To investigate feeding‐related decisions in Aedes aegypti (L.), adults are presented with simple diets of paired gustatory stimuli conveying information concerning energy content, nutrient richness, osmotic balance and food toxicity in a two‐diet matrix assay. Assessment of mosquito gut contents indicates that both sexes accept single sugar diets in a dose‐dependent manner. When presented with a choice between two different yet equimolar sugar solutions, more individuals of both sexes accept the disaccharides, sucrose and trehalose, than the monosacharrides, fructose and glucose. The combination of pyranose and furanose sugars in solution, either physically associated (as in sucrose) or present as monomers (as glucose and fructose), is accepted over solutions containing a single sugar moiety. Using the two‐diet matrix assay, mosquito diet‐choice is also tested between two equimolar sucrose ‘driver’ solutions in which one is presented with various concentrations of another potential feeding cue ‘test’ compound (i.e. each of the 20 naturally‐occurring amino acids, sodium chloride, quinine or caffeine). Diet‐choice between the ‘driver’ sucrose‐only solution and the solution of the ‘driver’ sucrose containing a ‘test’ amino acid is influenced by sex, amino acid concentration and sucrose concentration. There is also an example of synergism between the diet components, leucine and sucrose. Mosquitoes demonstrate a dose‐dependent acceptance of sucrose‐only diets over sodium chloride‐containing sucrose when presented together. Interestingly, the sucrose‐only diet is accepted by more mosquitoes than all concentrations of the saline‐containing sucrose diets except those approximately isotonic to mosquito haemolymph, at which concentration mosquitoes show no clear choice between the diets. More individuals of both sexes accept sucrose‐only diets than the diets of caffeine‐containing sucrose in a dose‐dependent manner. Only females, however, respond to quinine‐containing sucrose diets and modulate this behaviour in relation to the energetic reward: more females imbibed quinine‐containing sucrose at the higher sucrose concentration (1 m ). A systematic characterization of diet selection behaviour of A. aegypti is presented for 27 putative feeding cues potentially involved in nectar/honeydew feeding. This study will be used as a basis from which to investigate further the mosquito's assessment of food quality and ultimately host choice.  相似文献

Trissolcus brochymenae (Hymenoptera: Platygastridae) is an egg parasitoid that could be used to control stink bugs like Murgantia histrionica (Heteroptera: Pentatomidae), a pest of brassicaceous crops. Before laying their eggs, parasitoid females spend considerable time examining the substrate with their antennae, which are also used during feeding and mating behaviours. This suggests that contact chemoreception plays a prominent role in many aspects of parasitoid ecology. Therefore, we examined the sensitivity of antennal taste sensilla chaetica to several classical tastants including those that are appetitive or aversive. In addition we studied the taste preferences of wasps when presented with these chemicals alone or mixed. The sensilla chaetica of T. brochymenae responded to serial concentrations of sucrose, salts, and quinine, but no concentration-dependent effect was observed when testing sinigrin, a secondary metabolite found in many brassicaceae. However, both sinigrin and quinine inhibited responses to 0.1 M sucrose when mixed with this sugar. Behavioural taste preference assays confirmed that wasps showed a dose dependent preference for sucrose over agarose. In addition, a behavioural avoidance of sucrose solutions containing quinine was observed. This effect was not observed when sinigrin was used as a feeding deterrent. In the two-choice tests the wasp did not discriminate between sucrose solutions mixed with salts and sucrose alone. Further no preference for salts or sinigrin compared to agarose alone was observed. This work represents the first step towards the identification of gustatory receptor neurons implicated in the detection of different types of chemical cues in egg parasitoids.  相似文献

Although the function of nectar is to attract and reward pollinators, secondary metabolites produced by plants as anti‐herbivore defences are frequently present in floral nectars. Greater understanding is needed of the effects of secondary metabolites in nectar on the foraging behaviour and performance of pollinators, and on plant–pollinator interactions. We investigated how nectar‐feeding birds, both specialist (white‐bellied sunbirds Cinnyris talatala) and generalist (dark‐capped bulbuls Pycnonotus tricolor and Cape white‐eyes Zosterops virens), respond to artificial nectar containing the alkaloid nicotine, present in nectar of Nicotiana species. Preference tests were carried out with a range of nicotine concentrations (0.1–300 μM) in two sucrose concentrations (0.25 and 1 M), and for bulbuls also in two sugars (sucrose and hexose). In addition, we measured short‐term feeding patterns in white‐bellied sunbirds that were offered nicotine (0–50 μM) in 0.63 M sucrose. Both nicotine and sugar concentrations influenced the response of bird pollinators to nicotine. The birds showed dose‐dependent responses to nicotine; and their tolerance of high nicotine concentrations was reduced on the dilute 0.25 M sucrose diet, on which they increased consumption to maintain energy intake. White‐bellied sunbirds decreased both feeding frequency and feeding duration as the nicotine concentration in artificial nectar increased. Of the three species, bulbuls showed the highest tolerance for nicotine, and sugar type (sucrose or hexose) had no effect. The indifference of bulbuls to nicotine may be related to their primarily frugivorous diet. However, the response of white‐eyes to nicotine in the dilute sucrose solution was very similar to that of sunbirds, even though white‐eyes are generalist nectar‐feeders. Additional testing of other avian nectarivores and different secondary metabolites is required to further elucidate whether generalist bird pollinators, which utilise dilute nectars in which secondary metabolites have stronger deterrent effects, are more tolerant of ‘toxic’ nectar.  相似文献

In vitro-grown shoot tips of apples (Malus domestica Borkh. cv. Fuji) were successfully cryopreserved by vitrification. Three-week-old in vitro apple plantlets were cold-hardened at 5°C for 3 weeks. Excised shoot tips from hardened plantlets were precultured on a solidified Murashige & Skoog agar medium (MS) supplemented with 0.7 M sucrose for 1 day at 5°C. Following preculture shoot tips were transferred to a 2 ml plastic cryotube and a highly concentrated cryoprotective solution (designated PVS2) was then added at 25°C. The PVS2 contains (W/V) 30% glycerol, 15% ethylene glycol and 15% dimethylsulfoxide in medium containing 0.4 M sucrose. After dehydration at 25°C for 80 min, the shoot tips were directly plunged into liquid nitrogen. After rapid warming, the shoot tips were expelled into 2 ml of MS medium containing 1.2 M sucrose and then plated on agar MS medium. Direct shoot elongation was observed in approximately 3 weeks. The average rate of shoot formation was about 80%. This vitrification method was successfully applied to five apple species or cultivars and eight pear cultivars. Juvenile (3.0 ± 0.2 g) gibel carp (Carassius auratus gibelio) were fed to satiation for 8 weeks to investigate the effect of feeding frequency on growth, feed utilization and size variation. Five feeding frequencies were tested: two meals per day (M2), three meals per day (M3), four meals per day (M4), 12 meals per day (M12) and 24 meals per day (M24). The results showed that daily food intake increased significantly with the increase in feeding frequency and there was no significant difference between daily food intakes in M12 and M24 treatments. Growth rate, feed efficiency increased significantly with increasing feeding frequencies. Size variation was not affected by feeding frequency. Apparent digestibility of dry matter was not influenced by feeding frequency, while apparent digestibility of protein and energy increased significantly at high feeding frequencies. The feeding frequency had no significant effect on the moisture, lipid, protein, or energy contents of gibel carp, while the ash content decreased with increased feeding frequency. It was recommended that 24 meals per day was the optimal feeding frequency for juvenile gibel carp.  相似文献

Anther culture response was examined in five Solanum genotypes. Large genotypic differences exist in response to culture and liquid culture media were found to be superior to agar solidified media systems. Pre-treatment effects on embryoid induction were also investigated and two of the genotypes displayed differential response to temperature stress pretreatment. In general the beneficial effect of charcoal in the media was confirmed. Successful embryoid production and plantlet regeneration was obtained from the wild potato species, Solanum papita. The influence of sucrose concentration on embryoid production was also investigated. Large genotype by sucrose interactions were detected and this was mainly due to the differential response of the tuberosum clones to increasing sucrose concentration when compared with S. papita. Embryoids were produced from this species in media containing 15% sucrose although the optimum concentration of sucrose for embryoid production was 9%. The possible role of anther culture techniques in gene introgression and potato improvement is discussed.  相似文献

The effects of seven potential phagostimulants and of four molluscicidal compounds on feeding were examined by confining slugs with agar gels containing the chemicals in varying concentrations. Sugars generally increased the amount of gel ingested; sucrose more than glucose, lactose and fructose. Feeding increased with increasing sucrose concentration to a maximum and then fell progressively: the optimum concentration lay between 2.5% and 5%. The sweeteners saccharin and aspartame at concentrations up to 2.5% did not increase feeding. Addition of the molluscicides metaldehyde, methiocarb and ferric acetylacetonate to gels containing 2.5% sucrose progressively reduced feeding at concentrations of 0.001% and above. Metaldehyde reduced ingestion more than methiocarb and ferric acetylacetonate was intermediate. The molluscicidal herbicide Ioxynil deterred feeding completely at concentrations of 0.001%. The implications for slug bait development are discussed.  相似文献

This study investigated the influence of osmotic stress, induced by sorbitol and sucrose combinations, on growth and proline accumulation in callus cultures of rice (Oryza sativa L.). Dehusked mature seeds, cv. Hassawi, were induced to callus on MS medium supplemented with 4.52 µM 2,4-dichlorophenoxyacetic acid (2,4-D) and 2.32 µM 6-furfurylaminopurine (kinetin). The medium also contained 29.2, 58.4, 87.6, and 116.8 mM sucrose combined with 0, 54.9, 109.8, and 164.7 mM sorbitol. Callus formation was observed in about 35 % of the cultured seeds irrespective of the sugar treatment. An increase in callus mass was observed as sucrose concentration increased reaching a maximum growth at 87.6 mM. Callus growth was enhanced in response to 54.9 mM sorbitol but at higher concentration it was inhibitory. Best callus growth was obtained on a medium containing 54.9 mM sorbitol combined with 87.6 mM sucrose. Increasing osmotic stress, as a consequence of increasing sucrose and sorbitol concentrations, induced proline accumulation and the highest concentration of proline, 5.8 µmol g–1(f.m.), was obtained on 164.7 mM sorbitol combined with 116.8 mM sucrose.  相似文献

Some compounds that are bitter-tasting to humans, both alkaloidal (quinine, quinidine, atropine, caffeine) and non-alkaloidal (denatonium benzoate, sucrose octaacetate, naringin), deterred feeding and oviposition by Heliothis virescens (F.) in laboratory and field cage experiments. Preliminary electrophysiological studies of gustatory sensilla on the ovipositor of H. virescens provided evidence of 3 neurons, one of which is responsive to sucrose. Preliminary indications are that responses of this neuron may be inhibited by quinine and denatonium benzoate.  相似文献

Adults of Chortoicetes terminifera consumed larger meals of sucrose solution, applied as drops directly to their mouthparts, than of water. Meal size increased with increasing sucrose concentration up to 0·5 M but meals of 1·0 M sucrose were of lower volume. Locusts fed so that there was a short interval between each successive drop of sugar solution consumed greater volumes than those fed continuously, which indicates that sensory adaptation plays a part in bringing about the termination of meals of sugar solutions. Summary Larix occidentalis Nutt. can be micropropagated using whole excised mature embryos. The basal medium for induction (over 70% response) was half-strength Quoirin and LePoivre salts with Schenk and Hildebrandt organics and N6-benzyladenine singly or with kinetin at a final concentration of 10μM. Bud elongation was best on half-strength Schenk and Hildebrandt or Bornman’s mineral salts, and elongated shoots could be maintained on either half-strength Quoirin and LePoivre or Schenk and Hildebrandt formulations containing 0.05% activated charcoal, 2% sucrose, and 0.7% agar. Axillary buds developed naturally on elongating juvenile shoots, but a high sucrose treatment (6%) for 2 wk enhanced the number produced. Very good rooting (80 to 100%) was obtained by pulsing shoots for 2 to 3 h in a solution of indole butyric acid (1 mM, pH 4.5–5.0), or by planting shoots in peat-vermiculite moistened with basal medium containing 5μM α-naphthalene acetic acid, pH 5.0. Rooted shoots were hardened before transfer to the greenhouse, and initially were kept at low light and high humidity after transfer to ex vitro conditions. Abstract. The effects of single meals of different sucrose concentrations on feeding responses and survival of 8–24-h-old, 1–2-, 10–12- and 31–36-day-old female and male western cherry fruit flies, Rhagoletis indifferens Curran, were determined. Feeding time and food consumption response patterns in both sexes within age groups were curvilinear. Feeding times increased as sucrose concentrations increased, and were longest when the sucrose concentration was 100% (dry). Consumption of dilute wet sucrose was low, whereas consumption of concentrated wet sucrose was high. However, consumption of dry, 100% sucrose was also low. One to 2-day-old flies of both sexes that had not previously fed consumed more sucrose foods than unfed 8–24-h-old flies and 10–12- and 31–36-day-old flies that had been starved for 16–24 h. Females consumed more than males, but they consumed the same amount as males per mg bodyweight. When fed single 20% and 60% sucrose meals, 1–2-day-old flies survived longer compared to flies in all other age groups, with 31–36-day-old flies surviving shortest. Despite age-related differences in survival, in general, no sex differences in survival were seen in flies fed sucrose within any age groups, or in flies fed sucrose-yeast, cherry juice and honeydew foods. The results suggest that sugar-feeding behaviours and the energy invested in sugar 'seeking' by both sexes of R. indifferens should be the same throughout life.  相似文献

Injections of the melanocortin 3/4 receptor (MCR) agonist melanotan II (MTII) to a variety of brain structures produces anorexia, suggesting distributed brain MCR control of food intake. We performed a detailed analysis of feeding behavior (licking microstructure analysis) after a range of MTII doses (0.005 nM to 1 nM) was targeted to the forebrain (third ventricle, 3V) or hindbrain (fourth ventricle, 4V) regions. MTII (0.1 nM and 1 nM) delivered to the 3V or 4V significantly reduced 0.8 M sucrose intake. The anorexia was mediated by reductions in the number of licking bursts in the meal, intrameal ingestion rate, and meal duration; these measures have been associated with postingestive feedback inhibition of feeding. Anorexia after 3V but not 4V MTII injection was also associated with a reduced rate of licking in the first minute (initial lick rate) and reduced mean duration of licking bursts; these measures have been associated with taste evaluation. MTII effects on taste evaluation were further explored: In experiment 2, 3V MTII (1 nM) significantly reduced intake of noncaloric 4 mM saccharin and 0.1 M and 1 M sucrose solutions, but not water. The anorexia was again associated with reduced number of licking bursts, ingestion rate, meal duration, initial lick rate, and mean burst duration. In experiments 3 and 4, brief access (20 s) licking responses for sweet sucrose (0.015 M to 0.25 M) and bitter quinine hydrochloride (0.01 mM to 1 mM) solutions were evaluated. Licking responses for sucrose were suppressed, whereas those for quinine solutions were increased after 3V MTII, but not after 4V MTII injections (0.1 nM and 1 nM). The results suggest that multiple brain MCR sites influence sensitivity to visceral feedback, whereas forebrain MCR stimulation is necessary to influence taste responsiveness, possibly through attenuation of the perceived intensity of taste stimuli.  相似文献

In vitro morphogenesis via organogenesis was achieved from callus cultures derived from hypocotyl explants of Acacia sinuata on MS (Murashige and Skoog, 1962) medium. Calli were induced from hypocotyl explants excised from 7-day-old seedlings on MS medium containing 3% sucrose, 0.8% agar, 6.78 μM 2,4-dichlorophenoxyacetic acid and 2.22 μM 6-benzylaminopurine. Regeneration of adventitious buds from callus was achieved when they were cultured on MS medium supplemented with 10% coconut water, 13.2 μM 6-benzylaminopurine and 3.42 μM indoleacetic acid. Addition of gibberellic acid (1.73 μM) favored shoot elongation. Regenerated shoots produced prominent roots when transferred to half strength MS medium supplemented with 7.36 μM indolebutyric acid. Rooted plantlets, thus developed were hardened and successfully established in the soil. This protocol yielded an average of 20 plants per hypocotyl explant over a period of 4 months. A system consisting of a flow-through chamber connected to a commercial infrared gas analysis system was developed to measure homopteran respiration during feeding. Using this system, respiration rates of 202 and 206 μmol CO(2) h(-1) g(-1) (4.96 and 5.04 ml CO(2) h(-1) g(-1)) were determined for whiteflies and cotton aphids, respectively, at 25 degrees C on diets containing 15% sucrose. These rates were considerably higher than those of other stationary insects, indicating that whiteflies and aphids maintain a relatively high metabolic rate when feeding. Whitefly respiration increased with temperature from 25 to 46 degrees C with a Q(10) of about 2 on diets containing 10, 15 and 20% sucrose, but less than 2 on diets containing 2.5 and 5% sucrose. Respiration rates were similar on the diets containing >10% sucrose, but were generally lower on the diets containing <10% sucrose. Respiration rates decreased upon extended exposure to 47 degrees C; the rate of decrease was inversely related to the dietary sucrose concentration up to 15%. The results indicate that whiteflies require a sucrose concentration of between 5 and 10% (i.e. 0.15 and 0.3 M) for maximum rates of metabolism while feeding. Higher concentrations of sucrose in the diet delayed high-temperature mortality, possibly a reflection of the high sucrose requirement for sorbitol synthesis in whiteflies.  相似文献

Duckweed(Lemna gibba) is a useful model system for elucidating plant development, but the techniques needed for regenerating fronds from calli are not yet well established. This study examined the effects of auxin, sucrose, and gelling agents on callus and frond formation inL. gibba G3. After three weeks of culturing on a solid medium, two types of calli were observed: watery, pale-green, and undifferentiated; or white, compact calli that were organized into nodules and which resembled somatic embryogenie calli. Homogeneous callus lines were produced through selective subculture. To induce nodular calli, auxin (2,4-D) was absolutely required, with an effective concentration of 5 to 20 μM; induction was found to be possible with up to a maximum concentration of 4.4%. The calli were then maintained on a medium with a reduced 2,4-D concentration (1 μM), and were transferred every three weeks. Optimal callus induction and growth were obtained by using 3% sucrose with a combination of 0.15% Gelrite and 0.4% agar. Fronds, however, could be regenerated only on distilled water solidified with a combination of 0.4% agar and 0.15% Gelrite. On this medium, 87% of the callus expiants regenerated into fronds after four weeks of culture. These new fronds were morphologically normal but small, approximately 15 to 20% of the size of stock fronds. Continued culture of these fronds in an SH medium produced normal duckweeds, and histological examination of the cultures revealed several distinct types of callus nodules. Nonetheless, because zygotic embryogenesis inL. gibba does not produce distinct bipolar structures, the developmental pathway of frond regeneration from these nodular cultures remains unknown.  相似文献

Summary Conditions that allow regeneration of cells fromClostridium acetobutylicum strain B643 protoplasts were studied. Protoplast formation and stabilization in minimal media with 50 mM CaCl2, 50 mM MgCl2 and 0.3 M sucrose were crucial to subsequent regeneration on soft yeast extract agar containing 25 mM CaCl2 and 25 mM MgCl2. A conditioned flavor aversion (CFA) was used to measure thetaste characteristics of 0.005 and 0.025 M Na-saccharin in therat. At neither concentration did the rats show generalizationto NaCl, quinine HCl or SO4, sucrose octa-acetate or HCl, whereasgeneralization to sucrose was found at both concentrations.When the CFA was formed to 0.0001 M quinine HCl, no direct evidencewas found for generalization of the aversion to either saccharinconcentration. In vitro organogenesis was achieved from callus derived from hypocotyl explants of Cucumis sativus L. cv. Poinsett 76. Calli were induced from hypocotyl explants excised from 7-d-old seedlings grown on Murashige and Skoog (MS) medium containing 87.64 μM sucrose, 0.8 % agar, 3.62 μM 2,4-dichlorophenoxy acetic acid and 2.22 μM 6-benzyladenine (BA). Regeneration of adventitious buds from callus (25 shoots explant−1) was achieved on MS medium supplemented with 8.88 μM BA, 2.5 μM zeatin and 10 % coconut water after two subcultures in the same medium at 30-d interval. Gibberellic acid (1.75 μM) favoured shoot elongation and indole 3-butyric acid (7.36 μM) induced rooting. Rooted plants were hardened and successfully established in soil.  相似文献
