ATP-sensitiveK+(KATP) channels are therapeutictargets for several diseases, including angina, hypertension, anddiabetes. This is because stimulation ofKATP channels is thought toproduce vasorelaxation and myocardial protection against ischemia,whereas inhibition facilitates insulin secretion. It is well known that native KATP channels are inhibitedby ATP and sulfonylurea (SU) compounds and stimulated by nucleotidediphosphates and K+channel-opening drugs (KCOs). Although these characteristics can beshared with KATP channels indifferent tissues, differences in properties among pancreatic, cardiac,and vascular smooth muscle (VSM) cells do exist in terms of the actionsproduced by such regulators. Recent molecular biology andelectrophysiological studies have provided useful information towardthe better understanding of KATPchannels. For example, native KATPchannels appear to be a complex of a regulatory protein containing theSU-binding site [sulfonylurea receptor (SUR)] and aninward-rectifying K+ channel(Kir) serving as a pore-formingsubunit. Three isoforms of SUR (SUR1, SUR2A, and SUR2B) have beencloned and found to have two nucleotide-binding folds (NBFs). It seemsthat these NBFs play an essential role in conferring the MgADP and KCOsensitivity to the channel, whereas theKir channel subunit itselfpossesses the ATP-sensing mechanism as an intrinsic property. Themolecular structure of KATPchannels is thought to be a heteromultimeric (tetrameric) assembly ofthese complexes: Kir6.2 with SUR1(SUR1/Kir6.2, pancreatic type),Kir6.2 with SUR2A(SUR2A/Kir6.2, cardiac type), andKir6.1 with SUR2B(SUR2B/Kir6.1, VSM type)[i.e.,(SUR/Kir6.x)4]. It remains to be determined what are the molecular connections betweenthe SUR and Kir subunits thatenable this unique complex to work as a functionalKATP channel.

在室内用人工饲料连续饲养和未用药剂筛选条件下,测定了采自田间对氯氟氰菊酯产生高水平抗性甜菜夜蛾Spodoptera exigua (Hübner)种群的抗药性及其多功能氧化酶系活性的变化情况。用点滴法测定不同世代3龄幼虫抗性结果为:室内F1代LD50值为 0.9672 μg/头,抗性倍数为4 836.0倍,以后各世代逐渐降低,至F43代LD50值为0.0325μg/头,抗性倍数为162.5倍,抗性水平下降了29.8倍。用浸叶法测定不同世代3龄幼虫抗性结果为:室内F1代LC50值为185.6 mg/L,抗性倍数为964.7倍,以后各世代也逐渐降低,至F43代LC50值为9.2 mg/L,抗性倍数为47.8倍,抗性水平下降了20.2倍。与敏感品系相比,该田间种群室内饲养至F43代仍处于较高的抗性水平,抗性减退缓慢,很难恢复到敏感水平。测定甜菜夜蛾田间种群室内F2、F20和F41代及敏感品系5龄幼虫中肠微粒体甲氧试卤灵-O-脱甲基酶、乙氧试卤灵-O-脱乙基酶、芳香基羟基化酶及艾氏剂环氧化酶活性,结果表明:与敏感品系相比,田间种群甲氧试卤灵-O-脱甲基酶和艾氏剂环氧化酶的活性仅F2代显著较高,F20和F41代差异不显著,乙氧试卤灵-O-脱乙基酶和芳香基羟基化酶的活性F2、F20和F41代均显著较高。结果提示甜菜夜蛾抗性水平可能与其体内微粒体多功能氧化酶系活性有密切关系。  相似文献

The 2-pyridyl containing compounds (2-Py)2NH 1, (2-Py)2PH 2, Me2Al(2-Py)2N 3, Me2Al(2-Py)2P 4, Et2Al(2-Py)2N 5, Et2Al(2-Py)2P 6 and Et2Al(2-Py)2NAlEt3 7 have been synthesized and analyzed by solid state structure determination, FT-Raman spectroscopy and theoretical calculations in order to elucidate the charge density distribution. All di(2-pyridyl) amides and -phosphides coordinate the R2Al+ fragment via both ring nitrogen atoms, but the Lewis basicity of the central two-coordinated nitrogen atom in 5 is high enough to coordinate a second equivalent AlEt3 to form the Lewis acid base adduct Et2Al(2-Py)2NAlEt3 7. Several density functionals (BLYP, B3LYP, BPW91) have been examined in relation to various basis sets (6-31G, 6-31+G, 6-31G(d), 6-31+G(d)). This computational tool facilitates the unambiguous assignment of the Raman ring vibration frequencies. The shift to higher wavenumbers proceeding from the parent di(2-pyridyl)amine 1 and di(2-pyridyl)-phosphane 2 to the metal complexes 3 and 4 indicates partial double bond localization in the ring positions 3 and 5. This effect is more pronounced in the di(2-pyridyl)amide complexes than in the phosphide. Due to the higher electronegativity of the central nitrogen atom in 3, 5 and 7 compared to the bridging two-coordinated phosphorus atom in 4 and 6 the di(2-pyridyl)amide is the harder Lewis base. In the phosphides nearly all charge density couples into the rings leaving the central phosphorus atom only attractive for soft metals.  相似文献

为了更好地了解天然草原氮素矿化对全球氮沉降背景和草原施肥管理模式的响应, 从2000年起对内蒙古典型草原羊草 (Leymu schinensis) 群落开展了长期的氮素添加实验, 分别设置对照 (N0), 添加5gNH4NO3·m-2 (N1.75) 、30gNH4NO3·m-2 (N10.5) 和80gNH4NO3·m-2 (N28) 4个氮素添加梯度。2002年, 从相邻的同时进行施肥的两个生态系统类型, 即1979年围封的样地A和1999年围封的样地B进行土壤取样, 在最佳温度 (25℃) 和最适土壤湿度 (即60%田间持水量) 下进行5周的室内培养, 并用阶段性淋溶方法研究了氮素添加对土壤氮矿化动态的影响。在A和B两个样地内, 氮素添加都显著改变了土壤的累积氮矿化量。最高氮素处理N28对应于最低的累积氮矿化量, 而低氮素处理N1.75使得累积氮矿化量达到最高。在N0和N1.75处理中, 硝态氮的含量高于铵态氮;在N28处理中, 却表现出相反的趋势。氮素添加显著降低了土壤的pH值, 但累积氮矿化量与土壤pH值、有机碳和全氮均没有显著的相关性。大多数氮素添加处理水平在样地A具有比样地B更高的土壤累积氮矿化量。  相似文献

The rates of CO2 assimilation by potted spray carnation plants(cv. Cerise Royalette) were determined over a wide range oflight intensities (45–450 W m–2 PAR), CO2 concentrations(200–3100 vpm), and leaf temperatures (5–35 °C).Assimilation rates varied with these factors in a way similarto the response of single leaves of other temperate crops, althoughthe absolute values were lower. The optimal temperature forCO2 assimilation was between 5 and 10 °C at 45 W m–2PAR but it increased progressively with increasing light intensityand CO2 concentration up to 27 °C at 450 W m–2 PARand 3100 vpm CO2 as expressed by the equation TOpt = –6.47-h 2.336 In G + 0.031951 where C is CO2 concentration in vpmand I is photo-synthetically active radiation in W m–2.CO2 enrichment also increased stomatal resistance, especiallyat high light intensities. The influence of these results on optimalization of temperaturesand CO2 concentrations for carnation crops subjected to dailylight variation, and the discrepancy between optimal temperaturesfor growth and net photosynthesis, are discussed briefly  相似文献

The C3 species wheat and rice and the C4 species maize weregrown for 2–3 weeks in controlled environment growth chambersat 20 or 30 °C day and 15 °C night temperatures. CO2assimilation rates (at 20 and 30 °C) and several leaf parametersincluding total nitrogen, soluble protein, and RuBP carboxylaseprotein were determined. When the assimilation rates under atmosphericCO2 and O2 levels were expressed on a total nitrogen basis (=nitrogen use efficiency), the C4 species maize had a greaternitrogen use efficiency than either of the two C3 species examined,regardless of the combination of temperatures used for growthor measurement of CO2 assimilation. Maize is also shown to makemore efficient use of its soluble protein and RuBP carboxylaseprotein than either wheat or rice when measurements are madeat 320 parts 10–6 CO2 and 21% O2. Atmospheric CO2 enrichmentduring CO2 assimilation measurements increased the nitrogenuse efficiency in the C3 species. In one treatment (wheat grownand measured at 20 °C), CO2 assimilation under saturatingCO2 showed nitrogen, soluble protein, and RuBP carboxylase proteinuse efficiencies equal to or greater than that of the C4 species. These data indicate that C4 species may make more efficientuse of their nitrogen, soluble protein, and RuBP carboxylaseprotein than C3 species under atmospheric CO2 conditions. Thismay be due in part to the C4 cycle and CO2-concentrating mechanismin C4 photosynthesis.  相似文献

Exposure of renal proximal tubule cells to oxalate may play an important role in cell proliferation, but the signaling pathways involved in this effect have not been elucidated. Thus the present study was performed to examine the effect of oxalate on 3H-labeled thymidine incorporation and its related signal pathway in primary cultured rabbit renal proximal tubule cells (PTCs). The effects of oxalate on [3H]thymidine incorporation, lactate dehydrogenase (LDH) release, Trypan blue exclusion, H2O2 release, activation of mitogen-activated protein kinases (MAPKs), and 3H-labeled arachidonic acid (AA) release were examined in primary cultured renal PTCs. Oxalate inhibited [3H]thymidine incorporation in a time- and dose-dependent manner. However, its analogs did not affect [3H]thymidine incorporation. Oxalate (1 mM) significantly increased H2O2 release, which was blocked by N-acetyl-L-cysteine (NAC) and catalase (antioxidants). Oxalate significantly increased p38 MAPK and stress-activated protein kinase (SAPK)/c-Jun NH2-terminal kinase (JNK) activity, not p44/42 MAPK. Oxalate stimulated [3H]AA release and translocation of cytosolic phospholipase A2 (cPLA2) from the cytosolic fraction to the membrane fraction. Indeed, oxalate significantly increased prostaglandin E2 (PGE2) production compared with control. Oxalate-induced inhibition of [3H]thymidine incorporation and increase of [3H]AA release were prevented by antioxidants (NAC), a p38 MAPK inhibitor (SB-203580), a SAPK/JNK inhibitor (SP-600125), or PLA2 inhibitors [mepacrine and arachidonyl trifluoromethyl ketone (AACOCF3)], but not by a p44/42 MAPK inhibitor (PD-98059). Reactive oxygenspecies contribute to diaphragm dysfunction in certainpathophysiological conditions (i.e., sepsis and fatigue). However, the precise alterations induced by reactive oxygen species orthe specific species that are responsible for the derangements inskeletal muscle function are incompletely understood. In this study, weevaluated the effect of the superoxide anion radical (O2·), hydroxyl radical (·OH), and hydrogenperoxide (H2O2) on maximum calcium-activatedforce (Fmax) and calcium sensitivity of the contractileapparatus in chemically skinned (Triton X-100) single rat diaphragmfibers. O2· was generated using thexanthine/xanthine oxidase system; ·OH was generated using 1 mMFeCl2, 1 mM ascorbate, and 1 mMH2O2; and H2O2 wasadded directly to the bathing medium. Exposure to O2· or ·OH significantly decreasedFmax by 14.5% (P < 0.05) and 43.9%(P < 0.005), respectively. ·OH had no effect onCa2+ sensitivity. Neither 10 nor 1,000 µMH2O2 significantly altered Fmax orCa2+ sensitivity. We conclude that the diaphragm issusceptible to alterations induced by a direct effect of ·OH andO2·, but not H2O2, on thecontractile proteins, which could, in part, be responsible forprolonged depression in contractility associated with respiratorymuscle dysfunction in certain pathophysiological conditions.

In mammals, species with high sexual size dimorphism tend tohave highly polygynous mating systems associated with high variancein male lifetime reproductive success (LRS), leading to a highopportunity for sexual selection. However, little informationis available for species with weak sexual size dimorphism. Ina long-term study population, we used parentage analysis basedon 21 microsatellite markers to describe, for the first time,variance in male lifetime breeding success (LBS) of roe deer,a territorial ungulate where males weigh less than 10% morethan females. LBS ranged from 0 to 14 (mean = 4.54, variance= 15.5), and its distribution was highly skewed, with only afew males obtaining high LBS and many males failing to breedor siring only one fawn. As predicted for polygynous specieswith low sexual size dimorphism, the standardized variance inmale LBS was low (Im = 0.75) and was only slightly higher thanthe standardized variance in female LRS (If = 0.53), suggestinga low opportunity for sexual selection. The Im value reportedhere for roe deer is much lower than values reported for highlydimorphic ungulates such as red deer (Im > 3). We suggestthat, along a continuum of opportunity for sexual selection,roe deer occupy a position closer to monogamous and monomorphicterritorial ungulates than to highly polygynous, sexually dimorphicungulates with dominance rank–based mating systems suchas harems or roving mating systems.  相似文献

The nucleocapsid protein (NCP) from Mason-Pfizer monkey virus (MPMV) contains two evolutionary invariant Cys-X2-Cys-X4-His-X4-Cys retroviral-type zinc finger structures, where the Cys and His residues provide ligands to a tetrahedrally coordinated Zn(II) ion. The N-terminal zinc finger (F1) of NCP from MPMV contains an immediately contiguous Cys in the -1 position relative to the start of this conserved motif: Cys-Cys-X2-Cys-X4-His-X4-Cys. Metal complexes of 18-amino acid peptides which model the native zinc finger sequence, SER-Cys-X2-Cys-X4-His-X4-Cys (F1_SC), and non-native Cys-SER-X2-Cys-X4-His-X4-Cys (F1_CS) and SER-SER-X2-Cys-X4-His-X4-Cys (F1_SS) sequences have been spectroscopically characterized and compared to the native two-zinc-finger protein fragment, MPMV NCP 21-80. All Co(II)-substituted peptide complexes adopt tetrahedral ligand geometries and have S-MCo(II) ligand-to-metal charge-transfer (LMCT) transition intensities consistent with three Co(II)-S bonds for F1_SC and F1_CS. The non-native F1_CS peptide binds Co(II) with KCo=1.5᎒6 M-1, comparable to that of the native complex, and 걄-fold tighter than F1_SS. Like the Co(II) derivative, the absorption spectrum of Ni(II)-substituted NCP 21-80 is most consistent with tetrahedral Ni(II) complexes with multiple thiolate donors. In contrast, Ni(II) complexes of F1_SC and F1_CS exhibit a single absorption band in the 400-550 nm region ()겨-300 M-1 cm-1), distinct in the two complexes, assignable to a degenerate d-d transition envelope characteristic of non-native square-planar coordination geometry, and an intense LMCT transition in the UV ()255ᄾ,000 M-1 cm-1). Cd(II) complexes have intense absorption in the UV (5max=233 nm), with absolute intensities consistent with 񬩈 M-1 cm-1 per Cd(II)-S bond. 113Cd NMR spectroscopy of 113Cd MPMV NCP gives '=649 ppm, consistent with S3N coordination. Co(II) and Cd(II) complexes of non-native F1_CS peptides are more sensitive to oxidation by O2, relative to F1_SC, suggestive of a higher lability in the non-native chelate. The implications of these findings for the evolutionary conservation of this motif are discussed.  相似文献

虫酰肼是目前防治甜菜夜蛾Spodoptera exigua(Hübner)的一种主要杀虫剂,为评估其抗性风险,在室内进行了抗性筛选和交互抗性的研究。采用饲料感染法,在甜菜夜蛾饲养75代期间用虫酰肼筛选62代,与起始种群相比抗性上升39.2倍,与室内敏感品系相比抗性上升141.3倍。在筛选的早、中、后期,现实遗传力h2分别为0.1075(F0~F25)、0.2780(F26~F50)和0.0538(F51~F75),整个筛选62代现实遗传力为0.1556。抗性种群筛选43次(F55)后停止用药筛选,饲养21代后,与敏感品系相比,抗性水平由63.5倍下降到21倍,抗性下降3倍。说明甜菜夜蛾具有对虫酰肼产生抗性的风险,且抗性衰退缓慢,短期内很难恢复到敏感水平。交互抗性测定结果表明,上述室内选育的抗性品系对甲氧虫酰肼具有71.4倍的高水平交互抗性,对阿维菌素具有13.1倍的中等水平交互抗性,对甲维盐、茚虫威和呋喃虫酰肼分别具有7.0、8.4和4.7倍的低水平交互抗性,但对溴虫腈交互抗性不明显(1.9倍)。结果提示: 间断交替使用虫酰肼可以延缓抗性的发展,但除溴虫腈外,虫酰肼和其他几种新型杀虫剂之间的轮用可能不是甜菜夜蛾抗性治理的有效策略。  相似文献

The present study investigates the effects of triacontanol (CH3(CH2)28CH2OH),on plant growth (root and stem), peroxidase activity (apicalmeristem tissue), and auxin destruction (apical meristem tissue)in ‘Little Marvel’ dwarf (LM) and ‘Alaska’peas (AP). Triacontanol inhibited root growth in LM comparedto untreated controls. However, root growth in AP tissue wasenhanced by 1.0 mg I–1 triacontanol and inhibited by allother treatments, in comparison to untreated controls. Wateruptake in triacontanol-treated AP plants was greater than inuntreated controls, with the converse being the case for LM.Triacontanol treatment caused an increase in peroxidase activityin both LM and AP plants compared to untreated controls. Interms of (1–14C)IAA destruction, GA3 + 0.01 mg 1–1triacontanol caused appreciable auxin breakdown (40%) in LMtissue, with GA3 + 0.1 mg 1–1 triacontanol giving a 43%decrease compared to untreated controls. In AP tissue, 10 µMGA3 increased auxin destruction by 188% whereas 0.1 mg I–1triacontanol caused a 20% decrease compared to untreated controls.The effects of triacontanol on root and stem growth, peroxidaseactivity, and auxin destruction appear to be cultivar-specific,with respect to LM and AP varieties of peas.  相似文献

根据野外调查和文献资料研究了北京北部农牧交错区C4植物的种类组成、形态功能型组成及其与生境的关系.该区共有野生C4植物68种,分布在7科,40个属,其中禾本科43种,莎草科16种,藜科5种.1年生(ANG和ANF)C4物种占C4植物总数的62%,它们在盐碱地、沙地、弃耕干扰地、湿地均超过这些生境C4植物分布的半数,在草地ANG和ANF超过30%;C4植物数量和C4/total在该地区的生境分布中有两个方向的变化:一是从草地到弃耕干扰地,两者均呈增加趋势,二是从草地到沙地和盐碱地呈现前者减少而后者增加的不同变化趋势,体现了农牧交错区植被退化的复杂性.  相似文献

It has been predicted that the concentration of CO2in the aircould double during the 21st century. Though it is recognizedthat CO2-doubling could increase yield through its effects onplant photosynthesis and stomatal behaviour, it is unclear whetherCO2-doubling will change phasic development in wheat. A phytotronstudy was conducted with two contrasting cultivars of wheat,Condor (spring) and Cappelle Desprez (winter), to determinewhether development is affected by a season-long exposure to360 and 720 ppmv CO2. Plants were vernalized for 50 d (8/4 °C,8 h photoperiod) before their exposure to the CO2treatments. There were significant differences between cultivars in theduration of different phenophases as well as in the final numberof leaves. However, CO2concentration had no effect in eithercultivar on the duration of the early developmental phase toterminal spikelet initiation, or on the final number of leaves,though CO2-doubling did slightly increase the later phase fromterminal spikelet initiation to heading in Cappelle Desprez.Condor and Cappelle Desprez also differed markedly in the dynamicsof leaf appearance. While the former had a constant rate ofleaf appearance throughout development, the latter had a fastrate initially (between leaves 1 and 7), similar to that ofCondor, which was followed by a slower rate after the appearanceof leaf 7. Overall, CO2-doubling did not significantly affectthe rates of leaf appearance nor the shape of the relationship.Phyllochron for the first seven leaves was the same for bothCO2concentrations. However, the change in phyllochron associatedwith CO2-doubling for leaves 7–12 in Cappelle Desprez,although quite small (4%), accounts for part of the slightlyincreased duration of the phase from terminal spikelet initiationto heading under high CO2concentration in that cultivar. Flowering in Lemna gibba, a long-day duckweed, can be inducedunder a short-day condition when the photoperiodic regimes areR7FR3 (7 hr red followed by 3 hr far-red), R5FR5 and R3FR7.This indicates the necessity of a proper balance between redand far-red effects for flowering. The flowering induced bythese regimes is inhibited by a brief exposure to red givenat the start of darkness and this inhibition is reversed bysubsequent exposure to far-red. Thus, the red/far-red reversibleeffect is found only at the beginning of darkness for floweringof L. gibba. However, flowering of L. gibba is promoted by a red light breakgiven near the middle of a 14 hr dark period. The promotiveeffect is not reversed by subsequent exposure to far-red, i.e.,the effect of the red break converts from inhibition to promotionas when given later in the dark period, which suggests the involvementof a timing mechanism. Effects of plant hormones were examined on the dark- and light-inducedmovements of Cassia fasciculata. Indole-3-acetic acid (IAA),gibberellic acid (GA3) and 6-benzylaminopurine (6-BAP) inhibitedthe scotonastic movement whereas abscisic acid (ABA) enhancedit. After brief treatments (5 to 30 min), the ABA effect wasinhibitory rather than promotional. Hormonal treatment in theacidic range gave the best physiological response for ABA, butthe greatest efficiency of IAA, GA3 and 6-BAP was obtained withpH values close to neutrality. Three to 5 h were needed beforeexpression of the physiological effect triggered by GA3 and6-BAP, while 5 min treatments were sufficient for IAA and ABA.Light-induced movements were largely enhanced by IAA and slightlyby GA3 but inhibited by 6-BAP and ABA. The results are discussedin relation to the ionic changes in the pulvinar motor cells,regulating leaflet movements. 以盆栽枳椇和皂荚为试材,研究了不同浓度NaCl处理(CK、0.15%、0.30%、0.45%、0.60%)对枳椇和皂荚植株生长、叶绿素荧光特性和活性氧代谢的影响.结果表明:随着NaCl浓度的提高,植株生长量、叶片叶绿素含量、Fv/Fm、ΦPSⅡ、qP均逐渐下降,而qN逐渐升高.0.15% NaCl胁迫10 d后,枳椇叶片叶绿素含量、Fv/Fm、ΦPSⅡ、qP较对照处理分别降低了19.77%、2.94%、29.03%、8.16%,差异显著(P<0.05),而皂荚与对照处理差异不显著;0.30%NaCl胁迫下,皂荚Fv/Fm和ΦPSⅡ较对照处理分别降低了1.91%、14.66%,差异显著(P<0.05);0.45%NaCl胁迫下,皂荚叶片叶绿素含量和qP与对照处理差异显著(P<0.05),分别降低了29.28%和11.36%.随着NaCl浓度的增加,皂荚SOD活性持续升高,枳椇呈先升高后降低变化,在0.15% NaCl胁迫下达最大值;两种苗木细胞POD、CAT活性均呈先升后降趋势,且皂荚的升幅大于枳椇.二者的MDA含量均呈上升趋势,且枳椇的升高幅度大于皂荚,说明枳椇产生了较严重的膜脂过氧化.由此推断,皂荚的耐盐性较强与其具有较强的抗氧化保护酶系统有关.  相似文献

Sphingosine 1-phosphate (S1P) is a platelet-derived angiogenic lipid growth factor, modulating G-protein-coupled S1P1 receptors (S1P1-R) to activate endothelial nitric oxide synthase (eNOS), as well as MAPK pathways in endothelial cells. We explored whether and how hydrogen peroxide (H2O2), a representative reactive oxygen species, alters S1P1-R expression and influences S1P signaling in cultured bovine aortic endothelial cells (BAECs). When BAECs are treated with pathophysiologically relevant concentrations of H2O2 (150 µM for 30 min), S1P1-R protein expression levels are acutely augmented by 30-fold in a dose-dependent fashion. When BAECs have been pretreated with H2O2, subsequent S1P stimulation (100 nM) leads to a higher degree of eNOS enzyme activation (assessed as intracellular cGMP content, 1.7 ± 0.2-fold vs. no H2O2 pretreatment groups, P < 0.05), associated with a higher magnitude of phosphorylation responses of eNOS and MAPK ERK1/2. PP2, an inhibitor of Src-family tyrosine kinase, abolished the effects of H2O2 on both S1P1-R protein upregulation and enhanced BAEC responses to S1P. H2O2 does not augment S1P1 mRNA expression, whereas VEGF under identical cultures leads to increases in S1P1 mRNA signals. Whereas H2O2 attenuates proliferation of BAECs, addition of S1P restores growth responses of these cells. These results demonstrate that extracellularly administered H2O2 increases S1P1-R expression and promotes endothelial responses for subsequent S1P treatment. Interactions between three growth retardants and gibberellinA9 (GA9) in Chrysanthemum morifolium Ramat. cv. Bright GoldenAnne grown in short days have revealed that the particular retardantemployed modifies the response to this gibberellin. When plants were given a foliar spray of daminozide (a substitutedsuccinamic acid), the effects of a subsequent application ofGA9 to each lateral shoot, on both stem length and the rateof inflorescence development, were relatively small. However,GA9 was highly active on plants previously sprayed with piproctanylbromide (a quaternary ammonium, piperidinium compound) or onthose given a compost drench of chlorphonium chloride (a quaternaryphosphonium retardant). In the absence of GA9 all three retardantsreduced stem length and delayed flowering. Irrespective of the extent of the responses to various dosesof GA9 (1–50 µg per shoot), a close relationshipwas maintained between internode extension and earliness offlowering, as in previous studies with retardants and othergibberellins. On plants which had received chlorphonium chlorideGA3 and GA13 were highly active and poorly active, respectively. The data suggest that daminozide could act as a retardant notonly by blocking the biosynthesis of gibberellins but also,wholly or partly, by restricting the metabolism or action ofendogenous gibberellins.  相似文献

We constructed a mathematical model for simulating the relationshipsof extracellular concentration of dissolved inorganic carbon(DIC), the rates of photosynthetic CO2 fixation and glycolatesynthesis, and the concentrations of intrachloroplast CO2 andO2 in Chlamydomonas reinhardtii. When we compared the photosyntheticrates of I0W-CO2 (air)-grown C. reinhardtii measured experimentallyand the rates simulated with the incubation conditions in themodel, the model was found to function well. The calculatedrates for glycolate synthesis also matched the measured ratesbetween 80 to 200 µM extracellular DIC, found in the presenceof 1 mM aminooxyacetate. The conformity of the calculated ratesto the measured ones of the glycolate synthesis encouraged usto estimate the O2 concentration at the active site of ribulosebisphosphate carboxylase/oxygenase; the results were 0.36 and0.40 mM at 80 and 200 µM extracellular DIC, respectively.These high concentrations of O2 were due to stimulation of photosyntheticCO2 fixation and further O2 evolution by a CO2- concentratingmechanism in the low-CO2-grown cells. These cells were calculatedto consume 43% of ATP formed photosynthetically for CO2 concentrationat 200 µM extracellular DIC. The model modified to simulatethese relationships in high-CO2 (3 to 5% CO2)-grown C. reinhardtiipredicted O2 concentration in chloroplasts to be 0.36 mM ina 1% CO2 atmosphere. This high concentration of O2 caused activeglycolate synthesis at the measured rate in the high-CO2-growncells even in the presence of 1% CO2. The comparisons of themeasured and simulated rates of photosynthesis in low- and high-CO2-grownC. reinhardtii indicated that no matter how the CO2 accumulatedin the chloroplasts, it increased the O2 concentration in theorganelles, and consequently enhanced glycolate synthesis. 1This paper is the twenty-first in a series on glycolate metabolismin Euglena gracilis.
