Na+-K+-Cl- cotransport and Na+/K+ATPase were studied by immunohistochemistry in the kidney and urinary bladder of Trematomus bernacchii and Chionodraco hamatus. The activity was correlated to the density of mitochondria. The first segment of the renal proximal tubule was more active than the second one. In T. bernacchii and the temperate marine teleost Pagellus bogaraveo, the immunoreactivity for the antibody to cotransporters and to the !-subunit of the sodium pump was stronger than in the icefish. This difference indicates in the kidney of the icefish, a weaker secretory activity, a consequent lower osmolarity in the lumen and lower water loss, which correlates well with the need for a greater blood volume in the icefish. The epithelium of the urinary bladder in T. bernacchii, where intense immunostaining was observed, was composed of columnar cells. In C. hamatus the columnar cells, where the immunostaining was weaker, lined only a portion of the urinary bladder, the other region being composed of cuboidal cells.  相似文献

The main characteristic of Antarctic seawater is its low constant temperature and its high concentration of O(2), which can increase the formation rate of reactive oxygen species (ROS), together with a natural occurrence of elevated cadmium and copper levels. In the present paper, we studied the presence of cadmium, copper and zinc, metallothioneins (MTs) and glutathione (GSH), and antioxidant enzyme activities in the Antarctic teleosts Trematomus bernacchii and Trematomus newnesi, in order to determine the influence of the peculiar physico-chemical features of the Antarctic marine environment on these physiological defence systems in two species of teleosts. In both of them, cadmium and copper accumulation occurs mostly in the liver. T. bernacchii accumulates zinc mostly in the hepatic tissue, whereas T. newnesi does not show a preferential accumulation site. In addition to the intra-specific analysis, we decided to compare the two species of the Trematomus genus in order to verify if the different feeding habits and motility of these fish affects metal accumulation. Our results show that the liver of T. bernacchii accumulates cadmium and zinc at a higher extent with respect to T. newnesi. Glutathione (GSH) and metallothioneins (MTs) are present in great quantity in the liver of both species. Moreover liver is the tissue which generally showed the highest antioxidant enzyme levels. The results provide further insights in the physiological mechanisms evolved by animals living in this extreme environment.  相似文献

In this study, some of the mammalian kallikrein-kinin system (KKS)-like components were identified in two species of Antarctic notothenioid [Chionodraco hamatus (Channichthydae) and Trematomus bernacchii (Nothothenidae)]. The kidney and heart were assayed for kallikrein-like activity using the synthetic substrate d-Val-Leu-Arg-paranitroanilide. Values expressed as nmol p-nitroanilide/mg proteins, were in C. hamatus 15.5±1.5 and 15.2±1.4 in kidney and heart, respectively, and 15.8±2.2 and 15.9±1 in kidney and heart of T. bernacchii. Kallikrein-like activity was inhibited bvy aprotinin and phenylmethylsulfonyl fluoride (PMSF). The assay was stable at 20°C. Kininase II-like activity was performed on kidney, gills and heart using the substrate hippuryl-lhistidyl-l-leucine. The activity was inhibited by captopril, and in kidney and gills by high temperatures (20°C and 37°C); in the heart the enzymatic activity was measurable also at 20°C. Bradykinin-like immunoreactive cells were localized by immunohistochemistry in the nephron, in the gills, and in the arterial walls of the heart. These results show that Antarctic teleosts possess elements comparable to those of the KKS, including kallikrein-like, and kininase II-like activities, and the end product of the enzymatic cascade, bradykinin. The composition of tryptic peptides was determined for Hb (major or fast electrophoretic component) from Clethrionomys rutilus and from Ondatra zibethicus. Analysis of the separated and β-Hb chains made possible the definition of theT9 and βT12 peptides and the resolution of uncertainties in earlier Microtus data for peptides T9,T12b,βT9,βT12 and βT13. The previous postulate of two deletions in peptide βT5 is supported by the present data, and the independent dipeptide βT6 appears in the Ondatra Hb. A Microtus stem sequence is inferred and used for comparison with the new data, and cladograms are given to accomodate the extended phylogenetic progression.  相似文献

We investigated the presence of nitric oxide synthase (NOS) in brain of adult Antarctic teleosts by indirect immunofluorescence technique using a synthetic rat neuronal NOS (nNOS) antibody. The following species were examined: Trematomus bernacchii, Gymnodraco acuticeps, Histiodraco velifer, Cygnodraco mawsoni (haemoglobin-rich), Chionodraco hamatus and Pagetopsis macropterus (haemoglobin-free). Immunoreactive cell bodies were localized in dorsal telencephalon, in hypothalamus, in optic tectum of the mesencephalon as well as in Purkinje cells of the cerebellum. No differences were observed in the localization of the nNOS immunopositivity in the Antarctic teleosts brains examined and NOS distribution was similar to that described in other teleosts, suggesting that nitric oxide (NO) may also function as a neurotransmitter in the brain of Antarctic teleosts. A strong immunopositivity was observed in the cerebral blood vessels of the icefishes suggesting that NO may play a pivotal role in the regulation of the cerebral blood flow especially in these haemoglobin-free species.  相似文献

Regulatory peptides are short chains of amino acids that regulate cell-to-cell interactions in widely divergent animal groups. Evidence is accumulating to suggest that they mediate many aspects of physiology and behaviour, serving as neurotransmitters, neuromodulators and hormones. While most data in this field derive from studies on the mammalian nervous and endocrine systems, the last decade has witnessed an upsurge of interest in invertebrate peptide biology, not least because it is likely that many regulatory peptides originated in the nervous system of invertebrates. Platyhelminths, like other invertebrate groups investigated, contain numerous neuropeptides, and here David Halton and colleagues review the evidence that these putative signalling agents serve key roles in parasite motility, reproduction and morphogenesis. The physicochemical differences between host and parasite peptides raise the possibility that selective disruption of peptidergic control systems in parasites could be an exploitable target in future chemotherapeutic strategies.  相似文献

Metal bioaccumulation and metallothionein were investigated in different organs of the red-blooded teleost, Trematomus bernacchii and the haemoglobinless Chionodraco hamatus. Specimens of the two Antarctic fish were sampled from Terra Nova Bay (Ross Sea), and their levels of Cd, Cu and Zn in homogenates and in soluble fractions of liver, muscle, gills, heart and plasma were determined. Dosages of metallothioneins (MTs) or MT-like proteins were assayed by the silver saturation method in soluble fractions of the same organs. In both T. bernacchii and C. hamatus the highest MT contents were found in liver. Hepatic MT and Cd, Cu and Zn concentrations correlated positively in T. bernacchii, whereas in C. hamatus hepatic MT showed a positive correlation only with Cd. Positive correlations were also found between Cd and MT in gills of the two species. A metal-binding protein containing a high percentage of cysteine from C. hamatus was purified and compared with the MT from T. bernacchii and mammals. Accepted: 27 November 1999  相似文献

The presence and the comparative distribution of regulatory peptides and serotonin in the gut of four species of Antarctic notothenioid fishes [Cryodraco antarcticus and Chionodraco hamatus (Channichthyidae), and Trematomus bernacchii and Trematomus newnesi (Nototheniidae)], were immunohistochemically studied. In these species, numerous immunoreactive (IR) endocrine cells and nerve elements were detected. In the nototheniids most of the IR nerve fibres were of the intrinsic type, while most of the IR nerve fibres of the channichthyid intestine, besides insulin-like IR fibres, seemed to be of the extrinsic type. The intensity and frequency of immunopositivity are not the same in channnichthyids and nototheniids; the species belonging to the first family show many differences from the teleosts living in temperate water. The finding of insulin-like endocrine cells and nerve fibres in the gut wall of Cryodraco antarcticus is exceptional for vertebrates and deserves special attention.A preliminary account of this work was presented to the VII International Ichthyology Congress, Den Haag, 26–30 August 1991  相似文献

The patterns of colocalization of somatostatin (SOM), neuropeptide Y (NPY) and the catecholamine-synthesizing enzyme, dopamine beta-hydroxylase (DBH), were examined in intramural neurones in dissociated cell culture preparations from the detrusor muscle of the urinary bladder of the newborn guinea-pig using an elution-restaining immunocytochemical technique. Large numbers of the intramural neurones contained NPY-like (70-85% of the total neuronal population) and SOM-like (60-75%) immunoreactivities, in contrast to a small population (1-6%) of neurones containing immunoreactivity to DBH. Some neurones were immunoreactive to NPY (15-20%) and SOM (5-10%) alone, while 55-70% of the total neuronal population showed immunoreactivity to both NPY and SOM. NPY-like immunoreactive neuronal cell bodies that did not contain SOM were predominantly binucleate, whereas neuronal cell bodies immunoreactive to SOM alone were mainly mononucleate. Although not seen in every culture preparation, neuronal cell bodies containing both NPY-like and DBH-like immunoreactivities were also observed (less than 5% of the total neuronal population), and most, if not all, of these neuronal cell bodies were binucleate. SOM-like and DBH-like immunoreactivities were not seen in the same neuronal cell body throughout this study. These results show that intramural bladder neurones can be divided into distinct subpopulations based upon the coexistence of specific peptides and enzymes, and the possibility that they sustain local integrative and modulatory roles in bladder function is discussed.  相似文献

Regulatory peptides were immunolocalized in the midgut of the fruit fly Drosophila melanogaster. Endocrine cells were found to produce six different peptides: allatostatins A, B and C, neuropeptide F, diuretic hormone 31, and the tachykinins. Small neuropeptide-F (sNPF) was found in neurons in the hypocerebral ganglion innervating the anterior midgut, whereas pigment-dispersing factor was found in nerves on the most posterior part of the posterior midgut. Neuropeptide-F (NPF)-producing endocrine cells were located in the anterior and middle midgut and in the very first part of the posterior midgut. All NPF endocrine cells also produced tachykinins. Endocrine cells containing diuretic hormone 31 were found in the caudal half of the posterior midgut; these cells also produced tachykinins. Other endocrine cells produced exclusively tachykinins in the anterior and posterior extemities of the midgut. Allatostatin-immunoreactive endocrine cells were present throughout the midgut. Those in the caudal half of the posterior midgut produced allatostatins A, whereas those in the anterior, middle, and first half of the posterior midgut produced allatostatin C. In the middle of the posterior midgut, some endocrine cells produced both allatostatins A and C. Allatostatin-C-immunoreactive endocrine cells were particularly prominent in the first half of the posterior midgut. Allatostatin B/MIP-immunoreactive cells were not consistently found and, when present, were only weakly immunoreactive, forming a subgroup of the allatostatin-C-immunoreactive cells in the posterior midgut. Previous work on Drosophila and other insect species suggested that (FM)RFamide-immunoreactive endocrine cells in the insect midgut could produce NPF, sNPF, myosuppressin, and/or sulfakinins. Using a combination of specific antisera to these peptides and transgenic fly models, we showed that the endocrine cells in the adult Drosophila midgut produced exclusively NPF. Although the Drosophila insulin gene Ilp3 was abundantly expressed in the midgut, Ilp3 was not expressed in endocrine cells, but in midgut muscle.  相似文献

Two new genera, Reductosoma gen.n. (Desmosomatidae) and Desmostylis gen.n. (Desmosomatidae), and five new species are described from the Maud Rise and the Gunnerus Ridgc, Antarctica. Two new species of the Desmosomatidae (Eugerdellatinae and Desmosomatinae) are described: Reduetosoma gunnera gen. et sp.n. and Desmosoma hesslera sp.n.; one species of the Munnopsidae (Eurycopinae), Disconectes colernuni sp.n.; and two species of the Macrostylidae from the Antarctic deep sea, Desmostylis obscurus gen. et sp.n. and Macrostylis sarsi sp.n. Summary Immunohistochemistry was used to localize regulatory peptides in endocrine cells and nerve fibres in the pancreas of two species of elasmobranchs (starry ray,Raja radiata and spiny dogfish,Squalus acanthias), and in the Brockmann bodies of four teleost species (goldfish,Carassius auratus, brown troutSalmo trutta, rainbow trout,Oncorhynchus mykiss and cod,Gadus morhua). In the elasmobranchs, the classical pancreatic hormones somatostatin, glucagon and insulin were present in endocrine cells of the islets. In addition, endocrine cells were labelled with antisera to enkephalins, FMRF-amide, gastrin/cholecystokinin-(CCK)/caerulein, neurotensin, neuropeptide Y (NPY), and peptide YY (PYY). Nerve fibres were demonstrated with antisera against bombesin, galanin and vasoactive intestinal polypeptide (VIP). These nerve fibres innervated the walls of blood vessels, in the exocrine as well as the endocrine tissue. In the four teleost species immunoreactivity to somatostatin, insulin and glucagon was intense in the Brockmann bodies. Cells were labelled with antisera to enkephalin, neurotensin, FMRFamide, gastrin/CCK/ caerulein, NPY, PYY and VIP. Only a few nerve fibres were found with antisera against dopamine--hydroxylase (DBH, cod), enkephalin (met-enkephalin-Arg-Phe, cod), bombesin (cod), gastrin/CCK/caerulein (cod) and VIP. Galanin-like-immunoreactive fibres were numerous in the Brockmann bodies of all teleosts examined. Immunoreactivity to calcitonin gene-related peptide (CGRP), substance P, tyrosine hydroxylase (TH), and phenyl-N-methyl transferase (PNMT) could not be found in any of the species studied.  相似文献

Summary The gastrointestinal tract of the alligator Alligator mississipiensis has been investigated for the presence of immunoreactivity to fourteen regulatory peptides all known to occur in the mammalian gut system.Mucosal endocrine cells reacting specifically with the antisera to neurotensin, C-terminal gastrin, somatostatin, bombesin, secretin, pancreatic glucagon and enteroglucagon were detectable, the distribution of these cells being, in general, similar to the mammalian pattern. Peripheral nerve cell bodies and nerve fibres were detected with the antisera to vasoactive intestinal polypeptide, substance P, bombesin and somatostatin again with a distribution similar to that seen in mammals.No immunoreactivity was observed with the available antisera to glicentin, motilin, gastric inhibitory polypeptide, gastrin 34, cholecystokinin 9–20 and met-enkephalin.  相似文献

Blood chemistry and haematological parameters have been determined in the Antarctic teleosts, Notothenia neglecta Nybelin and Notothenia rossii Richardson at 2° C. Samples were taken using chronically implanted dorsal aortic cannulae following a minimum of 24–36 h recovery. Broadly similar results were obtained for the two species. In N. neglecta , routinely active specimens had high values of arterial pH (7.81) and PO 2 (9.26 kPa), and modest haemoglobin levels (5.6 g dl−1) relative to temperate species. Following 3 min strenuous activity there was a decrease in arterial pH (7.63) and a small rise in lactate from 0.41 to 0.68 mm, but no significant change in the calculated net metabolic acid load (δHm+). PaO2 and PaCO2 varied inversely during exercise, and oxygen content declined by 22%. pHa and most other haematological parameters returned to routine values between 1–3 h post-exercise. The results suggest that the major effect of strenuous activity in Notothenia spp. is a respiratory, rather than a metabolic acidosis.  相似文献
