Dormancy-associated gene expression in pea axillary buds.   总被引:10,自引:0,他引:10  
Pea (Pisum sativum L. cv. Alaska) axillary buds can be stimulated to cycle between dormant and growing states. Dormant buds synthesize unique proteins and are as metabolically active as growing buds. Two cDNAs, PsDRM1 and PsDRM2, were isolated from a dormant bud library. The deduced amino acid sequence of PsDRM1 (111 residues) is 75% identical to that of an auxin-repressed strawberry clone. PsDRM2 encodes a putative protein containing 129 residues, which includes 11 repeats of the sequence [G]-GGGY[H][N] (the bracketed residues may be absent). PsDRM2 is related to cold- and ABA-stimulated clones from alfalfa. Decapitating the terminal bud rapidly stimulates dormant axillary buds to begin growing. The abundance of PsDRM1 mRNA in axillary buds declines 20-fold within 6 h of decapitation; it quickly reaccumulates when buds become dormant again. The level of PsDRM2 mRNA is about three fold lower in growing buds than in dormant buds. Expression of PsDRM1 is enhanced in other non-growing organs (roots root apices; fully-elongated stems >elongating stems), and thus is an excellent “dormancy” marker. A mechanism involving transport, storage and retrieval of a symmetry-breaking message controls the relative growth rate of the cotyledonary buds of plantlets of Bidens pilosa L. The asymmetry was induced by administering a few needle pricks to one cotyledon of each plant. The storage of the symmetry-breaking message was independent of the number of pricks (all or nothing process) and irreversible. However, various treatments could render the plants either able to retrieve the stored symmetry-breaking message (in which case, the bud opposite to the pricked cotyledon began to elongate statistically sooner than the one associated with the stimulated cotyledon) or not (both buds then had an equal chance to be the first to start to grow). The retrieval process was also associated with a temporal oscillation. At the level of the whole plants, bud growth was observed only after the removal of apical dominance, and its degree of asymmetry was expressed by use of a parameter g ranging from zero (symmetrical case) to ± 1 (full asymmetry in favor of one of the cotyledonary buds). The highest g-values observed in the present contribution were of the order of 0.5. At the cellular level, the pricking of one cotyledon caused a number of cells, which were within the meristem of the bud associated with the pricked cotyledon and were in cell-cycle phases S or G2, to undergo cellular division and then be blocked in phase G1, whereas the cells of the opposite bud were practically unchanged.We thank Drs. A radioimmunoassay, combined with high-performance liquid chromatography, has been used to analyse the zeatin-type cytokinins of potato (Solanum tuberosum L. cv. Majestic) tubers and tuber buds throughout growth and storage. During tuber growth, zeatin riboside was the predominant cytokinin detected in all tissues. Immediately after harvest, the total cytokinin concentration fell dramatically in the storage tissue, largely as a consequence of the disappearance of zeatin riboside. During storage, levels of cytokinins in the storage tissue remained relatively constant, but increased in the tuber buds. In the buds of tubers stored at 2°C there was a 20-to 50-fold increase in total cytokinin over six weeks, coinciding with the natural break of innate dormancy. At 10°C the rise in the level of bud cytokinins was slower, correlating with the longer duration of innate dormancy. Injecting unlabelled cytokinins into tubers in amounts known to induce sprouting gave rise to increases in cytokinin concentrations in the buds of the same order as the increase associated with the natural break of dormancy. Metabolism of injected cytokinins was greater in non-dormant than in dormant tubers. Levels of free-abscisic acid and boundabscisic acid (alkaline hydrolyzable abscisic acidesters) in replicated samples of terminal vegetative buds of sycamore trees were measured during natural emergence from winter dormancy by gas chromatographic methods together with isotope dilution estimation of recovery rates. Not until after the buds had been released from true dormancy in January by winter chilling did any clear change occur in either abscisic acid (ABA) fraction, or in total ABA, on any basis of comparison. The percentage of total ABA present as the free acid declined at the end of true dormancy to approximately two-thirds of its value in the earlier winter months. It is concluded that glucosylation of ABA is unlikely to play a major part in the mechanism of release from dormancy in vegetative sycamore buds. At the end of true dormancy there was a large transient increase in what appeared to be phaseic acid, but this was not accompanied by any marked decrease in either free- or bound-ABA.Abbreviations ABA abscisic acid - TLC thin layer chromatography - GLC gas chromatography  相似文献

Polygalacturonase (PG, EC 3.2.1.15), an enzyme commonly found in ripening fruit, has also been shown to be associated with abscission. A zone-specific rise in PG activity accompanies the abscission of both leaves and flowers of tomato (Lycopersicon esculentum Mill.) plants. Studies of transgenic plants expressing an antisense RNA for fruit PG indicate that although the enzyme activity in transgenic fruit is < 1 % of that in untransformed fruit, the PG activity in the leaf abscission zone increases during separation to a similar value to that in untransformed plants. The timing and rate of leaf abscission in transgenic plants are unaffected by the introduction of the antisense gene. A polyclonal antibody raised against tomato fruit PG does not recognise the leaf abscission protein. Furthermore a complementary DNA (cDNA) clone (pTOM6), which has been demonstrated to code for fruit PG, does not hybridise to mRNA isolated from the abscission-zone region of tomato leaves. The direct double-antibody enzymelinked immunosorbent assay system was used in the detection and measurement of seed lectins from peanut (Arachis hypogaea L.) and soybean (Glycine max L.) plants (PSL and SBL, respectively) that had been inoculated with their respective rhizobia. Concentrations of PSL dropped to undetectable levels in peanut roots at 9 d and stems and leaves at 27 d after planting; SBL could no longer be detected in soybean roots at 9 d and in stems and leaves at 12 d. A lectin antigenically similar to PSL was first detected in root nodules of peanuts at 21 d reaching a maximum of 8 g/g at 29 d then decreasing to 2.5 g/g at 60 d. There was no evidence of a corresponding lectin in soybean nodules.Sugar haemagglutination inhibition tests with neuraminidase-treated human blood cells established that PSL and the peanut nodule lectin were both galactose/lactose-specific. Further tests with rabbit blood cells demonstrated a second mannosespecific lectin in peanut nodule extracts that was not detected in root extracts of four-week-old inoculated plants or six-week-old uninoculated plants, although six-week-old root extracts from inoculated plants showed weak lectin activity. The root extracts from both nodulated and uninoculated plants contained another peanut lectin that agglutinated rabbit but not human blood cells. A bundle of optical fibres and a photometer were used to measure light transmission through bud scales of Norway spruce [Picea abies (L.) Karst.] and Scots pine (Pinus silvestris L.). Percentage transmission increases with wavelength from none below 480 nm to 3% the far-red. Transmission of all wavebands measured was lowest in late summer and winter and highest in the autumn and spring. For most of the year in pine, and only from July to September in spruce, there is a depression of red light (660–670 nm) transmission. This affects the ratio of 660/730 nm light penetrating the apical domes. This ratio is lowest in July and August, at the time of initiation of female strobili. The seasonal variation in transmission of light by bud scales is discussed in relation to the possible consequences for the control of flowering.  相似文献

The systemic induction of expression of the gene for proteinase inhibitor II after wounding different parts of potato (Solanum tuberosum L.) plants was analysed at the RNA level. Wounding of either leaves or tubers led to an induction of expression of this gene in non-wounded upper and lower leaves as well as in the upper stem segment, whereas no expression was observed in nonwounded roots or in the lower stem segment. The signal mediating the systemic induction in nonwounded tissue must therefore be able to move both acropetally and basipetally. When the shoots of young hemp (Cannabis sativa L.) plants were cut off the roots, cultured as cuttings, and regenerating (adventitious) roots were removed as soon as appearing, ca. 80–90% of the plants became male (had staminate flowers) whereas if the roots were allowed to develop a similar percentage became female (pistillate flowers). Treatment of de-rooted cuttings with 6-benzylaminopurine (15 mg/l) restored the percent of female plants to ca. 80. It is suggested that the root system plays an essential role in sex expression in hemp and that this role is related to cytokinin synthesis in the root.  相似文献

Bud scales ofPicea abies (L.) Karst andFagus sylvatica L. selectively transmit the incident spectrum of electromagnetic radiation. It was shown that the scales absorb almost all radiation below 600 nm. Only the red and far-red light which is reduced to 0.01–2% reaches the apical domes. The center of a beech bud receives 0.11% and the center of a spruce bud 0.84% of the incident 300–2,000 nm light.  相似文献

Short-term applications of very high concentrations of 1-naphthaleneacetic acid (NAA) to expiants from flower stalks of tobacco (Nicotiana tabacum L. cv. Samsun) induced flower-bud regeneration to the same extent as longer or continuous incubation on lower concentrations. The maximum number of flower buds per explant after 15 d of culture was obtained not only by continuous culturing at 1 mol·l–1 NAA but also by 12 h of culturing at 22 mol·l–1 or 0.5 h at 220 mol· l–1, followed by incubation on medium without auxin for the remaining period. Continuous application of such high concentrations resulted in callus formation or caused the death of the explanted tissue. In all experiments in which auxin concentration and time of application were independently varied, the product of concentration and time determined the number of buds formed. Most, but not all, of the NAA taken up by the tissues was converted into conjugates. In expiants which had received a dose which was optimal for regeneration, the internal concentration of free NAA remaining beyond the pulse period was between 1.7 and 6.2 mol·l–1. Suboptimal applications led to lower values, supraoptimal treatments to much higher internal concentrations. Levels of endogenous growth substances (abscisic acid: ABA; indole-3-acetic acid: IAA) and applied benzyladenine (BA) were quantified during the eight first days of in vitro propagation of Wild Cherry (Prunus avium L.). Axillary buds from the middle part of the explants started to grow at day 2, thus were released from apical dominance. Hormone levels were measured in the apical, median and basal parts of the explants using an avidin-biotin based enzyme-linked immunosorbent assay (ELISA) after a purification of the extracts by high performance liquid chromatography (HPLC). All hormones showed rapid and considerable changes during the first eight days of growth. Exogenous IBA was probably transformed into IAA mainly in the basal part of the explant, and BA penetrated quickly. ABA levels were transiently enhanced in the apical part of the explants bearing young leaves. These phenomena are discussed in connection with the axillary bud reactivation.  相似文献

Potato (Solanum tuberosum L.) tuber buds normally remain dormant through the growing season until several weeks after harvest. In the cultivar Majestic, this innate dormancy persisted for 9 to 12 weeks in storage at 10° C, but only 3 to 4 weeks when the tubers were stored at 2° C. At certain stages, supplying cytokinins to tubers with innately dormant buds induced sprout growth within 2 d. The growth rate was comparable to that of buds whose innate dormancy had been lost naturally. Cytokinin-treatment did not accelerate the rates of cell division and cell expansion in buds whose innate dormancy had already broken naturally. Gibberellic acid did not induce sprout growth in buds with innate dormancy. The development of new strategies to enhance resistance of plants to pathogens is instrumental in preventing agricultural losses. Lesion mimic, the spontaneous formation of lesions resembling hypersensitive response lesions in the absence of a pathogen, is a dramatic phenotype occasionally induced upon expression of certain transgenes in plants. These transgenes simulate the presence of a pathogen and, therefore, activate the plant anti-pathogen defense mechanisms and induce a state of systemic resistance. Lesion mimic genes have been successfully used to enhance the resistance of a number of different plants to pathogen attack. However, constitutive expression of these genes in plants is associated with the spontaneous formation of lesions on leaves and stems, reduced growth, and lower yield. We tested the possibility of using a wound-inducible promoter to control the expression of bacterio-opsin (bO), a transgene that confers a lesion mimic phenotype in tobacco and tomato plants when constitutively expressed. We found that plants with inducible expression of bO did not develop spontaneous lesions. Nevertheless, under controlled laboratory conditions, they were found to be resistant to infection by pathogens. The activation of defense mechanisms by the bO gene was not constitutive, and occurred in response to wounding or pathogen infection. Furthermore, wounding of transgenic tobacco plants resulted in the induction of systemic resistance to pathogen attack within 48 h. Our findings provide a promising initial assessment for the use of wound-inducible promoters as a new strategy to enhance pathogen resistance in transgenic crops by means of lesion mimic genes.  相似文献

Tobacco (N. tabacum cv. Xanthi) cell lines contained two forms of anthranilate synthase (AS; EC 4.1.3.27) which could be partially separated by gel-filtration chromatography. One form was resistant to feedback inihibition by 10 M tryptophan (trp) while the other form was almost completely inhibited by trp at the same concentration. Cell lines selected as resistant to 5-methyltryptophan (5MT) had more of the trp-resistant AS form. Only the trp-sensitive form was detected in plants regenerated from both normal and 5MT-resistant cell lines. Overexpression of the trp-resistant form in 5MT-resistant tobacco cells disappeared during plant regeneration but reappeared when callus was initiated from the leaves of these plants. The trp-sensitive form was localized in the particulate fraction and the trp-resistant form in the cytosol of tobacco cultured cell protoplasts. The trp-resistant form of AS from tobacco had an estimated MW of 200 000, determined by Sephacryl S-200 chromatography, compared to an estimated MW of 150 000 for the trp-sensitive form. The estimated molecular weights of AS from carrot and corn were 160 000 and 150 000, respectively. Analysis of AS activity from the diploid Nicotiana species Nicotiana otophora (chromosome number 2n=24) by high-performance liquid chromatography showed two activity peaks identical in elution time and trp inhibition characteristics to the activity from N. tabacum (chromosome No. 48). Involvement of cytokinins (CKs) in axillary bud growth of miniature rose was studied. Variation in root formation and axillary bud growth was induced by two indole 3-butyric acid (IBA) pretreatments in two cutting sizes. At six physiological developmental stages around the onset of axillary bud growth, concentrations of CKs were determined in both root and axillary bud tissue by liquid chromatography combined with electrospray tandem mass spectrometry (LC-ESP-MS/MS). Chronological early onset of axillary bud growth occurred in long cuttings pretreated at low IBA concentration, whereas physiological early root formation was associated with long cuttings and high IBA concentration. The CKs zeatin (Z), isopentenyl adenine (iP), zeatin riboside (ZR), dihydrozeatin riboside (DHZR), isopentenyl adenosine (iPA), zeatin O-glucoside (ZOG), zeatin riboside O-glucoside (ZROG), zeatin riboside 5-monophosphate (ZRMP), and isopentenyl adenosine 5-monophosphate (iPAMP) were detected. Concentrations of CKs in axillary bud tissue far exceeded those in root tissue. Indole 3-butyric acid pretreatment influenced the concentration of CKs in axillary bud tissue more than did cutting size, whereas pretreatments only slightly affected CKs in root tissue. The dominant CKs found were iPAMP and ZR. An early and large increase in iPAMP indicated rapid CK biosynthesis in rootless cuttings, suggesting that green parts, including the axillary bud, can synthesize CKs. At the onset of axillary bud growth an increase in concentration of Z, ZR, ZRMP, ZOG, and ZROG was largely coincident with a decrease in iPAMP, iPA, iP, and DHZR. After the onset of axillary bud growth, CK content largely decreased. These results strongly indicate a positive role for CKs in axillary bud growth, and presumably ZRMP, ZR, and Z are active in miniature rose.  相似文献

The bud-inducing effect of the cytokinin N6-(2-isopentenyl)-adenine (i6-Ade) was examined in the moss Physcomitrella patens growing in liquid culture. Under these conditions, buds could be induced on chloronemata as well as on caulonemata. By application of i6-Ade, bud-formation was accelerated in both types of tissue. The number of buds, their size and their site of development were dependent on the concentration of the cytokinin in the range of 10-7 M to 10-5 M. Moreover, the percentage of caulonema cells increased with a cytokinin concentration of 10-5 M. These results indicate that chloronema cells may also function as target cells for exogenous cytokinins. The composition of proteins from caulonemata and chloronemata of two different species (P. patens and Funaria hygrometrica), grown on solid medium were compared.
