The stereochemical reaction course for the two C-3 hydrogens of leucine to produce a characteristic isoprenoidal lipid in halophilic archaea was observed using incubation experiments with whole cell Halobacterium salinarum. Deuterium-labeled (3R)- and (3S)-[3-2H]leucine were freshly prepared as substrates from 2,3-epoxy-4-methyl-1-pentanol. Incorporation of deuterium from (3S)-[3-2H]leucine and loss of deuterium from (3R)-[3-2H]leucine in the lipid-core of H. salinarum was observed. Taken together with the results of our previous report, involving the incubation of chiral-labeled [5-2H]leucine, these results strongly suggested an involvement of isovaleryl-CoA dehydrogenase in leucine conversion to isoprenoid lipid in halophilic archaea. The stereochemical course of the reaction (anti-elimination) might have been the same as that previously reported for mammalian enzyme reactions. Thus, these results suggested that branched amino acids were metabolized to mevalonate in archaea in a manner similar to other organisms.  相似文献

Moderately halophilic bacteria are a kind of extreme environment microorganism that can tolerate moderate salt concentrations ranging from 0.5 M to 2.5 M. Here, via a metagenomic library screen, we identified four putative Na+ transporters, designated H7-Nha, H16-Mppe, H19-Cap and H35-Mrp, from moderately halophilic community in the hypersaline soil of Huamachi Salt Lake, China. Functional complementation observed in a Na+(Ca2+)/H+ antiporter-defective Escherichia coli mutant (KNabc) suggests that the four putative Na+ transporters could confer cells a capacity of Na+ resistance probably by enhancing Na+ or Ca2+ efflux, but not Li+ or K+ exchange. Blastp analysis of the deduced amino-acid sequences indicates that H7-Nha has 71% identity to the NhaG Na+/H+ antiporter of Bacillus subtilis, while H19-Cap shows 99% identity to Enterobacter cloacae Ca2+ antiporter. Interestingly, H16-Mppe shares 59% identity to the metallophosphoesterase of Bacillus cellulosilyticus and H35-Mrp shows 68% identity to multidrug resistance protein of Lysinibacillus sphaericus. This is the first report that predicts a potential role of metallophosphoesterase in Na+ resistance in halophilic bacteria. Furthermore, everted membrane vesicles prepared from E. coli cells harboring H7-Nha exhibit Na+/H+ antiporter activity, but not Li+ (K+)/H+ antiporter activity, confirming that H7-Nha supports Na+ resistance mainly via Na+/H+ antiport. Our report also demonstrates that metagenomic library screen is a convenient and effective way to explore more novel types of Na+ transporters.  相似文献

The halophilic archaea Halococcus (Hc.) saccharolyticus, Haloferax (Hf.) volcanii, and Halorubrum (Hr.) saccharovorum were found to generate acetate during growth on glucose and to utilize acetate as a growth substrate. The mechanisms of acetate formation from acetyl-CoA and of acetate activation to acetyl-CoA were studied. Hc. saccharolyticus, exponentially growing on complex medium with glucose, formed acetate and contained ADP-forming acetyl-CoA synthetase (ADP-ACS) rather than acetate kinase and phosphate acetyltransferase or AMP-forming acetyl-CoA synthetase. In the stationary phase, the excreted acetate was completely consumed, and cells contained AMP-forming acetyl-CoA synthetase (AMP-ACS) and a significantly reduced ADP-ACS activity. Hc. saccharolyticus, grown on acetate as carbon and energy source, contained only AMP-ACS rather than ADP-ACS or acetate kinase. Cell suspensions of Hc. saccharolyticus metabolized acetate only when they contained AMP-ACS activity, i.e., when they were obtained after growth on acetate or from the stationary phase after growth on glucose. Suspensions of exponential glucose-grown cells, containing only ADP-ACS but not AMP-ACS, did not consume acetate. Similar results were obtained for the phylogenetic distantly related halophilic archaea Hf. volcanii and Hf. saccharovorum. We conclude that, in halophilic archaea, the formation of acetate from acetyl-CoA is catalyzed by ADP-ACS, whereas the activation of acetate to acetyl-CoA is mediated by an inducible AMP-ACS.Abbreviations.  The reaction of the macrocycles 1,4,7-tris (3,5-di-tert-butyl-2-hydroxy-benzyl)-1,4,7-triazacyclononane, L1H3, or 1,4,7-tris(3-tert-butyl-5-methoxy-2-hydroxy-benzyl)-1,4,7-triazacyclononane, L2H3, with Cu(ClO4)2·6H2O in methanol (in the presence of Et3N) affords the green complexes [CuII(L1H)] (1), [CuII(L2H)]·CH3OH (2) and (in the presence of HClO4) [CuII(L1H2)](ClO4) (3) and [CuII(L2H2)] (ClO4) (4). The CuII ions in these complexes are five-coordinate (square-base pyramidal), and each contains a dangling, uncoordinated pendent arm (phenol). Complexes 1 and 2 contain two equatorially coordinated phenolato ligands, whereas in 3 and 4 one of these is protonated, affording a coordinated phenol. Electrochemically, these complexes can be oxidized by one electron, generating the phenoxyl-copper(II) species [CuII(L1H)]+·, [Cu(L2H)]+·, [CuII(L1H2)]2+·, and [CuII(L2H2)]2+·, all of which are EPR-silent. These species are excellent models for the active form of the enzyme galactose oxidase (GO). Their spectroscopic features (UV-VIS, resonance Raman) are very similar to those reported for GO and unambiguously show that the complexes are phenoxyl-copper(II) rather than phenolato-copper(III) species. Background  

DNA ligases are required for DNA strand joining in all forms of cellular life. NAD+-dependent DNA ligases are found primarily in eubacteria but also in some eukaryotic viruses, bacteriophage and archaea. Among the archaeal NAD+-dependent DNA ligases is the LigN enzyme of the halophilic euryarchaeon Haloferax volcanii, the gene for which was apparently acquired by Hfx.volcanii through lateral gene transfer (LGT) from a halophilic eubacterium. Genetic studies show that the LGT-acquired LigN enzyme shares an essential function with the native Hfx.volcanii ATP-dependent DNA ligase protein LigA.  相似文献

Adaptive capacity of bacteria and archaea from salt lakes of the Altai Region are discussed. It is established that halophilic archaea (genus Halorubrum) and halotolerant bacteria (genus Halomonas) grow in a wide range of pH and mineralization (in the presence of Cl?, SO 4 2? , ClO 4 ? , Mg2+) and survive at low temperatures with a minor decrease in viability.  相似文献

In order to ascertain the existence of polyol dehydrogenase which seemed to take a part in polyalcohol production by Pichia miso, the preparation and the partial purification of polyol dehydrogenase from the cells of Pichia miso were carried out. Some properties of this enzyme preparation and the identification of the products formed by this enzyme action were also described. This enzyme preparation was found to catalyze the following reactions:

In spite of their common hypersaline environment, halophilic archaea are surprisingly different in their nutritional demands and metabolic pathways. The metabolic diversity of halophilic archaea was investigated at the genomic level through systematic metabolic reconstruction and comparative analysis of four completely sequenced species: Halobacterium salinarum, Haloarcula marismortui, Haloquadratum walsbyi, and the haloalkaliphile Natronomonas pharaonis. The comparative study reveals different sets of enzyme genes amongst halophilic archaea, e.g. in glycerol degradation, pentose metabolism, and folate synthesis. The carefully assessed metabolic data represent a reliable resource for future system biology approaches as it also links to current experimental data on (halo)archaea from the literature. Background and aims

The pH of the growth medium influences Cu speciation in solution, the negativity of plasma membrane (PM) surface potential, and hence the rhizotoxicity of Cu.

Methods

Solution culture experiments were conducted with wheat (Triticum aestivum L.) seedlings to examine the toxicity of various Cu species at pH values ranging from 4.50 to 8.25. The toxic species of Cu was identified, giving particular consideration to the electrical properties at the plant cell membrane and ion activities at the PM surface.

Results

The solution culture studies showed that at pH?<?6.60 (i.e., free Cu2+ >95 % of total Cu), the addition of cations (Ca2+ or H+) decreased the toxic effects of Cu by decreasing the negativity of the PM surface potential (and hence decreasing the activity of Cu2+ at the PM surface). For solutions with pH values from 7.50 to 8.25 (CuCO 03 >50 % of total Cu), an increase in pH significantly enhanced the toxicity of Cu, whilst the addition of Ca had negligible influence on toxicity.

Conclusions

Root growth in solution cultures was influenced primarily by the surface activities of free Cu2+ and CuCO 03 . Across all experiments, the data indicate that it was CuCO 03 , rather than CuOH+, that contributed Cu toxicity over pH?>?7.00. Although our data do not explore the mechanism of toxicity, we propose that CuCO 03 has an important role in Cu rhizotoxicity in alkaline growth media.

Proton (H+) and aluminum (Al3+) toxicities are major factors limiting crop production on acid soils. To study whether salicylic acid (SA) is functional in alleviating protein damage caused by H+ and Al3+ toxicities, an investigation of the antioxidant defense response regulated by SA was carried out on barley (Hordeum vulgare L.) seedlings under H+, Al3+, and combined stresses. It was found that the relative root elongation of seedlings, which grew in the solutions supplemented with SA, was significantly higher than that of seedlings without SA treatment after 24-h treatments with H+, Al3+, and combined stresses. The lesser amount of carbonylated proteins with molecular weights ranging from 14.4 to 97 kD, was accumulated in seedlings treated with SA than that in the seedlings without SA treatment. The higher activities of antioxidant enzymes and lesser content of MDA were observed in seedlings treated with SA compared with the seedlings without SA treatment. Moreover, the nitroblue tetrazolium staining of roots showed that ROS accumulation was decreased by SA treatments. This study suggested that SA could alleviate cell damage caused by H+ and Al3+ toxicities on acid soils by both activating antioxidant defense responses and reducing the contents of carbonylated proteins caused by ROS in barley seedlings.  相似文献

Halophilic archaea thriving in hypersaline environments, such as salt lakes, offer models for putative life in extraterrestrial brines such as those found on Mars. However, little is known about the effect of the chaotropic salts that could be found in such brines, such as MgCl2, CaCl2 and (per)chlorate salts, on complex biological samples like cell lysates which could be expected to be more representative of biomarkers left behind putative extraterrestrial life forms. We used intrinsic fluorescence to study the salt dependence of proteomes extracted from five halophilic strains: Haloarcula marismortui, Halobacterium salinarum, Haloferax mediterranei, Halorubrum sodomense and Haloferax volcanii. These strains were isolated from Earth environments with different salt compositions. Among the five strains that were analysed, H. mediterranei stood out as a results of its high dependency on NaCl for its proteome stabilization. Interestingly, the results showed contrasting denaturation responses of the proteomes to chaotropic salts. In particular, the proteomes of strains that are most dependent or tolerant on MgCl2 for growth exhibited higher tolerance towards chaotropic salts that are abundant in terrestrial and Martian brines. These experiments bridge together global protein properties and environmental adaptation and help guide the search for protein-like biomarkers in extraterrestrial briny environments.  相似文献

The effect of the putative K+/H+ ionophore, nigericin on the internal Na+ concentration ([Na i ]), the internal pH (pH i ), the internal Ca2+ concentration ([Ca i ]) and the baseline release of the neurotransmitter, GABA was investigated in Na+-binding benzofuran isophtalate acetoxymethyl ester (SBFIAM), 2′,7′-bis(carboxyethyl)-5(6) carboxyfluorescein acetoxymethyl ester (BCECF-AM), fura-2 and [3H]GABA loaded synaptosomes, respectively. In the presence of Na+ at a physiological concentration (147 mM), nigericin (0.5 μM) elevates [Na i ] from 20 to 50 mM, increases thepH i , 0.16 pH units, elevates four fold the [Ca i ] at expense of external Ca2+ and markedly increases (more than five fold) the release of [3H]GABA. In the absence of a Na+ concentration gradient (i.e. when the external Na+ concentration equals the [Na i ]), the same concentration (0.5 μM) of nigericin causes the opposite effect on thepH i (acidifies the synaptosomal interior), does not modify the [Na i ] and is practically unable to elevate the [Ca i ] or to increase [3H]GABA release. Only with higher concentrations of nigericin than 0.5 μM the ionophore is able to elevate the [Ca i ] and to increase the release of [3H]GABA under the conditions in which the net Na+ movements are eliminated. These results clearly show that under physiological conditions (147 mM external Na+) nigericin behaves as a Na+/H+ ionophore, and all its effects are triggered by the entrance of Na+ in exchange for H+ through the ionophore itself. Nigericin behaves as a K+/H+ ionophore in synaptosomes just when the net Na+ movements are eliminated (i.e. under conditions in which the external and the internal Na+ concentrations are equal). In summary care must be taken when using the putative K+/H+ ionophore nigericin as an experimental tool in synaptosomes, as under standard conditions (i.e. in the presence of high external Na+) nigericin behaves as a Na+/H+ ionophore.  相似文献

Rapid calcium exchange for protons and potassium in cell walls of Chara   总被引:3,自引:2,他引:1  
Net fluxes of Ca2+, H+ and K+ were measured from intact Chara australis cells and from isolated cell walls, using ion-selective microelectrodes. In both systems, a stimulation in Ca2+ efflux (up to 100 nmol m?2 s?1, from an influx of ~40 nmol m?2 s?1) was detected as the H+ or K+ concentration was progressively increased in the bathing solution (pH 7.0 to 4.6 or K+ 0.2 to 10mol m?3, respectively). A Ca2+ influx of similar size occurred following the reverse changes. These fluxes decayed exponentially with a time constant of about 10 min. The threshold pH for Ca2+ efflux (pH 5.2) is similar to a reported pH threshold for acid-induced wall extensibility in a closely related characean species. Application of NH4+ to intact cells caused prolonged H+ efflux and also transient Ca2+ efflux. We attribute all these net Ca2+ fluxes to exchange in the wall with H+ or K+. A theoretical treatment of the cell wall ion exchanges, using the ‘weak acid Donnan Manning’ (WADM) model, is given and it agrees well with the data. The role of Ca2+ in the cell wall and the effect of Ca2+ exchanges on the measured fluxes of other ions, including bathing medium acidification by H+ efflux, are discussed.  相似文献

Methyl jasmonate (MeJA) elicits stomatal closure in many plant species. Stomatal closure is accompanied by large ion fluxes across the plasma membrane (PM). Here, we recorded the transmembrane ion fluxes of H+, Ca2+ and K+ in guard cells of wild‐type (Col‐0) Arabidopsis, the CORONATINE INSENSITIVE1 (COI1) mutant coi1‐1 and the PM H+‐ATPase mutants aha1‐6 and aha1‐7, using a non‐invasive micro‐test technique. We showed that MeJA induced transmembrane H+ efflux, Ca2+ influx and K+ efflux across the PM of Col‐0 guard cells. However, this ion transport was abolished in coi1‐1 guard cells, suggesting that MeJA‐induced transmembrane ion flux requires COI1. Furthermore, the H+ efflux and Ca2+ influx in Col‐0 guard cells was impaired by vanadate pre‐treatment or PM H+‐ATPase mutation, suggesting that the rapid H+ efflux mediated by PM H+‐ATPases could function upstream of the Ca2+ flux. After the rapid H+ efflux, the Col‐0 guard cells had a longer oscillation period than before MeJA treatment, indicating that the activity of the PM H+‐ATPase was reduced. Finally, the elevation of cytosolic Ca2+ concentration and the depolarized PM drive the efflux of K+ from the cell, resulting in loss of turgor and closure of the stomata.  相似文献

【目的】从嗜盐古菌中筛选可产生生物絮凝剂的菌株,对发酵液、上清液、菌悬液、胞外聚合物的絮凝作用进行检测,筛选能够适应高盐废水处理,且具有广谱盐度及pH作用范围的微生物絮凝剂。【方法】以新疆乌勇布拉克干盐湖沉积物为研究对象,利用纯培养方法对嗜盐古菌进行分离,对絮凝菌株进行初筛及16S rRNA基因测序,构建系统进化树,初步判断菌株分类地位;复筛检测不同生物材料的絮凝效果;选择絮凝效果较好的生物材料,检测其盐度、pH的絮凝效果稳定性。【结果】采用纯培养方法共分离到28株嗜盐古菌,絮凝初筛共筛选出16株嗜盐古菌,分布于碱线菌属(Natrinema)、盐缓长菌属(Halopiger)和盐土生菌属(Haloterrigena)。菌株发酵液、上清液、菌悬液、胞外聚合物具有不同程度的絮凝效果。菌株A279-1、A133、RP33、NGA0064、RM-152、A389的发酵液、上清液的絮凝效果较好,其中菌株A389的发酵液絮凝率为61.06%,上清液为67.92%。所有菌株菌悬液的絮凝率达到80%以上。菌株所产胞外聚合物表现出较好的絮凝效果,菌株RM-152所产胞外聚合物的絮凝率最高,达89.86%,其次是A389 (81.53%)。菌株A389所产胞外聚合物的产量最大,达12.53 g/L,具有广泛的盐度和pH适应性。【结论】乌勇布拉克干盐湖沉积物中蕴含丰富的可产生微生物絮凝剂的嗜盐古菌资源。嗜盐古菌菌株发酵液、上清液、菌悬液及胞外聚合物均具有良好的絮凝作用,尤其是胞外聚合物表现出较好的絮凝效果,具有广谱的盐度和pH耐受性。嗜盐古菌所产生物絮凝剂的发现对于后续高盐废水功能材料开发具有重要应用价值。  相似文献

High concentration (1.0 M) of KSCN, but not of NaSCN, induced lysis of slightly halophilic Vibrio alginolyticus and moderately halophilic Vibrio costicolus, and the decrease in absorbance of the cell suspension was complete after 30 min at 25°C. Replacement of K+ with Na+ effectively prevented the lysis by SCN. K+ salts of NO3, Br, however, induced no significant lysis. In electron micrographs, a prolonged exposure of the cells of V. alginolyticus to 1.0 M KSCN displaced the nucleoplasm to maintain close contact with the cell membranes. After 40 min of interaction, 50% of the cellular protein, 96% of RNA and 94% of DNA were recovered in the lysed cells. In contrast to lysis in hypotonic conditions, the lysis induced by KSCN is due mainly to a partial release of protein from the cells. V. costicolus was more susceptible to SCN than V. alginolyticus, whereas nonhalophilic Escherichia coli was resistant to 1.0 M KSCN. Thus, lysis by SCN is characteristic of halophilic bacteria and cell membranes of more halophilic bacteria are more susceptible to chaotropic anions. The protective effect of Na+ observed here was considered to be manifested by specific interactions of Na+ with components of cell membranes, thereby rendering their structures resistant to the action of chaotropic anions.  相似文献

Abstract

Treatment of 4-(D-gluco-pentitol-l-y1)-2-pheny1–2H-1,2,3-triazole (1) with p-toluenesulfonyl chloride in pyridine solution, afforded the homo-C-nucleoside analog, 4-(2,5-anhydro-D-gluco-pentitol-1-yl)-2-phenyl-2H-1,2,3-triazole (2) as well as its partial p-toluenesulfonyl derivative (3). 4-(5-Chloro-5-deoxy-D-gluco-pentitol-1-yl)-2-phenyl-2H-1,2,3-triazole (8), was isolated as a byproduct from the reaction. The structure and anomeric configuration of 2 was determined by acylation, 1H, 13C NMR, and NOE, spectroscopy as well as mass spectrometry.

Abstract

A series of 5,7-dimethyl-oxazolo[5,4-d]pyrimidine-4,6(5H,7H)-dione derivatives, N5a–5l, was designed, synthesized and evaluated for their FGFR1-inhibition ability as well as cytotoxicity against three cancer cell lines (H460, B16F10 and A549) in vitro. Several compounds displayed good-to-excellent potency against these cancer cell lines compared to SU5402. Structure–activity relationship analyses indicated that compounds with a rigid structure and more heteroatoms at the side chain of the parent ring were more effective than those without these substitutions. The compound N5g (37.4% FGFR1 inhibition at 1.0?μM) was identified to have the most potent antitumor activities, with IC50 values of 5.472, 4.260 and 5.837?μM against H460, B16F10 and A549 cell lines, respectively. Together, our results suggest that 5,7-dimethyl-oxazolo[5,4-d]pyrimidine-4,6(5H,7H)-dione derivatives may serve as potential agents for the treatment of FGFR1-mediated cancers.  相似文献

Abstract

The formation of sparingly soluble Ca2+-phytate (Phy) species was studied using several techniques (potentiometry, calorimetry, thermoanalysis, ICP, SEM). Species with empirical formula CaxH(12_2x)Phy, (6 ≤ x ≤ 4.5) were formed in experiments performed at pH = 5.0, 7.5 and 10.5; at pH = 10.5 and in excess of Na+ the species Ca5Na2Phy was also found. Solubilities (s = 1.0 x 10-9and 1.1 x 10-10mol L-1 for the species Ca6Phy and Ca5Na2Phy respectively, at l = 0 mol L-1), thermogravimetric profiles (dehydratation, monohydrogen-phosphate and final pyrophosphate formation), and formation enthalpies (ΔΗ = 7.6 ± 1 and 3.7 ± 0.8 kJ mol-1for the equilibria 6Ca2+ + H3L9- = Ca6L0 + 3 H+ and 5Ca2+ + 2Na+ + H3L9- = Ca5Na2L0 + 3 H+, respectively), were obtained.  相似文献
