 38% of sugar consumed.  相似文献

The function of the flapping elytra was investigated in garden shafers (Melolontha melolontha L.) and rhinoceros beetles (Oryctes boas Fabr.).
1.  The movements of alae and elytra are determined. Both move with the same frequency and in the same phase, but the range of the elytral stroke never reaches below the frontal plane. In downstroke the elytra are at an angle positive to the airstream (horizontal flight).
2.  After exact investigation of the construction of the elytra (depth, width, length, surface, torsion and velocity of each point of the elytra) the most effective point in the production of lift during the downstroke was determined.
3.  After loss of one third of the surface of the elytra the beetles cannot fly horizontally. In tethered flight the wing stroke frequency of the alae does not increase when the elytra are shortened. Shortening of the elytra does not interfere with the resonance system of the pterothorax.
4.  The lift of the elytra results from active and passive components (more than 17% of the body weight). In the airstream (front 2.00 m/s, average body weight 944 mp) the passive lift is 14.3% for elytra in the down position; 3.1% of the lift is caused by the active downstroke.
5.  The elytra of these beetles are indispensible for horizontal flight. Primarily they produce lift passively with added help from the downstroke, and have no function for the drag. The effect of upstroke is discussed.

The nucleotide sequence of the previously cloned (Zabolotskaya, M. V., Nosovskaya, E. A., Kaplun, M. A., and Akimkina, T. V. (2001). Mol. Gen. Mikrobiol. Virusol. No 1, 32–34) DNA fragment from Thermoactinomyces sp. 27a (GenBank Accession No. AY280367) containing the metalloproteinase gene was determined. A continuous open reading frame encoding a polypeptide of 673 aa was revealed. Analysis of this sequence demonstrated that the metalloproteinase from Thermoactinomyces sp. 27a is synthesized as a preproprotein and includes a leader peptide (26 aa), N-terminal propeptide (215 aa), mature region (317 aa), and additional C-terminal domain (115 aa). The recombinant enzyme from Thermoactinomyces sp. 27a was expressed in Bacillus subtilis AJ73 cells and purified by anion exchange chromatography to an electrophoretically homogeneous state. The determined N-terminal amino acid sequence of the mature protein was identical to that deduced from the gene. The obtained data suggest that the mature protein should include 432 aa and have a calculated molecular weight of 46,262 Da. However, the molecular weight of the mature protein determined by mass spectrometry was 34,190 ± 70 Da indicating a C-terminal processing. Theproteinase was not inhibited by phenylmethyl sulfonyl fluoride but was inhibited by o-phenanthroline and ethylenediaminetetraacetic acid. The enzyme had maximum activity by azocasein hydrolysis at 55°C and pH 6.5–7.5; it was stable at pH 7.5–8.5 and remained stable at 50°C for several hours. The kcat/Km for 3-(2-furyl)acryloyl-glycyl-L-leucine amide hydrolysis was (2.8 ± 0.1) ×103 M–1×s–1.  相似文献

L-Lactate cytochrome c oxidoreductase (flavocytochrome b 2, FC b 2) from the thermotolerant methylotrophic yeast Hansenula polymorpha (Pichia angusta) is, unlike the enzyme form baker’s yeast, a thermostable enzyme potentially important for bioanalytical technologies for highly selective assays of L-lactate in biological fluids and foods. This paper describes the construction of flavocytochrome b 2 producers with over-expression of the H. polymorpha CYB2 gene, encoding FC b 2. The HpCYB2 gene under the control of the strong H. polymorpha alcohol oxidase promoter in a plasmid for multicopy integration was transformed into the recipient strain H. polymorpha C-105 (grc1 catX), impaired in glucose repression and devoid of catalase activity. A method was developed for preliminary screening of the transformants with increased FC b 2 activity in permeabilized yeast cells. The optimal cultivation conditions providing for the maximal yield of the target enzyme were found. The constructed strain is a promising FC b 2 producer characterized by a sixfold increased (to 3 μmol min?1 mg?1 protein in cell-free extract) activity of the enzyme.  相似文献

The author has reviewed the literature concerning the isolation and recognition of those bacteria variously namedMoraxella lwoffi, Mima polymorpha, B5W,Herellea vaginicola, Bacterium (Achromobacter, Acinetobacter)anitratum, Moraxella glucidolytica, Neisseria winogradskyi, Moraxella liquefaciens, Moraxella non-liquefaciens, Moraxella bovis, andMima polymorpha var.oxydans.He concludes that the saccharolytic and non-saccharolytic members may be extreme forms within a single group of bacteria. He does not think it yet feasible in the light of our present knowledge to split this group into species on the basis of sugar oxidation reactions, urease production or oxydase production. However, the first two are similar and may be tentatively namedMoraxella lwoffi; the next fiveMoraxella glucidolytica; and the last four also aMoraxella although the names liquefaciens and non-liquefaciens seem undesirable, duplex being a possible alternative. The final genus has yet to be decided from the possible choices ofAcinetobacter andMoraxella.  相似文献

Subcultures of SAG 211–9b and 1AM C-211, ultimately derived from CCAP 211/9b, a strain isolated by Pringsheim in 1939 and identified as Chlorella sac-charophila (Kruger) Migula were observed using light and electron microscopy. Their morphology proved to be basically identical. Both have two forms of cells, one (E-form) narrowly to broadly ellipsoidal, the other (S-form) ovoid to spheroidal. The cell wall of both forms is composed of a single smooth layer. The chloroplast of young cells is trough-like or saucer-shaped with a smooth margin, while that of mature cells is band- or cup-shaped with deep incisions. The thylakoid lamellae are loosely stacked and neither form has a pyrenoid. Both types of cells are capable of producing autospores: eight to 16 in E-form cells, two to four in S-form cells. These morphological features are different from those of C. saccharophila, which has a pyrenoid and produces only one form of autospores. In the absence of any existing genus that includes Chlorella-like algae with a simple cell wall, no pyrenoid, and two forms of mature cells and autospores, a new genus, Watanabea, is proposed with the type species W. reniformis.  相似文献

Exocelina baliem sp. n. is described from the Baliem Valley in the Central Mountain Range of New Guinea (Papua Province, Indonesia). Having striolate elytra, different structure and setation of the male and female genitalia and tarsomeres, and inhabiting swampy ponds, the new species differs from all known New Guinea species, which have smooth elytra and are stream associated. It forms a monophyletic group with the Australian Exocelina ferruginea (Sharp, 1882) and New Caledonian Exocelina inexspectata Wewalka, Balke & Hendrich, 2010, based on shape of the paramere and structure of the male tarsi. Habitus, protarsomeres, and male and female genitalia are illustrated, comparing some structures with Exocelina ferruginea and two New Guinea stream species. We briefly discuss the biogeographic relevance of this discovery.  相似文献
