Typing of 13 strains of fluorescent pseudomonads from the Belarusian collection of nonpathogenic microorganisms (BIM) by ERIC-PCR and BOX-PCR revealed high level of genetic heterogeneity in bacteria, most of which have been previously identified as Pseudomonas fluorescens according to the classical scheme. Evaluation of the similarities of the 16S rRNA gene sequences and their phylogenetic analysis excluded affiliation of the bacteria under study within the same species and allowed them to be distributed within three relatively distant clusters of the genus Pseudomonas phylogenetic tree. While eight strains fell into the phylogenetic group of P. fluorescens, only one of them could be identified as P. fluorescens. Four strains clustered within the P. vancouverensis phylogenetic group, formed by new species, which have been described mainly according to the evaluation of genome relationships. One bacterium was related to a stable branch that did not contain any type strains of the known Pseudomonas spp. These results indicate taxonomic heterogeneity of collection strains of the fluorescent pseudomonads and demonstrate the necessity of identification of them considering the requirements of phylogenetic bacterial taxonomy.  相似文献

A colchicine-resistant mutant of Chlamydomonas reinhardi (col10, 12) which appears blocked in the final stages of the cell division cycle is shown to have an RNA: protein ratio over four times that which is observed in wild-type cultures. This does not appear to be simply a consequence of reduced overall growth rate, because a comparable reduction in the overall growth rate of wild type by caffeine inhibition did not produce such a large rise in RNA content. The RNA levels in six other colchicine-resistant mutants, which have various abnormalities in colchicine-binding activity, has also been investigated. Two of them have elevated RNA levels.  相似文献

 We have generated 66 sequence-tagged-site (STS) markers from cDNA clones of Cryptomeria japonica, and 60% of them have already been mapped into C. japonica linkage groups. All of the STS markers showed a single fragment following polymerase chain reaction (PCR) amplification. We investigated by polymorphism of these STS markers in a mapped F2 population and 15 plus trees by means of a restriction endonuclease analysis. Polymorphism levels were 10.6% and 22.7% in the F2 population and the 15 plus trees, respectively. PCR amplification levels of the 66 STS markers in 14 conifer species varied depending on their genetic relationship with C. japonica. Taxodium, which is closely related to C. japonica, had the most amplifications (31.82%), followed by Sequoiadendron giganteum, which is of the same family. The average proportion of PCR amplifications in each family gradually declined in the following order: from Taxodiaceae to Cuppresaceae, Sciadopityaceae, Pinaceae, and Taxaceae. These results are in general agreement with a molecular phylogenetic relationship based on chloroplast DNA. The 66 STS markers will be useful as on anchor point for genome mapping and population genetics, and some of them will also be useful when studying other conifers. We first show how to formalize environments in which situations can be changed only by action, and in which a situation should be brought about that is acceptable. This leads us to the concept of a problem constellation that consists of an action system, a reaction system, a judgement system, and an initial situation. We then present a cybernetical system which we call Robbimat. It is capable of testing problem constellations, of modifying them according to the results of the tests, and of judging the final constellation. We designed Robbimat in order to have favourable problem constellations automatically generated. As an example we discuss a simple game. It constitutes a win-loss-environment the structure of which can be deduced from the payoff function. We show how Robbimat analyses such an environment, and utilizing its structure classifies the initial situation.

The composition, the hylogenesis and the sstematic arrangement of the family Calohypsibiidae are discussed. To this imil the genera Calohypsibius, Haplomacrobiotus, Parhexapodibius, Hexa-podibius and Haplohexapoibius are to be ascried. Within the family two main hyletic lines can be distinguished. In one of them the diloclaws and the legs do not tend to be reduced (Calohypsibius), in the other the diploclaws and the legs have been affected by a more or less advanced and differentiated process of reduction. Within the latter ghyletic line two evolutionary trends can be distinguished; in the former the process of reduction as mostly affected the claws of the first three airs of legs where, by loss of the secondary branches, they have become single claws (Haplomacrobiotus). In the latter evolutionary trend the process of reduction has affected mostly the claws of the hind legs and, more or less, also the very legs. In the most primitive stem of this evolutionary trend the claws are dipclaws and they are present in all legs (Parhexapodibius). From the primitive stem a phyletic line as arisen in which the claws of the hind legs are quite lacking and the Kind legs are reduced to short stumps. This hyletic line is subdivided in two sister lines: in one of them the claws of the first three pairs of legs are reduced in size but maintain the secondary branches and they are then diloclaws (Hexapoditius), in the second phletic line the claws, by loss of the secondary branches, have become single claws (Haplohexapodibius). The systematic position of Apodibius is discussed. Up to this time it is not absolutely sure that this genus is belonging to the family Calohypsibiidae.  相似文献

This paper describes a range of cooperative displays in the loriine parrot genus Trichoglossus. The displays tend to be performed jointly by the members of established monogamous pairs, and have evolved specialised characteristics which distinguish them from solo performances. The possible functions of these displays are discussed with reference to observations of apparently analogous performances in other monogamous avian and mammalian groups.  相似文献

Summary We report a detailed molecular analysis of three chromosomal rearrangements, which have been produced during I-R hybrid dysgenesis in Drosophila melanogaster. They all disrupt the yellow gene. One of them is a deletion; the other two are inversions, which may be interpreted as the results of recombination events between I elements inserted at their break points. These events appear to occur at the time of transposition and involve integrating rather than resident I elements. They are produced by a mechanism very similar to homologous ectopic recombination.  相似文献

Gammaglobulin groups (Gm and Inv) of various Southern African populations   总被引:9,自引:0,他引:9  
Data are presented on the distribution of the Gm and Inv groups in approximately 3500 individuals belonging to a number of diverse Southern African populations. The indigenous peoples show the presence of the Gm alleles known to occur in Negroes (Gm1, 5, 13, 14, Gm1, 5, 6, 14 and Gm1, 5, 6) but the Bushmen possess some of them in very low frequencies and have, in addition and in appreciable frequencies the Gm1 and Gm1, 13 alleles which have not been reported as occurring in West African populations. The distribution of the Gm1, 13 allele in various Bantu-speaking tribes of the sub-continent reveals a marked cline, increasing from north to south along the eastern seaboard. The correlation between the frequency of Gm1, 13 and the Khoisan morphological, features present in a number of the tribes, and with the linguistic evidence which has been used to group them is high. The Bushmen possess a Gm1, 5 allele and may also have a Gm1, 5, 13, 14, 17, 21 allele. A Gm1, 2, 5, 13, 14, 17 allele seems to be present in the Bantu. Its presence in Eastern New Guinea would also appear to be indicated by the population data presented here.  相似文献

Glucosinolates are sulphur‐containing secondary metabolites characteristic of Brassicaceous plants. Glucosinolate breakdown products, which include isothiocyanates, are released following tissue damage when hydrolytic enzymes act on them. The isothiocyanates have toxic effects on generalist herbivores when they attempt to feed on oilseed rape, Brassica napus, and also function as repellents. However, specialist herbivores such as Brevicoryne brassicae aphids, flea beetles, Psylliodes chrysocephala and the Lepidopteran pest, Pieris rapae, are adapted to the presence of glucosinolates and thrive on plants containing them. They may do this by avoiding tissue damage to prevent the formation of isothiocyanates or by metabolising or tolerating glucosinolates. For many specialist herbivores, the isothiocyanates function as attractants and glucosinolates can even be sequestered for defence against predatory insects. Thus, these herbivores have evolved resistance to host‐plant secondary metabolites and this type of evolutionary history may have given some insects an enhanced ability to adapt to xenobiotics. In an agricultural context, this may make pests better able to evolve resistance to artificially applied pesticides. The effect of increased glucosinolate content in making oilseed rape cultivars more susceptible to specialist pests was highlighted in a seminal article in the Annals of Applied Biology in 1995. This review of the literature considers developments in this area since then.  相似文献

Water Content and the Responsivity of Lettuce Seeds to Light   总被引:1,自引:0,他引:1  
Lettuce seed (Lactuca sativa L. cv. Grand Rapids) prevented from attaining maximum water content are still able to respond to irradiation if the seed moisture content is above 6%. When the limitation to water uptake is by immersion in a solution of either polyethylene glycol 400 (PEG 400), mannitol, or sucrose, the responsiveness of the seed is lessened. With PEG 400 the molarity which reduces germination by about 50% is 0.03 while mannitol and sucrose must be present at 0.07M and 0.05 M respectively to bring about the same reduction. While red light overcomes any additional effect 0.1 M mannitol and 0.025 M PEG 400 have on lettuce seed at 30°C (at which temperature the sample was positively photoblastic), irradiation did not overcome the effect of 0.1 M sucrose. Gibberellic acid did not have any effect on any of the treated seed. The solutes used cannot be considered as inert. Some chemical activity is associated with them and it is unwise to consider using these molecules to establish a particular level of water potential in the liquid bathing the seed.  相似文献

GTPase activating protein (GAP) is a known regulator of ras-p21 activity and is a likely target of ras-induced mitogenic signaling. The domains of GAP that may be involved in this signaling are unknown. In order to infer which domains of GAP may be involved, we have performed molecular dynamics calculations of GAP complexed to wild-type and oncogenic (Val 12–containing) ras-p21, both bound to GTP. We have computed and superimposed the average structures for both complexes and find that there are four domains of GAP that undergo major changes in conformation: residues 821–851, 917–924, 943–953, and 1003–1020. With the exception of the 943–953 domain, none of these domains is involved in making contacts with ras-p21, and all of them occur on the surface of the protein, making them good candidates for effector domains. In addition, three ras-p21 domains undergo major structural changes in the oncogenic p21-GAP complex: 71–76 from the switch 2 domain; 100–108, which interacts with SOS, jun and jun kinase (JNK); and residues 122–138. The change in conformation of the 71–76 domain appears to be induced by changes in conformation in the switch 1 domain (residues 32–40) and in the adjacent domain involving residues 21–31. In an accompanying paper, we present results from microinjection of peptides corresponding to each of these domains into oocytes induced to undergo maturation by oncogenic ras-p21 and by insulin-activated wild-type cellular p21 to determine whether these domain peptides may be involved in ras signaling through GAP.  相似文献

Intrafamilial relationships among clubtail dragonflies (Gomphidae) have been the subject of many morphological studies, but have not yet been systematically evaluated using molecular data. Here we present the first molecular phylogeny of Gomphidae. We include six of the eight subfamilies previously suggested to be valid, and evaluate generic relationships within them. We have included examples of all genera reported from the Nearctic except Phyllocycla. This sample includes all North American species of Ophiogomphus, which has allowed us to explore intrageneric relationships in that genus. Our particular focus is on the closest relatives of the genus Gomphus, especially those North American species groups that have been commonly treated as subgenera of Gomphus. The Gomphus complex is split into additional genera, supported by molecular and morphological evidence: Phanogomphus, Stenogomphurus, Gomphurus and Hylogomphus are here considered to be valid genera. The genus Gomphus, in our restricted sense, does not occur in the western hemisphere; in addition, G. flavipes is transferred to Stylurus.  相似文献

In the accompanying article, using molecular dynamics calculations, we found that the 66–77 and 122–138 domains in ras-p21 and the 821–827, 832–845, 917–924, 943–953, and 1003–1020 domains in GAP have different conformations in complexes of GAP with wild-type and oncogenic ras-p21. We have now synthesized peptides corresponding to each of these domains and coinjected them into oocytes with oncogenic p21, which induces oocyte maturation, or injected them into oocytes incubated with insulin that induces maturation by activating wild-type cellular ras-p21. We find that all of these peptides inhibit both agents but do not inhibit progesterone-induced maturation that occurs by a ras-independent pathway. The p21 66–77 and 122–138 peptides cause greater inhibition of oncogenic p21. On the other hand, the GAP 832–845 and 1003–1021 peptides inhibit insulin-induced maturation to a significantly greater extent. Since we have found that activated wild-type and oncogenic p21 activate downstream targets like raf differently, these GAP peptides may be useful probes for identifying elements unique to the wild-type ras-p21 pathway.  相似文献

In the period 1875–1920, a debate about the generality and applicability of evolutionary theory to all organisms was motivated by work on unicellular ciliates like Paramecium because of their peculiar nuclear dualism and life cycles. The French cytologist Emile Maupas and the German zoologist August Weismann argued in the 1880s about the evolutionary origins and functions of sex (which in the ciliates is not linked to reproduction), and death (which appeared to be the inevitable fate of lineages denied sexual conjugation), an argument rooted in the question of whether the ciliates and their processes where homologous to other cellular organisms. In the beginning of the twentieth century, this question of homology came to be less important as the ciliates were used by the British protozoologist Clifford Dobell and the American zoologist Herbert Spencer Jennings to study evolutionary processes in general rather than problems of development and cytology. For them, homology mattered less than analogy. This story illustrates two partially distinct problems in evolutionary biology: first, the question of whether all living things have common features and origins; and second, whether their history and current nature can be described by identical mechanisms. Where Maupas (contra Weismann) made the ciliates qualitatively the same as all other organisms in order to create a cohesive evolutionary theory for biology, Jennings and Dobell made them qualitatively different in order to achieve the same end. Abstract: Since the brachiopod Order Lingulida has been greatly revised in recent years, with particular emphasis on Cambrian and Ordovician genera, many well‐established Silurian linguloid species from the British Isles have been left without an appropriate genus in which to place them. This is rectified here by allocating the various species, mostly erected in the nineteenth or early twentieth centuries, to a variety of genera which have been mainly erected within the last forty years. Two new genera are erected, Mergliella, with type species Lingula bechei Davidson, 1866 , and Striatilingula, with type species Lingula? striata J. de C. Sowerby, 1839 .  相似文献

The complete nucleotide sequence of the cucumber (C. sativus L. var. Borszczagowski) chloroplast genome has been determined. The genome is composed of 155,293 bp containing a pair of inverted repeats of 25,191 bp, which are separated by two single-copy regions, a small 18,222-bp one and a large 86,688-bp one. The chloroplast genome of cucumber contains 130 known genes, including 89 protein-coding genes, 8 ribosomal RNA genes (4 rRNA species), and 37 tRNA genes (30 tRNA species), with 18 of them located in the inverted repeat region. Of these genes, 16 contain one intron, and two genes and one ycf contain 2 introns. Twenty-one small inversions that form stem-loop structures, ranging from 18 to 49 bp, have been identified. Eight of them show similarity to those of other species, while eight seem to be cucumber specific. Detailed comparisons of ycf2 and ycf15, and the overall structure to other chloroplast genomes were performed.  相似文献
