The nucleocytoplasmic hybrid combination performed by nuclear graft between B. bufo nucleus and B. calamita egg is 95% lethal during gastrulation mostly at the young gastrula stage (75%) and to a lesser extent at the yolk plug stage (20%). The heterologous B. calamita cytoplasm was treated or fractioned by different techniques including centrifugation, nucleic acid extraction, ammonium sulfate treatment and heating.
The treated cytoplasmic fractions were injected in B. bufo fertilized eggs in order to check their influence on morphogenetic capacities of the B. bufo nucleus. Injection of hyaline cytoplasmic fraction obtained from B. calamita centrifuged eggs blocks the B. Bufo fertilized egg gastrulation in similar rates as in nucleocytoplasmic association performed by interspecific nuclear grafting. Gastrulation, however, is not affected by injection of total nucleic acids extracted from this hyaline fraction. The soluble cytoplasmic fraction obtained after 60% ammonium sulfate precipitation blocks the development at the beginning of gastrulation, this property being inactivated by heating for 5 min at 60°C. Supernatant from 70% ammonium sulfate precipitation blocks the development at the yolk plug stage and is inactivated by heating for 5 min at 80°C.
These experiments suggest that two distinct factors in B. calamita eggs are involved in developmental arrest of injected B. bufo fertilized eggs, these two factors respectively acting on the beginning and on the end of gastrulation.  相似文献

Egg masses of Meloidogyne incognita and M. hapla were placed in soil at 10, 12, 16, and 20 C. At regular intervals, eggs from samples of egg masses were released from the gelatinous matrices and their developmental stages recorded. The number of days necessary to complete each stage from gastrulation to hatch is given for each temperature. The minimal temperature threshold for the development of eggs was computed by linear regression to be 8.26 C for M. incognita and 6.74 C for M. hapla.  相似文献

The egg-laying rate, number of egg masses, number of eggs/mass, number of eggs hatched/snail and egg viability of Biomphalaria glabrata exposed to different doses (5 and 50) of Echinostoma paraensei miracidia were analyzed as indicators of reproductive activity. Polystyrene plates were placed in aquariums containing the snails and every other day for four weeks after infection the plates were removed to count the number of egg masses and eggs laid. After this, the plates were numbered individually and placed in new aquariums free of snails and the egg masses were observed daily to determine the hatching rate. On average there was an increase in the parameters evaluated in the infected snails in relation to the controls (uninfected snails), except for egg viability, which was significantly lower in the groups infected with 50 miracidia. These findings indicate that when infected, this host snail is able to increase its reproductive activity, suggesting an ecological strategy to maintain the species.  相似文献

The number of young snails emerging from egg masses of Biomphalaria pfeifferi (Kampala strain) is reduced by exposure to an increasing number of miracidia of Schistosoma mansoni (West Nile strain). Snails exposed to four miracidia are rendered sterile. Survival of infected snails is inversely proportional to the number of miracidia to which an individual snail is exposed.  相似文献

The root-knot nematode Meloidogyne incognita was cultured monoxenically on excised tomato roots. Galls and egg masses were observed daily using a light microscope. Two phases were distinguished in the gelatinous matrix of the egg mass: a translucent, amorphous material on the surface of the egg mass and a denser, layered phase in which nematode eggs were deposited. Egg masses were also cryofixed, fractured, and observed as frozen, hydrated specimens on a cold stage in a scanning electron microscope (SEM). In the SEM, the layered phase appeared as a meshwork of fibrils that became more loosely associated as the gelatinous matrix aged: Small pearl-like bodies were observed along the fibers of gelatinous matrix. The egg shell surface and several stages of embryo development, including the one-cell stage, initial cleavages, blastula, gastrula, tadpole stage, elongation, and molt of the first-stage juvenile within the egg shell, were observed and photographed with this technique. The developmental events observed were consistent with those described in other nematode species with different techniques.  相似文献

Gastrulation in the maximum direct developing ascidian Molgula pacifica is highly modified compared with commonly studied "model" ascidians in that endoderm cells situated in the vegetal pole region do not undergo typical invagination and due to the absence of a typical blastopore the involution of mesoderm cells is highly modified. At the gastrula stage, embryos are comprised of a central cluster of large yolky cells that are surrounded by a single layer of ectoderm cells in which there is only a slight indication of an inward movement of cells at the vegetal pole. As a consequence, these embryos do not form an archenteron. In the present study, ultraviolet (UV) irradiation of fertilized eggs tested the possibility that cortical cytoplasmic factors are required for gastrulation, and blastomere isolation experiments tested the possibility that cell signaling beginning at the two-cell stage may be required for the development of the gastrula. Irradiation of unoriented fertilized eggs with UV light resulted in late cleavage stage embryos that failed to undergo gastrulation. When blastomeres were isolated from two-cell embryos, they developed into late cleavage stage embryos; however, they did not undergo gastrulation and subsequently develop into juveniles. These results suggest that cytoplasmic factors required for gastrulation are localized in the egg cortex, but in contrast to previously studied indirect developers, these factors are not exclusively localized in the vegetal pole region at the first stage of ooplasmic segregation. Furthermore, the inability of embryos derived from blastomeres isolated at the two-cell stage to undergo gastrulation and develop into juveniles suggests that important cell signaling begins as early as the two-cell stage in M. pacifica. These results are discussed in terms of the evolution of maximum direct development in ascidians.  相似文献

Eggs of Xenopus laevis were exposed to ultraviolet (uv) radiation (2537 Å) on the vegetal hemisphere soon after fertilization at doses sufficient to impair greatly the subsequent development of dorsal structures. It was found that temporary orientation of irradiated eggs 90° off the natural vertical axis rescues these eggs, allowing them to develop into normal embryos. Complete rescue results when oblique orientation is initiated well before first cleavage, and eggs remain in this position until the 16-cell stage. Significant rescue is seen, however, in eggs which remain off axis for shorter periods of time or when eggs are obliquely oriented, even after first cleavage. Furthermore, a period of oblique orientation prior to uv irradiation results in insensitivity of eggs to irradiation. Ultraviolet irradiation is found to randomize the position of the dorsal side with respect to the sperm entrance point, whereas the position of the dorsal side of rescued embryos is strongly specified by the orientation of the egg during the rescue period, and not by the sperm entrance point. Other effects of uv irradiation on early development include decreased pigmentation differences among 4-cell stage blastomeres and delayed gastrulation. It is proposed (1) that oblique orientation promotes in irradiated eggs a set of internal rearrangements mimicking those normally accomplished by the unirradiated egg in a period prior to first cleavage and as part of an early dorsalization process, and (2) that the uv-sensitive targets are part of the morphogenic machinery used by the egg for internal rearrangements in this period and are not elements of a system of transmitted particulate dorsal determinants.  相似文献

Gene expression regulation during embryo development is under strict regulation to ensure proper gene expression in both time and space. The involvement of microRNAs (miRNA) in early vertebrate development is documented and inactivation of different proteins involved in miRNA synthesis results in severe malformations or even arrests vertebrate embryo development. However, there is very limited information on when and in what tissues the genes encoding these proteins are expressed. Herein, we report a detailed characterization of the expression patterns of DROSHA, DGCR8, XPO5 and DICER1 in the developing chick embryo, from HH1 (when the egg is laid) to HH25 (5-days incubation), using whole mount in situ hybridization and cross-section analysis. We found that these genes are co-expressed in multiple tissues, mostly after stage HH4. Before early gastrulation DICER1 expression was never detected, suggesting the operation of a Dicer-independent pathway for miRNA synthesis. Our results support an important role for miRNAs in vertebrate embryo development and provide the necessary framework to unveil additional roles for these RNA processing proteins in development.  相似文献

Ultraviolet irradiation of the vegetal hemisphere of the fertilized amphibian (Xenopus laevis) egg prior to first cleavage results in the embryo developing an incomplete set of neural structures. The effects of irradiation on various morphogenetic processes, including cell division, formation of the dorsal lip, invagination at gastrulation, and neural induction by the primary organizer, were examined. A decrease in the capacity for invagination during gastrulation and a diminution in the neural inducing capacity of the primary organizer were found to account for defective neurulation in irradiated embryos. Consequently, irradiation of the uncleaved egg leads to interference with the events of both gastrulation and neurulation.  相似文献

The hatchability of eggs and the fecundity and survival of adult Bulinus (Physopsis) africanus was investigated in different salinities. Experimental results revealed egg masses and hatchlings to be considerably more sensitive to salinity than the adult snails. Egg-laying was recorded in salinities 4·5 ‰ and further increases in salinity resulted in a progressive reduction in the hatching success up to a lethal concentration of 5·25 ‰ Survival of these hatchings was adversely affected by salinities as low as 1·0 ‰ and a salinity of 4·5 ‰ was lethal within 6 days. In contrast, adult survival was unaffected in salinities < 3·5 ‰ while further increases in salinity resulted in significant reductions in survival up to a lethal salinity of 8·7 ‰, which caused 100% mortality within 24 h. The survival of B. africanus infected with Schistosoma haematobium and Schistosoma mattheei was lower in the different salinities and control than that of their uninfected counterparts.  相似文献

The present study includes the effect of temperature on the survival of young and adult snails, embryonic development, embryonic growth and egg laying of Indoplanorbis exustus. In Indoplanorbis exustus the bottom lethal temperature was 7.0°C and 7.5°C for young and adults respectively. while the upper lethal temperature was 34.0°C and 32.0°C for young and adult snails respectively. Between the temperatures 12.5°C and 36.5°C the embryonic development was accelerated and the incubation period was shortened. The growth of embryos was found to be faster at 25.0°C. Summary

A study was made of the interaction of the photoperiod and the availability of food in influencing egg laying of the freshwater snail Lymnaea stagnalis. At a long day photoperiod (LD = 16 h light/8 h dark) egg laying of fed snails had increased compared with that of fed animals kept at a medium day photoperiod (MD = 12 h light/12 h dark). In MD snails oviposition ceased within a week of the beginning of a starvation period. This is most probably due to a reduction in the activities of the neuroendocrine caudodorsal cells which secrete an ovulation hormone. In contrast, in starved LD snails a low rate of ovipository activity continued, indicating that a lowered frequency of caudodorsal cell release cycles occurred under these conditions. The decreased mean size (number of eggs) of the egg masses in starved LD snails indicates that the activities of the endocrine dorsal bodies, which control vitellogenesis and synthetic activities in the female accessory sex organs, had decreased.

All MD snails survived, but nearly all LD snails died during the course of the experiment. Determinations of the mantle glycogen stores of LD snails suggest that the high mortality of LD snails is due to exhaustion of the animal's energy reserves.  相似文献

为了弄清福寿螺在自然条件下的繁殖特性,在湖南省资兴市水稻田内进行了为期3年的人工养殖试验.结果表明:福寿螺在湘南自然条件下1年可发生不完全3代.第1代、第2代和第3代(越冬代)雌螺的平均性成熟期分别为59.3、45.4和213.0 d.雄螺的平均性成熟期比雌螺早4.3d.福寿螺的自然性比(♀∶♂)为1.54:1.每次交配持续时间平均为19.2h,但并非每次交配后都会产卵.田间福寿螺月产卵块数与月平均气温呈显著正相关(r=0.756).福寿螺卵块的平均孵化期(d)与日平均气温(℃)呈显著负相关(r=-0.980),其线性回归方程为y=-0.726x+23.064.福寿螺卵块的平均孵化期为20.7d,平均孵化率为44.1%.雌螺与雄螺的平均寿命分别为2.40和1.98 a.平均每头雌螺一生可产卵13764粒,可繁殖幼螺6070头.  相似文献

A polysaccharide preparation, isolated from egg masses deposited by snails of an Ampullarius species, was purified via precipitation with Cetavlon in the presence of sodium borate, and found to contain d-galactose and a smaller proportion of d-glucose, and to have two components with sedimentation coefficients of 10S and 40S. A polysaccharide, isolated from freshly laid egg masses, was highly branched and was shown to contain nonreducing α-d-glucopyranosyl and β-d-galactopyranosyl end-groups, and 3,6-di-O-substituted β-d-galactopyranosyl residues. One or more of the polysaccharide components was a d-glucopyrano-d-galactopyranan with non-reducing α-d-glucopyranosyl end-groups (1→4)-linked to β-d-galactopyranosyl residues. The polysaccharide preparations, obtained from freshly laid egg masses and from those that were left for 10 and 15 days after being laid, were structurally different from each other. With the passage of time, progressive diminution of the 10S component and the proportion of d-glucose in the polysaccharide took place, suggesting that each constituent was consumed preferentially by the snail embryos as an energy source.  相似文献

Egg masses of the European corn borer, Ostrinia nubilalis, were collected from caged corn plants to study the bacterial diseases of egg masses and first-instar larvae. In 1972, a reduction in the percentage hatch, indicative of a disease epizootic, was noted among the first-generation egg masses. Bioassays of the bacteria on first-instar larvae and egg masses showed that Bacillus thuringiensis var. kurstaki was pathogenic only to larvae. The differences in the lethal freezing temperatures of the foot muscles of the marine snails used in this study were related to the vertical distributions of the snails on the shore. The muscles of the subtidal species Thais lapillus and Nassarius obsoletus were injured at temperatures that were significantly higher than those of the muscles of the intertidal species Littorina obtusata, Littorina littorea, and Littorina saxatilis. The lethal freezing temperatures also varied among the intertidal species. The foot muscle of the high-intertidal species, L. saxatilis, was injured at a significantly lower temperature than the foot muscles of the low-intertidal species L. obtusata.Calorimetry was used to show that the differences in the lethal freezing temperatures between the subtidal and intertidal snails were related to the amounts of tissue ice formed. The ability of the muscles of the intertidal snails to tolerate lower subfreezing temperatures was associated with an increased tolerance to greater quantities of tissue ice. In contrast, the differences in the lethal freezing temperatures among the intertidal species were independent of the amounts of tissue ice formed. The percentage of water frozen in the muscles of these snails at their respective lethal freezing temperatures were not significantly different and were equal to 82%. Thus, the physiological mechanism responsible for the differences in the lethal freezing temperatures of the muscles of the intertidal snails is associated with an increased tolerance to a factor other than the amounts of tissue ice formed.  相似文献
