During our work on psychrophilic microorganisms we obtained a large collection of new isolates. In order to identify six of these, we examined their growth properties, cell wall compositions, and their 16S rRNA gene sequences. The results showed that all of the isolates are gram-positive, aerobic, contain lysine in their cell walls, and belong to the high mol% G+C Arthrobacter subgroup. Phylogenetic analysis of the 16S rRNA genes grouped five isolates obtained from a small geographical region into a monophyletic clade. Isolate B7 had a 16S rRNA sequence that was 94.3% similar to that of Arthrobacter polychromogenes and 94.4% similar to that of Arthrobacter oxydans. Primary characteristics that distinguish isolate B7 from the Arthrobacter type strain (Arthrobacter globiformis) and A. polychromogenes include lack of growth at 37 degrees C, growth at 0-5 degrees C, the ability to use lactose as a sole carbon source, and the absence of blue pigments. Because of these differences, isolate B7 was chosen as a type strain representing a new Arthrobacter species, Arthrobacter psychrolactophilus. The sixth isolate, LV7, differed from the other five because it did not have the rod/ coccus morphological cycle and was most closely related to Arthrobacter agilis.  相似文献

抗锌细菌Sphingomonas sp. DX-T3-03分离、鉴定及性质   总被引:1,自引:0,他引:1  
从江西德兴铜矿重金属污染土壤中筛选得到一株对重金属锌具有极强抗性的菌株,命名为DX-T3-03。对该菌株进行形态观察、生理生化试验,采用16S rRNA序列分析,鉴定该菌为鞘氨醇单胞菌属(Sphingomonas sp.)。研究其最佳生长条件及抗重金属特性。试验结果表明:该菌株的最适应生长条件为温度35°C,pH约6.7,转速150r/min;对重金属锌有极高抗性,可以达到25mmol/L及以上,并能够在多种单一及复合重金属(Cu70mg/L、Cd300mg/L、Pb400mg/L、Ni60mg/L)中生长。  相似文献

拟双角斯氏线虫共生细菌的分离与鉴定   总被引:2,自引:0,他引:2  
从我国东北地区采集的拟双角斯氏线虫(Steinernema ceratophorum)肠道内分离到1株具有较强生物活性功能的致病杆菌菌株CB43。形态特征及生理生化特征测定结果表明,CB43菌株与致病杆菌属(Xe-norhabdus)的基本特性相似;对寄主线虫的产量有明显的促进作用,其代谢物对细菌和真菌均有较强的抑制活性,符合线虫共生菌的基本特性和功能。16S rRNA序列及根据16S rRNA序列构建的系统发育树中,CB43菌株与Xenorhabdus budapestensis序列同源性最高,形成一个类群。但CB43菌株不能产生吲哚,在麦芽糖、海藻糖、D-葡萄糖酸和乙酸利用等生化特征与X.budapestensis存在一定的差异,可能是由于菌株的生态差异造成的。根据形态及生理生化特征,结合16S rRNA序列分析,CB43菌株属于Xenorhabdus budapestensis。  相似文献

高效降解环己酮的无色杆菌JDM-3-03株的分离和鉴定   总被引:2,自引:0,他引:2  
目的:分离、鉴定高耐受和高效降解环己酮的菌株。方法:从采自岳阳巴陵石化公司环己酮生产车间总出水口的污泥中,通过逐步驯化筛选环己酮降解菌株;通过形态观察、生理生化特征检测和基于16S rRNA基因序列的系统发育分析对分离到的菌株进行初步鉴定。结果:分离得到一株环己酮降解菌株JDM-3-03,初步鉴定其为无色杆菌Achromobacter insolitus的一个菌株;该菌能以环己酮为惟一碳源,且能耐受5000mg/L的环己酮;当环己酮的质量浓度为2000mg/L时,在温度为30℃、转速为150r/min的条件下,72h内该菌株对环己酮的降解率达到90.17%。结论:菌株JDM-3-03是一株可高效降解环己酮的无色杆菌。  相似文献

从处理硫酸盐废水厌氧折流板反应器(Anaerobic battqe reactor,ABR)的污泥中分离到1株硫酸盐还原菌,对该菌株进行了形态、生理生化特性方面的研究,并对16S rDNA序列进行了分析。该菌株为杆状或弧状,大小为(0.5~0.7)μm×(1.4~1.9)μm,革兰染色阴性,芽胞染色阴性,能运动,具有硫酸盐还原功能。菌株最适生长pH为7.0~8.0,喜中性偏碱环境;初始[SO4^2-]为2000mg/L,OD600nm。值为1.206,SO4^2-去除率达到71%;该菌株能分别利用乳酸、丙酮酸、丁酸、乙酸、乙醇、甲醇、葡萄糖作为电子供体,进行硫酸盐异化还原,乳酸最有利于该菌SO4^2-的去除,SO4^2-去除率为91.4%,其次为丙酮酸,达到51.2%。基于16SrDNA序列同源性构建了系统发育树,结果表明此株菌是属于脱硫弧菌属(Desulfovibrio)的硫酸盐还原菌,与Desulfovibrio具有96.0%的序列相似性。  相似文献

从黄浦江淀山湖的水底沉积物中筛选分离得到一株产纤维素酶的菌株。经细菌形态观察,生理生化实验并结合16SrRNA序列分析,鉴定该菌为蜡状芽孢杆菌(Bacillus cereus),同时发现其在系统发育树中处于一个独立的分支,推测该菌为Bacillus属中一个新的亚种。对该菌株产酶及酶活特性进行了初步研究,发现纤维素酶的产生与细菌的生长密切相关。该菌株在37°C,pH7.0的条件下,发酵66h后纤维素酶活达到最高值4.58U/mL。  相似文献

从云南轮马热泉下游淤泥中筛选得到了一株产耐热普鲁兰酶菌株LM14-2.根据形态特征及16S rRNA序列同源性分析,初步判定为Anoxybacillus sp.LM14-2.该菌株发酵上清液中有耐热普鲁兰酶积累,其反应最适pH值为6.0,最适温度为70℃.利用染色体步移技术获得了完整的普鲁兰酶编码基因(HQ660582),经序列相似性进一步分析,确定该蛋白与Ⅰ型普鲁兰酶保守区b相吻合.通常的普鲁兰酶在高温下很快失活,难以满足淀粉加工,洗涤剂等相关工业的需求,而该新型的耐热普鲁兰醇的作用温度广泛,热稳定性较好,65℃保温55 h后达到其半衰期,具有广阔的开发应用前景.  相似文献

从中国农业科学院北京畜牧兽医研究所鸡舍附近土壤中分离到一株芽孢杆菌P-25,并进行了分子鉴定。通过形态鉴定、革兰氏染色、生理生化测定、16SrRNA序列分析和系统发育树构建,确定该菌株为蜡状芽孢杆菌(Bacillus cereus),其16SrRNAGenBank登录号为GU271135。  相似文献

从河底污泥中分离到一株产氢菌,通过16S rDNA序列和系统发育分析,鉴定为克雷伯氏菌属,命名为Klebsiella sp. HQ-3.运用单因子试验考察了摇瓶发酵培养基中碳源、氮源、无机盐等因素对克雷伯氏菌HQ-3发酵产氢的影响,并对其产氢特性做了初步研究.结果表明,克雷伯氏HQ-3在葡萄糖浓度为24 g/L,蛋白胨为18 g/ L,KH2PO4为 8g/ L时具有较高产氢量,其最佳产氢初始pH为8.0左右.  相似文献

从四川自贡某盐井壁植物根系土壤中分离得到1株中度嗜盐解磷菌QW1011。该菌细胞呈线状, 大小为0.8 μm×30 μm~100 μm, 革兰氏染色为阳性、最适NaCl生长浓度为10%, NaCl最高耐受浓度15%。好氧生长, 酪素水解、硝酸还原和接触酶阴性。菌株的16S rRNA基因序列(接受号:EF647207)与Bacillus megatherium ATCC 14581的16S rRNA相似性为100%, 其16S-23S rRNA间区(ISR)的PCR扩增片的PAGE指纹图谱与参考菌株Baci  相似文献

Abstract The phylogenetic position of the obligate fish pathogenic Gram-positive eubacterium Renibacterium salmoninarum was determined by the 16S ribosomal RNA cataloguing approach. Comparison of the catalogue to those of more than 165 Gram-positive organisms from 50 genera revealed that R. salmoninarum is a member of the actinomycetes subdivision. As derived from similarity coefficients (SAB values), this organism is related to a subgroup harbouring morphologically and chemotaxonomically rather heterogeneous taxa, including Anthrobacter, Micrococcus, Cellulomonas, Jonesia, Promicromonospora, Stomatococcus and Brevibacterium . Thus, the inclusion of organisms characterized by regular rods increases the phenotypic variability of this phylogenetically coherent group.  相似文献

Two Gram-positive, catalase-positive, oxidase-negative, motile, endospore-forming, rod-shaped bacteria, designated as 0911MAR22V3T and 0911TES10J4, were isolated from air samples collected in two show caves, located in Andalusia, Southern Spain. Phylogenetic analysis based on 16S rRNA gene sequences indicated that both strains were indistinguishable and they were most closely related to Bacillus humi DSM 16318T (98%). DNA–DNA hybridization values of the strain 0911MAR22V3T with respect to strain 0911TES10J4 and B. humi DSM 16318T were 76.8% (73.9%, reciprocal) and 56.9% (63.3%, reciprocal analysis), respectively. Whole genome average nucleotide identity (ANI) values of both strains were in the threshold value for species delineation and less than 85% with B. humi. Strains 0911MAR22V3T and 0911TES10J4 grew at 10–47 °C (optimum 37 °C), at pH 6–9.5 and with 0–8% (w/v) NaCl (optimum 1%). In both strains the dominant isoprenoid quinone was MK-7, the major cellular polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, and two more phospholipids, the predominant fatty acids were iso-C15:0 and anteiso-C15:0 and the DNA G + C content was 38 mol%. On the basis of their phylogenetic relatedness and their phenotypic and genotypic features, the strains 0911MAR22V3T and 0911TES10J4 should be attributed to a novel species within the genus Bacillus, for which the name Bacillus onubensis sp. nov. is proposed. The type strain is 0911MAR22V3T (=LMG 27963T = CECT 8479T); and strain 0911TES10J4 (CECT 8478) is a reference strain.  相似文献

Novel orange pigmented, Gram-negative-staining, rod-shaped, non-motile, strictly aerobic strains designated NIO-S1(T) and NIO-S2 were isolated from the water sample of a pond adjacent to the coast and an algal mat from a fish pond, respectively, at Kakinada, India. Both strains were positive for oxidase, catalase and β-galactosidase activities. The predominant fatty acids in NIO-S1(T) were iso-C(15:0) (39.6%), anteiso-C(15:0) (9.9%), iso-C(17:0) 3OH (10.9%) and C(16:1)ω7c/C(16:1)ω6c (summed feature 3) (5.7%). The strains contained MK-7 as the major respiratory quinine, and diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and three unidentified lipids as the polar lipids. Phylogenetic analysis indicated that strain NIO-S1(T) was a member of the family "Cyclobacteriaceae" of the class "Sphingobacteriia" and it clustered with the genera Fontibacter, Cecembia and Aquiflexum with phylogenetic distances of 6.8, 9.0 and 12.2% (93.2, 91.0 and 87.8% similarity), respectively. DNA-DNA hybridization between strains NIO-S1(T) and NIO-S2 showed a relatedness of 93% and rep-PCR banding patterns were similar. Based on data from the current polyphasic study, it is proposed that the new isolates be placed in a new genus and species with the name Shivajiella indica gen. nov., sp. nov. The type strain of Shivajiella indica is NIO-S1(T) (= KCTC 19812(T)=MTCC 11065(T)).  相似文献

A novel genus and species within the order Glissmonadida (Cercozoa, Rhizaria), Saccharomycomorpha psychra n. g., n. sp., is described from lichen in the Ny-Ålesund region (High Arctic) and moss in the Fildes peninsula of King George Island (Maritime Antarctica). Cells were spherical and did not appear to present flagella in organic-rich Potato Dextrose Agar medium where they were able to feed osmotrophically. Molecular phylogenetic analyses based on 18S rRNA gene sequence demonstrated that Saccharomycomorpha psychra belong to “clade T” within the order Glissmonadida (Cercozoa, Rhizaria). All three investigated strains could grow at 4 °C and had an optimum growth temperature of 12 °C, 20 °C, and 20 °C, while a maximum growth temperature of 20 °C, 20 °C, and 25 °C, respectively. In conclusion, we established the phenotypic identity of “clade T,” which until now was exclusively detected by environmental sequences, and erect a new family Saccharomycomorphidae for “clade T.” Nomenclatural, morphological and ecological aspects of this novel species are discussed.  相似文献

肠道微生物与寄主具有复杂的、多方面的相互依存效应,这种依存效应所产生的共生关系或协同进化关系既可反映寄主间的系统演化关系,也可显示肠道微生物间的系统演化关系,共生关系或协同进化关系是由于寄主与肠道微生物两者之间存在着相互自然选择作用所形成的,在长期的进化历程中逐步发生的共生关系信息很可能被记录在DNA序列中。本文通过检测鱼鲤鱼科8种鱼中9种肠道菌群的分布含量对这9种菌群进行分析,且利用从GenBank调取这9种肠道细菌菌属的43个种或亚种的16S DNA序列的构建NJ树和MP树,将这6个科9个属43个种或亚种分为革兰氏阴性和革兰氏阳性两大类群(一级分枝)。在这两类群中,又以科为单位分为6个亚类群(二级分枝),而肠杆菌科中则以属为单位分为4个小类群(三级分枝),此外球状菌与杆状菌也能截然分开。将16S DNA的NJ树隐去所有的种,以属为单位所得到的以分枝形式的无根树在拓扑结构上与菌群分布含量(寄主范围)所构建的无根树相近,但芽孢杆菌在两种无根树的位置中有较大的差异。如果提高检测水平,扩大所检测的寄主对象,这种差异有可能消除。  相似文献

利用平板筛选法从盐场卤水池中筛选到一株高效降解Tween 20的中度嗜盐菌DF-B6菌株。通过形态学、生理生化及16S rDNA序列分析确定该菌株为Idiomarina属的成员。底物特异性实验确定DF-B6菌株分泌的脂裂解酶为酯酶, 而非脂肪酶。在含8% NaCl的0.05 mol/L Tris-HCl (pH 8.0)缓冲液中, 50°C条件下作用, 酶活性最高。当反应液中金属离子浓度为10 mmol/L时, Mg2+、Ca2+和Mn2+对酶活有激活作用, 而Zn2+、Fe3+和Cu2+则对酶反应有抑制作用。  相似文献

目的:为了探讨3株Pseudomonas sp.对吡啶降解存在多样性.方法:基于16S rRNA和ISR序列分析,对3株分离菌株进行初步鉴定,进而通过Touch -Down PCR,对3株细菌降解吡啶的多样性进行分析.结果:3株细菌XJUHX -1、XJUHX - 12和XJUHX - 16初步鉴定为Pseudomonas,3株实验菌株的部分降解基因的扩增条带有差异.结论:同属的3株 Pseudomonas在吡啶降解上存在多样性.  相似文献

The aerobic degradation of 3- N -trimethylamino-1-propanol (homocholine) as a sole source of carbon and nitrogen has been found for a Rhodococcus sp. bacterium isolated from soil. The isolate was identified as Rhodococcus sp. strain A2 based on its phenotypic features, physiological and biochemical characteristics, and results of phylogenetic analysis. The washed cells of strain A2 completely degraded homocholine within 6 h, with concomitant formation of several metabolites. Analysis of the metabolites using capillary electrophoresis, fast atom bombardment–MS, and GC–MS showed that trimethylamine was the major metabolite, in addition to β-alanine betaine (β-AB) and trimethylaminopropionaldehyde. Therefore, the possible degradation pathway of homocholine in the isolated strain is through consequent oxidation of the alcohol group (-OH) to aldehyde (-CHO) and acid (-COOH). Thereafter, the cleavage of β-AB C–N bonds yielded trimethylamine and alkyl chain.  相似文献
