Invertebrates from the Sea of Okhotsk were studied as a source of proteins that are capable of binding to lipopolysaccharides (LPSs) of gram-negative bacteria. A DOT analysis using dansyl-labeled LPS that we developed revealed LPS-binding proteins in blood-cell lysates of 21 out of 33 investigated species of invertebrates. Most of the investigated species with positive LPS-binding activity were decapod crustaceans (class Malacostraca, phylum Arthropoda). Hemocyte lysates from the red king crab Paralithodes camtschaticus and the sculptured shrimp Sclerocrangon boreas contained several LPS-binding proteins with different molecular weights. LPS-binding proteins were found for the first time in echinoderms (classes Holothuroidea, Asteroidea, and Echinoidea), sipunculans, and brachiopods.  相似文献

This review outlines current knowledge of mechanisms effecting metamorphosis in decapod crustaceans and insects. The comparative approach demonstrates some of the complexities that need resolving to find an answer to the question raised frequently by ecologists: “What triggers metamorphosis in spiny lobsters?” It is evident that crustacean moulting and metamorphosis are genetically controlled through endocrine systems that mediate gene expression. The molecular mechanisms underlying these developmental processes have been studied intensively in insects, particularly in the fruitfly, Drosophila melanogaster (Diptera), and some lepidopteran species. Comparatively, there is minimal information available for a few decapod crustacean species, but none for spiny lobsters (Palinuridae). Nothing was known of hormone signalling transduction pathways, via nuclear receptors (NRs) and gene activation during larval moults in palinurids—until a recent, ground-breaking study of early phyllosomal development of Panulirus ornatus by Wilson et al. (Rock Lobster Enhancement and Aquaculture Subprogram. FRDC Project 2000/263, Australian Govt, Fisheries Research and Development Corporation and Australian Institute of Marine Science, Nov 2005). Their study not only identified homologues of five hormone NRs of D. melanogaster, but also patterns of gene regulation showing strong similarities to those of gene expression found in insect larval development. Their results indicated that control of moulting and metamorphosis in palinurids closely parallels that in insects, suggesting that insects can serve as model systems for elucidating molecular mechanisms in larval decapods. In insects and crustaceans, the steroid hormone, ecdysone, (20E) initiates moulting. In insects, juvenile hormone (JH) mediates the type of larval moult that occurs, either anamorphic or metamorphic. The latter results when the level of JH in the haemolymph drops in the final larval instar. High levels of JH inhibit the metamorphic moult during insect larval development. The interaction of 20E and JH is not fully understood, and the operative molecular mechanisms are still being elucidated. No nuclear receptor for JH has been identified, and alternative JH signalling pathways await identification. In decapod crustaceans, methyl farnesoate (MF), a precursor of JH, replaces the latter in other functions mediated by JH in insects; but there is little evidence indicating that MF plays a similar ‘antimetamorphic’ role in decapod larval moults.  相似文献

During spermatogenesis in most animals, the basic proteins associated with DNA are continuously changing and somatic-typed histones are partly replaced by sperm-specific histones, which are then successively replaced by transition proteins and protamines. With the replacement of sperm nuclear basic proteins, nuclei progressively undergo chromatin condensation. The Chinese Mitten Crab (Eriocheir sinensis) is also known as the hairy crab or river crab (phylum Arthropoda, subphylum Crustacea, order Decapoda, and family Grapsidae). The spermatozoa of this species are aflagellate, and each has a spherical acrosome surrounded by a cup-shaped nucleus, peculiar to brachyurans. An interesting characteristic of the E. sinensis sperm nucleus is its lack of electron-dense chromatin. However, its formation is not clear. In this study, sequences encoding histones H3 and H4 were cloned by polymerase chain reaction amplification. Western blotting indicated that H3 and H4 existed in the sperm nuclei. Immunofluorescence and ultrastructural immunocytochemistry demonstrated that histones H3 and H4 were both present in the nuclei of spermatogonia, spermatocytes, spermatids and mature spermatozoa. The nuclear labeling density of histone H4 decreased in sperm nuclei, while histone H3 labeling was not changed significantly. Quantitative real-time PCR showed that the mRNA expression levels of histones H3 and H4 were higher at mitotic and meiotic stages than in later spermiogenesis. Our study demonstrates that the mature sperm nuclei of E. sinensis contain histones H3 and H4. This is the first report that the mature sperm nucleus of E. sinensis contains histones H3 and H4. This finding extends the study of sperm histones of E. sinensis and provides some basic data for exploring how decapod crustaceans form uncondensed sperm chromatin.  相似文献

The spermatozoa from the shrimp Palaemon serratus have been studied with respect to their morphological and biochemical characters.At the ultrastructural level the acrosomal vesicle of the nail-shaped spermatozoon displays a periodically striated bundle. This appearance seems to be characteristic for the Palaemonidae.The biochemical parameters of the chromatin, extracted from the spermatozoa, have been compared with those obtained from the testes and the hepatopancreas. The basic proteins of testes and sperm have been characterized by polyacrylamide gel electrophoresis. Protamines appear to be absent in the sperm of P. serratus. On the contrary, histones with a somatic character remain present. Moreover, supplementary histone fractions have been identified. These proteins have been further studied after purification on Biogel P-100. Their amino acid composition shows similarities to that of the H2A and H2B histone and has a molecular weight of 15,500±300.  相似文献

Tropomyosin represents a major allergen of decapod crustaceans such as shrimps and crabs, and its highly conserved amino acid sequence (>90% identity) is a molecular basis of the immunoglobulin E (IgE) cross-reactivity among decapods. At present, however, little information is available about allergens in edible crustaceans other than decapods. In this study, the major allergen in two species of edible crustaceans, Antarctic krill Euphausia superba and mantis shrimp Oratosquilla oratoria that are taxonomically distinct from decapods, was demonstrated to be tropomyosin by IgE-immunoblotting using patient sera. The cross-reactivity of the tropomyosins from both species with decapod tropomyosins was also confirmed by inhibition IgE immunoblotting. Sequences of the tropomyosins from both species were determined by complementary deoxyribonucleic acid cloning. The mantis shrimp tropomyosin has high sequence identity (>90% identity) with decapod tropomyosins, especially with fast-type tropomyosins. On the other hand, the Antarctic krill tropomyosin is characterized by diverse alterations in region 13–42, the amino acid sequence of which is highly conserved for decapod tropomyosins, and hence, it shares somewhat lower sequence identity (82.4–89.8% identity) with decapod tropomyosins than the mantis shrimp tropomyosin. Quantification by enzyme-linked immunosorbent assay revealed that Antarctic krill contains tropomyosin at almost the same level as decapods, suggesting that its allergenicity is equivalent to decapods. However, mantis shrimp was assumed to be substantially not allergenic because of the extremely low content of tropomyosin.  相似文献

The three species investigated, the European eel, Anguilla anguilla (L.), the Mediterranean moray, Muraena helena L., and the conger eel, Conger coner (L.), represent three different superfamilies of the suborder Anguilloidei (infradivision Elopomorpha). Their hemoglobin systems show peculiar structural properties, which distinuish them from all other teleost species studied. They present acidic and basic components differing greatly in their isoelectric points; the basic components have the highest isoelectric points detected in teleost hemoglobins. While there is one major basic component, multiplicity is present in the acidic components in Muraena and Conger. The polyeptides of the acidic comonents show the same electrophoretic mobility in 8 M urea. In the three species, the electroploretic mobility with urea-SDS of the hemoglobin polypeptides shows a shorter β polypeptide in the basic components. These features had never been investigated among Elopomorpha, and are likely to be phylogenetically relevant.  相似文献

The origin and early diversification of decapod crustaceans and their expansion from marine to continental environments are key events in arthropod evolution. Rare fossil decapods are known from the Palaeozoic, and the earliest eumalacostracans with undoubted decapod affinities are the Late Devonian Palaeopalaemon and Aciculopoda, found in offshore marine deposits. Here, we describe a new species of the shrimp Tealliocaris found in floodplain and temporary pond deposits from the Famennian (Late Devonian) of Belgium, together with a rare Palaeozoic assemblage of other crustaceans (conchostracans, notostracans and anostracans) and chelicerates (eurypterids). Tealliocaris walloniensis sp. nov. documents the earliest occurrence of continental decapod crustaceans and indicates that decapods have been part of continental ecosystems at least since the Late Devonian.  相似文献

The study of the rate of oxygen consumption by Nephrops norvegicius (L.) at each moulting stage has been carried out in closed respiration chambers, using an oxygen electrode to measure the oxygen concentration.Although the mean respiration rate seems to be low when compared with other decapod crustaceans of similar weight, the general trends of the variation of the oxygen consumption during the moulting cycle are similar to those described for other species of decapods. The spermatozoon of the Carib grackle, Quiscalus lugubris, a member of the family Icteridae, is generally similar in organization to the passerine-type of spermatozoon, in being highly elongated and displaying a helical structure of the acrosome, nucleus and principal piece of the tail. There are subtle variations in acrosomal structural features between this organelle in the grackle and that in some of the very few passerine species of birds in which the spermatozoon has been studied. The proximal centriole is present, and, thus, the Carib grackle is the third passeridan bird in which this organelle, hitherto regarded as absent in passerine birds, has been described in the spermatozoon. The spermatozoon of this bird also possesses a granular helix, which feature has been found variably even in the scanty available reports on passerine spermatozoa. It is advocated that the spermatozoon be studied in many more species of this large clade of birds. This report provides a basis for the study of spermiogenesis in the Carib grackle, with the aim of exposing, inter alia, a number of developmental features and processes of certain organelles that have received attention, recently, in the spermatozoa of passerine birds.  相似文献

The keeping of home aquaria is one of the most popular hobbies globally. In contrast to the ornamental fish trade, decapod crustaceans such as shrimps, crayfish and crabs are relatively new to the pet trade. Nevertheless, the popularity of ornamental shrimp in freshwater aquaria has rapidly increased in recent years. Indonesia is one of the leading producers and exporters of ornamental aquatic animals globally and the Czech Republic is a gateway for these animals into the European Union territory. The pathway for introductions of organisms unintentionally moved in association with ornamental shrimps via the international trade has to date not been evaluated. We examined a small number of shrimps imported from Indonesia into the Czech Republic in May 2015 and found large numbers of the protozoan Vorticella sp., one species of scutariellid temnocephalidan (Caridinicola sp.), and one species of bdelloid rotifer, associated with two species of atyid shrimps, indicating an invasion risk from fauna carried unintentionally by this vector. Although our observations were limited in scale, we estimate the total number of commensal fauna imported into the Czech Republic with ornamental shrimps via the pet trade to be in the order of hundreds of thousands per month. As attached organisms can directly or indirectly cause diseases in certain species of decapod crustaceans, we recommend five steps to reduce risks of introduction of “hitchhikers” to aquaria and wildlife.  相似文献

The neuroanatomy of the olfactory pathway has been intensely studied in many representatives of Malacostraca. Nevertheless, the knowledge about bilateral olfactory integration pathways is mainly based on Decapoda. Here, we investigated the olfactory projection neuron pathway of two marine isopod species, Saduria entomon and Idotea emarginata, by lipophilic dye injections into the olfactory neuropil. We show that both arms of the olfactory globular tract form a chiasm in the center of the brain, as known from several other crustaceans. Furthermore, the olfactory projection neurons innervate both the medulla terminalis and the hemiellipsoid body of the ipsi- and the contralateral hemisphere. Both protocerebral neuropils are innervated to a comparable extent. This is reminiscent of the situation in the basal decapod taxon Dendrobranchiata. Thus, we propose that an innervation by the olfactory globular tract of both the medulla terminalis and the hemiellipsoid body is characteristic of the decapod ground pattern, but also of the ground pattern of Caridoida.  相似文献

To add to our understanding of dendrocoelid spermatozoa and to describe additional phylogenetic characters, the ultrastructure of the testis was investigated in the subterranean freshwater planarian Dendrocoelum constrictum. This is the first study investigating spermatogenesis and spermatozoon ultrastructure in a subterranean freshwater planarian species. We found that the basic structure of spermatozoa in D. constrictum is similar to that of other Tricladida that have been studied previously. In fact, D. constrictum spermatozoa possess an elongated nucleus, one giant mitochondrion, and two subterminal flagella with a 9 + ‘1’ pattern. The flagella emerge together from one side of the spermatozoon. However, D. constrictum has some characteristics that have not yet been described for other freshwater planarians. In fact, the number of cortical microtubules reaches the maximum number in the anterior and middle part of region I, and then decrease until they disappear towards the posterior extremity of the spermatozoon. The extreme tip of the anterior region of the spermatozoon exhibits a specific external ornamentation of the plasma membrane.  相似文献

Antarctic marine invertebrates live in a cold, thermally stable environment and cannot tolerate large changes in body temperature (i.e. they are stenothermal). Their temperate relatives, by contrast, are eurythermal, living in warmer and thermally more variable environments. Have these different environments influenced how specific behaviours are affected by changes of temperature? This question was addressed in two temperate crustaceans, the decapod Carcinus maenas and isopod Ligia oceanica, and two Antarctic crustaceans, the isopod Glyptonotus antarcticus and amphipod Paraceradocus gibber. The thermal dependence of walking speed was analysed by contrasting the slopes of the linear part of each species’ behavioural curve. Over the temperature ranges analysed, the temperature sensitivity of walking speed in the stenotherms was 13–23% that of the eurytherms when measured in body lengths s?1. There was a linear relationship between walking speed and temperature up to +4.5°C in the Antarctic species G. antarcticus and P. gibber. Elevating temperature by up to 3.5°C above the maximum temperature experienced in the Antarctic (+1°C), does not lead to an acute breakdown of motor coordination. We describe for the first time the righting behaviour of G. antarcticus. The mean time-to-right tended to a minimum on warming from ?2 to+5°C, but this trend was not statistically significant. Our results suggest that the physiological adaptations which permit continued activity at low Antarctic temperatures have resulted in a lower thermal dependence of activity in Antarctic species, compared to related temperate species.  相似文献

Phylogenetic relationships within decapod crustaceans are highly controversial. Even recent analyses based on molecular datasets have shown largely contradictory results. Previous studies using mitochondrial genomes are promising but suffer from a poor and unbalanced taxon sampling. To fill these gaps we sequenced the (nearly) complete mitochondrial genomes of 13 decapod species: Stenopus hispidus, Polycheles typhlops, Panulirus versicolor, Scyllarides latus, Enoplometopus occidentalis, Homarus gammarus, Procambarus fallax f. virginalis, Upogebia major, Neaxius acanthus, Calocaris macandreae, Corallianassa coutierei, Cryptolithodes sitchensis, Neopetrolisthes maculatus, and add that of Dromia personata. Our new data allow for comprehensive analyses of decapod phylogeny using the mitochondrial genomes of 50 species covering all major taxa of the Decapoda. Five species of Stomatopoda and one species of Euphausiacea serve as outgroups. Most of our analyses using Maximum Likelihood (ML) and Bayesian inference (BI) of nucleotide and amino acid datasets revealed congruent topologies for higher level decapod relationships: (((((((Anomala, Brachyura), Thalassinida: Gebiidea), Thalassinida: Axiidea), (Astacidea, Polychelida), Achelata), Stenopodidea), Caridea), Dendrobranchiata). This result corroborates several traditional morphological views and adds new perspectives. In particular, the position of Polychelida is surprising. Nevertheless, some problems can be identified. In a minority of analyses the basal branching of Reptantia is not fully resolved, Thalassinida are monophyletic; Polychelida are the sister group to Achelata, and Stenopodidea are resolved as sister group to Caridea. Despite this and although some nodal supports are low in our phylogenetic trees, we think that the largely stable topology of the trees regardless of different types of analyses suggests that mitochondrial genomes show good potential to resolve the relationship within Decapoda.  相似文献

Several tissues of different protostome species were tested in respect to the occurence of sialic acid. There is no sialic acid in the turbellarian Euplanaria gonocephala and the mollusc Helix pomatia. In contrast to other tissues the digestive gland of the decapod crustaceans Astacus leptodactylus and Uca tangeri contains 60–80 μg of sialic acid per gram wet weight. Radioactive tracer experiments showed that Astacus leptodactylus cannot synthesize sialic acid by using the specific precursor N-acetyl-mannosamine but obviously resorbs exogenous sialic acid taken up with the food and stores it in the digestive gland. It is suggested that protostomes in general are devoid of endogenous sialic acid whereas it is abundant in deuterostomes. Contradictory to existing results low amounts of a resorcinol positive substance, probably a sialic acid, were detected in the deuterostome tunicate Phallusia mammilata.  相似文献

Proteome maps of hepatopancreas (midgut gland) and ovarian tissues of the crustacean, Cancer pagurus (Decapoda; edible crab) have been produced by 2D-PAGE and identification of proteins, following trypsin proteolysis, by electrospray MS/MS and database searching. Owing to the lack of sequence information on proteins and fully sequenced genomes amongst the decapod crustaceans and given the evolutionary distance to the nearest full genome database (Daphnia), it was necessary to adopt a non-conventional identification approach. Thus, a strategy was developed for effective identification of decapod proteins by sequence similarity, homology-based cross-species database searching, using various algorithms and a combination of NCBI Crustacea and Arthropoda databases, together with the Arthropoda PartiGene database (Blaxter, University of Edinburgh). In both hepatopancreas and ovary tissues, the largest group of proteins identified were a variety of enzymes, followed by a smaller number of storage/transport proteins [including vitellogenin (yolk protein), several subunits of hemocyanin, cryptocyanin, ferritin and calreticulin], with fewer structural proteins (actin, tubulin) and heat-shock proteins, in addition to a number of proteins of miscellaneous functions. Such protein identifications allow the development of tools, such as antibodies and RNA/DNA probes, to investigate the functions of the proteins in specific tissues during development.  相似文献
