It is apparent that the fluoroquinolones represent an important advancement in infectious disease therapy. However, like other anti-microbial compounds, they are not without side effects. An understanding and appreciation of fluoroquinolone-drug interactions is essential for their optimal clinical use. Many questions concerning the potential of the fluoroquinolones with regard to drug interactions still remain.In addition, the majority of research on fluoroquinolone-drug interactions has been done in the research center environment using normal, healthy adult volunteers. The clinical significance of fluoroquinolone-drug interactions needs to be based upon observations in various patient populations, where feasible. Many of the aforementioned interactions could be influenced by race, dosage regimen design, smoking status of the patient, diet composition, and underlying organ function. Future research needs to assess the multiple factors of the clinical setting that might have an influence on patient outcome as a result of combining a fluoroquinolone with another interacting drug. Because norfloxacin and ciprofloxacin are commercially available in the United States, an active surveillance program in the community and institutional settings will help to identify and clarify many of the issues related to flouroquinolone-drug interactions.  相似文献

木质纤维素广泛存在于自然界中,因结构复杂,其高效降解需要多种微生物的协同互作,由于参与木质纤维素降解的微生物种类繁多,其协同降解机理尚不完全明确。随着微生物分子生物学和组学技术的快速发展,将为微生物协同降解木质纤维素机制的研究提供新的方法和思路。笔者前期研究发现,细菌复合菌系在50℃下表现出强大的木质纤维素降解能力,菌系由可分离培养和暂时不可分离培养细菌组成,但是可分离培养细菌没有降解能力。通过宏基因组和宏转录组研究表明,与木质纤维素降解相关的某些基因表达量发生显著变化,通过组学方法有可能更加深入解释微生物协同降解木质纤维素的微生物学和酶学机理。文中从酶、纯培养菌株和复合菌群三个方面综述了木质纤维素微生物降解研究进展,着重介绍了组学技术在解析复合菌群作用机理方面的现状和应用前景,以期为探索微生物群落协同降解木质纤维素的机理提供借鉴。  相似文献

For the maxillofacial region, there are various indications that cannot be interpreted from 2D images and will benefit from multiplanar viewing. Dental cone beam CT (CBCT) utilises a cone- or pyramid-shaped X-ray beam using mostly flat-panel detectors for 3D image reconstruction with high spatial resolution. The vast increase in availability and amount of these CBCT devices offers many clinical benefits, and their ongoing development has potential to bring various new clinical applications for medical imaging. Additionally, there is also a need for high quality research and education. European guidelines promote the use of a medical physics expert for advice on radiation protection, patient dose optimisation, and equipment testing. In this review article, we perform a comparison of technical equipment based on manufacturer data, including scanner specific X-ray spectra, and describe issues concerning CBCT image reconstruction and image quality, and also address radiation dose issues, dosimetry, and optimisation. We also discuss clinical needs and what type of education users should have in order to operate CBCT systems safely. We will also take a look into the future and discuss the issues that still need to be solved.  相似文献

The development of subunit vaccines against most parasitic helminth infections will require a better understanding of the different components of a natural rejection process including (1) recognition of parasite antigens; (2) induction of protective immune response phenotypes; and (3) activation of appropriate immune effector mechanisms. While novel technologies have allowed significant progress to be made in the identification of candidate vaccine antigens, the large scale production of these antigens and their presentation to the host with appropriate adjuvant systems remains a major problem in vaccine research. Identification of the molecular interactions involved in the innate immune response to helminth infections and the application of new genomic and proteomic technologies are likely to lead to major advances in these research fields. Gastrointestinal nematode parasites and liver fluke are the most important helminth parasites of production animals. In recent years, a lot of new knowledge has been gathered on the immunobiology of the host-parasite interactions in these two infection systems, which has allowed new vaccination strategies to be considered. Functional genomic technologies such as gene expression analysis by microarrays, promise to further advance our understanding of the molecular pathways leading to protection against parasite infections. This will not only have implications for vaccine research, but also provide novel targets for drug development and genetic selection.  相似文献

20世纪90年代中期以来,国际景观生态学发展迅速.景观生态学研究最为活跃的地区集中在北美、欧洲、大洋洲(澳大利亚)、东亚(中国),表明景观生态学理论、方法和应用的广泛性和越来越高的认知度.从研究内容上看,景观生态评价、规划和模拟一直占居主导地位,其次是景观生态保护与生态恢复、景观生态学的理论探讨.随着景观生态学研究的深入,以科学和实践问题为导向的学科交叉与融合不断加强,促进了景观生态学新的学科生长点的形成和发展,主要包括水域景观生态学、景观遗传学、多功能景观研究、景观综合模拟、景观生态与可持续性科学.在全球化背景下,中国的景观生态学研究也已经取得了长足进展,国际同行开始关注并在重要学术刊物上评介中国的景观生态学研究,标志着中国景观生态学已逐步走上国际舞台.然而,中国景观生态学进一步发展的挑战和机遇并存.中国科学家需要抓住人类对可持续发展和可持续性科学的共同夙求,放眼国际前沿、服务本土需求,从中国自身的特色出发,关注受人类干扰的和以人为主导的景观,以景观格局与生态过程的多尺度、多维度耦合研究为核心,区域综合与区内分异并重,推动综合整体性景观生态学的建立和完善.  相似文献

Scientific and technological advances derived from the genomics revolution have a central role to play in dealing with continuing infectious disease threats in the developing world caused by emerging and re-emerging pathogens. These techniques, coupled with increasing knowledge of host-pathogen interactions, can assist in the early identification and containment of outbreaks as well as in the development of preventive vaccination and therapeutic interventions, including the urgent need for new antibiotics. However, the effective application of genomics technologies faces key barriers and challenges which occur at three stages: from the research to the products, from the products to individual patients, and, finally, from patients to entire populations. There needs to be an emphasis on research in areas of greatest need, in facilitating the translation of research into interventions and, finally, the effective delivery of such interventions to those in greatest need. Ultimate success will depend on bringing together science, society and policy to develop effective public health implementation strategies to provide health security and health equity for all peoples.  相似文献

Current work in experimental biology revolves around a handful of animal species. Studying only a few organisms limits science to the answers that those organisms can provide. Nature has given us an overwhelming diversity of animals to study, and recent technological advances have greatly accelerated the ability to generate genetic and genomic tools to develop model organisms for research on host–microbe interactions. With the help of such models the authors therefore hope to construct a more complete picture of the mechanisms that underlie crucial interactions in a given metaorganism (entity consisting of a eukaryotic host with all its associated microbial partners). As reviewed here, new knowledge of the diversity of host–microbe interactions found across the animal kingdom will provide new insights into how animals develop, evolve, and succumb to the disease.  相似文献

The principles of modern aquaculture encourage the development of fish feeds containing low fish meal content and several types of plant ingredients plus nutrients to avoid depleting global fish stocks and to reduce costs. However, food constituents can affect animal nutrition and feeding behavior, so the effect of different diets on fish behavior and growth needs to be understood to optimize the use of nutrients and to improve fish welfare. The development of multiple-choice self-feeding systems led to a new perspective for investigating these issues in aquaculture species. Our purpose with this review is to summarize the information that has been published to date on this topic and to identify gaps in knowledge where research is needed. Key subjects are assessed under the following major headings: How do we study dietary selection in fish? What food signals do fish use to choose the right diet? and How do fish respond to food challenges? The present review will provide a picture of the main results obtained to date in these studies in aquaculture fish species, as well as perspectives for future research in the field.  相似文献

This essay attempts to raise some ethical problems related to biomedical research as they revolve around the issues of knowledge, power, man and justice. The arguments connected with such matters will not provide answers, but rather soundings for unchartered waters. Much more can and needs to be developed about the nature of knowledge and the politics of its use and abuse. A more detailed analysis and critique of man's nature might be engaged in to determine many concerns, not least of which is whether we can even talk about human nature. The idea of justice is elusive at the best of times and is particularly complicated at a time in history when social and economic interpretation is so complex. Much more must be debated before we can produce a detailed map. Fundamental to all of this, though, hopefully will be a dialogue among the many disciplines concerned with human research. We can not afford to work in isolation. The consequences of our decisions are too great.  相似文献

Biobanks of human biospecimens involving tissue taken from surgery require close relationships with diagnostic pathology practices. As most of the tissue will be analysed using genetic or genomic technologies there is the possibility that new information is created that could be of relevance to the donors. Although attention has been recently focused on the responsibilities that may arise from researchers and biobanks in terms of giving back individual genetic research results (IGRRs) to research participants, little has been said in relation to the role of pathology services. In this Commentary, we summarise the issues with respect to pathology services and what guidelines and professional practice documents say about their responsibilities. We also provide points to consider in the development of an ethically defensible plan for giving back individual research results.  相似文献

Since its establishment as a model organism, Caenorhabditis elegans has been an invaluable tool for biological research. An immense spectrum of questions can be addressed using this small nematode, making it one of the most versatile and exciting model organisms. Although the many tools and resources developed by the C. elegans community greatly facilitate new discoveries, they can also overwhelm newcomers to the field. This Review aims to familiarize new worm researchers with the main resources, and help them to select the tools that are best suited for their needs. We also hope that it will be helpful in identifying new research opportunities and will promote the development of additional resources.  相似文献

Every year, tens of thousands of people fall victim to one of invalidating neurological pathologies. Acquired brain injury leads to cognitive impairment and heavy loss of autonomy. Rehabilitation interventions are needed to enable people to recover capacity and return to Instrumental Activities of Daily Living (iADL), such as grocery shopping. Unfortunately, the resources made available in cognitive rehabilitation are insufficient for the growing needs of victims of brain damage. The goal of the AGATHE project is to develop a tool that will provide therapists with an innovative means of dealing with cognitive rehabilitation and offer patients customized rehabilitation sessions, on the basis of simulated iADL. AGATHE fits into accessibility prospects of the tool (for everyone, everywhere and anywhere) and reduction of the cost of rehabilitation. AGATHE allows the strengthening and diversification of skills and expertise of the project's clinical and research partners as well as the opening of a new application field to the technology of the industrial partners. Initial tests of the AGATHE tool have been performed among therapists and patients after brain injury in order to validate usability issues. Perspective of efficacy trials has been identified and development projects are explored.  相似文献

The future holds the possibility to link and network biobanks, existing biorepositories and reference databases for research purposes in ways that have not been possible before. There is the potential to develop 'research portals' that will enable researchers to access these research resources that are located around the globe with the click of a mouse. In this paper, I will argue that our current governance system for research is unable to provide all of the oversight and accountability mechanisms that are required for this new way of doing research that is based upon flows of data across international borders. For example, our current governance framework for research is nationally based, with a complex system of laws, policies and practice that can be unique to a jurisdiction. It is also evident that many of the nationally based governance bodies in this field do not have the legal powers or expertise to adjudicate on the complex issues, such as privacy and disclosure risks that are raised by cross-border data sharing. In addition, the conceptual underpinning of this research governance structure is based on the "one researcher, one project, one jurisdiction" model. In the conclusion of this paper, I lay out some preliminary ideas as to how this system has to change to accommodate research that is based on networks. I suggest that a move to digital governance mechanisms might be a start to making research governance systems more appropriate for the 21st century.  相似文献

The global initiatives to eliminate lymphatic filariasis as a public health problem by the year 2020 have generated a great deal of debate in India, the largest endemic country. This has led to a shift in the focus from control to elimination of the disease. Although the campaign to eliminate filariasis has begun, much more needs to be done. Several recent research studies have provided an insight into various operational issues and prospects of elimination of lymphatic filariasis. RNA binding proteins (RBPs) and RNA interaction is an emerging topic in molecular biology. Many reports showed that such interactions contribute to many cellular processes as well as disease development. Several standard in vitro and in vivo methods were developed to fulfill the needs of this RBP–RNA interaction study to explore their biological functions. However, these methods have their limitations in terms of throughput. In this review, we emphasize two important high throughput methods to studying RBP–RNA interactions, affinity purification and protein microarray. These methods have recently become robust techniques regarding their efficiency in systematically analyzing RBP–RNA interactions. Here, we provide technique overviews, strategies and applications of these methods during biological research. Although these technologies are just beginning to be explored, they will be most important methods in this study.  相似文献

Community ecology and ecosystem ecology provide two perspectives on complex ecological systems that have largely complementary strengths and weaknesses. Merging the two perspectives is necessary both to ensure continued scientific progress and to provide society with the scientific means to face growing environmental challenges. Recent research on biodiversity and ecosystem functioning has contributed to this goal in several ways. By addressing a new question of high relevance for both science and society, by challenging existing paradigms, by tightly linking theory and experiments, by building scientific consensus beyond differences in opinion, by integrating fragmented disciplines and research fields, by connecting itself to other disciplines and management issues, it has helped transform ecology not only in content, but also in form. Creating a genuine evolutionary ecosystem ecology that links the evolution of species traits at the individual level, the dynamics of species interactions, and the overall functioning of ecosystems would give new impetus to this much-needed process of unification across ecological disciplines. Recent community evolution models are a promising step in that direction.  相似文献

There is increasing interest in the need to address the ethical dilemmas related to the engagement of adolescents in sexual and reproductive health (SRH) research. Research projects, including those that address issues related to STIs and HIV, adverse pregnancy outcomes, violence, and mental health, must be designed and implemented to address the needs of adolescents. Decisions on when an individual has adequate capacity to give consent for research most commonly use age as a surrogate rather than directly assessing capacity to understand the issues and make an informed decision on whether to participate in research or not. There is a perception that adolescents participating in research are more likely to be coerced and may therefore not fully comprehend the risk they may be taking when engaging in research. This paper examines the various ethical issues that may impact stakeholders' decision making when considering engaging adolescents in SRH research in Nigeria. It makes a case for lowering the age of consent for adolescents. While some experts believe it is possible to extrapolate relevant information from adult research, studies on ethical aspects of adolescents' participation in research are still needed, especially in the field of sexual and reproductive health where there are often differences in knowledge, attitudes and practices compared to adults. The particular challenges of applying the fundamental principles of research ethics to adolescent research, especially research about sex and sexuality, will only become clear if more studies are conducted.  相似文献

Unique ethical issues can be associated with research outside the customary laboratory setting. Protocols involving wild animals must consider that any infringement on the wild nature of the species can be disruptive and may involve pain, fear, anxiety, and frustration, all of which constitute ethical harm that must be balanced with anticipated benefit. Agricultural and companion animal research, however, take place in a human-engineered environment and involves domesticated species adapted to human contact. Special animal welfare issues can be related to agricultural production goals that fail to deal adequately with moral concerns. Human/companion animal relationships, on the other hand, present unique moral obligations to animal owners. Other factors may present additional ethical issues when research is performed outside the laboratory. These factors include a required sensitivity to the environment of wild animals and an awareness that this outside research may to quite public and, therefore, vulnerable to community perception. The institutional animal care and use committee(IACUC) has the responsibility to ensure that research in outside settings is ethical and properly implemented. This responsibility requires that IACUC members have knowledge of the needs of a wide range of species and that a process is in place to allow effective monitoring of research in remote locations. Finally, and most important, there must be a sensitivity to the unique ethical considerations outlined here. Armed with these strengths, the IACUC will be effective in what may be unfamiliar surroundings and will have a significant opportunity to cause improvements in animal welfare.  相似文献

Drivers behind food security and crop protection issues are discussed in relation to food losses caused by pests. Pests globally consume food estimated to feed an additional one billion people. Key drivers include rapid human population increase, climate change, loss of beneficial on-farm biodiversity, reduction in per capita cropped land, water shortages, and EU pesticide withdrawals under policies relating to 91/414 EEC. IPM (Integrated Pest Management) will be compulsory for all EU agriculture by 2014 and is also being widely adopted globally. IPM offers a 'toolbox' of complementary crop- and region-specific crop protection solutions to address these rising pressures. IPM aims for more sustainable solutions by using complementary technologies. The applied research challenge now is to reduce selection pressure on single solution strategies, by creating additive/synergistic interactions between IPM components. IPM is compatible with organic, conventional, and GM cropping systems and is flexible, allowing regional fine-tuning. It reduces pests below economic thresholds utilizing key 'ecological services', particularly biocontrol. A recent global review demonstrates that IPM can reduce pesticide use and increase yields of most of the major crops studied. Landscape scale 'ecological engineering', together with genetic improvement of new crop varieties, will enhance the durability of pest-resistant cultivars (conventional and GM). IPM will also promote compatibility with semiochemicals, biopesticides, precision pest monitoring tools, and rapid diagnostics. These combined strategies are urgently needed and are best achieved via multi-disciplinary research, including complex spatio-temporal modelling at farm and landscape scales. Integrative and synergistic use of existing and new IPM technologies will help meet future food production needs more sustainably in developed and developing countries, in an era of reduced pesticide availability. Current IPM research gaps are identified and discussed.  相似文献

Although the concept of intelligence is difficult to define, research has provided evidence for a significant genetic component. Attempts are now being made to use molecular genetic approaches to identify genes contributing to intelligence, and to determine the ways in which they interact with environmental variables. This research is then likely to determine the developmental pathways of intelligence, in an effort to understand mental handicap and learning disorders and develop new treatment strategies. This paper reviews research on the genetic basis of intelligence, and discusses the ethical concerns, including the role of genetic information, the value we place on intelligence and the allocation of resources. It will be argued that the objections raised are problematic, and that because of the value of this knowledge and the prospect of improving lives, this research is morally required. We will then provide a brief analysis of the issues raised by enhancement of intelligence using genetic technology, and will argue that there is no intrinsic difference between this and other means of optimising intelligence.  相似文献
