Bacillus cereus strain XZM002 isolated from high arsenic aquifer sediments of Datong Basin was applied to examine the effects of arsenate stress on antioxidant enzyme activities, lipid peroxidation levels and cell growth inhibition rate. After 2 d exposure, the cell growth inhibition rate enhanced with an increase of As(V) concentrations (0, 800, 1600 μg/l). Reactive oxygen species and glutathione contents, lipid peroxidation levels, and antioxidant enzymes (glutathione peroxidase, and other three) activities of the treated cells were significantly higher than those of the controls during 3 d exposure (p < 0.05). Besides, the levels of nine parameters reached maximum after 2 d exposure and increased significantly with increasing arsenate stress (p < 0.05). However, they returned to levels similar to those of the control on the fourth day of exposure. The results suggested that the antioxidant defense system in B. cereus strain XZM002 could protect the cells from oxidative damage induced by arsenate.  相似文献

An in vitro plant regeneration system was established from the spores of Pteris vittata and identification of its tolerance, and accumulation of gametophytes and callous, to arsenic (As) and copper (Cu) was investigated. The highest frequency (100%) of callus formation was achieved from gametophyte explants treated with 0.5 mg l?1 6-benzylaminopurine (6-BA) + 0.5 mg l?1 gibberellin acid (GA). Furthermore, sporophytes were differentiated from the callus tissue derived from gametophyte explants on MS medium supplemented with 0.5 mg l?1 6-BA, 0.5–1.0 mg l?1 GA and additional 300 mg l?1 lactalbumin hydrolysate (LH) for 4 weeks. The optimum combination of ½ MS + 1.0 mg l?1 GA + 0.5 mg l?1 6-BA + 300 mg l?1 LH promoted sporophyte formation on 75 ± 10% of the callus. Every callus derived from gametophyte explants could achieve 3–4 sporophytes. The in vitro growth of gametophyte and callus was accelerated in the medium containing Na3AsO4 lower than 0.5 mM, but this growth was inhibited with 2 mM Na3AsO4. And with the increase of Na3AsO4 in the culture medium from 0 to 2 mM, the As accumulation in gametophytes and callus increased and achieved a level of 763.3 and 315.4 mg kg?1, respectively. Gametophytes and calluses transplanted to culture medium, supplemented with different concentrations of CuSO4, are similar to those in Na3AsO4, and the Cu accumulation in gametophytes could achieve 7,940 mg kg?1 when gametophytes were subcultured in medium containing 3 mM CuSO4. These results suggested that the high efficiency propagation system could be a useful and rapid means to identify other heavy metal tolerance and accumulation. Further, the regeneration ability of callus made it possible for genetic transformation of this fern.  相似文献

High-arsenic groundwater in inland basins usually contains high concentrations of fluoride. In the present study, the effects of fluoride on arsenic uptake by Pteris vittata and on arsenic transformation in growth media were investigated under greenhouse conditions. After P. vittata was hydroponically exposed to 66.8 μM As (V) in the presence of 1.05 mM F? in the form of NaF, KF, or NaF+KF for 10 d, no visible toxicity symptoms were observed, and there were not significant differences in the dry biomass among the four treatments. The results showed that P. vittata tolerated F? concentrations as high as 1.05 mM but did not accumulate fluoride in their own tissues. Arsenic uptake was inhibited in the presence of 1.05 mM F?. However, in hydroponic batches with 60 μM As (III) or 65 μM As (V), it was found that 210.6 and 316.0 μM F? promoted arsenic uptake. As(III) was oxidized to As(V) in the growth media in the presence and absence of plants, and F? had no effect on the rate of As(III) transformation. These experiments demonstrated that P. vittata was a good candidate to remediate arsenic-contaminated groundwater in the presence of fluoride. Our results can be used to develop strategies to remediate As-F-contaminated water using P. vittata.  相似文献

This study aimed to evaluate the pH, phosphate, and nitrate in the process of arsenic absorption by Eichhornia crassipes (water hyacinth), using the surface response methodology, in order to optimize the process. The plants were exposed to a concentration of arsenic of 0.5 mg L?1 (NaAsO2) over a period of 10 days. The results indicated optimal levels for the absorption of arsenic by E. crassipes at pH equal to 7.5, absence of phosphate, and minimum nitrate level of 0.0887 mmol L?1. For the tested concentration, E. crassipes was able to accumulate 498.4 mg kg?1 of As (dry base) in its plant tissue and to reduce 83% of the initial concentration present in the aqueous medium where it was cultivated. The concentration of phosphorus in solution linearly increased the phosphorus content in the plants and negatively influenced the absorption of arsenic. The concentration of 0.5 mg L?1 of As did not significantly affect the relative growth rate (RGR) and the tolerance index (TI). 94% of As (III) initially solubilized in water was converted by the end of the experiment period into As (V). The water hyacinth was important in the phytoremediation of arsenic when cultivated under optimal conditions for its removal.  相似文献

The influence of cytokinins and culture conditions including medium volume, harvest time and elicitation with abiotic elicitors (SA/MeJ) have been studied for the optimal production of biomass and withanolides in the multiple shoot culture of Withania somnifera. Elicitation of shoot inoculum mass (2 g l?l FW) with SA at 100 μM in the presence of 0.6 mg l?l BA and 20 mg l?l spermidine for 4 h exposure time at the 4th week in 20 ml liquid medium recorded higher withanolides production (withanolides A [8.48 mg g?l DW], withanolides B [15.47 mg g?l DW], withaferin A [29.55 mg g?l DW] and withanone [23.44 mg g?l DW]), which were 1.14 to 1.18-fold higher than elicitation with MeJ at 100 μM after 5 weeks of culture. SA-elicited cultures did not exhibit much variation in biomass accumulation when compared to control. This cytokinin induces and SA-elicited multiple shoot culture protocol provides a potential alternative for the optimal production of biomass and withanolides utilizing liquid culture.  相似文献

Arsenic speciation and cycling in the natural environment are highly impacted via biological processes. Since arsenic is ubiquitous in the environment, microorganisms have developed resistance mechanisms and detoxification pathways to overcome the arsenic toxicity. This study has evaluated the toxicity, transformation and accumulation of arsenic in a soil microalga Scenedesmus sp. The alga showed high tolerance to arsenite. The 72-h 50 % growth inhibitory concentrations (IC50 values) of the alga exposed to arsenite and arsenate in low-phosphate growth medium were 196.5 and 20.6 mg? L?1, respectively. When treated with up to 7.5 mg? L?1 arsenite, Scenedesmus sp. oxidised all arsenite to arsenate in solution. However, only 50 % of the total arsenic remained in the solution while the rest was accumulated in the cells. Thus, this alga has accumulated arsenic as much as 606 and 761 μg? g?1 dry weight when exposed to 750 μg? L?1 arsenite and arsenate, respectively, for 8 days. To our knowledge, this is the first report of biotransformation of arsenic by a soil alga. The ability of this alga to oxidise arsenite and accumulate arsenic could be used in bioremediation of arsenic from contaminated water and soil.  相似文献

This study analyzed the accumulation of arsenic and its influence on the content of P, Mn, Fe, Cu, Zn, S, Ca, Cl, and K in fronds of Azolla filiculoides via X-ray fluorescence (XRF). Azolla was exposed to increased concentrations of sodium arsenate (0, 5, 10, 20, and 30 μg ml ? 1) in Yoshida's nutrient solution. After 96 h, fronds were collected and taken to constant weight to calculate the relative growth rate (RGR), the biomass duplication rate, the bioaccumulation factor (BAF), and the elemental concentration via XRF analysis. By increasing As concentrations the RGR and BAF were reduced. The greatest accumulation of As in fronds was achieved at 20 μg As ml ? 1, which did not show significant differences with the highest concentration. Accumulation of As in fronds diminished concentrations of Mn, Fe, Zn, S, Ca, and K, while the concentration of P remained low but unchanging. The P content was significantly lower than the As content, thus the As:P ratio in fronds enhanced as the As concentration increased. Concentrations of Cl and Cu were unaffected due to As. This is one of the first reports about the influence of As-accumulation on the elemental content in Azolla fronds.  相似文献

Different nitrogen sources, abscisic acid (ABA), gellan gum at various concentrations, and osmotica were evaluated for their effects on maturation of somatic embryo (SE) in Japanese larch (Larix leptolepis). Different concentrations of l-glutamine or casein hydrolysate (CH) in the medium were also compared. The highest number of matured embryos was obtained with ½ Litvay (LM) medium supplemented with 1.71 mM l-glutamine and 250 mg l?1 CH. In terms of osmoticum effect, the highest number of cotyledonary SEs was produced in medium containing 0.2 M maltose. As for the effects of ABA and gellan gum concentration, the highest number of cotyledonary SEs was achieved on a medium containing 60 μM ABA and 0.8% gellan gum. In addition, the best plantlet conversion frequency (35.5%) was obtained with SEs derived from the treatment with 60 μM ABA and 0.8% gellan gum.  相似文献

Worldwide threats of fuel shortages in the near future and climate change because of greenhouse gas emissions are posing severe challenges and therefore it is vital to search for sustainable ways of preventing the consequences. The dual use of microalgae for phycoremediation and biomass production for sustainable biofuel production is a viable choice. Phycoremediation of As(III) and As(V) ions using microalgae was investigated in a two-staged batch reactor. Accumulation and toxicity of inorganic arsenic forms (As(III) and As(V)) to green microalgae Botryococcus braunii depend on environmental factors. Dissolved oxygen and pH cycles did not significantly differ due to the absence or presence of arsenic (either As(III) or As(V)) ions in the culture. Monod model was utilized for representing the growth kinetics of microalgae in pure media containing various concentrations of nitrate ions. Maximum specific growth rate and saturation constant were found to be 0.14788 d?1 and 0.00105 g/L, respectively. With the increase in concentration of phosphate in growth medium, the growth of microalgae increased. Media with NaCl (1.0 g/L) and NaHCO3 (1 g/L) resulted in higher maximum biomass concentration. Effect of coexisting ions on phycoremediation of As(III) and As(V) ions using microalgae was studied.  相似文献

The transformation of sodium arsenite and sodium arsenate by the rhizospheric nitrogen-fixing bacterium Azospirillum brasilense Sp245 in association with wheat (Triticum aestivum L. ‘Saratovskaya 29’) was studied. The effect produced by the A. brasilense strain on the morphological parameters of wheat in an As-polluted environment was examined. The plants were cultivated in a hydroponic system, with glass beads serving as a support for root growth. The plant-growth medium (an artificial soil solution) was deficient in P and Fe. The total initial As concentrations used were 75, 750, and 7500 μg l−1. The As compounds used contained sodium arsenate and sodium arsenite at an As(V):As(III) ratio of 1:3.6 (in terms of As) in all experiments. Inoculation of A. brasilense Sp245 led to a decrease in the overall root length and to the formation of lateral roots; both effects are possibly related to the bacteria’s ability to synthesize auxins. Inoculation also changed the As(V): As(III) ratio of the plant-growth medium. In all experiments, the concentration of As(V) in the nutrient medium increased relative to the initial one and was approximately 1.5-fold higher than that in the medium of uninoculated plants. This value slightly decreased (1.6 > 1.5 > 1.4) with increasing concentration of As in the medium. Azospirillum-inoculated plants accumulated less As than did the surface-sterilized uninoculated plants. This study shows that A. brasilense Sp245 in association with wheat changes the speciation, bioavailability, and plant uptake of As.  相似文献

An efficient transformation protocol was developed for vanilla (Vanilla planifolia) using protocorm-like bodies (PLBs) derived from shoot tips as explants. Of the ten media tested, Murashige and Skoog (MS) medium containing 0.45 μM thidiazuron (TDZ) produced maximum PLBs per shoot tip. Genetic fidelity of PLB-derived plantlets was confirmed by random amplified polymorphic DNA (RAPD) using 23 random primers. PLBs were co-cultured with Agrobacterium tumefaciens strain EHA105 harbouring the binary vector pBI121 containing the β-glucuronidase (gusA) and neomycin phosphotransferase II (npt II) genes for 3 days in MS medium supplemented with acetosyringone and transferred to selective regeneration medium containing 4.43 μM benzyladenine (BA), 2.68 μM naphthalene acetic acid (NAA) supplemented with 50 mg l?1kanamycin and 250 mg l?1 cefotaxime. After 15 days of culture, the surviving explants were transferred to the same regeneration medium but with a higher concentration of kanamycin (75 mg l?1). Finally, explants surviving after 30 days were subjected to more stringent selection in the regeneration medium supplemented with 100 mg l?1 kanamycin. Strong β glucuronidase activity was detected in the transformed plantlets by histochemical assay. Integration of T-DNA into the nuclear genome of transgenic plants was confirmed by polymerase chain reaction and Southern hybridization, while expression of transgene was confirmed by northern hybridization. This protocol allows effective and high frequency transformation of vanilla.  相似文献

Nitrogen and light are critical determinants of biomass accumulation and secondary metabolite production under in vitro culture conditions. In this study, we analyzed the effects of varied concentrations of total nitrogen in Murashige and Skoog (MS) medium and light intensity on the production of biomass, anthocyanin pigments, and bioactive antioxidants in callus cultures of Abelmoschus esculentus cv. ‘Hongjiao’. Maximum callus biomass accumulation (3 g FW) was achieved when calluses were cultured on MS medium containing 60 mM nitrogen under 40 μmol m??2 s??1 light intensity. In contrast, maximum values of total anthocyanin accumulation (TA; 7.3 CV/g FW), total phenolic content (TP; 12.07 mg/100 g FW), total flavonoid content (TF; 2.47?±?0.15 mg/100 g FW), and total antioxidant activity (TAA; 56.10 μmol Trolox/g FW) were observed when calluses were cultured on MS medium containing 40 mM total nitrogen under 80 μmol m??2 s??1 light intensity. In addition, callus grown under same culture condition exhibited high flavonoid content along with increased phenolic content and antioxidant activity. High performance liquid chromatography (HPLC) was performed for qualitative and quantity analysis of callus cultures. Most of the pigments from the callus extracts were identical with pod anthocyanins, and appeared on the ODS-column HPLC with lower concentration than the main pigments of the pod tissues. These findings indicate that callus cultures of red-pod okra represent a potential source of bioactive compounds with antioxidant properties for industrial applications.  相似文献

In this study, the degradation of tetradecyltrimethylammonium bromide (TTAB) by freely suspended and alginate-entrapped cells from the bacteria Pseudomonas putida (P. putida) A ATCC 12633 was investigated in batch cultures. The optimal conditions to prepare beads for achieving a higher TTAB degradation rate were investigated by changing the concentration of sodium alginate, pH, temperature, agitation rate and initial concentration of TTAB. The results show that the optimal embedding conditions of calcium alginate beads are 4 % w/v of sodium alginate content and 2 × 108 cfu ml?1 of P. putida A ATCC 12633 cells that had been previously grown in rich medium. The optimal degradation process was carried out in pH 7.4 buffered medium at 30 °C on a rotary shaker at 100 rpm. After 48 h of incubation, the free cells degraded 26 mg l?1 of TTAB from an initial concentration of 50 mg l?1 TTAB. When the initial TTAB concentration was increased to 100 mg l?1, the free cells lost their degrading activity and were no longer viable. In contrast, when the cells were immobilized on alginate, they degraded 75 % of the TTAB after 24 h of incubation from an initial concentration of 330 mg l?1 of TTAB. The immobilized cells can be stored at 4 °C for 25 days without loss of viability and can be reused without losing degrading capacity for three cycles.  相似文献

The effect of sulphate nutrition on arsenic (As) concentration, photosynthetic and chlorophyll fluorescence parameters of rice was investigated in hydroponically grown rice seedlings (Oryza sativa L.), using three sulphate levels (1.8 μM, 0.7 mM, or 1.5 mM). The results showed that sulphate deficiency decreased As accumulation in root, but increased the translocation of As from root to shoot. Sulphate deficiency reduced maximum quantum yield (Fv/Fm), minimum fluorescence and electron transport rate (ETR) of a dark-adapted leaf. Compared with low sulphate treatments (1.8 μM), significant increases were observed in the parameters of rapid light curves, rETRmax and I k of photosystem I (PSI) and photosystem II (PSII) of rice grown in the high sulphate treatments (1.5 mM) regardless of As additions. Therefore, an adequately high sulphate supply may result in less As translocation from root to shoot, and protecting the reaction pathways of PSI and PSII of rice seedlings grown in higher As-contaminated medium.  相似文献

Drinking water contaminated with arsenic poses serious threat to the human health. The present study was aimed for quality assessment of the groundwater of Khairpur Mir's in respect with arsenic and other elemental contamination like Fe, Cu, Co, and Ni. The presence of the trace elements in groundwater from different sources in the study area was measured by using atomic absorption spectroscopy. For arsenic analysis hydride generation technique (MHS-15) was used with detection limit of 0.02 µg l?1. Elevated level of arsenic was observed in most of the samples as compared to recommended value of World Health Organization (WHO) guidelines (10 µg l?1). However, levels of Fe, Cu, Co, and Ni in hand pump (HP) water samples was found in the range of 4–1610 µg l?1, 0–556 µg l?1, 0–230 µg l?1, and 0–700 µg l?1, respectively. Whereas in tube well (TW) water samples the observed values are 5–1620 µg l?1, 0–50 µg l?1, 4–110 µg l?1, and 0–360 µg l?1 for Fe, Cu, Co, and Ni, respectively. Significant difference was observed between TW and HP water samples. It was concluded that the level of arsenic found was very high up to 13 fold more than the WHO recommended limit in study area. While the levels of other elements was noted within the safe limit.  相似文献

In an attempt to scale-up of adventitious root cultures of Echinacea angustifolia for the production of biomass and caffeic acid derivatives, i.e. echinacoside, chlorogenic acid, cichoric acid, caftaric acid, and cynarin, the effects of Murashige and Skoog (MS) medium dilutions, and initial sucrose concentrations were investigated in a 5-L airlift bioreactor. In addition, the kinetics of adventitious root growth and accumulation of secondary metabolites were also studied. The greatest root dry weight (6.50 g L?l) and accumulation of total phenolics [22.06 mg g?1 DW (dry weight)], total flavonoids (5.77 mg g?1 DW) and total caffeic acid derivatives (10.63 mg g?1 DW) were obtained at quarter-strength MS medium. Of the various gradients of sucrose tested, 5 % sucrose supplementation was regarded as an optimal concentration for enhancing productivity of biomass and bioactive compounds. Neither higher salt strength (3/4–2 MS) nor sucrose concentrations (7 and 9 %) showed promotive effect on root growth and metabolite production. The kinetic studies revealed that 4 weeks of culture period is the optimal time to achieve highest productivity of metabolites. Based on these results, a large-scale (20 L) and a pilot-scale (500 L) adventitious root culture system was established. In the pilot-scale bioreactor, adventitious roots were elicitor-treated with 100 μM methyl jasmonate (MJ) on day 28. After 1 week of elicitation, 1.75 kg dry root biomass was harvested containing 60.41 mg g?1 DW of total phenolics, 16.45 mg g?1 DW of total flavonoids, and 33.44 mg g?1 DW of total caffeic acid derivatives. Among the caffeic acid derivatives, the accumulation of echinacoside (the major bioactive compound) in MJ-treated adventitious roots grown in the 500-L bioreactor was the highest (12.3 mg g?1 DW), which is approximately threefold more than the non-MJ-treated roots cultured in 5- and 20-L bioreactors.  相似文献

An effective protocol was developed for in vitro propagation of Psoralea corylifolia via somatic embryogenesis in cell suspension culture. Embryogenic callus was obtained on Murashige and Skoog (MS) medium supplemented with 6 μM naphthaleneacetic acid (NAA) and 30 μM glutamine from transverse TCLs from 10-day-old hypocotyl explants with a 96.4% frequency. Embryogenic callus produced a higher number of somatic embryos (123.7 ± 1.24 per gram fresh weight callus) on MS medium containing 30 g l?1 sucrose, 1 μM NAA, 4 μM benzyladenine (BA), 15 μM glutamine and 2 μM abscisic acid (ABA) after 4 weeks of culture. Somatic embryos successfully germinated (97.6%) on ½ MS medium containing 20 g l?1 sucrose, 8 g l?1 agar and supplemented with 2 μM BA, 1 μM ABA and 2 μM gibberellic acid (GA3) within 2 weeks of culture. Somatic embryos developed into normal plants, which hardened with 100% efficiency in soil in a growth chamber. Plants were successfully transferred to greenhouse and subsequently established in the field. Plant survival percentage in the field differed with seasonal variations. Average psoralen content of 12.9 μg g?1 DW was measured in different stages of somatic embryo development by high-performance liquid chromatography (HPLC). This protocol will be helpful for efficient propagation of elite clones on a mass scale, conservation efforts of this species and for secondary metabolites production studies.  相似文献

Arsenic (As) is a toxic metalloid that has gained special interest in the past years as a global environmental problem. Groundwater in Córdoba province (Argentina) presents high As concentrations which can be absorbed by plants or be used for artificial irrigation. The aim of this research was to elucidate the differential responses of symbiotic interactions established with three bacterial strains and soybean plants to realistic doses of arsenic. The reference strain Bradyrhizobium diazoefficiens USDA110 and the native isolate Bradyrhizobium sp. Per 3.64 were able to grow up to 13 mM As(V) whereas the native strain Bradyrhizobium sp. Per 3.61 grew up to 9.5 mM As(V). Metalloid addition did not modify the soybean plant growth at 6 μM As(V). Nevertheless, it was enough to induce oxidative stress as observed by an increase on lipid peroxidation. The soybean-Bradyrhizobium sp. assay at 6 μM As(V) showed no changes in growth variables (shoot and root dry weight) in plants inoculated with the reference microsymbiont or Bradyrhizobium sp. Per 3.61. Regarding As uptake by plants, metalloid accumulation followed the same distribution pattern among strains. Remarkably, at 6 μM As(V), soybean inoculation with Bradyrhizobium sp. Per 3.61 revealed a significantly lower translocation factor (TF) in comparison to other inoculated strains promoting As phytostabilization. At the highest As(V) concentration tested, only Bradyrhizobium diazoefficiens USDA110 was able to nodulate the legume, however, a significant decrease in the number and dry weight of nodules and nitrogen content was observed. In conclusion, the inoculation of soybean plants with the reference strain Bradyrhizobium diazoefficiens USDA110 exposed to high As(V) concentration represents an effective and promising symbiotic interaction that allows the development of the legume due to the minimal effects on plant growth. However, in low As(V) concentration environments, the native isolate Bradyrhizobium sp. Per 3.61, is shown to be the best inoculant among the tested strains, owing to the limitation of metalloid translocation and accumulation to edible parts of the legume, avoiding fruit contamination and human poisoning.  相似文献

The identification of plants with high arsenic hyperaccumulating efficiency from water is required to ensure the successful application of phytoremediation technology. Five dominant submerged plant species (Vallisneria natans (Lour.) Hara., Potamageton crispus L., Myriophyllum spicatum L., Ceratophyllum demersum L. and Hydrilla verticillata (L.f.) Royle) in China were used to determine their potential to remove As from contaminated water. V. natans had the highest accumulation of As among them. The characteristics of As accumulation, transformation and the effect of phosphate on As accumulation in V. natans were then further studied. The growth of V. natans was not inhibited even when the As concentration reached 2.0 mg L?1. After 21 d of As treatment, the bioconcentration factor (BCF) reached 1300. The As concentration in the environment and exposure time are major factors controlling the As concentration in V. natans. After being absorbed, As(V) is efficiently reduced to As(III) in plants. The synthesis of non-enzymic antioxidants may play an important role under As stress and increase As detoxication. In addition, As(V) uptake by V. natans was negatively correlated with phosphate (P) uptake when P was sufficiently supplied. As(V) is probably taken up via P transporters in V. natans.  相似文献

Gentiana dinarica Beck, native to the Balkan Dinaric Mountains, was established in vitro from axillary shoot buds. It was maintained in the form of shoot cultures on MS medium supplemented with 1.0 mg l?1 6-benzyladenine (BA) and 0.1 mg l?1 α-naphthaleneacetic acid and excised root cultures were maintained on ½ MS medium with 0.5 mg l?1 indole-3-butyric acid (IBA). Shoot cultures, adventitious roots and excised root cultures were analysed by HPLC techniques for the presence of secoiridoids and xanthones. Gentiopicrin and swertiamarin, the dominant components of shoot cultures, could not be detected in root cultures. Xanthones were present in both shoot and root cultures with norswertianin-1-O-primeveroside as the dominant metabolite. The secoiridoid and xanthone content, although characteristic for certain plant organs, was dependent on the concentration of plant growth regulators (BA and IBA) added to the medium. BA in the shoot multiplication stage strongly increased the secondary metabolite (SEM) content of shoot cultures. IBA had little effect on SEM accumulation in shoots during rooting, while it moderately stimulated SEM accumulation in excised root cultures.  相似文献
