The present paper deals with the karyotype analysis of 10 species of Cupressus including C. arizonica, C. bakeri ssp. mathewsii, C. chengiana, C. duclouxiana, C. funebris, C. goveniana, C. lusitanica, C. macrocarpa, C. pygmaea and C. sargentii. Except for C. arizonica, C. chengiana and C. macrocarpa they are all reported for the first time.The karyotype formula of the 10 species is K(2n)=22=22m which belongs to Stebbins'1A type. After making comparison among the karyotypes of 13 representive species in Cupressus, combined with data from geography, fossil, palaeogeography and palaeoclimate, the author thinks that C. sempervirens in Mediterranean region might be the most promitive one, the species in East Asia-China intermediate, those in North America advanced and C.lusitanica in Mexico and Guatemala the most advanced. The genus may have originated in the Mediterranean during the Jurassic or even earlier and then migrated from East Asia to North America via the Beringia in the Miocene or Pliocene. The regions, covering California, New Mexico, Mexico, Guatemala etc. are the present distribution center and the differentiation center of Cupressus.

Quantitative genetic variation was assessed in a population of Crepis tectorum subsp. pumila , a winter annual confined to calcareous grassland on the Baltic island of öland (SE Sweden). Plants from 40 maternal sibships were grown in a greenhouse and scored for a large number of traits representing all stages of the life cycle. The study included several characters that have been subject to ecotypic differentiation as well as traits known to be under current selection. Highly significant family differences were found for all but one character, suggesting that past selection was too weak to eliminate the genetic variability of characters presumed to be adaptive and there is a potential for further adaptive change in most traits. Two additional traits treated as qualitative were also found to differ significantly among families. A parallel cultivation experiment showed that the extent of population divergence in a quantitative trait was positively correlated with the amount of inter- family variation, implying stability of the relative variability for substantial periods of time, a possible reflection of phenotypic constraints being expressed both within and between populations. Additional data indicated that genetic trade-offs among traits under simultaneous selection, year-to- year variation in selection in combination with a long-lived seed bank, and genotype × environment interactions, could prevent the erosion of genetic variability in some characters connected with fitness. 相似文献

Ehrendorferiana and its two included new species, E. austrocedri (the type species) and E. fitzroyae, are diagnosed. The new genus is a member of the predominantly temperate Neotropical subfamily Aphalaroidinae (Psyllidae). Based on the absence of metabasitarsal spurs it forms a likely monophyletic group with Baccharopelma, Panisopelma and Russelliana. The absence or paucity of long dorsal or marginal setae on the wing pads and caudal plate suggests a probable sister-taxon relationship with Russelliana. Apart from two species of Triozidae from New Zealand on Podocarpaceae, Ehrendorferiana is unique within Psylloidea in its host association with conifers, viz. Austrocedrus and Fitzroya (both Cupressaceae).

An electrophoretic study of genetic variation at 31 loci in each of nine closely related gobiid species was carried out in order to assess their systematic relationships. The species used were: Pomatoschistus canestrinii, P. lozanoi, P. marmoratus, P. microps, P. minutus, P. norvegkus, P. pictus, Knipowitschia caucasica, Neogobius melanostomus. Genetic distances ( D ) and identities ( I ) were calculated by Nei's method for all 36 pairwise comparisons and dendrograms were constructed.
The results show that the very similar and cross-fertile species pair, P. lozanoi and P. minutus , have the highest I value; P. norvegkus has high I values with both species. Pomatoschistus marmoratus and P. microps also display high genetic similarity with each other. The remaining Pomatoschistus species and Knipowitschia caucasica are broadly and moderately equidistant from each other and from the five species refferred to above, while Neogobius melanostomus is considerably more distant from all other species.
These findings essentially agree with the pattern of relationships established using classical taxonomic approaches. However, the observation that Knipowitschia caucasica is as similar to Pomatoschistus spp. as the latter are to each other suggests that the generic distinction of Knipowitschia could usefully be reconsidered. 相似文献

Use of isozymes (including allozymes) in studies of population genetics and systematics of seaweeds has increased sufficiently in the last decade to allow some generalization. Only a single locus has been observed for about half the enzymes analysed in seaweeds, compared with 29% in vascular plants. Compared with higher plants, macroalgal species generally have low amounts of electrophoretically detectable genetic variation; the lowest levels of genetic variation found in natural populations are those reported for seaweeds. Nonetheless, seaweeds show an association between levels of genetic diversity as revealed by isozymes and species-specific attributes, such as mating system and predominance of asexual versus sexual reproduction. In systematic studies, isozymes have revealed cryptic species and identified pairs of sibling taxa. The quaternary structure of enzymes appears to be conserved at the phylum level. With the current availability of improved techniques for enzyme electrophoresis and for data interpretation, we expect future studies utilizing isozyme electrophoresis to provide further insight into population and evolutionary processes in seaweeds. 相似文献

We used plastid sequences (trnL, trnL‐trnF, petN‐psbM and trnT‐trnL) to infer the phylogenetic relationships and inter‐island connections of the Canarian Juniperus cedrus, and AFLP fingerprints to assess its genetic diversity patterns. Maximum Likelihood, Maximum Parsimony and Bayesian methods suggest independent colonization events for the three Macaronesian junipers and support the monophyly of J. cedrus. Plastid sequences reveal a low genetic diversity (three haplotypes) and do not provide sufficient information to resolve its temporal and geographical origin. AFLPs indicate a greater isolation in J. cedrus than in other Macaronesian trees with similar distributions and dispersal syndromes. Gran Canaria harbours the least genetically diverse population, which justifies immediate conservation actions. This island and Tenerife also show independent genetic structure, meaning that genetic exchange from other islands should be avoided in eventual reinforcements. Populations from La Palma and La Gomera show the highest genetic diversity levels and number of polymorphic AFLPs, probably because a lower incidence of felling has allowed a less dramatic influence of genetic bottlenecks. We suggest that management efforts should prioritize populations from these islands to preserve the evolutionary potential of the species, but we also stress the importance of knowledge of the evolutionary history, genetic structure and ecological interactions in conservation strategies. © 2014 The Linnean Society of London, Botanical Journal of the Linnean Society, 2014, 175 , 376–394. 相似文献

The development of spatial genetic structure (SGS) in seed plants has been linked to several biological attributes of species, such as breeding system and life form. However, little is known about SGS in ferns, which together with lycopods are unique among land plants in having two free‐living life stages. We combined spatial aggregation statistics and spatial genetic autocorrelation analyses using five plastid microsatellites and one nuclear gene to investigate SGS in two populations of the outcrossing tree fern Alsophila firma (Cyatheaceae). We assessed how the observed patterns compare with those estimated for other ferns and seed plants. Populations of A. firma exhibited strong SGS, spatial clustering of individuals, substantial clonal diversity and no inbreeding. SGS in ferns appears to be higher than in most seed plants analysed to date. Contrary to our expectations, an outcrossing breeding system, wind dispersal and an arborescent life form did not translate into weak or no SGS. In ferns, SGS is probably being affected by the life cycle with two free‐living life stages. The reproductive biology of ferns appears to be more complex than previously thought. Population genetics of the hispid cotton rat (Sigmodon hispidus): patterns of genetic diversity at the major histocompatibility complex 总被引:2,自引:0,他引:2
The hispid cotton rat, Sigmodon hispidus, is a common rodent widely distributed across the southern USA and south into South America. To characterize major histocompatibility complex (MHC) diversity in this species and to elucidate large-scale patterns of genetic partitioning, we examined MHC genetic variability within and among 13 localities, including a disjunct population in Arizona and a population from Costa Rica that may represent an undescribed species. We also tested the hypothesis that populations within the USA are at equilibrium with regard to gene flow and genetic drift, resulting in isolation-by-distance. Using single-strand conformation polymorphism (SSCP) analysis we identified 25 alleles from 246 individuals. Gene diversity within populations ranged from 0.000 to 0.908. Analysis of molecular variance (AMOVA) revealed that 83.7% of observed variation was accounted for by within-population diversity and 16.3% was accounted for by among-population divergence. The disjunct population in Arizona was fixed for a single allele. The Costa Rican population was quite divergent based on allelic composition and was the only population with unique alleles. Within the main portion of the geographical distribution of S. hispidus in the USA there was considerable divergence among some populations; however, there was no significant pattern of isolation-by-distance overall (P = 0.090). Based on the significant divergence of the only sampled population to its east, the Mississippi River appears to represent a substantial barrier to gene flow. 相似文献

A method for estimating and comparing population genetic variation using random amplified polymorphic DNA (RAPD) profiling is presented. An analysis of molecular variance (AMOVA) is extended to accomodate phenotypic molecular data in diploid populations in Hardy-Weinberg equilibrium or with an assumed degree of selfing. We present a two step strategy: 1) Estimate RAPD site frequencies without preliminary assumptions on the unknown population structure, then perform significance testing for population substructuring. 2) If population structure is evident from the first step, use this data to calculate better estimates for RAPD site frequencies and sub-population variance components. A nonparametric test for the homogeneity of molecular variance (HOMOVA) is also presented. This test was designed to statistically test for differences in intrapopulational molecular variances (heteroscedasticity among populations). These theoretical developments are applied to a RAPD data set in Vaccinium macrocarpon (American cranberry) using small sample sizes, where a gradient of molecular diversity is found between central and marginal populations. The AMOVA and HOMOVA methods provide flexible population analysis tools when using data from RAPD or other DNA methods that provide many polymorphic markers with or without direct allelic data. 相似文献

To examine the validity of this classification and the degree of morphological differentiation among closely related endemic taxa with disjunct distributions, we undertook analyses of three quantitative and eight qualitative morphological traits of flowers and leaves, most of which have previously been used without statistical comparison in the classification of the different taxa. Cyclamen balearicum showed high levels of morphological differentiation from all other taxa in subgenus Psilanthum consistent with its specific status. In contrast, morphological differentiation between C. creticum and subspecies of C. repandum was similar to that among the three subspecies of C. repandum . When compared with a recent molecular phylogeny of this subgenus, our results suggest that C. creticum may best be described as a geographical subspecies of C. repandum . The two varieties of C. repandum ssp. peloponnesiacum showed few significant differences for individual traits and their overall morphology is very similar to each other. Californian vernal pools, a patchy, island-like habitat, are endangered as a result of habitat destruction. Conservation of the remaining vernal pool habitat is essential for the persistence of several endangered species. We present the first study examining DNA-level genetic diversity within and among populations of a vernal pool plant species. We investigated genetic variation across eight populations of the US federally endangered vernal pool endemic Lasthenia conjugens (Asteraceae) using intersimple sequence repeat (ISSR) markers. Genetic diversity within the species was high (Nei's gene diversity estimate was 0.37), with moderate differentiation among populations (Bayesian F ST analog of 0.124). Using an amova analysis, we found that the majority of the genetic variation (84%) was distributed within populations. There is a significant relationship between geographical distance and pairwise genetic differentiation as measured by the Bayesian estimate θB . The alternative hypotheses of historic geological processes within the Central Valley and contemporary gene flow are discussed as explanations of the data. Because of the vulnerability of the populations, we calculated a probability of loss for rare alleles (fragments) in the populations. Calculations show that sampling only one of the eight populations for ex-situ conservation or restoration will capture approximately 54% of the sampled rare fragments. We believe that one of the sampled populations has become extinct since it was sampled. When removing this population from the above-mentioned calculations, sampling one population will capture only 41.3% of the sampled rare fragments. We recommend sampling strategies for future conservation and restoration efforts of L. conjugens. 相似文献

Acorus gramineus Soland. (Araceae) is a rheophyte and is distributed in southeastern Asia. Its populations are restricted to riparian habitats. The discontinuous distribution might result in high genetic diversification among plants of different river systems. In the present study, leaf samples were collected from populations along six river systems in western Taiwan and the genetic variation was investigated by employing RAPD markers. A total of 255 samples from 17 sampling sites was studied. Eighty random 10-mer primers were screened and six of them, which showed better amplification results, were selected to analyse all of the samples. Data of 34 high-intensity and highly reproducible polymorphic fragments were used in statistical analyses. The results of AMOVA analyses indicated that, of the total variation, 46.84% was attributable to differences among river systems, 16.88% to differences among sampling sites within river systems, and 36.28% to differences among individuals within sampling sites. The results of cluster analysis and principal coordinate analysis revealed that sampling sites of each river system formed distinct clusters and the sampling sites of six river systems were clustered into three main groups according to latitudinal relationships. The results of the present study indicated that the population genetic structure of the plants of different river systems is highly diversified, which seems to imply that the gene flow among them is very limited. 相似文献

The fruit fly Bactrocera latifrons (Hendel) is an important pest of commercially significant plants such as chili, tomato and eggplant. The species is native to South and Southeast Asia, but has now invaded Japan, Hawaii and Africa. In this study, mitochondrial DNA sequences were used to infer genetic structure and demographic history of B. latifrons. The efficiency of DNA barcodes for identification of B. latifrons was also tested. Ninety‐three specimens infesting four host‐plant species were obtained from 11 sampling locations in Thailand. The mitochondrial haplotype network revealed no major divergent lineage, which was consistent with a phylogenetic analysis that found strong support for the monophyly of B. latifrons. Population pairwise FST revealed that most (65%) comparisons were not significantly different, suggesting a high rate of gene flow. Analysis of molecular variance (amova ) found no significant genetic differentiation among populations from different host‐plant species. Sharing of several haplotypes among flies from different host‐plants indicates that the flies were moved freely across the plant species. Demographic history analysis revealed that the population has undergone recent expansion dating back to the end of the last glaciation. Thus, the results indicate that both ongoing and historical factors have played important roles in determining the genetic structure and diversity of B. latifrons. DNA barcoding analysis revealed that B. latifrons specimens were clearly differentiated from other species with 100% correct identification. Therefore, cytochrome oxidase I (COI) barcoding sequences could be effectively used to identify this important pest species, which could encourage monitoring and control efforts for this species. 相似文献

Despite Juniperus spp. being an important component of Mediterranean arid and semi‐arid ecosystems, there is a lack of complex studies on their biogeographical patterns. Using 16 morphological cone and seed traits and three nuclear microsatellite markers, we investigated the morphological and genetic variability of seven Mediterranean and Macaronesian Juniperus taxa (J. oxycedrus ssp. oxycedrus, J. oxycedrus ssp. badia, J. brevifolia, J. cedrus, J. deltoides, J. macrocarpa and J. navicularis) to identify biogeographical trends and interspecific genetic relationships. The highest gene diversity was measured in J. oxycedrus ssp. oxycedrus (HE = 0.716) and the lowest in J. brevifolia (HE = 0.441). The west Mediterranean was characterized by a higher level of genetic diversity than the east Mediterranean. A lack of significant genetic differences between European and African populations of J. oxycedrus suggests that the Strait of Gibraltar was not a significant barrier to gene flow, but has promoted some morphological differentiation. 应用 RAPD标记分析了桫椤分布在海南尖峰岭及霸王岭 ,广东塘朗山、黑石顶及大西山孑遗种群的遗传变异。 2 8个引物共检测到 1 1 8个位点 ,其中多态位点 33个 ,多态位点比率 2 7.97%。Shannon多样性、Nei遗传分化和分子方差分析 ( AMOVA)结果一致显示尖峰岭种群遗传多样性最大 ,霸王岭种群次之、塘郎山种群第三 ,黑石顶种群第四 ,而大西山种群最小。种群的遗传变异主要发生在种群间 ;Shannon指数测出的种群间遗传多样性所占比率为 79.1 0 % (基于表型频率 )和 77.85 % (基于基因频率 ) ,Nei基因分化系数 ( Gst)达 80 .69% ,分子方差分析也表明种群分化极为显著 ( Φst=0 .82 86,P<0 .0 0 1 )。桫椤的种内遗传多样性水平很低 ,种内 Shannon多样性仅为 0 .0 641 (基于表型频率 )和 0 .1 1 2 4 (基于基因频率 ) ,总基因多样性只有 0 .0 770。另外 ,Jaccard相似性系数的 UPGMA分析结果显示 5个种群分为两个分支 :尖峰岭种群和霸王岭种群组成一分支 ;黑石顶、塘郎山和大西山种群组成另一分支 ,并且三者中前两者的遗传关系更为密切。聚类分析结果还得到主成分分析的支持。根据桫椤种群的遗传变异特点 ,初步探讨了保护策略。 相似文献

Eleven microsatellites were isolated from the vairone Leuciscus souffia (Risso 1826), an endangered fish that inhabits river systems in and around the Alps in Europe. The level of genetic diversity was assessed in 29 individuals of the subspecies L. s. souffia, and their variability was further estimated in seven individuals of a different subspecies, L. s. muticellus. Eight of these microsatellite loci were also applied to seven closely related cyprinid species. Availability of the reported microsatellite loci will facilitate the investigation of population genetic structure of these species with applications for the development of conservation strategies and phylogeographical approaches. 相似文献

Strong ocean current systems characterize the Southern Ocean. The genetic structure of marine phytoplankton species is believed to depend mainly on currents. Genetic estimates of the relatedness of populations of phytoplankton species therefore should provide a proxy showing to what extent different geographic regions are interconnected by the ocean current systems. In this study, spatial and temporal patterns of genetic diversity were studied in the circumpolar prymnesiophyte Phaeocystis antarctica Karsten using seven nuclear microsatellite loci. Analyses were conducted for 86 P. antarctica isolates sampled around the Antarctic continent between 1982 and 2007. The results revealed high genetic diversity without single genotypes recurring even among isolates within a bloom or originating from the same bucket of water. Populations of P. antarctica were significantly differentiated among the oceanic regions. However, some geographically distant populations were more closely related to each other than they were to other geographically close populations. Temporal haplotype turnover within regions was also suggested by the multilocus fingerprints. Our data suggest that even within blooms of P. antarctica genetic diversity and population sizes are large but exchange between different regions can be limited. Positive and significant inbreeding coefficients hint at further regional substructure of populations, suggesting that patches, once isolated from one another, may not reconnect. These data emphasize that even for planktonic species in a marine ecosystem that is influenced by strong currents, significant breaks in gene flow may occur. 相似文献
