The application of copolymerized agarose-polyacrylamide gels as the support for immobilized Cibacron Blue F3G A is demonstrated for the analytical electrophoresis of proteins possessing an affinity for this dye. Bovine serum albumin was used as a model protein to develop this technique. The optimal conditions for preparing matrices are described. These conditions produce gels with suitable mechanical strength and which allow rapid electrophoresis of proteins. The dye-agarose-polyacrylamide gels permit the determination of dissociation constants. The ease of preparation of these matrices recommends them for a variety of quantitative analytical investigations.  相似文献

A sensitive and versatile method for the qualitative and quantitative determination of glycosaminoglycans (GAGs) is described. An enriched GAG fraction was subjected to nuclease enzyme treatment and to an appropriate sequence of GAG degrading enzymes—Streptomyces hyaluronidase, chondroitinase AC and ABC, and endo-β-d-galactosidase—and nitrous acid treatment. To determine the result of each degradative procedure, the remaining GAG polymers were subjected to cellulose acetate electrophoresis. The combination of sequential degradation and the monitoring of each step by electrophoresis and densitometry permitted the identification and quantitation of all the GAGs on a microscale basis.  相似文献

Mouse fibroblast (L-929) cells, in culture, synthesized and secreted into the growth medium a vitamin B12-binding substance which was identical to mouse transcobalamin II (TC II) as judged by the following criteria: (i) gel filtration on Sephadex G-200, (ii) ion-exchange chromatography on DEAE-cellulose and CM-cellulose, and (iii) the ability to facilitate cellular B12 uptake by L-929 cells. The secretion of mouse fibroblast binder was blocked by cycloheximide and puromycin; and in both cases the cells' ability to secrete this binder was partially restored when the inhibitor was removed. Within 30 h after the cells were exposed to [57Co]B12 bound to mouse serum TC II (Mr ~ 38,000) the [57Co]B12 was bound to a large molecular weight intracellular binder (Mr ~ 120,000) which was not released into the culture medium. During this same incubation period, the cells released free [57Co]B12 and [57Co]B12 bound to a protein which had the same elution volume as mouse serum TC II on Sephadex G-200.  相似文献

Malate dehydrogenase was adsorbed onto monomolecular lipid films, using a multicompartment trough. The quantity of adsorbed protein and its enzymatic activity were studied with monolayers of various electrical charge densities and subphases of various electrolyte compositions. A closely packed layer of enzyme molecules was adsorbed onto negatively charged films, whereas considerably less protein was adsorbed onto neutral and positively charged monolayers. Electrolytes reduce the quantity of adsorbed protein. The adsorption was found to be irreversible even at high ionic strength. When adsorbed to uncharged lipid films the enzyme is nearly inactive, whereas negatively charged lipid headgroups enhance the specific activity of the enzyme.  相似文献

Lactoperoxidase-catalyzed cell surface radioiodination was used to study the size distribution and physiological characteristics of cell surface proteins of Cloudman melanoma cells maintained in vivo (An cell) or in vitro (TC cells). Slight qualitative and quantitative differences were observed between detergent-extracted radiolabeled membrane proteins obtained from An and TC cells when the proteins were resolved by polyacrylamide gel electrophoresis in sodium dodecyl sulfate. Striking differences were observed in the membrane residence times of radioiodinated proteins of the An and TC cells when labeled cells were cultured 24 hr. Many labeled surface proteins are lost more rapidly from An cells than from TC cells. The possibility that the release of surface proteins from tumor cell membranes may be an important factor which determines the nature of the host immune response against incipient cancers is considered.  相似文献

Peanut agglutinin (PNA), a lectin specific for terminal d-galactosyl residues, was found to react with embryonal carcinoma cells, but not with their differentiated derivatives. Receptors for PNA were detectable at the surface of all cells of the quasinullipotent F9 line and on only 50% of the multipotent PCC3/A/1 line. The fraction of the PCC3/A/1 population which expresses the F9 antigen was found to be included in the subpopulation carrying the PNA receptors. PNA+ and PNA? subpopulations of PCC3/A/1 were separated by a PNA-mediated reversible agglutination of PNA+ cells with rabbit erythrocytes. These subpopulations were essentially F9+ and F9?, respectively.  相似文献

Changes in the physical properties and molecular structure of isolated nucleohistone, induced by binding magnesium or other ions, have been studied. Nucleohistone has a net negative charge. Added magnesium binds tightly to the DNA phosphate groups that are not already complexed with histone. This results in neutralization of the net negative charge, a reduction in intermolecular repulsion, aggregation and compaction of the nucleohistone gel. The “unit thread” of nucleohistone observed in the electron microscope changes diameter from 110 Å to 230 Å on addition of magnesium before drying. However, X-ray diffraction studies fail to detect any changes in regular tertiary structure on adding divalent ions. The methods for dehydration commonly used in specimen preparation for electron microscopy appear to cause a complete loss of regular tertiary structure.We conclude that the DNA in hydrated nucleohistone is supercoiled, that the degree of regular supercoiling is independent of the presence of ions and that both the 110 Å and 230 Å threads observed in the electron microscope probably contain the distorted remnant of a single supercoil.  相似文献

Aprocedure is described for the synthesis of radioactive 3-methylthiopropionate, a recently isolated metabolite of mammalian methionine metabolism. The method is a two-step synthesis whereby correspondingly labeled methionine is degraded by ninhydrin to 3-methylthiopropionaldehyde and then specifically oxidized to 3-methylthiopropionate without oxidation of the sulfur atom by the yeast enzyme, aldehyde dehydrogenase. Radiochemical purity of the isolated product was established by paper, thin-layer, and gas-liquid chromatography. This procedure is economical and readily applicable to the synthesis of other alkylthio fatty acids for the study of S-methylcysteine and ethionine metabolism and probably for the synthesis of radioactive intermediates of branched chain amino acids.  相似文献

Chicken hepatocytes synthesize glucose and fatty acids at rates which are faster than rat hepatocytes. The former also consume exogenous lactate and pyruvate at a much faster rate and, in contrast to rat hepatocytes, do not accumulate large quantities of lactate and pyruvate by aerobic glycolysis. α-Cyano-4-hydroxycinnamate, an inhibitor of pyruvate transport, causes lactate and pyruvate accumulation by chicken hepatocytes. Glucagon and N6,O2′-dibutyryl adenosine 3′,5′-monophosphate (dibutyryl cyclic AMP) convert pyruvate kinase (EC 2.7.1.40) of rat hepatocytes to a less active form. This effect explains, in part, inhibition of glycolysis, inhibition of lipogenesis, stimulation of gluconeogenesis, and inhibition of the transfer of reducing equivalents from the mitochondrial compartment to the cytoplasmic compartment by these compounds. In contrast, pyruvate kinase of chicken hepatocytes is refractory to inhibition by glucagon or dibutyryl cyclic AMP. Rat liver is known to have predominantly the type L isozyme of pyruvate kinase and chicken liver predominantly the type K. Thus, only the type L isozyme appears subject to interconversion between active and inactive forms by a cyclic AMP-dependent, phosphorylation-dephos-phorylation mechanism. This explains why the transfer of reducing equivalents from the mitochondrial compartment to the cytoplasmic compartment of chicken hepatocytes is insensitive to cyclic AMP. However, glucagon and dibutyryl cyclic AMP inhibit net glucose utilization, inhibit fatty acid synthesis, inhibit lactate and pyruvate accumulation in the presence of α-cyano-4-hydroxycinnamate, and stimulate gluconeogenesis from lactate and dihydroxyacetone by chicken hepatocytes. Thus, a site of action of cyclic AMP distinct from pyruvate kinase must exist in the glycolytic-gluconeogenic pathway of chicken liver.  相似文献

The intravenous injection of killed BCG in an oil-in-saline emulsion (BCG-E) results in the development of intense chronic granulomatous inflammation in the lungs and spleen of C57B1/6 (B6) but not CBA mice. B6 mice injected intravenously with BCG-E exhibited marked suppression of antibody responsiveness and delayed hypersensitivity to sheep erythrocytes, as well as proliferation in response to PPD. In contrast, CBA mice similarly treated with BCG-E were not suppressed in any of these reactivities. The spleen is an important organ in this phenomenon since suppression was reversed by splenectomy and could be transferred to normal recipients with spleen cells from BCG-treated mice. Spleen cells responsible for suppression were adherent to plastic petri plates, removed with carbonyl iron, and were not eliminated with either anti-Thy-1 or anti-immunoglobulin serum + C. This study indicates that macrophages from BCG-inflamed spleen are capable of potent suppression of both antibody- and cellular-mediated immunologic reactivity.  相似文献

Pig heart citrate synthase and mitochondrial malate dehydrogenase interact in polyethylene glycol solutions as indicated by increased solution turbidity. A large percentage of both enzymes sediments when mixtures of the two in polyethylene glycol are centrifuged, whereas little if any of either enzyme sediments in the absence of the other. The observed interaction is highly specific in that neither cytosolic malate dehydrogenase nor nine other proteins showed evidence of specific interaction with either pig heart citrate synthase or mitochondrial malate dehydrogenase. Escherichia coli citrate synthase did not interact with pig heart citrate synthase, but did show evidence of interaction with pig heart mitochondrial malate dehydrogenase. The relation between enzyme behavior in polyethylene glycol solution and in the mitochondrion and the significance of possible in vivo interactions between citrate synthase and mitochondrial malate dehydrogenase are discussed.  相似文献

Dichloroacetate (2 mm) stimulated the conversion of [1-14C]lactate to glucose in hepatocytes from fed rats. In hepatocytes from rats starved for 24 h, where the mitochondrial NADHNAD+ ratio is elevated, dichloroacetate inhibited the conversion of [1-14C]lactate to glucose. Dichloroacetate stimulated 14CO2 production from [1-14C]lactate in both cases. It also completely activated pyruvate dehydrogenase and increased flux through the enzyme. The addition of β-hydroxybutyrate, which elevates the intramitochondrial NADHNAD+ ratio, changed the metabolism of [1-14C]lactate in hepatocytes from fed rats to a pattern similar to that seen in hepatocytes from starved rats. Thus, the effect of dichloroacetate on labeled glucose synthesis from lactate appears to depend on the mitochondrial oxidation-reduction state of the hepatocytes. Glucagon (10 nm) stimulated labeled glucose synthesis from lactate or alanine in hepatocytes from both fed and starved rats and in the absence or presence of dichloroacetate. The hormone had no effect on pyruvate dehydrogenase activity whether or not the enzyme had been activated by dichloroacetate. Thus, it appears that pyruvate dehydrogenase is not involved in the hormonal regulation of gluconeogenesis. Glucagon inhibited the incorporation of 10 mm [1-14C]pyruvate into glucose in hepatocytes from starved rats. This inhibition has been attributed to an inhibition of pyruvate dehydrogenase by the hormone (Zahlten et al., 1973, Proc. Nat. Acad. Sci. USA70, 3213–3218). However, dichloroacetate did not prevent the inhibition of glucose synthesis. Nor did glucagon alter the activity of pyruvate dehydrogenase in homogenates of cells that had been incubated with 10 mm pyruvate in the absence or presence of dichloroacetate. Thus, the inhibition by glucagon of pyruvate gluconeogenesis does not appear to be due to an inhibition of pyruvate dehydrogenase.  相似文献

Kidneys from rabbits and dogs were perfused with one of several DMSO concentrations (0.0, 0.7, 1.4, 2.1 m) in a K+-Mg2+-rich perfusate, frozen, and then thawed with equipment providing electromagnetic (EM) illumination. Electrical properties (dielectric constant and loss tangent) of kidneys were measured both before and after EM thawing. The kidneys thawed were evaluated by simple anatomical (macroscopic and microscopic) and physiological observations rather than by transplantation.Rabbit kidneys which are no thicker than 2 cm could be optimally (uniformly and rapidly) thawed by use of illumination at 2450 MHz, a frequency which has a penetration depth of 2.1 cm at 0 °C, Optimal thawing of canine kidneys, which are twice as thick as rabbit kidneys, required the insertion of steel spheres (electroseeds) into the renal pelvis prior to freezing and illumination at 7 MHz in addition to that at 2450 MHz. Increasing the DMSO concentration (0.0 to 2.1 m) in renal tissue illuminated with 2450 MHz increased the conductivity and the permittivity regardless of whether the renal tissue was frozen or thawed. The use of DMSO decreased the time for thawing with EM illumination and yielded kidneys with improved post-thaw morphology.  相似文献

When isolated frog skeletal muscles were incubated with 14C-labeled adenosine, the nucleoside was rapidly taken up by the cells and was either immediately incorporated into adenine nucleotides or deaminated to inosine. Incorporation was predominant at low (micromolar) concentrations whereas, deamination was the major route of metabolism at high (millimolar) concentrations. When muscles were incubated with 14C-labeled inosine the nucleoside, after entry into the cells, was metabolized to a lesser extent than adenosine. ATP and hypoxanthine were the major products of its metabolism. Intracellular concentrations were calculated using 3H-labeled sorbitol to measure the extracellular space.Because of its lower rate of intracellular metabolism inosine was used to investigate the characteristics of the nucleoside transport system. The uptake of inosine was saturable at high concentrations and was specifically inhibited by the presence of adenosine or uridine in the incubation media. Persantin, a well known specific inhibitor of nucleoside transport, also competitively inhibited inosine uptake, as did theophylline [1, Woo et al. Can J. Physiol. Pharmacol. 52, 1063, 1974]. These data, along with the knowledge that in a well-oxygenated muscle, inosine entry follows a downhill chemical potential gradient, strongly support the view that the transport mechanism is facilitated diffusion.The muscle cell membrane does not appear to be permeable to 14C-labeled ATP under the conditions studied. Investigations of the permeability to the major extracellular degradation products of ATP suggest that AMP was the compound most likely to cross the cell membrane.  相似文献

Human serum transferrin, bovine lactoferrin, and hen conalbumin were investigated with respect to the ability of the bound metal to catalyze thiol oxidation. All three proteins were able to stimulate the oxidation of thiols in both reduced lysozyme and reduced glutathione. The efficiency of the metal in catalyzing thiol oxidation was not decreased by binding to transferrin, suggesting that transferrin-bound metals are completely available to both low and high molecular weight thiols. A 5 × 10?7m concentration of transferrin isolated from serum was able to catalyze the formation of 70% of the theoretical lysozyme activity from reduced inactive lysozyme by 60 min. Increased rates of lysozyme activity formation were observed with copper-saturated transferrin. Decreased lysozyme regeneration rates were observed with the iron-saturated molecule compared to native transferrin. The results presented suggest that metalloproteins may aid in the maintenance of the steady-state cellular concentrations of low molecular weight disulfide by catalyzing the autooxidation of thiols.  相似文献

The activities of ornithine decarboxylase (ODC) and thymidine kinase (TK) and the rates of DNA synthesis were determined in hepatomas and livers of rats bearing Morris hepatoma 5123-C or 7800 and entrained to a schedule of 12 hours of light followed by 12 hours of darkness, with food (60% protein) available only during the first 2 hours of the dark period. ODC activity in hepatoma 5123-C displayed a diurnal oscillation, increasing 2-fold during the feeding period and then rapidly decaying to 20% of the peak level. The livers of rats bearing hepatoma 5123-C exhibited a similar oscillation of ODC activity, with peak values lower than in the hepatomas but higher than in the livers of control (non-tumor bearing) animals. TK activity and the rate of DNA synthesis in hepatoma 5123-C were low during most of the dark period but increased rapidly towards the end of the dark period. DNA synthesis reached a plateau at the dark-light interface and then rapidly declined, but TK activity remained high during the light period. Similar studies on hepatoma 7800 established that ODC activity in this hepatoma did not oscillate but remained at low levels throughout the day. Similarly, host livers of rats bearing hepatoma 7800 did not exhibit the diurnal oscillation of ODC activity characteristic of liver from control rats, but showed a slow increase in activity followed by a plateau and a slow decline to base-line levels. DNA synthesis in hepatoma 7800 was constant throughout the day, whereas TK activity may have increased during the dark period. In the livers of control rats and animals bearing hepatoma 5123-C or 7800, TK activity and rate of DNA synthesis were at low levels at all times studied and appeared not to oscillate.  相似文献

Benzoic acid, p-tert.-butylbenzoic acid, and a structurally related hypolipidemic agent SC-33459 were found to inhibit glucose synthesis by hepatocytes isolated from 48-h fasted rats as well as fatty acid synthesis by hepatocytes isolated from meal-fed rats. Glucose synthesis was less sensitive than fatty acid synthesis. Benzoic acid was the least effective inhibitor of both processes; SC-33459 and p-tert.-butylbenzoic acid were very potent inhibitors with similar efficacy. Glycine prevented the inhibition of fatty acid synthesis caused by benzoic acid, but had no effect on that caused by p-tert.-butylbenzoic acid. Octanoate opposed the inhibitory effects of both benzoic acid and p-tert.-butylbenzoic acid. Oxidation of [1-14C]oleate to ketone bodies and acid-soluble radioactive products was inhibited by both p-tert.-butylbenzoic acid and SC-33459. Preincubation of hepatocytes with SC-33459 was required for the latter effect, suggesting catabolism of this compound may be involved. SC-33459 is a p-tert.-butylphenyl derivative which should be readily converted to p-tert.-butylbenzoic acid by β oxidation. Both p-tert.-butylbenzoic acid and SC-33459 decreased citrate levels dramatically. All three compounds reduced CoA and acetyl-CoA levels and increased medium-chain acyl-CoA ester levels. p-tert.-Butylbenzoic acid and SC-33459 also increased long-chain acyl-CoA ester levels. The increase in medium-chain acyl-CoA levels presumably reflects benzoyl-CoA formation from benzoic acid and p-tert.-butylbenzoyl-CoA formation from p-tert.-butylbenzoic acid and SC-33459. Inhibition of glucose and fatty acid synthesis by these compounds may be due to effects on specific enzymes or to CoA sequestration.  相似文献

We have observed that preincubation of 48 hour-fasted or alloxan diabetic rat liver slices, with no exogenous energy supply, for 3 hours resulted in an increased rate of incorporation of [1-14C] acetate into fatty acids and cholesterol during the following 2 hours. This preincubation effect was enhanced by the presence of glucose (25mM) in or prevented by the addition of dibutyryl cyclic adenosine 3′,5′ monophosphate (10?4M) to the preincubation medium. Preincubation of normal rat liver slices did not change their rate of incorporation of [1-14C] acetate into fatty acids or cholesterol. The rate of 14CO2 synthesized by normal, fasted or diabetic liver slices was little affected by preincubation. The preincubation effect, i.e. enhanced fatty acid synthesis was also observed in suspensions of hepatocytes from fasted and diabetic rats, preincubated for 2 hours, followed by a 1 hour incubation with either [1-14C] acetate or [3H] H2O as precursor. We conclude from these data that there is concurrent and coordinated short- and long-term regulation of fatty acid biosynthesis in fasted and diabetic rat livers. Further, we suggest that the release of inhibition by preincubation of these tissues provides a useful tool for studying the coordinated control  相似文献
