We report the synthesis of covalent conjugates of nanodiamonds with doxorubicin and a cytostatic drug from the class of 1,3,5-triazines. The obtained conjugates were identified using a number of physicochemical methods (IR-spectroscopy, NMR-spectroscopy, XRD, XPS, TEM). As a result of our study, it was found that ND-СONH-Dox and ND-COO-Diox showed good hemocompatibility, since they did not affect plasma coagulation hemostasis, platelet functional activity, and erythrocyte membrane. The ND-COO-Diox conjugates are also capable of binding to human serum albumin due to the presence of ND in their composition. In the study of the cytotoxic properties of ND-СONH-Dox and ND-COO-Diox in the T98G glioblastoma cell line, indicating that ND-СONH-Dox and ND-COO-Diox demonstrate greater cytotoxicity at lower concentrations of Dox and Diox in the composition of the conjugates compared to individual drugs; the cytotoxic effect of ND-COO-Diox was statistically significantly higher than that of ND-СONH-Dox at all concentrations studied. Greater cytotoxicity at lower concentrations of Dox and Diox in the composition of conjugates compared to individual cytostatics makes it promising to further study the specific antitumor activity and acute toxicity of these conjugates in models of glioblastoma in vivo. Our results demonstrated that ND-СONH-Dox and ND-COO-Diox enter HeLa cells predominantly via a nonspecific actin-dependent mechanism, while for ND-СONH-Dox a clathrin-dependent endocytosis pathway. All data obtained provide that the synthesized nanomaterials show a potential application as the agents for intertumoral administration.  相似文献

Four-year monitoring of phenology of the blowfly Caliphora vicina under quasi-natural conditions in the environs of St. Petersburg demonstrated development of two complete generations and overwintering of the third generation. The duration of preimaginal development (from egg laying to adult emergence) of the 1st and 2nd generations varied from 19 to 32 days; the required sum of effective temperatures (SET) was 326 ± 31 degree-days with a threshold of 5°С. Reproductive maturation of females (the period of time from adult emergence to laying of the first eggs) lasted from 15 to 31 days (SET was 262 ± 39 degree-days with a threshold of 5°С). The duration of development of one complete generation (from egg stage to the beginning of oviposition) during the period of observation varied from 34 to 57 days (SET was about 600 degree-days with a threshold of 5°С). The first diapausing larvae hatched from the eggs laid in the middle of August. In September, at shorter day lengths and at a temperature of 10–11°С, 80–100% of the larvae entered diapause. A small fraction of the larvae of the 1st and 2nd generation and most of the larvae of the 3rd generation overwintered. Successful overwintering of adults in the environs of St. Petersburg is hardly possible.  相似文献

In natural populations, insects regularly face an adverse impact of different natures: harsh weather swings, lack of food resources, the insecticidal treatment. We studied the effect of repeated episodes of mild heat stress of different frequencies on stress resistance of Drosophila melanogaster females. We found out that the mild heat stress (38°С, 1 hr) repeated daily within 2 weeks resulted in (a) an increased activity of the dopamine (DA) metabolism enzymes, DA‐dependent arylalkylamine N‐acetyltransferase and alkaline phosphatase, which suggested a decrease in DA level, and (b) an increased survival rate under acute heat stress (38°С, 4 hr). The same mild heat stress repeated weekly had no effect on these parameters.  相似文献

The effect of temperature on sexual reproduction and the rate of vegetative division were investigated in laboratory experiments with Haslea ostrearia (Gaillon) Simonsen from the Atlantic and H. karadagensis Davidovich, Gastineau et Mouget from the Black Sea. In the range of 10–26°С, a temperature increase was favorable for vegetative growth; the maximum growth rate was recorded at the highest temperature of the range. In contrast to vegetative growth, sexual reproduction of both algal species was effective in the temperature range of 10–18°С, but did not occur at temperatures of 23°С. The optimal temperature for auxosporulation was in H. karadagensis somewhat lower than in H. ostrearia. The determined physiological optimums for sexual reproduction can explain the cell-size distribution that is characteristic of the natural population of H. karadagensis in different seasons of the year.  相似文献

The Sonogashira reaction can be applied for the preparation of acetylenic derivatives of betulonic acid where the triterpenoid moiety can serve as either the halo- or the acetylenic component. This reaction opened access to the first derivatives of betulonic acid containing either the arylethynyl (СС-Ar(Het) or the ethynyl (ССН) moieties. From the fundamental perspective, this work illustrates the possibility of selective Pd-catalyzed cross-coupling at terminal acetylenes in the presence of a terminal alkene. Hepatoprotective and anti-inflammatory properties of selected acetylenic derivatives of betulonic acid were investigated using the CCl4-induced hepatitis and carrageenan-induced edema models, respectively.  相似文献

Устойчивость каппа-фага (Serratia marcescens) и его мутанта «С» исследовалась в буферных растворах при pH от 2.5 до 12 с целью подтверждения, или опровержения, возможности их инактивирования в результате кратковременного доведения кислотности основной суспензии в течение приготовления концентрата фага до pH 4. Было установлено, что каппа-фаг и его с-мутант в среде с pH 4 и выше остаются относительно устойчивыми и что в этих пределах pH между обоими фагами нет существенных различий. Однако при низких pH (2,5–3,5) титр обоих фагов вскоре довольно резко падает, причем с-мутантинакмпбпруемся бысмрее, чем каппа-фаг.  相似文献

2,3-Indolotriterpenic alcohols have been synthesized for the first time by successive modification of 3-oxo triterpenic acids (Fisher reaction, reduction of С17-СOOH, cyanoethylation) and characterized by physicochemical methods of analysis. It has been found that 2,3-indolouvaol and 2,3-indolo-28-cyanoethoxybetulin exhibit antitumor activity in vitro toward NCI-H522 lung cancer (–12.65%) and COLO 205 colon cancer cells (–42.78%), respectively. The activity of 2,3-indolooleanolic and 2,3-indolobetulinic acids toward 19 and 9 cell lines of six and four human cancers, respectively, has been revealed. Indole-fused triterpenoids have been shown to hold promise as objects in the search for novel antitumor agents.  相似文献

Microvesicles were isolated from blood plasma and total blood of healthy females and breast cancer patients by filtration and ultracentrifugation. According to flow cytometry, different subpopulations of exosomes were represented in blood of healthy donors and cancer patients at different levels with median fluorescence intensity (MFI) values in both groups arranged in the following order: CD24/СD9 > СD9/СD81 > CD9/CD63 = CD24/CD63. Concentration of exosomes in blood plasma of healthy females estimated by nanoparticle tracking analysis (NTA) did not exceed (3.71 ± 1.15) × 107 particles/mL of blood and did not differ from that in plasma of breast cancer patients, which averaged (3.99 ± 1.03) × 107 particles/mL of blood. Concentration of total exosomes in blood (including exosomes from plasma and blood cell surface-bound exosomes) did not depend on the presence/absence of a tumor; the values were (7.66 ± 0.7) × 107 particles/mL of healthy blood and (9.4 ± 1.24) × 107 particles/mL of blood from cancer patients. Comparative analysis of exosomes using 2-D electrophoresis with subsequent analysis of 2-D proteomic maps revealed proteins missing in blood or differentially expressed in healthy females and breast cancer women. The data presented provide the possibility for identification of exosomal proteomic markers and isolation of tumor-specific exosomes, which contributes to the development of breast cancer diagnostics.  相似文献

Pacific halibut Hippoglossus stenolepis is registered in the Anadyr estuary (Bering Sea basin) for the first time: 20 specimens, 18.5–160.5 cm in length, were caught by trawl and longline within the sea portion of the Anadyr River estuary in August 2010 and September 2015 in highly desalted (23.5‰ on average) and heated (6.5°С) water at a depth of 14–30 meters. Halibut is suggested to form feeding groups within the Anadyr Estuary. An opportunity for organized fishing in this region is discussed.  相似文献

Prototypes of lithium emitters and collectors designed on the basis of capillary-porous systems (CPS) are being tried-out at the T-11M tokamak within the concept of continuous lithium circulation, previously proposed for stationary fusion neutron sources (FNSs). One of the goals of the T-11M tokamak research program are to implement the closure of the lithium circulation circuit and to develop the technology of recuperation (extraction and removal from the tokamak chamber) of hydrogen isotopes by using special lithium CPS-based collectors in order to prevent hydrogen isotope accumulation on the inner surface of the discharge chamber. In order to determine the optimal temperature regime for the future FNS operation, the effect of the temperature of the collector surface on its ability to capture lithium and hydrogen isotopes was studied. It is ascertained that the efficiency of capturing lithium and hydrogen isotopes by metal (12Cr18Ni10Ti) collectors in the operating regimes of the T-11M tokamak depends on the temperature of their collecting surfaces; namely, it remains almost constant in the temperature range from–196 to 50°С and then gradually decreases as the temperature rises to 300–400°С. In this case, the amount of the collected lithium is reduced by no more than sixfold, whereas the amount of collected hydrogen isotopes decreases by more than two orders of magnitude. Thus, the wall of a tokamak reactor chamber, which will be heated to 400°С but still coated with residual lithium (or its chemical compounds), will serve as a “mirror” for the incident hydrogen isotopes. It is found that the liquid lithium surface of the CPS-based collector can efficiently capture hydrogen isotopes falling onto it. As the lithium CPS-based collector is heated from 100 to 240°С, the amount of captured deuterium is reduced only twofold. This means that, in a steady-state mode, if, e.g., an MHD pump is used to transport liquid lithium from the collector back to the emitter, then, together with lithium, the captured hydrogen isotopes can also be transported into the recuperation zone.  相似文献

Ratios of 13С/12C and 15N/14N isotopes were identified in different parts and organs of drooping birch (Betula pendula Roth) in preforest-steppe and pine-birch forests of the Middle Urals by mass spectrometry. The data were analyzed and interpreted from the perspective of biochemical processes of carbon and nitrogen metabolism in the leaf, cambial tissue, trunk wood, branches, roots, and in the soil. The lighter isotopic composition of carbon is characteristic for the leaves, trunk cambium as well as fine (<2 mm) roots. The trunk wood is characterized by the basal trend for 13C enrichment. The heavier carbon isotopic composition inversely related to metabolic activity of organs and tissues, in addition, 13С/12C ratio corresponds to the nitrogen content in the organs and tissues, indicating the metabolic control of carbon fractionation in woody plants. The isotopic composition of nitrogen in the aboveground parts of the plant (leaves, trunk cambium, wood) and in the medium and fine roots was significantly depleted in 15N (δ15N varies from 0 to–3‰), while main roots (δ15N = 0.6 ‰) and soil (δ15N = 2.4–6.7‰) were more enriched. The ratio of stable isotopes of carbon and nitrogen is an integrating index of carbon and nitrogen metabolism in plants.  相似文献
