The ribsome-deficient plastids of the albino leaves of the barley mutant albostrians divide at about the same rate as normal plastids and contain similar levels of plastids DNA to the normal plastids. Double-ring structures were observed around the neck of constricting dumbbell-shaped, ribosome-deficient plastids in the basal intercalary meristem of albino leaves. In the distal region of albino leaves the ribosome-deficient plastids contain a rudimentary thylakoid system often closely associated with DNA nucleoids. It is suggested that nuclear coded proteins synthesized within the cytoplasm are responsible for the formation of the double-ring structures and the rudimentary thylakoids of albino plastids.  相似文献

Summary In monogenic, recessive chloroplast mutants of maize which contain chlorophylls, and lycopene or -carotene but no normal carotenoids, great variability in the size of plastids was associated with a number of ultrastructural abnormalities. In the mutant accumulating lycopene some plastids contain dense bundles of lamellae, whereas the chloroplasts of the -carotene mutant show poor thylakoid development. Neither of the mutants was able to form normal grana.A comparison of chlorophyll/carotenoid ratios in different chloroplast fractions of normal and mutant leaves showed that plastids of small size and delicate structure contain relatively less chlorophyll than fully differentiated chloroplasts.  相似文献

Zusammenfassung In den Palisadenzellen von Keim- und Rosettenblättern einer rezessiven xantha-Mutante von Arabidopsis thaliana fehlt den Plastiden die für normale, grüne Chloroplasten typische Schichtenstruktur. Licht- und elektronenmikroskopische Untersuchungen ergaben, daß sich dieser genabhängige Differenzierungsdefekt während der Plastidenentwicklung beim Übergang von den Tubuli des Prolamellarkörpers zu Thylakoiden manifestiert. Es entstehen aus dem persistierenden Prolamellarkörper nur einzelne abnorme Thylakoide, die zuweilen napfoder taschenartig verformt oder zu Magnograna vereinigt sein können. Besonders charakteristisch für die Mutante aber ist, daß solchen Thylakoiden häufig parallel laufende Züge einzelner Tubuli oder Fibrillen beidseitig dicht aufliegen. Auf Grund Dieser Anordnung wird die Möglichkeit eines Thylakoidwachstums durch laminalen Einbau von Tubuli erörtert.In den Blättern der Mutante wurden nur Carotinoide, jedoch keine Chlorophylle festgestellt. Zugleich finden sich in den entwicklungsgehemmten Plastiden, neben Phytoferritin-Partikeln, zahlreiche osmiophile Globuli sowie Matrixeinschlüsse unbekannter Natur. Daher wird die Frage diskutiert, ob die beobachteten Störungen der Thylakoid-Differenzierung in der Mutante auf den Chlorophyllmangel oder auf eine blockierte Strukturproteid-Synthese zurückgehen.Alle übrigen Zellorganelle sind in der Mutante ebenso gestaltet wie in der grünen Normalform. Besonders wird auf das Vorkommen von dense bodies hingewiesen und ihre Beziehung zu Sphärosomen erörtert.
Disturbed thylakoid-formation in chloroplasts of a xantha-mutant of Arabidopsis thaliana (L.) Heynh.
Summary A monohybrid recessive xantha-mutant of Arabidopsis thaliana was grown aseptically on mineral agar +2% sucrose up to maturity. The cotyledons and rosette leaves were non-green during all developmental stages, their yellow colour bleaching with age.Palisade cells of various leaves of mutant plants were investigated light- and electron microscopically. All of the organelles of these cells, except the plastids, had a normal appearance. Special attention was paid to the existence of dense bodies, and their relationship to sphaerosomes and lysosomes was outlined.In the mutant plastids the typical thylakoid differentiation present in the chloroplasts of the green Arabidopsis plant is entirely lacking. Within the palisade cells the plastids are normal in number and size. Under the light microscope they appear as hyaline vesicles with usually two primary grana. Their outline is characterized by numerous invaginations and protrusions; however, it is still uncertain to what extent this amoeboidy is only an artifact.The specific gene dependent block in the plastid development is apparently effective during the conversion of tubules of the prolamellar body to thylakoids. From the persistent prolamellar bodies in the mutant only single abnormal thylakoids originate. These often have a deformed cup- or ventricle-like shape and occasionally are piled up as magnograna. In a characteristic manner tubules and fibrils are closely associated in parallel rows with the outsides of the thylakoids. The fibrils having a mean diameter of 75 Å are connected with the tubules which are derived from the prolamellar body. From this striking aggregation the possibility of a laminal fusion of tubules as a process of normal thylakoid growth is deduced.By pigment analysis of mutant leaves no chlorophyll but only carotenoids were detected. However, many osmiophilic globules as well as large bodies of unknown composition (proteinaceous?) appear in the plastid matrix. Therefore the question is discussed whether the disturbances of the thylakoid differentiation is due primarily to a blocked chlorophyll formation or to an inhibition of the synthesis of some structural membrane proteids.Phytoferritin particles which up to now have not been observed either in normal or in mutant plastids of Arabidopsis were frequently found in the plastid matrix of this xantha-mutant.

A volume of tissue of lettuce (Lactuca sativa L.) plants extending 2 mm basipetally from the apical meristem and including leaf primordia and young expanding leaves was surveyed using electron-probe microanalysis (EPMA) on both frozen-hydrated and freeze-dried samples. This analysis was carried out either 2 or 5 d following NaCl salinization of the medium from the 10 mol · m–-3 control level up to 80 mol · m–-3. The objective was the investigation of possible changes in the nutritional status of the apical meristem that might account for some aspects of salt-induced growth inhibition. Sodium and chloride increased significantly in tissues basal to the apical meristem, while both phosphorus and potassium decreased in the same region. These changes were evident in specimens collected just 2 d after the commencement of salinization (20 h after completion of the salinization) and were not exacerbated by an additional 3 d of treatment; they were present in tissue as close as 100 m to the meristem and extending down to 500 m. The apical 10–50 m were relatively protected from both the increase in sodium and chloride and the decrease in phosphorus and potassium that occurred in more basal regions. Young leaves (up to 1.5 mm in length) appear to control their own mineral nutrient levels when challenged by salinization of the medium, presumably because of altered growth. A decrease in the concentration of total Ca as a result of salinization was significant in cells 500 m basal to the meristem, but was evident as a tendency in the data even within the first 50 m. Using an improved automatic method for the analysis of calcium by EPMA, it was found that total Ca was reduced by salinization, especially in basal regions (500 m below the apex) and also in young leaves (1–1.5 mm in length). We suggest that the nutrition of the shoot apical meristem may be disturbed soon after salinization and that the shoot meristem might be the source of a signal to expanding leaves, as well as exerting its own direct influence over leaf emergence.Abbreviation EPMA electron-probe microanalysis This work was supported by U.S. Department of Agriculture grant 87-CRCR-1-2462.  相似文献

Antlers are periodically cast and re-grown cranial appendages of deer. Both endochondral and intramembranous ossification are involved in the formation of antler bone. After velvet shedding, antlers are completely bony structures which are referred to as hard antlers. Growing antlers accumulate substances with an affinity to mineralized tissues. Among these substances are lead and fluoride. Due to the seasonally limited life span of antlers, the concentrations of lead and fluoride in hard antlers reflect uptake by the forming bone during a defined, species-specific period of some months. Antlers can thus be viewed as naturally standardized environmental samples that are well suited as biomonitors of environmental pollution by bone-seeking contaminants. Because hard antlers are collected by hunters as trophies and kept in private or public collections, material for study can be obtained rather easily. So far, lead concentrations in hard antlers have been reported only from Europe, whereas data for fluoride are available from both Europe and North America. Some studies compared contaminant concentrations in antlers from different regions, while others analyzed time-trends in contaminant levels in antlers from a single region. Using the latter approach, a pronounced drop of lead concentrations during recent decades has been reported for antlers from various European countries. This indicates a marked decline of environmental lead levels that can be attributed mainly to the phase-out of leaded gasoline and, in addition, to a reduction of lead emissions also from stationary sources. In Germany, a corresponding drop has also been recorded for fluoride concentrations in antlers, which is attributed to a decline of fluoride emissions from stationary sources due to improved emission-control measures. There is some evidence that exposure to higher levels of fluoride may lead to an impaired antler mineralization. Using antlers as biomonitors has been shown to be an efficient method for assessing environmental pollution by lead and fluoride at low cost. Further studies using this now well established approach are therefore encouraged.This revised version was published online in May 2005. The following errors were eliminated:1. Myelin from adult rat brains was separated on a discontinuous sucrose gradient into three subfractions. Analysis of light, heavy and membrane fraction lipid classes was performed by HPTLC and densitometry while fatty acid composition was determinated by GLC. The more interesting results observed are: i) the membrane fraction resembles in its lipid and fatty acid composition other cell membranes (particulary oligodentrocytes); ii) light and heavy myelin are quite similar between them but the former has a higher content of sphingomyelin, a lower hydroxy/nohhydroxy cerebrosides ratio and a lower content of monoenoic fatty acids than the heavy subfraction. The results obtained could explain the different structures observed in each myelin subfraction since fatty acid composition, hydroxy fatty acids, sphingomyelin and cholesterol play a key role in the stability and structure of membranes.  相似文献

For the structural analysis of the carbohydrate chains ofN-,O-glycoproteins a straightforward strategy was developed based on the cleavage of theN-linked chains with immobilized peptide-N 4-(N-acetyl--glucosaminyl) asparagine amidase-F (PN-Gase-F) fromFlavobacterium meningosepticum, followed by alkaline borohydride treatment of the remainingO-glycoprotein material. This methodology was applied to the isolation of the Asn- and Ser-linked carbohydrate chains of human chorionic gonadotrophin. The structures of the isolated oligosaccharides were verified by 500-MHz1H-NMR spectroscopy. The Asn-linked sugar chains were shown to be: NeuAc2-3Gal1-4GlcNAc1-2Man1-6[NeuAc2-3Gal1-4GlcNAc1-2Man1-3]Man 1-4GlcNAc1-4[Fuc1-6]0-1GlcNAc and Man1-6[NeuAc2-3Gal1-4GlcNAc1-2Man 1-3]Man1-4GlcNAc1-4GlcNAc. Also some minor constituents occurred. Aeonium domesticum cv. variegatum is a mesochimera of the constitution green/white/green with normal proplastids and chloroplasts in the unaffected tissues and ribosome-deficient colourless mutant plastids in the white leaf tissues. All the different plastid types contain succulent protein crystalloids (SPC). For more detailed characterization, the SPC elements were freed from the plastids and purified by gel filtration. Electron microscopy of different fractions revealed five levels of structural organization. Beginning with the most complex state, the levels are designated as succulent protein (SP) organizational state V (hexagonally arranged and closely packed tubules in the stroma of intact plastids) to I (globular protomers of 5 nm diameter as the basic structure of SPCs). Highly purified SP-fractions were shown by means of sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) to consist of two or three proteins of Mr 56 kdalton, 58 kdalton and 60 kdalton, depending on the buffer medium used for SP isolation and the duration of storage of leaves in the frozen state. In the urea/SDS-PAGE system, these proteins show similar mobilities to - and -tubulin, but no immunoreaction against antitubulin. The proteolytic cleavage pattern of tubulin subunits and SP proteins are different. Their locations on two-dimensional isoelectric focusing-SDS gels show some overlappings because of microheterogeneities in both proteins in the pH gradient from pH 4.5 to 6.5. Summary Ultrastructure of the constricting neck of dividing proplastids and young chloroplasts in the first leaves ofAvena sativa was examined by electron microscopy. An electron-dense, double ring structure (plastid-dividing ring doublet; PD ring doublet) with a width of 15–40 nm was revealed around the narrow neck of the constricted and dividing plastids by serial section technique. The inner and outer ring of the doublet coated the inside (stromal side) of the inner envelope membrane and the outside (cytoplasmic side) of the outer envelope membrane, respectively. However, electron-dense materials were not observed within the lumen between the outer and inner envelope membranes.Although the PD ring doublet was commonly observed in the constricted plastids with a 70–140 nm wide neck, they could be scarcely observed in the constricted plastids with a 160 or more nm wide neck. The components of the PD ring were assumed not to be concentrated enough to identify by electron microscopy in the early stage of constriction and the PD ring may be formed and recognized at the final stage.The significance of the formation of the PD ring and its role in plastokinesis (plastid kinesis) were discussed.  相似文献

Summary Several TEM and SEM techniques were applied to examine developing structures in valves of the centric diatomThalassiosira eccentrica (Ehrenb.) Cleve after cytokinesis. It was possible to confirm that in each stage of the silicification process there is a distinction between a growing zone with a loose assemblage of silica spheres and a compacting zone in an older phase of development. The spherical structure of the silica in the growing zone results from the addition of silica by small cytoplasmic vesicles of about 300 to 400 Å in diameter. The vesicle membrane fuses with the silicalemma and the vesicle content is released into the silica-deposition vesicle. The origin of these vesicles, named STV, is still unknown.  相似文献

Summary Electron-microscopic studies of plastids in cortical cells of poplar (Populus euramericana cv. gelrica) were carried out to examine whether any structural changes were initiated after breaking of dormancy in midwinter under non-growing conditions. After the breaking of dormancy, ultrastructural changes became evident and the profiles of plastids became heterogeneous. Organelles resembling the plastid initials proposed by Mühlenthaler and Frey-Wyssling in 1965 were frequently observed concomitant with changes in the plastid envelope. The formation of plastid initials appeared to be initiated by the formation of septa in pre-existing plastids. After this stage, narrow connections appeared between the initials and the parent plastids. Approximately 50 days after the breaking of dormancy in late March, further heterogeneity in the profiles of plastids was observed. At this stage, young plastids (plastids without starch granules) were frequently observed and the formation of plastid initials was hardly ever observed. These observations suggest that the plastid initials may be present for only a limited period in the cortical cells of the poplar and may be the precursors of the proplastids. Similar ultrastructural profiles were found in cortical cells of mulberry and in leaf buds of apple trees, suggesting that such changes in the ultrastructure of plastids are a general feature of perennials.  相似文献

Molecular and Functional Studies of the Gamma Subunit of the Sodium Pump   总被引:6,自引:0,他引:6  
This article reviews our studies of the subunit of the sodium pump. is a member of the FXYD family of small, single transmembrane proteins and is expressed predominantly in the kidney tubule. There are two major variants of which function similarly to bring about two distinct effects, one on KATP and the other, on K K, the affinity of the pump for K + acting as a competitor of cytoplasmic Na+. In this way, is believed to provide a self-regulatory mechanism for maintaining the steady-state activity of the pump in the kidney. Our studies also suggest that K+ antagonism of cytoplasmic Na+ activation of the pump is relevant not only to the presence of in the kidney, but probably some hitherto undefined factor(s) in other tissues, most notably heart. The interesting possibility that not only but other members of the FXYD family regulate ion transport in a tissue-specific manner is discussed.  相似文献

In the present study we investigated the TNF- induced signal transduction mechanism in human neutrophil. Exogenously added TNF- affects both PKC activity and its translocation from cytosol to the membrane. Endogenous protein phosphorylation pattern is inhibited in TNF- induced neutrophil in Ca-dependent and Ca-independent manner, including a major 47 and 66 kDa cytosolic proteins, which may be implicated in superoxide anion generation. However TNF- dose dependently enhances the expression of -PKC isotype but not the -PKC. Morphology and cell cytotoxicity are studied in TNF- treated neutrophil to understand the TNF- induced cell death or apoptosis and these experiment is further confirmed by DNA fragmentation analysis. These results clearly demonstrate that TNF- induces cellular death of human neutrophil at least in part by enhanced expression of Ca-independent -PKC. These observations provide an insight towards understanding the function of -PKC in apoptotic pathway.  相似文献

Summary Chloroplast differentiation in relation to increasing leaf age has been investigated in maize plants exposed to continuous illumination. In the young leaves the proplastids differentiate into chloroplasts containing well organized grana as well as prolamellar bodies. In the older leaves, while plastids differentiate, the prolamellar bodies are no longer detectable. Chloroplast ability to build up prolamellar bodies does not seems so much a light dependent process as it is affected by cell differentiation rate.Supported by a grant of C.N.R.  相似文献

The localization of photosynthetic activity in developing maize (Zea mays L.) chloroplasts was studied in situ by two electron-microscopic-cytochemical methods. The activity of photosystem I was detected by photooxidation of 3,3-diaminobenzidine (DAB) and the activity of the photosystem II by photoreduction of thiocarbamyl nitrotetrazolium blue (TCNBT). During the transformation of proplastids into chloroplasts, at the base of the leaf blade the DAB reaction appeared before the TCNBT reaction. A positive DAB reaction was observed in the single thylakoids of plastids in cells located only about 0.5 mm above the base. Dark, osmiophilic DAB polymers accumulated in the lumina of the thylakoids. Plastid envelopes and tubules of the prolamellar bodies in immature chloroplasts were DAB-negative. In fully differentiated leaf tissue the DAB reaction was intense in the thylakoids of bundle-sheath chloroplasts, as well as in the stroma thylakoids and the peripheral grana thylakoids of mesophyll chloroplats. The photoreduction of TCNBT started in leaf tissue about 1 mm above the base. Dark granular material of reduced TCNBT appeared mostly in the partitions of grana, i.e. interthylakoidally, but some granules were also attached to the stroma thylakoids. The membranes of plastid envelopes and the tubules of prolamellar bodies showed a negative TCNBT reaction. Young bundle-sheath chloroplasts contained some reduced TCNBT in their grana; these deposits largely disappeared in the course of further differentiation. Summary Up to now Antirrhinum was classified as a typical example for a uniparentalmaternal inheritance of the plastids. However, the findings reported here prove that also the male gametophyte of Antirrhinum may occasionally transmit plastids into the egg. This conclusion is based on genetic experiments involving a form of the plastom mutant prasinizans which is described as gelbgrüne prasinizans. In contrast to all other plastid mutations known in Antirrhinum majus this mutant originated in Sippe 50 is completely viable. In plants containing plastids of this mutant type only, the mutant character is manifested during early growth stages. Cotyledons and first foliage leaves which are initially white or white yellow, slowly turn green and become indistinguishable from normal Sippe 50. Reciprocal crosses of green Sippe 50 with gelbgrüne prasinizans gave few variegated descendants; the others were exclusively plants identical with the maternal parent as far as leaf colour is concerned (Table). The variegated individuals cannot be gene mutants since selfing and crossing experiments showed non-mendelian inheritance. Furthermore it could be ruled out that in the cross Sippe 50 x gelbgrüne prasinizans the three variegated descendants represent spontaneous new plastom mutants because the pale tissue in these plants turned green in the same way as the paternal parent. Because of the typical greening of this mutant and since plastid mutations could be ruled out we have to conclude that plastids were transmitted by the pollen parent into the egg. There these plastids multiplied together with the maternal plastids giving rise to the chimeras after sorting-out of the two plastid types. This interpretation is supported by the observation of mixed cells in tissues where the leaf variegation is finely mosaiced. The results were possible only because the plastids of the pollen parent can be unequivocally recognised.  相似文献

Protein crystalloids are typical constituents of Aeonium domesticum plastids. They are composed of hexagonally arranged tube-like elements situated in the stroma without a bordering membrane. The single tubule has an external diameter of about 20 nm and an internal one of about 10 nm. The green-white-green mesochimera Ae. domesticum cv. variegatum contains normal chloroplasts in the green tissue and colourless plastids in the pale tissue. The defective plastids have a double-layered envelope, scarce internal membrane structures and contain, in the mature stage, a large vacuole. Plastid ribosomes can be detected only rarely in proplastids. They lose their ribosome complement entirely in the course of development. Polyacrylamide gel electrophoresis of total nucleic acids extracted from white tissue revealed the absence of the 23S and 16S rRNA normally present in plastids. Despite the loss of ribosomes, the plastids contain large protein crystalloids, which are structurally identical with those of normal green chloroplasts. Summary Transmission of paternal chloroplasts was observed in Nicotiana, considered to inherit organelles in a strictly maternal way. Plants carrying streptomycin resistant plastids were used as pollen donors. Cell lines with paternal plastids in the offspring were selected as green (resistant) sectors on calli induced from the seedlings on streptomycin-containing media. The presence of paternal plastids in the regenerated plants was confirmed by restriction analysis. In the Nicotiana plumbaginifolia xN. plumbaginifolia Np(SR1)3 and the N. plumbaginifolia Np(gos)29 xN. tabacum SR1 crosses 2.5% and 0.07% of the offspring were found to contain paternal (tabacum) plastids, respectively. These plants, however, carried maternal mitochondria exclusively. This sexual cybridization method offers a simple way to transfer chloroplasts solely, a goal not accessible by protoplast fusion.  相似文献

Zusammenfassung Im Dunkeln angezogene Zellkulturen von Nicotiana tabacum var. Samsun enthalten zahlreiche kleine Plastiden mit Stärkekörnern. In den Plastiden sind keine Pigmente nach weisbar. Im Stroma der Plastiden sind nur wenige isolierte Thylakoide und einige Tubuli und Vesikel zu erkennen. Prolamellarkörper, die für die Plastiden im Dunkeln angezogener Blätter charakteristisch sind, wurden nicht beobachtet.Im Licht ergrünen die Kulturen, und gleichzeitig wandeln sich die Plastiden in photosynthetisch aktive Chloroplasten um, die kleine Granastapel aus 4–8 Thylakoiden enthalten.Diese Umwandlung wird — im Gegensatz zur Chloroplastenentwicklung in Blättern — nur durch den blauen Anteil des sichtbaren Lichtes induziert.

Summary Dark grown cells in cultures of Nicotiana tabacum var. Samsun contain numerous small plastids with starch granules and without any detectable traces of pigments. The submicroscopic structure of these plastids consists of only a few isolated thylacoids, some vesiculi, and tubuli. Prolamellar bodies, characteristic of plastids in dark grown leaves, were not observed. In light the cells start to synthesize chlorophyll and the plastids are transformed into photosynthetic active chloroplasts which possess grana composed of stacks of 4–8 lamellae. It has been shown that this transformation is — in contrast to the development of the chloroplasts in leaves — caused only by the blue region of the visible light.

Summary An extracellular adenosine responsive site that stimulates adenylate cyclase activity has been identified in several tissues. There is limited information on the presence and physiologic significance of adenosine receptors in well-defined segments of the mammalian nephron. We therefore examined the effect of adenosine and selected analogues on basal hydraulic conductivity in rabbit cortical collecting tubules (CCT) perfused in vitro. Adenosine and analogues with an intact ribose moiety produced a significant, sustained increase in hydraulic conductivity. No increase in hydraulic conductivity was seen in either time control CCT's or CCT's exposed to an adenosine analogue with an altered ribose moiety. These experiments are compatible with the presence of a functional adenosine receptor which requires an intact ribose moiety and acts to increase hydraulic conductivity in the mammalian CCT.An intracellular adenosine responsive site, termed the P site, which inhibits adenylate cyclase activity, has also been described in several tissues. We therefore examined the effect of aP site agonist on hydraulic conductivity responses to arginine vasopressin, forskolin and cAMP.P site stimulation with 25 dideoxyadenosine inhibited the effect of AVP and of forskolin but not of cAMP to increase hydraulic conductivity. These results are compatible with a functionalP site in the rabbit CCT which acts at the catalytic subunit of adenylate cyclase to inhibit hydraulic conductivity. Together, these results demonstrate purinergic modulation of basal and arginine vasopressin-stimulated water flux in the mammalian collecting tubule.  相似文献
