Some epiphytic species accumulate airborne particles and are suitable biological indicators for monitoring urban and industrial pollution. The species Tillandsia recurvata L. was studied as a monitor of air pollution in an urban area from Mexico. Individuals were collected in 25 sites which are exposed to different pollution degree and sources.The magnetic particle concentration, particle size, and mineralogy were determined and compared with chemical contents for all samples. The highest values of magnetic concentration dependent parameters were observed in industrial and heavy traffic sites (e.g., mass specific magnetic susceptibility of up to 171.5 × 10−8 m3 kg−1). In contrast, sites with low or without vehicular traffic reached low values (e.g., mass specific magnetic susceptibility of down to 1.8 × 10−8 m3 kg−1). The integrated magnetic analysis (King's and Day's plots, remanent magnetization parameters and thermomagnetic measurements) revealed the presence of ferromagnetic minerals, mostly magnetite-like with fine grain sizes (0.1–1 μm) and subordinate presence of high-coercivity minerals. Selected samples were observed by SEM and EDS analysis and revealed the presence of Fe-rich particles, as well as trace elements, among others, As, Sb, S, Cr, Mo, V, Zn, Ba, Hg, Pt and Cu. Most of the elements detected by EDS were also quantified by ICP-MS measurements.Multivariate statistical analyses prove a high correlation between magnetic parameters and elements, as well as allow us classifying sites in clusters (fuzzy c-means clustering) with different pollution degree. These results demonstrate the usefulness of the species T. recurvata L. as a passive pollution monitor, with an affordable and immediate application. This species is abundant not only in Mexico, but also in other cities from America.  相似文献

α-type phospholipase A2 inhibitory protein (PLIα) isolated from the serum of the venomous snake Glyoidius brevicaudus, GbPLIα, is a homotrimer of subunits having a C-type lectin-like domain. The serum protein from nonvenomous snake Elaphe quadrivirgata, EqPLIα-LP, is homologous to GbPLIα, but it does not show any inhibitory activity against PLA2s. When a mixture of denaturant-treated monomeric forms of GbPLIα and EqPLIα-LP was used to reconstitute their trimers, no significant amounts of heterotrimers composed of GbPLIα and EqPLIα-LP subunits could be formed. On the other hand, when a mixture of denaturant-treated monomeric forms of GbPLIα and the recombinant chimeric EqPLIα-LP, Eq13Gb37Eq, in which the residues 13–36 were replaced by those of GbPLIα, was used to reconstitute their trimers, significant amounts of their heterotrimers were observed. Furthermore, when a mixture of denaturant-treated monomeric forms of EqPLIα-LP and the recombinant chimeric GbPLIα, Gb13Eq37Gb, in which the residues 13–36 were replaced by those of EqPLIα-LP, was used, significant amounts of their heterotrimers were observed. By comparison of the respective inhibitory activities of the heterotrimeric subspecies, it was suggested that the inhibitory activity of the trimer was governed by one subunit with the highest activity, and not affected by the number of these subunits. The intermolecular electrostatic interactions between Glu23 and Lys28 of GbPLIα were also suggested to be important in stabilizing the trimeric structure. The importance of the electrostatic interaction was supported by the less stability of the homotrimeric structure of a mutant GbPLIα with a single amino acid substitution, GbPLIα(K28E).  相似文献

The cDNA encoding of a phospholipase A2 inhibitor (PLIα) of the Chinese mamushi, Agkistrodon blomhoffii siniticus, was identified from a liver cDNA library by use of a probe prepared by polymerase chain reaction (PCR) on the basis of the amino acid sequence of PLIα. It encoded a polypeptide of 166 amino acid residues, including 19 residues of the signal sequence and 147 residues of the complete mature sequence of PLIα. The PLIα cDNA was subcloned into the expression vector pET-16b and used to transform Escherichia coli strain BL21(DE3)pLysS. The recombinant PLIα expressed as a fusion protein was solubilized and purified to homogeneity by use of a metal affinity resin. The purified PLIα fusion protein underwent folding to form a trimeric structure like the intact PLIα, and showed inhibitory activity against the group II acidic PLA2 from A. blomhoffii siniticus venom; although its binding constant (1/Ki) value was 30-fold lower than that of the natural PLIα. The elimination of the N-terminal additional peptide from the fusion protein resulted in a marked increase in the inhibition activity with a binding constant comparable to that of the natural PLIα against the acidic PLA2. Furthermore, the carbohydrate chains of the natural PLIα were found to play an important role in the inhibitory activity against the basic PLA2.  相似文献

Oil pollution is an environmental problem of increasing importance. Hydrocarbon-degrading microorganisms, adapted to grow and thrive in oil-containing environments, have an important role in the biological treatment of this pollution. One of the limiting factors in this process is the bioavailability of many fractions of the oil. The hydrocarbon-degrading microorganisms produce biosurfactants of diverse chemical nature and molecular size. These surface-active materials increase the surface area of hydrophobic water-insoluble substrates and increase their bioavailability, thereby enhancing the growth of bacteria and the rate of bioremediation.  相似文献

From autumn 2005 to spring 2008, a study focusing on the collection of Trichoptera larvae was conducted in the catchment of the Guadiamar River (SW Spain). The distribution of caddisfly species in relation with environmental factors was examined. Four groups of species were recognised according to the environmental gradient. A direct ordination analysis (CCA) used to describe assemblage changes among sites corroborated that conductivity, land-use, geomorphological gradient, and temporality were the variables that best explain the Trichoptera distribution, but to establish the relative influence of the land-use variables was difficult. On the other hand, mining pollution appeared to be the main factor explaining Trichoptera community structure. In April 1998 this river system underwent an accidental release of a large mass of toxic mine waste, which exterminated macroinvertebrates in the middle and lower parts and floodplain. Several years later, caddisfly communities in these areas were different from those of unaffected upper reaches. The species assemblage was especially poor in lower river reaches that underwent the permanent, diffuse urban and agricultural pollution. These conditions were tolerated only by Hydropsyche exocellata Duföur.  相似文献

The response of cells to changes in their physico-chemical micro-environment is essential to their survival. For example, bacterial magnetotaxis uses the Earth''s magnetic field together with chemical sensing to help microorganisms move towards favoured habitats. The studies of such complex responses are lacking a method that permits the simultaneous mapping of the chemical environment and the response of the organisms, and the ability to generate a controlled physiological magnetic field. We have thus developed a multi-modal microscopy platform that fulfils these requirements. Using simultaneous fluorescence and high-speed imaging in conjunction with diffusion and aerotactic models, we characterized the magneto- aerotaxis of Magnetospirillum gryphiswaldense. We assessed the influence of the magnetic field (orientation; strength) on the formation and the dynamic of a micro-aerotactic band (size, dynamic, position). As previously described by models of magnetotaxis, the application of a magnetic field pointing towards the anoxic zone of an oxygen gradient results in an enhanced aerotaxis even down to Earth''s magnetic field strength. We found that neither a ten-fold increase of the field strength nor a tilt of 45° resulted in a significant change of the aerotactic efficiency. However, when the field strength is zeroed or when the field angle is tilted to 90°, the magneto-aerotaxis efficiency is drastically reduced. The classical model of magneto-aerotaxis assumes a response proportional to the cosine of the angle difference between the directions of the oxygen gradient and that of the magnetic field. Our experimental evidence however shows that this behaviour is more complex than assumed in this model, thus opening up new avenues for research.  相似文献

This work analyses the distribution of living benthic foraminiferal assemblages of surface sediments in different intertidal areas of Ria de Aveiro (Portugal), a polihaline and anthropized coastal lagoon. The relationships among foraminiferal assemblages in association with environmental parameters (temperature, salinity, Eh and pH), grain size, the quantity and quality of organic matter (enrichment in carbohydrates, proteins and lipids), pollution caused by metals, and mineralogical data are studied in an attempt to identify indicators of adaptability to environmental stress. In particular, concentrations of selected metals in the surficial sediment are investigated to assess environmental pollution levels that are further synthetically parameterised by the Pollution Load Index (PLI). The PLI variations allowed the identification of five main polluted areas. Concentrations of metals were also analysed in three extracted phases to evaluate their possible mobility, bioavailability and toxicity in the surficial sediment. Polluted sediment in the form of both organic matter and metals can be found in the most confined zones. Whereas enrichment in organic matter and related biopolymers causes an increase in foraminifera density, pollution by metals leads to a decline in foraminiferal abundance and diversity in those zones. The first situation may be justified by the existence of opportunistic species (with high reproduction rate) that can live in low oxic conditions. The second is explained by the sensitivity of some species to pressure caused by metals. The quality of the organic matter found in these places and the option of a different food source should also explain the tolerance of several species to pollution caused by metals, despite their low reproductive rate in the most polluted areas. In this study, species that are sensitive and tolerant to organic matter and metal enrichment are identified, as is the differential sensitivity/tolerance of some species to metals enrichment.  相似文献

Current technologies for removal and recovery of both toxic and industrial interest metals usually produce wastes with high concentrations of those substances. They are an important source of environmental pollution, specially when they contain heavy metals. This is one of the most important environmental problems, and of the most difficult to solve. So far, there have been a number of studies considering the possibility of removing and recovering heavy metals from diluted solutions. These are due, principally, because of the commercial value of some metals as well as the environmental impact caused by them. The traditional methods for removing have several disadvantages when metals are present in concentrations lower than 100?mg/l. Biosorption, which uses biological materials as adsorbents, has been considered as an alternative method. In this work, several variables that affect the capacity for copper biosorption by T. ferrooxidans have been studied. Particularly, the effect of pH, chemical pretreatment, biomass concentration and temperature have been considered. Results indicate that a capacity as high as 119?mg of Cu/g of dry biomass can be obtained at a temperature of 25?°C.  相似文献

Bacterial chemotaxis has the potential to increase the rate of degradation of chemoattractants, but its influence on degradation of hydrophobic attractants initially dissolved in a non-aqueous-phase liquid (NAPL) has not been examined. We studied the effect of chemotaxis by Pseudomonas putida G7 on naphthalene mass transfer and degradation in a system in which the naphthalene was dissolved in a model NAPL. Chemotaxis by wild-type P. putida G7 increased the rates of naphthalene desorption and degradation relative to rates observed with nonchemotactic and nonmotile mutant strains. While biodegradation alone influenced the rate of substrate desorption by increasing the concentration gradient against which desorption occurred, chemotaxis created an even steeper gradient as the cells accumulated near the NAPL source. The extent to which chemotaxis affected naphthalene desorption and degradation depended on the initial bacterial and naphthalene concentrations, reflecting the influences of these variables on concentration gradients and on the relative rates of mass transfer and biodegradation. The results of this study suggest that chemotaxis can substantially increase the rates of mass transfer and degradation of NAPL-associated hydrophobic pollutants.  相似文献

Ozone exposure during growth affects the feeding value of rice shoots   总被引:2,自引:0,他引:2  
Rising tropospheric ozone concentrations have been observed in many Asian countries in recent years. Ozone pollution reduces the yield of agricultural crops but may also affect crop quality. This study aimed at estimating the effect of ozone exposure on feeding quality of rice shoots for ruminant herbivores. Rice plants from two genotypes differing in ozone tolerance were exposed to ozone at a concentration of 120 nl/l for 18 days, and feeding value was determined by chemical analyses and in vitro incubation in rumen fluid. Rice biomass was reduced by an average of 24% in the ozone treatment as compared to the control. Moreover, ozone exposure affected various feed quality parameters. Crude protein content was lower in ozone treated plants (P<0.05). Potential gas production during the in vitro incubation for 96 h also dropped (P<0.01) due to ozone treatment, indicating reduced digestibility of the plant materials. This was explained with an increase in the antinutritive components lignin (P<0.05) and phenolics (P<0.001) due to ozone exposure. An ozone tolerant genotype exhibited a more pronounced increase in phenolics, suggesting that this may constitute a stress defense mechanism. Our results suggest that ozone may affect the feeding value of cereal straws and calls for further research in this direction.  相似文献

Liver, spleen and heart tissues of DBA/2 Hfe knockout mice have been characterised by low temperature AC magnetic susceptibility measurements together with Transmission Electron Microscopy (TEM) and Selected Area Electron Diffraction in order to investigate the chemical iron speciation in a murine model of iron overload diseases. With emphasis on ferritin-like species, the temperature dependent in-phase and out-of-phase susceptibility profiles agree with the elemental analysis in that, in this model, iron accumulation takes place in the hepatic tissue while in the spleen and heart tissues no differences have been observed between knockout and wild type animals. The comparison of the magnetic properties between perfused and non-perfused liver tissues has made it possible to estimate the magnetic contribution of usually present blood remains. The TEM observations reveal that, besides the isolated ferritins and ferritin-containing lysosomes–siderosomes present in the hepatocytes, other iron deposits, of heterogeneous size, morphology and crystalline structure (haematite and/or goethite), are present in the cytoplasm, near the membrane, and in extracellular spaces.  相似文献

The primary goal of this study was to determine the conditions required for the effective inactivation of Bacillus anthracis spores on materials by using methyl bromide (MeBr) gas. Another objective was to obtain comparative decontamination efficacy data with three avirulent microorganisms to assess their potential for use as surrogates for B. anthracis Ames. Decontamination tests were conducted with spores of B. anthracis Ames and Geobacillus stearothermophilus, B. anthracis NNR1Δ1, and B. anthracis Sterne inoculated onto six different materials. Experimental variables included temperature, relative humidity (RH), MeBr concentration, and contact time. MeBr was found to be an effective decontaminant under a number of conditions. This study highlights the important role that RH has when fumigation is performed with MeBr. There were no tests in which a ≥6-log10 reduction (LR) of B. anthracis Ames was achieved on all materials when fumigation was done at 45% RH. At 75% RH, an increase in the temperature, the MeBr concentration, or contact time generally improved the efficacy of fumigation with MeBr. This study provides new information for the effective use of MeBr at temperatures and RH levels lower than those that have been recommended previously. The study also provides data to assist with the selection of an avirulent surrogate for B. anthracis Ames spores when additional tests with MeBr are conducted.  相似文献

Development of biologically relevant crowding solutions necessitates improved understanding of how the relative size and density of mobile obstacles affect probe diffusion. Both the crowding density and relative size of each co-solute in a mixture will contribute to the measured microviscosity as assessed by altered translational mobility. Using multiphoton fluorescent correlation spectroscopy, this study addresses how excluded volume of dextran polymers from 10 to 500 kDa affect microviscosity quantified by measurements of calmodulin labeled with green fluorescent protein as the diffusing probe. Autocorrelation functions were fit using both a multiple-component model with maximum entropy method (MEMFCS) and an anomalous model. Anomalous diffusion was not detected, but fits of the data with the multiple-component model revealed separable modes of diffusion. When the dominant mode of diffusion from the MEMFCS analysis was used, we observed that increased excluded volume slows probe mobility as a simple exponential with crowder concentration. This behavior can be modeled with a single parameter, β, which depends on the dextran size composition. Two additional modes of diffusion were observed using MEMFCS and were interpreted as unique microviscosities. The fast mode corresponded to unhindered free diffusion as in buffer, whereas the slower agreed well with the bulk viscosity. At 10% crowder concentration, one finds a microviscosity approximately three times that of water, which mimics that reported for intracellular viscosity.  相似文献

化工废水的排放是导致水环境毒物污染的重要来源,以原生动物为靶生物的微型生物群落监测——PFU法因能快速而真实地评价水体受污染程度而被广泛应用.盐城沿海化工园区是至今江苏省环保部门批准建设的苏北地区规模最大的以发展精细化工和医药化工为主导的专业园区,以该园区附近清洁水源中的原生动物为种源,用PFU法评价该园区化工废水的静态毒性.结果表明,原生动物群落对化工废水效应浓度(EC)变化非常敏感.在低的化工废水EC下,原生动物群落群集的物种多样性指数和群集种类均随毒性时间的延长而快速增加,群集速度也较快;随着化工废水EC的增加,原生动物群落群集的物种多样性指数随毒性时间的延长而增加缓慢甚至下降,群集种类则明显减少,群集速度也减缓,说明化工废水EC有较强的生物胁迫效应.在反映原生动物群集过程的3个参数Seq、G和T90%中,Seq与化工废水EC均呈负相关,而G随化工废水EC增加先呈负相关后呈正相关.根据其回归方程Seq=-0.141 EC+19.05 (R2=0.941,P<0.01)推算出该化工废水的效应浓度EC5、EC20和EC50分别为7.1%、27.3%和67.7%.最终确定盐城沿海化工园区化工废水对其附近清洁水源原生动物群落的安全浓度为7.1%,最大允许浓度为27.3%.盐城沿海化工园区化工废水必须处理至其毒性削减72.7%以上才能排放.  相似文献

We examined two aspects of the social control of nest climate in bumblebee colonies: which parameters of nest climate bumblebees actively down-regulate by fanning and the dynamics of the colony response as colony size increased. Colonies of Bombus terrestris were exposed to an increase in carbon dioxide, temperature or relative humidity. We performed 70 temperature trials (six colonies), 58 CO2 trials (four colonies) and four humidity trials (two colonies). An increase in CO2 concentration and temperature elicited a fanning response whereas an increase in relative humidity did not. This is the first report of fanning in bumblebee colonies to control respiratory gases. The number of fanning bees increased with stimulus intensity. The colony response to a CO2 concentration of 3.2% was comparable to the colony response to a temperature of 30°C. A marked fanning response occurred at 1.6% CO2, a concentration never exceeded in a large field nest during a pilot measurement of 10 days. We investigated the colony response over a wide range of colony sizes (between 10 and 119 workers). The proportion of the total workforce invested by colonies in nest ventilation did not change significantly; thus, the number of fanning workers increased with colony size. Furthermore, as colony size increased, the dynamics of the colony response changed: colonies responded faster to perturbations of their environment when they were large (60 or more individuals) than when they were small. This analysis was conducted to explore the association between 5 birth size measurements (weight, length and head, chest and mid-upper arm [MUAC] circumferences) as dependent variables and 10 maternal factors as independent variables using canonical correlation analysis (CCA). CCA considers simultaneously sets of dependent and independent variables and, thus, generates a substantially reduced type 1 error. Data were from women delivering a singleton live birth (n = 14506) while participating in a double-masked, cluster-randomized, placebo-controlled maternal vitamin A or β-carotene supplementation trial in rural Bangladesh. The first canonical correlation was 0.42 (P<0.001), demonstrating a moderate positive correlation mainly between the 5 birth size measurements and 5 maternal factors (preterm delivery, early pregnancy MUAC, infant sex, age and parity). A significant interaction between infant sex and preterm delivery on birth size was also revealed from the score plot. Thirteen percent of birth size variability was explained by the composite score of the maternal factors (Redundancy, RY/X = 0.131). Given an ability to accommodate numerous relationships and reduce complexities of multiple comparisons, CCA identified the 5 maternal variables able to predict birth size in this rural Bangladesh setting. CCA may offer an efficient, practical and inclusive approach to assessing the association between two sets of variables, addressing the innate complexity of interactions.  相似文献

The aim of this study was to prepare biodegradable sustained release magnetite microspheres sized between 1 to 2 μm. The microspheres with or without magnetic materials were prepared by a W/O/W double emulsion solvent evaporation technique using poly(lactide-co-glycolide) (PLGA) as the biodegradable matrix forming polymer. Effects of manufacturing and formulation variables on particle size were investigated with non-magnetic microspheres. Microsphere size could be controlled by modification of homogenization speed, PLGA concentration in the oil phase, oil phase volume, solvent composition, and polyvinyl alcohol (PVA) concentration in the outer water phase. Most influential were the agitation velocity and all parameters that influence the kinematic viscosity of oil and outer water phase, specifically the type and concentration of the oil phase. The magnetic component yielding homogeneous magnetic microspheres consisted of magnetite nanoparticles of 8 nm diameter stabilized with a polyethylene glycole/polyacrylic acid (PEG/PAA) coating and a saturation magnetization of 47.8 emu/g. Non-magnetic and magnetic microspheres had very similar size, morphology, and size distribution, as shown by scanning electron microscopy. The optimized conditions yielded microspheres with 13.7 weight% of magnetite and an average diameter of 1.37 μm. Such biodegradable magnetic microspheres seem appropriate for vascular administration followed by magnetic drug targeting.  相似文献

Aggregation of embryos in clutches that lack internal circulation can increase the risk of hypoxia by limiting gas exchange. As a result, limits on oxygen solubility and diffusion in water can constrain the size and embryo concentration of aquatic egg clutches. Hypoxia in egg masses can slow embryo development, increase mortality, and reduce size at hatching. The risk of hypoxia for embryos, however, can be reduced by association with photosynthetic organisms. We examined whether embryo development in egg ribbons of the cephalaspidean mollusk Haminoea vesicula is significantly influenced by oviposition on eelgrass (Zostera marina). Association with the photosynthetic substrate had marked effects on development relative to association with non-photosynthetic substrates, and the direction of these effects was mediated by light conditions. Under intermediate and high light levels, association with eelgrass accelerated embryo development, while under dim light, the presence of the macrophyte increased development rate and reduced hatchling shell size. Benefits of association with eelgrass at higher light levels likely result from oxygen production by eelgrass photosynthesis, while we attribute costs under low light to oxygen depletion by eelgrass respiration. Association with Z. marina also limited microphyte growth in egg ribbons of H. vesicula. In the field, measurements of light attenuation within an eelgrass bed showed that conditions under which benefits accrue to embryos are ecologically relevant and correspond to spatial patterns of oviposition on eelgrass in the field. The choice of a photosynthetic oviposition substrate under appropriate light conditions can improve embryo fitness by accelerating embryo development without compromising hatchling size and by reducing the potential for excessive and harmful fouling by microphytes.  相似文献

Abstract

Increasing levels of heavy metals in soil have become a serious concern for human health because they can be easily transferred into the human body through contaminated food web. It is imperative to evaluate pollution levels, origin and ecological risks of heavy metals. The geoaccumaualtion (Igeo), contamination factor (CF), pollution load index (PLI) and human health risk were estimated to determine the soil pollution in Faisalabad, a heavily-populated and industrialized city of Pakistan. The maximum CF (1.58) and PLI (1.22) values were estimated for Cd and Pb, respectively, and maximum Igeo (?0.19) value was observed for Cd. Correlation analysis and principal component analysis suggested that common industrial sources for Cd and Pb were identified in the study sites. It clearly indicates that the significant levels of heavy metals pollution arise from local industries, busy commercial centers and heavy traffic load in the last few decades in heavily-populated and industrialized city. Further, soil heavy metals concentration were used to evaluate the human health risk such as chronic or non-carcinogenic including hazard indexes HIexP (ingestion, inhalation and dermal and carcinogenic) and cancer risk (CR). The HIexP values of Pb (10.30) and Cd (4.56) were found above the permissible limit (HI = 1) for children. The CR due to carcinogenic metals (Co, Cr and Cd) are within the safe limit (1E-06 to 1E-04). However, CR was comparatively higher in adults as compared to children. The results from the current investigation can help to develop a sustainable strategy in the study region to minimize the entry of heavy metals in food chain through source identification and pollution abatement techniques.  相似文献

The evolution of the investment in exaggerated secondary sexual traits is a topic of great interest for scientists. Despite antlers in the family Cervidae being one of the most interesting allometric structures, the nature of the relationships between antler and body size, and the influence of physiological factors driving the evolution of these characters, still remain unclear. In this paper, I examine these relationships in depth using the largest sample size ever studied (43 species). Under the hypothesis that antler growth may be limited by skeleton size as this process requires the allocation of huge amounts of mineral resources to the antlers, skeleton-related variables may more accurately explain these allometric relationships. The existence of physiological constraints should therefore be more clearly highlighted when studying the relationships between body size variables and the relative investment in the antler (measured as length or mass of antler per kg of skeleton). Results show that antler length is best described as being linearly related to head-body length rather than other measurements of size, and antler weight has a quadratic relationship with body mass. However, the relative investment in antler length (related to skeleton mass) is independent of body size variables, while the relative investment in antler mass has a quadratic relationship with shoulder height. The results obtained for antler mass reflect the existence of physiological constraints in the evolution of antlers, which are greater for larger sized species. On the other hand, the evolution of antler length may be linked to other factors, most probably sociobiological and biomechanical ones.  相似文献
