The physiological reasons associated with differential sensitivity of C3 and C4 plant species to soil compaction stress are not well explained and understood. The responses of growth characteristics, changes in leaf water potential and gas exchange in maize and triticale to a different soil compaction were investigated. In the present study seedlings of triticale and maize, representative of C3 and C4 plants were subjected to low (L – 1.10 g cm−3), moderate (M – 1.34 g cm−3) and severe (S – 1.58 g cm−3) soil compaction level. Distinct differences in distribution of roots in the soil profile were observed. Plants of treatments M or S in comparison to treatment L, showed a decrease in leaf number, dry mass of stem, leaves and roots, and an increase in the shoot to root ratio. A drastic decrease in root biomass in M and S treatments in the soil profile on depth from 15 to 40 cm was observed. Any level of soil compaction did not influence the number of seminal and seminal-adventitious roots but decreased their length. The number and total length of nodal roots decreased with compaction. Changes of growth traits in M and S treatments in comparison to the L were greater for maize than for triticale and were accompanied by daily changes in water potential (ψ) and gas exchange parameters (PN, E, gs). Differences between M and S treatments in daily changes in ψ for maize were in most cases statistically insignificant, whereas for triticale, they were statistically significant. Differences in the responses of maize and triticale to soil compaction were found in PN, E and gs in particular for the measurements taken at 12:00 and 16:00. The highest correlation coefficients were obtained for the relationship between leaf water potential and stomatal conductance, both for maize and triticale, which indicates the close association between stomata behavior and changes in leaf water status.  相似文献

This study investigates erythromycin toxicity toward activated sludge as a function of exposure time and antibiotic concentration. Batch experiments were conducted and microscopic techniques ranging from bright-field microscopy to epifluorescence and confocal laser scanning microscopy (CLSM), combined with a fluorescent viability indicator (BacLight® Bacterial Viability Kit, Molecular Probes), allowed us to study erythromycin time-kill activity. The erythromycin toxicity was observed at lower concentration when exposure time increased. A 4 μg/L erythromycin concentration was toxic to heterotrophic bacteria on a 5-day time exposure, and a 5 mg/L concentration inhibited nitrification. These findings are in agreement with the microscopic studies, which showed a latency time before the lower antibiotic concentrations began to kill bacteria. Microscope slide wells were used as micro-reactors in which erythromycin concentration ranged from 0.1 to 1 mg/L. After 45 min there were 94% (SD 3.8) of living bacteria in control micro-reactors, 67% (SD 3.1) in micro-reactors that contained 0.1 mg/L erythromycin and 37% (SD 18.6) in micro-reactors that contained 1 mg/L erythromycin. CLSM allowed visualization of isolated stained cells in the three-dimensional structure of damaged flocs.  相似文献

In this study, we evaluated the hydrocarbon removal efficiency and microbial diversity of different soil layers. The soil layers with high counts of recoverable hydrocarbon degrading bacteria had the highest hydrocarbon removal rate compared with soil layers with low counts of hydrocarbon degrading bacteria. Removal efficiency was 48% in the topsoil, compared with 31% and 11% at depths of 1.5 and 1 m, respectively. In the 1 and 1.5 m soil layers, there was no significant difference between total petroleum hydrocarbon (TPH) removal in nutrient amended treatments and controls. The respiration rate reflected the difference in the number of bacteria in each soil layer and the availability of nutrients. High O2 consumption corresponded positively with high TPH removal. Analysis of the microbial diversity in the different soil layers using functional diversity (community-level physiological profile, via Biolog) and genetic diversity using polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) of 16S rDNA revealed differences in, respectively, substrate utilisation patterns and DGGE profiles of 16S rDNA fragments. Microbial diversity as revealed by DNA fragments was lower in the highly contaminated soil layer (1.5 m) than in the topsoil and at 1 m.  相似文献

The objective was to look for the promising tree leaves, for feeding to livestock. The preliminary screening of tree leaves (nine species) by in vitro gas production technique revealed that leaves of Azadirachta indica, Melia azedarach, Morus alba and Leucaena leucocephala could serve as promising, alternate feed resource for ruminants. Therefore, in vivo evaluation of these tree leaves (except A. indica) along with that of Toona ciliate was assessed. Each species of the fresh tree leaves, supplemented with mineral mixture and common salt, was offered ad lib as complete feed to three bucks (Beetle × Anglo Nubian × French Alpine; 6 years old of 56.7 ± 1.12 kg BW). The data were analyzed by using completely randomized design. Bucks relished all the tree leaves (except T. ciliate) as indicated by higher (P < 0.05) voluntary DM intake and digestibility of nutrients. All the animals were in positive N-balance except those fed Toona ciliate. The N-retention was maximum in animals fed M. alba followed by that in L. leucocephala and M. azedarach. However, the DCP content of M. alba, M. azedarach and L. leucocephala was statistically comparable, while ME availability was higher (P < 0.05) from leaves of M. azedarach followed by that of M. alba, clearly indicated that leaves of M. azedarach, M. alba or L. leucocephala, supplemented with mineral mixture and common salt, could serve as an excellent complete feed for small ruminants.  相似文献

Twelve novel fenfuram-diarylether hybrids were designed, synthesized and characterized by 1H NMR and MS. Their in vitro antifungal activities were evaluated against five phytopathogenic fungi by mycelial growth inhibition method. Most compounds showed significant antifungal effect on Rhizoctonia solani and Sclerotinia sclerotiorum. Compound 1c exhibited the most potent antifungal effect on R. solani with an EC50 value of 0.242 mg/L, superior to the commercial fungicide boscalid (EC50 = 1.758 mg/L) and the lead fungicide fenfuram (EC50 = 7.691 mg/L). Molecular docking revealed that compound 1c featured a higher affinity for succinate dehydrogenase (SDH) than fenfuram. Furthermore, it was shown that the 2-chlorophenyl group of compound 1c formed a π-π stacking with D/Tyr-128 and a Cl-π interaction with B/His-249, which made compound 1c more active than fenfuram against SDH.  相似文献

Pike-perch Sander lucioperca is currently considered as one of the most promising candidates for production in freshwater recirculation aquaculture systems (RAS). Here, due to the lack of studies on nitrite (NO2?) toxicity in pike-perch, a flow-through exposure at 0, 0.44, 0.88, 1.75, 3.5, 7, 14 and 28 mg/L NO2?–N was carried out to determine the acute and chronic toxicity over a period of 32 days. In juvenile pike-perch, 120 h LC50 was 6.1 mg/L NO2?–N and at ≥ 14 mg/L NO2?–N all fish had died within 24 h. Chronic exposure revealed a significant build up of NO2? in the plasma as well as in the muscles at ≥ 0.44 mg/L NO2?–N peaking in fish exposed to the highest concentration of 3.5 mg/L NO2?–N after 32 days. Still, due to high individual variation methemoglobin (MetHb) was only significantly increased (p < 0.01) at 3.5 mg/L NO2?–N. No adverse effects on red blood cells (RBC) and hematocrit were observed in any of the treatments. In a second experiment, compensation of NO2? toxicity at increasing chloride concentrations (40 (freshwater), 65, 90, 140, 240, 440 mg/L Cl?) was observed at a constant exposure of 10 mg/L NO2?–N for 42 days. At ≥ 240 mg/L Cl?, NO2? build-up in blood plasma and muscle was completely inhibited. At lower Cl? concentrations (≤ 140 mg/L), NO2? was significantly increased in plasma, but only insignificantly elevated in muscle due to high individual variation. MetHb was increased significantly difference only at 40 mg/L Cl? (freshwater control) compared to the control. Again, high individual variations were observed. As a conclusion, S. lucioperca is moderately sensitive towards NO2? and acceptable levels in RAS should hence not exceed 1.75 mg/L NO2?–N to avoid MetHb formation. However, based on the 120 h LC50 and a factor of 0.01 according to Sprague (1971), a NO2? concentration of ≤ 0.061 mg/L NO2?–N is considered as “safe.” Thereby, no NO2? should accumulate in the plasma or muscle tissue during chronic exposure. For 10 mg/L NO2?–N, ≥ 240 mg/L chloride compensates for NO2? uptake in plasma and muscle.  相似文献

During our screening program for agrochemicals from Chinese medicinal herbs and wild plants, the essential oil of Litsea cubeba fruits was found to possess strong contact toxicity against the cigarette beetle Lasioderma serricorne adults and the booklouse Liposcelis bostrychophila, with LD50 values of 27.33 μg/adult and 71.56 μg/cm2, respectively, and also showed strong fumigant toxicity against the two stored product insects with LC50 values of 22.97 and 0.73 mg/L, respectively. The essential oil obtained by hydrodistillation was investigated by GC MS. The main components of the essential oil were identified to be E-citral (geranial) (27.49%), Z-citral (neral) (23.57%) and d-limonene (18.82%) followed by β-thujene (3.34%), β-pinene (2.85%), α-pinene (2.57%), 6-methyl-5-hepten-2-one (2.40%) and linalool (2.36%). Citral (Z/E-citral), d-limonene, β-pinene, α-pinene and linalool were separated and purified by silica gel column chromatography and preparative thin layer chromatography, and further identified by means of physicochemical and spectrometric analysis. Citral and linalool showed strong contact toxicity against L. serricorne and L. bostrychophila (LD50 = 11.76, 12.74 μg/adult and 20.15, 99.97 μg/cm2, respectively) and fumigant toxicity against L. serricorne and L. bostrychophila (16.54, 18.04 mg/L air and 0.14, 0.71 mg/L air, respectively). Otherwise, citral, d-limonene and linalool were strongly repellent against the cigarette beetle L. serricorne as the essential oil whereas β-pinene and α-pinene exhibited weaker repellency against the cigarette beetle compared with the positive control, DEET. Moreover, except α-pinene and linalool, the other three compounds as well as the essential oil exhibited comparable repellency against the booklouse relative to DEET.  相似文献

Biomass production, dry matter content, specific leaf area and pigment content of Chinese cabbage were all quite similar, when plants were grown in the absence or presence of UV-A + B (2.2 mW cm−2). Elevated Cu2+ concentrations (2–10 μM) in the root environment and UV radiation had negative synergistic effects for Chinese cabbage and resulted in a more rapid and stronger decrease in plant biomass production and pigment content. The quantum yield of photosystem II photochemistry (Fv/Fm) was only decreased at ≥5 μM Cu2+ in the presence of UV radiation, when leaf tissue started to become necrotic. The enhanced Cu toxicity in the presence of UV was largely due to a UV-induced enhanced accumulation of Cu in both roots and shoots. An enhanced Cu content strongly affected the uptake and assimilation of sulfur in plants. The total sulfur content of the root increased at ≥2 μM Cu2+ in presence of UV and at 10 μM Cu2+ in absence of UV and that of the shoot increased at ≥2 μM Cu2+ in presence of UV and at ≥5 μM Cu2+ in absence of UV. In the shoot it could be attributed mainly to an increase in sulfate content. Moreover, there was a strong increase in the water-soluble non-protein thiol content upon Cu2+ exposure in the root and, to a lesser extent in the shoot, both in the presence and absence of UV. The regulation of the uptake of sulfate responded to the occurrence of Cu toxicity directly, since it was more rapidly affected in the presence than in the absence of UV radiation. For instance, the expression and activity of the high affinity sulfate transporter, Sultr1;2, were enhanced at ≥2 μM in the presence of UV, and at ≥5 μM Cu2+ in the absence of UV. In the shoot, the expression of the vacuolar sulfate transporter, Sultr4;1, was upregulated at ≥5 μM Cu2+ in the presence and absence of UV whilst the expression of a second vacuolar sulfate transporter, Sultr4;2, was upregulated at 10 μM Cu2+ in the presence of UV. It is suggested that high Cu tissue levels may interfere/react with the signal compounds involved in the regulation of expression and activity of sulfate transporters. The expression of adenosine 5′-phosphosulfate reductase in the root was hardly affected and was slightly down-regulated at 2 μM in the presence of UV and at 10 μM in the absence of UV. The expression and activity of sulfate transporters were enhanced upon exposure at elevated Cu2+ concentrations; this may not be simply due to a greater sulfur demand at higher Cu levels, but more likely is the consequence of Cu toxicity, since it occurred more rapidly in the presence compared to the absence of UV.  相似文献

BackgroundSilver nanoparticles (Ag-NPs) are widely used nowadays in a variety of commercial applications including medical, health care, textiles and household supplies.ObjectivesThe current study was designed to determine the median lethal dose (LC50) of Ag-NPs on Oreochromis niloticus and Tilapia zillii.MethodsAcute and sub-acute toxicity study of the Ag-NPs on brain tissues was carried out using different concentrations of the NPs at 2 mg L and 4 mg L. These concentrations were dispersed in deionized water with the exception of the control groups in the experiments. Biochemical and molecular analysis were conducted on tissue homogenates in order to evaluate the potential effects of NPs on the antioxidant system.ResultsThe Ag-NP acute toxicity (96 h LC50) values of 19.5 ± 2 and 20 ± 2.4 mg/L were reported for O. niloticus and T. zillii respectively. Fish exposed to 2 mg/L Ag-NPs did not show any significant change in the levels of reduced glutathione (GSH), total glutathione (tGSH) levels, superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR), glutathione peroxidase (GPx) and glutathione-S-transferase (GST) activity or genes expression and malondialdehyde (MDA) level. In contrary, a dose of 4 mg/L showed a significant reduction in the levels all the above-mentioned parameters except in MDA level where it was significantly induced.ConclusionResults indicate that exposure of O. niloticus and T. zillii to Ag-NPs (4 mg/L) has deleterious effects on brain antioxidant system, whereas a dose of 2 mg/L has no effects.  相似文献

Methane (CH4) and nitrous oxide (N2O) are important greenhouse gases, because of their contribution to the global greenhouse effect. The present study assessed emissions of N2O and CH4 from constructed wetland microcosms, planted with Phragmites australis and Zizania latifolia, when treating wastewater under different biological oxygen demand (BOD) concentration conditions. The removal rate was 95% for BOD and more than 80% for COD in all three pollutant concentrations, both plants’ removal rates of pollutants were at almost the same level, and both were found to resist BOD concentrations as high as 200 mg L−1. When BOD concentrations fell below 200 mg L−1, the soil plant units reached an average of 80–92% T-N and T-P removal rates; however, as the concentrations increased to 200 mg mg L−1 or when during the initial phases of winter, the removal rates for T-N and T-P decreased to less than 70%. With NH3-N removal, the influences of BOD concentrations and air temperature were more obvious. When BOD concentrations increased to 100 mg L−1 after October, an obvious decrease in NH3-N removal was detected; almost no nitrification occurred beginning in December at BOD concentrations of 200 mg mg L−1. N2O and CH4 emissions showed obvious seasonal changes; higher emissions were observed with higher BOD concentrations, especially among Z. latifolia units. The enumeration of methane-oxidizing bacteria and methane-producing bacteria was also conducted to investigate their roles in impacting methane emissions and their relationships with plant species. The pollutant purification potentials of P. australis and Z. latifolia plant units during wastewater treatment of different pollutant concentrations occurred at almost the same levels. The nutrient outflow and methane flux were consistently higher with Z. latifolia units and higher concentrations of BOD. The more reductive status and higher biomass of methanogens may be the reason for the lower nitrification and higher CH4 emissions observed with Z. latifolia units and higher concentration systems. The Z. latifolia root system is shallow, and the activity of methanotrophs is primarily confined to the upper portion of the soil. However, the root system of P. australis is deeper and can oxidize methane to a greater depth. This latter structure is more favorable as it is better for reducing methane emissions from P. australis soil plant systems.  相似文献

The aim of this field investigation was to study the enrichment of biodiversity of the slope at an early phase of succession, initiated by selected pioneers, and to study how this enrichment related to enhancement of the slope stability. Four experimental plots, with differing plant pioneers and number of species (diversity), were designed in order to assess the effects of plant succession on slope stability. Plant growth pattern was assessed by observing the increment in species diversity (number), species frequency and plant biomass. Higher vegetation biomass in a mixed culture situation (LLSS) in the field with Leucaena leucocephala as a pioneer, marked an increase in species diversity after 24 months of observation. In contrast, G (grasses and legume creepers) plot revealed the slowest rate of succession and the lowest above-ground biomass amongst the plots. The mixed-culture plot without L. leucocephala (SS) had also shown a lower biomass, a similar phenomenon observed in a plot grown by L. leucocephala (LL) with low plant diversity. Consequently, these plant growth patterns gave a positive effect on slope stability where the regression study showed that the shear strength was much affected by plant biomass. Meanwhile, throughout the succession process in LLSS plot, root length density reached the highest value amongst the plots, 23 Km m?3. In relation to this, the saturation level of the slope indicates the unsaturated condition of the soil which resulted in the enhancement of both soil penetrability and soil shear strength of the plot. These attributes reveal a strong positive relationship between the process of natural succession and the stability of slopes.  相似文献

A purification process for the manufacture of a recombinant C-terminus heavy chain fragment from botulinum neurotoxin serotype C [rBoNTC(Hc)], a potential vaccine candidate, has been defined and successfully scaled-up. The rBoNTC(Hc) was produced intracellularly in Pichia pastoris X-33 using a three step fermentation process, i.e., glycerol batch phase, a glycerol fed-batch phase to achieve high cell densities, followed by a methanol induction phase. The rBoNTC(Hc) was captured from the soluble protein fraction of cell lysate using hydrophobic charge induction chromatography (HCIC; MEP HyperCel?), and then further purified using a CM 650M ion exchange chromatography step followed by a polishing step using HCIC once again. Method development at the bench scale was achieved using 5–100 mL columns and the process was performed at the pilot scale using 0.6–1.6 L columns in preparation for technology transfer to cGMP manufacturing. The process yielded approximately 2.5 g of rBoNTC(Hc)/kg wet cell weight (WCW) at the bench scale and 1.6 g rBoNTC(Hc)/kg WCW at the pilot scale. The purified rBoNTC(Hc) was stable for at least 3 months at 5 and ?80 °C as determined by reverse phase-HPLC and SDS–PAGE and was stable for 24 months at ?80 °C based on mouse potency bioassay. N-Terminal amino acid sequencing confirmed that the N-terminus of the purified rBoNTC(Hc) was intact.  相似文献

The present work evaluated the synergistic effects of soil fertilization with rock P and K materials and co-inoculation with P and K-dissolving bacteria [PDB (Bacillus megaterium var. phosphaticum) and KDB (Bacillus mucilaginosus and B. subtilis)] on the improvement of P and K uptake, P and K availability and growth of maize plant grown under limited P and K soil conditions (calcareous soil). The experiment was establishment with eight treatments: without rock P and K materials or bacteria inoculation (control), rock P (RP), rock K (RK), RP + PDB, RK + KDB and R(P + K)+(P + K)DB. Under the same conditions of this study, co-inoculation of PDB and KDB in conjunction with direct application of rock P and K materials (R(P + K)) into the soil increased P and K availability and uptake, and the plant growth (shoot and root growth) of maize plants grown on P and K limited soils.  相似文献

Rodents develop a marked elevation in core temperature (Tc) when they are placed in an open field environment (OFE). Open field-induced hyperthermia (OFH) involves activation of thermoregulatory systems that are quiescent under baseline conditions. Cholinergic stimulation arising from exposure to organophosphates (OP) is expected to modulate OFH; subtle effects of OPs may be observed during OFH that would not occur in the absence of stress. We have found that methyl scopolamine (MS), a peripheral muscarinic antagonist, attenuates stress-induced hyperthermia. To this end, we assessed if a low dose of the OP pesticide chlorpyrifos (CHP) would alter OFH with and without administration of MS. Tc and motor activity (MA) of male, Sprague-Dawley rats were monitored by telemetry while housed in their home cage at an ambient temperature of 22°C. At 9 AM the rats were gavaged with corn oil or 10 mg/kg CHP that resulted in a ∼40% inhibition in plasma cholinesterase activity. Tc and MA were the same in the control and CHP groups prior to OFE. The rats were then dosed IP with saline or 1.0 mg/kg MS at 12 noon and subjected to OFE (61×61×61 cm box) for 1 h while Tc and MA were monitored every 2 min. Tc of the corn oil/saline group increased by 1.2°C during OFE, whereas Tc of the CHP/saline group was significantly attenuated. Administration of MS attenuated the OFH in rats treated with corn oil and CHP. Overall, exposure to OFE exacerbated the effects of a relatively low dose of CHP that had no effect in the unstressed animal. In addition, the OFH response to MS suggests a peripheral (i.e., outside of CNS) cholinergic pathway is operative in the control of OFH.  相似文献

This study investigates the potential of using Normalized Difference Vegetation Index (NDVI) and Enhanced Vegetation Index (EVI) to estimate root zone soil moisture at native in-situ measured sites, and at distant sites under the same climatic setting. We obtained in-situ data from Soil Climate Analysis Network (SCAN) sites near the Texas-New Mexico border area, and NDVI and EVI products from the Moderate Resolution Imaging Spectroradiometer (MODIS) sensor on board the Terra satellite. Results show that soil moisture values of the same depth are highly correlated (r = 0.53 to 0.85) among sites as far as 150 km apart, and that NDVI and EVI are highly correlated at the same site (r = 0.87 to 0.91). Correlation based on raw time series of NDVI and soil moisture is in general higher than that based on deseasonalized time series at every depth. The correlation reaches maximum value when vegetation index (VI) lags soil moisture by 5 to 10 days. NDVI shows a slightly higher correlation with soil moisture than EVI does by using the deseasonalized time series of NDVI and soil moisture. It is found that deseasonalized time series of NDVI and soil moisture are correlated at native sites (r = 0.33 to 0.77), but not at sites where soil moisture is very low. Regression analysis was conducted using deseasonalized time series soil moisture and deseasonalized time series NDVI with a 5-day time lag. Regression models developed at one site and applied to a similar distant site can estimate soil moistures, accounting for 50–88% of the variation in observed soil moistures.  相似文献

A series of 23 novel bis-phosphonium salts based on pyridoxine were synthesized and their antibacterial activities were evaluated in vitro. All compounds were inactive against gram-negative bacteria and exhibited the structure-dependent activity against gram-positive bacteria. The antibacterial activity enhanced with the increase in chain length at acetal carbon atom in the order n-Pr > Et > Me. Further increasing of length and branching of alkyl chain leads to the reduction of antibacterial activity. Replacement of the phenyl substituents at the phosphorus atoms in 5,6-bis(triphenylphosphonio(methyl))-2,2,8-trimethyl-4H-[1,3]-dioxino[4,5-c]pyridine dichloride (compound 1) with n-butyl, m-tolyl or p-tolyl as well as chloride anions in the compound 1 with bromides (compound 14a) increased the activity against Staphylococcus aureus and Staphylococcus epidermidis up to 5 times (MICs = 1–1.25 μg/ml). But in practically all cases chemical modifications of compound 1 led to the increase of its toxicity for HEK-293 cells. The only exception is compound 5,6-bis[tributylphosphonio(methyl)]-2,2,8-trimethyl-4H-[1,3]dioxino[4,5-c]pyridine dichloride (10a) which demonstrated lower MIC values against S. aureus and S. epidermidis (1 μg/ml) and lower cytotoxicity on HEK-293 cells (CC50 = 200 μg/ml). Compound 10a had no significant mutagenic and genotoxic effects and was selected for further evaluation. It should be noted that all bis-phosphonium salt based on pyridoxine were much more toxic than vancomycin.  相似文献

In soil, high variability of U bioavailability results in large range of apparent U toxic levels for plants. U toxicity on hairy roots of carrot was studied in nutrient gel with a standardized in vitro device. After exposure to 2.5 and 20 mg U L?1 for 34 days, U concentration ranged between 4 and 563 mg U kg?1 fresh weight which was in good accordance with U accumulation by roots of plant from contaminated soils. Threshold of U toxicity for root length decreased with time and a transient hormesis occurred for exposure to 2.5 and 5 mg U L?1. After 34 days and with root length as endpoint, significant toxicity appeared at a gel contamination level above 7.5 mg U L?1 corresponding to a maximum U concentration in the liquid phase of 0.8 mg L?1. The calculated EC50 for root length as a function of gel contamination was 9.4 mg U L?1. Lower threshold and EC50 were observed for biomass as endpoint (resp. 5 and 7.3 mg U L?1). The low values observed in this study could result from high sensitivity of carrot to U, high bioavailability of U in gel or absence of interferences with microorganisms. This in vitro device appeared adapted to study toxicity of U to plant roots in optimal conditions of both exposure and observations and is recommended to examine further physiological processes and the influence of microorganism interactions.  相似文献

Responses of Japanese mustard spinach (JM-spinach; Brassica rapa L. var. pervirdis) were investigated at elevated levels of arsenic (As). Plants were grown hydroponically in the greenhouse under 0, 6.7, 33.5 and 67 μM As (equal to 0, 0.5, 2.5 and 5 mg L?1 As, respectively) for 14 days. Arsenic was used as sodium meta-arsenite (NaAsO2). Toxicity symptom was solely shown as shoot growth repression at 33.5 and 67 μM As exposures. Dry weight (DW) enhanced by 19.4% in shoot and 38.9% in root in the 6.7 μM As level as compared to control but decreased by 48.1% and 72.1% DW in shoot and 24.1% and 61.1% DW in root in the 33.5 and 67 μM As levels, respectively. This result indicated that As at lower concentration might have slight stimulating effect on JM-spinach growth, but toxicity increased with increasing As. Based on the regression lines between growth and As concentration in the plant tissues, the critical toxicity level (CTL) of As in JM-spinach shoot was 7.85 μg g?1 DW considering 10% DW reduction. The CTL for the root was almost 2110 μg As g?1 DW, indicating that shoot of JM-spinach was more sensitive to As-toxicity than that of root. Arsenic concentrations increased in plant parts with increasing As in the medium. Arsenic concentrations were also compared in DW and fresh weight (FW) basis. The JM-spinach concentrated unaccepted level of As in shoots for human consumption in the higher As levels without showing visible toxicity symptom. In spite of decreasing iron (Fe) concentration in shoot in the highest As level, chlorophyll index did not decrease accordingly. Phosphorus (P) concentration also decreased. Phosphorus concentration decreased much more than Fe concentration. Low P might help to mobilize Fe in shoots, resulting in higher chlorophyll index at 67 μM As level. Phosphorus might compete with Fe in shoot tissues of As-stressed JM-spinach.  相似文献

Forest vegetation is known to enhance the stability of slopes by reinforcing soil and increasing its shear resistance through root system. The effects of root reinforcement depend on the morphological characteristics of the root system, the tensile strength of single roots, and the spatial distribution of the roots in soil. In the present study the results of research carried out in order to evaluate the biotechnical characteristics of the root system of Persian Ironwood (Parrotia persica), in northern Iran are presented. Profile trenching method was used to obtain root area ratio (RAR) values for uphill and downhill sides of the individual trees. For each species, single root specimens were sampled and tested for their tensile strength. It was found that root density generally decreases with depth according to an exponential law. Maximum RAR values were located within the first 0.1 m, with maximum rooting depth at about 0.65 m. RAR values ranged from 0.001% at lower depths to 1.39% near the surface, at upper 0.1 m depth. Significant differences of RAR values, rooting depth and root cohesion between uphill and downhill were observed, however, the differences were not significant for number of roots (ANCOVA). Downhill profiles had higher RAR values, rooting depth and root cohesion. In general, root tensile strength tends to decrease with diameter according to a power law, as observed by other researchers. Downhill roots were significantly stronger in tensile strength than uphill ones. Inter-species variation of tensile strength in downhill roots was also observed. The resulting data were used to evaluate the reinforcing effects in terms of increased shear strength of the soil, using Wu/Waldron Model. The root reinforcement provided by Persian Ironwood is about 46.0 kPa in the upper layers and 0.3 kPa in the deeper horizons. The results of Spearman test revealed a significant correlation between RAR and cr and that best followed by a power law. The results presented in this paper contribute to expanding the knowledge on biotechnical characteristics of Persian Ironwood on slope reinforcement.  相似文献

Numerous species of soil bacteria which flourish in the rhizosphere of plants or around plant tissues stimulate plant growth and reduce nematode population by antagonistic behavior. These bacteria are collectively known as PGPR (plant growth promoting rhizobacteria). The effects of six isolates of PGPR Pseudomonas putida, Pseudomonas fluorescens, Serratia marcescens, Bacillus amyloliquefaciens, Bacillus subtilis and Bacillus cereus, were studied on tomato plant growth and root knot nematode reproduction after 45 days from nematode infection. The highest number of shoot dry weight/g (43.00 g) was detected in the plant treated with S. marcescens; then P. putida (34.33 g), B. amyloliquefaciens (31.66 g), P. fluorescens (30.0 g), B. subtilis (29.0 g), B. cereus (27.0 g) and nematode alone (untreated) 20 g/plant. While the highest number of plant height was observed when plant was treated with S. marcescens, P. fluorescens, P. putida, B. amyloliquefaciens and P. putida 52.66, 50.66, 48 and 48 cm respectively. No significant differences were seen between previous treatments but only had significant differences compared with untreated plant. The highest number of fruit/plant was observed when plants were treated with S. marcescens (10.66), then B. amyloliquefaciens (8.66), P. putida (8), P. fluorescens (8) and B. cereus (7.66). No significant differences between the last 4 treatments, but all had significant differences compared with untreated plants. The highest weight of plant yield (g) was observed with S. marcescens (319.6 g/plant) and the lowest weight of plant yield was observed in plants treated with nematode alone (untreated). On the other hand, the lowest numbers of J2/10 g of soil (78), galls/root, (24.33) galls/root, egg masses/root (12.66) and egg/egg masses were observed in the plants treated with S. marcescens.  相似文献
