During the course of studies on the oxidative metabolism of d-sorbitol by acetic acid bacteria, it was found that d-sorbitol was almost quantitatively converted to 5-keto-d-fructose via l-sorbose by a certain strain of Gluconobacter suboxydans. In addition to 5-keto-d-fructose, three γ-pyrone compounds, kojic acid, 5-oxymaltol, and 3-oxykojic acid, 2-keto-l-gulonate, and several organic acids such as succinic, glycolic, and glyceric acids were confirmed in the culture filtrate of this bacterium.
  • The most suitable carbon source for 5-ketofructose fermentation by Gluconobacter suboxydans Strain 1 was confirmed to be d-sorbitol or l-sorbose using growing and resting cells. d-Fructose had little effect on the formation of this dicarbonylhexose.

  • The optimal pH for the formation from l-sorbose by intact cells was found to be at 4.2.

  • The activity of the pentose phosphate cycle in the resting cells was calculated as 13~17 μatoms/hr/mg of dry cells by the use of the manometric techniques.

  • There was no strain tested so far which could accumulate a large amount of 5- keto-d-fructose from d-sorbitol except this bacterium.

  • The experimental results shown in this paper makes the prediction that a certain dehydrogenating system of l-sorbose is functional in the organism, and the metabolic pathways of d-sorbitol via l-sorbose and 5-keto-d-fructose is proposed.

A simple procedure is described for preparing sugar orthocarbonates. It is based on treating the corresponding thionocarbonate in pyridine with cupric acetate and an alcohol, such as methanol, ethanol, or isopropyl alcohol. Treatment of 1,2:5,6-di-O-isopropylidene-D-mannitol 3,4-thionocarbonate with diols, such as 1,2-ethanediol, 1,2-propanediol, or 1,2:5,6-di-O-isopropylidene-D-mannitol, also gave orthocarbonates. Methyl thionocarbonate, S-methyl xanthate, and dithiobis(thioformate) derivatives of 1,2:3,4-di-O-isopropylidene-α-D-galactopyranose all gave the trimethyl orthocarbonate upon treatment with methanol in the presence of pyridine and cupric acetate. The structure of the orthocarbonates was proved by elemental analysis, n.m.r., and mass spectra, and by treatment with mild acid to form carbonates. Treatment of 1,2:5,6-di-O-isopropylidene-3-thio-D-altritol 3,4-thionocarbonate with methanol or ethanol gave the corresponding orthothiocarbonate, but on treatment with 1,2-ethanediol or with sodium ethoxide the 3,4-episulfide resulted.  相似文献

Brevibacterium flavum mutants defective in the phosphoenolpyruvate (PEP)-dependent glucose phosphotransferase system (PTS) were selected with high frequency by 2-deoxyglucose-resistance. Most of them (DOGr) still had the fructose-PTS and grew not only on fructose but also on glucose like the wild-type strain. A mutant having 1 /8th the fructose-PTS activity of the wild strain but normal glucose-PTS activity was isolated as a xylitol-resistant mutant. It grew on glucose but not on fructose. The glucose-PTS was active on glucose, glucosamine, 2-deoxyglucose and mannose, and slightly on methyl-a-glucoside and N-acetylglucosamine, but not on fructose or xylitol. The fructose-PTS acted on fructose and xylitol, and to some extent on glucose but not on glucosamine or 2-deoxyglucose. Mutants unable to grow on glucose (DOGrGlc-) derived from a DOGr mutant were all defective in the fructose-PTS. All revertants able to grow on glucose derived from a DOGrGlc“ mutant had the fructose-PTS. The glucokinase activity was about 2/3rds the glucose activity of the fructose-PTS. All the DOGrGlc- mutants had normal levels of glucokinase. One of these mutants grew on maltose and sucrose, which were hydrolyzed to glucose. Thus, glucokinase seems to contribute to the phosphorylation of glucose liberated inside the cell. The fructose-PTS was induced by fructose and repressed by glucose. The glucose repression was not observed in a mutant defective in the glucose-PTS.  相似文献

The presence and some properties of the key enzymes of the glyoxylate cycle, isocitrate lyase (threo-Ds-isocitrate glyoxylate-lyase, EC 4.1.3.1) and malate synthase (L-malate glyoxylate-lyase (CoA-acetylating) EC 4.1.3.2), were investigated in Leptospira biflexa. Isocitrate lyase activity was found for the first time in the organism. The enzyme was induced by ethanol but not by acetate. The optimum pH was 6.8. The activity was inhibited by phosphoenolpyruvate, a specific inhibitor of isocitrate lyase. The optimum pH of malate synthase of L. biflexa was about 8.5. The Km value for glyoxylate was 3.0 × 10?3 M and the activity was inhibited by glycolate, the inhibitor. The results strongly suggested the presence of a glyoxylate cycle in Leptospira. The possibility that the glyoxylate cycle plays an essential role in the synthesis of sugars, amino acids and other cellular components as an anaplerotic pathway of the tricarboxylic acid cycle in Leptospira was discussed.  相似文献

The unicellular acidophilic red alga Galdieria sulphuraria is a facultative heterotroph with a complex uptake system for sugars and polyols, consisting of at least 14 transporters. Upon transfer to heterotrophic conditions, these transporters were induced simultaneously. Once induced, transporters for common hexoses and pentoses are apparently not down-regulated under heterotrophic conditions. Uptake of deoxysugars (FUC/Rha), however, was repressed by substrates metabolized via gluco-, galacto-, glycero-, or hexokinase, whereas substrates phosphorylated by xylulokinase had no effect. This indicates that several sugar kinases play a key role in sugar sensing. In contrast, polyol transporters were repressed only by glucose or its analogs but not by other sugars. This repression does not involve the activity of kinases. Most likely this type of glucose sensing is independent of metabolism and takes place prior to or during uptake. In its natural environment, these two different sensing mechanisms would enable the alga to utilize a mixture of different substrates in a most economic way by repressing dispensible transporters.  相似文献

Effects of irradiated sugar solutions on the growth and viability of E. coli B were investigated on glucose, fructose and sucrose. The antibacterial effect was demonstrated to be classified into two types; the bactericidal effect was developed in every sugar solutions irradiated in air, while the bacteriostatic effect was found especially in fructose irradiated in air free. The bactericidal activity was abolished by heating, pH change to alkali, and addition of catalase or ferrous ions, suggesting its entity is a peroxide products(s). No appreciable activity was observed with the radiation produced hydrogen peroxide and low molecular carbonyl compounds with each alone and even with their combination. Behaviours of the bacteriostatic activity on similar treatments indicate that the entity is unlikely a peroxide compound but a more thermostable and thiol reactive product.  相似文献

A product easily converted to glyoxal was found in an early stage of the reaction of sugar with amine in ethanol. Glyoxaldicyclohexylimine was isolated from the reaction mixture of d-glucose with cyclohexylamine. This finding suggested the formation of a similar type glyoxaldialkylimine in other reactions of sugar with amine. This two-carbon compound was assumed to be produced directly from sugar or glycosylamine, and a new pathway for sugar fragmentation was proposed.  相似文献

From the methanolic extract of the underground parts of Globularia orientalis, a new antioxidant sugar ester was isolated. The structure of the new compound, globularitol (1), was identified as 6-O-feruloyl-beta-D-glucopyranosyl-(1-->6)-glucitol by spectroscopic methods (1D and 2D NMR, ESI- and FAB-MS) and confirmed by chemical means.  相似文献

Aquaspirillum (Spirillum) gracile is one of the few spirilla that cause acidification of the medium when cultured with sugars. Acidic reactions have been reported only for d-glucose, d-galactose, and l-arabinose, and the mode of attack of these sugars has not been previously investigated. The soluble portion of extracts of glucose-cultured cells of A. gracile ATCC 19624 was found by spectrophotometric methods to contain enzyme activities characteristic of the Entner-Doudoroff and Embden-Meyerhof-Parnas pathways. No activity for 6-phosphogluconate dehydrogenase (EC 1.1.1.44) was detected. Pyridine nucleotide-linked dehydrogenase activities for l-arabinose and d-galactose (EC 1.1.1.46 and EC 1.1.1.48) occurred in the soluble fraction of cells cultured with either sugar. Glucose-cultured cells contained not only glucokinase (EC 2.7.1.2) and glucose-6-phosphate dehydrogenase (EC 1.1.1.49) activities but also glucose dehydrogenase (EC 1.1.1.47) activity. Enzymes capable of oxidizing gluconate were not detectable, but gluconokinase (EC 2.7.1.12) activity was present. Paper chromatographic analysis of the spent culture supernatant media from glucose-cultured cells indicated an accumulation of gluconic acid, and this was confirmed by enzymatic methods. Evidence is presented for the production of d-galactonic and l-arabonic acids in cultures containing d-galactose or l-arabinose, respectively.  相似文献

介绍了一种用1-甲基咪唑为溶剂和催化剂、盐酸羟胺和乙酸酐为肟化和乙酰化试剂,对植物样品中糖与糖醇进行乙酰化衍生化后的气相色谱分离和质谱鉴定的分析方法.并对糖与糖醇乙酰化影响较大的反应温度、反应时间、反应物组成和反应物浓度等条件进行了比较研究,确定了糖与糖醇乙酰化各步反应的最佳反应温度和反应时间,分析了各组分间相互作用及其用量对衍生化效率的影响.对多种糖与糖醇乙酰化产物进行毛细管气相色谱分离、FID检测及GC-MS结构鉴定.研究证明,在合适条件下应用此方法对糖与糖醇进行乙酰化,反应完全,产物单一,能得到理想的分离、检测和定量分析效果.适用于微量植物组织中多种单糖、双糖及其糖醇的定量分析.  相似文献

This work is focused on the research and verification of new processes of sucrose crystallization in the sugar technology. A major part of the work deals with testing of different conditions at crystallization of sucrose either from sugar raw juice treated by micro‐ and ultrafiltration or from technical sugar solution extracted from continuous chromatographic separation. The equilibrium data were obtained by means of a crystallization test and the width of metastable zone was determined by the ultrasonic sensor. Juice thickening to demanded dry matter content was carried out on a climbing film evaporator under reduced pressure. Results approved possibility of sucrose crystallization from sugar juice without any previous exacting chemical procedure and thus reduce energy and environmental heftiness of sugar production.  相似文献

Mycoplasma gallisepticum cells were found to contain two different sugar transport systems, one for d-glucose and alpha-methyl-d-glucoside (alpha-MG) and the other for d-mannose and d-fructose. Both systems were noninducible, stereospecific, dependent on temperature and pH, and sensitive to sulfhydryl-blocking reagents. The rate of sugar uptake depended on its external concentration, obeying Michaelis-Menten kinetics. The sugar accumulated in the cells against a concentration gradient, and an energy requirement for accumulation was demonstrated with alpha-MG. Both transport systems thus meet the criteria of active transport. The exit of alpha-MG from the cells, like its entry, depended on temperature and was accelerated by energy supplied by the oxidizable d-mannose. d-Glucose accelerated alpha-MG exit, apparently by an exchange reaction. A method for measuring the intercellular space and intracellular free-water volume of Mycoplasma was devised, and several of its applications are described.  相似文献

The detection and discrimination of chemical compounds in potential foods are essential sensory processes when animals feed. The fruit fly Drosophila melanogaster employs 68 different gustatory receptors (GRs) for the detection of mostly nonvolatile chemicals that include sugars, a diverse group of toxic compounds present in many inedible plants and spoiled foods, and pheromones [1-6]. With the exception of a trehalose (GR5a) and a caffeine (GR66a) receptor [7-9], the functions of GRs involved in feeding are unknown. Here, we show that the Gr64 genes encode receptors for numerous sugars. We generated a fly strain that contained a deletion for all six Gr64 genes (DeltaGr64) and showed that these flies exhibit no or a significantly diminished proboscis extension reflex (PER) response when stimulated with glucose, maltose, sucrose, and several other sugars. The only considerable response was detected when Gr64 mutant flies were stimulated with fructose. Interestingly, response to trehalose is also abolished in these flies, even though they contain a functional Gr5a gene, which has been previously shown to encode a receptor for this sugar [8, 9]. This observation indicates that two or more Gr genes are necessary for trehalose detection, suggesting that GRs function as multimeric receptor complexes. Finally, we present evidence that some members of the Gr64 gene family are transcribed as a polycistronic mRNA, providing a mechanism for the coexpression of multiple sugar receptors in the same taste neurons.  相似文献

To study the behavior of Bifidobacterium toward oxygen, oxygen uptake was investigated in detail. The cells of Bifidobacterial strains absorbed considerable amounts of oxygen. The exogenous oxygen uptake activity changed depending upon the period of incubation. Bifidobacterial cells also had high endogenous oxygen uptake, which was, in B. longum strains, as high as about 80% of the exogenous oxygen uptake activity. Bifidobacterial cells accumulated considerable amounts of polysaccharide, which was associated with cellular growth. By incubating the cultivated cells in a glucose-free medium, the endogenous oxygen uptake activity was decreased with a decrease of intracellular polysaccharide. Therefore it was postulated that the high endogenous oxygen uptake activity of Bifidobacterium was owing to the metabolism of intracellular polysaccharide. The enzymatic activity, which was involved in the mechanism of oxygen uptake, was also investigated.  相似文献

The fermentative production of uridine diphosphate N-acetylglucosamine (UDPAG) from 5′-UMP and glucosamine by dried cells of baker’s yeast was studied. UDPAG was found to accumulate in a reaction system containing 5′-UMP, glucosamine, fructose, inorganic phosphate and magnesium ions with dried baker’s yeast as an enzyme source. UDPAG was separated from the reaction mixture by means of anion exchange column chromatography and was identified by several biochemical methods.

The reaction conditions for the fermentative production of UDPAG were examined. The yield of UDPAG was about 40~66% based on 5′-UMP added when fermentation conditions were optimized. The concentration of glucosamine and potassium phosphate buffer, and pH as well as the water content of dried cells greatly affected the formation of UDPAG.  相似文献

A comparative study was carried out on the sugar composition of lipopolysaccharides (LPS) isolated from representative strains of members of the family Vibrionaceae including all of the constituting genera, i.e., Vibrio, Aeromonas, Photobacterium, Plesiomonas, and Lucibacterium. More than 100 strains were examined. It was found that, with the exception of Vibrio parahaemolyticus 06, 2-keto-3-deoxyoctonate (KDO), known generally as a component sugar in the core region of usual gram-negative bacterial LPS, is virtually absent from LPS of the Vibrionaceae strains so far examined. Furthermore, mannose was also lacking in LPS of Vibrionaceae strains with the exception of only one strain, A. anaerogenes (ATCC 15467). Instead, some KDO-like substances were found in LPS from Vibrio (“Beneckea”) nereida (ATCC 25917) and Plesiomonas shigelloides including the type strain (ATCC 14029), the same as those found in LPS from V. parahaemolyticus O7 and O12, and three strains of V. alginolyticus. These substances were strongly positive in the periodate-thiobarbituric acid test, yielding a color with maximum absorption at 549 nm. The spectra were identical to that of KDO, whereas substances differed from KDO at least in behavior in high-voltage paper electrophoresis and thin-layer chromatography. A particularly interesting feature from the chemotaxonomical point of view was found in the sugar composition of LPS isolated from V. cholerae. Fructose was present exclusively in LPS of V. cholerae (both O1 and non-O1 groups and classical and eltor biotypes) with the exception of one strain of Photobacterium phosphoreum (NCMB 844). In addition, a pair of rarely occurring amino sugars, perosamine and quinovosamine, was found in LPS from O1 group V. cholerae regardless of either the biotype (classical or eltor) or the serotype (Inaba or Ogawa), whereas this pair was not present in non-O1 group V. cholerae (the so-called NAG vibrios). This feature was confirmed with LPS from more than 30 additional strains of O1 group V. cholerae isolated from patients. The virtual absence of KDO in LPS of the family Vibrionaceae was demonstrated for the first time in this study. These results are compatible with the interpretation that the absence of KDO in LPS can be used as one of the taxonomical characteristics of Vibrionaceae in addition to (G+C) content, DNA (or RNA) homology, and numerical analysis data.  相似文献
