Reduviidae and some other groups of cimicomorphan Heteroptera possess a hairy attachment structure on the apex of the tibia called “fossula spongiosa”. The fossula spongiosa was never studied comparatively across Reduviidae, its fine structure and mode of function is not well documented, and attachment structures in immature stages are virtually unknown. Here, a sample of 171 species of Reduviidae representing 22 subfamilies is examined for presence-absence of the fossula spongiosa on the three pairs of legs. Representatives of 11 of the 22 subfamilies are shown to possess a fossula spongiosa. The fine structure of the fossula spongiosa is examined for a more limited sample of Reduviidae and several Pachynomidae and Nabidae. In addition, scanning micrographs for the fossula spongiosa in other Cimicomorpha are given, among them Anthocoridae, Cimicidae, Microphysidae (first record of a fossula spongiosa), and Thaumastocoridae. The fossula spongiosa in Reduviidae consists of tenent hairs (acanthae) with spatulate or tapering apices interspersed with sensory setae, both of which are embedded in a thick and flexible cuticle, underlain by a hemolymph cavity separated almost entirely from the interior of the remaining tibia by a cuticular invagination. Judging from comparison with non-reduviid Cimicomorpha, this separation of the fossula spongiosa cavity from the tibial interior may be unique to Reduviidae. A simple experiment using live specimens of Platymeris biguttata (Reduviinae) revealed a liquid on the tip of the tenent hairs that might be involved in the attachment of the fossula spongiosa by adhesion mechanisms. The nymphs of Reduviidae whose adults have a fossula spongiosa are here documented for the first time to possess pads of ventrally barbed setae instead of tenent hairs and their tibia lacks the internal cuticular invagination. The nymphal attachment structures seem to rely on increase of friction rather than the adhesion mechanism proposed to be present in the adult. Combined with the tenent setae on the third tarsomere known in some Emesinae and here documented for Saicinae, three types of hairy attachment structures occur on the legs of Reduviidae: tenent hairs (acanthae), which form the fossula spongiosa in many Reduviidae, barbed setae that substitute the fossula in the immatures, and tenent setae on the tarsus which are restricted to only a few taxa.  相似文献

The trichome in ant-feeding Holoptilinae (Insecta: Heteroptera: Reduviidae) comprises remarkable modifications of abdominal sternites 2 and 3. It has been hypothesized that this structure plays a role in attracting and drugging ants. In the present study the trichome of 14 species of Holoptilini, comprising 11 species of Ptilocnemus Westwood and representatives of three additional genera of Holoptilini, is examined using scanning electron and light microscopy. The astoundingly diverse species-level modifications of sternites and vestiture are described and primary homology hypotheses are proposed. The trichome provides species-specific diagnostic characters within Ptilocnemus and evidence for species-groups within the genus, but also for the sistergroup relationship of Ptilocnemus and Smiliopus Bergroth. The comparative morphology establishes a framework for investigations into systematics, functional morphology, and behavioral ecology of these myrmecophagous assassin bugs.  相似文献

Glutathione peroxidase (GPX) activity was measured in several tissues of the blood‐sucking bug, Rhodnius prolixus. In contrast to the pattern found in vertebrates, where GPX is predominantly intracellular, the highest levels of this enzyme in Rhodnius were found in the hemolymph. The hemolymph glutathione‐dependent peroxidase accepted both H2O2 and t‐butyl hydroperoxide as substrates. This fact, together with the absolute glutathione dependence, inhibition by mercaptosuccinate, insensitivity to cyanide, and a molecular mass (100.7 kDa) similar to vertebrate GPXs, led us to attribute this peroxidatic activity to a Se‐dependent enzyme. Hemolymph GPX specific activity increases during development and a twofold stimulation was observed after an oxidative challenge with hemin, suggesting that enzyme synthesis is under regulatory control. A role for extracellular GPX as an antioxidant protection against oxidative damage produced by heme derived from digestion of blood hemoglobin is discussed. Arch. Insect Biochem. Aim The tribe Rhodniini is one of six comprising the subfamily Triatominae (Heteroptera: Reduviidae), notorious as blood‐sucking household pests and vectors of Trypanosoma cruzi throughout Latin America. The human and economic cost of this disease in the American tropics is considerable, and these bugs are unquestionably of great importance to man. Studies of the evolution, phylogeny, biogeography, ecology, physiology and behaviour of the Rhodniini are needed to help improve existing Chagas’ disease control programmes. The objective of the study reported here was to propose biogeographical hypotheses to explain the modern geographical distribution of the species of Rhodniini. Location Neotropical region. Methods We employed mitochondrial rDNA sequences (16S) currently available in GenBank to align sequences of Rhodniini species using ClustalX. The analyses included 16S sequences from predatory reduviid subfamilies (Stenopodainae, Ectrichodiinae, Harpactorinae, Reduviinae and Salyavatinae) present in GenBank as an outgroup. Cladistic analysis used the program PAUP to derive trees based on maximum parsimony (MP) and maximum likelihood (ML). Known distribution data for Rhodniini species were obtained from reviews and plotted on maps of South and Central America using the program iMap. An area cladogram was derived from the cladistic result to show the historical connections among the studied taxa and the endemic areas. The program TreeMap (Jungle Edition) was used to deduce taxon–area associations where the optimal solutions to explain the biogeographical hypothesis of the Rhodniini in the Neotropics were those with lowest total cost. Results Parsimony and maximum‐likelihood analysis of 16S rDNA sequences included 14 species of Rhodniini, as well as five species of predatory Reduviidae representing five of the predatory subfamilies. Tanglegrams were used to show the relationship between the Neotropical areas of endemism and Rhodniini species. When TreeMap with codivergence (vicariance) events were weighted as 0 and duplication (sympatry), lineage losses (extinction) and host switching (dispersal) were all weighted as 1, 20 scenarios were found to explain the biogeographical history of Rhodniini in the Neotropical region. Twelve of the optimal solutions with the lowest total cost were used to explain the biogeography of the Rhodniini in the Neotropics. These optimal reconstructions require six vicariance events, 20 duplications (sympatry), at least three dispersals, and at least one extinction event. Main conclusions The Rhodniini have a complex biogeographical history that has involved vicariance, duplications (sympatry), dispersal and extinction events. The main geological events affecting the origin and diversification of the Rhodniini in the Neotropics were (1) uplift of the Central Andes in the Miocene or later, (2) break‐up of the Andes into three separate cordilleras (Eastern, Central and Western) in the Plio‐Pleistocene, (3) formation of a land corridor connecting South and North America in the Pliocene, and (4) uplift of the Serra do Mar and Serra da Mantiqueira mountain systems between the Oligocene and Pleistocene. The relationships and biogeographical history of the species of Rhodniini in the Neotropical region probably arose from the areas of endemism shown in our work.  相似文献

The uptake of RHBP (Rhodnius heme-binding protein) by the ovaries of Rhodnius prolixus was characterized. RHBP purified from oocyte was labeled with 125I and used to study the process of uptake by the ovary in vivo and in vitro. After injection, the [125I]RHBP was readily removed from the hemolymph and accumulated especially in the ovary. The capacity of the ovary to take up [125I]RHBP from the hemolymph varied during the days following blood meal. It increased up to day 2, remained stable until day 5, and then decreased up to the end of oogenesis. In vitro, the uptake of [125I]RHBP was linear at least up to 60 min. The uptake was dependent on [125I]RHBP concentration and showed to be a saturable process. The addition of a molar excess of non-related proteins such as Vitellin (Vt), Lipophorin (Lp), and Bovine Serum Albumin (BSA) did not reduce [125I]RHBP uptake. Using immunogold technique the RHBP was localized at the microvilli, coated pits, and yolk granules. The main yolk protein, Vt, did not compete with RHBP for the uptake. Thus, it is discussed here that they bind to independent binding sites of the oocytes, and are directed later on to the same compartment. The need of both proteins for the completion of mature oocyte was verified in vivo. The reduction of heme-RHBP in the hemolymph, by changing the diet, decreased the number of eggs laid. Increasing the concentration of heme-RHBP in the hemolymph, the number of eggs produced increased in a dose dependent manner. In vitro, both apo-RHBP and heme-RHBP can be taken up by the oocyte. We analysed pretarsal characters of 87 species of Leiodidae (including 10 cholevines and representatives of all tribes and ca. 60% of the genera of non‐cholevines), five species of Agyrtidae, and nine representatives of outgroup taxa (Hydraenidae, Staphylinidae, Hydrophilidae, and Histeridae) using scanning electron microscopy. We focused our observations on the architecture of the empodium (including the sclerites and associated setae), the shape and composition of the medial projection of the distal margin of the terminal tarsomere, and the armature of the claws, which were considered a promising source of information for delimiting supraspecific taxa in our previous study. We identified several diagnostic features and recognize potential synapomorphies at the tribal, subtribal and generic levels. The internal systematic arrangement and/or even the monophyletic status of most of the subfamilies of Leiodidae (Camiarinae, Catopocerinae, Leiodinae, and Platypsyllinae) are challenged. We identified potential synapomorphies for Camiarinae (Camiarini and Agyrtodini) and Leiodinae. The non‐monophyly of Cholevinae is possible because part of the tribe (Anemadini, Eucatopini, and Oritocatopini) shares potentially apomorphic features with Leiodinae (e.g., a triangular medial projection with a diagonal row of conical spines), whereas another part (Leptodirini and Ptomaphagini) shares a potentially apomorphic feature with Coloninae and Platypsyllinae (a typical medial projection with two distinct triangular projections).  相似文献

We investigated whether metasternal gland (MG) products mediate male aggregation during mating in Rhodnius prolixus (Heteroptera: Reduviidae) Stål, 1859, through selective occlusion of female MGs. Pairs of insects presenting intact MGs mated and other males aggregated around them confirming conclusions from previous studies. However, when female MGs were occluded, males did not aggregate. Conversely, the occlusion of male MGs did not affect aggregation of males. The present experiments demonstrated that the products emitted by the MGs of a mating R. prolixus female induce other males to aggregate around the mating pair. Aggregation behaviour of males probably results from the eventual release of volatiles from the female's MGs. We reinforce the hypothesis suggesting that the compounds produced by the MGs of R. prolixus act as sexual communication signals for this species.  相似文献

The yolk platelets from Rhodnius prolixus, a blood-sucking bug, are composed mostly of vitellin and here are shown to contain at least two hydrolytic enzymes, a phosphatase and a cathepsin D-like proteinase. Both the proteinase and the phosphatase have an acid pH optimum. No hydrolytic activity was observed under alkaline or neutral conditions. Among several proteinase inhibitors tested, only pepstatin could abolish vitellin breakdown in vitro. The proteinase appears to be bound to the yolk platelet membranes. The phosphatase activity, using p-nitrophenyl phosphate as substrate, was enhanced after disruption of the platelet membrane by Triton X-100. This activity could be inhibited by tartrate but not by p-cloromercuribenzoate.  相似文献

Triatomines are of great concern in public health because they are vectors of Chagas' disease. This study presents an analysis of the species Triatoma melanosoma. The cytogenetic characteristics of triatomines include holocentric chromosomes, post-reductional meiosis in the sex chromosomes and nucleolar fragmentation in the meiotic cycle. The methodology utilized consisted of the techniques of lacto-acetic orcein staining and silver ion impregnation. The organs analyzed were adult testicles. The results enabled to classify the chromosomes by number and size, being three large, eight medium and one small heterochromosome. The three largest chromosomes and the heterochromosomes showed heteropyknotic chromatin in meiosis. The heterochromosomes in 8.05% of the cells in metaphase I behaved as pseudobivalents, contrasting with 91.95% of the cells with individualized sex chromosomes, confirming the achiasmatic nature of these chromosomes. However, the pseudobivalents occurred prominently in metaphase II (78.38%), this fact probably is related to the post-reductional nature of the sex chromosomes. The nucleolus in T. melanosoma persisted until the diplotene phase after which it began to fragment. Nucleolar corpuscles were observed in metaphases I and II and during anaphases I and II, these characteristics being related to the phenomenon of nucleolar persistence. In the initial spermatids, peripheral silver ion impregnation occurred, which could be analogous to the pre-nucleolar corpuscles observed after fragmentation. Thus, this study extends our knowledge of the characteristics of triatomines, in particular, heteropyknotic degree, kinetic activity, formation of sex chromosome achiasmatic pseudobivalency, confirmation of the fragmentation phenomenon, and post-meiotic nucleolar reactivation.  相似文献

The objective of this work was to study the pattern of salivation of triatomines during feeding in mouse skin. Rhodnius prolixus was fed with a solution of the dye acridine orange or fluorescein. The saliva was efficiently labelled with acridine orange, probably due to the difference in pH between the salivary gland (6.0) and the hemolymph (6.5-7.0). This procedure was not effective at labelling the saliva of Triatoma infestans, however, fluorescent labelling of R. prolixus saliva allowed us to demonstrate that salivation occurs during entire feeding process. The saliva is released soon after the bite. In the probing phase, saliva is pumped continuously in the host skin, including around the blood vessels. During the engorgement phase, saliva is observed in a bolus within the blood vessel and some of it is sucked up by the insect, together with blood. The frequency of saliva emission inside the vessels was low (0.51+/-0.18 Hz). The saliva deposition in the microcirculation is continuous and modulated by the frequency of the cibarial pump because, when functioning at high frequency, cibarial pump sucks almost all saliva to the insect gut. This mechanism would determine the quantity of saliva deposited in the microcirculation as necessary, and consequently minimizing the host's immune response to salivary antigens.  相似文献

Plastron and spiracular features strongly resemble those of Aphelocheirus aestivalis (Fab.), described by Messner et al. (1981). Plastronal foliose hairs, somewhat irregular shaped are regularly distributed in the plastron. Very numerous tiny openigs on sternites an tergites connect platronal air with a subcuticular air film limited internally apparently by a membrane. Aplicated, radial, structure at the proximal end of the tracheae, almost occluding the extremely fine spiracular openings, appear with lesser variations in both species, suggesting close relationships than accepted until now.  相似文献

N,N-diethyl-3-methylbenzamide (DEET) is widely used as an insect repellent; however, little is known about its mode of action. On the other hand, nitric oxide (NO) participates in the olfaction transduction pathway of insects. In this work, nitroso-acetyl-cysteine (SNAC), a nitric oxide donor, or dibutyril-cyclic-GMP (db-cGMP), the cyclic nucleotide analog, were applied on fifth instar nymphs of Rhodnius prolixus before exposing them to DEET, to obtain information about the possible role of NO/cGMP system in the olfaction process. In the first place, we exposed the nymphs to several DEET concentrations (70, 700, 1,750, and 3,500 microg/cm2). All these concentrations produced a repellent effect. A decrease in repellency during the course of the experiment was observed when the nymphs were exposed to high concentrations of DEET (700 and 1,750 microg/cm2), suggesting an adaptation phenomenon. The pre-treatment of the insects with 15 microg /insect of SNAC or 2 microg/insect of db-cGMP produced a reduction of the repellency. An increase in locomotor activity was observed in insects exposed to 350 or 700 microg/cm2 DEET. Although exposure to 70 microg/cm2 DEET produced a high repellency response, it did not modify the insects' locomotor activity. Insects treated with two doses of SNAC before being exposed to 350 microg/cm2 of DEET showed no differences in locomotor activity compared to controls.  相似文献

Apiomerus, the charismatic bee-assassins (>108 spp.), belong to the New World resin bugs in the harpactorine tribe Apiomerini (12 extant genera) that is characterized by a novel predation strategy, resin trap predation. Apiomerini also exhibit striking genitalic diversity that has shaped subgeneric classifications within the genus Apiomerus and females of some species of Apiomerus are known to engage in unique maternal care behaviors. The lack of a phylogenetic framework currently hinders evolutionary interpretations of genitalic morphology and maternal care. We here present a molecular phylogeny based on 4, 477 bp of six ribosomal and protein coding genes and 95 terminal taxa using parsimony and maximum likelihood approaches as a way of addressing these shortcomings. Apiomerini are monophyletic, with Heniartes being the sistergroup to all remaining taxa that form the monophyletic Manicocoris (Calliclopius, Manicocoris, Micrauchenus, and Ponerobia) and Apiomerus (Agriocoris, Apiomerus, and Sphodrolestes) clades. Previously proposed subgeneric groups are polyphyletic, but several proposed species groups are recovered as monophyletic. Ancestral state reconstruction of the metatibial comb indicates that this structure evolved in the ancestor of all Apiomerini where it was present in males and in females; it became strongly sexually dimorphic (better developed in females than in males) in the Apiomerus clade (Apiomerus + Agriocoris + Sphodrolestes). Genitalic features reveal a pattern of homoplasy, but frequently are nonetheless useful to diagnose supraspecific groups within Apiomerus. The complex genitalia found within Apiomerus are derived for that clade. We conclude that, using the metatibial comb as a proxy, maternal care is relatively common in the tribe Apiomerini and propose that it likely evolved at the base of the Apiomerus clade if not at the base of Apiomerini.  相似文献

Abstract.  With more than 6600 species worldwide, Reduviidae (Insecta: Heteroptera), or assassin bugs, form the second largest and one of the most diverse groups of true bugs. The poor condition of the higher-level classification of Reduviidae is reflected by the facts that different authors recognize between 21 and 32 subfamily-level names and that Reduviidae were never subjected to a rigorous cladistic analysis using an exemplar approach. In the present study, a cladistic analysis of higher-level taxa of Reduviidae based on 162 morphological characters and 75 ingroup and outgroup species is presented. Twenty-one subfamily-level taxa of Reduviidae were examined, accounting for 28 tribes. In addition to characters previously used for diagnosis in Reduviidae, information on recently published character complexes is used in the present analysis, supplemented with new character information gathered specifically for this project. Reduviidae are supported as a monophyletic group with Pachynomidae as their sister taxon. The major results of this study are the support of a sistergroup relationship of Hammacerinae with the remaining Reduviidae, the monophyly of the Phymatine Complex, the relatively basal position of Harpactorinae within Reduviidae as well as a novel hypothesis on the relationships within this group, and the sistergroup relationship of Ectrichodiinae + Tribelocephalinae and their placement in a clade that also contains Emesinae, Saicinae, and Visayanocorinae. The analysis further supports a clade formed by paraphyletic Salyavatinae + Sphaeridopinae, renders Vesciinae non-monophyletic, and demonstrates the polyphyly of Reduviinae. Pseudocetherinae are shown to group with some Reduviinae. Triatominae are supported as a monophyletic group and are nested among additional Reduviinae and Stenopodainae.  相似文献

Here we investigated H2O2 production and detoxification in the hematophagous hemiptera, Rhodnius prolixus. Superoxide dismutase (SOD) catalyzes the dismutation of superoxide radical (O2-). This reaction produces hydrogen peroxide, which is scavenged by antioxidant enzymes such as catalase (CAT). SOD and CAT activities were found in all tissues studied, being highest in the midgut. CAT was dose-dependently inhibited in vivo by injections of 3-amino-1,2,4-triazole (AT). Insects treated with AT showed a twofold increase in H2O2 levels. Injection of DL-buthionine-[S, R]-sulfoximine (BSO), an inhibitor of glutathione synthesis, also resulted in a fourfold increase in H2O2, together with stimulation of CAT activity. Simultaneous administration of both AT and BSO had a synergistic effect on midgut H2O2 content. Taken all together, our results suggest that CAT and glutathione-dependent mechanisms cooperate to control H2O2 concentration in the midgut cell and prevent hydroxyl radical generation by Fenton reaction in this tissue.  相似文献
