Unsaturated fatty acids undergo peroxidation processes. The presence of elevated levels of metabolites of lipid peroxidation processes may cause alterations in metabolic pathways which may lead to disturbances and clinical symptoms of illnesses. One such illness may be retention of fetal membranes (RFM) in cows. The aim of the study was the determination of thiobarbituric acid (TBA) reactive substances, conjugated dienes and hydroperoxides in bovine retained and not retained placenta in order to describe the oxidative status of cows affected with RFM. Placental samples were collected immediately after spontaneous delivery or caesarian section before term and at term and divided into 6 groups, with 16 animals in each group, as follows: A) caesarian section before term without RFM: B) caesarian section before term with RFM: C) caesarian section at term without RFM: D) caesarian section at term with RFM: E) spontaneous delivery at term without RFM; F) spontaneous delivery with RFM. The levels of TBA reactive substances, conjugated dienes and hydroperoxides were measured spectrophotometrically. The levels of all parameters were statistically significantly higher in the maternal than in the fetal part of placenta and the lowest in preterm groups. Statistically significantly higher concentrations in retained placenta samples than in control animals were observed. In conclusion, the metabolites of lipid peroxidation processes are elevated in cases of retained placenta and their presence might influence directly and/or indirectly the improper release of fetal membranes in cows.  相似文献

Acyloxyacyl hydrolase (AOAH) is an enzyme of bovine polymorphonuclear neutrophil leukocytes (PMN) that is capable of detoxifying endotoxin (25). The activity of AOAH in PMN isolated from the blood was investigated in dairy cows that expelled the fetal membranes normally (Group NFM) and in cows with retained fetal membranes (Group RFM) to obtain better insight into the role of the AOAH enzyme of neutrophils in endotoxin-related diseases, which occur frequently in dairy cows during the early postpartum period, especially in RFM cows. Twenty early postpartum dairy cows were used in the study: 13 NFM cows and 7 RFM cows. In the RFM cows, the percentage of PMN in blood (29+/-4%) was significantly (P<0.05) lower than in NFM cows (43+/-4%). The average AOAH activity in RFM cows (mean +/- SEM = 89+/-13 pmol fatty acid/10(7) PMN/h) was lower than in NFM cows (107+/-6 pmol fatty acid/10(7) PMN/h), but the difference in neutrophil AOAH activity between the 2 groups was not significant. There was also a higher percentage of immature neutrophils in isolated leukocyte suspensions from RFM cows (22+/-8%) than from NFM cows (15+/-4%), so that impairment of AOAH activity in early postpartum cows could be explained, in part, by immaturity of the neutrophils. These results suggest that the decreased AOAH activity of PMN could play a role in the pathogenesis of endotoxin-related diseases in dairy cows during the early postpartum period.  相似文献

Sixteen primiparous Holstein cows with retained fetal membranes (RFM) were studied for postpartum prostaglandin release, uterine infection and resumption of estrous cyclicity after manual removal of RFM (eight cows) versus leaving the RFM untreated (eight cows). The RFM were results of induced parturition on Day 274 of gestation. Seventeen non-RFM primiparous cows were controls. The 15-keto-13, 14-dihydro-metabolite of prostaglandin F(2alpha) (PGFM) was measured in daily blood samples. Aerobic and anaerobic bacteria were cultured from weekly uterine swabs from Week 3 until results were negative. Resumption of estrous cyclicity was determined by milk progesterone three times weekly. Manual removal caused an immediate and large but short-lived increase in PGFM, probably due to the physical damage of uterine tissue. No sustained difference in postpartum PGFM release between cows with RFM manually removed and cows with RFM left untreated was detected. Non-RFM controls had lowest PGFM concentrations. Uterine infections were more frequent and more severe after manual removal of RFM. Untreated RFM-cows and controls were similarly affected. Most infections involved Actinomyces (formerly Corynebacterium ) pyogenes and/or Fusobacterium necrophorum . Actinomyces pyogenes was isolated in the third week postpartum in 5 8 cows with RFM manually removed versus 2 8 cows with RFM left intact and in 2 17 controls. Manual removal prolonged the interval from calving to first functional corpus luteum by 20 d. This study, using RFM resulting from induced parturition, shows that manual removal of RFM can delay the postpartum return to normal reproductive status without altering PGFM profiles.  相似文献

Retention of fetal membranes (RFM) is believed to be associated with conditions of oxidative stress. In this study, 8-hydroxy-2'-deoxyguanosine (8-OH-dG) was used for the determination of spontaneous oxidative DNA lesions in maternal and fetal parts of bovine retained and nonretained placentas. Placental specimens were collected directly after spontaneous delivery or during cesarean section from cows divided into 6 groups: (A) cesarean section before term without RFM, (B) with RFM, (C) cesarean section at term without RFM, (D) with RFM, (E) spontaneous delivery at term without RFM and (F) with RFM. Isolated DNA was hydrolyzed and analyzed by HPLC; native nucleosides were monitored at 254 nm and 8-OH-dG by electrochemical detection. No significant differences in 8-OH-dG levels between retained and nonretained placental tissues were found in all samples from preterm groups (mean concentrations between 13 and 42 micromol/mol deoxyguanosine (dG)). In the term cesarean section group with RFM a significant increase in 8-OH-dG concentration in DNA from maternal (8-fold) and fetal (18-fold) membranes were detected when compared to the respective nonretained tissues. Also, in the term spontaneous delivery groups maternal nonretained placental tissues showed increased levels of 8-OH-dG in comparison to the respective tissues of the retained placenta group. In placental tissues oxidative DNA lesions appear to be controlled by responsive mechanisms which, possibly following exhaustion, give rise to increased 8-OH-dG levels.  相似文献

The objective of this study was to determine the effect of a monensin controlled-release capsule on the incidence of retained fetal membranes (RFM), milk yield and conception rate at first service (CRFS). The study was conducted during May to August 2004 in a large dairy herd in Chihuahua, Mexico. Cows (n = 2,025) from 20 to 30 d before expected parturition were randomly assigned to either a treatment or a control group. The treated group (n = 1,010) received orally a capsule of monensin that released 335 mg/d for 95 d. Control cows (no capsule, n = 1,015) were randomly assigned and matched by parity. Outcome variables were the incidence of RFM, first three test day milk and CRFS. Monensin decreased the incidence of RFM by 3.9% only in multiparous cows. Therefore, multiparous cows treated with monensin were 0.78 times (95% C.I. = 0.71-0.86) as likely as control multiparous cows to develop RFM. Overall, milk yield was negatively affected by the RFM condition, but it was improved by monensin only in multiparous cows with RFM at test day 1. Conception rate at first service was depressed by 10.6% in multiparous cows with RFM. However, in RFM cows treated with monensin, CRFS improved to the level of treated cows without RFM.  相似文献

Incidence of assisted births, retained fetal membranes (RFM), and metritis were recorded in one hundred dairy cows from parturition through 14 days post-calving. Manual removal of RFM was not attempted. All RFM were excised inside the vulva and observations of natural RFM expulsion were recorded. Fifteen of 100 cows (15%) had assisted births, 27 (27%) had RFM, 8 (8%) had primary metritis not associated with other postpartum reproductive problems, and 26 (26%) had secondary metritis. Uterine swabs for culture were collected during the study from cows with postpartum reproductive problems. E . coli was the most common organism isolated (69.4%). Sensitivities of all isolates to penicillin, tetracycline, and triple sulfa were 44.0%, 59.5%, and 36.9% respectively. One of two antibiotic treatments were administered to cows with these postpartum reproductive problems. Treated animals received either 5 g. tetracycline powder IU on day 1 of treatment plus 10.5 million units procaine penicillin G IM on days 1, 2, and 3; or 4 Sulfaurea boluses IU on day 1. Despite the antibiotic treatments, 26 of 34 cows having either assisted birth and/or RFM developed metritis (76%). Neither treatment regimen was superior to the other. The poor results of antimicrobial therapy suggested the futility of routine administration of therapeutic agents for postpartum reproductive problems. Treatment failure was attributed to ineffective drugs or inadequate dosage regimens.  相似文献

The objective of this study was to determine the effect of a calcium-energy supplement at calving on the incidence of calving-related disorders (CRD), fertility, BCS and milk yield in cows fed anionic diets and to establish any associations among outcome variables. In Florida, from October to December 1997, 479 cows were assigned to three groups and treated at calving as follows: Group 1: 160 nontreated cows; Group 2: 158 cows, treated orally with 60g Ca as CaCl2; Group 3: 161 cows, treated orally with 110g Ca as calcium propionate (510g) plus propylene glycol (400g). No treatment effect was detected for any of the outcome variables. An association was found between dystocia and age and retained fetal membranes (RFM). Age and RFM were associated with metritis. RFM and displacement of the abomasum were associated with ketosis. Ketosis and age were related to displacement of the abomasum. Parity, BCS, ovarian cysts, RFM and metritis were associated with fertility.  相似文献

Peripheral blood plasma concentrations of progesterone and the main metabolite of prostaglandin F, (15-keto-13,14-dihydro-PGF) PGFM, were determined in 10 Holstein cows with retained foetal membranes (RFM) and 12 Holstein cows without RFM (NRFM) during the peripartal period. The rate of uterine involution in the postpartum cows was monitored.There was no difference in the rate of uterine involution between cows with or without RFM. Cyclical ovarian activity was resumed within a month after parturition in both group. Increases in the mean peripheral plasma PGFM concentrations were evident in the RFM cows 6 days before parturition, compared to 48 h before parturition in the NRFM cows. A gradual decline in PGFM to prepartum concentrations occurred in both groups by Day 12 after parturition, although in the RFM cows, PGFM concentrations remained high until the placenta was shed.In both groups, the mean peripheral plasma concentrations of progesterone showed a marked decline beginning 48 h before partusition. The mean plasma progesterone concentrations were less than 1 ng/ml during the immediate postpartum period.  相似文献

Twenty-one mature F1 Brahman-Hereford cows were treated with 25 mg of dexamethasone (DEX) on day 279 or 280 of gestation to induce birth prematurely. Eigth cows were untreated (UT). Blood was sampled on day 279 or 280 of gestation just prior to treatment of cows with DEX (0 hr), at least daily thereafter to calving and within 1 hr postpartum. Concentrations of progesterone (P4), estrone (E1) and estradiol-17β (Eβ) and -17α (Eα) in blood serum were measured by radioimmunoassay. Among 21 cows treated with DEX, 16 (76%) calved within 78 hr (52±3 hr). Eleven of the 16 cows retained fetal membranes more than 12 hr (RFM) and five cows did not retain fetal membranes (NRFM). Five cows (24%) treated with DEX calved 266±46 hr later (NOR) on day 290±1 of gestation compared to day 286±2 for cows in group UT. No cow in groups NOR or UT had RFM. Failure of group NOR to calve prematurely appeared due to elevated serum P4 (P<.05), low serum Eβ (P<.10) and other estrogens (P>.10) pretreatment, and to only a 32% decrease in serum P4 within 72 hr after treatment. Serum estrogens, especially Eβ, were next lowest pretreatment in group RFM. However, in group RFM, all serum estrogens increased (P<.10 to P<.01) within 48 hr after treatment, reached higher concentrations and peaked later in relation to calving than in other groups (NRFM, NOR and UT). Synchronization of placental maturation and parturition may require a longer period of elevated serum estrogens prior to calving than was observed in group RFM. Treatment of cows prepartum with DEX had no effect on gain of calves, milk yield or yields of fat, total protein and total solids in milk during the first 12 weeks of lactation.  相似文献

The objectives were to: (i) determine the effect of prepartum supplementation of Vitamin E (Vit E) and selenium (Se) on plasma cortisol, erythrocyte peroxidation and the incidence of retained fetal membranes (RFM); (ii) estimate myeloperoxidase (MPO), lysozyme, elastase, and acid phosphatase (ACP) enzyme activities in the cotyledons of cows with or without RFM; and (iii) determine the molecular weight (SDS-PAGE) of proteins present in the cotyledons of cows with or without RFM. Fifty dairy (Friesian x Sahiwal) cows were equally allocated to one of two treatments, given as an im injection 3 week before calving: 1100 IU of DL alpha-tocopherol acetate (Vit E) and 30 mg of sodium selenite (Se), or saline (control). Concentrations of plasma cortisol (20 cows) were determined on days 21, 7, 3, 2, 1, and 0 prepartum, and erythrocyte lipid peroxide (all cows) was determined on days 21 and 7 prepartum. Treatment with Vit E and Se did not affect (P = 0.23) the incidence of RFM (12% versus 0%, respectively) but decreased (P < 0.05) erythrocyte lipid peroxide concentrations on day 7 prepartum compared with day 21 prepartum. Plasma cortisol concentration increased (P < 0.05) from day 21 prepartum to the day of parturition in Vit E+Se and control cows. However, on day 0, plasma cortisol concentrations were lower (P<0.05) in cows given Vit E+Se than in control cows (with or without RFM). To investigate enzyme activity and peptides in cotyledons, cotyledons were collected (from cows that were not part of the principal experiment), homogenised with PBS, and the supernatant used for the estimation of cationic peptides. Cotyledons of cows with RFM (n = 8) had lower (P < 0.01) MPO and greater (P < 0.05) lysozyme and ACP enzyme activities than those from non-RFM cows (n = 6). A band at <10 kDa in the SDS-PAGE indicated the presence of cationic peptides. In conclusion, a single treatment of Vit E and Se at 3-week prepartum reduced concentrations of plasma cortisol and erythrocyte peroxide. Altered enzyme activities in the fetal membranes indicated the involvement of leukocytes and trauma at the fetomaternal junction and warrant further investigation.  相似文献

Peripheral plasma levels of 15-keto-13,14-dihydro-PGF2α, progesterone, Cortisol, LH and prolactin were studied in 6 primiparous postpartum dairy cows. The cows were followed by hormone measurements and clinical examinations from parturition until pregnancy was established. Blood was collected 3 times per day. The cervix, uterus and ovaries were examined by rectal palpation at 6–10 days intervals. The cows were observed for signs of oestrus twice daily and were additionally teased with a bull to provoke standing heat. Four cows had a normal parturition and dropped their fetal membranes shortly afterwards. (NR group). The remaining 2 retained their fetal membranes for more than 24 h following parturition (RFM group). One out of 6 cows showed standing oestrus at the first ovulation, 4 animals were in oestrus at the second ovulation and all cows showed signs of oestrus at the third ovulation. Although the length of the first luteal phase varied from 9 to 22 days a corpus luteum was in all cases palpated. The secretion of progesterone during the first luteal phase was terminated by a PGF2α release. A significant difference in 15-keto-13,14-dihydro-PGF2α levels between the 2 groups was found on days 0–4 (2.39 vs 6.87 nmol/1 at Ρ < 0.06). Postpartum prostaglandin F2α release as reflected by the level of 15-keto-13,14-dihydro-PGF2α lasted shorter in the NR group than in the RFM group (15–17 vs 21 days). Significant positive correlations between 15-keto-13,14-dihydro-PGF2α and Cortisol as well as between prolactin and Cortisol during the first 24 days postpartum were noted only in cows having normal parturition. The most pronounced daily prolactin variations occurred during the second luteal phase (NR group), when a significant difference between the times 8.00, 12.00 and 15.00 was recorded (14.7, 31.5 and 19.7 μg/l, respectively). Moreover, a partial negative correlation between log value of prolactin and arithmetical value of LH was found in these cows only during the first luteal phase after parturition.  相似文献

Ante- and postpartum antioxidative/oxidative profiles in blood plasma of cows with and without retained placental membranes (RFM) were investigated. Twenty-two healthy pregnant cows were included in the study. Seven animals out of 22 suffered from RFM. Blood samples were obtained at 4, 3, 2, 1 weeks and 5 days antepartum (a.p.), at parturition and 1, 3, and 5 weeks postpartum. The following antioxidative parameters were measured using spectrophotometric methods: total antioxidant activity (TAC), β-carotene, vitamin A, vitamin C, and ceruloplasmin. The oxidative profile was based on the content of intermediates and end products of lipid and protein peroxidative processes which were measured by spectrophotometric and spectrofluorimetric methods. The examined parameters revealed a dynamic profile within the experimental period. The highest antioxidant and oxidant activity was noted at 2 and 1 week a.p. with a drop towards parturition suggesting the presence of oxidative stress during this time period and an apparent appropriate metabolic response of the macroorganism. Except for TAC and vitamin A, the contents of oxidative and antioxidative blood constituents did not differ between cows with and without RFM. A TAC and vitamin A by time interaction indicated higher antepartal concentrations of TAC and vitamin A in cows without RFM than in cows with RFM suggesting a possible role of antioxidative/oxidative imbalances in the aetiology of RFM.  相似文献

Retention of foetal membranes (RFM) in cows is supposed to be associated with the imbalance between production and neutralisation of reactive oxygen species (ROS). The consequence of uncontrolled ROS increase is oxidative damage to tissues, cells, and macromolecules. 8-iso-prostaglandin F2alpha (8-iso-PGF2alpha) is considered as a marker of oxidative tissue damage. The aim of the study was to investigate whether the concentrations of 8-iso-PGF2alpha, in caruncles and cotyledons from the bovine placenta differ between retained and properly released foetal membranes. Placentomes were collected immediately after either spontaneous delivery at term via the vagina or caesarean section before as well as at term through the incision and divided into six groups consisting of eight cows each as follows: A-preterm caesarean section without RFM, B-preterm caesarean section with RFM, C-term caesarean section without RFM, D-term caesarean section with RFM, E-term spontaneous delivery without RFM, F-term spontaneous delivery with RFM. The concentrations of free and total 8-iso-PGF2alpha, were determined in caruncles as well as cotyledons by enzyme immunoassay and expressed in picogram per gram of wet weight of tissue. The concentrations of free and total 8-iso-PGF2alpha were lower (P < 0.05) in cotyledons than in caruncles in all groups examined, as well as they were higher (P < 0.05) in retained than in released placenta. The concentrations of both parameters were lower (P < 0.05) in term spontaneous delivery groups than in term caesarean section groups. The results indicate that oxidative tissue damage, which may be the result of ROS imbalance, appears during RFM. However, the dynamics of this damage requires further elucidation.  相似文献

ABSTRACT: BACKGROUND: Transient receptor potential channel type 6 (TRPV6) and Calbindin-D9k (CaBP-9 k) are involved in the active calcium (Ca2+) transport mechanism in many tissues including placenta and uterus, suggesting a role in the establishment and maintenance of pregnancy. Moreover, TRPV6 and CaBP-9 k seem to support the materno-fetal Ca2+ transport that is crucial for fetal Ca2+ homeostasis, bone growth and development. However, it is unknown if these proteins are also involved in the aetiology of pathologies associated with parturition in cows, such as retained fetal membranes (RFM). The aim of the current study was to create an expression profile of uterine and placentomal TRPV6 and CaBP-9 k mRNAs and proteins during pregnancy and postpartum in cows with and without fetal membrane release. METHODS: Uteri and placentomes of 27 cows in different stages of pregnancy and placentomes of cows with and without RFM were collected. Protein and mRNA expression of TRPV6 and CaBP-9 k was investigated by real-time PCR, immunohistochemistry and Western blot. RESULTS: In the uterine endometrium, highest TRPV6 and CaBP-9 k expression was found in the last trimester of pregnancy, with a particular increase of protein in the glandular epithelium. In the placentomes, a gradual increase in TRPV6 mRNA was detectable towards parturition, while protein expression did not change significantly. Placentomal CaBP-9 k expression did not change significantly throughout pregnancy but immunohistochemistry revealed an increase in staining intensity in the maternal crypt epithelium. Immunohistochemical, stronger placental CaBP-9 k signals were seen in animals with RFM compared to animals with an undisturbed fetal membrane release, while protein levels, measured by Western blot analyses did not change significantly. CONCLUSIONS: The results of the present study demonstrate a dynamic expression of TRPV6 and CaBP-9 k during pregnancy in the bovine uterine endometrium and placentomes, suggesting a functional role for these proteins in Ca2+ metabolism during pregnancy. The temporal and spatial expression patterns indicate that TRPV6 and CaBP-9 k may be involved in materno-fetal Ca2+ transport, mainly through an interplacentomal transport, and that both proteins may participate in physiological processes that are crucial for fetal and placental development. However, neither TRPV6 nor CaBP-9 k seem to be causative in the retention of fetal membranes.  相似文献

The metabolism of arachidonic acid (AA) by caruncular and allantochorionic tissues and its regulation was studied in normal cows (n=13) and those with retained fetal membranes (RFM; n=9). Tissues were taken via the vagina about 6 hours postpartum and incubated for 6 hours in minimum essential medium containing tritiated AA alone or in the presence of oxytocin, platelet activating factor (PAF), epidermal growth factor (EGF) or ionophore calcium (A23187). The metabolites of AA were separated by reverse phase-high pressure-liquid chromatography. Tissue concentrations of prostaglandin F (PGF) and prostaglandin E2 (PGE2) and plasma 13,14-dihydro-15-keto-PGF (PGFM) concentration were also measured by radioimmunoassay. For caruncular tissue, less thromboxane B2 (TXB2) and more 6-keto prostaglandin F (PGIM) was synthesized in tissue from the animals with RFM than in the controls. Oxytocin, PAF, EGF and A23187 increased only PGIM production in the control animals; A23187 also decreased TBX2 synthesis. For the allantochorion, more PGE2, leukotriene B4 (LTB4) and PGIM and less TXB2, PGF and hydroxyecosatetranoic acids (HETE) was synthesized in tissue from cows with RFM than from animals that delivered normally. All of the substances used in this study increased PGIM, PGF and LTB4 and decreased TXB2 production by the allantochorionic tissue in control animals. The metabolism of AA by the allantochorionic tissue seems quantitatively under hormonal control. The metabolism of AA at the level of both maternal and fetal components of the placenta in cows with RFM differed from that seen in animals that expelled the membranes normally.  相似文献

Profiles of certain hormones measured by radioimmunoassay in 41 Holstein cows and heifers with retained fetal membranes (RFM; >12 hr postpartum) were compared to 41 peers without RFM (NRFM). Peers were matched by parity, season of calving, gestation length, dystocia and parturient paresis within prepartum diet group. Linear covariates of natural photoperiod, mean daily temperature, calf birth weight, length of gestation, and age and body weight of cow were included in the leastsquares analyses of data. Plasma profiles of prolactin and estrone were nearly identical from day 8 prepartum to day 2.5 postpartum. Plasma estradiol-17α was approximately one-third higher each day in group RFM (P<.05 across days) but estradiol-17β (Eβ) tended to be lower until day 2 prepartum (not significant). Also, plasma progesterone (P) was higher in group RFM between days 8 to 3 prepartum (p<.05 across days).Relationships between plasma P and Eβ were indicative of subsequent RFM (>24 hr rather than >12 hr), but only on day 6 prepartum. Three of four cases of 12 to 24 hr RFM had P and Eß profiles similar to NRFM. Either a combination of low P (<3.0 ng/ml) and low Eß (<100 pg/ml) or only high P (>7.9 ng/ml) were associated with a ten-fold higher rate of RFM (>24 hr) than when P was intermediate (4 to 8 ng/ml) and Eß exceeded 99 pg/ml. These results are in agreement with prior data wherein RFM were induced at premature births either by ovariectomy during pregnancy or by glucocorticoids.  相似文献

Postpartum ovarian activity, uterine involution and plasma concentrations of calcium and 15-keto-13, 14 dihydro-prostaglandin F2alpha (PGFM) were assessed in dairy cows with retained fetal membranes (n=10) and milk fever (n=10) at parturition. In addition, calcium and PGFM were evaluated in dairy cows affected with uterine prolapse (n=10) and pyometra (n=14). Cows with retained fetal membrane averaged 24.2+/-3.7 d until their first postpartum ovulation, while controls averaged 29.0+/-3.7 d (P>0.10). In cows with retained fetal membranes, the difference in follicular activity between the contralateral and ipsilateral ovaries in relation to the previously gravid uterine horn was appreciably greater post partum when compared with that of the controls. Cows with milk fever had an average of 30.8+/-3.1 d until their first postpartum ovulation, while control cows had an average of 20.4+/-3.3 d (P<0.05). The mean diameter of the uterine horns in cows with milk fever was greater (P<0.05) compared with that of the controls between Days 15-32 post partum. Concentrations of plasma calcium were lower in cows with retained fetal membranes within 24 h after parturition and during the first week post partum than in the controls (6.27+/-0.18 vs 7.40+/-0.18 mg/100ml, P<0.05). Concentration of calcium was lower (P<0.05) in cows with milk fever on Day 1 prior to treatment (4.68+/-0.40 < 5.8+/-0.45 mg/100ml) than in control cows; however, the calcium (Ca) level was not different during the subsequent 7 d post partum after treatment. Cows with uterine prolapse had lower concentrations of Ca during the first 7 d post partum than the controls (6.10+/-0.15 vs 7.33+/-0.12mg/100ml; P<0.01). Cows with pyometra had higher (P<0.05) concentrations of plasma PGFM than the controls (208.+/-13.2 > 138.1+/-15.2).  相似文献

Retained fetal membranes (RFM) and clinical metritis (CM) are frequently diagnosed disease conditions in dairy cows and considered of major economic impact due to negative effect on reproduction and milk production. The objective of this study was to evaluate the efficacy of i.u. tetracycline for the treatment of RFM and CM in dairy cows. Affected cows were randomly assigned to two groups; treatment group animals received i.u. 5 g chlortetracycline twice weekly for 2 wks, and no treatment group.A total of 1416 cows and 804 heifers in 5 herds calved during the study period. CM was diagnosed in 18.6% (inter farm range; 15.2–23.5%) and 30% (19.4–42.3%) of cows and heifers, respectively. RFM was diagnosed in 13.1% (9.4–18.1%) and 9.2% (3.6–13.8%) of cows and heifers, respectively. Conception rates after first insemination were 38.3%, 42.5% and 18% in normal, treated and non-treated CM cows, respectively. Numbers of days open were 140.5, 136.2 and 165.5 in normal, treated and non-treated CM cows, respectively. Based on 305-d corrected milk yield, cows and heifers affected by RFM and CM produced 300–500 kg less milk compared with their normal herd mates. Cows treated for CM produced 654 kg more milk per 305-d corrected lactation compared to non-treated control cows. Treatment of RFM had no effect on reproductive performance or milk production. In conclusion, i.u. chlortetracycline treatment was proven to prevent the detrimental effect of CM on reproductive performance in heifers and cows and on milk production in cows only.  相似文献

Prostaglandin E(2) 9-keto reductase (9-KPR) activity shifts reversibly PGE(2) into PGF(2 alpha) and may be responsible for the control of prostaglandins (PGs) levels in, among others, placental tissues. The retention of fetal membranes in cows is the postpartum disorder where the disturbances in PGs metabolism have been reported. It has been argued whether these disturbances are due to alterations in 9-KPR activity. In this study, the activity of the enzyme was determined in maternal and fetal bovine placental tissues which were divided into 6 groups as follows: (A) caesarian section before term without retained fetal membranes (n=10), (B) caesarian section before term with retained fetal membranes (n=10), (C) caesarian section at term without retained fetal membranes (n=12), (D) caesarian section at term with retained fetal membranes (n=12), (E) spontaneous delivery at term without retained fetal membranes (n=12), (F) spontaneous delivery at term with retained fetal membranes (n=12). The enzyme activity was measured spectrophotometrically and expressed in nanokatals (nkat) per protein content. The activity increased towards parturition and was significantly higher in maternal than in fetal part of placenta in all groups examined. The significantly higher values in retained than in not retained placental tissues were observed in the samples examined. The present results indicate that the disturbances in 9-KPR activity in bovine retained placenta exist but their reasons still require further experiments.  相似文献
