Twenty-six induced revertants of Scutoid (Sco), a dominant mutation of Drosophila melanogaster, have been characterized genetically. Sco is an unusual mutation, involving two small reciprocal transpositions within the region 35A4 to 35C5 of chromosome arm 2L. One of these transpositions juxtaposes the noc and l(2)br28 loci. We suggested previously that the Sco phenotype results from the "fusion" of noc and l(2)br28. In support of this idea we now show that 23 of 26 revertants of Sco are noc-, indeed the majority are either chromosome aberrations broken between noc and l(2)br28 or deletions of these loci from the mutant chromosome. However, some revertants of Sco are rather more complex, and their properties suggest an interaction between the pu-noc and l(2)br28-l(2)br37 regions of chromosome arm 2L and also demonstrate the genetic complexity of the el-noc region.  相似文献

The Scutoid mutation of Drosophila melanogaster is associated with two small transpositions within region 35 of the polytene chromosome arm 2L. A number of induced revertants of Sco have been recovered and the majority of these are chromosome aberrations. The localisation of at least one breakpoint of these aberrant revertants to the proximal breakpoint of the proximal transposition of Sco, a breakpoint that juxtaposes two loci (noc and sna), normally separated by several loci is evidence that the Scutoid phenotype results from a fusion of noc and sna.  相似文献

AnAdh duplication is described in the medflyCeratitis capitata. Evidence is presented for two separateAdh 1 andAdh 2 structural loci mapping at a distance of 0.49 recombination unit from each other. By deletion mapping theAdh region has been cytologically located near the free end of the left arm of the second chromosome within an area between 2C;3A segments of the polytene chromosome. The genetic analysis of the region aroundAdh has identified seven neighboring genes (Acon 1,Mpi, Est 6,Aox, Xdh, Mdh 2,Lsp I) which identify the linkage group D. The orientation of loci with regard to the centromere sets the origin of the map of the left arm of the second chromosome close to the twoAdh loci.Research supported by National Research Council of Italy, Special Project RAISA, Subproject N.2, Paper N.200. Summary The nopaline catabolism (noc) genes are located in a 14.4 kb region on the pTiC58 plasmid of A. tumefaciens C58. These genes permit the bacterium to grow on nopaline N2-(1,3-dicarboxylpropyl) arginine, a substrate produced in plant tumors initiated by strain C58. The functions of the noc genes include the use of nopaline and L-ornithine as sole carbon and nitrogen sources. Using Tn5 insertional mutants, we have identified and mapped the positions of the genes that are responsible for nopaline catabolism (NopC), ornithine catabolism (OrnC) and nopaline uptake (NopU). A polar relationship was found between these phenotypes, which extended leftward over the noc region to the T-region. The NopC mutants were also deficient in nopaline oxidase, an enzyme that liberates free arginine from nopaline.The noc region also encodes the synthesis of a periplasmic protein, n1 that was induced by nopaline. Tn5 insertional mutations and molecular cloning were used to map the n1 production locus. The recombinant plasmids, pSa4480 and pSa4481, containing the 8.9 kb right-hand end of the noc region, conferred n1 production when introduced into a pTi-free strain of A. tumefaciens. Production of n1 by the strains carrying these plasmids required nopaline induction.We have identified in toto three noc loci: nocB, nocC, and nocA, which confer n1 production, nopaline oxidase production and ornithine catabolism respectively. A model is proposed whereby the noc genes of pTiC58 are contained on a leftward reading operon in the order nocB, nocC, and nocA.  相似文献

Background

Xeroderma Pigmentosum (XP) is a rare skin disorder characterized by skin hypersensitivity to sunlight and abnormal pigmentation. The aim of this study was to investigate the genetic cause of a severe XP phenotype in a consanguineous Pakistani family and in silico characterization of any identified disease-associated mutation.

Results

The XP complementation group was assigned by genotyping of family for known XP loci. Genotyping data mapped the family to complementation group A locus, involving XPA gene. Mutation analysis of the candidate XP gene by DNA sequencing revealed a novel deletion mutation (c.654del A) in exon 5 of XPA gene. The c.654del A, causes frameshift, which pre-maturely terminates protein and result into a truncated product of 222 amino acid (aa) residues instead of 273 (p.Lys218AsnfsX5). In silico tools were applied to study the likelihood of changes in structural motifs and thus interaction of mutated protein with binding partners. In silico analysis of mutant protein sequence, predicted to affect the aa residue which attains coiled coil structure. The coiled coil structure has an important role in key cellular interactions, especially with DNA damage-binding protein 2 (DDB2), which has important role in DDB-mediated nucleotide excision repair (NER) system.

Conclusions

Our findings support the fact of genetic and clinical heterogeneity in XP. The study also predicts the critical role of DDB2 binding region of XPA protein in NER pathway and opens an avenue for further research to study the functional role of the mutated protein domain.  相似文献

A molecular-genetic approach has been taken to identify genes involved in olfactory transduction in Drosophila melanogaster. Two independent lines of research led to the finding that the dominant Scutoid (Sco) mutation causes a diminshed extracellular electroantennogram response to the odorants ethyl acetate (EtAC) and acetone (AC). Sco flies showed about 4- and 2.5-fold reduced responses to EtAC and AC, respectively, compared to Canton-S wild-type and sibling control flies lacking the Sco mutation when electroantennogram recordings were made from the proximal anterior third antennal segment. The responses to five other odors from three different chemical classes were unaltered. The maximum response to either EtAC or AC was decreased with no change in apparent affinity. Responses to short-chain (but not long-chain) acetate esters and ketones were dramatically affected at all antennal locations tested. Only in the proximal quadrants were responses to ethyl acetoacetate also reduced. Most Sco revertants tested had a normal olfactory response; duplications of the region including no-ocelli partially suppress the Sco bristle as well as olfactory phenotypes. Sco adults had an impaired behavioral response to EtAC but not to banana or propionate. There was no effect of the mutation on larval chemosensory behavior or extracellularly recorded adult compound eye and ocellar visual responses. These findings suggest the involvement of Sco in an olfactory pathway in adults which is specific for short-chain acetate esters and ketones. © 1995 John Wiley & Sons, Inc.  相似文献

Genetic Control of Adh Expression in DROSOPHILA MELANOGASTER   总被引:5,自引:4,他引:1  
Natural variants displaying different levels of expression of the gene for alcohol dehydrogenase (Adh) were subjected to genetic mapping experiments. The strains studied carry one of the two common electrophoretic forms of the enzyme. The difference among Adh-fast strains appears to be due to multiple loci with trans-acting effects. Differences among Adh-slow strains are due to modifiers or quantitative sites located very close to the structural gene (less than 0.05 map unit) or part of it. The modifiers detected in the Adhs strains seem to operate only on the structural allele in the cis-position.—A modifier that affects the ratio of ADH levels in larvae and adults was also detected in the Adhs strains. This modifier is also closely linked to Adh and is cis-acting.  相似文献

TE146 is a transposing element (TE) consisting of six polytene chromosome bands that has inserted into the no-ocelli (noc 250) locus. This member of Ising's TE family carries two copies of the white and roughest loci. TE146 is lost from noc with a spontaneous frequency of approximately 1 in 22000 chromosomes. All spontaneous losses are accompanied by the reversion of the noc mutation associated with the TE. The TE is associated with fold-back (FB) sequences. The losses of TE146 retain fold-back homology at noc. Of 26 -ray-induced losses of TE146, 16 are gross deletions, removing loci neighboring noc and ten are not. The non-deleted -ray-induced losses are either noc and rst + or noc + and rst . The white+ genes of TE146 are dosage compensated since w/Y; TE146/+ and w/w; TE146/+ flies are sexually dimorphic for eye color. These w + genes are also suppressed by zeste since z w; TE146/+ flies have zeste-colored eyes.  相似文献

Alcohol dehydrogenase (ADH) isozymes in annual sunflowers (Helianthus annuus) are dimers whose subunits are produced by two genes, Adh 1 and Adh 2 .The codominant F and S alleles of Adh 1 produce the slower-migrating set of three isozymes. The faster-migrating set of three isozymes is controlled by Adh 2 , which also has at least two alleles, F and S. Hybridization experiments indicated that the Adh 2 alleles segregate in expected Mendelian fashion and that Adh 1 and Adh 2 are not linked. A third common 1-locus allele is designated early (E) because when homozygous it results in a blank at the 1FF isozyme position in mature seeds, but in developing seeds produces a normal-appearing band at the 1FF position. Hybridization studies showed that the early alleles segregated normally. Correlation between genotype and presence or absence of isozymes electrophoretically intermediate between those of Adh 1 and Adh 2 suggests that four intergenic isozymes may be formed as a result of dimerization of the four basic subunits. Studies of zymograms of developing seeds suggest that the remaining but inconstant zymogram bands are mature seed isozymes which have altered charges during early morphogenesis and thus are developmental artifacts.  相似文献

Summary Spore control (Sco) mutants were isolated after nitrosoguanidine-induced mutagenesis of germinated spores. They were recognized as colonies showing high proteolytic activity on protein-agar (generally elastin-agar) test plates. Fourteen such mutants were isolated. The Sco mutations were transferred by transformation into an isogenic collection of genetically marked strains. Most of them appeared to be single mutations. Transduction experiments permitted the localisation of six Sco mutants in three loci, all in the argC-metC region. ScoA is located between argC and metC, ScoB is to the right of metC and ScoC is to the left of argC. ScoC and the previously described catA mutation are probably placed in the same gene.Two ScoC strains also appear to carry a second mutation, ScoD, probably localised in the same locus as ScoB or in a locus close to it. Eight other Sco mutations, apparently unlinked to the argC-metC region, were not localised. The results indicate complex regulation of sporulation-associated products such as the proteases, dependent on several genes.  相似文献

A dominant eye color mutation was found associated with a third chromosome inversion broken distally at or near the karmoisin (kar) locus in 87C and proximally within centric heterochromatin. Suppressibility of the mutant phenotype by an extra Y chromosome indicated that this was an example of dominant position-effect variegation. When heterozygous with deficiencies uncovering the kar locus, this inversion chromosome was found to be lethal unless a region in 87EF was also deleted. Extra Y chromosomes rescued inversion/deletion heterozygotes, while removal of the Y chromosome from heterozygous males deficient for the region in 87EF was lethal. Thus, a variegating lethal lies near the breakpoint in 87C, and a wild-type gene that enhances its variegation lies in 87EF. Furthermore, deletion of the region in 87EF was found to strongly suppress white-mottled-4 (wm4) variegation, while deletion of another region in 87BC suppressed less strongly. These results indicate that essential genes on autosomes are sensitive to position effects, and loci that enhance variegation, as defined by deficiency mapping, are very common.  相似文献

A genetic map of the Y chromosome of Drosophila hydei has been constructed from deletion/complementation experiments, with the aid of male sterile mutants of the Y chromosome. A central conclusion of our experiments is that not more than a single complementation group can be detected in each of the lampbrush loop forming sites. Additional complementation groups, functionally independent of lampbrush loops, reside between these loci. Six complementation groups have been defined by several methods of mapping. An additional ten complementation groups are indicated, but their exact definition requires further investigation. The "synthetic sterility" of mutations in these ten loci contributes to the difficulty in unequivocally establishing their individual boundaries. Mapping problems also arise from the instability of certain mutants.  相似文献

The mutagenicity of methyl methanesulfonate (MMS) was studied in a genetically marked two-component heterokaryon of Neurospora crassa. Types of genetic alterations detectable in this system are (I) point mutations in the ad-3A and ad-3B loci; (2) multilocus (chromosome) deletions in the ad-3 region, and (3) recessive lethal mutations in the whole genome. Study of the inactivation kinetics of the heterokaryotic and homokaryotic conidial fractions has made it possible to distinguish between nuclear and cytoplasmic inactivation.Forward mutations in the ad-3 region induced by MMS in the heterokaryotic fraction of conidia were obtained by a direct method with the following results: (I) The overall ad-3 forward mutation frequency increases in proportion to the 1.91 power of the concentration of MMS. (2) The forward mutation frequency of point mutations at the ad-3A and ad-3B loci increases in proportion to the 1.68 power of the concentration. (3) The forward mutation frequency of chromosome deletions in the ad-3 region increases more than exponentially with increasing concentrations of MMS. (4) After treatment for 300 min with 20 mM MMS, 15.5% of the ad-3 mutations are multilocus deletions. Tests for genotype and allelic complementation of the point mutations showed that (I) the ratio between ad-3B and ad-3A mutants was 1.75, (2) 52.1% of the ad-3B mutants showed allelic complementation, with 39.2% non-polarized and 12.9% polarized complementation patterns and 47.9% noncomplementing mutants, and (3) both the ratio between point mutations in the ad-3A and ad-3B loci and the spectrum of complementation patterns among the ad-3B mutants were independent of MMS concentration.  相似文献

The Minute phenotype results from mutations at?>50 loci scattered throughout the genome of Drosophila. Common traits of the Minute phenotype are short and thin bristles, slow development, and recessive lethality. Here, we report a novel P-element induced Minute mutation, P{lacW}M(3)66D 1 , that maps to region 66D on chromosome 3L. Flies heterozygous for P{lacW}M(3)66D 1 have a strong Minute phenotype. Molecular characterisation of the chromosomal region revealed three previously undescribed Drosophila genes clustered within a 5-kb genomic fragment. Two of the genes have significant sequence homology to genes for the mammalian ribosomal proteins L14 and RD, respectively, and share a joint 240-bp promoter region harbouring the P-element insert. Quantitative Northern blot analyses showed the mutation to affect RPL14 mRNA levels only. Interestingly, the reduction in abundance of RPL14 mRNA is not constitutive, indicating that the promoter function abolished by the inserted P-element is utilised with different efficiencies in different developmental situations. Remobilisation of the P element produced wild-type flies with normal levels of RPL14 mRNA, demonstrating that the mutant phenotype is caused by the insertion. P{lacW}M(3)66D 1 joins a growing list of Minute mutations associated with ribosomal protein-haploinsufficiency.  相似文献

Sex determination in Caenorhabditis elegans is controlled by the X chromosome: autosome ratio, i.e. 2A;XX animals are hermaphrodite, and 2A;XO animals are male. A procedure for isolating 2A;XO animals that are transformed into hermaphrodites has been developed. Nine mutations causing this transformation have been obtained: eight are recessive, and all of these fall into a new autosomal complementation group, her-1 V. The remaining mutation (her-2) is dominant and has a genetic map location similar to that of tra-1 III. Recessive mutations of tra-1 cause the reverse transformation, transforming 2A;XX animals into males. Therefore, the her-2 mutation may result in constitutive expression of tra-1. Mutations in her-1 are without effect on XX animals, but the her-2 mutation prevents sperm production in both XX and XO animals, in addition to its effect on the sexual phenotype of XO animals. The epistatic relationships between tra and her genes are used to deduce a model for the action of these genes in controlling sex determination.  相似文献

Copper has an important role in the life cycle of many streptomycetes, stimulating the developmental switch between vegetative mycelium and aerial hyphae concomitant with the production of antibiotics. In streptomycetes, a gene encoding for a putative Sco-like protein has been identified and is part of an operon that contains two other genes predicted to handle cellular copper. We report on the Sco-like protein from Streptomyces lividans (ScoSl) and present a series of experiments that firmly establish a role for ScoSl as a copper metallochaperone as opposed to a role as a thiol-disulphide reductase that has been assigned to other bacterial Sco proteins. Under low copper concentrations, a Δsco mutant in S. lividans displays two phenotypes; the development switch between vegetative mycelium and aerial hyphae stalls and cytochrome c oxidase (CcO) activity is significantly decreased. At elevated copper levels, the development and CcO activity in the Δsco mutant are restored to wild-type levels and are thus independent of ScoSl. A CcO knockout reveals that morphological development is independent of CcO activity leading us to suggest that ScoSl has at least two targets in S. lividans. We establish that one ScoSl target is the dinuclear CuA domain of CcO and it is the cupric form of ScoSl that is functionally active. The mechanism of cupric ion capture by ScoSl has been investigated, and an important role for a conserved His residue is identified.  相似文献

siren1 and siren2 are novel alcohol dehydrogenase (Adh)-derived chimeric genes in the Drosophila bipectinata complex. D. ananassae, however, harbors a single homolog of these genes. Like other Adh-derived chimeric genes, siren evolved adaptively shortly after it was formed. These changes likely shifted the catalytic activity of siren.  相似文献
