棉花咖啡酸-O-甲基转移酶基因的原核表达及蛋白纯化鉴定   总被引:1,自引:0,他引:1  
为获得大量高纯度的GhCOMT2蛋白以便研究其功能和性质,以pMD18-GhCOMT2质粒为模板,PCR扩增GhCOMT2基因的cDNA编码区,构建原核表达载体pET-28a-GhCOMT2,经酶切鉴定并测序后转化到大肠杆菌BL21 (DE3)中进行诱导表达,并采用Western blotting方法鉴定表达产物.结果表明:在大肠杆菌BL21(DE3)菌株中成功表达了与标签蛋白融合的GhCOMT2蛋白,大小约为40.062 kD,浓度为0.62 mg/mL.重组蛋白的最佳诱导条件为:0.2 mmol/L IPTG在16℃诱导12 h.重组蛋白以可溶形式高效表达,用蛋白标签亲和层析柱(His TrapTM HP)获得纯化重组蛋白,Western blotting分析表明其能与His多克隆抗体起特异性反应.  相似文献

[目的]构建K99、GFP基因重组质粒并在大肠杆菌BL21(DE3)感受细胞中表达。[方法]利用PCR扩增技术扩增K99基因和GFP基因,经连接和转化将目的片段克隆到pLA载体上,利用双酶切技术及PCR技术对重组质粒进行鉴定并测序分析,利用Western Blot技术分析蛋白质表达情况。[结果]测序分析结果表明,基因具有正确的阅读框;双酶切和PCR鉴定得到498 bp和720 bp的特异性条带;荧光检测表明重组菌表达了蛋白;Western Blot结果表明重组菌表达为融合蛋白。[结论]成功构建了大肠重组菌,并表达外源融合蛋白。  相似文献

克隆人基质金属蛋白酶-2基因(Mmp2)编码区并构建重组真核表达载体pEYFP-Mmp2,研究其在肝癌细胞中的作用。以肝癌细胞Hep G2的总RNA为模板,通过RT-PCR获得cDNA,并通过PCR获得Mmp2基因编码区。经TA克隆和测序鉴定将无任何突变的Mmp2基因编码区插入到真核表达载体pEYFPN1中,构建p EYFP-Mmp2重组真核表达载体并进行酶切鉴定和测序鉴定。pEYFP-Mmp2稳定性转染肝癌细胞,经G418筛选获得Mmp2稳转单克隆细胞株,并用Western blotting分析鉴定。用实时无标记细胞增殖分析技术(RTCA)分析Mmp2基因对肝癌细胞生长增殖的作用,细胞划痕愈合实验分析Mmp2基因对肝癌细胞迁移的作用。结果证明成功构建重组真核表达载体pEYFP-Mmp2,Western Blotting证实稳转株中高表达外源融合蛋白MMP2-YFP。细胞增殖和迁移结果表明,Mmp2基因可以抑制肝癌细胞增殖但能够促进肝癌细胞迁移。以上研究表明,Mmp2基因能够促进肝癌细胞迁移。  相似文献

旨在通过现代分子生物学技术制备水稻白叶枯病菌FtsZ蛋白。以水稻白叶枯病菌总DNA为模板,采用巢式PCR方法扩增获得水稻白叶枯病菌fts Z基因,构建fts Z基因的表达载体p ET-22b-ftsZ,转化表达宿主E.coli BL21后,经PCR、Nde I/Xho I双酶切及测序鉴定、阳性克隆子经IPTG诱导表达,融合蛋白经镍柱纯化后,通过SDS-PAGE和Western blotting分析鉴定。结果显示,水稻白叶枯病菌ftsZ基因的重组表达载体构建成功,且阳性克隆子在IPTG的诱导下表达了Fts Z-6×His融合蛋白,并通过镍柱纯化获得了电泳纯的Fts Z-6×His融合蛋白。  相似文献

FADD是Fas/FasL系统的一个信号连接蛋白,通过传递凋亡信号,介导细胞凋亡.为了揭示FADD在牛卵泡发育过程中的调控作用,采用RT-PCR从牛卵巢组织中扩增FADD基因,将其cDNA终止密码子删除,采用定向克隆技术连接到带有水母绿色荧光蛋白(AcGFP)报告基因的真核表达载体pAcGFP-N1中,构建融合蛋白重组质粒,经BglⅡ/EcoR Ⅰ酶切、测序鉴定后,用脂质体介导质粒转染CHO-K1细胞,观察有无荧光的表达及用RT-PCR和Western blotting方法检测基因转录、表达情况.结果表明,成功克隆牛FADD基因,通过PCR方法在FADD阅读框两端引入了Bgl Ⅱ和EcoR Ⅰ克隆位点,并于起始位点前加入Kozak序列,成功构建pAcGFP- bFADD融合蛋白真核表达载体,重组质粒转染CHO-K1 24 h后在荧光显微镜下观察到绿色荧光,转染效率可达65%,通过RT-PCR扩增出654 bp的转录产物,并用Western blotting检测到51.4 kD目的蛋白的表迭.  相似文献

金黄色葡萄球菌femB基因与甲氧西林高水平耐药密切相关,可能成为开发抗MRSA药物的新靶位.以金葡菌基因组DNA为模板,PCR扩增femB全长基因,所得片段与pGM-T载体连接并转化感受态大肠杆菌DH5α,阳性克隆以PCR、双酶切及测序鉴定.将鉴定正确的目的片段定向克隆到pGEX-4T-1表达载体中,转化至大肠杆菌BL21后经IPTG诱导表达GST/FemB融合蛋白;采用SDS-PAGE及Western blot对融合蛋白进行鉴定.结果显示,重组质粒在宿主菌中获得了高效表达,融合蛋白相对分子质量为75 kD,该融合蛋白可与抗GST-tag抗体特异结合;表明femB基因的原核表达系统构建成功,为进一步研究其生物学功能奠定了基础.  相似文献

应用重叠延伸PCR方法扩增HIV-1 TAT蛋白转导结构域(PTD)与鼠源神经肽Y(NPY)的融合基因,克隆目的片段并插入酵母表达载体pPICZαA,构建成重组表达质粒pPICZα-PTD-NPY.PCR和酶切鉴定及测序正确后,经限制性内切酶Sac Ⅰ线性化重组表达质粒并通过电转化整合到巴斯德毕赤酵母菌GS115的染色体基因组中.阳性重组酵母菌用含1%甲醇的培养基诱导其分泌表达.经过120 h的诱导,取上清浓缩除盐后进行SDS-PAGE电泳,表明该系统成功表达了PTD-NPY融合蛋白,Western blotting实验证实表达产物具有特异性.获得真核表达的PTD-NPY融合蛋白,为下一步的应用研究提供了物质基础.  相似文献

目的克隆猪白细胞介素18(pIL-18)成熟蛋白基因,并在植物乳杆菌(Lb.plantarum)NC8中进行表达。方法通过RT-PCR方法从猪脾脏细胞中扩增出pIL-18成熟蛋白基因,克隆到T载体pMD18-T后测序;将阳性基因片段克隆至大肠埃希菌-乳酸菌穿梭表达载体pSIP-409构建重组表达载体pSIP-409-IL-18,进行酶切和PCR鉴定;应用电穿孔技术将其转化至Lb.plantarum NC8中,经SppIP诱导表达后,进行SDS-PAGE及Western-blot分析。结果经测序,pIL-18成熟蛋白基因核苷酸长度为579 bp,编码193个核苷酸;酶切和PCR鉴定证明成功构建了重组表达载体pSIP-409-IL-18;SDS-PAGE及Western-blot分析表明重组菌表达了18 kD的融合蛋白,该重组蛋白可以与鼠抗猪IL-18多克隆抗体反应。结论成功克隆了pIL-18成熟蛋白基因,并获得有生物活性的pIL-18重组乳酸菌,为研制开发IL-18重组乳酸菌制剂奠定基础。  相似文献

目的:在大肠杆菌中表达猪丹毒丝菌C43065株表面保护性抗原A(SpaA)N端保护区(SpaA-N),并检测其抗原性.方法:利用PCR方法从猪丹毒丝菌C43065株基因组中扩增出spaA基因片段,构建pMD18-spaA重组质粒并对插入片段进行测序;以pMD18-spaA重组质粒为模板,PCR扩增得到spoA-N基因片段,构建重组表达质粒pGEX-spaA-N,经序列测定证实正确后转化大肠杆菌BL21(DE3),再经IFrG诱导表达GST-SpaA-N融合蛋白并纯化.结果:扩增得到的spaA基因长1 881 bp,编码由626个氨基酸残基构成的多肽;SDS-PAGE和Western印迹检测结果表明,诱导表达获得相对分子质量约64 000的GST-SpaA-N融合蛋白,该融合蛋白能与相应抗体发生特异性反应.结论:获得了在大肠杆菌中可溶性表达的GST-SpaA-N融合蛋白,为进一步研究猪丹毒丝菌免疫保护性抗原奠定了基础.  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
