Hydrogen metabolism was studied in anoxic sediments of the stratified Lake Mendota; using a method which allowed the measurement of in situ H2 concentrations and the headspace-free analysis of turnover of dissolved H2. Addition of sulfate resulted in partial but immediate inhibition of H2-dependent methanogenesis. Sulfate addition did not result in an immediate decrease in the steady state concentration of dissolved H2, nor did it significantly stimulate the rate constant of H2 turnover. Sulfate-induced decrease in dissolved H2 was only observed after prolonged incubation or when endogenous H2 production was stimulated by added glucose. The turnover of the in situ H2 accounted for only 14% of the H2-dependent methanogenesis from bicarbonate. While rates of methanogenesis increased during the season, rates of H2 turnover decreased, accounting for only 2% of the H2-dependent methanogenesis at the end of summer stratification. These observations indicate that increasing proportions of CH4 were formed from H2 being directly transferred in syntrophic methanogenic associations. The rapid inhibition of H2-dependent methanogenesis by exogenous sulfate may be explained at least partially by assuming methanogenic associations in which syntrophic sulfate reducers change their metabolism from fermentative H2 production to sulfate reduction.  相似文献

Concentrations of various sulfur compounds (SO42−, H2S, S0, acid-volatile sulfide, and total sulfur) were determined in the profundal sediments and overlying water column of a shallow eutrophic lake. Low concentrations of sulfate relative to those of acid-volatile sulfide and total sulfur and a decrease in total sulfur with sediment depth implied that the contribution of dissimilatory sulfur reduction to H2S production was relatively minor. Addition of 1.0 mM Na235SO4 to upper sediments in laboratory experiments resulted in the production of H235S with no apparent lag. Kinetic experiments with 35S demonstrated an apparent Km of 0.068 mmol of SO42− reduced per liter of sediment per day, whereas tracer experiments with 35S indicated an average turnover time of the sediment sulfate pool of 1.5 h. Total sulfate reduction in a sediment depth profile to 15 cm was 15.3 mmol of sulfate reduced per m2 per day, which corresponds to a mineralization of 30% of the particulate organic matter entering the sediment. Reduction of 35S0 occurred at a slower rate. These results demonstrated that high rates of sulfate reduction occur in these sediments despite low concentrations of oxidized inorganic compounds and that this reduction can be important in the anaerobic mineralization of organic carbon.  相似文献

Degradation of dimethyl sulfide and methanethiol in slurries prepared from sediments of minerotrophic peatland ditches were studied under various conditions. Maximal aerobic dimethyl sulfide-degrading capacities (4.95 nmol per ml of sediment slurry · h−1), measured in bottles shaken under an air atmosphere, were 10-fold higher than the maximal anaerobic degrading capacities determined from bottles shaken under N2 or H2 atmosphere (0.37 and 0.32 nmol per ml of sediment slurry · h−1, respectively). Incubations under experimental conditions which mimic the in situ conditions (i.e., not shaken and with an air headspace), however, revealed that aerobic degradation of dimethyl sulfide and methanethiol in freshwater sediments is low due to oxygen limitation. Inhibition studies with bromoethanesulfonic acid and sodium tungstate demonstrated that the degradation of dimethyl sulfide and methanethiol in these incubations originated mainly from methanogenic activity. Prolonged incubation under a H2 atmosphere resulted in lower dimethyl sulfide degradation rates. Kinetic analysis of the data resulted in apparent Km values (6 to 8 μM) for aerobic dimethyl sulfide degradation which are comparable to those reported for Thiobacillus spp., Hyphomicrobium spp., and other methylotrophs. Apparent Km values determined for anaerobic degradation of dimethyl sulfide (3 to 8 μM) were of the same order of magnitude. The low apparent Km values obtained explain the low dimethyl sulfide and methanethiol concentrations in freshwater sediments that we reported previously. Our observations point to methanogenesis as the major mechanism of dimethyl sulfide and methanethiol consumption in freshwater sediments.  相似文献

The vertical distribution of hydrogen was measured in the Loclat, a eutrophic and holomictic lake near Neuchâtel, Switzerland, before and during summer stratification. H2 concentrations decreased with depth in the anaerobic hypolimnion and were often below the detection limit (2.5 nl of H2 liter−1) in the water adjacent to the lake sediment. H2 was apparently not released from the lake sediment. The highest H2 concentrations (>4 μl of H2 liter−1) were observed in the aerobic water of the epilimnion and metalimnion. There, the H2 concentrations changed with time, indicating a turnover of H2. The H2 production processes could not be studied in the laboratory since incubation of water samples in light or darkness did not result in H2 production but rather always in H2 consumption. The possible role of cyanobacteria and algae for H2 production is discussed. Aerobic or anaerobic H2 consumption activities were observed at all depths of the water column, with highest activities in the hypolimnion. Aerobic H2 consumption activity was insensitive to azide inhibition, but sensitive to heat, mercuric chloride, or cyanide. It was restricted to a particle fraction of 0.2 to 3.0 μm in size, so that it must be due to single bacterial cells. Aerobic hydrogen bacteria, on the other hand, occurred in clusters of >3.0 μm. Therefore, the hydrogen bacteria could not have caused the H2 consumption in lake water. The aerobic H2 consumption activity followed Michaelis-Menten kinetics, with a Km of 67 nM H2. This is an exceptionally low value compared with Km values of hydrogenases in hydrogen bacteria and other species, but is similar to that for H2-decomposing abiontic soil hydrogenases.  相似文献

The kinetic parameters Km, Vmax, Tt (turnover time), and v (natural velocity) were determined for H2 and acetate conversion to methane by Wintergreen Lake sediment, using short-term (a few hours) methods and incubation temperatures of 10 to 14°C. Estimates of the Michaelis-Menten constant, Km, for both the consumption of hydrogen and the conversion of hydrogen to methane by sediment microflora averaged about 0.024 μmol g−1 of dry sediment. The maximal velocity, Vmax, averaged 4.8 μmol of H2 g−1 h−1 for hydrogen consumption and 0.64 μmol of CH4 g−1 h−1 for the conversion of hydrogen to methane during the winter. Estimated natural rates of hydrogen consumption and hydrogen conversion to methane could be calculated from the Michaelis-Menten equation and estimates of Km, Vmax, and the in situ dissolved-hydrogen concentration. These results indicate that methane may not be the only fate of hydrogen in the sediment. Among several potential hydrogen donors tested, only formate stimulated the rate of sediment methanogenesis. Formate conversion to methane was so rapid that an accurate estimate of kinetic parameters was not possible. Kinetic experiments using [2-14C]acetate and sediments collected in the summer indicated that acetate was being converted to methane at or near the maximal rate. A minimum natural rate of acetate conversion to methane was estimated to be about 110 nmol of CH4 g−1 h−1, which was 66% of the Vmax (163 nmol of CH4 g−1 h−1). A 15-min preincubation of sediment with 5.0 × 10−3 atm of hydrogen had a pronounced effect on the kinetic parameters for the conversion of acetate to methane. The acetate pool size, expressed as the term Km + Sn (Sn is in situ substrate concentration), decreased by 37% and Tt decreased by 43%. The Vmax remained relatively constant. A preincubation with hydrogen also caused a 37% decrease in the amount of labeled carbon dioxide produced from the metabolism of [U-14C]valine by sediment heterotrophs.  相似文献

In isolated Elodea densa leaves, the relationships between H+ extrusion (-ΔH+), K+ fluxes and membrane potential (Em) were investigated for two different conditions of activation of the ATP-dependent H+ pump. The ‘basal condition’ (darkness, no pump activator present) was characterized by low values of-ΔH+ and K+ uptake (ΔK+), wide variability of the ?ΔH+/ΔK+ ratio, relatively low membrane polarization and Em values more positive than EK for external K+ concentrations (|K+]o of up to 2mol m?3. A net K+ uptake was seen already at [K+]o below 1 mol m?3, suggesting that K+ influx in this condition was a thermodynamically uphill process involving an active mechanism. When the H+ pump was stimulated by fusicoccin (FC), by cytosol acidification, or by light (the ‘high polarization condition’), K+ influx largely dominated K+ and C? efflux, and the ?ΔH+/ΔK+ ratio approached unity. In the range 50 mmol m?3?5 mol m?3 [K+]0, Em was consistently more negative than EK. The curve of K+ influx at [K+]0 ranging from 50 to 5000mmol m?3 fitted a monophasic, hyperbolic curve, with an apparent half saturation value = 0–2 mol m?3. Increasing |K+]0 progressively depolarized Em, counteracting the strong hyperpolarizing effect of FC. The effects of K+ in depolarizing Em were well correlated with the effects on both K+ influx and ?ΔH+, suggesting a cause-effect chain: K+0 influx → depolarization → activation of H+ extrusion. Cs+ competitively inhibited K+ influx much more strongly in the ‘high polarization’ than in the ‘basal’ condition (50% inhibition at [Cs+]/[K+]0 ratios of 1:14 and 1:2, respectively) thus confirming the involvement of different K+ uptake systems in the two conditions. These results suggest that in E. densa leaves two distinct modes of interactions rule the relationships between H+ pump, membrane polarization and K+ transport. Denitrification in aquatic sediments was measured by an N2O reductase assay. Sediments consumed small added quantities of N2O over short periods (a few hours). In experiments with sediment slurries, N2O reductase activity was inhibited by O2, C2H2, heat treatment, and by high levels of nitrate (1 mM) or sulfide (10 mM). However, ambient levels of nitrate (<100 μM) did not influence activity, and moderate levels (about 150 μM) induced only a short lag before reductase activity began. Moderate levels of sulfide (<1 mM) had no effect on N2O reductase activity. Nitrous oxide reductase displayed Michaelis-Menten kinetics in sediments from freshwater (Km = 2.17 μM), estuarine (Km = 14.5 μM), and alkaline-saline (Km = 501 μM) environments. An in situ assay was devised in which a solution of N2O was injected into sealed glass cores containing intact sediment. Two estimates of net rates of denitrification in San Francisco Bay under approximated in situ conditions were 0.009 and 0.041 mmol of N2O per m2 per h. Addition of chlorate to inhibit denitrification in these intact-core experiments (to estimate gross rates of N2O consumption) resulted in approximately a 14% upward revision of estimates of net rates. Pore water and solid phase distributions of C, N, P and Si in sediments of the Arctic Ocean (Svalbard area) have been investigated. Concentrations of organic carbon (Corg) in the solid phase of the sediment varied from 1.3 to 2.8% (mean 1.9%), with highest concentrations found at shallow stations south/southwest of Svalbard. Relatively low concentrations were obtained at the deeper stations north/northeast of Svalbard. Atomic carbon to nitrogen ratios in the surface sediment ranged from below 8 to above 10. For some stations, high C/N ratios together with high concentrations of Corg suggest that sedimentary organic matter is mainly of terrigenous origin and not from overall biological activity in the water column. Organic matter reactivity (defined as the total sediment oxygen consumption rate normalized to the organic carbon content of the surface sediment) correlated with water depth at all investigated stations. However, the stations could be divided into two separate groups with different reactivity characteristics, representing the two most dominant hydrographic regimes: the region west of Svalbard mainly influenced by the West Spitsbergen Current, and the area east of Svalbard where Arctic polar water set the environmental conditions. Decreasing sediment reactivity with water depth was confirmed by the partitioning between organic and inorganic carbon of the surface sediment. The ratio between organic and inorganic carbon at the sediment-water interface decreased exponentially with water depth: from indefinite values at shallow stations in the central Barents Sea, to approximately 1 at deep stations north of Svalbard. At stations east of Svalbard there was an inverse linear correlation between the organic matter reactivity (as defined above) and concentration of dissolved organic carbon (DOC) in the pore water. The more reactive the sediment, the less DOC existed in the pore water and the more total carbonate (Ct or ΣCO2) was present. This observation suggests that DOC produced in reactive sediments is easily metabolizable to CO2. Sediment accumulation rates of opaline silica ranged from 0.35 to 5.7 μmol SiO2 m−2d−1 (mean 1.3 μmol SiO2 m−2d−1), i.e. almost 300 times lower than rates previously reported for the Ross Sea, Antarctica. Concentrations of ammonium and nitrate in the pore water at the sediment-water interface were related to organic matter input and water depth. In shallow regions with highly reactive organic matter, a pool of ammonium was present in the pore water, while nitrate conoentrations were low. In areas where less reactive organic matter was deposited at the sediment surface, the deeper zone of nitrification caused a build-up of nitrate in the pore water while ammonium was almost depleted. Nitrate penetrated from 1.8 to ≥ 5.8 cm into the investigated sediments. Significantly higher concentrations of “total” dissolved nitrogen (defined as the sum of NO3, NO2, NH4 and urea) in sediment pore water were found west compared to east of Svalbard. The differences in organic matter reactivity, as well as in pore water distribution patterns of “total” dissolved nitrogen between the two areas, probably reflect hydrographic factors (such as ice coverage and production/import of particulate organic material) related to the dominant water mass (Atlantic or Arctic Polar) in each of the two areas. We developed new techniques to measure dissolved H2 and H2 consumption kinetics in anoxic ecosystems that were not dependent on headspace measurements or gas transfer-limited experimentation. These H2 metabolism parameters were then compared with measured methane production rates, and estimates of H2 production and interspecies H2 transfer were made. The H2 pool sizes were 205 and 31 nM in sewage sludge from an anaerobic digestor and in sediments (24 m) from Lake Mendota, respectively. The H2 turnover rate constants, as determined by using in situ pool sizes and temperatures, were 103 and 31 h−1 for sludge and sediment, respectively. The observed H2 turnover rate accounted for only 5 to 6% of the expected H2-CO2-dependent methanogenesis in these ecosystems. Our results are in general agreement with the results reported previously and are used to support the conclusion that most of the H2-dependent methanogenesis in these ecosystems occurs as a consequence of direct interspecies H2 transfer between juxtapositioned microbial associations within flocs or consortia.  相似文献

Elongation of subapical segments of maize (Zea mays) roots was greatly inhibited by 2H2O in the incubation medium. Short-term exposure (30 min) to 2H2O slightly reduced O2 uptake and significantly increased ATP levels. 2H2O inhibited H+ extrusion in the presence of both low (0.05 mm) and high (5 mm) external concentrations of K+ (about 30 and 53%, respectively at 50% [v/v] 2H2O). Experiments on plasma membrane vesicles showed that H+-pumping and ATPase activities were greatly inhibited by 2H2O (about 35% at 50% [v/v] 2H2O); NADH-ferricyanide reductase and 1,3-β-glucan synthase activities were inhibited to a lesser extent (less than 15%). ATPase activities present in both the tonoplast-enriched and submitochondrial particle preparations were not affected by 2H2O. Therefore, the effect of short incubation time and low concentration of 2H2O is not due to a general action on overall cell metabolism but involves a specific inhibition of the plasma membrane H+ -ATPase. K+ uptake was inhibited by 2H2O only when K+ was present at a low (0.05 mm) external concentration where absorption is against its electrochemical potential. The transmembrane electric potential difference (Em) was slightly hyperpolarized by 2H2O at low K+, but was not affected at the higher K+ concentrations. These results suggest a relationship between H+ extrusion and K+ uptake at low K+ external concentration.  相似文献

Biogeochemical processes in the groundwater discharge zone of urban streams   总被引:1,自引:0,他引:1  
The influence of biogeochemical processes on nitrogen and organic matter transformation and transport was investigated for two urban streams receiving groundwater discharge during the dry summer baseflow period. A multiple lines of evidence approach involving catchment-, and stream reach-scale investigations were undertaken to describe the factors that influence pore water biogeochemical processes. At the catchment-scale gaining stream reaches were identified from water table mapping and groundwater discharge estimated to be between 0.1 and 0.8 m3 m?2 d?1 from baseflow analysis. Sediment temperature profiles also suggested that the high groundwater discharge limited stream water infiltration into the sediments. At the stream reach-scale, dissolved organic carbon (DOC) and dissolved organic nitrogen (DON) concentrations were higher in stream water than in groundwater. However, DOC and DON concentrations were greatest in sediment pore water. This suggests that biodegradation of sediment organic matter contributes dissolved organic matter (DOM) to the streams along with that delivered with groundwater flow. Pore water ammonium (NH4 +) was closely associated with areas of high pore water DOM concentrations and evidence of sulfate (SO4 2?) reduction (low concentration and SO4:Cl ratio). This indicates that anoxic DOM mineralization was occurring associated with SO4 2? reduction. However the distribution of anoxic mineralization was limited to the center of the streambed, and was not constrained by the distribution of sediment organic matter which was higher along the banks. Lower sediment temperatures measured along the banks compared to the center suggests, at least qualitatively, that groundwater discharge is higher along the banks. Based on this evidence anoxic mineralization is influenced by groundwater residence time, and is only measurable along the center of the stream where groundwater flux rates are lower. This study therefore shows that the distribution of biogeochemical processes in stream sediments, such as anoxic mineralization, is strongly influenced by both the biogeochemical conditions and pore water residence time.  相似文献

Low dissolved oxygen (DO) levels often occur during summer in tidal creeks along the southeastern coast of the USA. We analyzed rates of oxygen loss as water-column biochemical oxygen demand (BOD5) and sediment oxygen flux (SOF) at selected tidal creek sites monthly over a 1-year period. Ancillary physical, chemical and biological data were collected to identify factors related to oxygen loss. BOD5 rates ranged from 0.0 mg l?1 to 7.6 mg l?1 and were correlated positively with organic suspended solids, total suspended solids, chlorophyll a concentrations, temperature, and dissolved oxygen, and negatively with pH and nitrate + nitrite. SOF rates ranged from 0.0 to 9.3 g O2 m?2 d?1, and were positively correlated with temperature, chlorophyll a, and total suspended solids, but negatively with dissolved oxygen. Both forms of oxygen uptake were seasonally dependent, with BOD5 elevated in spring and summer and SOF elevated in summer and fall. Average oxygen loss to sediments was greater and more variable than oxygen loss in the water column. Oxygen deficits at three of five locations were significantly related to BOD5 and SOF, but not at two sites where ground water discharges were observed. Correlation and principal component analyses suggested that BOD5 and SOF responded to somewhat different suites of environmental variables. BOD5 was driven by a set of parameters linked to warm season storm water inputs that stimulated organic seston loads, especially chlorophyll a, while SOF behaved less strongly so. Runoff processes that increase loads of organic material and nutrients and ground water discharges low in dissolved oxygen contribute to occurrences of low dissolved oxygen in tidal creeks.  相似文献

The (Ca2+ + Mg2+)-dependent ATPase (ATP phosphohydrolase, EC 3.6.1.3) from human erythrocytes occurred in two different states, A-state and B-state, depending on the membrane preparation.The A-state showed low maximum activity (V) and the Ca2+ activation was characterized by a Hill coefficient, nH, of about 1 and a Michaelis constant, KCa, about 30 μM.The B-state showed high V, a nH above 1, which indicates positive cooper-activity of Ca2+ activation, and a KCa of about 1 μM.With varying ATP concentrations, both the A-state and the B-state showed negative cooperativity and slightly different values of Km.The B-state was shifted to the A-state when the membranes were exposed to low Ca2+ concentrations. The shift reached 50% at approx. 0.5 μM Ca2+. At the low Ca2+ concentrations an activator was released from the membranes.The A-state was shifted to the B-state when the membranes were exposed to Ca2+ in the presence of the activator. The shift reached 50% at about 30 μM Ca2+. The recovery of high V was time dependent and lasted several minutes. Increasing concentrations of Ca2+ and activator accelerated the recovery.It is suggested that the A-state and the B-state correspond to enzyme free of activator and enzyme associated with activator, respectively. Furthermore, the two states may represent a resting and an active state, respectively, of the calcium pump.  相似文献

Activity of extracellular phosphatases (phosphomonoesterases) was measured in sandy streambed sediments of the Breitenbach, a small unpolluted upland stream in Central Germany. Fluorigenic 4-methylumbelliferyl phosphate served as a model substrate. Experiments were conducted using sediment cores in a laboratory simulation of diffuse groundwater discharge through the stream bed, a natural process occurring in the Breitenbach as well as many other streams.Streambed sediments contained high levels of particulate phosphorus, but concentrations of dissolved phosphorus in the interstitial water were 3 to 4 orders of magnitude lower. These interstitial concentrations were similar to those in the stream and groundwater. Extracellular phosphatase activity was high in the streambed sediments. These enzymes probably contribute significantly to the flux of phosphorus in sediment by hydrolyzing phosphomonoesters, making free phosphate available to the sediment microorganisms.Factors influencing the kinetic parameters V max (maximum activity) and apparent K m (enzyme affinity) of phosphatase were discharge rates of water through the sediment, water quality (ground- or stream water), and substrate (phosphomonoesters) as well as dissolved ortho-phosphate concentrations. Enzymes are supposed to be effective at limiting substrate concentrations, where, in this study, changes in discharge rates had little influence on rates of hydrolysis. Higher V max and lower K m values were found during percolation of groundwater through the sediment cores, compared with stream water. This indicates that rates of hydrolysis were higher with groundwater, both at substrate limitation and at substrate saturation. This was probably a consequence of the lower levels of dissolved ortho-phosphate in the groundwater.  相似文献

We measured sediment production of carbon dioxide (CO2) and methane (CH4) and the net flux of CO2 across the surfaces of 15 boreal and subarctic lakes of different humic contents. Sediment respiration measurements were made in situ under ambient light conditions. The flux of CO2 between sediment and water varied between an uptake of 53 and an efflux of 182 mg C m−2 day−1 from the sediments. The mean respiration rate for sediments in contact with the upper mixed layer (SedR) was positively correlated to dissolved organic carbon (DOC) concentration in the water (r2 = 0.61). The net flux of CO2 across the lake surface [net ecosystem exchange (NEE)] was also closely correlated to DOC concentration in the upper mixed layer (r2 = 0.73). The respiration in the water column was generally 10-fold higher per unit lake area compared to sediment respiration. Lakes with DOC concentrations <5.6 mg L−1 had net consumption of CO2 in the sediments, which we ascribe to benthic primary production. Only lakes with very low DOC concentrations were net autotrophic (<2.6 mg L−1) due to the dominance of dissolved allochthonous organic carbon in the water as an energy source for aquatic organisms. In addition to previous findings of allochthonous organic matter as an important driver of heterotrophic metabolism in the water column of lakes, this study suggests that sediment metabolism is also highly dependent on allochthonous carbon sources.  相似文献

The acetylene block technique was employed to study denitrification in intertidal estuarine sediments. Addition of nitrate to sediment slurries stimulated denitrification. During the dry season, sediment-slurry denitrification rates displayed Michaelis-Menten kinetics, and ambient NO3 + NO2 concentrations (≤26 μM) were below the apparent Km (50 μM) for nitrate. During the rainy season, when ambient NO3 + NO2 concentrations were higher (37 to 89 μM), an accurate estimate of the Km could not be obtained. Endogenous denitrification activity was confined to the upper 3 cm of the sediment column. However, the addition of nitrate to deeper sediments demonstrated immediate N2O production, and potential activity existed at all depths sampled (the deepest was 15 cm). Loss of N2O in the presence of C2H2 was sometimes observed during these short-term sediment incubations. Experiments with sediment slurries and washed cell suspensions of a marine pseudomonad confirmed that this N2O loss was caused by incomplete blockage of N2O reductase by C2H2 at low nitrate concentrations. Areal estimates of denitrification (in the absence of added nitrate) ranged from 0.8 to 1.2 μmol of N2 m−2 h−1 (for undisturbed sediments) to 17 to 280 μmol of N2 m−2 h−1 (for shaken sediment slurries).  相似文献

Michaelis-Menten kinetic parameters for H2 consumption by three methanogenic habitats were determined from progress curve and initial velocity experiments. The influences of mass transfer resistance, endogenous H2 production, and growth on apparent parameter estimates were also investigated. Kinetic parameters could not be determined for undiluted rumen fluid and some digestor sludge from gas-phase measurements of H2, since mass transfer of H2 across the gas-liquid interface was rate limiting. However, accurate values were obtained once the samples were diluted. H2 consumption by digestor sludge with a long retention time and by hypereutrophic lake sediment was not phase transfer limited. The Km values for H2 uptake by these habitats were similar, with means of 5.8, 6.0, and 7.1 μM for rumen fluid, digestor sludge, and sediment, respectively. Vmax estimates suggested a ratio of activity of approximately 100 (rumen fluid):10 (sludge):1 (sediment); their ranges were as follows: rumen fluid, 14 to 28 mM h−1; Holt sludge, 0.7 to 4.3 mM h−1; and Wintergreen sediment, 0.13 to 0.49 mM h−1. The principles of phase transfer limitation, studied here for H2, are the same for all gaseous substrates and products. The limitations and errors associated with gas phase determination of kinetic parameters were evaluated with a mathematical model that combined mass transport and Michaelis-Menten kinetics. Three criteria are described which can be used to evaluate the possibility that a phase transfer limitation exists. If it does not exist, (i) substrate consumption curves are Michaelis-Menten and not first order, (ii) the Km is independent of initial substrate concentration, and (iii) the Km is independent of biomass (Vmax) and remains constant with dilution of sample. Errors in the Michaelis-Menten kinetic parameters are caused by endogenously produced H2, but they were <15% for rumen fluid and 10% for lake sediment and digestor sludge. Increases in Vmax during the course of progress curve experiments were not great enough to produce systematic deviations from Michaelis-Menten kinetics.  相似文献

Oxyleghemoglobin was used to supply low concentrations of O2 to H2-oxidizing bacteroids from Rhizobium japonicum USDA 122 DES. The H2 oxidation system of these bacteroids was capable of effectively utilizing O2 at the low concentrations of O2 expected to be found in soybean nodules. Apparent Km values of approximately 10 nanomolar O2 have been calculated for the oxyhydrogen reaction. These values include the Km values for both H2 oxidation and endogenous substrate oxidation. Even in the presence of oxyleghemoglobin, H2 additions stimulated C2H2 reduction, reduced the rate of endogenous respiration and maintained the ATP contents of bacteroids. In our reconstituted oxyleghemoglobin and bacteriod system, we estimate that the H2 oxidation system is capable of recycling all of the H2 evolved during the N2 fixation process.  相似文献

We measured potential rates of bacterial dissimilatory reduction of 75SeO42− to 75Se0 in a diversity of sediment types, with salinities ranging from freshwater (salinity = 1 g/liter) to hypersaline (salinity = 320 g/liter and with pH values ranging from 7.1 to 9.8. Significant biological selenate reduction occurred in all samples with salinities from 1 to 250 g/liter but not in samples with a salinity of 320 g/liter. Potential selenate reduction rates (25 nmol of SeO42− per ml of sediment added with isotope) ranged from 0.07 to 22 μmol of SeO42− reduced liter−1 h−1. Activity followed Michaelis-Menten kinetics in relation to SeO42− concentration (Km of selenate = 7.9 to 720 μM). There was no linear correlation between potential rates of SeO42− reduction and salinity, pH, concentrations of total Se, porosity, or organic carbon in the sediments. However, potential selenate reduction was correlated with apparent Km for selenate and with potential rates of denitrification (r = 0.92 and 0.81, respectively). NO3, NO2, MoO42−, and WO42− inhibited selenate reduction activity to different extents in sediments from both Hunter Drain and Massie Slough, Nev. Sulfate partially inhibited activity in sediment from freshwater (salinity = 1 g/liter) Massie Slough samples but not from the saline (salinity = 60 g/liter) Hunter Drain samples. We conclude that dissimilatory selenate reduction in sediments is widespread in nature. In addition, in situ selenate reduction is a first-order reaction, because the ambient concentrations of selenium oxyanions in the sediments were orders of magnitude less than their Kms.  相似文献

The denitrification rates in a marine sediment, estimated by using 15N-nitrate, Vmax, Km, and sediment nitrate concentrations, were 12.5 and 2.0 nmol of N2-N cm−3 day−1 at 0 to 1 and 1 to 3 cm, respectively, at 12°C. The total rate was 165 nmol of N2-N m−2 day−1.  相似文献
