Four 13-hydroxygibberellins, gibberellin A1 (GA1), 3-epi-GA1,GA19 and GA20 were identified by full-scan GC/MS in extractsof lettuce seedlings (Lactuca sativa L. cv. Grand Rapids). Theresults suggest that the early-13-hydroxylation biosyntheticpathway to GA1 functions in the lettuce seedlings. It was alsofound that GA1 is active per se in the control of hypocotylelongation in lettuce seedlings. To investigate the relationshipbetween control by light of hypocotyl elongation and levelsof endogenous GAs in lettuce, endogenous levels of GAs werequantified by radioimmunoassay in seedlings that had been grownfor 5 days in the dark (5D) and in those that had been grownfor 4 days in the dark and then under white light for 1 day(4D1L). The endogenous level of GA1 in the upper and lower partsof hypocotyls in 5D seedlings was about four times higher thanthat in 4D1L seedlings. The response of explants (hypocotylsegments with cotyledons) from dark-grown seedlings to GA1 isknown to be similar in the dark and under white light when theexplants are treated with inhibitors of the biosynthesis ofGA. Therefore, the above information suggests that the highlevel of GA1 in hypocotyls of dark-grown seedlings is responsiblefor the rapid elongation of hypocotyl, while irradiation bywhite light decreases the endogenous level of GA1 in the hypocotylswith a resultant decrease in the rate of hypocotyl elongation. The presence of GA9, GA19 and GA20 was demonstrated by gas chromatography/massspectrometry (GC/MS) and the presence of GA44 strongly indicatedby GC/MS in selected ion monitoring mode (GC/SIM) in extractsof shoots of light-grown tall peas (Pisum sativum L.). Usingthe rice seedling bioassay with cv. Tan-ginbozu, the levelsof gibberellins in pea shoots were monitored from early shootgrowth through to apical senescence in a tall pea line. Levelsof activity corresponding to GA20, GA19 and GA44 remained relativelystable in the shoot despite reproductive development and apicalsenescence. The level of GA1-like activity increased to a maximumwhen the leaves had between 7 and 9 leaves expanded and decreasedonly with apical senescence. The na gene which blocks the productionof biologically active gibberellins in shoots but not in developingseed, was also operative in pod walls, with na pods containinglittle or no significant gibberellin-like activity when comparedto na pods at contact. This occurred despite the presence ofrelatively high levels of gibberellins in developing seed atthe same time. The results suggest that there is little or nosignificant leakage of biologically active gibberellins fromdeveloping seed to pods or shoots. Extracts of pods of tallpeas with Na contained low levels of gibberellin-like activitybut like developing seed, contained little or no significantGA1-like activity despite the presence of significant GA1-likeactivity in shoot extracts of tall peas. An antiserum recognizing free gibberellins (GAs) was preparedby immunizing rabbits with a GA4-BSA conjugate. A radioimmunoassay(RIA) and an enzyme-linked immunosorbent assay (ELISA) wereset up using this antiserum. This antiserum showed high cross-reactivityto the so-called active GAs, such as GA1, GA3, GA4 and GA7.The range for measurements of these gibberellins extended from30 fmol to 3 pmol in both RIA and ELISA. Extracts from immature seeds of P. vulgaris were subjected todetermination of GA, by RIA and GC/SIM. The change of endogenous gibberellin and ABA content was measuredduring the fruit development of Prunus persica. GA5, GA32 andtwo unidentified gibberellins, were found throughout the developmentalstages. Endogenous levels of gibberellins in shoots and ears of twodwarf rice (Oryza sativa L.) cultivars, Tan-ginbozu (dx mutant)and Waito-C (dy mutant), were analyzed and compared with thoseof normal rice cultivar, Nihonbare. The endogenous levels of13-hydroxylated gibberellins in Tan-ginbozu were much lowerthan those in Nihonbare. In Waito-C, the levels of GA19 andGA20 in the shoots were higher but that of GA1 was lower thanthe levels of these gibberellins in Nihonbare. These resultssupport the hypothesis that the dy gene controls the 3ß-hydroxylationof GA20 to GA1 while the dx gene controls a much earlier stepin the gibberellin biosynthesis. Our results indicate that GA1is the active gibberellin that regulates the vegetative growthof rice. The endogenous levels of GA4 in the ears of the twodwarf cultivars of rice were higher than the level of GA4 inthe ears of the normal cultivar, Nihonbare suggesting that thebiosynthesis of gibberellin is not blocked in the anthers ofthe dwarf rice. Auxin activates pumping of protons from the symplast to theapoplast and causes hyperpolarization of the symplast membranein the elongation zone of Vigna stems prior to the accelerationof growth. This auxin-induced hyperpolarization has been studiedin most cases in hypocotyl segments excised from the elongationzone. In the present study, mature-zone segments were perfusedwith IAA by the xylem perfusion technique in an effort to determinewhether or not IAA has any effects in the mature zone. Althoughno hyperpolarization of the symplast membrane was observed uponthe perfusion with auxin alone, auxin-induced hyperpolarizationwas observed when mature-zone segments had been pretreated withGA3, in the absence of an increase in the growth rate. Theseresults suggest that cells in the mature zone have lost theability to activate the proton-pumping machinery in responseto auxin but that this ability can be restored by treatmentwith GA3. This effect of GA3 suggests the possibility that theconcentration of gibberellin in a tissue controls one of thecell's responses to auxin, namely, activation of the protonpump. Maize (Zea mays L., hybrid Cargill 147) seedlings, grown inaseptic conditions, were inoculated with three strains of Azospirillumlipoferum (Al op 33, Al iaa 320, and ATCC 29708) or culturedin different concentrations of indol-3-acetic acid (IAA) orgibberellin A3 (GA3). After 48 h, root length, root surfacearea, root dry weight, and shoot dry weight were measured inall treatments. Gibberellin content was evaluated in selectedroots of control and Azospirillum inoculated seedlings by gaschromatography-mass spectrometry-selected ion monitoring withthe use of deuterated gibberellins as internal standards. Thethree strains of A. lipoferum, IAA (2 ng ml–1), and GA3(40 to 400 pg ml–1) significantly enhanced root growth.Improvement of root hair growth and density was obtained mainlywith A. lipoferum strain Al op 33 and GA3 40 pg ml–1.GA3 was identified by gas chromatography-mass spectrometry (byboth, full scan and selected ion monitoring) in a free acidfraction from roots of the seedlings inoculated with A. lipoferum.In the roots of the non inoculated seedlings GA3 was found afterhydrolysis of a fraction expected to contain glucosyl conjugates. Endogenous gibberellins (GAs) in several kinds of crown gallcells and cultured cells derived from normal tissue of Nicotianatabacum were systematically analyzed by gas chromatography-selectedion current monitoring (GC-SICM) after chromatographic purifications,and GA1, GA9, GA19 and GA20 were identified. Agrobacterium tumefaciens,a pathogen of crown gall, was confirmed not to produce GAs inits culture. Radioimmunoassays and enzyme-linked immunosorbent assays formethyl esters of gibberellins A1, A3, A4, and A7 were establishedusing an antiserum specific for GA1-Me. The antiserum was characterizedby high titer and specificity for such C19-GAs with 3ß-hydroxylgroup as GA1, GA3, GA4 and GA7. Combination of this antiserumand HPLC enabled us to identify and quantify GA, and GA4 fromthe pollen of Zea mays with a high degree of reliability. Similarly,identification and quantification of GA9 and GA20 were alsomade possible by use of an antiserum specific for GA20-Me. Combineduse of immunoassays and GC/MS enabled us to identify nine GAsfrom the pollen and four from the anthers of Zea mays. The identificationof non-13-hydroxylated GAs, such as GA4 and GA9, in additionto 13-hydroxylated GAs from the pollen and the anthers suggeststhat the early-non-hydroxylation pathway, as well as the early-13-hydrox-ylationpathway, operates in the male reproductive organs of Zea mays,and that the organ-specific biosynthesis and/or localizationof GAs in Zea mays is similar to that in Oryza saliva. Hypocotyl length was found to vary between cucumber plants carryingdifferent genes controlling sex expression. Among lines havingonly unisexual flowers (genotype M/M), the homozygous monoeciousplants (st+/st+) had significantly longer hypocotyls than theirgynoecious counterparts (st/st), heterozygous gynoecious plants(st+/st) being intermediate. Similarly, hypocotyls of plantsof an andromonoecious line (st+/st+ m/m) were significantlylonger than in their hermaphrodite counterparts (st/st m/m).Differences in intemode length were also significant and inthe same direction. Since stem and particularly hypocotyl elongationin cucumber is known to be very sensitive to applied gibberellin,these findings suggest the existence of differences in the effectivelevels of endogenous gibberellins in the different sex types,higher levels being correlated with stronger male tendency.This conclusion is in good agreement with the known effect ofapplied gibberellin on sex expression (enhancement of the maletendency) in cucumber. Application of gibberellin (GA4+GA7) and exposure to ‘summer’conditions (long days and relatively high temperature) inhibitedthe development of pistillate flowers while ‘winter’conditions (short days, lower temperature) had a similar effecton staminate flowers. The effect was in either case specific,that is, limited to flower development. It is concluded thatexogenous and endogenous gibberellins affect not only the initiation,but also the further development, of flowers in the cucumber.  相似文献

Endogenous levels of gibberellins, auxins and cytokinins incrown gall tissues of Catharanthus roseus G. Don. (Vinca roseaL.) of two different tumor types, induced by a nopaline-typeand an octopine-type Ti-plasmid, were examined. The levels of gibberellins were higher in nopaline-type V208cells than in octopine-type V277 cells. GA12 and GA24 were identifiedfrom V208 cells. The level of IAA was 20-fold higher in V208cells than in V277 cells. The level of trans-ribosylzeatin washigher in V277 cells than in V208 cells, whereas trans-zeatinwas present at higher levels in V208 cells than in V277 cells. Our results revealed that the production of gibberellins, aswell as of IAA and cytokinins, in the crown gall tissues ofCatharanthus roseus differed between the octopine- and nopaline-typetumors, even though the crown galls apparently grow as unorganizedaggregates of cells in both cases. 4Present address: Facultad de Ciencias, Pontificia UniversidadJaveriana, Bogota, Colombia. The sympton of dwarfing in the sea campion, Silene vulgaris(Moench) Garcke sub sp. maritima(With.) A. & D. Löveinfected with the anther smut fungus Ustilago violacea (Pers.)Fuckel, a systemic, perennial parasite, has been investigated. Extracts of both healthy and diseased plants contain IAA andGA3, but diseased plants contain less gibberellins than healthyplants. Neither IAA nor gibberellins were detected in significantquantities in the medium when U. violacea was grown in pureculture, but IAN was present. IAN was also found m extractsof diseased plants and it is tentatively suggested that it isformed by the fungus and may accumulate in the host owing tothe inability of the plant to convert IAN to IAA The exogenousapplication of IAN to healthy plants does not produce any diseasesymptoms. The dwarfing symptom of the diseased plant may be due to thelower levels of gibberellins which it contains compared withhealthy plants since the exogenous application of GA3, restoresthe diseased plant to normal growth  相似文献

Gibberellic acid (GA3), kinetin, and indole-3yl-acetic acid(IAA) were applied to roots of Phaseolus vulgaris under twodifferent light intensities and when either young or old leaveswere removed In all cases GA3, promoted stem and lateral growth,especially when light intensity was reduced. Promotion by GA3,of stem growth under reduced light was reduced if IAA and kinetinwere present; promotion of lateral growth under reduced lightwas reduced if IAA was added and eliminated if kinetin or kinetinplus IAA were added to GA3. Removal of young and mature leavesreduced main stem growth; removal of young leaves promoted,and of mature leaves reduced, lateral shoot growth. We suggestthat shoot growth and apical dominance are governed by the balanceof hormones present in elongating internodes. There may be twoways of modifying this balance; firstly by altering light, temperature,or nutrients, or by applying hormones generally to the plant.Secondly, local modifications can be made by removing apicesor young leaves, or applying hormones in lanolin to specificareas. Knowledge of both the general and local conditions maybe necessary for a complete understanding of apical dominance.  相似文献

Exogenous gibberellin A3(GA3) reduced the number of leaf nodesat flowering and time to flowering and increased the stem heightat flowering in three genotypes of spring rape (Brassica napusvar.annua L.). The responses to GA3were similar to those forlong days (LD) and low-temperature treatments, suggesting thatthe effect of photoperiod and the vernalization response areprobably mediated through gibberellins. The response to exogenousGA3was greatest in non-cold-treated plants in short days (SD)suggesting that endogenous GAs are limiting in these conditions.CCC, an inhibitor of gibberellin biosynthesis, caused a smallincrease in the number of leaf nodes at flowering and time toflowering and a small decrease in the stem height at flowering,but unexpectedly, its effect was hardly influenced by the applicationof exogenous GA3. Genotypes that showed the clearest responsesto the treatments with regard to the number of leaf nodes atflowering and time to flowering did not show the clearest responseswith regard to the stem height at flowering; the pattern ofresponses of the number of leaf nodes at flowering and timeto flowering was distinct from that of stem height at flowering.This indicates that flower formation and stem elongation areseparable developmental processes which may be controlled bydifferent endogenous gibberellins, different levels of a specificendogenous gibberellin, or different responses to gibberellin.Copyright 1999 Annals of Botany Company Brassica napus var. annua, gibberellin, photoperiod, spring rape, vernalization.  相似文献

C I 666) infected with barley yellow dwarf virus(BYDV) showed a marked decrease in the mitotic activity of theinfected plants Assay of endogenous gibberellins revealed adecrease in the level of a substance corresponding to gibberellicacid (GA3) in BYDV-infected plants No significant differencein the level of endogenous auxins was observed Application ofgibberellic acid to infected plants reversed the dwarfing effectbut the response was not significantly different from the responseof healthy plants and was found to be due to increased cellelongation.
