We describe a protocol for somatic embryogenesis of Protea cynaroides, with potential for high frequency production of this important horticultural species. Somatic embryos formed directly on both P. cynaroides mature zygotic embryos and excised cotyledons cultured on MS medium without growth regulators. The addition of growth regulators such as naphthalene acetic acid (NAA) (5; 13 and 27 μM) and 2,4-dichlorophenoxyacetic acid (2,4-D) (5; 11 and 23 μM), in combination with thidiazuron (TDZ) (1 μM), benzylaminopurine (BAP) (1 μM) or kinetin (1 μM) suppressed the formation of somatic embryos. After eight weeks in culture, formation of somatic embryos was observed. Zygotic explants formed the most embryos when cultured in a 12-h photoperiod in comparison to explants cultured in the dark. Up to 83% of these embryos germinated after transferal to the germination medium containing 0.3 μM GA3. Significantly fewer embryos germinated in MS medium with no growth regulators, or supplemented with higher concentrations of GA3, while low germination percentages were also observed in MS media containing casein hydrolysate and coconut water. The germination of normal somatic embryos (two separate cotyledons and a single radicle) was observed only in media containing either no growth regulators, 0.3 μM GA3 or 1 μM GA3. All embryos that germinated in high concentrations of GA3 were malformed.  相似文献

The etiology and epidemiology of concentric leaf spot and seedling blight diseases of yam (Dioscorea spp) was investigated at Jbadan, south western Nigeria, in the low land humid tropics. Sclerotium rolfsii was associated with the concentric leaf spot and seedling blight of yam. The pathogen was also harbored by weeds namely Mucuna pruriensis, Commelina erecta, ipomea triloba, I. involucrataand Ipomoea sp found growing within the yam plots. A high mean inoculum densities of 3.0 ± 0.5 × 106 colony forming unit/g of soil was estimated in the soil samples obtained from the yam plots This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

Aims: To investigate the inhibition potential of leaf‐associated bacteria against the pathogen of bacterial leaf spot of Euphorbia pulcherrima. Methods and Results: Seven out of 200 bacterial strains were effective antagonists by in vitro screening and the two strains PAB241 and PAB242 significantly reduced the disease incidence and severity as foliar treatments of E. pulcherrima. The two effective strains, PAB241 and PAB242, were both identified as Bacillus amyloliquefaciens by a polyphasic approach including phenotypic feature, carbon source utilization profile, fatty acid methyl esters and analysis of 16S rRNA gene sequence. In addition, the suspensions of B. amyloliquefaciens PAB241 and PAB242 showed antibacterial activities against the pathogen of bacterial leaf spot of E. pulcherrima under different treatments. Conclusions: The leaf‐associated bacteria, B. amyloliquefaciens PAB241 and PAB242, markedly inhibited the growth of X. axonopodis pv. poinsettiicola under different treatments and protected E. pulcherrima from pathogen infection in growth chamber conditions. Significance and Impact of the Study: This is the first study that showed B. amyloliquefaciens from plant leaves was a potential bactericide against bacterial leaf spot of E. pulcherrima.  相似文献

Genetic mapping of gray leaf spot (GLS) resistance genes in maize   总被引:3,自引:0,他引:3  
Bulked segregant analysis was used to identify amplified fragment length polymorphism markers (AFLPs) linked to quantitative trait loci (QTLs) involved in the resistance to gray leaf spot (GLS) in maize. By using ten AFLP primer combinations 11 polymorphic markers were identified and converted to sequence- specific PCR markers. Five of the 11 converted AFLPs were linked to three GLS resistance QTLs. The markers were mapped to maize chromosomes 1, 3 and 5 using existing linkage maps of two commercially available recombinant inbred-line populations. Converted restriction fragment length polymorphism markers and microsatellite markers were used to obtain a more-precise localization for the detected QTLs. The QTL on chromosome 1 was localized in bin 1.05/06 and had a LOD score of 21. A variance of 37% was explained by the QTL. Two peaks were visible on chromosome 5, one was localized in bin 5.03/04 and the other in bin 5.05/06. Both peaks had a LOD score of 5, and 11% of the variance was explained by the QTLs. A variance of 8–10% was explained by the QTL on chromosome 3 (bin 3.04). The consistency of the QTLs was tested across two F2 populations planted in consecutive years. 2013年在内蒙古赤峰市喀喇沁旗发现桔梗匍柄霉叶斑病,该病害在2014-2017年均再次发生。采用单孢分离法在病叶上得到病原菌,于PCA培养基上培养后,在光学显微镜下观察记录病原菌形态学特征。同时将病原菌的rDNA ITS、gpd和EF-1α的PCR产物测序后做BLAST分析。结果表明该菌与桔梗匍柄霉Stemphylium platycodontis基本一致。这是国内桔梗匍柄霉叶斑病的首次详细报道。  相似文献

Bacterial leaf spot of lettuce, caused by Xanthomonas campestris pv. vitians, is a devastating disease of lettuce worldwide. Since there are no chemicals available for effective control of the disease, host-plant resistance is highly desirable to protect lettuce production. A total of 179 lettuce genotypes consisting of 29 leaf, 15 crisphead, one stem, 21 romaine, and 113 butterhead types were evaluated for response to X. c. vitians. One source of high resistance and five sources of moderate resistance were identified with four being butterhead lettuce and two leaf lettuce. The population genetic structure based on 350 expressed-sequence-tag-derived single nucleotide polymorphism generated two clades: Clade I and Clade II. The butterhead type was genetically distinct from romaine and crisphead types, while the leaf type was found to frequently exchange genes with other types through the history of breeding. Association mapping identified one single nucleotide polymorphism (QGB19C20.yg-1-OP5) associated with disease severity in Q general linear model and Q + K mixed linear model. Two SNP markers (Contig15389-1-OP1 and Contig6039-19-OP1) were associated with the resistance in the leaf lettuce PI 358000-1 which had no disease symptoms. The marker QGB19C20.yg-1-OP5 is in linkage group 2, while both of Contig15389-1-OP1 and Contig6039-19-OP1 are in linkage group 4. The resistant lines and the associated SNPs should be useful to develop resistant cultivars to battle against the devastating disease in lettuce.  相似文献

Of twenty-one fungicides tested at recommended rates of application for their effectiveness in controlling leaf spot of celery grown in Ireland on peat and on a clay-loam soil, three formulations containing fentin acetate with maneb and formulations of fentin hydroxide and fentin chloride were most effective. Benomyl was superior to the other non-tin fungicides tested and, at moderate disease levels, it resulted in marketable yields of celery equal to those recorded after treatment with the organotin compounds. Generally, the organotin compounds and benomyl were followed in descending order of effectiveness by copper oxychloride, captafol, Daconil 2787 and maneb. None of the fungicides tested in the three seasons caused visible symptoms of phytotoxicity and, on disease-free celery in 1969, none had a significant effect on yield.  相似文献

Xanthomonas campestris pv. mori causes bacterial leaf spot (BLS) of mulberry (Morus spp.) worldwide. Twenty mulberry lines with contrasting response to BLS showed significant (p?<?0.01) variation between resistant and susceptible groups for stomata frequency (SF), trichome density (TD), total soluble sugar (TSS) and polyphenol (PP) content, while non-significant for chlorophyll content. Heritability (broad-sense) estimates obtained from the variance component analysis were high (≥0.72) for these traits. The disease severity index values of the lines were significantly negatively correlated with TD (r?=??0.66; p?<?0.01), TSS (r?=??0.73; p?<?0.01) and PP (r?=??0.61; p?<?0.01), while positively correlated with SF (r?=?0.65; p?<?0.01). The cluster analysis was based on highly significant four variables and DSI values, and classified the lines into two major clusters. The resistant lines that come under cluster-I had high TD, TSS and PP, while those in susceptible category had high SF in cluster-II. These findings suggest that direct selection for SF, TD, TSS and PP will be effective for development of host resistance against BLS in mulberry.  相似文献

Pollen protoplasm is very nutritious, but the hard and highly resistant outer wall (exine) of the pollen grain presents an obstacle that pollen-feeders must overcome to benefit from the valuable protoplasm. Pollen digestion in three pollen-eating scarab beetles, the green protea beetle Trichostetha fascicularis and two monkey beetle species, Peritrichia cinerea and Pachynema flavolineata, was investigated. Adult beetles were collected for observations of feeding mechanisms, scanning electron microscopy of mouthparts, and histological examination of gut contents. Serial sections of different regions along the gut were stained for different nutrients and examined to determine the appearance of the pollen grains and the removal of carbohydrates, proteins and lipids from the pollen. During feeding, beetles used dense brushes of setae on the tips of their maxillae to mop up nectar and sweep pollen into their mouths. Grains were ingested intact. Nutrients were effectively removed from the grains as they passed along the gut and most pollen grains in the hindgut were empty, with broken exines. All the beetles excreted the undigested exine. Comparison with other studies on pollen digestion by a variety of insect and vertebrate pollen-feeders suggests that digestive enzyme penetration of ingested pollen grains and exudation of the products of digestion is the most likely method used by these beetles.  相似文献

The relationship between development of light leaf spot and yield loss in winter oilseed rape was analysed, initially using data from three experiments at sites near Aberdeen in Scotland in the seasons 1991/92, 1992/93 and 1993/94, respectively. Over the three seasons, single-point models relating yield to light leaf spot incidence (% plants with leaves with light leaf spot) at GS 3.3 (flower buds visible) generally accounted for more of the variance than single-point models at earlier or later growth stages. Only in 1992/93, when a severe light leaf spot epidemic developed on leaves early in the season, did the single-point model for disease severity on leaves at GS 3.5/4.0 account for more of the variance than that for disease incidence at GS 3.3. In 1991/92 and 1992/3, when reasonably severe epidemics developed on stems, the single-point model for light leaf spot incidence (stems) at GS 6.3 accounted for as much of the variance. Two-point (disease severity at GS 3.3 and GS 4.0) and AUDPC models (disease incidence/severity) accounted for more of the variance than the single-point model based on disease incidence at GS 3.3 in 1992/93 but not in the other two seasons. Therefore, a simple model using the light leaf spot incidence at GS 3.3 (x) as the explanatory variable was selected as a predictive model to estimate % yield loss (yr): yr= 0.32x– 0.57. This model fitted all three data sets from Scotland, When data sets from Rothamsted, Rosemaund and Thurloxton in England were used to test it, this single-point predictive model generally fitted the data well, except when yield loss was clearly not related to occurrence of light leaf spot. However, the regression lines relating observed yield loss to light leaf spot incidence at GS 3.3 often had smaller slopes than the line produce, by the model based on Scottish data.  相似文献

It has been assumed that species of the large African genus Protea have strong self‐incompatibility systems. However, this assumption was based largely on studies conducted on a clade of bird‐pollinated species that occur in the shrubby fynbos vegetation of the Cape region of southern Africa. To test whether self‐incompatibility occurs in a grassland/savanna Protea clade, which is largely insect‐pollinated, we performed controlled pollination experiments on four species, P. caffra, P. dracomontana, P. simplex and P. welwitschii. Although pollen–ovule ratios of all four species fall within the range for outcrossers, all four species are self‐compatible and capable of autonomous seed production. Using fluorescence microscopy, we found that self‐pollen tubes had the same probability of reaching ovules as cross‐pollen tubes. In the small tree P. caffra, selfed progeny had rates of germination and survivorship that were identical to those of crossed progeny. The grassland Protea spp. studied are likely to have mixed mating systems on account of being both visited by insects and capable of autonomous selfing. If one assumes previous reports of self‐incompatibility in Protea to be reliable, there have been at least five losses of self‐incompatibility and two gains of autonomous selfing in this genus. Six criteria for evaluating fungicides for control of celery leaf spot (Septoria apiicola) were compared. Two of these criteria consisted of yield measurements, of which marketable yield gave better statistical separation of the fungicides than gross yield. Of the other four criteria, assessment of leaf spot on a whole-plot basis was the most valuable. Overall disease rating on individual intact plants, rating of each petiole and numbers of diseased petioles per plant were less useful assessment criteria.  相似文献

In order to develop weather-based forecasting model of bacterial leaf spot (BLS) disease of mulberry caused by Xanthomonas campestris pv. mori, weekly disease severity data were recorded for three years on the ruling cultivar S-1. Daily meteorological data viz. maximum temperature, minimum temperature, maximum relative humidity, minimum relative humidity, rainfall and number of rainy days were also recorded. It was observed that BLS appeared in April/May and continued up to November with maximum severity in July. The correlation coefficient between disease severity and meteorological parameters revealed that the BLS disease severity has significant positive correlation with minimum temperatures, maximum and minimum relative humidity, rainfall and number of rainy days and negative correlation with maximum temperature. Multiple regressions analysis revealed that average of maximum temperature, minimum temperature and rainfall of preceding seven days and maximum relative humidity, minimum relative humidity of previous 9–15 days was found to maximally influence BLS disease severity. The contribution of the meteorological factors was found to be highest of minimum temperature (40.65%) followed by maximum temperature (24.20%), maximum relative humidity (16.41%), minimum relative humidity (8.07%), rainfall (5.29%) and number of rainy days (5.38%).  相似文献

从福建多地采集了55份感染叶斑病的枫香植物病样,包含叶片、叶柄、枝条和树皮。通过分离纯化得到了12个类拟盘多毛孢属真菌菌株,综合形态特征以及3个基因(ITS、β-tubulin和tef1)的分子系统发育分析结果,分别鉴定为Neopestalotiopsis cocoesN. chryseaPestalotiopsis neglectaP. neolitseae,均为枫香上首次报道,其中N. cocoes在国内首次被发现。通过柯赫氏法则验证,发现N. cocoes能够侵染叶片、叶柄和枝条,N. chrysea能侵染叶片和枝条,P. neglecta能侵染叶柄和枝条,而P. neolitseae不能侵染枫香植物组织。  相似文献

Light leaf spot lesions were generally first observed as light green areas on leaves of UK winter oilseed rape crops in January or February and later became brittle and bleached. Elongated lesions, which were brown with indistinct edges, developed on stems in the spring and summer, when lesions were also observed on flower buds, pedicels and pods. Development of diagnostic white pustules (spore masses of Pyrenopeziza brassicae, which erupt through surfaces of infected tissues) for confirmation of light leaf spot infection on symptomless plants or plants with indistinct or ambiguous symptoms in the autumn, winter or spring was enhanced by incubating plants in polyethylene bags. In experiments with artificially inoculated plants, glasshouse-grown plants exposed in infected crops and plants sampled from crops, white pustules developed at all incubation temperatures from 2oC to 20oC on infected leaves of different cultivars. The period of incubation required before the appearance of pustules decreased as the time that had already elapsed since the initial infection increased. The longest periods of incubation were required at the lowest temperatures (2oC or 5oC) but leaves senesced and abscised from plants most quickly at the highest temperatures (15oC or 20oC), suggesting that the optimal incubation temperature was between 10oC and 15oC.  相似文献

Development of phoma leaf spot (caused by Leptosphaeria maculans) on winter oilseed rape (canola, Brassica napus) was assessed in two experiments at Rothamsted in successive years (2003–04 and 2004–05 growing seasons). Both experiments compared oilseed rape cultivars Eurol, Darmor, Canberra and Lipton, which differ in their resistance to L. maculans. Data were analysed to describe disease development in terms of increasing numbers of leaves affected over thermal time from sowing. The cultivars showed similar patterns of leaf spot development in the 2003–04 experiment when inoculum concentration was relatively low (up to 133 ascospores m−3 air), Darmor developing 5.3 diseased leaves per plant by 5 May 2004, Canberra 6.6, Eurol 6.8 and Lipton 7.5. Inoculum concentration was up to sevenfold greater in 2004–05, with Eurol and Darmor developing 2.4 diseased leaves per plant by 16 February 2005, whereas Lipton and Canberra developed 2.8 and 3.0 diseased leaves, respectively. Based on three defined periods of crop development, a piece-wise linear statistical model was applied to the progress of the leaf spot disease (cumulative diseased leaves) in relation to appearance (‘birth’) and death of leaves for individual plants of each cultivar. Estimates of the thermal time from sowing until appearance of the first leaf or death of the first leaf, the rate of increase in number of diseased leaves and the area under the disease progress line (AUDPL) for the first time period were made. In 2004–05, Canberra (1025 leaves ×°C days) and Lipton (879) had greater AUDPL values than Eurol (427) and Darmor (598). For Darmor and Lipton, the severity of leaf spotting could be related to the severity of stem canker at harvest. Eurol had less leaf spotting but severe stem canker, whereas Canberra had more leaf spotting but less severe canker.  相似文献

Summary Rooted cuttings ofSeverinia buxifolia were inoculated with the vesicular-arbuscular mycorrhizal (VAM) fungusGlomus intraradices or provided an inoculum filtrate (non-VAM plants) and grown in one of seven media combinations of fired montmorillonite clay (FMC) and Canadian peatmoss (CP) at ratios of 100%, 80%, 67%, 50%, 33%, 20%, or 0% FMC. Mycorrhizal infection increased with higher proportions of FMC, but the growth of both VAM and non-VAM plants was reduced with increased FMC amendment. The growth benefit (top and root fresh-dry weights) conferred by mycorrhizal infection was greater at higher levels of FMC in the media. Improved phosphorus uptake by inoculated severinia plants appeared at least partially responsible for increased growth compared to non-VAM plants under conditions of high soluble salts and pH associated with high FMC composition.
