We investigated the genetic differentiation of Rosa canina at regional and continental scales to delineate provenance regions for seed collection. Using RAPD technique, we examined eight populations in Brandenburg (Germany) and 13 samples from Bavaria and Hungary. Using RAPD technique, we examined eight populations in Brandenburg (Germany) and 13 samples from Bavaria and Hungary. AMOVA and -statistics revealed a significant differentiation ( = 0.23, P < 0.0001) between accessions from Brandenburg and the two other regions. The interpopulation differentiation in Brandenburg was low ( = 0.13). We found the greatest interpopulation distance between Hungary and Brandenburg (Hoher Fl?ming, = 0.47) and the lowest between Dahmeland and Hoher Fl?ming (both Brandenburg, = 0.01). UPGMA dendrogram and PCA showed a clear disjunction between Brandenburg and other regions. Due to the outcrossing mating and seed dispersal system, the genetic variation within R. canina populations is moderately high (87% for Brandenburg, 76% for Bavaria and Hungary). The significant genetic differentiation at the continental scale suggests lower gene flow with increasing geographical distance and underlines the usefulness of regional provenances for planting.  相似文献

Quantifying foliar stable carbon isotope discrimination (Δ) is a powerful approach for understanding genetic variation in gas exchange traits in large populations. The genetic architecture of Δ and third-year height is described for more than 1,000 clones of Pinus taeda tested on two contrasting sites. for Δ was 0.14 (±0.03), 0.20 (±0.07), and 0.09 (±0.04) at Florida, Georgia, and across sites, respectively. for stable carbon isotope discrimination ranged from 0.25 (±0.03) at the Florida site to 0.33 (±0.03) at the Georgia site, while the across-site estimate of was 0.19 (±0.02). For third-year height, ranged from 0.13 (±0.05) at the Georgia site to 0.20 (±0.06) at the Florida site with an across-site estimate of 0.09 (±0.05). Broad-sense heritability estimates for third-year height were 0.23 (±0.03), 0.28 (±0.03), and 0.13 (±0.02) at the Florida site, Georgia site, and across sites, respectively. Type B total genetic correlation for Δ was 0.70 ± 0.06, indicating that clonal rankings were relatively stable across sites, while for third-year height, rankings of clones were more unstable across the two trials . Third-year height and Δ were negatively correlated at the parental , full-sib family , and clonal levels, suggesting that genetic variation for Δ in P. taeda may be a result of differences in photosynthetic capacity. We conclude that Δ may be a useful selection trait to improve water-use efficiency and for guiding deployment decisions in P. taeda.  相似文献

Marine invertebrate populations usually show high levels of genetic variability that has frequently been associated with spatial and temporal environmental heterogeneity. One of the most heterogeneous marine environments is the intertidal zone, the habitat of Collisella subrugosa, the most widespread and abundant Brazilian limpet. C. subrugosa has planktonic larvae that can disperse over long distances, what can promote gene flow among shores, working against interpopulational differentiation. In this study we investigated the genetic variability and populational substructure of C. subrugosa through analysis of 24 allozyme loci in 14 samples (590 individuals) collected along 2,700 km of the Brazilian coast. The genetic variability was high ( and ), as expected for intertidal species. Genetic differentiation among samples was low (F ST = 0.03) what may reflect intensive gene flow associated with larval dispersal. However, we detected an isolation-by-distance pattern of population substructure in one sampled region. High levels of heterozygote deficiency were also observed for many loci in each sample. Alternative hypothesis are discussed, and the “breeding groups” is suggested to explain these pattern, indicating the main cause as environmental heterogeneity.  相似文献

Analysis of linkage disequilibrium (=mean squared correlation of allele frequencies at different gene loci) provides a means of estimating effective population size (N e) from a single sample, but this method has seen much less use than the temporal method (which requires at least two samples). It is shown that for realistic numbers of loci and alleles, the linkage disequilibrium method can provide precision comparable to that of the temporal method. However, computer simulations show that estimates of N e based on for unlinked, diallelic gene loci are sharply biased downwards ( in some cases) if sample size (S) is less than true N e. The bias is shown to arise from inaccuracies in published formula for when S and/or N e are small. Empirically derived modifications to for two mating systems (random mating and lifetime monogamy) effectively eliminate the bias (residual bias in % in most cases). The modified method also performs well in estimating N e in non-ideal populations with skewed sex ratio or non-random variance in reproductive success. Recent population declines are not likely to seriously affect , but if N has recently increased from a bottleneck can be biased downwards for a few generations. These results should facilitate application of the disequilibrium method for estimating contemporary N e in natural populations. Summary Outcrossing rate estimates for eight accessions of Clarkia tembloriensis indicate that this annual plant species has a wide interpopulational range of outcrossing rate ( ). Populations' t estimates were significantly correlated with observed heterozygosity and mean number of alleles per locus. Estimated fixation indices, , for most populations were very close to their expected values, Feq, for a given Nei's gene diversity statistics showed that the group of outcrossing populations have more total genetic variation and less differentiation among populations than does the group of selfing populations. These results indicate that the breeding system of C. tembloriensis has had a strong influence on the amount and distribution of genetic variation within and among its populations.  相似文献

A steady supply of water is indispensable for leaves to fulfil their photosynthetic function. Understanding water movement in leaves, especially factors that regulate the movement of water flux from xylem to epidermis, requires that the nature of the transport pathway be elucidated. To determine the hydraulic linkage between xylem and epidermis, epidermal cell turgor pressure (P t) in leaves of Tradescantia fluminensis was monitored using a cell pressure probe in response to a 0.2 MPa step change in xylem pressure applied at the leaf petiole. Halftime of P t changes were 10–30 times greater than that of water exchange across an individual cell membrane suggesting that cell-to-cell water transport constitutes a significant part of the leaf hydraulic path from xylem to epidermis. Furthermore, perfusion of H2O2 resulted in increases of both and by a factor of 2.5, indicating that aquaporins may play a role in the xylem to epidermis hydraulic link. The halftime for water exchange did not differ significantly between cells located at the leaf base (2.5 s), middle (2.6 s) and tip (2.5 s), indicating that epidermal cell hydraulic properties are similar along the length of the leaf. Following the pressure application to the xylem (0.2 MPa), P t changed by 0.12, 0.06 and 0.04 MPa for epidermal cells at the base, middle and the tip of the leaf, respectively. This suggests that pressure dissipation between xylem and epidermis is significant, and that the pressure drop along the vein may be due to its structural similarities to a porous pipe, an idea which was further supported by measurements of xylem hydraulic resistance using a perfusion technique.  相似文献

The negative effects of inbreeding depression on fragmented small populations are likely to be expressed more strongly after restoration efforts if regeneration processes have been highly restricted in degraded habitats. We examined the potential influences of inbreeding depression on a population of Nymphoides peltata (Menyanthaceae) restored from the remnant soil seed bank. A hand-pollination experiment demonstrated self-compatibility of a single remaining homostyle genet and significant inbreeding depression in selfed progeny, especially in parameters related to seedling growth (–0.6 for biomass, and –0.4 for relative growth rate). Our genetic analysis indicated that the presumed number of parents contributing to the current soil seed bank was only 2–8 genets and that a single sib-family dominated at each of three sampling sites. The results also showed that the selfed progeny of the homostyle genet were overwhelmingly dominant at two sites (86.8 and 94.7%). As a result, the growth performance of the seed bank seedlings was significantly reduced, to a level as low as that of the selfed progeny. Active restoration efforts to minimize the negative effects of the genetic bottleneck and continuous monitoring based on genetic and demographic study are recommended.  相似文献

For long-lived iteroparous vertebrates that annually produce few young, life history theory predicts that reproductive output (R) and juvenile survival should influence temporal variation in population growth rate (λ) more than adult survival does. We examined this general prediction using 15 years of mark–recapture data from a population of California spotted owls (Strix occidentalis occidentalis). We found that survival of individuals ≥1 year old (ϕ) exhibited much less temporal variability , where CV is coefficient of variation, than R and that R was strongly influenced by environmental stochasticity. Although λ was most sensitive ( ; log-transformed sensitivity) to ϕ and much less sensitive to either R or juvenile survival (survival rate of owls from fledging to 1 year old; ), we estimated that R contributed as much as ϕ to the observed annual variability in λ. The contribution of juvenile survival to variability in λ was proportional to its These results are consistent with the hypothesis that natural selection may have favored the evolution of longevity in spotted owls as a strategy to increase the probability of experiencing favorable years for reproduction. Our finding that annual weather patterns that most affected R (temperature and precipitation during incubation) and ϕ (conditions during winter related to the Southern Oscillation Index) were equally good at explaining temporal variability in λ supports the conclusion that R and ϕ were equally responsible for variability in λ. Although currently accepted conservation measures for spotted owl populations attempt to enhance survival, our results indicated that conservation measures that target R may be as successful, as long as actions do not reduce ϕ.  相似文献

The electrophoretic variability of nine enzymatic loci was studied in 31 populations of eleven species of the genus Orchis (fam. Orchidaceae). This orchid group shows a higher degree of genetic variability ( ) than reported by Gottlieb (Progr. Phytochem. 7:1–46, 1981) for other outbreeding plant species ( ).Furthermore, the Nei coefficients of genetic identity show a higher biochemical-genetic similarity between species pairs of which we found the hybrids ( ) than between other species pairs ( ). These results seem to indicate a correlation between hybridizing aptitude and degree of genetic similarity. High values could be partially attributed to the presence of a gene flow between two species which, by hybridizing, would join their genetic pools. But they could also indicate that hybridizing species are not well differentiated: the isozyme similarity would reflect a more general similarity of the gene pool that could lead to hybrid formation.  相似文献

A non-autonomous dynamical system, in which the seasonal variation of a mosquito vector population is modeled, is proposed to investigate dengue overwintering. A time-dependent threshold, R(t), is deduced such that when its yearly average, denoted by , is less than 1, the disease does not invade the populations and when is greater than 1 it does. By not invading the population we mean that the number of infected individuals always decrease in subsequent seasons of transmission. Using the same threshold, all the qualitative features of the resulting epidemic can be understood. Our model suggests that trans-ovarial infection in the mosquitoes facilitates dengue overwintering. Photosynthetic limitations under moderate water deficit were evaluated in ‘Valência’ orange trees grafted on three different rootstocks, in pots. Net CO2 assimilation rate (A N), stomatal conductance (g s), and photosystem II (PS II) operating efficiency ( ) in response to changing intercellular CO2 partial pressure (C i) were analyzed under controlled conditions. Drought decreased A N and g s, whereas remained unchanged. This resulted in a higher ratio between electron transport rate (ETR) and gross CO2 assimilation rate (A G). Since the comparison of A NC i gas exchange curves can lead to incorrect conclusions, a normalization of C i values () of stressed leaves was applied. Then, the relationship established for irrigated trees between the ETR/A G ratio and C i was used to estimate the from ETR/A G ratios measured under water stress. The response of A N to suggests that the CO2 diffusional restriction is the main factor that limits photosynthesis in orange leaves under moderate water deficit.  相似文献

During a survey of Guatemalan herpetofauna in the summers of 1998–2000, 29 presumed new species of Eimeria Schneider, 1875 were found, seven of which have a distinct elongate-ellipsoidal shape (L/W ratio ≥ 1.7) and are described herein. Six of the seven new species are similar in oöcyst length, width and L/W ratio and sporocyst length, width and L/W ratio, lack a micropyle, oöcyst residuum, Stieda body, sub- and parastieda bodies, have a polar granule and sporocyst residuum, and their sporocysts appear to have dehiscence sutures. The seventh is slightly smaller and has sporocysts with a Stieda body. The new species are: E. coniophanes n. sp – whose sporulated oöcysts from Coniophanes fissidens are 29.2×14.9 (27–31×13–16) m, with sporocysts m; E. coniophis n. sp. –from Conophis lineatus are 32.0×16.5 (30–34×14–18) m, with sporocysts m; E. dryomarchoni n. sp. – from Drymarchon corais are 32.2×17.7 (31–34×17–19) m, with sporocysts m; E. leptophis n. sp. – from Leptophis mexicanus are 29.5×17.0 (28–31×16–18) m, with sporocysts m; E. oxybelis n. sp. – from Oxybelis aeneus are 31.8×16.5 (29–33×15–18) m, with sporocysts m; and E. scaphiodontophis n. sp. – from Scaphiodontophis annulatus are 30.0×15.3 (28–33×14–16) m, with sporocysts m. Sporulated oöcysts of E. siboni n. sp. from Sibon nebulata are 24.3×14.2 (21–27×13–16) m, with sporocysts m and with a Stieda body. We conclude that until all aspects of each life-cycle are known, it is prudent at this time to name all tetrasporocystic dizoic coccidia from snakes as members of Eimeria rather than place some of them in Choleoeimeria Paperna & Landsberg, 1989.  相似文献

The conservation status of small breeding areas of the Goosander (Mergus merganser merganser) in Central Europe is unclear. Geographic isolation of these areas suggests restricted gene flow to and from large North-European populations. On the other hand, migrating Goosanders from northern Europe join the Central European breeding population for wintering. To evaluate the conservation status of the small breeding areas we assessed the genetic structure of M. merganser populations in Europe by examining two nuclear marker systems (microsatellites and Single Nucleotide Polymorphisms, SNP) and mitochondrial (mtDNA) control region sequence variation for Goosanders in 11 sampling areas representing three of five distinct breeding areas and two subspecies (M. m. merganser and M. m. americanus). Overall population differentiation estimates including both subspecies were high, both based on mtDNA () and nuclear markers (θ ST = 0.219; 95% CI 0.088–0.398, SNP and microsatellites combined). Within Europe, mtDNA revealed a strong overall () and significant pairwise population differentiation between almost all comparisons. In contrast, both nuclear marker systems combined revealed only a small overall genetic differentiation (θ ST = 0.022; 95% CI 0.003–0.041). The strong genetic differentiation based on female-inherited mtDNA but not on biparentally inherited nuclear markers can be explained by sex-biased dispersal and strong female philopatry. Therefore, small breeding areas in Europe are endangered despite large male-mediated gene-flow, because when these populations decline, only males—but due to strong philopatry not females—can be efficiently supplemented by migration from the large North European populations. We therefore propose to manage the small breeding areas independently and to strengthen conservation efforts for this species in Central Europe.  相似文献

A method for evaluating the average shear rate () in a stirred and aerated tank bioreactor has been proposed for non-Newtonian fluids. The volumetric oxygen transfer coefficient (k L a) was chosen as the appropriate characteristic parameter to evaluate the average shear rate (). The correlations for the average shear rate as a function of N and rheological properties of the fluid (K and n) were obtained for two airflow rate conditions (ϕair). The shear rate values estimated by the proposed methodology lay within the range of the values calculated by classical correlations. The proposed correlations were utilized to predict the during the Streptomyces clavuligerus cultivations carried out at 0.5 vvm and four different rotational impeller speeds. The results show that the values of the average shear rate () varied from 437 to 2,693 s−1 by increasing with N and flow index (n) and decreasing with the fluid consistency index (K).  相似文献

Genetically engineered pacemaking in ventricular cells has been achieved by down-regulation of the time independent inward rectifying current (I K1), or insertion of the hyperpolarisation-activated funny current (I f). We analyse the membrane system (i.e. ionic concentrations clamped) of an epicardial Luo-Rudy dynamic cell model using continuation algorithms with the maximum conductance () of I K1 and I f as bifurcation parameters. Pacemaker activity can be induced either via Hopf or homoclinic bifurcations. As K1 is decreased by ≈74%, autorhythmicity emerged via a homoclinic bifurcation, i.e., the periodicity first appear with infinitely large periods. In contrast, the insertion of f induced periodicity via a subcritical Hopf bifurcation at f≈ 0.25 mSμF−1. Stable autorhythmic action potentials occurred at f > 0.329 mSμF−1.  相似文献

Less than a decade after being observed off Florida, the invasive Indo-Pacific lionfish is now widely distributed off the southeast coast of the United States. As a step towards measuring invasion impacts to native communities, we examine the magnitude and extent of this invasion by first, compiling reports of lionfish to provide range information and second, estimate lionfish abundance from two separate studies. We also estimate native grouper (epinepheline serranids) abundance to better assess and compare lionfish abundances. In the first study we conducted SCUBA diver visual transect surveys at 17 different locations off the North Carolina coast in water depths of 35–50 m. In the second study, we conducted 27 Remote Operated Vehicle (ROV) transect surveys at five locations from Florida to North Carolina in water depths of 50–100 m. In both studies, lionfish were found to be second in abundance only to scamp (Mycteroperca phenax). Lionfish were found in higher abundance in the shallower North Carolina SCUBA surveys ( ha−1) than in the deep water ROV surveys ( ha−1). Lionfish reports continue to expand most recently into the Bahamas, raising the specter of further spread into the Caribbean and Gulf of Mexico. The potential impacts of lionfish to native communities are likely to be through direct predation, competition and overcrowding. The high number of lionfish present in the ecosystem increases the potential for cascading impacts throughout the food chain. Within the southeast region the combined effects of climate change, overfishing and invasive species may have irreversible consequences to native communities in this region. The self-complementary subset ∪{AAA,TTT} with = {AAC, AAT, ACC, ATC, ATT, CAG, CTC, CTG, GAA, GAC, GAG, GAT, GCC, GGC, GGT, GTA, GTC, GTT, TAC, TTC} of 22 trinucleotides has a preferential occurrence in the frame 0 (reading frame established by the ATG start trinucleotide) of protein (coding) genes of both prokaryotes and eukaryotes. The subsets ∪{CCC} and ∪{GGG} of 21 trinucleotides have a preferential occurrence in the shifted frames 1 and 2 respectively (frame 0 shifted by one and two nucleotides respectively in the 5′-3′ direction). and are complementary to each other. The subset contains the subset which has the rarity property (6 × 10−8) to be a complementary maximal circular code with two permutated maximal circular codes and in the frames 1 and 2 respectively. is called a C3 code. A quantitative study of these three subsets in the three frames 0, 1, 2 of protein genes, and the 5′ and 3′ regions of eukaryotes, shows that their occurrence frequencies are constant functions of the trinucleotide positions in the sequences. The frequencies of in the frame 0 of protein genes are 49, 28.5 and 22.5% respectively. In contrast, the frequencies of in the 5′ and 3′ regions of eukaryotes, are independent of the frame. Indeed, the frequency of in the three frames of 5′ (respectively 3′) regions is equal to 35.5% (respectively 38%) and is greater than the frequencies and , both equal to 32.25% (respectively 31%) in the three frames. Several frequency asymmetries unexpectedly observed (e.g. the frequency difference between and in the frame 0), are related to a new property of the subset involving substitutions. An evolutionary analytical model at three parameters (p, q, t) based on an independent mixing of the 22 codons (trinucleotides in frame 0) of with equiprobability (1/22) followed by t ≈ 4 substitutions per codon according to the proportions p ≈ 0.1; q ≈ 0.1 and r = 1 − pq ≈ 0.8 in the three codon sites respectively, retrieves the frequencies of observed in the three frames of protein genes and explains these asymmetries. Furthermore, the same model (0.1, 0.1, t) after t ≈ 22 substitutions per codon, retrieves the statistical properties observed in the three frames of the 5′ and 3′ regions. A dual isotope approach was used to assess the relative importance of terrestrial vegetation detritus and other primary producers in the trophic web of Flamengo Sound (Ubatuba, SP), SE Brazil, surrounded by the Atlantic Rain Forest. Primary producers showed distinct C signatures and the observed values suggest that little terrestrial or bulk sediment organic matter enter the food web of the sound. Suspended particulate organic matter (POM, supports the bulk of the consumers, with some contribution by macroalgae . Consumers C values ranged from −17.4 to . At least three trophic levels were detectable in the food web. The N value of POM was , while that of sediment and detritus was . The N values of suspension feeding benthic invertebrates were 8.2–, deposit feeders 8.3–, and carnivores 10.7–. Values for fishes were for detritivore, 11.4– for benthic feeders, 12.4– for zooplanktivores, and for piscivores/benthic invertebrate feeders. Squid mean value was . There is a reasonable agreement between feeding habits information from the literature and N values from this study. In the sound, the first and second trophic steps seem to be about 1– higher than those of similar organisms studied in temperate waters and this may reflect an input of allochtonous anthropogenic nitrogen enriched in 15N from human activities.  相似文献
