It has been demonstrated that the differences observed in the aetiological structure of the individual foci of leptospirosis can be explained not only by the affinity of leptospiral serogroups to certain animal species, but also by different mechanisms of transmission of the causative agent of leptospiral infection which can be transferred both by sexual and alimentary routes (in water). It has been demonstrated that mostly one serotype of leptospires predominates in natural foci of leptospirosis, but several in anthropurgic ones. In the author's opinion, leptospiral infection in natural foci is mainly spread by the sexual route through the background species of animals--carriers of leptospirosis, and by the alimentary route in the anthropurgic foci. It is presumed that leptospires of the serogroups Javanica, Australis, Icterohaemorrhagiae, transmitted by the shrew-mice, hedgehogs and rats by the sexual route, are by their origin "ancient" serogroups of leptospires while the serogroups of leptospires isolated from domestic animals, showing predominantly the alimentary route of transmission of infection in the focus, are representatives of the "younger" forms of the evolutional development of leptospires.  相似文献

Monitoring studies during 1996-2001 have shown a natural foci of tick-borne borreliosis with high abundance of Ixodes ricinus and high infection rate with an agent: 19.5-38.4 tick individuals per 1 km of route, infection rate 18.0 +/- 1.7-22.5 +/- 1.5%. A risk to be infected with the Lyme disease in the ricinus focus of the tick-borne borreliosis during the epizootic season did not vary much during the period of study, 3.5-8.2 in different year, mean 6.0 infected ticks per 1 km of route.  相似文献

The cycle of Echinococcus multilocularus in natural and synanthropic hosts was investigated during 10 yr in an endemic focus of alveolar hydatid disease in the Massif Central of France. The natural cycle, involving red foxes, Vulpes vulpes, and voles, Arvicola terrestris, existed immediately surrounding a village in which human cases of alveolar hydatid disease occurred. Both foxes and free-ranging dogs could serve as the source of infection for the human population.  相似文献

The role of natural hybridization in the evolutionary history of numerous species is well recognized. The impact of introgressive hybridization and hybrid speciation has been documented especially in plant and animal assemblages. However, there remain certain areas of investigation for which natural hybridization and its consequences remain under-studied and under-appreciated. One such area involves the evolution of organisms that positively or negatively affect human populations. In this review, I highlight exemplars of how natural hybridization has contributed to the evolution of (i) domesticated plants and animals; (ii) pests; (iii) human disease vectors; and (iv) human pathogens. I focus on the effects from genetic exchange that may lead to the acquisition of novel phenotypes and thus increase the beneficial or detrimental (to human populations) aspects of the various taxa.  相似文献

The infecting ability of the fleas Citellophilus tesquorum altaicus loff, 1936, the main plague vectors in the Tuva natural focus, was experimentally studied in different periods of the epizootic season. Seasonal dynamics in the efficiency of infecting the long-tailed Siberian souslik with the plague causative agent through flea bites was noticed. Seasonal differences in infectivity of the "blocked" flea bites are revealed. An increase of infected experimental animals with a generalization of infection process in the period of epizooty activation in the natural focus was observed. A resistance of the long-tailed Siberian souslik to the plague agent infection through flea bites in the spring season was registered.  相似文献

During 60 years passed after the creation of the conception on natural focus of human infections by the academician E. N. Pavlovski? our knowledge on a role of blood-sucking insects and ticks in this phenomenon have been significantly enlarged. It has been recovered, that these arthropods serve not only as vectors of infection agents, but also as natural reservoirs and amplificators of these agents. In the process of the infection agent circulation in the natural focus there are several additional pathways of circulations without a participation of vertebrates. These pathways are as follows: the dispersion of infection agent in cases of simultaneous feeding of infected and "clean" ticks, and also the sexual, transovarial and transphase transmissions of pathogens. These mechanism of circulation are most often occur in the ixodid ticks, which can play the main role in supporting the infection agent in the focus.  相似文献

The Plasmodium falciparum parasite is an obligate intracellular pathogen whose invasion and remodelling of the human erythrocyte results in the clinical manifestations of malarial disease. The functional analysis of erythrocyte determinants of invasion and growth is a relatively unexplored frontier in malaria research, encompassing studies of natural variation of the erythrocyte, as well as genomic, biochemical and chemical biological and transgenic approaches. These studies have allowed the functional analysis of the erythrocyte in vitro, resulting in the discovery of critical erythrocyte determinants of Plasmodium infection. Here, we will focus on the varied approaches used for the study of the erythrocyte in Plasmodium infection, with a particular emphasis on erythrocyte invasion.  相似文献

Q fever occurs in all geographical landscape zones and in most administrative regions of the Ukrainian SSR. The disease was diagnosed in those areas where no such cases had ever been registered. In the Kharkov region the examination of 425 fever patients with unclear diagnosis resulted in diagnosing 5 cases of Q fever. The proportion of seropositive persons among different professional groups of the population in the eastern part of the republic was determined (6.9% among workers of meat-packing factories, 8.7% among workers of fur- and wool-treating establishments, 5.2% among stock breeders). At the farms of the Kharkov, Poltava and Sumy regions the spread of infection among large cattle was 2.3% and among small cattle 5.6%. Rickettsial carriership revealed in hard ticks collected in the Kharkov region indicated the possibility of the existence of the natural focus of Q fever in this zone and increased the available information on the presence of the territories with the natural foci of this infection in the Ukraine.  相似文献

In this paper, Eiji Konishi reviews general features of naturally occurring (natural) antibodies that react with protozoan parasites. Several functions of natural antibodies have been identified in relation to their multireactivity, but reports on protozoan infection have dealt mainly with the role of natural antibodies in the innate immunity of the host, These antibodies lyse cells in the presence of complement and have opsonizing activity, in vitro. Studies of their origin have shown the possibilities of (1) continuous polyclonal stimulation by gastrointestinal bacteria, and (2) there being multireactive antibodies secreted by CD5(+) B cells. The protective functions of natural antibodies are important in the interpretation of the host range, the mode of infection, and the course of the disease of certain protozoan parasites.  相似文献

Sigurdsson's three criterions for a slow viral infection are quoted and discussed and the background of his work briefly described. A fourth criterion for a slow viral infection, is suggested, infection of the hosts lymphoid tissues, which is a common feature of slow infections that have been observed. The general properties of the maedi-visna virus, the diseases and the immune response it causes are discussed. Sheep and goats are susceptible to natural maedi-visna infection. In the goat an identical retrovirus causes arthritis. Arthritis has not been found in maedi-visna infected sheep. Two subacute spongiform encephalopathies of animals are shortly reviewed, scrapie in sheep and goats and transmissible mink encephalopathy (TME). General properties of the very unusual scrapie agent are mentioned briefly. The third type of a slow viral infection is mentioned, Aleutian mink disease, an immunopathological disorder of certain minks caused by a selective infection of lymphoid cells.  相似文献

Rift Valley fever virus (RVFV) is a formidable pathogen that causes severe disease and abortion in a variety of livestock species and a range of disease in humans that includes hemorrhagic fever, fulminant hepatitis, encephalitis and blindness. The natural transmission cycle involves mosquito vectors, but exposure can also occur through contact with infected fluids and tissues. The lack of approved antiviral therapies and vaccines for human use underlies the importance of small animal models for proof-of-concept efficacy studies. Several mouse and rat models of RVFV infection have been well characterized and provide useful systems for the study of certain aspects of pathogenesis, as well as antiviral drug and vaccine development. However, certain host-directed therapeutics may not act on mouse or rat pathways. Here, we describe the natural history of disease in golden Syrian hamsters challenged subcutaneously with the pathogenic ZH501 strain of RVFV. Peracute disease resulted in rapid lethality within 2 to 3 days of RVFV challenge. High titer viremia and substantial viral loads were observed in most tissues examined; however, histopathology and immunostaining for RVFV antigen were largely restricted to the liver. Acute hepatocellular necrosis associated with a strong presence of viral antigen in the hepatocytes indicates that fulminant hepatitis is the likely cause of mortality. Further studies to assess the susceptibility and disease progression following respiratory route exposure are warranted. The use of the hamsters to model RVFV infection is suitable for early stage antiviral drug and vaccine development studies.  相似文献

The generation of protective immunity by various stages in the life-cycle of Strongyloides ratti and the phases against which resistance is directed has been examined in murine strongyloidiasis. Mice were exposed to natural, complete infections, were treated with thiabendazole (which largely resembles the natural infection), were treated with cambendazole (which restricts infection to the larval stage), or infected directly by oral transfer of adult worms. Mice that were infected with infective larvae alone did not become resistant to infective larvae or the complete infection but were resistant to adult worms implanted directly into the gut. Mice exposed to adult worms alone were resistant to natural infections and adults worms implanted directly but were not resistant to infective larvae. On the other hand, mice that had received prior natural infections showed evidence of resistance to infective larvae, adult worms, and natural, complete infections. It is concluded that there is immunological cross-reactivity between infective larvae and adult worms but that under certain circumstances the infective larvae are able to evade the host's protective immune response.  相似文献

Recent studies revealed an amazing phenotypic heterogeneity between genetically identical individual cells within populations of microbial pathogens. During the course of an infection, subpopulations occur, which differ in certain virulence-relevant factors, stress adaptation functions or physiological and metabolic abilities. The mechanisms driving this heterogeneity are divergent reactions of the pathogens to differences in host tissue microenvironments. In addition, certain genetic regulatory circuits with positive feedback loops and stochastic differences in gene expression can generate endogenous fluctuations in regulatory components leading to bistable expression of virulence-associated functions. Here, we focus on the occurrence of phenotypic heterogeneity in populations of well-studied examples of pathogens, which enables cooperative, social behavior where a subpopulation of producers shares fitness- and/or virulence-relevant goods and traits with non-producers. We further highlight that this strategy allows preadaptation of a subgroup of cells to recurrent and thus predictable changes of the environment that they encounter during the different stages of the infection. The diversity within bacterial communities has a significant influence on the survival of the pathogens within their hosts and the progression of the disease.  相似文献

Before maternally inherited bacterial symbionts like Wolbachia, which cause cytoplasmic incompatibility (CI; reduced hatch rate) when infected males mate with uninfected females, can be used in a program to control vector-borne diseases it is essential to understand their dynamics of infection in natural arthropod vector populations. Our study had four goals: (1) quantify the number of Wolbachia strains circulating in the California Culex pipiens species complex, (2) investigate Wolbachia infection frequencies and distribution in natural California populations, (3) estimate the parameters that govern Wolbachia spread among Cx. pipiens under laboratory and field conditions, and (4) use these values to estimate equilibrium levels and compare predicted infection prevalence levels to those observed in nature. Strain-specific PCR, wsp gene sequencing, and crossing experiments indicated that a single Wolbachia strain infects Californian Cx. pipiens. Infection frequency was near or at fixation in all populations sampled for 2 years along a >1000-km north-south transect. The combined statewide infection frequency was 99.4%. Incompatible crosses were 100% sterile under laboratory and field conditions. Sterility decreased negligibly with male age in the laboratory. Infection had no significant effect on female fecundity under laboratory or field conditions. Vertical transmission was >99% in the laboratory and approximately 98.6% in the field. Using field data, models predicted that Wolbachia will spread to fixation if infection exceeds an unstable equilibrium point above 1.4%. Our estimates accurately predicted infection frequencies in natural populations. If certain technical hurdles can be overcome, our data indicate that Wolbachia can invade vector populations as part of an applied transgenic strategy for vector-borne disease reduction.  相似文献

SUMMARY

Tuberculosis (TB) is a leading cause of death worldwide despite the availability of effective chemotherapy for over 60 years. Although Mycobacterium bovis bacillus Calmette-Guérin (BCG) vaccination protects against active TB disease in some populations, its efficacy is suboptimal. Development of an effective TB vaccine is a top global priority that has been hampered by an incomplete understanding of protective immunity to TB. Thus far, preventing TB disease, rather than infection, has been the primary target for vaccine development. Several areas of research highlight the importance of including preinfection vaccines in the development pipeline. First, epidemiology and mathematical modeling studies indicate that a preinfection vaccine would have a high population-level impact for control of TB disease. Second, immunology studies support the rationale for targeting prevention of infection, with evidence that host responses may be more effective during acute infection than during chronic infection. Third, natural history studies indicate that resistance to TB infection occurs in a small percentage of the population. Fourth, case-control studies of BCG indicate that it may provide protection from infection. Fifth, prevention-of-infection trials would have smaller sample sizes and a shorter duration than disease prevention trials and would enable opportunities to search for correlates of immunity as well as serve as a criterion for selecting a vaccine product for testing in a larger TB disease prevention trial. Together, these points support expanding the focus of TB vaccine development efforts to include prevention of infection as a primary goal along with vaccines or other interventions that reduce the rate of transmission and reactivation.  相似文献

Plague (Yersinia pestis infection) is a highly virulent rodent disease that persists in many natural ecosystems. The black rat (Rattus rattus) is the main host involved in the plague focus of the central highlands of Madagascar. Black rat populations from this area are highly resistant to plague, whereas those from areas in which the disease is absent (low altitude zones of Madagascar) are susceptible. Various lines of evidence suggest a role for the Major Histocompatibility Complex (MHC) in plague resistance. We therefore used the MHC region as a candidate for detecting signatures of plague-mediated selection in Malagasy black rats, by comparing population genetic structures for five MHC-linked microsatellites and neutral markers in two sampling designs. We first compared four pairs of populations, each pair including one population from the plague focus and one from the disease-free zone. Plague-mediated selection was expected to result in greater genetic differentiation between the two zones than expected under neutrality and this was observed for one MHC-class I-linked locus (D20Img2). For this marker as well as for four other MHC-linked loci, a geographic pattern of genetic structure was found at local scale within the plague focus. This pattern would be expected if plague selection pressures were spatially variable. Finally, another MHC-class I-linked locus (D20Rat21) showed evidences of balancing selection, but it seems more likely that this selection would be related to unknown pathogens more widely distributed in Madagascar than plague.  相似文献

On the basis of experimental and clinical study of infections with the aerosol mechanism of infection there was revealed a relationship between the fractional-dispersive composition of the microbial aerosol, the porta of infection and the clinico-pathogenetic peculiarities of the course of the disease. On the example of tularemia, plague and other nosological forms it was demonstrated that coarse-dispersive aerosol caused development of oculo-bubonic and anginous-bubonic form of the disease, whereas the high-dispersive aerosol led to the appearance of primary pneumonia. In experimental aerosol infection with the causative agents in which the infection under natural conditions is not air-borne (botulism, American horse encephalo-myelitis, etc.) specific disease as a rule develops without any primary affection of the respiratory organs.  相似文献
