Decidual cell induction in the pseudopregnant rat was examined in this study using the lectin concanavalin A (ConA). The histochemical binding of the lectin to the uterine cell surface at the time of deciduomatic induction was also studied. ConA was found to induce significant deciduomata (decidual-like tissue) in the uterine horn when injected intraluminally on day 5 of pseudopregnancy (PSP). ConA-induced deciduomata appeared as a series of discrete nodules in the uterine horn, reminiscent of the anatomical appearance of normal embryo implantation sites. Deciduoma induction by ConA was greatly reduced by pre-absorption of the lectin with its competitive sugar. Lectin histochemistry revealed binding of ConA to the cell surface on day 5 of PSP. Pre-absorption of the lectin with its competitive sugar also significantly reduced surface binding of the lectin, and this finding may be correlated with the greatly reduced ability of the pre-absorbed lectin to induce deciduomata. Possible mechanisms for the induction of deciduomata by lectins are considered.  相似文献

The modifications of enzymatics activities which involved dehydrogenases (LDH, MDH and G6PDH) alkalines phosphatases (Ph. Alc., ATP/as at Ph 7,2 and Ph 9,4) and hydrolases (Ph Ac. and beta Glu) were studied during the experimental deciduomata evolution in the Rat from the 5th to the 9th day. One the 5th day the uterus were used as control. Dehydrogenases and alkalines phosphatases activites appeared in isolated cells of sub-epithelial and middle antimesometrial stroma on the 5th day. Then, on the 6th day, they distributed themselves in strings of 8 to 10 cells. On the 7th day, they were regulary arranged in the decidual area. The growing decidual tissue was always surrounded by an hydrolysis activity.  相似文献

Summary An NADH-ferricyanide reductase activity resistant to inactivation by cytochemical procedures was examined during decidualization of rat endometrium. Resistant activity was restricted to plasma membranes, distal elements of the Golgi apparatus, and discoid cisternae and cytoplasmic vesicles of decidual cells of endometrium of the pseudopregnant rat on days 3, 4, 5, 7, and 9, after mating. The procedure reduced or eliminated any evidence of NADH-ferricyanide reductase activity from other cellular components such as endoplasmic reticulum, nuclei, and mitochondria. The observations of the glutaraldehyde-resistant reductase in both plasma membranes and discoid cisternae may indicate a role for the latter in the biosynthesis of plasma membranes during decidualization when massive cell proliferation and membrane biosynthesis occur. The origin of the discoid cisternae is tentatively ascribed to the mature faces of the Golgi apparatus.Work supported in part by a grant from the NIH CA1880101 to D.J.M.  相似文献

The influence of deciduoma-induced differentiation on the lipid peroxidation in the rat uterus was investigated. The wet weight of uterus and its protein content increased during deciduoma progression. Content of the thiobarbituric acid reactive substances (TBARS) as well as lipid peroxidation induced by ascorbate and cumene hydroperoxide showed significant decreases during deciduoma growth. Restoration of normalcy was observed during regression. The activity of superoxide dismutase, an inhibitor of lipid peroxidation showed an opposite pattern namely increase during deciduoma development and decline during the regressive phase. We conclude that cell differentiation during deciduoma induction is accompanied by a temporary and reversible decrease in the peroxidative potential of the rat uterus.  相似文献

Gamma glutamyl transpeptidase (GGT) is significantly enhanced in the rat uterus following implantation and during deciduoma morphogenesis. Uteri of gravid rats (Day 7 and 8 of pregnancy) show higher levels of this enzyme in the regions of implantation than in the interimplantation areas. Most of the GGT activity of induced deciduoma is present in the endometrium and the enzyme activity increases gradually to reach a peak five days post induction. Myometrial GGT activity is barely detectable during deciduoma growth. Results are discussed in relation to endometrial alterations during nidation and uterine glutathione content.  相似文献
