Chytridiomycosis is an emerging infectious disease caused by the pathogen Batrachochytrium dendrobatidis (Bd) and is the cause of the decline and extinction of amphibian species throughout the world. We surveyed the distribution of Bd within and around the Tasmanian Wilderness World Heritage Area (TWWHA), a 1.38 million ha area of significant fauna conservation value, which provides the majority of habitat for Tasmania's 3 endemic frog species (Litoria burrowsae, Bryobatrachus nimbus and Crinia tasmaniensis). Bd was detected at only 1 (3%) of the 33 sites surveyed within the TWWHA and at 15 (52%) of the 29 sites surveyed surrounding the TWWHA. The relatively low incidence of the disease within the TWWHA suggests that the majority of the TWWHA is currently free of the pathogen despite the fact that the region provides what appears to be optimal conditions for the persistence of Bd. For all survey sites within and around the TWWHA, the presence of Bd was strongly associated with the presence of gravel roads, forest and < 1000 m altitude--factors that in this study were associated with human-disturbed landscapes around the TWWHA. Conversely, the presence of walking tracks was strongly associated with the absence of Bd, suggesting an association of absence with relatively remote locations. The wide distribution of Bd in areas of Tasmania with high levels of human disturbance and its very limited occurrence in remote wilderness areas suggests that anthropogenic activities may facilitate the dissemination of the pathogen on a landscape scale in Tasmania. Because the majority of the TWWHA is not readily accessible and appears to be largely free of Bd, and because Tasmanian frogs reproduce in ponds rather than streams, it may be feasible to control the spread of the disease in the TWWHA. 相似文献

The dissemination of the virulent pathogen Batrachochytrium dendrobatidis (Bd) has contributed to the decline and extinction of many amphibian species worldwide. Several different strains have been identified, some of which are sympatric. Interactions between co-infecting strains of a pathogen can have significant influences on disease epidemiology and evolution; therefore the dynamics of multi-strain infections is an important area of research. We stained Bd cells with 2 fluorescent BODIPY fatty acid probes to determine whether these can potentially be used to distinguish and track Bd cell lines in multi-strain experiments. Bd cells in broth culture were stained with 5 concentrations of green-fluorescent BODIPY FL and red-fluorescent BODIPY 558/568 and visualised under an epifluorescent microscope for up to 16 d post-dye. Dyed strains were also assessed for growth inhibition. The most effective concentration for both dyes was 10 pM. This concentration of dye produced strong fluorescence for 12 to 16 d in Bd cultures held at 23 degrees C (3 to 4 generations), and did not inhibit Bd growth. Cells dyed with BODIPY FL and BODIPY 558/568 can be distinguished from each other on the basis of their fluorescence characteristics. Disease can be an important driver of host population dynamics and epizootics can cause severe host population declines. Batrachochytrium dendrobatidis (Bd), the pathogen causing amphibian chytridiomycosis, may occur epizootically or enzootically and can harm amphibian populations in many ways. While effects of Bd epizootics are well documented, the effects of enzootic Bd have rarely been described. We used a state-space model that accounts for observation error to test whether population trends of a species highly susceptible to Bd, the midwife toad Alytes obstetricans, are negatively affected by the enzootic presence of the pathogen. Unexpectedly, Bd had no negative effect on population growth rates from 2002-2008. This suggests that negative effects of disease on individuals do not necessarily translate into negative effects at the population level. Populations of amphibian species that are susceptible to the emerging disease chytridiomycosis can persist despite the enzootic presence of the pathogen under current environmental conditions. 相似文献

We studied the historical prevalence of the invasive and pathogenic chytrid fungus Batrachochytrium dendrobatidis (Bd) among amphibians from the Bolivian Andes. Our aim was also to determine its geographic pattern of dispersion, and a potential host taxonomic signature. We collected frog tissue samples from nine museum collections covering a period from 1863 to 2005 and from the field during 2009–2016. Bd was diagnosed via quantitative PCR in 599 individuals of 17 genera and 54 species. We found an overall Bd prevalence of 41% among 44 species tested. The first incidence of Bd was from a Telmatobius culeus in 1863; this is the earliest report of detection for this pathogen in the world. Results reveal a non-random historical and geographical pattern of Bd occurrence and amphibian declines that suggests the presence of two different invasive strains, an ancient endemic and a more recent introduction. Prevalence of Bd increased significantly by the mid-1990s, particularly in the cloud-forests, and this is coincident with the timing of drastic amphibian declines. In contrast, amphibians occurring in drier altiplano habitats have persisted in spite of Bd presence. We hypothesize that the early 1990s, and the cloud-forests in central Bolivia were the center of an epidemic surge of Bd that took its toll on many species, especially in the genus Telmatobius. Further sampling of cloud-forest species, and ongoing genetic studies of Bd isolates from Bolivia should help resolve the history of this invasive pathogen and test hypotheses on the differential response of endangered hosts. 相似文献

Since the early 1980s, the southern corroboree frog Pseudophryne corroboree and northern corroboree frog P. pengilleyi have been in a state of decline from their sub-alpine and high montane bog environments on the southern tablelands of New South Wales, Australia. To date, there has been no adequate explanation as to what is causing the decline of these species. We investigated the possibility that a pathogen associated with other recent frog declines in Australia, the amphibian chytrid fungus Batrachochytrium dendrobatidis, may have been implicated in the decline of the corroboree frogs. We used histology of toe material and real-time PCR of skin swabs to investigate the presence and infection rates with B. dendrobatidis in historic and extant populations of both corroboree frog species. Using histology, we did not detect any B. dendrobatidis infections in corroboree frog populations prior to their decline. However, using the same technique, high rates of infection were observed in populations of both species after the onset of substantial population declines. The real-time PCR screening of skin swabs identified high overall infection rates in extant populations of P. corroboree (between 44 and 59%), while significantly lower rates of infection were observed in low-altitude P. pengilleyi populations (14%). These results suggest that the initial and continued decline of the corroboree frogs may well be attributed to the emergence of B. dendrobatidis in populations of these species. 相似文献

The pathogenic fungus Batrachochytrium dendrobatidis (Bd) was recently detected in Missouri hellbender Cryptobranchus alleganiensis populations that have declined precipitously for unclear reasons. The objective of this study was to determine whether Bd occurred historically in Missouri hellbender populations or is a relatively novel occurrence. Epidermal tissue was removed from 216 archived hellbenders collected from 7 Missouri streams between 1896 and 1994. Histological techniques and an immunoperoxidase stain were used to confirm historic occurrence of Bd infection in hellbenders from the North Fork of the White (1969, 1973, 1975), Meramec (1975, 1986), Big Piney (1986), and Current rivers (1988). Bd was not detected in hellbenders from the Niangua, Gasconade or Eleven Point rivers. The study detected no evidence for endemism of Bd in Missouri hellbender populations prior to 1969, despite the fact that nearly one third of the hellbenders sampled were collected earlier. Our findings are consistent with the hypothesis that Bd is a non-endemic pathogen in North America that was introduced in the second half of the twentieth century. 相似文献

Chytridiomycosis, caused by the amphibian chytrid fungus Batrachochytrium dendrobatidis (Bd), is an emerging infectious disease responsible for amphibian declines on several continents. In laboratory conditions, optimal temperatures for Bd growth and survivorship are between 17 and 25 degrees C. We investigated the effect of different storage temperatures, both in field and laboratory conditions, on detection of Bd from swabs stored for 7 d. We sampled 52 wild Litoria wilcoxii males for Bd by simultaneously running 2 cotton swabs along the skin of the frog. One group of swabs was stored in a freezer within 2 h of sampling and the other was kept in a car in an exposed environment for 7 d before being stored in the freezer. In the laboratory experiment, swabs were inoculated with zoospores of Bd and underwent one of 4 treatments: immediate DNA extraction, or storage at 27, 38 or 45 degrees C for 7 d prior to DNA extraction. Swabs from all treatments were analyzed by quantitative (real-time) PCR test. Though prevalence of Bd did not differ significantly between swabs that were frozen and those that remained in a car for 7 d (19.2 vs. 17.3%, respectively), the number of Bd zoospores detected on car swabs taken from infected frogs was, on average, 67% less than that detected on the corresponding frozen swab. In the laboratory experiment, the number of zoospore equivalents varied significantly with treatment (F(3,35) = 4.769, p = 0.007), indicating that there was reduced recovery of Bd DNA from swabs stored at higher temperatures compared with those stored at lower temperatures or processed immediately. We conclude that failure to store swabs in cool conditions can result in a significant reduction in the amount of Bd DNA detected using the PCR assay. Dramatic declines and extinctions of amphibian populations throughout the world have been associated with chytridiomycosis, an infectious disease caused by the pathogenic chytrid fungus Batrachochytrium dendrobatidis (Bd). Previous studies indicated that Bd prevalence correlates with cooler temperatures in the field, and laboratory experiments have demonstrated that Bd ceases growth at temperatures above 28°C. Here we investigate how small-scale variations in water temperature correlate with Bd prevalence in the wild. We sampled 221 amphibians, including 201 lowland leopard frogs (Rana [Lithobates] yavapaiensis), from 12 sites in Arizona, USA, and tested them for Bd. Amphibians were encountered in microhabitats that exhibited a wide range of water temperatures (10-50°C), including several geothermal water sources. There was a strong inverse correlation between the water temperature in which lowland leopard frogs were captured and Bd prevalence, even after taking into account the influence of year, season, and host size. In locations where Bd was known to be present, the prevalence of Bd infections dropped from 75-100% in water <15°C, to less than 10% in water >30°C. A strong inverse correlation between Bd infection status and water temperature was also observed within sites. Our findings suggest that microhabitats where water temperatures exceed 30°C provide lowland leopard frogs with significant protection from Bd, which could have important implications for disease dynamics, as well as management applications.There must be quite a few things a hot bath won't cure, but I don't know many of them--Sylvia Plath, "The Bell Jar" (1963). 相似文献

The underlying functional neuroanatomy of tinnitus remains poorly understood. Few studies have focused on functional cerebral connectivity changes in tinnitus patients. The aim of this study was to test if functional MRI "resting-state" connectivity patterns in auditory network differ between tinnitus patients and normal controls. Thirteen chronic tinnitus subjects and fifteen age-matched healthy controls were studied on a 3 tesla MRI. Connectivity was investigated using independent component analysis and an automated component selection approach taking into account the spatial and temporal properties of each component. Connectivity in extra-auditory regions such as brainstem, basal ganglia/NAc, cerebellum, parahippocampal, right prefrontal, parietal, and sensorimotor areas was found to be increased in tinnitus subjects. The right primary auditory cortex, left prefrontal, left fusiform gyrus, and bilateral occipital regions showed a decreased connectivity in tinnitus. These results show that there is a modification of cortical and subcortical functional connectivity in tinnitus encompassing attentional, mnemonic, and emotional networks. Our data corroborate the hypothesized implication of non-auditory regions in tinnitus physiopathology and suggest that various regions of the brain seem involved in the persistent awareness of the phenomenon as well as in the development of the associated distress leading to disabling chronic tinnitus. 相似文献

【目的】探索我国粘虫Mythimnaseparata(Walker)种群周年北迁南回的动态规律。【方法】2014-2017年分别在全国16、25、24、23个省(市、区)17、28、27、26个县(市、区)设立了17、35、34、31台高空测报灯,系统观测了全年或主要发生期逐日蛾量。【结果】基本掌握了粘虫在我国全年各代发生区域、发生时间和发生数量,为全国区域预报提供了重要依据。【结论】我国华南江南、西南、长江中下游、黄淮、华北、东北和西北等区域是我国粘虫成虫发生区域,每年循环发生5个代次;30?N以南区域诱测到越冬粘虫,27?N及以南的观测点虫量较多,冬季未观测到区域间虫源交流;浙江象山、上海奉贤、江苏东台、山东长岛、河北滦县是粘虫北迁南回的主要路径;华南江南、长江中下游、黄淮、华北、东北等区域间观测到虫源交流,华北和东北地区2代、3代可进行虫源交流;东南亚国家或许为我国提供较多虫源,我国粘虫存在种群重建的可能,这有待于今后进行国际合作、深入研究。 相似文献

Modelling the influence of landscape connectivity on animal distribution: a functional grain approach 总被引:1,自引:0,他引:1
Landscape change may reduce the connectivity of landscapes and impact the movement of animals. If movement processes have been influenced by landscape connectivity, we hypothesize that animals may distribute themselves in larger connected regions of the landscape in order to minimize the movement costs associated with obtaining required resources and avoiding predators. We adopt the term functional grain to describe a set of functionally connected regions. In this spatial pattern, each region describes a contiguous area of the landscape within which an animal may move freely below a threshold amount of movement cost. We used telemetry data from woodland caribou Rangifer tarandus caribou to test hypothetical functional grains where connectivity was determined by the spatial configuration of resource patches (patch only), by the resistance to movement presented by landscape features (resistance only), and by a combination of the two (patch + resistance). To identify these functional grains, we used a grains of connectivity approach, and introduced a novel lattice‐based variant of this method to build the resistance only model. We developed a measure of fit that describes caribou distribution with respect to larger functionally connected regions in the grain, and used this to ask: 1) are seasonal caribou locations consistent with a random functional grain, implying that landscape connectivity has not shaped their distribution? 2) Given a functional grain model, are seasonal caribou locations distributed in larger functionally connected regions than random points, implying a response to the shape, size, and location of the connected regions. We found support for landscape connectivity influencing animal distribution using grains based on a landscape resistance model, and that support varied between behaviourally defined seasons. We also discuss how our novel lattice approach may be valuable for highly mobile mammals and other species where the identification of resource patches is a limitation. 相似文献

Because cytotoxic T-lymphocytes (CTLs) have been shown to play a role in controlling human immunodeficiency virus (HIV) infection and because CTL-based simian immunodeficiency virus (SIV) vaccines have proved effective in non-human primates, one goal of HIV vaccine design is to elicit effective CTL responses in humans. Such a vaccine could improve viral control in patients who later become infected, thereby reducing onwards transmission and enhancing life expectancy in the absence of treatment. The ability of HIV to evolve mutations that evade CTLs and the ability of these 'escape mutants' to spread amongst the population poses a challenge to the development of an effective and robust vaccine. We present a mathematical model of within-host evolution and between-host transmission of CTL escape mutants amongst a population receiving a vaccine that elicits CTL responses to multiple epitopes. Within-host evolution at each epitope is represented by the outgrowth of escape mutants in hosts who restrict the epitope and their reversion in hosts who do not restrict the epitope. We use this model to investigate how the evolution and spread of escape mutants could affect the impact of a vaccine. We show that in the absence of escape, such a vaccine could markedly reduce the prevalence of both infection and disease in the population. However the impact of such a vaccine could be significantly abated by CTL escape mutants, especially if their selection in hosts who restrict the epitope is rapid and their reversion in hosts who do not restrict the epitope is slow. We also use the model to address whether a vaccine should span a broad or narrow range of CTL epitopes and target epitopes restricted by rare or common HLA types. We discuss the implications and limitations of our findings. 相似文献

This study modelled the probability of introduction of Hyalomma marginatum into Europe by predicting the potential migratory routes of 28 bird species and the probability to carry immatures of the tick. Flyways were modelled as a spatio-temporal feature, at weekly intervals, using satellite-derived data of temperature and vegetal phenology, together with cost surfaces derived from speed and direction of the wind (years 2002–2018). The expected period of activity of tick immatures defined the probability of ticks being carried by birds along the modelled flyways. The probability of moulting of the engorged nymphs was modelled as a linear relationship of the daily sum of temperatures after tick introduction by birds. Positive probabilities of tick introduction extend the known northern range of permanent populations to central and western France, and large portions of central Europe. The flight of birds into an area and thence the risk of introduction of H. marginatum is very heterogeneous, with sites receiving “waves” of different bird species at diverse times of the year. Therefore, there is not a clear period of time for introduction, as it depends on the modelled behaviour of the bird species. The probability of introduction into Baltic and Nordic countries is small. We hypothesise that conditions of a warmer climate might support permanent populations of H. marginatum if a high number of immatures is introduced. Active surveys in risky territories, where the tick is not yet established, are advisable for rapid intervention. 相似文献

Landscape connectivity is a key issue of nature conservation and distance parameters are essential for the calculation of patch level metrics. For such calculations the so-called Euclidean and the least cost distance are the most widespread models. In the present work we tested both distance models for landscape connectivity, using connectivity metrics in the case of a grassland mosaic, and the ground beetle Pterostichus melas as a focal species. Our goal was to explore the dissimilarity between the two distance models and the consequent divergence from the calculated values of patch relevance in connectivity. We found that the two distance models calculated the distances similarly, but their estimations were more reliable over short distances (circa 500 m), than long distances (circa 3000 m). The variability in the importance of habitat patches (i.e. patch connectivity indices) was estimated by the difference between the two distance models (Euclidean vs. least cost) according to the patch size. The location of the habitat patches in the matrix seemed to be a more important factor than the habitat size in the estimation of connectivity. The uncertainty of three patch connectivity indices (Integral Index of Connectivity, Probability of Connectance and Flux) became high above a habitat size of 5 ha. Relevance of patches in maintaining connectivity varied even within small ranges depending on the estimator of distance, revealing the careful consideration of these methods in conservation planning. 相似文献

Recent evidence suggests that the core abnormality of cerebral function in schizophrenia is a disruption of functional connectivity
between diverse cerebral sites. Functional connectivity is defined as the correlation between neuronal activity at remote
sites. It can be measured using functional imaging techniques such as positron emission tomography (PET). This paper reports
an analysis using a neural network to discriminate between the patterns of functional connectivity in schizophrenic patients
and healthy subjects. The data was derived from a PET study of regional cerebral blood flow during word generation in 6 healthy
subjects and 16 schizophrenic patients with established illness, in whom the clinical diagnosis could be made with confidence.
After training on data from two healthy subjects and seven schizophrenic patients, the neural network successfully assigned
all members of a test set of four healthy subjects and nine schizophrenic patients to the correct diagnostic category. While
this result should be interpreted with caution on account of the small sample size, it indicates that neural network analysis
is potentially of value in the diagnosis of schizophrenia.
