 <正> 单葡萄糖甘油二酯(MGDG)和双葡萄糖甘油二酯(DGDG)是莱氏衣原体膜上主要的极性脂。糖脂的生理功能过去很少研究,至今仍不清楚。近年来实验结果表明,MGDG在膜上容易形成六角形Ⅱ结构,而DGDG则形成脂双层结构。六角形Ⅱ结构的出现与生物膜的生理功能的关系是当前瞩目的研究内容。本文首次研究了外源脂肪酸和胆固酵对莱氏衣原体AIH089菌株膜上糖脂含量的影响。  相似文献

支原体是许多疾病的病原及组织培养和细胞培养中常见的污染物。目前常用抗生素抑制其生长。本文以莱氏衣原体AIH089为材料,用电子顺磁共振波谱技术研究红霉素和土霉素对莱氏衣原体膜脂流动性的影响,并用薄板层和气色谱技术分析膜上磷脂和脂肪酸的组成,发现红霉素和土霉素能使莱氏衣原体膜上心磷脂和硬脂酸的含量减少,使棕榈酸和油酸的含量增加,提高膜脂的流动性。改变膜脂组成,提高膜脂的流动性可能是这两种抗生素抑制莱氏衣原体生长的机制之一。  相似文献

本文以莱氏衣原体AIH089为材料,用DPH荧光偏振等技术研究红霉素和土霉素对莱氏衣原体膜流动性和Mg~(2+)-ATPase活性的影响,并用聚丙烯酰胺梯度凝胶电泳技术进一步分析膜蛋白的组成,发现红霉素和土霉素能使莱氏衣原体膜的流动性显著增加,使Mg~(2+)-ATPase活性显著降低。红霉素和土霉素对莱氏衣原体膜流动性和膜上Mg~(2+)-ATPase活性的影响与它们的抑菌能力有很好的相关性。  相似文献

DPH标记细胞膜的动力学与膜脂流动性的荧光偏振校正测量   总被引:1,自引:0,他引:1  
用稳态荧光技术测得经过校正的荧光成分,由此算出用DPH标记的细胞膜的偏振度。方法是作荧光偏振值在随时间变化的曲线,将其外推至零标记时间求出该时间的荧光偏振值。用此法测定了艾氏腹水癌细胞的膜流动性。结果表明流动性比用整个细胞测得之值小,说明膜脂的有序程度和包装密度比胞浆中的脂大。实验结果和用三房空模型分析所得的理论值符合较好,提示荧光探剂的标记过程主要受分子扩散所控制。  相似文献

应用荧光探剂 1,6-二苯基-1,3,5-己三烯(DPH)观察外源胆固醇对水稻极端线粒体膜流动性的影响。结果表明,无论外源胆固醇通过水培根系吸收或是直接添加给予水稻根端线粒体,都能使DPH与线粒体结合后的荧光强度减弱,使荧光偏振度和微粘度降低,增加水稻根端线粒体膜流动性。  相似文献

拟菊酯是一类新型农药,其生物活性与结构有关,如含氰基拟菊酯在哺乳动物脑组织中与γ-氨基丁酸受体-离子载体复合物空间特异性结合,而不含氰基拟菊酯并非如此。Stelzer等用荧光法研究了氯菊酯、氯氰菊酯等对大鼠淋巴细胞膜的影响,认为上述两类拟菊酯基本上以同样的方式影响膜脂排列。为深入探讨氯菊酯和氯氰菊酯对膜脂排列的影响,及研究北京菊酯和杀灭菊酯对膜脂结构的影响,本文用荧光法探讨了上述四种拟菊酯对二肉豆蔻酰磷脂酰胆碱  相似文献

本研究使用ATP特异性荧光共振能量转移(Fluorescence resonance energy transfer,FRET)为基础的荧光蛋白传感器(Ateam1.03-nD/nA),分析了4种外源信号分子(细胞外ATP、Ca2+、H2O2和NO)对拟南芥(Arabidopsis thaliana(L.)Heynh.)幼苗叶绿体和细胞质中ATP水平的影响。结果显示,细胞质ATP水平整体高于叶绿体,在4种不同浓度的信号分子处理下,叶绿体Ateam1.03-nD/nA的FRET比值仅在1.2 ~ 1.8波动;细胞质Ateam1.03-nD/nA 的FRET比值仅在2.2 ~ 3.0之间波动,未产生显著变化。结果表明在以上外源信号分子的作用下,植物细胞质和叶绿体ATP均维持在较为稳定的水平。  相似文献

Green leaf tissues contain relatively higher proportions of unsaturated fatty acids, especially α-linolenic acid, than do etiolated or senescent tissues. There appear to be developmental changes in the fatty acid composition of leaves during maturation and senescence. The normal rate of development of spinach (Spinacia oleracea L.) and bean (Phaseolus vulgaris L.) leaf tissues was altered by the application of kinetin and antimetabolites. Spinach was used for the kinetin studies and bean for the antimetabolite studies. Supposedly the kinetin retarded senescence and the antimetabolites retarded normal development. Special emphasis was placed on the incorporation of acetate into palmitate, the most abundant saturated fatty acid, and into linolenate, the most abundant unsaturated fatty acid. Kinetin does not enhance linolenate synthesis, but kinetin-treated tissues contain proportionately more linolenate. In contrast, tissues treated with antimetabolites contain proportionately less linolenate. Actinomycin-D and puromycin seem to have a greater effect on the synthesis of linolenate than on the synthesis of palmitate. Chloramphenicol does not have this same differential effect. The possible influence of antimetabolites on the synthesis of unsaturated fatty acids is discussed.  相似文献

COMPARISON OF THE FATTY ACIDS OF LIPIDS OF SUBCELLULAR BRAIN FRACTIONS   总被引:6,自引:3,他引:3  
Abstract— Rat brain grey and white matter were fractionated to yield myelin, nerve terminal, synaptic vesicle, nerve terminal 'ghost', and microsomal fractions of white and grey matter. Ester-type glycolipids were found in all fractions except myelin, while cerebrosides occurred in significant concentrations only in myelin and white microsomes. Comparison of the fatty acid profile of the ethanolamine- and serine-containing phospholipids showed marked differences between myelin and the particles from grey matter, while the microsomes of white matter were of intermediate composition. Docosahexaenoic acid, a minor acid in myelin, was a major fatty acid in microsomes of grey and white matter. The fatty acid composition of sphingomyelin was distinctly different in the fractions derived from grey and white matter, clustering about stearate and nervonate in the latter, but only about stearate in the grey. Marked differences in the positional distribution of fatty acids were seen within phosphatidyl choline from myelin and nerve terminals. Ribonucleic acid was found in nerve terminal and synaptic vesicle fractions. The sphingosine found in the ganglioside from microsomes of both grey and white matter was similar with respect to distribution of the C18 and C20 homologues.
The possibility is discussed that microsomes furnish characteristic lipids for the synthesis or renewal of specific membranes, and that these lipids are accumulated somewhat before being released.  相似文献

由于鱼油资源短缺, 植物油在水产饲料中广泛使用。然而, 随之而来的鱼体脂肪异常沉积等问题也日益突出, 严重危害养殖鱼类健康。脂肪的沉积是一个复杂的过程, 主要包括脂肪的合成、转运和分解。到目前为止, 在鱼类中已经进行了大量关于植物油替代鱼油影响脂肪沉积的研究。但是, 这些研究主要集中于脂肪的合成和分解, 有关脂肪转运的研究十分缺乏。脂肪转运不仅是影响组织脂肪沉积的重要因素, 而且在机体脂稳态和能量平衡中起着重要作用。因此综述了鱼类脂蛋白的种类和组成, 鱼类对脂肪和脂肪酸的转运, 营养因素对脂肪和脂肪酸转运的影响, 指出了脂类转运研究的重要性和紧迫性, 并且提出了未来需要努力的方向。  相似文献

Abstract— Three dietary levels of essential fatty acids, 30, 0-75 and 007 calorie-%, with a linoleic: linolenic acid ratio of 4:1, were fed to rats for two generations. In the third generation the weight of the cerebrum and the concentration of its lipids and the fatty acid composition of phosphoglycerides were determined from term to 120 days of age. The cerebral weights and the concentrations of phospholipids, cholesterol and cerebrosides differed only slightly between the three dietary groups. The accretion of fatty acids of the linoleic acid series was independent of the dietary essential fatty acid level while the accretion of fatty acids of the linolenic acid series was markedly reduced in the groups with low essential fatty acid supply. The sum of the total polyunsaturated fatty acids in ethanolamine phosphoglycerides differed only slightly between the groups. The proportion of the major polyunsaturated fatty acid of the linoleic acid series was equal between the groups while that of 22:6 (n-3) was much lower in the groups fed 007 calorie % essential fatty acids. In these latter groups the relative concentrations of 22:5 (n-6), 20:3 (n-9) and 22:3 (n-9) were increased. The differences in the fatty acid composition were dependent on the age of the rats. They were largest in newborn rats and diminished with age after weaning.  相似文献

本文报道了樟科11个属49种种子油和‘种果肉油的脂肪酸成分。其中樟族和木姜子族的各属种(滇新樟除外)种子油脂肪酸成分以 C_(10)、C_(12)和 C_(14)饱和或烯酸为主。而厚壳桂属、润楠和楠属种子油脂肪酸成分主要为棕榈酸、油酸和亚油酸。山苍子、天目木姜子、大果山胡椒、山橙、红脉钓樟、鸭公树和锈叶新木姜子种子油中的 C_(10)、C_(12)和 C_(14)烯酸经分离鉴定均是顺式Δ~4烯酸。  相似文献

Bacteriol. 84:1260-1267. 1962.-Variations in the temperature of growth and in the composition of the medium alter the proportions of individual fatty acids in the lipids of Escherichia coli. As the temperature of growth is lowered, the proportion of unsaturated fatty acids (hexadecenoic and octadecenoic acids) increases. The increase in content of unsaturated acids with a decrease in temperature of growth occurs in both minimal and complex media. Cells harvested in the stationary phase contained large amounts of cyclopropane fatty acids (methylenehexadecanoic and methylene octadecanoic acids) in comparison with cells harvested during exponential growth. Cells grown in a chemostat, limited by the concentration of ammonium salts, show a much higher content of saturated fatty acids (principally palmitic acid) than do cells harvested from an exponentially-growing batch culture in the same medium. Cells grown in a chemostat, limited by the concentration of glucose, show a slightly higher content of unsaturated fatty acids than cells from the corresponding batch culture. The results do not indicate a direct relation between fatty acid composition and minimal growth temperature.  相似文献
