An impairment of muscle glutamine metabolism in response to dexamethasone (DEX) occurs with aging. To better characterize this alteration, we have investigated muscle glutamine release with regard to muscle glutamine production (net protein breakdown, de novo glutamine synthesis) in adult and old glucocorticoid-treated rats. Male Sprague-Dawley rats (3 or 24 mo old) were divided into seven groups: three groups received 1.5 mg/kg of DEX once a day by intraperitoneal injection for 3, 5, or 7 days; three groups were pair fed to the three treated groups, respectively; and one control group of healthy rats was fed ad libitum. Muscle glutamine synthetase activity increased earlier in old rats (day 3) than in adult rats (day 7), whereas an increase in muscle glutamine release occurred later in old rats (day 5) than in adult DEX-treated rats (day 3). Consequently, muscle glutamine concentration decreased later in old rats (day 5) than in adults (day 3). Finally, net muscle protein breakdown increased only in old DEX-treated rats (day 7). In conclusion, the impairment of muscle glutamine metabolism is due to a combination of an increase in glutamine production and a delayed increase in glutamine release.  相似文献

Intraperitoneal injection of deionized water (0.25ml/10 g body wt) produced a large increase in ornithine decarboxylase activity in cerebral cortex and heart of 6 day old rats, but had no effect on those activities in the 20 day old rat. Injection of the same dose of hypertonic (1.8%) saline caused a marked decline in the activity of this enzyme in both cerebral cortex and heart of the 6 day old rat and in the heart of 20 day old rats. In neonatal rats, the increase in heart ornithine decarboxylase elicited by the injection of water and the decline in activity which follows the injection of hypertonic saline were both evident within 30 minutes after injection; both effects were maximal two hours post-injection and both persisted for longer than four hours after injection. A decline in enzyme activity observed after injection of hypertonic saline was also found following the injection of hypertonic glucose, suggesting that osmotic effects, rather than specific ion effects, were mediating the loss of activity. The KM of ornithine decarboxylase in neonatal heart decreased following hypertonic saline injection, whereas that of cerebral cortex did not, supporting previous suggestions that the ornithine decarboxylase in heart may have unique regulatory controls.  相似文献

Catecholamines released by the adrenal medulla during birth play a key role in the adaptation of the newborn to extrauterine life. Respiratory, metabolic and cardiovascular adaptations to the hypoxia and other stresses associated with delivery are dependent upon a profound surge of adrenomedullary activity which occurs despite the immaturity of connections between the central nervous system and the adrenal. The "non-neurogenic" response seen in the fetus and neonate is thus essential to survival, and any interference either with catecholamine release or with catecholamine actions at adrenergic targets results in loss of the ability to survive hypoxia or other stressors. The immature secretory mechanism disappears as a result of development of neural connections, and factors which accelerate ontogeny of neural competence thus lead to premature loss of non-neurogenic secretory capabilities and a consequent increase in vulnerability. The fetus and neonate also have unusual proportions of adrenergic receptor subtypes in many tissues; these confer reactivity to specific stimuli associated with birth and with periods in which tissue differentiation may be under adrenergic control. Again, the ontogenetic switchover of receptor-mediated mechanisms appears to be a function of the development of neuronal competence, but in this case an important role may be played by a secondary surge in sympathetic tone occurring during the postnatal period. Through specialized mechanisms mediating catecholamine secretion and adrenergic responses, the adrenal medulla thus appears to provide both physiological and trophic signals to the fetus and neonate.  相似文献

Acute neurogenic or non-neurogenic inflammation was elicited in skin patches innervated by the saphenous nerve of anaesthetized Sprague Dawley rats. Lambda carrageenan was used to induce non-neurogenic inflammation, mustard oil (allyl-iso-thio-cyanate) or antidromic nerve stimulation to induce neurogenic inflammation. Antidromic nerve stimulation yielded plasma extravasation but no significant sensitization of unmyelinated nociceptor units. In contrast, mustard oil and carrageenan yielded plasma extravasation and sensitization of nociceptors, though carrageenan sensitized only part of them. Sensitization resulted in ongoing spike discharges and in a shift of response curves to lower temperatures when controlled radiant heat stimuli were applied to the receptive fields. Responses to mechanical stimuli with v. FREY hairs were not significantly altered. Effects of neurogenic and non-neurogenic inflammation on unmyelinated nociceptor units are compared.  相似文献

Summary ‘Pod washings’ were collected from groundnut at three hour intervals for 24 hours after feeding the plants with14CO2. Low radioactivity was found in the first four ‘Pod washings’ of 10 day old pods and the radioactivity increased with time. Highest radioactivity was found in washings of 20 and 30 day old pods and was twice the activity found in the washings of 50 day old pods. The radioactivity of both ethanol-soluble and insoluble fractions in pods was determined. The radioactivity mobilized into the pods increased with age and it was highest in 50 day old pods. The influence of the pod exudates on geocarposphere microflora during pod development is discussed.  相似文献

The effect of intrauterine iodine infusion on estrous cycle length was studied in four cows. The infusions were performed at various times of the estrous cycle: early, middle, late, and during luteolysis. Blood samples were drawn every third hour from the jugular vein. Progesterone and 15-keto-13,14-dihydroprostaglandin F2α (the main metabolite of PGF2α) were measured to monitor luteal activity and prostaglandin release. No release of prostaglandins was observed immediately following intrauterine infusion. Infusion in two cows on day 5 of the estrous cycle resulted in prostaglandin release after 54 and 69 hrs., respectively, followed by luteal regression and the occurrence of estrus at approx. five days after infusion. Infusions performed on days 11 or 12 resulted in prostaglandin release after 147 and 120 hrs., respectively, followed by luteolysis and heat after a 19 day estrous cycle. Infusion in two cows at days 16 and 17 resulted in prostaglandin release after 117 hrs. in both animals. One cycle was prolonged whereas the other cycle was normal in duration. One cow infused on day 20 following the occurrence of the first prostaglandin surge had a cycle length of 26 days, whereas another cow infused on day 20 was not affected because luteolysis was essentially complete by the time of infusion. One animal infused on day 5 did not respond to the iodine infusion. In this animal, however, the corpus luteum was not completely developed prior to the infusion. From this study it can be concluded: 1) intrauterine iodine infusions performed after the development of a progesterone secreting corpus luteum result in prostaglandin release within three to six days with the subsequent occurrence of luteolysis; 2) luteolysis wras in all cases observed in connection with prostaglandin F2α release of the same order of magnitude and duration as during normal luteolysis. kw|Keywords|k]prostaglandin release; k]progesterone; k]cow; k]es trous cycle; k]iodine infusion  相似文献

The concentration of 17-hydroxyprogesterone (17-OH-prog), but not 17-hydroxypregnenolone (17-OH-preg), was detectable in the sera of 5 day old female rats. The level of 17-OH-preg increased dramatically between day 5 and 10, remained high for 5 days and then decreased to low levels by day 25; a second increase was found on day 35. 17-OH-prog began to increase in the serum after day 10, reached a peak by day 20 and then decreased by day 25 and remained the same through day 35. Stimulation of the ovaries of intact females with 20 IU of pregnant mare's serum gonadotropin resulted in a prompt increase in both progestins, but a much larger increase in 17-OH-preg than in 17-OH-prog. Increases were similar, but quantitatively less, in hypophysectomized females. The results demonstrate that the ovaries of immature rats contain an active 17-hydroxylase system.  相似文献

The release of wild or captive-bred mammals within their historical ranges typically aims to reestablish populations in areas where they have become extinct or extirpated, to reinforce natural populations, or to resolve human–wildlife conflicts. Such programs, which also typically in parallel help foster the protection of the release site, concern a wide range of endangered mammalian species, including our closest living relatives: chimpanzees. In June 2008, the Chimpanzee Conservation Center (CCC), which is located in the High Niger National Park (HNNP) in Guinea, released a group of 12 chimpanzees (Pan troglodytes verus) comprised of 6 females and 6 males (8–20 yr old). The selected release site lies 32 km from the sanctuary in the Mafou, a core area of HNNP where wild chimpanzees are also known to occur. The purpose of this release was therefore to reinforce the natural chimpanzee population within the Mafou core area and to promote the protection of the HNNP. Nearly 2 yr postrelease, 9 chimpanzees still remain free-living. Two thirds of the release chimpanzees were equipped with VHF-GPS store-on-board tracking collars. We used data from retrieved collars to explore the release chimpanzees’ habitat use, individual day range, and core area use (50% and 80%) during the first year of the release. Males traveled significantly further than females. Although minimum day range did not differ between the sexes or vary seasonally, some release males were active for longer during the day than the females. Males also ranged over larger areas and used a wider network of core areas than the females. Habitat use was similar to that recorded in wild chimpanzees in the HNNP. As of September 2010, 2 males and 3 females form a group at the release site. Two of these females gave birth to healthy offspring respectively 16 and 20 mo postrelease. Another female successfully immigrated into a wild chimpanzee community. We suggest that the success of this chimpanzee release can be attributed to the CCC’s lengthy rehabilitation process and the savanna-mosaic habitat of the HNNP. This release demonstrates that under special socioecological circumstances, the release of wild-born adult chimpanzees of both sexes is a viable strategy, which can also function as an effective conservation tool.  相似文献

The effects of methadone (METH) on serum levels of prolactin (PRL), growth hormone (GH), corticosterone (CS) and TSH were determined in developing rats. METH increased PRL, GH and CS and decreased TSH at all ages tested, but the time course and magnitude of these effects changed during ontogeny. METH effects on day 10 were lower in magnitude than those observed in adults. In 20 day old pups, METH effects on GH and CS were comparable to those of adults, but TSH effects were still blunted. METH effects on hormone secretion in both 10 and 20 day old pups lasted longer than those observed in adults. Naloxone blocked all hormonal responses in adults, but did not completely block METH effects on CS secretion in 10 day old pups.  相似文献

Abstract In Gryllus bimaculatus, more digestive enzymes (amylase, trypsin, aminopeptidase) are secreted in the caecum of fed crickets than in unfed crickets, but the enzymes are released continuously at a basal rate in unfed animals. The rate of synthesis of the enzymes appears to parallel their rate of release. Digestive enzymes are released in response to a specific ratio of nutrients, although a high nutrient component in the food does not necessarily induce a high digestive enzyme release for that component. Rinsed flat‐sheet preparations of the caecum are incubated with specific nutrients (carbohydrates and proteins) and various concentrations of a neuropeptide (type‐A allatostatin), which affects generally the basal rates of secretion. Both maltose and glucose increase the release of amylase in vitro, but starch produces an inhibition of amylase release at lower concentrations. Bovine serum albumin (BSA), peptone and a mixture of amino acids have almost no effect on the release of aminopeptidase or carboxypeptidase, and only low concentrations of peptone increase trypsin release. High concentrations of both BSA and peptone strongly inhibit trypsin activity, perhaps by excess substrate binding to the trypsin active site. The allatostatin Grybi‐AST 5 elevates the release of amylase in vitro, but not of trypsin or aminopeptidase, in 2‐day‐old fed females. In the caeca from 1‐day‐old unfed crickets, both amylase and the trypsin release are stimulated in the presence of AST 5. The paracrine AST 5 is probably released from the gut endocrine cells and binds to the enzyme‐producing caecal cells.  相似文献

The net in vitro release of lactate, pyruvate, alanine, glutamate, glutamine and total amino acids from soleus muscles of suckling rats was studied. In all cases, the net output of metabolites was considerably higher in young animals than in 30 day old controls. Glutamine efflux of 1 day old pups was very low, probably due to lack of effectivity of the glutamine synthesizing system. The considerable output of amino acids in the younger animals was partly due to depletion of internal amino acid pools.  相似文献

This study was conducted in vitro to examine factors that may regulate prostaglandin release by bovine trophoblast and endometrial slices. Trophoblastic tissues and endometrial slices were recovered from superovulating and normally-ovulating cattle on day 16 or 20 of pregnancy and incubated for 24 h. Release of PGF2 alpha and 13,14-dihydro-15-keto-PGF2 alpha (PGFM), and incorporation of [14C]-leucine into proteins were quantified and expressed per microgram DNA, which gives a measure of cellular activity. Activity of trophoblastic tissue for synthesizing protein was decreased (P less than .05) and for releasing PGFM was increased (P less than .05) on day 20 compared to day 16 of pregnancy. Neither superovulation nor day of pregnancy altered trophoblastic activity for releasing PGF2 alpha. Superovulation increased (P less than .05) endometrial release of PGF2 alpha. Endometrial release of PGF2 alpha was less (P less than .05) on day 20 than on day 16 of pregnancy. When arachidonic acid (0, 100, 200 or 400 micrograms) was added at the start of incubation, trophoblastic release of PGF2 alpha changed (P less than .05) quadratically with dose of arachidonic acid. When arachidonic acid was added 8 h after the start of incubation, trophoblastic release of PGF2 alpha increased linearly (P less than .01) with dose of arachidonic acid. Adding arachidonic acid to incubation medium did not affect trophoblastic or endometrial protein synthesis. Endometrial slices suppressed (P less than .05) trophoblastic protein synthesis and release of PGF2 alpha. Apparently, endometrium can modulate trophoblastic release of prostaglandins and synthesis of proteins in vitro, and trophoblastic tissue from superovulated cattle 16 or 20 days pregnant can be used to study trophoblastic synthesis of prostaglandins and proteins.  相似文献

Egg-laying in hens exposed for more than 11 months to photostimulatory daylengths was intermittent and associated with a reduction in numbers of yellow-yolky ovarian follicles. Old laying hens (105 weeks) had lower concentrations of luteinizing hormone (LH) in the pituitary gland and plasma and reduced pituitary gland responsiveness to chicken LH-releasing-hormones (LHRH-I and II) in vivo when compared with young laying hens (28 weeks). Four weeks after transfer from 14 to 8 h light/day, egg production almost stopped in old, but not in young hens, although plasma LH concentrations decreased in all birds. After transfer from 14 to 20 h light/day, plasma LH increased in young, but not in old, hens, without a change in the rate of egg production. Reproductive function was enhanced in old hens returned to long days after induction of a moult and ovarian regression by reducing daylength and dietary restriction. Moulted hens had a greater rate of egg production, higher concentrations of plasma LH and a greater pituitary-gland responsiveness to LHRH-II in vivo than unmoulted control hens. After transfer from 14 to 8 h light/day, egg-laying decreased more rapidly in unmoulted than in moulted hens; transfer to 17 h light/day increased egg production in moulted, but not in unmoulted, birds. Induction of ovarian regression in old hens by dietary restriction alone also enhanced reproductive function after the dietary restriction was relaxed. Egg-laying was more persistent in hens brought into lay for a second year by transferring them from 3 to 11 h light/day than in hens transferred from 3 to 20 h light/day. Egg production was stimulated in hens maintained on 3 or 11 h light/day for 42 weeks, after transfer to 20 h light/day. Egg production ceased in hens maintained on 20 h light/day for 46 weeks, after transfer to 3 h light/day. These observations are consistent with the view that poor persistence of laying in hens less than 2 years old and exposed continuously to long days is caused, in part, by a reduction in hypothalamic-gonadotroph function. This reduction in neuroendocrine function may be due, in part, to the development of relative photorefractoriness.  相似文献

The aromatization of testosterone into 17 beta-estradiol (E2) was assessed in purified Leydig and Sertoli cells from rats aged 10-80 days. E2 was identified by gas chromatography-mass spectrometry (GC-MS) and measured both by radioimmunoassay (RIA) and GC-MS associated with stable isotope dilution. A potent competitive inhibitor of the aromatase activity, 4-hydroxyandrostenedione (4-OH-A) was used to test the enzymatic specificity. The basal aromatase activity was present in both cell types whatever the age of the animals. The basal E2 levels did not vary in Sertoli cells while a gradual increase was noted in Leydig cells until day 40, followed by a slight decrease in mature rats. In 10-day old animals, the aromatase activity was localized in Sertoli cells and highly stimulated by FSH; on day 20, both Sertoli and Leydig cells synthesized E2 although E2 from Sertoli cell origin was still predominant. Starting on day 20 until adulthood, the aromatase activity was under LH control in Leydig cells with a maximum around 40 days. The FSH and LH effects were mediated by cyclic AMP.  相似文献

The role of luteal oxytocin in the generation of luteolytic episodes of prostaglandin F2alpha at luteolysis was investigated. On day 10 of the cycle Dorset ewes underwent either surgical removal of the corpora lutea (lutectomy; n = 4) or sham operation (sham; n = 4). Lutectomised ewes were then administered progesterone by twice daily i.m. injection in corn oil (20 mg/day) until day 14 when treatment was ceased to simulate luteolysis. The concentration of 13, 14 dihydro-15-keto prostaglandin F2alpha (PGFM) was measured in peripheral blood samples collected at 20-min intervals for 8 h on days 12-16 of the cycle. Progesterone and oestradiol concentrations were similar in the two groups over the whole experimental cycle while oxytocin fell dramatically following lutectomy. No prostaglandin F2alpha release episodes were seen on day 12 or 13, while from days 14-16 both groups exhibited a similar episode frequency (lutectomy 0.9/ewe/8 h; sham 0.8/ewe/8 h). Analysis of episode characteristics revealed lower episode amplitude (p<0.05) but longer episode duration (p<0.05) in the lutectomy group. The results demonstrate that a normal frequency of prostaglandin F2alpha release episodes occurs independently of luteal oxytocin secretion. However, luteal oxytocin is involved in regulating the pattern of release, perhaps causing the release of episodes of the magnitude required for the successful completion of luteolysis.  相似文献

One or two 6–7 or 9–10 day old blastocysts were transferred to 39 pure-bred or cross-bred Charolais heifer recipients.The embryos were collected after slaughter of the donors. An insemination catheter (Cassou gun) was used to transfer the blastocysts through the cervix into the uterus. Placement was approximately 5cm beyond the body of the uterus.Pregnancy was determined by slaughter of the recipients on the 45th day of gestation. Nineteen of the 39 recipients were pregnant. The survival rate of embryos transferred was as follows: 6–7 day old embryos: 7 out of 23 (30.4%); 9–10 day old embryos: 18 out of 36 (50%). All embryos: 25 out of 59 (42.3%). The difference between the 6–7 day old embryos and the 9–10 day old embryos was not significant. Twelve of the 20 recipients receiving a bilateral transfer were pregnant, and 6 were carrying twins. This is a satisfactory technique for use in field conditions for the transfer of bovine blastocysts.  相似文献

Fetal and neonatal hypothyroidism was induced by treating pregnant rats with propylthiouracil (PTU) in the drinking water from day 12 of gestation to day 7 postnatally. The serum levels of thyroxine and triiodothyronine were lowered in both mother and offspring and the neonates showed a 20% reduction in weight gain. An i.p. glucose tolerance test on the 7-day old neonates revealed no major disturbances in glucose tolerance or insulin response. The data suggest that thyroid hormones are not essential for the neonatal development of the B-cell secretory response to glucose.  相似文献

Abstract The potato psyllid, Bactericera (= Paratrioza) cockerelli (?ulc) (Hemiptera: Triozidae), is a major pest of potato. Studies were conducted to determine the age at which both males and females reach reproductive maturity and the effect of age and time of day on sex attraction. Adult B. cockerelli reach reproductive maturity within 48 h post‐eclosion, with females being mature on the day of eclosion and males at 1 day post‐eclosion. Oviposition generally began 2 days after mating but was delayed when females mated within 2 days post‐eclosion. In laboratory olfactometer assays, the age of females used as odor sources and the age of males assayed to these odors did not affect sex attraction, with both younger (1–4‐day‐old) and older (8–10‐day‐old) males being attracted to females, regardless of female age (1–4‐day‐old or 8–10‐day‐old). Males assayed to live females at different times during the photophase (between 8:00 and 20:00 hours) were attracted to females between 11:00 and 17:00 hours, showing a temporal periodicity in sex attraction with B. cockerelli at least during the photophase.  相似文献
